Seeds of Vigna mungo were allowed to germinate at 27, 18 and15°C, and time-course changes of hydrolytic enzyme activitiesand the mobilization rate of reserve components in cotyledonswere studied. The seeds germinated at 27 and 18°C grew normally,whereas the growth at 15°C was markedly retarded. In cotyledonsof seedlings grown at 27 and 18°C, amylolytic and proteolyticenzyme activities increased at early stages of growth and therates of starch and protein mobilization changed correspondingto the hydrolytic enzyme activities. At 15°C the enzymeactivities increased gradually during the experimental periodof 16 days, but the reserves in cotyledons remained almost unchangeduntil the end of the experimental period. Changes of zymogram patterns of amylolytic and proteolytic activitiesin cotyledons of seedlings grownat 27, 18 and 15°C wereexamined using polyacrylamide gel electrophoresis. The intensitiesof a main band of a-amylase and at least two bands of protease(gelatin-hydrolyzing activity) increased concurrently with invitro activities of amylolytic and proteolytic enzymes. At leastthree bands of starch phosphorylase were present in cotyledonsat early stages of germination and their intensities decreasedduring the growth of seedlings at 27, 18 and 15°C. Peroxidase and glucose-6-phosphate dehydrogenase become increasinglyactive in cucumber cotyledons excised from the plant althoughthere is a fall in total protein content. The increases areinhibited by dinitrophenol, D, and L-threo-chloramphenicol,cycloheximide, puromycinj and actinomycin D, and it is concludedthat the enzymes are synthesized de novo. Disc electrophoresisof cotyledon extracts revealed three bands of peroxidase, oneof which becomes much enhanced in excised cotyledons. Citrullineaccumulates markedly in excised cotyledons. If the shoot ofcucumber plants is cut away above the cotyledons, very largeincreases in peroxidase and glucose-6-phosphate dehydrogenaseare observed although yellowing and protein loss are delayed.  相似文献

The aim of this study was to describe the dehydrin content of mature Araucaria angustifolia embryos, a species of endangered and economically important conifers, native to southern Brazil, northeastern Argentina, and eastern Paraguay. The A. angustifolia seeds have been categorized as recalcitrant. Dehydrins were studied by western blot analysis and in situ immunolocalization microscopy using antibodies raised against the K segment, a highly conserved lysine-rich 15-amino acid sequence extensively used to recognize proteins immunologically related to the dehydrin family. Western blot analysis of the heat-stable protein fraction, as estimated by 15 % SDS-PAGE, revealed three main bands of approximately 20-, 26-, and 29-kDa; when 17.5 % SDS-PAGE was used, each band resolved into two other bands. Two thermosensitive dehydrin bands of around 16 and 35 kDa were common to the axis and cotyledons, and another thermosensitive band, with molecular mass of approximately 10 kDa, was present in the cotyledons only. Following alkaline phosphatase (AP) treatment, a gel mobility shift was detected for each one of the four main bands that can be due to phosphorylation. Dehydrins were detected in all axis and cotyledon tissues using in situ immunolocalization microscopy. At the subcellular level, dehydrins were immunolocalized in the nuclei, protein bodies, and microbodies. In the nucleus, dehydrins were found to be associated with chromatin. We concluded that the gel mobility shift for the four main bands (probably due to phosphorylation), the presence of thermosensitive bands, and the specific localizations in nuclei and protein bodies provide key starting points to understand the function of dehydrins in the embryo cells of this species.  相似文献

Three cinnamyl alcohol dehydrogenase (CAD) isoenzymes were separatedfrom etiolated wheat seedlings (Triticum aestivum L.) and examinedby native gel electrophoresis. Two of these enzymes (CAD-1 andCAD-2) were purified to apparent homogeneity. They exhibiteda marked difference in substrate affinity. On sodium dodecylsulphate-acrylamide gel the isolated isoenzymes showed onlyone protein band each with an Mr 45000 and 40000 daltons, respectively,whereas on native gel two bands were identified for each protein.Isoenzymes from a variety of diploid, tetraploid, and hexaploidwheats were compared. To determine which arginine residues are responsible for band 3-mediated anion transport, we analyzed hydroxyphenylglyoxal (HPG)-modified band 3 protein in native erythrocyte membranes. HPG-modification leads to inhibition of the transport of phosphoenolpyruvate, a substrate for band 3-mediated transport. We analyzed the HPG-modified membranes by reverse phase-HPLC, and determined that arginine 901 was modified by HPG. To determine the role of Arg 901 in the conformational change induced by anion exchange, we analyzed HPG-modification of the membranes when 4,4'-dinitrostilbene-2,2'-disulfonic acid (DNDS) or diethypyrocarbonate (DEPC) was present. DNDS and DEPC fix band 3 in the outward and inward conformations, respectively. HPG-modification was unaffected in the presence of DEPC but decreased in the presence of DNDS. In addition to that, 4,4'-diisothiocyanostilbene-2,2'-disulfonic acid (DIDS), which specifically reacts with the outward conformation of band 3, did not react with HPG-modified membranes. Furthermore, we expressed a band 3 mutant in which Arg 901 was replaced by alanine (R901A) on yeast membranes. The kinetic parameters indicated that the R901A mutation affected the rate of conformational change of the band 3 protein. From these results, we conclude that the most C-terminal arginine, Arg 901, has a functional role in the conformational change that is necessary for anion transport.  相似文献

The influence of the embryonic axis and cytokinins (CKs) onreserve mobilization has been examined in yellow lupin (Lupinusluteus L. cv. JSG 6167) seed during germination and during earlygrowth for up to 9 d in the dark. The study included determinationof starch, soluble sugars, proteins, and amino acid content.Amylolytic and proteolytic enzyme activity was also measuredin untreated cotyledons with intact embryo (attached) or detachedcotyledons (embryo removed), and in detached cotyledons followingtreatment with CKs namely, dihydrozeatin, (diH)Z, and 6-benzylaminopurine,BAP. Generally, the detached cotyledons showed reduced mobilizationand decreased enzymatic activity in comparison to attached cotyledons,indicating the importance of the embryonic axis in this process.However, a rise in protease activity and free amino acid contentwas detected in 9-d-old detached cotyledons suggesting thatthe end products do not necessarily inhibit enzyme activity.While (diH)Z was partially effective in inducing reserve mobilizationand enzymatic activity in detached cotyledons, the effect ofBAP was more pronounced and appeared to replace the embryonicaxis. The embryonic axis of this species has recently been shownto synthesize CKs which are transported to the cotyledons, arehighly stabe and induce cotyledon expansion and chlorophyllsynthesis. The results of the present investigation and previousstudies from this laboratory collectively indicate that theregulation of reserve mobilization in yellow lupin seeds appearsto be mediated, at least in part, by a stimulus, probably aCK, emanating from the embryonic axis. We have shown in a previous paper that plastids of watermelon ( Citrullus vulgaris Schrad., cv. Fairfax) cotyledons differentiate into amyloplasts when the cotyledons are grown in water and into prochloroplasts when they are grown in benzyladenine (BA) solution. In the present work we have tested whether this large difference in development of the plastids is accompanied by equally conspicuous changes in their polypeptide pattern. Cotyledons were grown for 4 days in the dark either on distilled water or on 10−5 M BA. Alternatively they were transfered to 10−5 M BA after 4 days of growth in water.
Plastids of control cotyledons had a rather simple polypeptide pattern. The only prominent protein bands were the two subunits of ribulose bisphosphate carboxylase (EC 4.1.1.39). Contamination with storage protein was present. Plastids from BA-treated cotyledons had a much more complex polypeptide pattern. No storage protein contamination was observed. Polypeptide bands present only as traces in the control and having molecular weights of 32, 44, 48 and 49 kDa increased in intensity after an exposure of only 6 h to BA. The 32-kDa band seemed to be most dependent on the presence of the hormone. Contrary to what was observed in plastids, BA seemed to have no influence on the polypeptide pattern of mitochondria.  相似文献

Accumulation of chlorophyll, development of photosystem I andII activities and contents of chloroplastic components wereinvestigated in greening radish seedlings germinated and grownwith 4-thiouridine (4SU). The development of photosystem I activityprior to that of photosystem II was observed also in the 4SU-culturedgreening radish cotyledons in which chlorophyll accumulationwas inhibited up to 60–80% of that of the control. Photochemicalactivities expressed on a plastid protein basis decreased withthe increase of 4SU in the culture medium. In contrast to ferredoxinand ferredoxin-NADP reductase, which were present in significantamounts in the treated cotyledons, chloroplastic cytochromes(f, b559 and b6 decreased in the plastids from 4SU-culturedcotyledons. These results suggest that 4SU interferes in partwith protein synthesis in plastids and thereby with chloroplastdevelopment. (Received December 4, 1979; )  相似文献

在库蚊Culex pipiens品系中,非专一性酯酶活性的升高是对有机磷杀虫剂产生抗性的重要机理之一。应用SDS/PAGE,比较淡色库蚊Culex pipiens pallens抗敌百虫品系(RD)、敏感型品系(S)和抗苄呋菊酯品系(PY)中可溶性总蛋白质带型,显示RD中含有一条特异蛋白带,其它两个品系中未检出。在RD成虫匀浆液总蛋白中含量高达2.1%。分子量测定为66 kD。应用柱层析法分离得到了较纯的纯品。以α-NA为底物测得Km=64.1 mmol/L,Vmax=249.8 mmol/(L·mg·min)。与羧酸酯酶相比较:其Km值小于已报道的抗性品系及非抗性品系A-酯酶和B-酯酶。Vmax值比已报道抗性品系A-酯酶低,比B-酯酶高。较高浓度的敌百虫并不能抑制其酶活,属于A-酯酶。在昆虫体内可能主要通过结合隔离作用(sequestration)提高昆虫对有机磷的耐受性,对有机磷杀虫剂水解作用的可能性也不能排除。  相似文献

Activated platelets are known to express P-selectin, a lectin-likeadhesion receptor (CD62), through which they bind to sialylLewis X (sLex) ligands displayed on the membranes of leukocytes.To determine whether direct platelet-platelet interactions viaP-selectin/sLex interactions are also possible, we have examinedthe ganglioside extract of human blood platelets for the presenceof sLex ligands. Using the sensitive method of high-performancethin-layer chromatography (HPTLC)-immunostaining with the monoclonalantibody (mAb) CSLEX or with sialidase followed by mAbs MC480or PM81, eight sLex bands were demonstrated at R1 0.01, 0.03,0.05, 0.06, 0.08, 0.10, 0.14 and 0.21 in the solvent 45:55:10chloroform-methanol-aqueous 0.02% CaCl2. The sensitivity ofall eight bands to sialidase or endoglycoceramidase confirmedthat they were gangliosides. Comparison of the HPTLC mobilitiesand densities of platelet bands with those from five other humantissues (granulocytes, monoblasts, kidney, aortic endotheliumand erythrocytes) in three different solvents revealed threemajor bands associated with platelets: 3 (R1 0.03), 6 (0.08)and 14 (0.21). Platelet bands were demonstrated not to haveresulted from granulocyte contamination. Partial purificationof platelet sLex gangliosides by high-performance liquid chromatographyand their reaction with 14 oligosaccharide-specific mAbs (FH4,FH5, LM112-161, LM-181, A5, 1B2, BR55-2, BE2, ES4, MC631, MH04,SH34, P001 and MC813-70) revealed that band 6 is a multifucosylatedneolacto ganglioside and band 14 is a branched, disialo neolactofucoganglioside. Platelet band 3 combined the features of bothbands 6 and 14, and reacted differently than granulocyte band3. These partial structures resemble gangliosides associatedwith adhesion in other cell systems. It is concluded that plateletsexpress tissue-specific sLex gangliosides (sLex ligands). 通过利用NaBr密度梯度超速离心对粘虫Mythimna separata幼虫的血脂蛋白进行了分离及理化特性分析。查明了幼虫血淋巴内主要是高密度脂蛋白(HDLp),而低密度脂蛋白(LDLp)的含量极低。这两种脂蛋白的密度分别为HDlp 1.123g/Ml,LDLp 1.059g/mL。HDLp和LDLp蛋白含量、脂类、氨基酸的种类测定结果表明HDLp,LDLp的蛋白含量分别是25.2μg/μL和11μg/μg。HDLp水解后可获得16种氨基酸。LDLp可获得15种。氨基酸的种类比较一致,其含量变化幅度也不大。HDLp脂类部分占最多的是二酰基甘油酯(DC)和磷脂(PC)。其余的脂类依次是碳氢化合物(HC)和胆固醇?。LDLp主要是磷脂和碳氢化合物。昆虫脂蛋白的主要中性甘油酯是二酰基甘油酯。  相似文献

NAD-specific pig heart isocitrate dehydrogenase is composed of three distinct types of subunits: α, β, and γ, which have molecular weights of about 40,000 but differ in amino acid composition and in isoelectric points. When the native enzyme is subjected to polyacrylamide gel electrophoresis under nondenaturing conditions, two major protein bands with Mr values of about 360,000 (band 1) and 100,000 (band 2) and two minor bands (bands 3 and 4) with Mr values of about 40,000 are consistently present. Enzymatic activity, as detected from NADH fluorescence, is distributed throughout the protein-staining region. Analytical isoelectric focusing in urea reveals that band 1 is composed of all three subunits in roughly the normal ratio of 2α:1β:1γ, and is probably an octamer, band 2 of an equal amount of α and β and is probably dimer, while bands 3 and 4 each consist of only the monomeric α subunit. The highest enzymatic specific activity is associated with a region intermediate between octamer and dimer, which includes the 160,000 tetramer. The protein pattern resulting from isoelectric focusing under nondenaturing conditions consists of protein bands comparable in pattern to those in the presence of urea along with bands of intermediate pI values, many of which are associated with enzymatic activity. Analysis of the subunit composition of these bands supports the activity of the α species in isolation and establishes the activity of the separated β component. No activity of the isolated γ subunit species has thus far been demonstrated. However, the highest apparent specific activity is observed when at least two types of subunits are present. These studies indicate that a range of oligomeric species of the enzyme are enzymatically active and that at least three of the four subunit chains comprising the minimum complete enzyme molecule (2α:1β:1γ) possess an active site.  相似文献

The effect of N supply on soybean (Glycine max L. Merrill) seedgrowth was investigated using an in vitro liquid culture system.Sucrose was maintained at 200 mM and N was supplied by asparagineand methion-ine in a 6.25:1 molar ratio. Media N concentrationsfrom zero to 270 mM had little effect on cultured cotyledondry matter accumulation rate for 7 or 14 d, but rates approachedzero after 21 d when there was no N in the media. Only 17 mMN was required for maximum cotyledon growth rate up to 21 d.Cotyledon N accumulation and concentration increased in directproportion to the N concentration in the media. The N concentrationin cotyledons from a high protein genotype was higher than anormal genotype at all media N levels (0–270 mM). Solublesugar and oil concentrations in the cotyledons were highestat zero media N and decreased as media N increased. These datasuggest that the concept of seed N demand, which is thoughtto cause senescence in soybean, is incorrect. Soybean seedscan accumulate dry matter without accumulating N and apparentlyneed only minimal supplies of N (17 mM) to maintain the metabolicenzymes necessary to sustain dry matter accumulation. Mung bean and tomato were in vitro selected on media containing 0, 25, 50, 100 and 150 mM NaCl. Two types of media (hormone supplemented media, CB and hormone free media, MS) were used for mung bean using cotyledon explants whereas two types of explants (cotyledons and shoot apices) were used for tomato on MS media. Total-N, protein content, nitrite reductase (NiR) activity and protein protein profiles were checked in selected plants and compared to original non selected ones. NaCl at low concentrations slightly increased total-N in shoots and roots of in vitro selected mung bean and tomato whereas higher concentrations induced significant reductions. Similar increases in protein content were detected at lower concentrations with no significant effects thereover. On the contrary, NaCl gradually inhibited NiR activity. Similar responses of total-N, protein and NiR activity, but with greater magnitudes, were detected in original plants. In addition, NaCl significantly reduced dry weights of shoots and roots of either in vitro selected or, in particular, original intact plants. Moreover, electrophoresis (SDS-PAGE) of protein from shoots of either in vitro selected or intact plants showed that NaCl induced new protein bands while some others were concomitantly disappeared. The induction of one or more of the 86.4, 79, 77.6, 77 and 71.5 kDa bands following in vitro selection and/or the disappearance of the 86 kDa band from intact plants seemed necessary for mung bean resistance. Also, the presence of 86.2 kDa band and/or the loss of the 85.8 and 57.5 kDa bands might be included in tomato resistance. Of these induced bands in mung bean selected on CB media, only two bands were detected in plants selected on MS media. In tomato, two bands lost following selection from cotyledons but only one band lost following selection from shoot apices. These changes in protein pattern therefore might serve as adaptive regulators for resistance to NaCl.  相似文献

Cellular behaviour was examined during embryogenesis in Brassicanapus to test whether or not polyploidy occurs in the cotyledonsduring the phase of oil deposition. Nuclear DNA content, nuclearand cell size, and the mitotic index were measured in the cotyledonson various days post anthesis (dpa). In squashed monolayersfrom 15 dpa cotyledons, a polyploid (>5C) population wasdetected together with a substantial number of cells in G2 (4C).Nuclear volume was measured on sectioned tissues and, at 15dpa, the range of values from the cotyledons (40–500 *m3)contrasted with that in the vestigial suspensor and endosperm(50–> 600 µm3). At 15 dpa the nuclear volumedata suggest that whilst cells in the cotyledons were in Gland G2 many endosperm and suspensor cells were polyploid. Thus,polyploidy observed in the squashed monolayers was probablydue to contaminating endosperm/suspensor cells. At 25 and 35dpa, polyploidy was not detected; all cells were in Gl (2C)and cell area increased. The mitotic index peaked at 20 dpabefore declining and given the narrower distribution of nuclearvolumes at 25 and 35 dpa (50–300 µm3), these dataare consistent with cell arrest in Gl. Thus, polyploidy wasnot detected in the cotyledons of B. napus which differs fromwhat is known about cellular development in legume cotyledons. Previous work has shown that vernalization acts at two sites,one in the cotyledons and one in the shoot, in young plantsof genotype Ife Sn Hr. During the present study the size ofthe vernalization responses in both the cotyledons and shootincreased as the temperature was lowered from 17 to 3 °C.This occurred regardless of whether the treatment was givenfor the same chronological period of time or for the same physiologicalperiod of time. Vernalization treatment was effective from thetime the seeds were developing in the pods on the maternal plantuntil at least 20 leaves were expanded and became graduallymore effective as the length of the treatment was increasedfrom 2 to 5 weeks. High pre– or post–vernalizationtemperatures can reduce the cotyledon effect and to a lesserextent the shoot effect of vernalization. Devernalization occurredto a larger extent in low light intensities and darkness thanin high light intensities. No stabilization of the vernalizationeffects in the cotyledons or shoot appeared to occur at normalgrowing temperatures (15–25 °C). In order to compare the effects of cis and trans unsaturation on the structure and packing of phospholipid bilayers, infrared spectra of aqueous dispersions of 1,2-dioleoyl-sn-glycero-3-phosphocholine (DOPC) and 1,2-dielaidoyl-sn-glycero-3-phosphocholine (DEPC) were measured in a diamond anvil cell at 28 degrees C as a function of pressure up to 36 kbar. The infrared spectra indicate that DEPC and DOPC undergo pressure-induced liquid-crystalline to gel phase transitions at critical pressures of 0.7 and 5.2 kbar, respectively. Below their respective critical pressures, the infrared spectra of DOPC and DEPC are essentially indistinguishable, whereas above these pressures, there are very pronounced differences in the barotropic behavior of these two lipids. Specifically, at the 5.2-kbar transition in DOPC, there are significant changes in the frequencies, intensities, and widths of bands associated with the interfacial C = O groups, the olefinic CH = CH groups, and the terminal CH3 groups, whereas the corresponding bands of DEPC are, by contrast, relatively insensitive to the pressure-induced phase transition. The unusual band shape changes in DOPC are attributed to a unique packing arrangement of the oleoyl acyl chains required to accommodate the bent geometries of adjacent cis double bonds. Moreover, above 5 kbar in DEPC, well-defined correlation field splittings of the CH2 scissoring and rocking modes are observed, with magnitudes very similar to those observed at comparable pressures in saturated lipid systems. The absence of correlation field splittings of the corresponding bands of DOPC up to 36 kbar suggests that the bent oleoyl acyl chains are closely packed with all chains oriented parallel to each other.  相似文献
