 Our objective was to evaluate the relative importance of gradients in light intensity and the isotopic composition of atmospheric CO2 for variation in leaf carbon isotope ratios within a Pinus resinosa forest. In addition, we measured photosynthetic gas exchange and leaf carbon isotope ratios on four understory species (Dryopteris carthusiana, Epipactus helleborine, Hieracium floribundum, Rhamnus frangula), in order to estimate the consequence of the variation in the understory light microclimate for carbon gain in these plants. During midday, CO2 concentration was relatively constant at vertical positions ranging from 15 m to 3 m above ground. Only at positions below 3 m was CO2 concentration significantly elevated above that measured at 15 m. Based on the strong linear relationship between changes in CO2 concentration and δ13C values for air samples collected during a diurnal cycle, we calculated the expected vertical profile for the carbon isotope ratio of atmospheric CO2 within the forest. These calculations indicated that leaves at 3 m height and above were exposed to CO2 of approximately the same isotopic composition during daylight periods. There was no significant difference between the daily mean δ13C values at 15 m (–7.77‰) and 3 m (–7.89‰), but atmospheric CO2 was significantly depleted in 13C closer to the ground surface, with daily average δ13C values of –8.85‰ at 5 cm above ground. The light intensity gradient in the forest was substantial, with average photosynthetically active radiation (PAR) on the forest floor approximately 6% of that received at the top of the canopy. In contrast, there were only minor changes in air temperature, and so it is likely that the leaf-air vapour pressure difference was relatively constant from the top of the canopy to the forest floor. For red pine and elm tree samples, there was a significant correlation between leaf δ13C value and the height at which the leaf sample was collected. Leaf tissue sampled near the forest floor, on average, had lower δ13C values than samples collected near the top of the canopy. We suggest that the average light intensity gradient through the canopy was the major factor influencing vertical changes in tree leaf δ13C values. In addition, there was a wide range of variation (greater than 4‰) among the four understory plant species for average leaf δ13C values. Measurements of leaf gas exchange, under natural light conditions and with supplemental light, were used to estimate the influence of the light microclimate on the observed variation in leaf carbon isotope ratios in the understory plants. Stable carbon isotope signatures are often used as tracers for environmentally driven changes in photosynthetic δ13C discrimination. However, carbon isotope signatures downstream from carboxylation by Rubisco are altered within metabolic pathways, transport and respiratory processes, leading to differences in δ13C between carbon pools along the plant axis and in respired CO2. Little is known about the within-plant variation in δ13C under different environmental conditions or between species. We analyzed spatial, diurnal, and environmental variations in δ13C of water soluble organic matter (δ13CWSOM) of leaves, phloem and roots, as well as dark-respired δ13CO213Cres) in leaves and roots. We selected distinct light environments (forest understory and an open area), seasons (Mediterranean spring and summer drought) and three functionally distinct understory species (two native shrubs—Halimium halimifolium and Rosmarinus officinalis—and a woody invader—Acacia longifolia). Spatial patterns in δ13CWSOM along the plant vertical axis and between respired δ13CO2 and its putative substrate were clearly species specific and the most δ13C-enriched and depleted values were found in δ13C of leaf dark-respired CO2 and phloem sugars, ~?15 and ~?33 ‰, respectively. Comparisons between study sites and seasons revealed that spatial and diurnal patterns were influenced by environmental conditions. Within a species, phloem δ13CWSOM and δ13Cres varied by up to 4 ‰ between seasons and sites. Thus, careful characterization of the magnitude and environmental dependence of apparent post-carboxylation fractionation is needed when using δ13C signatures to trace changes in photosynthetic discrimination.  相似文献

The assimilation of 14CO2 into the C4 acids malate and aspartate by leaves of C3, C4 and C3–C4 intermediate Flaveria species was investigated near the CO2 compensation concentration * in order to determine the potential role of phosphoenolpyruvate (PEP) carboxylase (EC 4.1.1.31) in reducing photorespiration in the intermediates. Relative to air concentrations of CO2, the proportion of CO2 fixed by PEP carboxylase at * increased in all six C3–C4 intermediate species examined. However, F. floridana J.R. Johnston and F. ramosissima Klatt were shown to be markedly less responsive to reduced external CO2, with only about a 1.6-fold enhancement of CO2 assimilation by PEP carboxylase, as compared to a 3.0- to 3.7-fold increase for the other C3–C4 species examined, namely, F. linearis Lag., F. anomala B.L. Robinson, F. chloraefolia A. Gray and F. pubescens Rydb. The C3 species F. pringlei Gandoger and F. cronquistii A.M. Powell exhibited a 1.5- and 2.9-fold increase in labeled malate and aspartate, respectively, at *. Assimilation of CO2 by PEP carboxylase in the C4 species F. trinervia (Spreng.) C. Mohr, F. australasica Hook., and the C4-like species F. brownii A.M. Powell was relatively insensitive to subatmospheric levels of CO2. The interspecific variation among the intermediate Flaverias may signify that F. floridana and F. ramosissima possess a more C4-like compartmentation of PEP carboxylase and ribulose-1,5-bisphosphate carboxylase/oxygenase (EC 4.1.1.39) between the mesophyll and bundle-sheath cells. Chasing recently labeled malate and aspartate with 12CO2 for 5 min at * resulted in an apparent turnover of 25% and 30% of the radiocarbon in these C4 acids for F. ramosissima and F. floridana, respectively. No substantial turnover was detected for F. linearis, F. anomala, F. chloraefolia or F. pubescens. With the exception of F. floridana and F. ramosissima, it is unlikely that enhanced CO2 fixation by PEP carboxylase at the CO2 compensation concentration is a major mechanism for reducing photorespiration in the intermediate Flaveria species. Moreover, these findings support previous related 14CO2-labeling studies at air-levels of CO2 which indicated that F. floridana and F. ramosissima were more C4-like intermediate species. This is further substantiated by the demonstration that F. floridana PEP carboxylase, like the enzyme in C4 plants, undergoes a substantial activation (2.2-fold) upon illuminating dark-adapted green leaves. Regional food web studies that fail to account for small-scale isotopic variability can lead to a mismatch between an organism’s inferred and true trophic position. Misinterpretation of trophic status may result, substantially limiting spatial and temporal comparability of food web studies. We sampled several carbon sources and consumers in a nested design to assess the variability of food web members across small spatial scales (100 s of m to several km) in regions around the Windmill Islands and Vestfold Hills in East Antarctica. For carbon sources, δ13C in sea ice POM was particularly variable between locations (km apart) and between sites (100 s of m apart) with replicate samples varying by up to 16‰. Macroalgae δ13C was less variable (replicate samples ranging up to 6.9‰ for the red alga Iridaea cordata), yet still differed between locations. Sediment POM and pelagic POM were the least variable, displaying minimal differences between locations or sites for δ13C and δ15N. Three out of eight consumers were significantly different between locations for δ13C, and five out of eight for δ15N, with the fish Trematomus bernacchii the most variable for both δ13C and δ15N. At smaller scales, the amphipod Paramorea walkeri showed significant variation between sites in δ13C but not in δ15N. We attribute small-scale variability to the dynamic physical environment for carbon sources in coastal systems and a close coupling of diet to habitat for consumers. We highlight the need to account for small-scale spatial variation in sampling designs for regional food web studies.  相似文献

The main aim of this study was to test various hypotheses regarding the changes in 13C of emitted CO2 that follow the addition of C4-sucrose to the soil of a C3-ecosystem. It forms part of an experimental series designed to assess whether or not the contributions from C3-respiration (root and microbial) and C4-respiration (microbial) to total soil respiration can be calculated from such changes. A series of five experiments, three on sieved (root-free) mor-layer material, and two in the field with intact mor-layer (and consequently with active roots), were performed. Both in the experiments on sieved mor-layer and the field experiments, we found a C4-sucrose-induced increase in C3-respiration that accounted for between 30% and 40% of the respiration increase 1 h after sucrose addition. When the course of C3-, C4- and total respiration was followed in sieved material over four days following addition of C4-sucrose, the initially increased respiration of C3-C was transient, passing within less than 24 h. In a separate pot experiment, neither ectomycorrhizal Pinus sylvestrisL. roots nor non-mycorrhizal roots of this species showed respiratory changes in response to exogenous sucrose. No shift in the 13C of the evolved CO2 after adding C3-sucrose to sieved mor-layer material was found, confirming that the sucrose-induced increase in respiration of endogenous C was not an artefact of discrimination against 13C during respiration. Furthermore, we conclude that the C4-sucrose induced transient increase in C3-respiration is most likely the result of accelerated turnover of C in the microbial biomass. Thus, neither respiration of mycorrhizal roots, nor processes discriminating against 13C were likely sources of error in the field. The estimated 13C of evolved soil CO2 in three field experiments lay between –25.2 and –23.6. The study shows that we can distinguish between CO2 evolved from microbial mineralisation of added C4-sucrose, and CO2 evolved from endogenous carbon sources (roots and microbial respiration).  相似文献

The stable carbon and oxygen isotope ratios in cellulose of C3 and C4 plants growing on the surface of a montane peat bog in the Nilgiri hills, southern India, were measured. The mean monthly δ13C values in cellulose of both C3 and C4 plants are found to be significantly related to rainfall, while the δ18O values are sensitive to changes in maximum temperature and relative humidity of the region. Further, higher δ18O values were observed in C4 plants compared to C3 plants, suggesting that C4 plants are probably less sensitive to relative humidity as compared to C3 plants and are able to photosynthesize even during drier conditions. The plant isotope-climate correlations thus established can be used for reconstructing the past temperature and rainfall conditions of the tropics from the isotopic ratios of peat deposits, derived from a mixture of C3 and C4 plants in the region.  相似文献

CO2 compenstation point (), the concentration of CO2 at which photosynthesis and respiration are at equilibrium, is a commonly used diagnostic for the C4 photosynthetic pathway, since it reflects the reduced photorespiration that is a property of C4 photosynthesis. Geographic variation for was examined within Flaveria linearis, a C3–C4 intermediate species. Collections from four widely separated Floridian populations were propagated in a greenhouse and measured for . Little differentiation among populations was found, but significant within-population variation was present. Temperature is a hypothesized selective agent for the C4 photosynthetic pathway. To test this hypothesis, plants exhibiting a range of were cloned and placed in growth chambers at 25°C and 40°C. After 7 weeks, valves were remeasured and plants were harvested and weighed. There was a poor correlation between initial and final measures of for a given genotype (r=0.38, P>0.1). Broad sense heritability for was computed to be 0.10. At 25°C, there was no relationship between final size and . At 40°C, more C4-like plants, as indicated by their low , had grown larger. Differences in relative growth rate were attributable more to differences in net assimilation rate than in leaf area ratio. Taken together, these results demonstrate that although significant plasticity exists in the amount of photorespiration in this C3–C4 species, high temperature appears to be an effective selective agent for the reduction of photorespiration and the enhancement of C4-like traits.  相似文献

We tested the reliability of herbivore faecal δ13C and δ15N values for reconstructing diet through review of an extensive database derived from a 3-year study of ungulates in South Africa's Kruger National Park. Faeces are a useful material for stable isotope studies of diet because they record dietary turnover at very short time scales, and because sampling is non-invasive. However, the validity of faecal isotope proxies may be questioned because they represent only undigested food remains. Results from Kruger Park confirm that free-ranging browsers have faecal δ13C consistent with C3 feeding, grazer faeces are C4, and mixed-feeder faeces intermediate. Although the respective ranges do not overlap, there is significant variation in faecal δ13C of browsers and grazers (~2.0–4.0‰) across space and through time. We demonstrate that most (~70%) of this variation can be ascribed to corresponding patterns of variation in the δ13C of C3 and C4 plants, respectively, re-enforcing the fidelity of faecal isotope proxies for diet but highlighting a need for mixing models that control for variations in plant δ13C in order to achieve accurate diet reconstructions. Predictions for the effects of climate (rainfall) and ecophysiology on 15N-abundance variations in mammals do not persist in faeces. Rather, faecal δ15N tracks changes in plant δ15N, with further fractionation occurring primarily due to variations in dietary protein (reflected by %N). Controlling for these effects, we show that a dual-isotope multiple source mixing model (Isosource) can extend diet reconstructions for African savanna herbivores beyond simplified C3/C4 distinctions, although further understanding of variations in mammal δ15N are needed for greater confidence in this approach.  相似文献

Stable isotope analysis is frequently used to infer resource use in natural populations of fishes. Studies have examined factors, other than diet, that influence δ15N and δ13C including tissue-specific rates of equilibration and starvation. Most such studies completed under laboratory conditions tightly control food quantity and its isotopic composition, but it is also necessary to evaluate the influence of these factors under more natural conditions. Using pumpkinseed sunfish (Lepomis gibbosus) we evaluated whether restricted rations below minimum daily requirements affects tissue equilibration to a change in diet by holding fish on two treatments that often reflect divergent resource use in natural populations (pelagic zooplankton or littoral macroinvertebrates). Over 42 days, δ15N values increased while δ13C values did not change, additionally neither were related to diet treatment. Increased δ15N values were negatively related to body condition while δ13C values were not, indicating that stable isotope values were more affected by decreasing body condition than by diet. Additionally, δ15N values changed more in the blood and liver tissues than in white muscle tissue, indicating that restricting food availability had greater effects on tissues with greater metabolic activity. We hypothesize that stable isotope values of consumers are subject to a tissue-specific trade-off between sensitivity to changes in resource use and resistance to the effects of low resource availability. This trade-off may require consideration in stable isotope studies of wild populations facing periodic limitations of food availability.  相似文献

Recent studies suggest that the direction and magnitude of changes in soil organic carbon (soil C) pools following forest-to-pasture conversion in the tropics are dependent upon initial soil conditions and local factors (e.g. pre-conversion soil C content, soil texture, vegetation productivity, and management practices). The goal of this study was to understand how landscape-scale variation in soil-forming factors influenced the response of soil C pools to forest clearing and pasture establishment in northeastern Costa Rica. We measured soil C and its stable isotopic composition in 24 paired pasture and reference forest sites distributed over large gradients of edaphic characteristics and slope throughout a 1400 km2 region. We used the large difference in stable C isotopic signatures of C3 vegetation (rain forest) versus C4 vegetation (pasture grasses) as a tracer of soil C dynamics. Soil C pools to 30 cm depth ranged from 26% lower to 23% higher in pastures compared to paired forests. The presence of non-crystalline clays and percent slope explained between 27 and 37% of the variation in the direction and magnitude of the changes in soil C storage following pasture establishment. Stable carbon isotopes (13C) in the top soil (0–10 cm) showed a rapid incorporation of pasture-derived C following pasture establishment, but the vegetation in these pastures never became pure C4 communities. The amount of forest-derived soil C in pasture topsoils (0–10 cm) was negatively correlated to both pasture age and the concentrations of non-crystalline iron oxides. Together these results imply that site factors such as soil mineralogy are an important control over soil C storage and turnover in this region.  相似文献

Much of the primatology literature on stable isotope ratios of carbon (δ13C) and nitrogen (δ15N) has focused on African and New World species, with comparatively little research published on Asian primates. Here we present hair δ13C and δ15N isotope values for a sample of 33 long-tailed macaques from Singapore. We evaluate the suggestion by a previous researcher that forest degradation and biodiversity loss in Singapore have led to a decline in macaque trophic level. The results of our analysis indicated significant spatial variability in δ13C but not δ15N. The range of variation in δ13C was consistent with a diet based on C3 resources, with one group exhibiting low values consistent with a closed canopy environment. Relative to other macaque species from Europe and Asia, the macaques from Singapore exhibited a low mean δ13C value but mid-range mean δ15N value. Previous research suggesting a decline in macaque trophic level is not supported by the results of our study.  相似文献

Wetlands are significant sources of the important greenhouse gas CH4. Here we explore the use of an experimental system developed for the determination of continuous fluxes of CO2 and CH4 in closed ecosystem monoliths including the capture of 14CO2 and 14CH4 following pulse labelling with 14CO2. We show that, in the ecosystem studied, ebullition (bubble emission) may account for 18 to 50% of the total CH4 emission, representing fluxes that have been difficult to estimate accurately in the past. Furthermore, using plant removal and 14C labelling techniques, we use the system to detail the direct influence of vascular plants on CH4 emission. This influence is observed to be dependent on the amount of vascular plants present. The results that may be produced using the presented experimental set-up have implications for an improved understanding of wetland ecosystem/atmosphere interactions, including possible feedback effects on climate change. In recent years much attention has been devoted to ascertaining and subsequently using the relationship between net ecosystem productivity and CH4 emission as a basis for extrapolation of fluxes across large areas. The experimental system presented may be used to study the complex relationship between vascular plants and CH4 emission and here we show examples of how this may vary considerably in nature between and even within ecosystems.  相似文献

The effect of NH4+/NO3 availability on nitrate reductase (NR) activity in Phragmites australis and Glyceria maxima was studied in sand and water cultures with the goal to characterise the relationship between NR activity and NO3 availability in the rhizosphere and to describe the extent to which NH4+ suppresses the utilization of NO3 in wetland plants.The NR activity data showed that both wetland helophytes are able to utilize NO3. This finding was further supported by sufficient growth observed under the strict NO3 nutrition. The distribution of NR activity within plant tissues differed between species. Phragmites was proved to be preferential leaf NO3 reducer with high NR activity in leaves (NRmax > 6.5 μmol NO2 g dry wt−1 h−1) under all N treatments, and therefore Phragmites seems to be good indicator of NO3 availability in flooded sediment. In the case of Glyceria the contribution of roots to plant NO3 reduction was higher, especially in sand culture. Glyceria also tended to accumulate NO3 in non-reduced form, showing generally lower leaf NR activity levels. Thus, the NR activity does not necessarily correspond with plant ability to take up NO3 and grow under NO3-N source. Moreover, the species differed significantly in the content of compounds interfering with NR activity estimation. Glyceria, but not Phragmites, contained cyanogenic glycosides releasing cyanide, the potent NR inhibitor. It clearly shows that the use of NR activity as a marker of NO3 utilization in individual plant species is impossible without the precise method optimisation.  相似文献

Background

The carbon (C) isotope signature of solid materials such as plants and soil, or gaseous samples (atmospheric or soil air), can be used as a useful tool for investigations of the C cycle. In gaseous samples, stability of δ13C of carbon dioxide (CO2) in air during storage represents a problem.

Methods

We tested the long-term storage effect of δ13C originated from CO2 in vials both capped with butyl or butyl coated with polytetrafluoroethylene (PTFE) on the internal surface. Therefore, pure CO2, depleted in 13C, was stored for up to 736 days. In addition, the relevance of long-term storage for ecological soil air data collected from a free-air CO2 enrichment (FACE) experiment located in Stuttgart (Germany) during one growing period with a maximum storage period of 210 days was judged.

Results

With increasing storage time, a change in isotopic composition towards less depleted δ13C was observed. The changes in δ13C were highest at the beginning of the storage period and decreased over time, which could be described with an asymptotic model. The maximum change in δ13C was less than 2?‰ and lower for vials capped with butyl/PTFE septa. In the FACE experiment, the comparison between corrected and uncorrected data showed that δ13C originated from soil air changed within this data set by up to 1?‰. The calculation of the fraction of plant derived C resulted in an underestimation of up to 10 %.

Conclusion

The storage effect should be taken into consideration when interpreting δ13C values in order to avoid miscalculations.  相似文献

Sea lion and seal populations in Alaskan waters underwent various degrees of decline during the latter half of the twentieth century and the cause(s) for the declines remain uncertain. The stable carbon (13C/12C) and nitrogen (15N/14N) isotope ratios in bone collagen from wild Steller sea lions (Eumetopias jubatus), northern fur seals (Callorhinus ursinus) and harbor seals (Phoca vitulina) from the Bering Sea and Gulf of Alaska were measured for the period 1951-1997 to test the hypothesis that a change in trophic level may have occurred during this interval and contributed to the population declines. A significant change in '15N in pinniped tissues over time would imply a marked change in trophic level. No significant change in bone collagen '15N was found for any of the three species during the past 47 years in either the Bering Sea or the Gulf of Alaska. However, the 15N in the Steller sea lion collagen was significantly higher than both northern fur seals and harbor seals. A significant decline in '13C (almost 2 ‰ over the 47 years) was evident in Steller sea lions, while a declining trend, though not significant, was evident in harbor seals and northern fur seals. Changes in foraging location, in combination with a trophic shift, may offer one possible explanation. Nevertheless, a decrease in '13C over time with no accompanying change in '15N suggests an environmental change affecting the base of the foodweb rather than a trophic level change due to prey switching. A decline in the seasonal primary production in the region, possibly resulting from decreased phytoplankton growth rates, would exhibit itself as a decline in '13C. Declining production could be an indication of a reduced carrying capacity in the North Pacific Ocean. Sufficient quantities of optimal prey species may have fallen below threshold sustaining densities for these pinnipeds, particularly for yearlings and subadults who have not yet developed adequate foraging skills.  相似文献

For 383 Poaceae species harvested over the Japanese islands and stored as herbarium specimens along several decades, we determined C3 and C4 types of photosynthesis from leaf stable carbon isotope ratio (δ13C). Then, we sought the relationships between C4 species richness and climatic factors or habitat types. Except for the two Panicum species (P. lanuginosum and P. scoparium) having the possibility of C3–C4 intermediate, 227 and 154 species were classified into C3 and C4. The C4 species richness increased from northern to southern islands in Japan, positively correlated with mean annual air temperature. Greater C4 species richness in the seashore habitats, and smaller C4 species richness in the shaded, wet and highland habitats would be related to the photosynthetic responses to local environmental factors such as irradiance level and temperature regime. No difference of leaf δ-value of C3 Poaceae was obtained between the habitats with different soil water availability, suggesting the less importance of soil water availability on leaf water-use efficiency in C3 Poaceae species in Japan having humid climate. Additionally, possible effects of human activity around the harvested time or site on leaf δ-value were estimated, because the habitat includes the sites with high human activity. Leaf δ-value was decreased with sampling year, and it was higher in the densely inhabited district for both C3 and C4. They are probably due to a historical decrease in the atmospheric δ-value via increasing human activity, and high gas emission at the districts of high human density.  相似文献

We studied Holocene speleothems and tufa samples collected in numerous caves and rivers in the Dinaric Karst of Croatia, Slovenia, Bosnia and Herzegovina, as well as Serbia and Montenegro. Differences in the formation process of tufa and speleothems are discussed in the context of their isotopic composition (14C, 13C and 18O), as well as the chemistry of surface water (rivers, lakes) and drip water (in caves). The physical and chemical parameters monitored in the surface water (tufa precipitation) and drip water (speleothem precipitation) show that more stable conditions accompany speleothem rather than tufa formation. This is particularly obvious in the water temperature variations (2-22°C in surface water and 7-12°C in drip water) and in saturation index variation (3-11 in surface water and 1-6 in drip water). The range of 14C ages recorded by Holocene speleothems (∼12?000 yr) is wider by several thousands years than that of Holocene tufa samples (∼6000 yr). δ13C values for tufa samples range from −12‰ to −6‰ and for speleothem samples from −12‰ to +3‰ reflecting higher soil carbon and/or vegetation impact on the process of tufa than on speleothem formation. The differences in δ18O values of tufa and speleothem samples from different areas reflect different temperature conditions and differing isotopic composition in the water. The study shows that speleothems from the Dinaric Karst can be used as global palaeoclimatic records, whereas tufa records changes in the local palaeoenvironment.  相似文献
