Continuous ethanol fermentation of cheese whey powder solution was realized using pure culture of Kluyveromyces marxianus (DSMZ 7239) at hydraulic residence times (HRT) between 12.5 and 60 h. Sugar utilization, ethanol and biomass formation were investigated as functions of HRT. Effluent sugar concentration decreased, but percent sugar utilization, ethanol and biomass concentrations increased with HRT. Ethanol productivity was maximum (0.745 gE l−1h−1) at an HRT of 43.2 h where the biomass productivity was almost minimum (0.18 gX l−1 h−1). The ethanol yield coefficient was almost constant at 0.4 gE g−1S up to HRT of 43.2 h and the growth yield coefficient was minimum at HRT of 43.2 h. Kinetic models were developed and the constants were determined by using the experimental data.  相似文献

AIMS: To test the suitability of cheese whey powder (CWP) solution for ethanol fermentation and to compare performances of different Kluyveromyces marxianus strains for ethanol fermentation from CWP solution. METHODS AND RESULTS: Batch ethanol fermentation of cheese whey (CW), CWP and lactose solutions with the same initial sugar contents were compared by using two different K. marxianus strains and the CWP solution was found to be the most suitable substrate. CWP solution was fermented to ethanol using three different yeast strains and DSMZ-7239 was found to be the most suitable one yielding the highest rate and extent (3.3%, v/v) of ethanol formation. CONCLUSIONS: CWP solution and K. marxianus strain of DSMZ-7239 were found to be more suitable for ethanol fermentation with the highest ethanol yield when compared with the other substrates and the yeast strains tested. SIGNIFICANCE AND IMPACT OF THE STUDY: CWP can be used as a concentrated form of CW for ethanol fermentations with considerable advantages.  相似文献

Due to its high content of lactose and abundant availability, cheese whey powder (CWP) has received much attention for ethanol production in fermentation processes. However, lactose‐fermenting yeast strains including Kluyveromyces marxianus can only produce alcohol at a relatively low level, while the most commonly used distiller yeast strain Saccharomyces cerevisiae cannot ferment lactose since it lacks both β‐galactosidase and the lactose permease system. To combine the unique aspects of these two yeast strains, hybrids of K. marxianus TY‐22 and S. cerevisiae AY‐5 were constructed by protoplast fusion. The fusants were screened and characterized by DNA content, β‐galactosidase activity, ethanol tolerance, and ethanol productivity. Among the genetically stable fusants, the DNA content of strain R‐1 was 6.94%, close to the sum of the DNA contents of TY‐22 (3.99%) and AY‐5 (3.51%). The results obtained by random‐amplified polymorphic DNA analysis suggested that R‐1 was a fusant between AY‐5 and TY‐22. During the fermentation process with CWP, the hybrid strain R‐1 produced 3.8% v/v ethanol in 72 h, while the parental strain TY‐22 only produced 3.1% v/v ethanol in 84 h under the same conditions.  相似文献

Cheese whey powder (CWP) solution with different CWP or sugar concentrations was fermented to ethanol in a continuous fermenter using pure culture of Kluyveromyces marxianus (DSMZ 7239). Sugar concentration of the feed CWP solution varied between 55 and 200 g l−1 while the hydraulic residence time (HRT) was kept constant at 54 h. Ethanol formation, sugar utilization and biomass formation were investigated as functions of the feed sugar concentration. Percent sugar utilization and biomass concentrations decreased and the effluent sugar concentration increased with increasing feed sugar concentrations especially for the feed sugar contents above 100 g l−1. Ethanol concentration and productivity (DP) increased with increasing feed sugar up to 100 g l−1 and then decreased with further increases in the feed sugar content. The highest ethanol concentration (3.7%, v v−1) and productivity (0.54 gE l−1 h−1) were obtained with the feed sugar content of 100 g l−1 or 125 g l−1. The ethanol yield coefficient (YP/S) was also maximum (0.49 gE gS−1) when the feed sugar was between 100 and 125 g l−1. The growth yield coefficient (YX/S) decreased steadily from 0.123 to 0.063 gX gS−1 when the feed sugar increased from 55 to 200 g l−1 due to adverse effects of high sugar contents on yeast growth. The optimal feed sugar concentration maximizing the ethanol productivity and sugar utilization was between 100 and 125 g l−1 under the specified experimental conditions.  相似文献

Dark fermentative hydrogen gas production from cheese whey powder solution was realized at 55°C. Experiments were performed at different initial biomass concentrations varying between 0.48 and 2.86 g L?1 with a constant initial substrate concentration of 26 ± 2 g total sugar (TS) per liter. The highest cumulative hydrogen evolution (633 mL, 30°C), hydrogen yield (1.56 mol H2 mol?1 glucose), and H2 formation rate (3.45 mL h?1) were obtained with 1.92 g L?1 biomass concentration. The specific H2 production rate decreased with increasing biomasss concentration from the highest value (47.7 mL g?1 h?1) at 0.48 g L?1 biomass concentration. Total volatile fatty acid concentration varied beetween 10 and 14 g L?1 with the highest level of 14.2 g L?1 at biomass concentration of 0.48 g L?1 and initial TS content of 28.4 g L?1. The experimental data were correlated with the Gompertz equation and the constants were determined. The most suitable initial biomass to substrate ratio yielding the highest H2 yield and formation rate was 0.082 g biomass per gram of TS. © 2012 American Institute of Chemical Engineers Biotechnol. The simultaneous saccharification and fermentation (SSF) of pretreated sugar cane leaves to produce ethanol using a cellulolytic enzyme complex from Trichoderma reesei QM 9414 and Saccharomyces cerevisiae NRRL-Y-132 was optimized. Enzymic saccharification parameters were evaluated prior to SSF studies. A 92% conversion of 2·5% substrate (alkaline hydrogen peroxide pretreated) to sugars was achieved at 50°C and pH 4·5, using T. reesei cellulase (40 FPU/g substrate), in 48 h. The pretreated substrate was then subjected to an SSF process using the cellulase complex and S. cerevisiae cells. Optimization of the SSF system is described.  相似文献

Both intrinsic and observed kinetic investigations for those ethanol fermentations using self-flocculated yeast strains have been hindered by the lack of real online monitoring techniques and proper characterization methods for the flocs. An optical detecting technique, the focused beam reflectance measurement probe developed by Lasentec (Redmond, WA) was inserted into a fermentor to monitor the floc chord length distributions. Using a simulating system composed of the floc-buffer suspensions, the total floc chord length counts per second were directly correlated with the floc biomass concentrations so that the floc biomass concentrations can be in situ detected. Furthermore, a characterization method of the flocs was established by properly weighted treatments of the detected floc chord length distributions. When a real yeast floc ethanol fermentation system was detected during its intrinsic kinetic investigations in which the floc size needed to be controlled at a level of micrometer scale to eliminate inner mass transfer limitations, it was found and validated that CO(2) produced during fermentation exerted significant disturbances. By applying 1/length-weighted treatment, these disturbances were effectively overcome.  相似文献

Cheese whey contains a high organic content and causes serious problems if it is released into the environment when untreated. This study aimed to investigate the optimum condition of lactic acid production using the microaerobic sequencing batch reactor (microaerobic SBR) in a nonsterile system. The high production of lactic acid was achieved by immobilized Lactobacillus plantarum TISTR 2265 to generate an acidic pH condition below 4.5 and then to support single-cell protein (SCP) production in the second aerobic sequencing batch reactor (aerobic SBR). A hydraulic retention time (HRT) of 4 days and a whey concentration of 80% feeding gave a high lactic acid yield of 12.58 g/L, chemical oxygen demand (COD) removal of 62.38%, and lactose utilization of 61.54%. The microbial communities in the nonsterile system were dominated by members of lactic acid bacteria, and it was shown that the inoculum remained in the system up to 330 days.  相似文献

Taking continuous ethanol fermentation with the self‐flocculating yeast SPSC01 under very high concentration conditions as an example, the fermentation performance of the yeast flocs and their metabolic flux distribution were investigated by controlling their average sizes at 100, 200, and 300 µm using the focused beam reflectance online measurement system. In addition, the impact of zinc supplementation was evaluated for the yeast flocs at the size of 300 µm grown in presence or absence of 0.05 g L?1 zinc sulfate. Among the yeast flocs with different sizes, the group with the average size of 300 µm exhibited highest ethanol production (110.0 g L?1) and glucose uptake rate (286.69 C mmol L?1 h?1), which are in accordance with the increased flux from pyruvate to ethanol and decreased flux to glycerol. And in the meantime, zinc supplementation further increased ethanol production and cell viability comparing with the control. Zinc addition enhanced the carbon fluxes to the biosynthesis of ergosterol (28.6%) and trehalose (43.3%), whereas the fluxes towards glycerol, protein biosynthesis, and tricarboxylic acid cycle significantly decreased by 37.7%, 19.5%, and 27.8%, respectively. This work presents the first report on the regulation of metabolic flux by the size of yeast flocs and zinc supplementation, which provides the potential for developing engineering strategy to optimize the fermentation system. Spontaneously hypertensive rats (SHR) were administered either 2.4 g/kg ethanol or an isocaloric glucose daily for 4 weeks and the levels of norepinephrine (NE), epinephrine (EP), dopamine (DA), serotonin (5-HT) and 5-hydroxyindoleacetic acid (5-HIAA) in different brain regions were determined. Results indicated a 3-fold increase in NE level in brain stem and hypothalamus and more than 2-fold increase in DA in corpus striatum in alcohol-treated rats as compared to controls. There was a significant increase in the level of DA in the corpus striatum but the levels in cerebral cortex, brain stem and hippocampus were decreased instead. Decreases in 5-HT levels were found in hypothalamus, brain stem, cortex and cerebellum of alcohol-treated brain as compared to untreated controls. These results indicate alterations of the biogenic amine contents in different regions of the SHR brain after chronic ethanol ingestion. Since stimulated release of biogenic amines in the SHR brain has been implicated in the regulation of blood pressure, changes due to ethanol ingestion may be a risk factor in hypertensive patients.  相似文献

【目的】探讨植物发酵液提取物(plant fermentation extract,PFE)对铜绿假单胞菌生物膜的抑制作用,为临床上铜绿假单胞菌感染相关疾病的治疗提供参考。【方法】通过划线法分离临床标本中的铜绿假单胞菌并进行鉴定,通过报告菌株测定铜绿假单胞菌的毒力因子,采用试管法和激光共聚焦扫描显微镜测定生物膜的形成。【结果】在分离出的16株铜绿假单胞菌中,PFE对PA007菌株的作用效果最好,1%PFE显著降低PA007菌株生物膜、绿脓菌素和N-(3-oxododecanoyl)-HSL(3-oxo-C12-HSL)的产量(P0.05)。同时,也显著降低Las A蛋白酶的活性以及持留菌存活率(P0.05)。荧光定量PCR实验结果表明PFE能显著抑制las I和pqs A基因的表达(P0.05)。【结论】PFE具有抗铜绿假单胞菌感染能力,在临床上铜绿假单胞菌感染疾病的治疗中具有巨大的潜在价值。  相似文献

The feasibility of using soy skim, a co-product of the aqueous processing of soybeans, in ethanol production from corn was evaluated. Specific growth rates were compared when Saccharomyces cerevisiae was grown in soy skim and peptone-yeast extract media supplemented with glucose. Such soy skim was proved to be a good nitrogen source for yeast growth. Next, fermentation of dry-ground corn to ethanol using soy skim as the media was simulated on 1.5-L scale. Replacing water with soy skim increased the initial ethanol production rates by 4-32% while final ethanol yield was about 39 g/100 g dry corn, similar to the result when water was used. Solid and protein contents in the finished beer increased with the addition of soy skim. Thus, replacing water in corn-ethanol fermentation with soy skim is feasible, and may improve the economics of both aqueous soybean processing and corn ethanol fermentation.  相似文献

Experiments were conducted on14C-sorbitol, fructose, and glucose uptakeinto flesh discs, and sorbitol efflux from thediscs, with and without ABA application toexamine the effect of abscisic acid (ABA) onsugar accumulation in peach fruit flesh at thestart of the maturation stage in relation tomembrane transport. Total uptake of14C-sorbitol, fructose, and glucose intoflesh discs was effectively promoted by ABA ata concentration of 10–5 M. PCMBS(p-chloromercuribenzensulfonicacid)-sensitive uptake, which was considered ascarrier-mediated uptake, of sorbitol into thediscs was clearly stimulated by ABA at10–5 M, compared with glucose andfructose uptake. Sorbitol efflux from the discsacross the tonoplast was restricted by ABA at10–5 M. ABA application todeveloping fruit increased sugar accumulationin the fruit. Estimated ABA concentration inthis fruit was approximately 10–5 M. These results indicate that sugar accumulationin peach fruit flesh is stimulated by ABA at aconcentration of 10–5 M both invitro and in vivo. ABA stimulatesuptake of sugars, especially sorbitol, into theflesh by enhancing carrier-mediated transportpossibly across both tonoplast and plasmamembrane.  相似文献

Consolidated bioprocessing (CBP) is believed to be a potentially cost-efficient and commercially viable way to produce cellulosic biofuels. In this study, we have evaluated the performance of the CBP organism Clostridium phytofermentans (ATCC 700394) on AFEX-treated corn stover (AFEX-CS). Fermentation conditions including temperature, inoculation size, nutrients, and initial pH were investigated. At optimal conditions with 0.5% (w/w) glucan loading of AFEX-CS, C. phytofermentans hydrolyzed 76% of glucan and 88.6% of xylan in 10 days. These values reached 87% and 102% of those obtained by simultaneous saccharification and co-fermentation (SSCF) using commercial enzymes and S. cerevisiae 424A. Ethanol titer for CBP was found to be 2.8 g/L which was 71.8% of that yielded by SSCF (3.9 g/L). Decomposition products from AFEX-CS helped to increase ethanol yield somewhat during CBP. Particle size played a crucial role in the enhancement of sugar conversion by CBP.  相似文献

Abstract

Polyhydroxyalkanoates (PHAs) are intracellular carbon and energy storage reserve material stored by gram-negative bacteria under nutrient limitation. PHAs are best alternative biodegradable plastics (bio-plastics) due to their resemblance to conventional synthetic plastic. The present study investigated the synergistic effect of nutritional supplements (amino acid and vitamin) on the PHA production by Alcaligenes sp. NCIM 5085 utilizing a sugar refinery waste (cane molasses) under submerged fermentation process. Initially, the effect of individual factor on PHA yield was studied by supplementing amino acids (cysteine, isoleucine, and methionine), vitamin (thiamin), and cane molasses at varying concentration in the production medium. Further, the cultivation medium was optimized by varying the levels of cane molasses, methionine and thiamin using response surface methodology to enhance the PHA yield. The maximum PHA yield of 70.89% was obtained under the optimized condition, which was then scaled up on 7.5?L-bioreactor. Batch cultivation in 7.5?L-bioreactor under the optimized condition gave a maximum PHA yield and productivity of 79.26% and 0.312 gL?1 h?1, respectively. The PHA produced was subsequently characterized as PHB by FTIR. PHB extracted was of relatively high molecular weight and crystallinity index. DSC analysis gave Tg, Tm, and Xc of 4.2, 179?°C and 66%, respectively. TGA analysis showed thermal stability with maximized degradation occurring at 302?°C, which is above the melting temperature (179?°C) of the purified polymer. The extracted polymer, therefore, possessed desirable material properties to be used in food packaging.  相似文献

External nutrient supplementation and detoxification of hydrolysate significantly increase the production cost of cellulosic ethanol. In this study, we investigated the feasibility of fermenting cellulosic hydrolysates without washing, detoxification or external nutrient supplementation using ethanologens Escherichia coli KO11 and the adapted strain ML01 at low initial cell density (16 mg dry weight/L). The cellulosic hydrolysates were derived from enzymatically digested ammonia fiber expansion (AFEX)-treated corn stover and dry distiller's grain and solubles (DDGS) at high solids loading (18% by weight). The adaptation was achieved through selective evolution of KO11 on hydrolysate from AFEX-treated corn stover. All cellulosic hydrolysates tested (36-52 g/L glucose) were fermentable. Regardless of strains, metabolic ethanol yields were near the theoretical limit (0.51 g ethanol/g consumed sugar). Volumetric ethanol productivity of 1.2 g/h/L was achieved in fermentation on DDGS hydrolysate and DDGS improved the fermentability of hydrolysate from corn stover. However, enzymatic hydrolysis and xylose utilization during fermentation were the bottlenecks for ethanol production from corn stover at these experimental conditions. In conclusion, fermentation under the baseline conditions was feasible. Utilization of nutrient-rich feedstocks such as DDGS in fermentation can replace expensive media supplementation.  相似文献

番茄果实糖酸类物质的含量及比例直接影响其风味品质,前期研究表明,适宜浓度的外源5-氨基乙酰丙酸(ALA)能够促进果实的成熟并提高其芳香品质。该试验为探究外源ALA对番茄果实发育及其糖酸品质的影响,以番茄‘原味1号’(Solanum lycopersicum cv.Yuanwei No.1)品种为试材,于第4穗果授粉后10 d果实表面喷施0、100和200 mg·L^(-1)的ALA溶液,分析ALA对番茄果实形态、果皮色泽及果实不同部位组织中糖、酸类物质组分及含量的影响。结果表明:(1)外源ALA溶液能显著促进番茄果实横径、纵径的增加,提高果实单果重,还显著降低果实硬度,促进果实软化,提升果实口感,并提高了果实V_(C)和可溶性固形物含量。(2)果实不同部位组织(包括果肉、小柱和隔膜)糖类物质组分含量测定结果显示,外源ALA处理能够显著提高果实可溶性总糖含量(包括果糖、葡萄糖和蔗糖),并有利于糖类物质向果肉中积累。(3)在有机酸类物质中,除酒石酸含量增加外,外源ALA处理均能不同程度地降低果实各部位组织中酸类物质含量,从而显著提高番茄果实果肉部位糖酸比,提升果实糖酸品质。研究发现,在番茄果实发育过程中外源施用200 mg·L^(-1) ALA不仅能够促进果实发育及着色,提高单果重,提升果实的外观品质,还有利于果实糖酸品质的形成。  相似文献

为确定食物蛋白对长爪沙鼠行为和生理特征的影响,我们采用两种不同蛋白含量(9% 和27% )的食物饲喂不同年龄的雄性长爪沙鼠,测定了不同食物条件下长爪沙鼠的体重、睾丸重、脾脏重、血清抗体、社会行为和气味吸引力等指标。结果发现,食物蛋白含量没有影响长爪沙鼠的社会行为、生理特征及幼体气味的选择,但对成体气味的被选择程度影响显著,提示食物质量与其配偶选择过程相关联。  相似文献
