The phytoplankton productivity and biomass of two large, freshwater Antarctic lakes (Vestfold Hills, eastern Antarctica) were investigated over a 12-month period. Crooked Lake was sampled at one site, while Lake Druzhby, a complex lake with two shallow and one deep basin, was subject to a more detailed investigation. Concentrations of chlorophyll a were usually below 1 µg l-1, indicating ultra-oligotrophic conditions. Despite periodic low nutrient levels, low temperatures (range 0.4-2.8°C) and periodic poor light climate, some degree of photosynthesis was measurable throughout the year, including the dark winter phase. Snow cover had a pronounced impact on the light climate of the water column and inhibited photosynthesis. Mean rates of carbon fixation in the 0- to 15-m water column varied between 0 and 38.47 µg C l-1 day-1 in Crooked Lake and 0.24 and 37.68 µg C l-1 day-1 in Lake Druzhby. There were significant differences in the seasonal patterns of primary production between the basins of Lake Druzhby. The shallow basins had highest productivity in August, whereas the deep basin had highest rates in summer. Chlorophyll specific rates of photosynthesis or assimilation numbers [µg C (chl. a)-1 h-1] varied between 0.05 and 44.9, and photosynthetic efficiency [µg C (chl. a)-1 h-1 µmol m-2 s-1] between 0.02 and 5.19. The data suggest that the phytoplankton of these lakes is adapted to low irradiance levels, low temperatures and nutrient limitation.  相似文献

1. Viral and microbial loop dynamics were investigated over an annual cycle in three contrasting saline Antarctic lakes – Highway Lake (salinity 4‰), Pendant Lake (salinity 19‰) and Ace Lake, a meromictic system (with a mixolimnion salinity of 18‰) in order to assess the importance of viruses in extreme, microbially dominated systems. 2. Virus like particles (VLP) showed no clear seasonal pattern, with high concentrations occurring in both winter and summer (range 0.89 × 107 ± 0.038 to 12.017 × 107 ± 1.28 mL?1). VLP abundances reflected lake productivity based on chlorophyll a concentrations. Bacterial abundances and biomass did not correlate with VLP numbers except in Pendant Lake, the most productive of the three lakes studied. 3. Pendant Lake supported the highest bacterial biomass (range Highway: 18.44 ± 1.35 to 59.43 ± 2.80 ng C mL?1; Ace: 14.42 ± 2.69 to 68.39 ± 2.95 ng C mL?1; Pendant: 31.36 ± 3.94 to 115.95 ± 4.49 ng C mL?1) so that virus to bacteria ratios (VBR) (range 30.48 ± 7.96 to 96.67 ± 8.21) were higher in Ace Lake (range 30.58 ± 3.98 to 80.037 ± 1.60) and Highway Lake (range 18.63 ± 3.12 to 126.74 ± 6.50). 4. Negative correlations occurred between VLP and cryptophytes (dominant phototrophic nanoflagellates), suggesting that they were not hosts to lytic viruses. Among the other protists only the heterotrophic nanoflagellates of Highway Lake (dominated by the marine choanoflagellate Diaphanoeca grandis) showed a positive correlation with VLP. 5. The VLP was negatively correlated with photosynthetically active radiation (PAR) and temperature, both of which increased with ice thinning and breakout, increasing viral decay. In winter VLP probably persisted in cold, dark water. 6. High VLP concentrations and high VBR (values at the upper end of those reported for marine and lacustrine systems) indicated that viruses, most of which were probably bacteriophage, are a major element within the microbial communities in extreme, saline lakes.  相似文献

1. Virus‐like particle (VLP) abundances in nine freshwater to saline lakes in the Vestfold Hills, Eastern Antarctica (68° S) were determined in December 1999. In the ultra‐oligotrophic to oligotrophic freshwater lakes, VLP abundances ranged from 1.01 to 3.28 × 106 mL–1 in the top 6 m of the water column. In the saline lakes the range was between 6.76 and 36.5 × 106 mL–1. The lowest value was found in meromictic Ace Lake and the highest value in hypersaline Lake Williams. Virus to bacteria ratios (VBR) were lowest in the freshwater lakes and highest in the saline lakes, with a maximum of 23.4 in the former and 50.3 in the latter. 2. A range of morphologies among VLP was observed, including phages with short (Podoviridae) and long tails, icosahedric viruses of up to 300 nm and star‐like particles of about 80 nm diameter. 3. In these microbially dominated ecosystems there was no correlation between VLP and either bacterial numbers or chlorophyll a. There was a significant correlation between VLP abundances and dissolved organic carbon concentration (r=0.845, P < 0.01). 4. The data suggested that viruses probably attack a spectrum of bacteria and protozoan species. Virus‐like particle numbers in the freshwater lakes were lower than values reported for lower latitude systems. Those in the saline lakes were comparable with abundances reported from other Antarctic lakes, and were higher than most values published for lower latitude lakes and many marine systems. Across the salinity spectrum from freshwater through brackish to hypersaline, VLP concentrations increased roughly in relation to increasing trophy. 5. Given that Antarctic lakes have a plankton almost entirely made up of bacteria and protists, and that VLP abundances are high, it is likely that viruses play a pivotal role in carbon cycling in these extreme ecosystems.  相似文献

A shallow, saline lake (Rookery Lake) close to the sea and surrounded by a penguin rookery was investigated during the austral spring and summer of 1996/1997. The proximity to the sea means that the lake is likely to have been formed recently during isostatic uplift. Inputs of carbon and nutrients from the penguin rookery have rendered Rookery Lake eutrophic compared with other brackish and saline lakes in the Vestfold Hills. Chlorophyll a concentration, bacterioplankton, heterotrophic nanoflagellate and phototrophic nanoflagellate abundances were all significantly higher than in other non-enriched lakes. The high productivity created seasonal anoxia during winter and spring below ice cover. The ciliate community resembled the marine community, and was dissimilar to that seen in older saline lakes within the Vestfold Hills. Thus Rookery Lake provides valuable evidence of the impact of natural eutrophication on an Antarctic lake, as well as of the evolution of the typical microbial community which dominates the older lakes of the Vestfold Hills. This study assessed the effects of salinity changes over space and time upon leaf gland secretion in Avicennia germinans trees growing naturally in an area featuring markedly seasonal rainfall. Soil ? (, soil N MPa) during the wet season was -0.95ǂ.05 and -2.12ǂ.08 at low and high salinity sites, respectively. During the dry season, these values decreased to -3.24ǂ.09 at low salinity and to -5.75ǂ.06 at high salinity. Consequently, predawn and midday plant water potential were lowered during drought at both sites. The rates of secretion (mmol m-2 h-1 ) increased during drought from 0.91ǂ.12 during the wet season to 1.93ǂ.12 at low salinity, and from 1.69ǂ.12 during the wet season to 2.81ǂ.15 at high salinity. Conversely, stomatal conductance (gs) was lowered by both salinity and drought. As xylem osmolality increased during drought, secretion tended to rise exponentially, and gs decayed hyperbolically. Thus, a trade-off is obtained between enhancement in salt secretion and control of water loss suggested by gs.  相似文献

 Samples collected from Lake Fryxell, southern Victoria Land, Antarctica in January 1992 and 1994 were analysed for the abundance of bacterioplankton and the diversity and abundance of protistan plankton. At the times of sampling, 14 ciliate species and 10 species of autotrophic flagellate were recorded. The samples contained two species of rotifer (Philodina spp.), which formed the first record of planktonic metazoans in the Dry Valley lakes of this region of Antarctica. Bacterial concentrations ranged between 1.0 and 3.8×108 l-1 in the upper oxic waters increasing to 20×108 l-1 in the anoxic waters. Heterotrophic flagellates decreased in abundance down the oxygenated water column, disappearing completely at 9 m, and ranged between 0.28 and 7.39×105 l-1 in abundance. Autotrophic flagellates were much more abundant exhibiting a number of distinct peaks down the water column (1.89–25.3×108 l-1). The ciliated protozoa were very abundant (up to 7720 l-1) in relation to flagellate and bacterial numbers, typical of oligotrophic lakes world-wide. The distribution of the protistan plankton showed marked zonation, probably in response to the differing salinity and temperature gradients in the water column. Yarrowia lipolytica KCCM50506, which transforms isobutyric acid to L-#-hydroxy isobutyric acid (L-#-HIBA), was screened. Chemostat cultures were carried out in jar fermentors at dilution rates of 0.02 hу to 0.12 hу. L-#-HIBA fermentation-regulating factors were determined to be specific growth rate, and concentrations of glucose and isobutyric acid in fermentor from analysis of steady-state data. The specific productivity of L-#-HIBA increased as the specific growth rate increased, apparently as a growth-associated type of product formation. A fed-batch culture was carried out under optimum conditions where the concentrations of glucose and isobutyric acid in the fermentor were maintained at 23 g lу and 9 g lу, respectively. The concentrations of cells and L-#-HIBA obtained at the end of fermentation were 20 g lу and 49 g lу, respectively, corresponding to 2.0 and 2.7 times more than concentrations in batch culture.  相似文献

A system for the production of transgenic plants has been developed for the Liliaceous ornamental plant Muscari armeniacum Leichtl. ex Bak via Agrobacterium-mediated transformation of embryogenic cultures. Leaf-derived embryogenic cultures were co-cultivated with each of three A. tumefaciens strains, all of which harbored the binary vector carrying the neomycin phosphotransferase II (nptII), hygromycin phosphotransferase (hpt) and intron-containing #-glucuronidase (gus-intron) genes in the T-DNA region. Following co-cultivation, the embryogenic cultures were cultured on a medium containing 500 mg l-1 cefotaxime for 1 week followed by a medium containing 75 mg l-1 hygromycin in addition to cefotaxime. After 4-5 weeks, several hygromycin-resistant (Hygr) cell clusters were produced from the co-cultivated embryogenic cultures. The highest efficiency of production of Hygr cell clusters was obtained when embryogenic cultures were inoculated with A. tumefaciens EHA101/pIG121Hm in the presence of 100 µM acetosyringone (AS) and 0.1% (v/v) of a surfactant (Tween20) followed by co-cultivation in the presence of 100 µM AS. Hygr embryogenic cultures developed into complete plants via somatic embryogenesis, and most of them were verified to be transgenic by GUS histochemical assay and polymerase chain reaction analysis. Southern blot analysis revealed the integration of one to five copies of the transgene into the genome of transgenic plants, but most of them had one or two copies.  相似文献

The isotope 15N was used to examine nitrogen dynamics in LakesFryxell and Vanda, two permanently ice-covered Antarctic lakes.Half-saturation constants for NH4+. uptake in the shallow watersof both lakes were <10 µg N l–1; uptake kineticexperiments on populations forming the deep-chlorophyll layersof these lakes showed zero-order kinetics and could not be fittedwith the Michaelis-Menten equation. Elevated uptake within thefirst few minutes following pulses of NH4+. and NO3 occurredin both lakes. NH4+ regeneration, determined from isotope dilutionexperiments, exceeded uptake at 4.6 m in Lake Fryxell, was lessthan uptake at 9 m in Lake Fryxell and was equal to uptake at10 m in Lake Vanda under the experimental conditions. NO3uptake was suppressed by NH4+ levels as low as 2 µg NH4+-N l–1 in Lake Fryxell; the suppression was strongestin the near-surface populations. Substrate-saturated C:N uptakeratios (g:g) in Lake Fryxell decreased from 8.4 near the surfaceto 1.8 at the bottom of the trophogenic zone. Overall, the nitrogendynamics in these lakes are similar to other lakes and the openocean in that biological productivity during the austral summeris supported by regenerated nutrients.  相似文献

The consumption and assimilation rates of the woodlouse Armadillidium vulgare were measured on leaf litters from five herb species grown and naturally senesced at 350 and 700 µl l-1 CO2. Each type of litter was tested separately after 12, 30 and 45 days of decomposition at 18°C. The effects of elevated CO2 differed depending on the plant species. In Medicago minima (Fabaceae), the CO2 treatment had no significant effect on consumption and assimilation. In Tyrimnus leucographus (Asteraceae), the CO2 treatment had no significant effect on consumption, but the elevated CO2 litter was assimilated at a lower rate than the ambient CO2 litter after 30 days of decomposition. In the three other species, Galactites tomentosa (Asteraceae), Trifolium angustifolium (Fabaceae) and Lolium rigidum (Poaceae), the elevated CO2 litter was consumed and/or assimilated at a higher rate than the ambient CO2 litter. Examination of the nitrogen contents in these three species of litter did not support the hypothesis of compensatory feeding, i.e. an increase in woodlouse consumption to compensate for low nitrogen content of the food. Rather, the results suggest that in herbs that were unpalatable at the start of the experiment (Galactites, Trifolium and Lolium), more of the the litter produced at 700 µl l-1 CO2 was consumed than of that produced at 350 µl l-1 because inhibitory factors were eliminated faster during decomposition.  相似文献

The uptake and transport of salt ions (Na+, Cl-), macronutrients (K+, Ca2+, Mg2+) and abscisic acid (ABA) response to increasing soil salinity were examined in 2-year-old seedlings of Populus euphratica and a hybrid, P. talassica Kom 2 (P. euphratica + Salix alba L.). Leaf burn symptoms appeared in the hybrid after 8 days of exposure to salinity when soil NaCl concentration increased to 206 mM, whereas P. euphratica exhibited leaf damage after day 21 when soil NaCl exceeded 354 mM. Leaf necrosis was the result of excess salt accumulation since the injury followed an abrupt increase of endogenous salt levels. Compared with the hybrid, P. euphratica exhibited a greater capacity to exclude salt ions from leaves under increasing salinity, especially Cl-. Salt treatment altered nutrient balance of the hybrid, leaf K+, Ca2+ and Mg2+ concentrations significantly declined and the same trends were observed in roots with the exception of K+. Although K+ levels decreased in salinised P. euphratica, increasing salinity did not affect the levels of Ca2+ and Mg2+ in leaves, but did increase the uptake of these nutrients when salt stress was initiated. NaCl-induced increase of ABA concentration in xylem sap [ABA] was observed in the two tested genotypes, however xylem [ABA] increased more rapidly in P. euphratica and a fivefold increase of xylem [ABA] was recorded after the first day of exposure to salt stress. Therefore, we conclude that the increase of Ca2+ uptake may be associated with the rise of ABA, and thus contributes to membrane integrity maintenance, which enables P. euphratica to regulate uptake and transport of salt ions under high levels of external salinity in the longer term.  相似文献

The dynamics of bacterioplankton and protozooplankton in twomaritime Antarctic lakes (Heywood Lake and Sombre Lake, SignyIsland, South Orkneys) were studied during the phase of icebreak-out (December and early January 1994/95). The lakes aresuffering animal-induced (fur seal) eutrophication, though HeywoodLake is most severely affected. Both lakes had morphologicallydiverse bacterial communities which increased during the studyperiod, reaching maxima of 80 x 108 l–1 in Heywood Lakeand 31.8 x 108 l–1 in Sombre Lake. Heterotrophic nanoflagellates(HNAN) reached a peak in late December with maxima of 40.6 x108 l–1 in Sombre Lake and 174 x 105 l–1 in HeywoodLake. Phototrophic nanoflagellates (PNAN) peaked in late Decemberafter ice loss in Heywood Lake (63 x 105 l–1), which coincidedwith a peak in a bloom of Chroomonas acuta which reached abundancesof 1.0 x 108 l–1. In Sombre Lake, ice persisted for alonger period and here PNAN reached their highest density atthe end of the study period (around 70.0 x 105 l–1). Ciliateabundance reached high levels in Heywood Lake (>60001–1),while in Sombre Lake maximum abundance was 568l–1. Protozooplanktondiversity was greater in the less-enriched Sombre Lake. Grazingrates of ciliates averaged 70.6 bacteria indiv.–1 h–1in Heywood Lake and 119.3 bacteria indiv.–1 h–1in Sombre Lake. The difference was a reflection of the differenttaxonomic make-up of the community in the lakes. HNAN grazingrates varied between 0.51 and 0.83 bacteria indiv.–1 h–1in Sombre and Heywood Lakes, respectively. Specific growth rates(r) h–1 in Sombre Lake were 0.028 for ciliates and 0.013for HNAN, and in Heywood Lake 0.010 for ciliates and HNAN 0.012.These growth rates result in doubling times ranging between38 and 69 h for ciliates and around 55 h for HNAN.HNAN grazingon bacteria was curtailed in Heywood Lake in early January asa result of predation by microcrustacean larvae feeding on theplankton. Thus, for a short phase top-down control was apparentin the dynamics of Heywood Lake, a feature uncommon in Antarcticlake ecosystems. The impact of natural eutrophication on thesesystems is discussed in relation to other unaffected Antarcticlakes.  相似文献

The nitrogen composition and sugar and auxin concentrations of callus induction medium were optimized in order to improve the regeneration of green plants from two elite oat cultivars, Aslak and Veli. For both cultivars, the production of green plantlets was doubled by optimization. However, the results obtained also clearly demonstrated that cultivars of the same species may differ drastically in their requirements for essential media components. Veli clearly required higher total amounts of nitrogen (67.8 mM) than Aslak (44.9 mM) but less maltose and 2,4-dichlorophenoxyacetic acid (28 g l-1 and 0.6 mg l-1) than Aslak (38 g l-1 and 2 mg l-1). This result indicates that the optimal production of green plantlets through embryogenesis requires that media be optimized for each cultivar separately.  相似文献

Variation in the carbon isotopic composition of ecosystem respiration ('13CR) was studied for 3 years along a precipitation gradient in western Oregon, USA, using the Keeling plot approach. Study sites included six coniferous forests, dominated by Picea sitchensis, Tsuga heterophylla, Pseudotsuga menziesii, Pinus ponderosa, and Juniperus occidentalis, and ranged in location from the Pacific coast to the eastern side of the Cascade Mountains (a 250-km transect). Mean annual precipitation across these sites ranged from 227 to 2,760 mm. Overall '13CR varied from -23.1 to -33.1‰, and within a single forest, it varied in magnitude by 3.5-8.5‰. Mean annual '13CR differed significantly in the forests and was strongly correlated with mean annual precipitation. The carbon isotope ratio of carbon stocks (leaves, fine roots, litter, and soil organic matter) varied similarly with mean precipitation (more positive at the drier sites). There was a strong link between '13CR and the vapor saturation deficit of air (vpd) 5-10 days earlier, both across and within sites. This relationship is consistent with stomatal regulation of gas exchange and associated changes in photosynthetic carbon isotope discrimination. Recent freeze events caused significant deviation from the '13CR versus vpd relationship, resulting in higher than expected '13CR values.  相似文献

Broad bean (Vicia faba), an annual plant bearing extrafloral nectaries (EFN) at the base of the upper leaves, is regularly infested by two aphid species, Aphis fabae and Acyrthosiphon pisum. EFN and A. fabae are commonly attended by the ant, Lasius niger, while Ac. pisum usually remains uninfested. Sugar concentration and sugar composition of extrafloral nectar did not change significantly after aphid infestation. The sugar concentration was significantly higher in EFN (c. 271 µg µl-1) than in the honeydew of A. fabae (37.5 µg µl-1). The presence of small A. fabae colonies had no significant effect on ant attendance of EFN, which remained at the same level as that on plants without A. fabae. Obviously, there was no significant competitive effect between the two sugar sources. We suggest that the high sugar concentration in the extrafloral nectar may outweigh the higher quality (due to the presence of melezitose) and quantity of aphid honeydew. Ants and the presence of EFN influenced aphid colony growth. While A. fabae colonies generally grew better in the presence of ants, Ac. pisum colonies declined on plants with EFN or A. fabae colonies. We conclude that EFN may provide some degree of protection for V. faba against those sucking herbivores that are not able to attract ants.  相似文献

The production of ligninolytic enzymes by the fungus Phanerochaete chrysosporium BKM-F-1767 (ATCC 24725) in laboratory-scale bioreactors was studied. One bioreactor was filled with cubes of polyurethane foam and the other with cubes of nylon sponge, in order to determine the more suitable carrier to produce high ligninolytic enzyme activities by this fungus. Both cultivations were carried out in batch. Manganese-dependent peroxidase activities about 600 U lу were achieved in the bioreactor filled with cubes of nylon sponge, while up to 500 U lу were detected in that filled with cubes of polyurethane foam. Furthermore, quite high levels of laccase appeared in both cultures: maximum activities of 114 U lу and 62 U lу were obtained on nylon and polyurethane supports, respectively.  相似文献

The stable C isotope composition ('13C) of leaf and wood tissue has been used as an index of water availability at both the species and landscape level. However, the generality of this relationship across species has received little attention. We compiled literature data for a range of conifers and examined relationships among landscape and environmental variables (altitude, precipitation, evaporation) and '13C. A significant component of the variation in '13C was related to altitude (discrimination decreased with altitude in stemwood, 2.53‰ km-1 altitude, r2=0.49, and in foliage, 1.91‰ km-1, r2=0.42), as has been noted previously. The decrease in discrimination with altitude was such that the gradient in CO2 partial pressure into the leaf (Pa-Pi) and altitude were generally unrelated. The ratio of precipitation to evaporation (P/E) explained significant variation in Pa-Pi of stemwood (r2=0.45) and foliage (r2=0.27), but only at low (<0.8) P/E. At greater P/E there was little or no relationship, and other influences on '13C probably dominated the effect of water availability. We also examined the relationship between plant drought stress (O) and '13C within annual rings of stemwood from Pinus radiata and Pinus pinaster in south-western Australia. Differential thinning and fertiliser application produced large differences in the availability of water, nutrients and light to individual trees. At a density of 750 stems ha-1, O and '13C were less (more negative) than at 250 stems ha-1 indicating greater drought stress and less efficient water use, contrary to what was expected in light of the general relationship between discrimination and P/E. The greater '13C of trees from heavily thinned plots may well be related to an increased interception of radiation by individual trees and greater concentrations of nutrients in foliage - attributes that increase rates of photosynthesis, reduce Pi and increase '13C. '13C was thus modified to a greater extent by interception of radiation and by nutrient concentrations than by water availability and the '13C-O relationship varied between thinning treatments. Within treatments, the relationship between '13C and O was strong (0.38<r2<0.58). We conclude that '13C may well be a useful indicator of water availability or drought stress, but only in seasonally dry climates (P/E<1) and where variation in other environmental factors can be accounted for.  相似文献

 The autotrophic ciliate Mesodinium rubrum Lohmann was observed during winter and spring in saline lakes ranging in salinity from 2 to 78‰ in the Vestfold Hills, Antarctica. The ciliate remained active during winter, and contained chlorophyll even though the level of light available for photosynthesis was minimal. No evidence of encystment as a means of survival during winter was observed. A seasonal study in one of the lakes, Ace Lake, revealed that M. rubrum was present throughout the year at abundances ranging from 1×104 to 3.5×105 cells l-1. During the winter period, when little light penetrated the lake’s ice cover, cells were most common immediately under the ice at 2 m, where cell numbers were typically 8×104 cells l-1. A meso-scale oceanographic grid survey was conducted during the first cruise of the Marion Offshore Ecosystem Variability Study in the upstream region of the Prince Edward Islands in austral autumn (April/May) 2001. Mesozooplankton samples, collected using a Bongo net (fitted with 200-µm and 300-µm mesh nets), were separated into three size fractions, 200-500 µm, 500-1,000 µm, 1,000-2,000 µm, by reverse filtration. Total surface (depth<5 m) chlorophyll-a concentration during the study ranged between 0.11 and 0.34 µg l-1 and was always dominated by picophytoplankton (0.45-2.0 µm). Total mesozooplankton abundance and biomass during the survey ranged between 49 and 1,512 ind. m-3 and between 0.7 and 25 mg Dwt. m-3, respectively. Throughout the survey, the 200 to 500 µm class numerically dominated the mesozooplankton community, with an average of ~69% (SD=ᆠ.3%). The dominant species in the 200- to 500-µm size fraction were the copepods, Oithona similis, Calanus simillimus and Metridia lucens, and the pteropod, Limacina retroversa. However, in terms of biomass, the 1,000- to 2,000-µm group was predominant, with dry weight values constituting an average of ~66% (SD=ᆞ.2%). The most well-represented species in this group were the carnivorous Euphausia vallentini, Thysanoessa vicina, Sagitta gazellae and Eukrohnia hamata. Three distinct groupings of stations were identified by numerical analysis. The different station groupings identified reflect changes in the relative contributions of the dominant species, as opposed to the presence/absence of species.  相似文献

By manipulating hormone levels, light intensities and temperature, we have developed an efficient leaf-disc method for the regeneration of plants via embryogenesis and for transformation in four genotypes of Vitis vinifera L. In MS basal medium supplemented with 1 mg l-1 6-benzylaminopurine (BAP) and 0.1 mg l-1 2,4-dichlorophenoxyacetic acid, leaf discs cultured for 2 weeks under dark conditions produced calli in over 80% of the cultures. These subsequently differentiated into pro-embryos and embryos only if kept under conditions of low light intensity (15 µE m-2 s-1) for 2 weeks before being transferred to conditions of high light intensity (60 µE m-2 s-1). If the calli were directly transferred to high light intensity, the differentiation into embryos was blocked and the calli turned pink. The somatic embryos germinated at a frequency of about 10% on NN basal medium and about 32% on NN medium supplemented with 1 mg l-1BAP and 0.1 mg l-1 indole-3-butyric acid. The embryos, however, germinated when pre-exposed to a low temperature of 4°C for 2 weeks. If they were transferred directly to room temperature under conditions of high light intensity (60 µE m-2 s-1), shoot buds were produced, whereas under conditions of low light intensity (15 µE m-2 s-1) secondary embryogenesis was induced. About 90-95% of the in vitro grown plantlets could be successfully transferred to soil. The above method was also applicable for developing transgenic embryos whose transgenic nature was monitored using #-glucuronidase as a reporter gene.  相似文献
