Papillary muscles of rat and dog hearts were fixed in such a way as to prevent excessive shortening during the procedure. The material was embedded in either araldite or methacrylate and was stained in various ways. The filamentous fine structure of mammalian cardiac muscle is similar to that previously described for striated skeletal muscle. The sarcomeres are composed of a set of thick and thin filaments which interdigitate in the A band proper. The filament ratios and the filamentous array are in accord with those found in skeletal muscle. The functional significance of this twofold array of filaments is not entirely clear. Various other structural aspects of cardiac cells such as surface membranes, mitochondria, nuclei, and cytoplasmic granules are described. The sarcoplasmic reticulum is discussed in detail as are the various structural components forming the intercalated discs. Fairly frequent deep invaginations of the sarcolemma with basement membrane are observed in addition to the intercalated discs. These surface membrane invaginations probably explain the branching appearance of cardiac fibers as seen under the light microscope.  相似文献

The fine structure of desmosomes and intercalated discs in the toad heart is discussed. A definite relationship between the dense components of these structures and the dense region of the Z band is demonstrated. The dense region of the Z band characteristically widens at its approach to the plasma membrane, and often terminates beneath it in a distinct discoidal plaque. Cardiac desmosomes appear to be structures which result from the intimate apposition of plaques of Z band material. These desmosomes retain the Z band function as sites of attachment for myofilaments. The suggestion is made that rotation of a desmosome through 90° and splitting of filaments from the adjacent sarcomere could result in the formation of a simple step-like intercalated disc. Intermediate stages in this process are illustrated. Complex discs present in the toad probably represent the alignment of groups of simple discs produced by contractile forces. Possible physiologic functions of the disc and desmosome are discussed. Other morphologic features of toad cardiac cells include a distinct amorphous outer coat to the sarcolemma, a prominent N band, and a granular sarcoplasm with poorly developed reticulum.  相似文献

With a view to indicating that the α excitatory state in muscle is not of a special nature it is shown that the α strength-duration curves are of the same form as those determined for nerve and other tissues except that in about two-thirds of all cases the rheobase appears to be slightly too low. Also from experiments in latent addition it is found that the α excitatory state following an inadequate stimulus subsides exponentially at a rate which is related to the α excitability in the same way, approximately, as the subsidence rate in nerve is related to the nerve''s excitability. In both tissues the subsidence as measured directly is 2–3 times as fast as it appears to be from the strength-duration curve. The α refractory period is at least as short as that of nerve so the α chronaxie is unusually long compared to the refractory period. There is no reason at present, however, to consider this as having any bearing on the problem at issue. It is concluded therefore, that the α excitability differs from others in the rates of its reactions rather than in its fundamental nature and that any conclusions about excitability drawn from its study will probably be valid quite generally.  相似文献

Electron microscopic evidence is presented that the early response to denervation ("simple atrophy") of the semitendinosus m. of the frog is characterized by a greater prominence of the sarcoplasmic reticulum and by the presence, in the interfibrillar spaces, of mitochondria which are more numerous and smaller than in normal muscle. In contrast with the dynamic changes of the sarcoplasmic structural components, the myofibrils showed a progressive decrease in diameter after denervation and throughout the period studied. By carrying out tissue fractionation experiments, the yield of microsome-protein was found significantly greater in the denervated muscles, as compared with the contralateral controls, in this initial stage. Under the conditions attending the overdevelopment of the sarcoplasmic reticulum (SR), denervated semitendinosus m. incorporated valine-C14 into proteins more actively than the control pairs. The denervated muscles also showed an increase in the number of freely scattered and membrane-bound ribosomes and of polyribosomes, suggesting a more active synthesis of the SR membranes. Pronounced atrophy of the myofibrils, disorganization of the SR, and an increased number of ribonucleoprotein particles lying in the enlarged interfibrillar spaces were the main ultrastructural features of "degenerative atrophy" in frog muscle in the late periods after denervation. The probably adaptive character of the early changes occurring on denervation of frog muscle is discussed.  相似文献

In cross-sections of single fibers from the frog semitendinosus muscle the number of thick myofilaments per unit area (packing density) is a direct function of the sarcomere length. Our data, derived from electron microscopic studies, fit well with other data derived from in vivo, low-angle X-ray diffraction studies of whole semitendinosus muscles. The data are consistent with the assumption that the sarcomere of a fibril maintains a constant volume during changes in sarcomere length. The myofilament lattice, therefore, expands as the sarcomere shortens. Since the distance between adjacent myofilaments is an inverse square root function of sarcomere length, the interaction of the thick and the thin myofilaments during sarcomere shortening may occur over distances which increase 70 A or more. The "expanding-sarcomere, sliding-filament" model of sarcomere shortening is discussed in terms of the current concepts of muscle architecture and contraction.  相似文献

The length-tension relationship was determined for strips of guinea pig taenia coli and correlated with the length and ultrastructural organization of the component fibers. The mean fiber length in "stretched" strips (passive ≥ active tension) was 30% greater than that for fibers in "unstretched" strips (active >> passive tension). In stretched fibers the dense bodies and 100 A diameter myofilaments were consolidated into a mass near the center of fibers in cross-sectional profile. The thick myofilaments were segregated into the periphery of the fiber profiles. In unstretched fibers the dense bodies-100 A diameter filaments and the thick myofilaments were uniformly distributed throughout cross-sectional profiles. A tentative model is proposed to account for the change in fiber length and ultrastructural organization that accompanies stretch. The basic features of the model require the dense bodies to be linked together into a network by the 100 A diameter filaments. The functional consequences of stretching the fibers are discussed in relation to the model proposed for this network.  相似文献

1. A study was made of the electrolyte changes which occur when frog muscles are immersed in a Ringer solution with 1/5 of the Na replaced by NH4Cl. Analyses were made in the solution and in the muscles for K and NH3, and the muscles were also analyzed for Cl, HCO3, and Na. Control muscles were immersed in normal Ringer''s solution and similarly analyzed. 2. The amount of ammonia taken up was about equal to the K and Na lost. There was also a small increase in chloride content. The bicarbonate content was the same in both experimental and control muscles, indicating no change in the muscle pH due to the NH3 which penetrated. An increased loss of K due to the penetration of NH3 was also demonstrated by the use of radioactive K. 3. After 5 hours, the concentration of ammonia per gram of muscle is about the same as the concentration in the solution. After 4 or 5 days, the concentration in the muscle is about 1.5 times that in the solution. The inside to outside NH3 ratio is about equal to the corresponding H ion ratio, but is much less than the K ratio. 4. The rate of penetration of the NH3 is increased by a rise of temperature, by stirring the solution, and by decrease in the concentration of Na, K, Ca, or Mg in the solution; it is decreased by increasing the size of the muscles or by killing them with chloroform or boiling. 5. Liver, smooth muscle, skin, and kidney, in a few experiments, behaved much like muscle except that there was a formation of urea in the case of liver. 6. The injection of NH4Cl into anesthetized cats causes an increase in the level of K in the blood plasma.  相似文献

The organisation of the myofibrils and the sarcoplasmic reticulum in frog slow muscle fibres has been compared with that in twitch fibres. It has been found that the filaments have the same length in the two types of fibre, but that there are differences in their packing: (a) in contrast to the regular arrangement of the I filaments near the Z line in twitch fibres, those in slow fibres are irregularly packed right up to their insertion into the Z line; (b) the Z line itself shows no ordered structure in slow fibres; (c) the fine cross-links seen between the A filaments at the M line level in twitch fibres are not present in slow fibres. The sarcoplasmic reticulum in slow fibres consists of two separate networks of tubules. One set of tubules (diameter about 500 to 800 A) is oriented mainly in a longitudinal direction. The tubules of the other network (diameter about 300 A) are oriented either transversely at approximately Z line level or longitudinally, connecting the transverse tubules. Triads are very rarely found, occurring at only every 5th or 6th Z line of each fibril. The central element of these triads is continuous with the thin tubules. Slow fibres from muscles soaked in ferritin-containing solutions contain ferritin particles in the network of thin tubules, the rest of the sarcoplasm remaining free of ferritin.  相似文献

The potassium contraction of skeletal muscle and relaxation of cardiac muscle have been correlated with the carbon dioxide and total acid production of these tissues. 1. The immersion of surviving sartorius muscles of the frog in isotonic potassium chloride solution causes a marked increase in the rate of acid production. 2. It is probable that carbon dioxide is the principal acid involved in the above effect. 3. The immersion of surviving cardiac muscle of the frog in isotonic potassium chloride solution causes a pronounced depression in the rate of survival acid production. 4. Reasons are given for believing that these changes in metabolism may be independent of the stimulation and inhibition of contraction which potassium simultaneously produces in these tissues.  相似文献

本研究报道了裸露心肌标本的制备及研究肌钙蛋白Ca~(2 )敏感性的技术方法。10mmol/L EGTA裸露液浸浴大鼠右室乳头肌,其肌膜通透性明显增高,表现为去除舒张液中ATP和磷酸肌酸后,心肌立即挛缩。最适裸露时间为24h。此时最大Ca~(2 )激活张力(T_(max)最高;收缩和舒张至50%T_(max)所需时间T_(1/2)和R_(1/2)最短。张力-pCa(Ca~(2 )摩尔浓度的负对数)关系曲线是S形,且同一标本先后两次实验的张力-pCa关系及50%T_(max)时的pCa(pCa_(50))均无明显差别,而茶碱可使张力-pCa关系左移,pCa_(50)增高。表明高通透心肌标本功能稳定,且可反映肌钙蛋白Ca~(2 )敏感性的改变。  相似文献

The walls of the gastrointestinal tract and urinary bladder of rats were fixed in osmium tetroxide, embedded in methacrylate, and sectioned for electron microscopy. The examination of sections of smooth muscle tissue with the electron microscope reveals the presence of bundles of unmyelinated nerve fibers within the intercellular spaces. In addition, vesiculated nerve processes, bounded on their outer surfaces by delicate plasma membranes and typically containing varying quantities of synaptic vesicles and mitochondria, make intimate contact with the surface of smooth muscle cells. These nerve processes are similar in structure and disposition to nerve endings previously described in skeletal muscle, in the central nervous system, in peripheral ganglia, in receptors, and in glands. It is concluded that the relationships existing between vesiculated nerve processes and the surface of smooth muscle cells constitute neuromuscular junctions. Profiles of protrusions of smooth muscle cells are often seen protruding into the intercellular spaces. Here they occur singly or in groups, originating from one or more cells. Because of the plane of section the protrusions may sometimes appear as individual entities between the muscle cells. In such cases care must be exercised in their identification because they have characteristics similar to sectioned nerve processes which also occur in the intercellular spaces.  相似文献

以大肠癌裸鼠移植瘤为动物模型,观察了分叉双歧杆菌的完整肽聚糖体内对大肠癌生长的抑制作用,并从细胞增殖活性方面探讨了它的抑瘤机制。结果表明完整肽聚糖注射组大肠癌移植瘤的生长速度、平均瘤重以及增殖细胞核抗原(PCNA) 阳性细胞密度均明显低于肿瘤对照组(P< 0.01) 。提示分叉双歧杆菌的完整肽聚糖体内能明显抑制大肠癌的生长,其抑瘤机制之一是降低肿瘤细胞的增殖活性。  相似文献
