The kinetics of continuous l-sorbose fermentation using Acetobacter suboxydans with and without cell recycle (100%) were investigated at dilution rates (D) of 0.05, 0.10, 0.15 and 0.3 h–1. The biomass and sorbose concentrations for continuous fermentation without recycle increased as the dilution rate was increased from 0.05 to 0.10 h–1. A maximum biomass concentration of 8.44 g l–1 and sorbose concentration of 176.90 g l–1 were obtained at D=0.10 h–1. The specific rate of sorbose production and volumetric sorbose productivity at this dilution rate were 2.09 g g–1 h–1 and 17.69 g l–1 h–1. However, on further increasing the dilution rate to 0.3 h–1, both biomass and sorbose concentrations decreased to 2.93 and 73.20 g l–1 respectively, mainly due to washout of the reactor contents. However, the specific rate of sorbose formation and volumetric sorbose productivity at this dilution rate increased to 7.49 g g–1 h–1 and 21.96 g l–1 h–1 respectively. Continuous fermentation with 100% cell recycle served to further enhance the concentration of biomass and sorbose to 28.27 and 184.32 g l–1 respectively (in the reactor at a dilution rate of 0.05 h–1). Even though, there was a decline in the biomass and sorbose concentrations to 6.8 and 83.40 g l–1 at a dilution rate of 0.3 h–1, the specific rates of sorbose formation and volumetric sorbose productivity increased to 3.67 g g–1h–1 and 25.02 g l–1 h–1.  相似文献

The ribulose monophosphate cycle methylotroph Methylobacillus flagellatum was grown under oxyturbidostat conditions on mixtures of methanol and formaldehyde. Formaldehyde when added at low concentration (50 mg/l) increased the methanol consumption and the yield of biomass. The presence of 150–300 mg/l of formaldehyde resulted in an increase of the growth rate from 0.74 to about 0.79–0.82 h-1. The presence of 500 mg/l of formaldehyde in the inflow decreased culture growth characteristics. Activities of methanol dehydrogenase and enzymes participating in formaldehyde oxidation and assimilation were measured. Summary The growth parameters ofPenicillium cyclopium have been evaluated in a continuous culture system for the production of fungal protein from whey. Dilution rates varied from 0.05 to 0.20 h–1 under constant conditions of temperature (28°C) and pH (3.5). The saturation coefficients in the Monod equation were 0.74 g l–1 for lactose and 0.14 mg l–1 for oxygen, respectively. For a wide range of dilution rates, the yield was 0.68 g g–1 biomass per lactose and the maintenance coefficient 0.005 g g–1 h–1 lactose per biomass, respectively. The maximum biomass productivity achieved was 2 g l–1 h–1 biomass at dilution rates of 0.16–0.17 h–1 with a lactose concentration of 20 g l–1 in the feed. An unsaturated fatty acid auxotroph of the oleaginous yeast Apiotrichum curvatum, named UfaM3, blocked in the conversion of stearic to oleic acid was cultivated in single-stage continuous culture. The influence of consumed carbon to nitrogen ratios (C/N ratios, g g–1) obtained at various dilution rates (D) on fatty acid (FA) accumulation and its profiles were studied. In continuous culture in N-limited medium a maximum FA accumulation of 45.6% (g g–1 of dry biomass) was obtained at an optimal D of 0.049 h–1, recording an efficiency of substrate conversion of 0.48 g g–1 and 0.22 g g–1 for biomass and lipids, respectively. The quality of lipid approached cocoa butter at an optimal C/N ratio of between 20 and 30. The C/N ratio in the incoming medium was 38.5 g g–1 with 30 g l–1 of glucose and both C and N sources were completely consumed at a critical D of 0.07 h–1. Summary The production of l-lactic acid from whey permeate, a waste product of the dairy industry, by fermentation with the lactic acid bacterium Lactobacillus casei subsp. casei was investigated. A fermentation medium consisting of permeate and supplements, which enables exponential growth of the organisms, was developed. A fast method for determination of free and immobilized biomass in solid-rich media, based on measurement of cellular ATP, was evolved. Continuous fermentations in a stirred tank reactor (STR) and in a fluidized bed reactor (FBR) with immobilized biomass were compared. In the STR a volumetric productivity of 5.5 g/l per hour at 100% substrate conversion [dilution rate (D) = 0.22 h–1] was determined. In the FBR porous sintered glass beads were used for immobilization and a maximum biomass concentration of 105 g/kg support was measured. A productivity of 10 g/l per hour was obtained at D = 0.4 h–1 (substrate conversion 93%) and of 13.5 g/l per hour at D = 1.0 h–1 (substrate conversion 50%). Offprint requests to: W. Summary A new methylotrophic strain (T15), which employs the ribulose monophosphate (RuMP) cycle of formaldehyde assimilation, was isolated on the basis of high in vitro activities of formaldehyde and formate dehydrogenases (19 and 678 mU per mg protein, respectively). Serial subculturing of the strain in batch cultures, on 4 g/l CH3OH for 6 months, led to loss of substantial percentages of the NAD-linked formaldehyde (25%) and formate (98%) dehydrogenases. The activities of these two enzymes were partially recovered when cells were grown continuously at very low dilution rate (0.03 h–1). We found large variations (40 to 1000%) in the activities of other key enzymes of carbon-substrate oxidation (both linear and cyclic) and assimilation, in batch cultures with pure and mixed substrates, and in continuous cultures of different dilution rates. Key intracellular reaction rates, including those of the cyclic and linear substrate oxidation, were measured in vivo using a 14C-tracer technique in both continuous and batch cultures. The results indicate significant variations in these reaction rates, particularly those of linear and cyclic carbon oxidation. Overall, the cyclic oxidation appears to be employed to a larger (although not predominant) extent in strain T15 compared with another RuMP strain (L3) we have previously examined. T15 exhibits high biomass yields (up to 0.63 g cells per g CH3OH) and growth rates (up to 0.46 h–1) on CH3OH in batch cultures. CH3NH2 can also be utilized as a substrate. In continuous culture, T15 could be grown at dilution rates up to 0.36 h–1 with a corresponding biomass yield of 0.4. Examination of a large number of data on the biomass yields of strains T15 and L3 reveals that the large variations in yields derive from the variable branching of carbon flow between linear and cyclic oxidation and assimilation, rather than changes in the biosynthetic efficiency of carbon incorporation into biomass.  相似文献

Cells of the propionate-tolerant strain Propionibacterium acidipropionici P200910, immobilized in calcium alginate beads, were tested for propionic and acetic acid production both in a semidefined laboratory medium and in corn steep liquor in batch, fed-batch, and continuous fermentation. Cell density was about 9.8 × 109 cells/g (wet weight) of beads, and beads were added to the medium at 0.1 g (wet weight) beads/ml. Beads could be reused for several consecutive batch fermentations; propionic acid production in the tenth cycle was about 50%–70% of that in the first cycle. In batch culture complete substrate consumption (glucose in semidefined medium, lactate in corn steep liquor) and maximum acid production were seen within 36 h, and acid yields from the substrate were higher than in free-cell fermentations. Fed-batch fermentations were incubated up to 250 h. Maximum propionic acid concentrations obtained were 45.6 g/l in corn steep liquor and 57 g/l in semidefined medium; this is the highest concentration achieved to date in our laboratory. Maximum acetic acid concentrations were 17 g/l and 12 g/l, respectively. In continuous fermentation of semidefined medium, dilution rates up to 0.31 h–1 could be used, which gave higher volumetric productivities (0.96 g l–1 h–1 for propionic acid and 0.26 g l–1 h–1 for acetic acid) than we have obtained with free cells. Summary Some environmental affects on cell aggregation described in the literature are briefly summarized. By means of a biomass recirculation culture (Contact system), using the yeast Torulopsis glabrata, the aggregation behavior of cells in static and in dynamic test systems is described. Sedimentation times required to obtain 50 g · l–1 yeast dry matter in static systems were always higher than in dynamic ones.In addition to, influencing the biomass yield, the specific growth rate of the yeast also affected cell aggregation. Reassessement of bioenergetic growth yield of Arthrospira platensis was performed by using continuous culture under both autotrophic and mixotrophic conditions. Continuous culture was carried out at dilution rates of 0.017, 0.023 and 0.030 h–1. Under these dilution rates bioenergetic yields ranged between 4.45–6.03 × 10–3 g biomass kJ–1 and between 5.42–7.46 × 10–3 g biomass kJ–1, under autotrophic and mixotrophic conditions respectively. A maximum bioenergetic yield of 8.1 × 10–3 g biomass kJ–1 using an autotrophic culture can be calculated. Pigment accumulation (chlorophyll a and carotenoids) may be related to light irradiance, reaching a maximum pigment concentration under light saturation irradiance. Phycocyanin concentration increased during light limitation.  相似文献

Kinetic parameters and physiological states of Corynebacterium glutamicum at the growing and l-lysine-overproducing phase were characterised in continuous culture on threonine-limited complex and minimal media. High l-lysine productivity occurred at dilution rates ranging from 0.1 h–1 to 0.3 h–1 on threonine-limited complex medium, and at dilution rates ranging from 0.1 h–1 to 0.15 h–1 in minimal medium. l-Lysine yields of 0.25 g/g (0.31 g/g as l-lysine hydrochloride) in complex medium, and of 0.17 g/g (0.21 g/g as l-lysine hydrochloride) in minimal medium, corresponding respectively to intrinsic yields of 0.533 g/g and 0.572 g/g were obtained. These intrinsic yield factors are closed to the theoretical ones (0.608 g/g, 0.75 mol/mol). Intrinsic biomass yields were calculated as 0.658 g/g in complex medium and 0.283 g/g in minimal medium. CO2 production has been clearly related to l-lysine production. According to our results on specific uptake rates and specific productivities in complex medium, metabolic rearrangement should occur during the transition from the growing phase to the l-lysine-overproducing phase. A thermotolerant methylotrophicBacillus sp. (KISRI TM1A, NCIMB 40040), isolated from the Kuwaiti environment and belonging to the group II spore-forming, bacilli, could not be correlated with any knownBacillus sp. It may, therefore, be a new species. It grew at temperatures from 37° to 58°C from pH 6.5 to 9.0 and on methanol up to 40 g l–1. It grew well in a chemostat. Its biomass yield coefficient was improved by about 30% by optimization of medium and growth conditions, reaching a maximum of 0.44g g–1 at 45°C pH 6.8 to 7.0, dilution rate 0.25 h–1 with methanol at 10 g l–1. Average crude protein and amino acid content were 84% and 60%, respectively, and maximum productivity attained under laboratory conditions was 5.06 g l–1h–1. It was concluded that this strain has good potential for use in single-cell protein production.  相似文献

Depending on the biomass yield on glucose and the cell morphology ofBacillus thuringiensis, three different metabolic states were observed in continuous culture. At dilution rates between 0.18 h–1 and 0.31 h–1 vegetative cells, sporulating bacteria and spores coexisted, while glucose and amino acids were consumed. Only vegetative cells were observed at dilution rates between 0.42 h–1 and 0.47 h–1 and glucose was used as the main carbon and energy source. AtD = 0.50 h–1 the biomass yield on glucose decreases sharply. To define better the specific growth rate range in which the microorganism uses mainly glucose, a dilution rate of 0.25–0.45 h–1 was studied. The experimental data could be adjusted to a Monod model and the following rate coefficients and growth yields were determined: maximum specific growth rate 0.54 h–1, saturation constant 0.56 mg glucose ml–1, biomass growth yields 0.43 g cells (g glucose)–1, and 0.76 g cells (g oxygen)–1, and maintenance coefficients 0.065 g glucose (g cells)–1 h–1 and 0.039 g oxygen (g cells)–1 h–1.  相似文献

Summary A salicylate-hydroxylase-producing strain of Pseudomonas putida with an unusual capability to grow at toxic levels of salicylate up to 10 g l–1 has been isolated. It grew well under continuous culture conditions, with optimum growth at pH 6.5 and a temperature of 25° C. The use of an ammonium salt as a nitrogen source, instead of nitrate, resulted in a 30–40% increase in its biomass yield coefficient. Optimum growth under continuous culture conditions was achieved using 4 g l–1 salicylate at 25° C, pH 6.5 and 0.2 h–1 dilution rate. High salicylate hydroxylase enzyme activity [236 units (U) l–1] and productivity (424.8 U h–1) were obtained at a dilution rate of 0.45 h–1 using a mineral medium containing 4 g l–1 of salicylate. Operating under continuous culture conditions with oxygen limitation and a slight accumulation of residual salicylate (0.2 g l–1) resulted in a decrease in culture performance and enzyme productivity. Lactobacillus delbrueckii subsp.bulgaricus ATCC 11842 was grown in a chemostat at 45°C and pH 5.5 using glucose as the carbon source, with the aim of optimizing biomass production. Cells were grown in a complex medium under nitrogen. At dilution rates lower than 0.18h–1, it was difficult to keep steady-state conditions and pleomorphic forms were observed. The addition of 30mM Ca2+ and Mn2+ reverted the cells to normal shape: 30mM Mg2+ had no effect. Increasing the dilution rate resulted in normal morphology without the addition of any cations. Under these conditions, a maximum productivity of 1.24g dry biomass 1–1 h–1 was obtained. The maximum growth yield, corrected for maintenance, was 30g biomass mol–1 glucose and the maintenance energy was 0.26g glucose g–1 biomass h–1. Lactate was the main fermentation product at all glucose concentrations used in the fed medium. Cells grown at high dilution rates had normal technological properties (acid production and proteolysis) when tested in milk.  相似文献

Several biofilters and biotrickling filters were used for the treatment of a mixture of formaldehyde and methanol; and their efficiencies were compared. Results obtained with three different inert filter bed materials (lava rock, perlite, activated carbon) suggested that the packing material had only little influence on the performance. The best results were obtained in a biotrickling filter packed with lava rock and fed a nutrient solution that was renewed weekly. A maximum formaldehyde elimination capacity of 180 g m–3 h–1 was reached, while the methanol elimination capacity rose occasionally to more than 600 g m–3 h–1. Formaldehyde degradation was affected by the inlet methanol concentration. Several combinations of load vs empty bed residence time (EBRTs of 71.9, 46.5, 30.0, 20.7 s) were studied, reaching a formaldehyde elimination capacity of 112 g m–3 h–1 with about 80% removal efficiency at the lowest EBRT (20.7 s).  相似文献

Summary Candida tropicalis S001 was grown on the lipid fraction of a protein-containing waste-water in order to (i) remove fat from the water, and (ii) produre yeast biomass for feed. The yeast cells were separated from the waste-water by sedimentation. Defatted waste-water was used for methane production and gave a yield of a 0.3 m3 methane/kg reduced chemical oxygen demand. The maximum specific growth rate (µmax) of C. tropicalis growing on waste-water fat at pH 4.0 was 0.35 h–1; the fat content was decreased from 8 g/l to about 0.1 g/l within 24 h. In continous culture a corresponding reduction was maintained at dilution rates up to 0.36 h–1. The effect on growth of pH, temperature and CO2 concentration was studied with triolein as the major carbon source. The µmax was nearly constant (0.16 h–1) in the pH and temperature range of 3.2–4.0 and 30°–38° C, respectively; 10% CO2 was optimal for growth. Summary Growth of Propionibacterium acidi-propionici was studied on lactose as substrate and in acid whey permeate in a three-electrode poised-potential system with cobalt sepulchrate as artificial electron donor. In batch culture experiments in a stirred-tank reactor the substrate was fermented completely to propionic acid up to 6.5 g 1–1 lactose in a supplemented whey permeate medium. No acetic acid was produced during the growth of P. acidi-propionici. An electron flow of 80–100 mA was obtained and the electron balance was 101%. In continuously growing cultures with 3 g 1–1 of lactose as the substrate, propionate was formed as the only fermentation product up to a dilution rate (D) of 0.04 h–1. With D>0.04 h–1 the bacteria immobilized on the working electrode surface. It was examined whether an electron transfer occurred between the platinum working electrode and the immobilized cells. Summary A methanol/nitrate-medium and anaerobic conditions yielded an enrichment culture which consisted ofHyphomicrobium andParacoccus. This mixed culture proved to be very effective in denitrification of solutions containing high concentrations of nitrate and free nitric acid when grown in a chemostat (D=0.04 h-1). With 0.1 mol/l nitric acid solution as feed medium the pH in the culture vessel adjusted itself to 5.8. For the reduction of 1 g NO3–N 2.6 g methanol were consumed and 0.56 g cells were produced.  相似文献

Summary A mixed methanol-utilizing bacterial culture was utilized to produce bacterial biomass as milk replacer. This culture comprised three pure strains: KISRI-5 (NCIB 12135), KISRI-512 (NCIB 12137) and KISRI-5112 (NCIB 12138). Optimal concentrations of methanol (15 g 1–1) and medium elements as well as optimal growth conditions, e.g., pH (6.8), temperature (38°C), dissolved oxygen and dilution rate, were established. The maximum biomass yield coefficient obtained under optimized conditions was 0.48 g g–1.· Large-scale production was successfully carried out in a 1500 1 fermenter under chemostat conditions. A good product was obtained having high true protein content (59–62%) and low polysaccharides (5%) without microbial contamination.  相似文献
