Two groups of 16 rats each were fed the same diet with 12.9 ppm Zn. Nine days after each animal was injected with65Zn for assessing fecal zinc of endogenous origin, zinc intake and excretion were determined for a six-day period at the age of about five (group I) and nine (II) weeks. At mean growth rates of 5.1 and 5.2 g/day, food consumption per gram of gain was 2.01 g in group I vs 2.86 g in II. Overall, zinc retention amounted to 21 vs 25 μg Zn/g of gain. Apparent absorption averaged 92 vs 74% of Zn intake (132 vs 189 μg/day), while true absorption averaged 98 vs 92%. It was concluded that endogenous fecal zinc excretion was limited to the indispensable loss (F em) in group I (7 μg/day), while it exceeded this minimum loss in group II (33 μg/day). True retention, which reflected total zinc utilization (true absorption times metabolic efficiency), was derived from apparent absorption plusF em (11 μg/day for group II according to the greater metabolic body size of the rats). It averaged 98% of Zn intake in group I vs 80% in group II. The mean metabolic efficiency was 100% vs 87%. The conclusion was that these marked differences between age groups in utilizing the dietary zinc reflected the efficient homeostatic adjustments in absorption and endogenous excretion of zinc to the respective zinc supply status.  相似文献

A conventional balance study with 48 male weanling rats was conducted to determine true absorption and endogenous fecal excretion of manganese (Mn) in relation to dietary Mn supply, following the procedures of a previously adapted isotope dilution technique. After 10 d on a diet with 1.5 ppm Mn, eight animals each were assigned to diets containing 1.5, 4.5, 11.2, 35, 65, or 100 ppm Mn on a dry-matter basis. Three days later, each rat was given an intramuscular54Mn injection and kept on treatment for a balance period of 16 d. Apparent Mn absorption assessed for the final 8 d, averaged 8.6 μg/d without significant treatment effects, although Mn intake ranged from 18.6 to 1200 μg/d, in direct relation to dietary Mn concentrations. Mean fecal excretion of endogenous Mn for the six treatments was 0.9, 2.7, 7.4, 11.0, 16.3, and 17.7 μg/d, respectively. These values delineate the rates to which true absorption exceeded apparent rates. True absorption, as percent of Mn intake, averaged 28.7, 15.9, 11.7, 6.1, 3.4, and 2.0, respectively, as compared with mean values of 23.9, 10.9, 6.2, 3.4, 1.2, and 0.5 for percent apparent absorption. It was concluded that both true absorption and endogenous fecal excretion markedly responded to Mn nutrition and that the reduction in the efficiency of true absorption was quantitatively the most significant homeostatic response for maintaining stable Mn concentrations in body tissues.  相似文献

At the end of a 49-d experiment with 32 growing male rats, a period of 8 d was used to determine endogenous excretion and true absorption as well as apparent absorption and retention of cobalt with the aid of the isotope dilution technique. For this purpose, a single im dose of58Co was applied at d 35 of the experiment. After that, urine and feces were collected separately from d 8 to 15 after injection of the isotope. The specific cobalt activity of the liver was used as an endogenous reference source. The basal diet provided 5.9 ppb cobalt, the different treatment groups were obtained by supplementing the diet with 0, 10, 50, 250, or 1250 ppb cobalt. The different diets were offered from the beginning of the experiment. In the balance period, apparent and true absorption as well as fecal excretion behaved similar to cobalt intake, whereas urinary excretion increased more rapidly with increasing cobalt supply. Endogenous fecal excretion accounted for 3.5 ng Co/d in the groups fed the diets without and with 10 ppb cobalt. An increase was not observed until supplementing the diet with 50 ppb cobalt. This increase between 250 and 1250 ppb cobalt was higher than the corresponding increase in the dietary cobalt supply. This indicates that endogenous fecal excretion might be more important for homeostatic regulation at a higher dietary cobalt concentration. Endogenous renal excretion as calculated from the results of the isotope dilution technique showed a similar kind of response to increasing cobalt supply as endogenous fecal loss. Nevertheless, the elimination of excessive cobalt mainly took place by adjusting urinary excretion, whereas the variations in true absorption and endogenous fecal excretion had no quantitative importance. Apparent and true absorption were on average 28.0 and 29.8%, respectively, of the cobalt intake. In the case of retention, a marked decline was observed from 19% in the depletion group to 3% with 1250 ppb cobalt, again demonstrating the importance of urinary excretion for controlling the cobalt content of the organism.  相似文献

A conventional balance study with growing rats was conducted to evaluate experimental conditions for determining endogenous fecal manganese (Mn) excretion and, hence, true Mn absorption by the isotope-dilution technique. Thirty-four rats, with a mean initial live weight of 60 g, allotted to three groups of 8 animals and one group of 10 animals, were injected intramuscularly with a54Mn tracer dose and sacrificed after 4, 8, 12, and 16 d, respectively. In liver and serum, the specific radioactivity of Mn was the lowest among the tissues analyzed and its exponential rate of decrease over the period of d 4–16 was the highest. During the 8-d period, from d 9–16, apparent Mn absorption averaged 14.1% of intake (128.5 μg Mn/d). Assuming that the specific activity of Mn in liver of d 11 or, alternatively, in serum of d 16, was on the average representative of that of endogenous Mn in feces of d 9–16, it was computed that Mn of endogenous origin accounted for 9.0 and 9.3% of the total fecal excretion of the metal, and that true absorption amounted to 21.9 and 22.1% of Mn intake, respectively.  相似文献

Copper metabolism in male Nagase analbuminaemic (NA) rats was compared with that in male Sprague Dawley (SD) rats fed purified diets containing either 5 or 100 mg Cu/kg diet. Dietary copper loading increased hepatic and kidney copper concentrations in both strains to the same extent, but baseline values were higher in the NA rats. There was no strain difference in true and apparent copper absorption nor in faecal endogenous and urinary copper excretion. NA rats had higher levels of radioactivity in kidneys at 2 hr after intraperitoneal administration of 64Cu. As based on the distribution of added 64Cu, about 70% of plasma copper appeared to be in the non-protein compartment in the NA rats, whereas in SD rats, it was only about 1%. It is concluded that the NA rats are able to maintain a relatively normal metabolism of copper, even after dietary copper challenge. In the NA rats, zinc concentrations in kidneys, liver and urinary zinc excretion were elevated when compared with SD rats. The high-copper diet did not affect tissue zinc concentrations and apparent zinc absorption in both strains of rats.  相似文献

Evidence suggests that rats can tolerate a dietary phytate/Zn molar ratio greater than 15 if the dietary Zn concentration is high. High dietary Ca exacerbates the effect of phytic acid on Zn utilization by rats. In a short term (15 d) balance trial with adult men, we observed slightly greater Zn balance when whole compared to dephytinized wheat bran was consumed (molar ratios 12 and 1.2, respectively). There was, however, greater fecal excretion of Zn during the first 5 d whole bran was consumed. In a second study, Na phytate was the major source of phytic acid and Zn balance was less when the phytate/Zn molar ratio was greater than 16 compared to 4. The difference was not significant, however, and there was evidence of physiological adjustments to maintain homeostasis when the high ratio diet was consumed. Mean Zn intake averaged 17 mg (0.26 mmole) and 11 mg (0.17 mmole) daily for the bran and Na phytate studies, respectively. The level of Zn intake may influence the response of humans to varying phytate/Zn ratios. Comparison of isotope retention studies and the balance data is discussed. Some information on the relationship of dietary Ca to the phytate/Zn effect in human diets is gathered from current literature. The phytate/Zn molar ratio is a useful index of Zn bioavailability.  相似文献

This study aimed to investigate the effects of a combination of a dairy product fermented by lactobacilli (DFL) and galactooligosaccharides (GOS) on mineral balances in growing rats with hypochlorhydria induced by a proton pump inhibitor (PPI). Three-week-old male rats were assigned to receive one of six diets: a control diet, control diets containing 1.6 or 5.0 % GOS, a DFL diet and DFL diets containing 1.6 or 5.0 % GOS for 9 days. From day 5 of the feeding period, half of the rats fed with control diets were subcutaneously administered with saline, whereas the remaining rats were administered with PPI for 5 days. Calcium (Ca), phosphorus (P), magnesium (Mg), iron (Fe) and zinc (Zn) balances were determined from days 6 to 9. PPI administration significantly decreased the apparent absorption of Ca and Fe and increased urinary P excretion, resulting in decreased Ca, Fe and P retention. GOS dose-dependently increased the apparent absorption of Ca, Mg and Fe and urinary Mg excretion and decreased urinary P excretion. DFL significantly increased the apparent absorption of Ca and Mg and urinary Mg excretion. The combination of DFL and GOS additively affected these parameters, resulting in increased Ca, P and Fe retention, and it further increased the apparent absorption and retention of Zn at 5.0 % GOS. In conclusion, the combination of DFL and GOS improves Ca, P and Fe retention in an additive manner and increases the Zn retention in growing rats with hypochlorhydria induced by PPI.  相似文献

Copper absorption was measured at two levels of dietary zinc in six healthy young men who were confined to a metabolic unit for a 75 d study of zinc utilization. A diet of conventional foods was fed, providing either 16.5 or 5.5 mg zinc and 1.3 mg copper daily. Copper absorption was determined by feeding65Cu, a stable isotope of copper, once during the 16.5 mg Zn diet and near the beginning and end of the 5.5 mg Zn diet. Apparent copper absorption averaged 48.1% when the 16.5 mg Zn diet was fed. This was significantly higher than the averages of 37.2 and 38.5% when the 5.5 mg Zn diet was fed. Absorption also differed significantly among subjects. Fecal copper did not differ between diets or among subjects. All subjects were in positive copper balance at both levels of dietary zinc. These results suggest that a dietary zinc intake slightly above the Recommended Dietary Allowance of 15 mg/d does not increase fecal copper loss and does not interfere with copper absorption.  相似文献

We examined zinc (Zn) metabolism in rats given diets containing excess calcium (Ca). Rats were given phytate-free diet containing 5 g Ca/kg (control), 12.5 g Ca/kg, or 25 g Ca/kg for 4 wk in Experiment 1. The dietary treatment did not affect Zn concentration in the plasma, testis, kidney, spleen and liver; however, Zn concentration in the femur and its cortex was significantly higher in rats given diet containing 25 g Ca/kg than in other rats. Rats were given phytate-free diet containing 5 g Ca /kg or 25 g Ca /kg for 4 wk in Experiment 2. After 12-h food deprivation, rats were given a diet extrinsically labeled by 67Zn with dysprosium as a fecal marker for 4 h. Feces were collected from 1 d before administration of the labeled diet to 5 d after administration. Excess Ca did not affect the true absorption of Zn and its endogenous excretion but increased femoral Zn. These results suggest that excess Ca improves Zn bioavailability without affecting Zn absorption when diets do not contain phytate.  相似文献

The fecal monitoring technique for measuring the absorption of Zn, Mn, Se, and Fe was studied in eight male piglets (mean±SEM birthweight (bw)=1695±50 g) using high resolution gamma spectrometry. Four d old piglets were fed a complete liquid milk diet for five d prior to the orogastric administration of an isotope dose (75Se,54Mn,59Fe, and65Zn) equilibrated with the liquid milk diet.51CrCl3 was used as a fecal marker but was found to be partially absorbed. Stool samples were collected daily for 15 d, counted, and then the daily fecal excretion was calculated. Results indicate that endogenous excretion for each of the isotopes was not constant but decreased exponentially with time. The pattern of endogenous excretion varied between elements. An improved method for calculating the endogenous excretion was therefore developed. This method is based on the pattern of endogenous excretion in three-four d old male piglets (mean±SEM bw=2060±75 g) injected intravenously with the same isotopes and on the level of endogenous excretion in orally fed animals in the postabsorptive phase of excretion. These findings have important implications for the estimation of endogenous excretion in future fecal monitoring absorption studies in order to minimize underestimation of true absorption.  相似文献

The major purpose of this study was to determine whether acute or chronic Pb exposure would increase urinary excretion of zinc in the rat. Four groups of unanesthetized rats were given 0, 0.03, 0.3, or 3 mg Pb (as acetate) kg intravenously, and urinary excretion of zinc, sodium, and potassium was monitored for 6 h. Only at the highest dose was urinary Zn excretion significantly elevated; there were no significant changes in sodium and potassium excretion at any dose. Two other groups of rats were studied for 9 weeks in metabolism cages before and during administration of either 500 ppm Pb (as acetate) or equimolar Na acetate in the drinking water. Two days after Pb treatment and continuing through day 35, Zn excretion was elevated in the Pb-exposed animals; beyond this day, zinc excretion became similar in the two groups. The difference in Zn excretion was not the result of lower water intake by the Pb-treated animals. At sacrifice (70 days after starting Pb exposure), Pb-exposed animals had lower Zn content of the plasma and testis, but there was no difference in kidney Zn. Plasma renin activity was significantly higher in Pb-exposed animals. We conclude that chronic Pb exposure in rats can result in some degree of decreased tissue zinc, which is, at least in part, secondary to increased urinary losses of zinc.  相似文献

1. The maintenance crude protein (CP) requirements of subadult nilgai antelope (Boselaphus tragocamelus) were determined by nitrogen balance. Four isocaloric pelleted diets, ranging from 8.4 to 23.8% CP, were fed. 2. Subadults maintained a positive nitrogen balance on all diets. Linear regression showed that 95.8% of diet CP was truly digested and estimated metabolic fecal nitrogen (NFN) excretion at 0.388 gN/W0.75/day. 3. Estimates for endogenous urinary nitrogen excretion and maintenance nitrogen requirement, derived through linear regression, were erroneous. Consequently, the former was calculated based on metabolic size (0.126 gN/W0.75/day) and added to MFN to obtain an 0.514 gN/W0.75/day estimate of required nitrogen for maintenance. This value is between the levels recommended for maintenance of yearling deer and cattle. 4. Given the same level of dry matter intake (68.6 g/W0.75/day) and true CP digestibility, study animals would be able to meet N equilibrium on a 4.89% CP (dry matter basis) diet. 5. Based on data from other sources, natural diets containing 6.98% CP would need to be consumed by free-ranging subadults to maintain body weight.  相似文献

Histidine has been reported to affect body zinc status by increasing urinary zinc excretion. The effects of experimental histidinemia on distribution of65Zn in anesthetized rats were studied. Infusion ofl-histidine at a rate sufficient to raise plasma concentrations to approximately 2mm for 6h starting 48 h after a single intraperitoneal65Zn injection did not alter65Zn activities in a variety of tissues when compared with anesthetized uninfused animals. However, plasma65Zn and erythrocyte65Zn were decreased, and liver65Zn was increased. If65Zn was injected intravenously during histidine infusion, net accumulation of zinc by some tissues was increased, but uptake by others was reduced relative to uninfused animals. In all cases, however, uptake expressed relative to plasma65Zn levels was increased when allowance was made for the more rapid fall in plasma65Zn during histidine infusion. Similar infusions ofd-histidine produced quantitatively similar effects. Since enzymatic mechanisms and amino acid carriers would be expected to show stereoselectivity, such processes are unlikely to be involved in the zinc distribution changes described. The possibility of zinc transport by a hitherto unidentified carrier is discussed. These experiments confirm that histidinemia can affect zinc status, but any associated changes in urinary zinc excretion do not seem adequate to account for the tissue changes found.  相似文献

The objective of this investigation was to evaluate the effect of 47 mg zinc supplementation on deficiency of zinc in rats during 98 d of restriction of motor activity (hypokinesia), which appeared by higher plasma zinc concentration. One Hundred 13-week-old Sprague-Dawley male rats weighing 360–390 g were used to perform the studies: They were equally divided into four groups: 1. Unsupplemented control animals (UCA); 2. Unsupplemented hypokinetic animals (UHA); 3. Supplemented control animals (SCA); and 4. Supplemented hypokinetic animals (SHA). For the simulation of the effect of hypokinesia (HK), the UHA and SHA were kept in small individual cages made of wood, which restricted their movements in all directions without hindering food and water intake. The SCA and SHA received daily with their food an additional amount of zinc. Before and during the experimental period of 98 d, plasma, urinary and fecal zinc, balance of zinc, food intake, and body weight were determined at different intervals. In the SHA and UHA, the concentration of zinc in plasma, and the elimination of zinc in urine and feces increased significantly when compared with the SCA and UCA, whereas the balance of zinc was negative. The body weight and food intake decreased significantly in the SHA and UHA when compared with the SCA and UCA. The increased plasma concentration of zinc in both the SHA and UHA groups was in contrast to the observed hypozincnemia during prolonged immobilization as during prolonged hospitalization. This reaction suggests that there may be some other mechanisms that are affecting the process of control and regulation of zinc metabolism during prolonged HK. It was concluded that exposure to prolonged restriction of motor activity of rats induces significant increases in plasma concentration, fecal and urinary elimination of zinc in the presence of negative zinc balance and regardless the daily intake of large amounts of zinc with their food, leading to zinc deficiency.  相似文献

The effects of histidine on the zinc status are controversial. In mice, we studied the effects of a moderate histidine supplement on the regulation of the zinc status using subcutaneously administered 65Zn. In animals fed a zinc-adequate diet, histidine supplement did not cause changes in the zinc status (zinc concentrations, 65Zn tissue distribution, and tissue specific activities). Neither effects on the regulation of the zinc status (65Zn retention, excretion and biological half-life) could be demonstrated. However, the combination of a low zinc diet and moderate histidine supplementation caused changes in the regulation of the zinc status (lower 65Zn retention, associated with increased fecal excretion and a shorter biological half-life), aggravating the dietary deficiency (lower bone zinc, a shift in the 65Zn tissue distribution). Reviewing the literature, it seems that only a molar histidine/zinc ration of 2,000 or higher will cause zinc deficiency.  相似文献

We studied the effects of dietary inclusion of freeze-dried goat and cow milk on the utilization of copper, zinc and selenium, and on the metabolic fate of copper and zinc, in rats using a standard (non-milk) control diet recommended by the American Institute of Nutrition and diets based on goat or cow milk. For animals given the goat milk diet, the apparent digestibility coefficient (ADC) of copper is similar to that obtained with the standard diet and higher than that in animals given the cow milk diet. The copper balance was higher among the rats given the goat milk and the standard diets than among those given cow milk. The ADC and retention of zinc and selenium were higher for the goat milk diet than for the other two diets. The copper content in the kidneys and in the femur was greater when the animals consumed a goat milk diet than a cow milk diet. Zn deposits in femur, testes, liver, kidney, heart and longissimus dorsi muscle were greatest with the goat-milk diet, followed by the standard diet and were lowest for the rats given cow-milk diet. This study shows that the goat-milk has an important and beneficial effect on the bioavailability of copper, zinc and selenium.  相似文献

Four lactating dairy cows were used in two experiments to study the effects of the roughage to concentrate ratio in the diet on nitrogen balance, plasma urea, urinary urea, milk urea and urinary purine derivatives. The use of the allantoin to creatinine ratio in spot samples of urine as an index of the urinary allantoin excretion was also evaluated. Four isoenergetic and isonitrogenous diets were formulated according to a 2 × 2 factorial arrangement. Factor I was concentrate content. The roughage to concentrate ratios were 65:35 and 35:65 for the high roughage and high concentrate diets, respectively. Factor II was fat content, which was 2.8% and 5.8% for the low and high fat diets, respectively. In Experiment 1 cows were fed diets with low fat content, and in Experiment 2 cows were fed diets with high fat content. In both experiments, diets were fed according to a change-over design. Nitrogen balance was not affected by the treatments. In cows fed high concentrate diets the amount and the proportion of nitrogen excreted in milk, as well as milk production was higher than in cows fed the high roughage diets. In both experiments, as an overall effect, the urea levels in plasma, urine and morning milk were higher, although the total urinary excretion of urea was lower, for the high concentrate diets. Urinary allantoin excretion was higher, although not significantly in Experiment 1, for the high concentrate diets. The allantoin to creatinine ratio in spot samples of urine showed the same pattern as the total allantoin excretion. Urinary creatinine excretion appeared to be affected by the diet.  相似文献

Six African green monkeys were labeled intravenously with [1,2-(3)H]cholesterol while consuming a cholesterol-free liquid formula diet. The plasma cholesterol specific activity was compared with the specific activity of the biliary cholesterol and bile acids and with the fecal neutral steroids in order to determine whether the traditional isotopic balance method was valid for the calculation of endogenous cholesterol excretion. The specific activity of biliary cholesterol and bile acids averaged 10-15% lower than plasma cholesterol specific activity. Fecal cholesterol and coprostanone specific activities were similar to that of the biliary cholesterol, but the specific activity of fecal coprostanol was approximately 25% lower. This suggests that biliary cholesterol and bile acids were derived from a pool of hepatic cholesterol that did not completely equilibrate with the whole body exchangeable cholesterol pool. In addition, there was further reduction in the specific activity of coprostanol, the major fecal neutral steroid, presumably by cholesterol synthesized in the lower intestine and preferentially converted to coprostanol. As a result, the traditional isotopic balance procedure underestimated endogenous neutral steroid excretion by 46% and bile acid excretion by 31% in African green monkeys fed the cholesterol-free diet. Within 7 days after the addition of 1 mg cholesterol/kcal to the diet, the specific activities of plasma and biliary cholesterol and biliary bile acids were identical and there was no difference in the specific activities of the individual fecal neutral steroids. Thus, the traditional isotopic balance procedure (DPM fecal neutral steroids + bile acids/specific activity [DPM/mg] plasma cholesterol) can be used for calculation of endogenous cholesterol excretion in cholesterol-fed animals during the nonsteady state when plasma cholesterol concentrations are rapidly increasing, as well as after a new steady state has been achieved.-Henderson, G. A series of experiments was conducted to determine how dietary protein, alfalfa, or zeolite influence the excretory patterns of zearalenone (Z), a uterotropic mycotoxin synthesized by Fusarium fungi. Rats were fed diets containing 16.3% casein, 40% casein, 11.2% casein + 25% alfalfa, or 25% casein + 25% alfalfa. Also fed were diets containing 0, 1, 2, or 5% anion exchange zeolite. Tracer doses of [3H]Z were administered either as a constituent of the diet or as a topical application on the skin at the base of the skull. When Z was administered orally, no differences were seen in the fraction of the dose excreted in urine or feces as a result of varying dietary levels of alfalfa and protein. Topical doses resulted in rats fed 25% casein + 25% alfalfa or 40% casein excreting more Z in urine than those fed 25% alfalfa or 16.3% casein. Fecal excretion of Z was greatest for rats fed 25% casein + 25% alfalfa whereas rats fed 40% casein excreted more fecal Z than those fed 16.3% casein. Feeding Z to rats receiving dietary zeolite resulted in a positive correlation between dietary zeolite and fecal excretion of Z but a negative correlation with urinary excretion of Z. Topical administration of Z produced a positive correlation between dietary zeolite and fecal Z excretion but no effect on urinary excretion. It may be concluded that protein and alfalfa treatments alleviate Z toxicosis through increased metabolism whereas zeolite binds Z in the digestive tract to prevent absorption.  相似文献
