Withdrawal of a utilizable nitrogen source during mid G1 of the cell cycle induces gametic differentiation in synchronously grown vegetative cultures of Chlamydomonas reinhardi. Cell division accompanies gametic differentiation in such cultures, and the ability of mid G1 vegetative cells to form gametes is matched by their ability to undergo a round of cell division after nitrogen withdrawal. Synchronously grown cultures require up to 19 hr in nitrogen-free medium to complete a round of division and to form mating-competent cells. Asynchronously grown liquid cultures require less time after nitrogen withdrawal (generally 5–8 hr) to achieve mating competency. In these cultures cell division did not necessarily accompany gametic differentiation since gametic differentiation took place in induced cultures at high cell concentrations which prevented cell division. Maximum mating competency was achieved in less than 2 hr after induction of vegetative cells grown on agar plates. Little cell division was observed during that short induction interval. The relationship between the attainment of mating competency (gametogenesis) and other physiological events resulting from nitrogen withdrawal is discussed.  相似文献

Routinely prepared gametes (by flooding 3 week-old agar cultures) showed about 80% mating competence if the opposite sexual partners were mixed together. The mating competence exhibited a strict dependence on the composition of the solution in which the cells were suspended before mixing; it decreased progressively with increasing concentration of nitrates. In contrast, no inhibiting effect was found if urea was used as the source of nitrogen. Other ions present in nutrient media did not show any effect. Mating activity varied according to the spectral composition of light, being higher with a blue light than with a red one. Blue light caused accumulation ofvis-à-vis pairs, which were blocked to form zygotes. Freshly released daughter cells in vegetatively grown synchronous cultures had a dual nature—vegetative and sexual one. In these daughter cells, similar rules were found for governing of mating competence to those valid for standard gametes obtained from flooded agar cultures. High mating competence was found in daughter cells released the during dark period in distilled water, nitrate-free media, in the presence of Mg2+ or Ca2+ ions, or in media containing urea. The conditions during which daughter cells are released and the conditions under which they mate can be considered crucial for expression of gametic nature as a mating competence.  相似文献

Nitrogen-replete cells of Phaeodactylum tricomutum Bohlin assimilated ammonium and the amino acid l-arginine simultaneously. Arginine was taken up at rates expected to supply at least 30% of the cells' requirement for nitrogen; arginme-carbon mainly entered protein but, when uptake was in darkness, ≈40% was respired. Cells grown in a 12:12 h light:dark cycle with ammonium as the sole nitrogen source took up ammonium throughout the growth cycle, whereas cells grown with the addition of arginine took up little ammonium during the dark phase. The uptake of ammonium over the course of the cycle was reduced by 30% when arginine was present. Cells grown with arginine as the sole nitrogen source took up the amino acid at the rate required for growth. In contrast, cells grown on ammonium, while growing at the same rate as those on arginine, assimilated nitrogen at twice the rate. Cells grown with both sources of nitrogen present, took up arginine at the same rate as before, but more of the arginine-carbon was respired (60% as compared with 40% when ammonium was absent). The uptake of ammonium was reduced by 30%, but the total nitrogen assimilation again exceeded immediate requirements. A high uptake rate of arginine was indicative of cells assimilating ammonium only; a low uptake rate of ammonium during the dark phase of growth was indicative of cells assimilating arginine. It is not known whether the findings with P. tricomutum are applicable to other marine phytoplankton. If they are, arginine may be of greater significance as a natural source of nitrogen for phytoplankton than is generally thought.  相似文献

During sexual differentiation, Chlamydomonas reinhardtii changes its chemotactic behavior in response to ammonium. Just like gamete formation, the change in chemotaxis mode is controlled by the sequential action of two environmental cues, removal of ammonium or nitrate from the medium and light. Thus, vegetative cells and mating incompetent pre-gametes, the latter being generated by nitrogen starvation in the dark, exhibit chemotaxis towards ammonium. Irradiation of pre-gametes results in a loss of chemotaxis and the gaining of mating competence. Incubation of these gametes in the dark resulted in their regaining chemotactic activity; re-illumination again resulted in its loss. Blue light was shown to be most effective in switching-off chemotaxis. RNA-interference strains with reduced levels of the blue-light receptor phototropin showed an attenuated inactivation of chemotaxis that could be partially compensated by the application of higher fluence rates, suggesting that these light responses are mediated by phototropin. Chlamydomonas lytic enzyme of the cell wall (gamete wall-autolysin) is responsible for shedding of cell walls during mating of opposite mating-type gametes. This paper reports some topographic aspects of lytic enzyme in cells. Both vegetative and gametic cells contain the same wall lytic enzyme. The purified enzyme is a glycoprotein with an apparent molecular mass of 67 kD by gel filtration and 62 kD by SDS PAGE, and is sensitive to metal ion chelators and SH-blocking agents. These properties are the same as those of the gamete wall-autolysin released into the medium by mating gametes. However, the storage form of the enzyme proves to be quite different between the two cell types. In vegetative cells, the lytic enzyme is found in an insoluble form in cell homogenates and activity is released into the soluble fraction only by sonicating the homogenates or freeze-thawing the cells, whereas gametes always yield lytic activity in the soluble fractions of cell homogenates. When vegetative cells are starved for nitrogen, the storage form of enzyme shifts from its vegetative state to gametic state in parallel with the acquisition of mating ability. Adding nitrogen to gametes converts it to the vegetative state concurrently with the loss of mating ability. We also show that protoplasts obtained by treatment of vegetative cells or gametes with exogenously added enzyme have little activity of enzyme in the cell homogenates, suggesting that lytic enzyme is stored outside the plasmalemma. When the de-walled gametes or gametes of the wall-deficient mutant, cw-15, of opposite mating types are mixed together, they mate normally but the release of lytic enzyme into the medium is practically negligible. When the de-walled vegetative cells are incubated, the lytic enzyme is again accumulated in the cells after the wall regeneration is almost complete.  相似文献

Studies on the mean cellular carbohydrate contents of Platymonas striata Butcher under conditions of nitrogen-starvation, and after refeeding these starved cultures with either nitrate or ammonium ions (growing under continuous illumination or with an alternating light/dark regime) have shown that nitrogen-starved cells accumulated abnormal amounts of cellular carbohydrate and that nitrogen refeeding produced a marked drop in the cellular carbohydrate. Cells grown in a light/dark regime accumulated less carbohydrates than those grown in continuous light. The mean cellular carbohydrate levels 16 h after nitrogen refeeding were still much in excess of those of cells grown with normal nutrition. It was therefore suggested that the differences in nitrogen uptakes in this period — when comparing either the uptake of cells grown in continuous light with that of cells grown in a light/dark regime; or when comparing the uptakes of cells presented with either nitrate or ammonium ions and grown in a light/dark regime —cannot be directly due to shortages of carbohydrate for the provision of carbon skeletons for nitrogen assimilation.  相似文献

Laboratory strains of Chlamydomonas reinhardtii, which are descendantsof a 1945 isolate by G.M. Smith (Harris 1989), were dividedinto two groups depending upon whether the vegetative cellsrequire light to differentiate into gametes under ammonium ion-starvedconditions. Light-dependent (LD) strains were unable to becomegametes in the dark, while light-independent (LI) strains coulddo so. All the wild-type strains isolated recently from thefield showed light-dependency, suggesting that the LD-phenotypeis the wild-type. The LD-cells failed to acquire flagellar agglutinability,to accumulate cell body agglutinins, or to form mating structuresin the dark, but did so rapidly after transfer to light. Moreover,the light-induced LD-gametes, but not the Li-gametes, lost theirmating ability, cell body agglutinins, and mating structuresafter transfer to darkness, indicating that the LD-cells requirelight not only for gametic differentiation but also for maintenanceof gametic activity. Cells of Spirogyra filaments grown in darkness become longer than light grown cells. This elongation can be prevented by a few minutes of red light given with intervals of 24 h under the dark period. Far-red light given after the red light pulse counteracts the red light effect. Cells which have finished their elongation in light do not elongate further in darkness. Along with the cell elongation the chloroplasts become less spirally wound and ultimately shortened to straight bands.  相似文献

Biddulphia aurita, a centric diatom, can grow on either nitrate, nitrite, or ammonia as its sole nitrogen, source. Cells remove ammonium nitrogen from the medium 2.3–2.4 times faster than either nitrate or nitrite nitrogen and, when grown for 24 hr in the ammonium medium, contain higher levels of non-protein nitrogen than cells grown in the nitrate or nitrite medium for the same period of time. The nitrogenous compounds in the nonprotein nitrogen fraction from cells grown in the nitrate, nitrite, or ammonium medium contain the same level of soluble-free amino nitrogen, combined amino nitrogen, and ammonium nitrogen. The high level of soluble nonprotein nitrogen in the medium of the cells grown in the ammonium medium is due to soluble amide nitrogen which represents 18% of the total soluble nitrogen present in these cells, whereas it represents only 2% in cells from the nitrite medium, and its level is negligible in cells from the nitrate medium. Cells grown in the nitrate medium have both nitrate- and nitrite-reductase activity. Cells grown in the nitrite medium have only nitrite-reductase activity in significant levels, while cells grown in the ammonium medium lack both enzymes.  相似文献

Azospirillum lipoferum RG6xx was grown under conditions similar to those resulting in encystment of Azotobacter spp. A. lipoferum produced cells of uniform shape when grown on nitrogen-free β-hydroxybutyrate agar. Cells accumulated poly-β-hydroxybutyrate and often grew as chains or filaments that eventually lost motility and formed capsules. Within 1 week, vegetative A. lipoferum inocula were converted into microflocs arising from filaments or chains. Cells within microflocs were pleomorphic, contained much poly-β-hydroxybutyrate, and were encapsulated. Some cells had a cystlike morphology. Up to 57% of the dry weight of encapsulated flocs was poly-β-hydroxybutyrate, whereas vegetative cells grown in broth with combined nitrogen had only 3% of their dry weight as poly-β-hydroxybutyrate. Neither encapsulated cells in flocs nor nonencapsulated vegetative cells were significantly desiccation resistant. Under starvation conditions (9 days) only 25% of encapsulated cells remained viable, whereas vegetative cells multiplied severalfold. In short-term germination experiments with encapsulated flocs, nitrate, ammonium, and soil extract promoted formation of motile vegetative cells. Most cells in treatments lacking combined nitrogen eventually depleted their visible poly-β-hydroxybutyrate reserves without germinating. The remaining cells retained the reserve polymer and underwent size reduction.  相似文献

A newly isolated denitrifying strain, Rhodobacter sphaeroides NII2 was immobilized in polyvinyl alcohol (PVA) gel, and the properties of the cells in the gel were examined. The immobilized cells had low or almost no denitrification activity, but the cells were activated by incubation in light with culture medium for denitrification containing 0.5% nitrate and no other nitrogen source. Cells grown in the dark were activated by incubation at an earlier stage and to a higher rate than the light-grown cells. The activation was markedly enhanced in the PVA gel with a low cell concentration. The immobilized cells consumed nitrate with a temporary accumulation of NO2 and evolved nitrogen gas. The immobilized cells could use various organic compounds as electron donors for denitrification. Thus, the immobilized cells were applied to a continuous treatment of synthetic wastewater using an aparatus devised by this laboratory. The results showed an efficient removal of NO3-N from the test water.  相似文献

Arginase, ornithine carbamoyl transferase (OCT) and arginine deiminase activities were found in cell-free extracts of Nostoc PCC 73102, a free-living cyanobacterium originally isolated from the cycad Macrozamia. Addition of either arginine, ornithine or citrulline to the growth medium induced significant changes in their in vitro activities. Moreover, growth in darkness, compared to in light, induced higher in vitro activities. The in vitro activities of arginase and arginine deiminase, two catabolic enzymes primarily involved in the breakdown of arginine, increased substantially by a combination of growth in darkness and addition of either arginine, or ornithine, to the growth medium. The most significant effects on the in vitro OCT activities where observed in cells grown with the addition of ornithine. Cells grown in darkness exhibited about 6% of the in vivo nitrogenase activity observed in cells grown in light. However, addition of external carbon (glucose and fructose) to cells grown in darkness resulted in in vivo nitrogenase activity levels similar to, or even higher than, cells grown in light. Growth with high in vivo nitrogenase activity or in darkness with the addition of external carbon, resulted in repressed levels of in vitro arginase and arginine deiminase activities. It is suggested that nitrogen starvation induces a mobilization of the stored nitrogen, internal release of the amino compound arginine, and an induction of two catabolic enzymes arginase and arginine deiminase. A similar and even more pronunced induction can be observed by addition of external arginine to the growth medium.  相似文献

Haploid cells of opposite mating type of Saccharomyces cerevisiae conjugate to form zygote. During the conjugation process, the degradation or reorganization of the cell wall and the fusion of the two plasma membranes take place. Since chloroquine inhibits cellular events associated with the reorganization of the plasma membrane, the effect of the drug on conjugation was studied. Chloroquine at a concentration, at which cell growth was not retarded, inhibited zygote formation, while it did not affect other mating functions, such as sexual agglutination, production of and response to mating pheromone. Cells in a mating culture containing chloroquine formed no "prezygote" suggesting that they were not prepared for entering into fusion process. The inhibitory effect of chloroquine was reversible as cells formed zygote when they were washed after treatment with chloroquine. Zygote formation was unaffected in cells possessing chloroquine within vacuoles after incubation with the drug in complete medium (YPD) at pH 7.5, followed by washing. This suggests that chloroquine inhibits zygote formation by adsorbing to the plasma membrane of S. cerevisiae.  相似文献

Cell cycle dependent photosynthesis in the marine dinoflagellate Amphidinium carteri was studied under constant illumination and light/dark (L/D) photocycles to distinguish intrinsic cell cycle control from environmental influences. Cells were grown in constant light and on a 14:10 L:D cycle at light intensities that would yield a population growth rate of 1 doubling per day. In the former case division was asynchronous, and cells were separated according to cell cycle stage using centrifugal elutriation. Cells grown on the L:D cycle were synchronized, with division restricted to the dark period. Cell cycle stage distributions were quantified by flow cytometry. Various cell age groups from the two populations were compared as to their photosynthetic response (photosynthetic rate versus irradiance) to determine whether or not the response was modulated primarily by cell cycle constraints or the periodic L/D cycle. Cell cycle variation in photosynthetic capacity was found to be determined solely by the L/D cycle; it was not present in cells grown in constant light.  相似文献

Morphology and sexual reproduction in Chlorogonium capillatum Nozaki, Watanabe & Aizawa sp. nov. (Volvocales, Chlorophyta) originating from Miyatoko Mire, Japan, were studied under controlled laboratory conditions. Vegetative cells of this new species were fusiform with blunt anterior and posterior ends, and they had a massive parietal chloroplast and numerous contractile vacuoles distributed throughout the protoplast. Several to many pyrenoids were randomly distributed in the chloroplast, but they disappeared under the light microscope when grown photoheterotrophically. During asexual reproduction, the first division took place transversely without a preceding rotation of the parental protoplast. In sexual reproduction, the parental protoplast divided successively to form 32 or 64 small, biflagellate isogametes. After gametogenesis, the gametes did not escape from the parental cell (gametangial) wall, within which pairs of the adjoining gametes fused to form quadriflagellate zygotes. Such zygotes were then released from the parental cell wall and developed into hypnozygotes, which at maturity developed numerous thin spines or hairs on the zygote wall. On zygote germination, four biflagellate germ cells were released from the zygote wall separately. This type of gametic union, "paedogamy," has not previously been described in the green algae except for Chlorococcum echinozygotum Starr . Chlorogonium capillatum can be clearly distinguished from other described species of Chlorogonium by its numerous contractile vacuoles and blunt anterior and posterior ends in vegetative cells as well as by its unique sexual reproduction, in which paedogamous conjugation occurs, and numerous thin spines or hairs that develop on the hypnozygote walls .  相似文献

Cells of certain Chlamydomonas reinhardtii strains dramatically swell and ultimately disintegrate when grown in the dark but not in the light. This effect of darkness is delayed: cells undergo about two divisions before swelling. Measurements on O2 uptake and ATP content indicate that the cells grown in the dark are not deficient in energy supply. Dim blue light not effective in photosynthesis prevents `dark-lethality'. Inhibition of protein synthesis during the latent period with cycloheximide also prevents dark-lethality. We propose that blue light stimulates synthesis of one or more proteins which are part of or are important in assembly of an osmoregulatory complex.  相似文献
