目的克隆猪白细胞介素18(pIL-18)成熟蛋白基因,并在植物乳杆菌(Lb.plantarum)NC8中进行表达。方法通过RT-PCR方法从猪脾脏细胞中扩增出pIL-18成熟蛋白基因,克隆到T载体pMD18-T后测序;将阳性基因片段克隆至大肠埃希菌-乳酸菌穿梭表达载体pSIP-409构建重组表达载体pSIP-409-IL-18,进行酶切和PCR鉴定;应用电穿孔技术将其转化至Lb.plantarum NC8中,经SppIP诱导表达后,进行SDS-PAGE及Western-blot分析。结果经测序,pIL-18成熟蛋白基因核苷酸长度为579 bp,编码193个核苷酸;酶切和PCR鉴定证明成功构建了重组表达载体pSIP-409-IL-18;SDS-PAGE及Western-blot分析表明重组菌表达了18 kD的融合蛋白,该重组蛋白可以与鼠抗猪IL-18多克隆抗体反应。结论成功克隆了pIL-18成熟蛋白基因,并获得有生物活性的pIL-18重组乳酸菌,为研制开发IL-18重组乳酸菌制剂奠定基础。  相似文献

GumC is one of nine enzymes involved in the biosynthesis of fastidian gum, an exopolysaccharide produced by Xylella fastidiosa that may be linked directly to the pathogenicity of the microorganism. GumC may be responsible for gum polymerization or secretion through the membrane of X. fastidiosa. To perform structure and functions studies, we developed an expression system for the production of GumC as a fusion protein with maltose binding protein (MBP) using pMAL-c2x vector. The GumC-MBP fusion protein was expressed as a 94 kDa protein, which strongly reacts with anti-MBP antibodies. GumC-MBP was isolated by affinity chromatography through an amylose column and used to produce antibodies against the fusion protein. After the enzymatic cleavage of MBP, GumC was purified on a Q Sepharose Fast Flow column. GumC showed a molecular weight corresponding to the expected one (52 kDa) and its N-terminal sequence was identical to that deduced from the DNA. The shape of the circular dichroism spectrum was compatible with a folded protein that contains alpha-helical regions in its structure. Therefore, in this study we describe, for the first time, the production of GumC recombinant protein.  相似文献

The hemA gene encoding 5-aminolevulinic acid synthase (ALAS) was cloned from the genomic DNA of photosynthetic bacterium Rhodopseudomonas palustris KUGB306. The deduced protein (ALAS) of this gene contained 409 amino acids. The hemA gene was subcloned into an expression vector pGEX-KG and the encoded protein was overexpressed as a fusion protein with glutathione-S-transferase (GST) in Escherichia coli BL21. The recombinant ALAS was purified and isolated free of the fusion partner (GST) by affinity purification on glutathione-Sepharose 4B resin and cleavage of the purified fusion protein by thrombin protease. The optimum pH and temperature of the recombinant ALAS was found to be at pH 7.5-8.0 and 35-40 degrees C, respectively. The Km value of the enzyme was 2.01 mM for glycine and 49.55 microM for succinyl-CoA. The enzyme activity was strongly inhibited by Pb2+, Fe2+, Co2+, Cu2+, and Zn2+ at 1 mM, but slightly affected by Mg2+ and K+. The recombinant ALAS required pyridoxal 5'-phosphate (PLP) as a cofactor for catalysis. Removal of this cofactor led to complete loss of the activity. Ultraviolet-visible spectroscopy with the ALAS suggested the presence of an aldimine linkage between the enzyme and PLP.  相似文献

在肝脏疾病的基因治疗过程中,为把目的基因定向导入靶细胞,构建了具有靶向性的逆转录病毒的包装细胞.即应用RT-PCR方法分别反转录并扩增env,pres2基因,将它们分别克隆至pGEM-T载体上,经测序正确后,将一目的基因亚克隆在pcDNA3.1(-)表达载体上,然后将另一目的片段从T载体上切下,克隆至经同样酶切的重组表达载体中。从而成功地构建了肝细胞靶向性系统的表达载体pcDNA3.1(-)-env-pres2和pcDNA3.1(-)-pres2-env。  相似文献

目的构建表达绿色荧光蛋白融合人核糖核酸酶抑制因子的表达载体pEGFP—C1—hri,为探讨人核糖核酸酶抑制因子抗肿瘤作用的分子机制打下基础。方法用亚克隆法,将目的片段从表达载体pGEX-6p-1-hri克隆到pEGFP-C1上,用双酶切筛选得到阳性重组质粒pEGFP—C1—hri,用脂质体法将其瞬时转染到小鼠黑色素瘤细胞B16中,在荧光显微镜下检测其表达。结果pEGFP—C1—hri中插入了hri序列,绿色荧光高效表达于B16细胞浆中。结论pEGFP—C1—hri表达载体已成功构建。  相似文献

A lambda placMu1 insertion was made into araE, the gene for arabinose-proton symport in Escherichia coli. A phage containing an araE'-'lacZ fusion was recovered from the lysogen and its restriction map compared with that of the 61-min region of the E. coli genome to establish the gene order thyA araE orf lysR lysA galR; araE was transcribed toward orf. A 4.8-kilobase SalI-EcoRI DNA fragment containing araE was subcloned from the phage lambda d(lysA+ galR+ araE+) into the plasmid vector pBR322. From this plasmid a 2.8-kilobase HincII-PvuII DNA fragment including araE was sequenced and also subcloned into the expression vector pAD284. The araE gene was 1416-base pairs long, encoding a hydrophobic protein of 472 amino acids with a calculated Mr of 51,683. The amino acid sequence was homologous with the xylose-proton symporter of E. coli and the glucose transporters from a human hepatoma HepG2 cell line, human erythrocytes, and rat brain. The overexpressed araE gene product was identified in Coomassie-stained sodium dodecyl sulfate-polyacrylamide gel electrophoresis gels of cell membranes as a protein of apparent Mr 35,000 +/- 1,150. 应用重叠延伸PCR方法扩增HIV-1 TAT蛋白转导结构域(PTD)与鼠源神经肽Y(NPY)的融合基因,克隆目的片段并插入酵母表达载体pPICZαA,构建成重组表达质粒pPICZα-PTD-NPY.PCR和酶切鉴定及测序正确后,经限制性内切酶Sac Ⅰ线性化重组表达质粒并通过电转化整合到巴斯德毕赤酵母菌GS115的染色体基因组中.阳性重组酵母菌用含1%甲醇的培养基诱导其分泌表达.经过120 h的诱导,取上清浓缩除盐后进行SDS-PAGE电泳,表明该系统成功表达了PTD-NPY融合蛋白,Western blotting实验证实表达产物具有特异性.获得真核表达的PTD-NPY融合蛋白,为下一步的应用研究提供了物质基础.  相似文献

通过RT-PCR方法直接从猪脾脏淋巴细胞中扩增出猪白细胞介素18(pIL-18)成熟蛋白基因的cDNA, 克隆到pGEM-T载体, 构建重组质粒pGEM-T-IL18, 转化E.coli JM109感受态细胞, 取PCR和酶切鉴定为阳性的重组质粒进行序列测定。测序结果表明, pIL-18成熟蛋白基因核苷酸长度为474 bp, 编码157个氨基酸。将其克隆到表达载体pGEX6P-1中, 构建重组质粒pGEX-IL18, 转化E.coli BL21感受态细胞, 用IPTG诱导表达。重组菌菌体裂解物SDS-PAGE可检测到相对分子质量为45 kD的重组蛋白, 占菌体总蛋白的28%左右, 以包涵体形式存在。对包涵体进行洗涤, 用MTT法测定表明, 重组蛋白能明显刺激猪脾脏T淋巴细胞增殖反应的活性。  相似文献

目的:构建重组原核表达载体pGEX-5x-1-p65,诱导GST-p65融合蛋白的表达并观察其包涵体的显微结构.方法:应用PCR技术扩增得到p65全长序列,并亚克隆至带有GST标签的pGEX-5x-1载体中.经酶切、测序鉴定后,在原核细胞中诱导表达GST-p65融合蛋白并将诱导后的菌体制作透射电镜标本,观察菌体内部显微结构.结果:成功构建表达载体pGEX-5x-1-p65,原核细胞中诱导表达、凝胶电泳后未见可溶性融合蛋白的高效表达.透射电镜观察到在承载有重组载体的菌体内部出现大量高电子密度的包涵体.结论:成功构建了原核表达载体pGEX-5x-1-p65,电子显微镜观察并证实在原核细胞内p65蛋白诱导表达形成包涵体.  相似文献

通过PCR从已构建的猪源戊型肝炎病毒全基因克隆扩增ORF3全基因,将扩增产物插入到pMD18-T载体中,亚克隆至原核表达载体pET28a(+),构建pET28a-ORF3表达载体,转入E.coli BL21 (DE3),IPTG诱导表达。Ni-NTA层析柱纯化表达蛋白,用SDS-PAGE、免疫印迹、ELISA等方法分析鉴定表达产物。结果成功扩增到345 bp的目的基因;构建了重组表达载体pET28a-ORF3;转化宿主菌E.coli BL21 (DE3)后表达产物的相对分子质量在6.50~16.5 kDa之间,与预期表达的目的蛋白相对分子质量相符;表达的目的蛋白能与阳性猪源和人源血清发生特异性反应,证实其具有较好的反应原性。  相似文献

以米根霉菌基因组DNA为模板,根据GenBank上已公布的米根霉L-乳酸脱氢酶基因(ldhL)序列设计特异性引物,PCR扩增得到963 bp的DNA片段,经序列分析后将其亚克隆到原核表达载体pET30a上,构建成重组质粒pET30a-ldhL.将pET30a-ldhL转化到BL21感受态细菌中,经IPTG诱导表达后进行SDS-PAGE分析,可见约43 kD的与预期大小一致的目的蛋白条带,结果表明ldhL基因在大肠杆菌中进行了表达,经酶活分析产物的酶活力为98 U/mL,证明了表达产物具有预期的酶活性,这为进一步研究利用乳清为发酵原料高产L-乳酸的米根霉基因工程菌株奠定了基础.  相似文献

Mal+ lacZ operon fusions, inducible by maltose, were isolated in Escherichia coli, strain MC4100. One fusion strain, SF1707, was analyzed in detail. This fusion did not map in any of the known genes of the malA or malB region, but its expression was under control of malT, the positive regulator gene of the maltose regulon. The gene in which the fusion occurred mapped between xyl and mtl at 80 min on the linkage map and was transcribed clockwise. We define this gene as malS. The malS-lacZ fusion was transferred onto a phage lambda vector and the 5' portion of malS was subcloned into pBR322. The resulting plasmid was used as a probe to identify the intact malS gene in a lambda library of E. coli chromosomal HindIII fragments. The phage that hybridized with the probe contained a 12-kilobase insert. The malS containing portion was subcloned into pBR322 as a 4-kilobase ClaI-HindIII fragment. This plasmid directed the malT and maltose-dependent synthesis of a periplasmic protein of 66,000 apparent molecular weight. The purified enzyme hydrolyzed maltodextrins longer than maltose including cyclic dextrins. The primary products of hydrolysis were glucose, maltose, and maltotriose, even when maltotetraose was used as a substrate. These properties differentiate this periplasmic enzyme from the cytoplasmic amylomaltase and define it as an alpha-amylase.  相似文献

鸡传染性支气管炎病毒(Infectious bronchitis virus,IBV)是禽类一种变异性很强的冠状病毒,为获得高度纯化的IBV核蛋白,建立监测IBV抗体的方法,通过RT-PCR从IBV中扩增IBV-N基因,定向连接到表达载体pGEX-KG中,转化E. coli BL21(DE3)菌株,诱导获得表达产物,SDS-PAGE分析产物大小、Western blot分析其免疫学活性,通过亲和层析法获得高度纯化的表达蛋白,建立一种检测IBV抗体的方法,并应用于临床监测。结果显示,IBV-N基因全长1230bp,GST融合蛋白表达产物大小约为80kDa,表达量约占菌体总蛋白的25%,具有良好的免疫学活性;通过GST亲和层析柱成功获得纯化蛋白,以该蛋白包被酶标板,成功地建立了一种检测血清中IBV抗体的间接ELISA方法。研究表明重组N蛋白作为IBV诊断抗原,具有制备简便、特异性强、敏感性高、重复性好的特点,可用于该病的临床监测,为该病疫情监测、发病机制研究奠定了基础。  相似文献

以猪A组轮状病毒mRNA为模板,应用反转录聚合酶链式反应(RT-PCR)技术,扩增了1194bp的Vp6基因,通过T-A克隆技术,将PCR产物克隆至克隆载体pGEM-TVector中,构建克隆质粒pGEM-T-Vp6。用SacⅠ和KpnⅠ双酶切pGEM-T-Vp6和以胸苷酸合成酶基因(thymidylate synthase,thyA)为选择压力的非抗生素抗性的穿梭表达载体pW425t,并将纯化的Vp6基因亚克隆至表达载体pW425t中,构建出可以在乳酸菌与大肠杆菌之间穿梭表达的原核表达重组质粒pW425t-Vp6。将pW425t-Vp6转化至thyA基因缺陷型的大肠杆菌感受态E.coliX13中,经生长功能弥补筛选阳性克隆,通过SDS-PAGE分析,可见约44.88kD的融合蛋白。由Western blot分析,表明该蛋白具有与轮状病毒多克隆抗体的反应原性,从而为pW425t-Vp6在乳酸菌受体菌株中表达提供理论基础和实验依据。  相似文献

通过基因工程的方法构建奈瑟氏淋球菌表面蛋白A(Neisseria gonorrhoeae surface protein A,nspA)和大肠杆菌不耐热肠毒素B亚单位(B subunit of Escherichia coli heat-labile enterotoxin,ltB)融合基因的原核表达载体,对其进行表达与鉴定,为后续融合蛋白LTB-NspA的生物活性分析及其作为淋球菌粘膜免疫疫苗的研究奠定基础.用PCR法从标准菌株分别扩增出nspA、ltB基因,用重组PCR法通过接头将ltB与nspA融合,将其插入pET-30a中,转入BL21中表达.经测序、SDS-PAGE和Western blot分析,证实成功构建了1tB-nspA融合基因的原核表达载体,并在BL21中表达.ltB-nspA融合基因的成功表达,为进一步研究其生物活性及淋球菌粘膜免疫疫苗的研究奠定了一定基础.  相似文献

A xylanase gene (xynZF-2) from the Aspergillus niger XZ-3S was cloned and expressed in Escherichia coli. The coding region of the gene was separated by only one intron with the 68 bp in length. It encoded 225 amino acid residues of a protein with a calculated molecular weight of 24.04 kDa plus a signal peptide of 18 amino acids. The amino acid sequence of the xynZF-2 gene had a high similarity with those of family 11 of glycosyl hydrolases reported from other microorganisms. The mature peptide encoding cDNA was subcloned into pET-28a(+) expression vector. The resultant recombinant plasmid pET-28a-xynZF-2 was transformed into E. coli BL21(DE3), and finally the recombinant strain BL21/xynZF-2 was obtained. A maximum activity of 42.33 U/mg was gained from cellular of E. coli BL21/xynZF-2 induced by IPTG. The optimum temperature and pH for recombinant enzyme which has a good stability in alkaline conditions were 40 °C and 5.0, respectively. Fe3+ had an active effect on the enzyme obviously.  相似文献

构建嗜麦芽寡养单胞菌D2株荧光素样单加氧酶基因表达克隆载体,并进行融合表达。PCR扩增出嗜麦芽寡养单胞菌D2菌株基因组DNA中包含单加氧酶基因约1300bp的核酸片段,将其克隆到T载体pMD-18中进行序列测定,所得序列申请并获得GenBank登记号(GQ122330)。DNA star软件分析发现该基因片段中含有一个996bp的完整开放读码框架(ORF),与GenBank中收录的S.maltophilia R551-3(CP001111/GenomeProject17107)和K279a(AM743169)的MO基因核酸序列同源性分别为90%和89%,氨基酸序列同源性分别为93%和90%。根据该ORF序列设计分别含有BamHⅠ和HindⅢ酶切位点的表达克隆扩增引物,PCR扩增、双酶切后将产物亚克隆到pET32a载体中,经过双酶切验证,证实成功获得了表达重组载体pET32a/MO;将其转化宿主菌E.coli BL21,IPTG诱导后成功表达出54.2ku的MO融合蛋白,为该酶进一步的功能研究和开发奠定了基础。  相似文献

The whole sec2 DNA fragment was obtained by polymerase chain reaction with total genomic DNA extracted from Staphylococcus aureus. The newly discovered gene contains 717 bp (GenBank Accession number AY450554) and encodes 239 amino acids in accordance with the protein sequence reported in GenBank. The sec2 gene was fused to anti-Her-2 scFv gene with a DNA linker at the upstream to construct the fusion gene of immunotoxin which was subcloned to expression vector pET-32a(+) and expressed efficiently in Escherichia coli. The purified fusion immunotoxin could target the HER-2 overexpressed by breast tumor cell SK-Br-3 in vitro and demonstrate tumor-inhibition effect on SK-Br-3.  相似文献
