Abstract— Three groups of six mice each were killed 1, 4 and 7 days after an intracerebral injection of [1,2-14C]ethanolamine. The specific radioactivities of the acid-labile ethanolamine phosphoglycerides (ethanolamine plasmalogens) and of the acid-stable ethanolamine phosphoglycerides (diacyl and alkyl acyl glycerophosphoryletholamines) from myelin and microsomal fractions were determined. All of these brain ethanolamine phosphoglycerides turn over rapidly with an apparent half-life of less than 3 days. The biosynthesis of alkenyl acyl glycerophosphorylethanolamines from diacyl glycerophosphorylethanolamines in mouse brain myelin or microsomes is unlikely.  相似文献

An ADP-ribosyltransferase has been identified in compact myelin and in several white matter fractions which contain less compact myelin, fractionated on the basis of increasing protein/lipid ratios. One fraction the P3A contained the greatest activity although the activity in compact myelin was only slightly less. The ADP-ribosyltransferase activity of solubilized myelin was stimulated by increasing amounts of GTPS and was specific for the -isomer of NAD. Although ADP-ribosylation was demonstrated with the heterotrimeric G proteins in the 40–50 kDa range, the substrate for the ADP-ribosyltransferase in the 20 kDa range was identified as MBP. A thin-layer chromatographic procedure for the isolation of tissue phospholipids and their subsequent analysis is described. The method has been applied to the determination of the fatty acids of phosphoglycerides in human brain from the early fetal stage to old age. The study shows changes in the distribution and fatty acid composition of each phosphoglyceride in normal brain, although they are quite small after early childhood. A lipid-specific fatty acid pattern for each of the four major phosphoglycerides was found. Besides this, the pronounced differences between fatty acids of the lipids from the cerebral cortex and from the adjacent white matter justify speaking of a tissue-specific fatty acid pattern for brain phosphoglycerides. The phospholipids of cerebral white matter contained more monoenoic acid but much less polyunsaturated fatty acid than those of cerebral cortex. The brain phosphoglycerides also showed an age-dependent fatty acid pattern. With increasing age the concentration of the fatty acids of the linoleate family diminished while that of the linolenate family increased. Brain inositol phosphoglycerides, the fatty acid composition of which has not been studied systematically before, were characterized by a large concentration of arachidonate which was nearly as high for white as for gray matter and showed only small changes with age.  相似文献

A ‘Monte Carlo’ method was developed for characterizing matrilines of rhesus monkey infants (Macaca mulatta) born to their mothers within six similar captive social groups, according to behavioural measures, and for characterizing social groups according to their matrilines' median scores. Eight- and 16-week-old infants differed according to their groups, but not according to their matrilines, in measures of their activity in the vicinity of their mothers, and the frequencies with which social companions other than the mother initiated social contacts with them while they were out of contact with their mothers. Sixteen-week-olds differed according to their groups in the frequencies with which their mothers rejected them, and 8-week-olds in the frequencies of the social contacts they initiated with companions other than mother while off their mothers. When tested with their mothers, in a mildly disturbing situation away from other social companions, 52-week-olds differed according to their group of origin in time spent out of contact with their mothers. Different groups were seen as producing infants differing in ‘enterprise’ from their eighth week, in the sense of being ready to initiate social contacts with others while off their mothers at 8 weeks, and of being off their mothers in the test at 52 weeks. Cases where the 8-week measures did not reflect infant enterprise could be explained in terms of other aspects of the relationships involving the infants and their mothers in the social group. In particular, matrilines differed consistently in dominance status, and some mothers also received high levels of aggression from other adults. Mothers receiving high levels of aggression were more responsible for maintaining proximity with their 8-week-old infants, and their 8-week-olds were less often involved in social contacts with others.  相似文献

The phospholipid composition of the main proteolipid complexes of the nervous system was studied in myelin and synaptosomal membranes from brains of representatives of various vertebrate classes. 2. The relative content of acid phospholipids was much higher in proteolipid complexes from myelin and synaptosomal membranes of all vertebrates studied as compared to their content in the initial lipid extract (28-80% and 11-20% of total phospholipid content, respectively). 3. The relative content of acid phospholipids in proteolipid complexes of myelin membranes was much lower in brain of fishes and amphibia as compared to higher vertebrates. 4. The main acid phospholipids of proteolipid complexes was phosphatidylserine, phosphatidic acid being characteristic for myelin proteolipids and diphosphatidyl glycerol for synaptosomal proteolipids of all vertebrates studied.  相似文献

Myelin phospholipids have been examined by a separation-reaction-separation procedure for two-dimensional thin-layer chromatography on silica gel. After separation in one dimension, alk-1-enyl groups are cleaved by exposure of the plates to HCl fumes. Development in the second dimension quantitatively separates acid-labile and acid-stable phosphoglycerides as well as the aldehydes released from the acid-labile phosphoglycerides. Myelin phospholipids from the central nervous systems of the rhesus monkey, squirrel monkey, ox, and mouse contain 32-36% acid-labile ethanolamine phosphoglycerides (ethanolamine plasmalogens) and 8-14% acid-stable ethanolamine phosphoglycerides. Acid-labile choline and serine phosphoglycerides account for less than 1% of the myelin phospholipids.  相似文献

The glycerophospholipid (GPL) content and acyl group compositions of isolated brain endothelial fractions have been determined in the developing rat. During development there is a marked change in proportions of ethanolamine glycerophospholipids (EGP) to choline glycerophospholipids (CGP), the former rising while CGP falls with age. The acyl group compositions of plasmenylethanolamine (P-GPE) and 1,2-diacyl-sn-glycero-3-phosphocholine (D-GPC) alter significantly during development; both show a decline in saturated fatty acids (SFAs) and a rise in then-6/SFA ratio, in contrast to a constancy in composition of 1,2-diacyl-sn-glycero-3-phosphoethanolamine (D-GPE). The degree of change in the acyl group composition in a particular GPL fraction is related to the rate of its accumulation and to the proportional increase in concentration, fractions accumulating most rapidly or increasing markedly in concentration showing the greatest acyl group compositional change. The possible significance of the high proportion of SFAs in P-GPE and D-GPC fractions in the developing brain endothelial fraction is discussed in relation to the altering blood-brain barrier capacities observed with age.  相似文献

The purpose of this study was to examine the relationship of age to body composition, glucoregulation, activity, and energy expenditure in male and female rhesus monkeys. The animals were studied in three groups, young adults (YA, 7-9 years), middle-aged adults (MA, 13-17 years), and older adults (OA, > 23 years) adults. OA had a lower (P < 0.05) lean body mass than the YA and MA. OA also had the lowest values (P < 0.06) for energy expenditure (kJ/minute). Age-related differences (P < 0.05) were observed in time spent resting and moving. The OA spent the most time resting and the least time in vertical movement. There was a trend towards an age-related decrease in acute insulin response to glucose, while other glucoregulatory parameters were not changed with age. These results are similar to findings in humans, providing further evidence that the rhesus monkey is an appropriate model of human aging.  相似文献

The influence of hyperphenylalaninemia on the lipid composition of brain myelin has been investigated in 19-day-old chick embryos. CNP-ase activity was used as myelin marker enzyme for myelin isolation. CNP-ase activity was significantly lower in hyperphenylalaninemic myelin when compared with control. No significant differences were observed after experimental treatment in the total lipid content of myelin as well as in the proportion of cholesterol:phospholipid:galactolipid. Nevertheless, a clear increase in the percentage of esterified cholesterol was found. No appreciable alterations were observed in the phospholipid composition of brain myelin from both control and hyperphenylalaninemic chick embryos. However, the ratio of unsaturated to saturated fatty acids in serine plasmalogen and sphingomyelin was considerably increased by this treatment. This ratio in choline and ethanolamine phosphatides from treated embryos did not differ from that of controls.  相似文献

Sex differences in toy preferences in children are marked, with boys expressing stronger and more rigid toy preferences than girls, whose preferences are more flexible. Socialization processes, parents, or peers encouraging play with gender-specific toys are thought to be the primary force shaping sex differences in toy preference. A contrast in view is that toy preferences reflect biologically-determined preferences for specific activities facilitated by specific toys. Sex differences in juvenile activities, such as rough-and-tumble play, peer preferences, and infant interest, share similarities in humans and monkeys. Thus if activity preferences shape toy preferences, male and female monkeys may show toy preferences similar to those seen in boys and girls. We compared the interactions of 34 rhesus monkeys, living within a 135 monkey troop, with human wheeled toys and plush toys. Male monkeys, like boys, showed consistent and strong preferences for wheeled toys, while female monkeys, like girls, showed greater variability in preferences. Thus, the magnitude of preference for wheeled over plush toys differed significantly between males and females. The similarities to human findings demonstrate that such preferences can develop without explicit gendered socialization. We offer the hypothesis that toy preferences reflect hormonally influenced behavioral and cognitive biases which are sculpted by social processes into the sex differences seen in monkeys and humans.  相似文献

Ultrastructural disruption of myelin sheaths and a loss of myelin with age are well-documented phenomena in both the human and rhesus monkey. Age-dependent activation of calpain-1 (EC 3.4.22.52) has been suggested as a plausible mechanism for increased proteolysis in the white matter of the rhesus monkey. The present study documents activation of calpain-1 throughout brain white matter in aged animals, evidenced by immunodetection of the activated enzyme as well as a calpain-derived spectrin fragment in both tissue section and Triton X-100-soluble homogenate of subcortical white matter from the frontal, temporal, and parietal lobes. Separation of myelin fractions from brain stem tissue into intact and floating myelin confirmed previous reports of an age-related increase in activated calpain-1 in the floating fraction. Measurements of calpain-1 activity using a fluorescent substrate revealed an age-related increase in calpain-1 proteolytic activity in the floating myelin fraction consistent with immunodetection of the activated enzyme in this fraction. Double-immunofluorescence demonstrated co-localization of activated calpain-1 with human leukocyte antigen-DR (HLA-DR), a marker for activated microglia, suggesting that these cells represent the major source of the increase in activated calpain-1 in the aging brain. These data solidify the role of calpain-1 in myelin protein metabolism and further implicate activated microglia in the pathology of the aging brain.  相似文献

The extent of availability of the four major classes of phospholipids in freshly isolated bovine brain myelin has been studied using phospholipase C (Clostridium perfringens), phospholipase A (bee venom), sphingomyelase (Staphalococcus aureus) and trinitrobenzenesulphonate. The results have been compared with those of a previous study (Gwarsha et al., 1984). Taken together, the data suggest that 47–67% of the phosphatidylcholine, 45–66% of the ethanolamine-containing phosphoglycerides, 52–55% of the spingomyelin and 10–32% of the phosphatidylserine + phosphatidylinositol in the fresh myelin is available to these probes and hence may be located at the external surface of the membrane bilayer.  相似文献

Sex differences in the behavior of 2.5- to 4.5-year-old rhesus monkeys, living in two social groups approximating natural compositions, were studied over a period of 3 years. Both sexes interacted significantly more often with members of their own sex in agonistic and affiliative interactions even when total rates and durations for male and female subjects did not differ. Strong sexual segregation was also seen in proximity, nonspecific contact, and huddling partners. Males were more involved in play and sex than were females and engaged in these activities primarily with other males. Females did more grooming than males, but groomed both male and female partners. Females also appeared to interact with a wider age range of partners than did males. Although total participation in aggressive interactions did not differ between the two sexes, females used more active forms of agonistic expression than did males. These differences in the behavior of adolescent rhesus are hypothesized to lead to social bonding among adolescent males, while females remain bonded to their matri-lines, including younger males and some fully adult males associated with matrilineal relatives. Adolescent males emigrate from their natal groups but retain sociality and bond to males and females in new groups as they become adult. © 1993 Wiley-Liss, Inc.  相似文献

The efficiency of ethyleneglycol-bis (β-amino-ethyl ether) N,N′-tetra-acetic acid (EGTA) in removing possible contamination from myelin was tested. Myelin fractions were isolated in the presence or absence of EGTA. An axolemma-enriched fraction was also prepared. Gel electrophoresis showed no important alteration of the protein pattern of myelin treated with EGTA. Only a minor band of about 41,000 daltons was selectively removed when EGTA was used during the two density gradient and differential centrifugation steps. EGTA, when used in the final washes, did not remove this band. It was absent from axolemma-enriched fractions. Different hypotheses are considered to explain these findings.  相似文献
