An empirical and generalized model is presented, based on a modified Arrhenius equation, for predicting the combined effect of temperature and water activity on the growth rate of bacteria. When it was applied to seven separate sets of wide ranging published results, spanning some 50 years and including a spore-former and a silage micro-organism, predictions explained between 92.9 and 99.0% of the variation in the results with an overall mean of 96.6%. Advantages over existing models are that it is relatively easy to fit to data using least squares regression and requires only five coefficients. These, together with its simplicity and demonstrated wide application, will facilitate its practical use.  相似文献

Growth rates of tsetse populations were estimated by calculating the dominant eigenvalues of appropriate Leslie matrices. The individual effects of four variables (pre-adult and adult survival probability, interlarval period and pupal duration), have been investigated by varying each one over a wide range of values, while the other three are held constant. R, the log of the growth rate, was found to vary approximately linearly with adult and pre-adult death rate; a 1% change in the adult death rate causes approximately a 10-fold change in R. R varies linearly with the log of fecundity and of the pupal duration. An increase in the pupal duration results in a decrease in the growth rate for populations which have a positive growth rate, but an increase for populations which have a negative growth rate. For a population at equilibrium, a change in the pupal duration has no effect. Small changes in fecundity have less effect on the growth rate than small changes in the death rate; this fact is advanced as an important contributor to the generally very cautious nature of female tsetse, and their aversion to man, particularly as a potential host. A simple linear model is described which relates R to all four variables and their first order interactions. The model is used to produce a set of graphs which encapsulate the relationship between the growth rate and the vital parameters over a wide range of values. It is also used to draw the loci on one side of which tsetse populations grow, and on the other of which they decline. Population resilience is discussed in relation to the problem of tsetse eradication; it is concluded that if one can impose and sustain an added mortality of 4% per day on any female tsetse population then it must go extinct, regardless of the strength of the density dependent processes; and it seems likely that in most field conditions only an added 2-3% is required. It is pointed out that ground and aerial spraying techniques produce much higher daily mortalities than this, but they may often not be sustained for sufficiently long to achieve eradication. When odour-baited targets are used the increased death rate is much smaller, but it can be sustained as required; recent work in Zimbabwe shows that there is a good correspondence between the calculated imposed death rate and the observed rate of decline of tsetse populations.  相似文献

Epidermal growth factor (EGF) known as a urgastrone is a powerful mitogen with a wide variety of possibilities for medical usages. A mature EGF coding region was isolated from human prepro-EGF sequence by a conventional PCR and cloned into pQE vector in which the gene product was supposed to be expressed with 6×His tag for the subsequent purification. The recombinant mature EGF was expressed in M15[Rep4], anEscherichia coli host strain, in amount of 30–40% of total proteins present inE. coli extract by the addition of isopropylthio-β-galactopyranoside (IPTG). The recombinant EGF purified using a Ni2+-NTA affinity column chromatography was active in its ability to induce phosphorylation on tyrosine residues of several substrate proteins when murine NIH3T3 and human MRC-5 fibroblast cells were stimulated with it. This work may provide the basic technology and information for the production of recombinant EGF.  相似文献

Summary A survey was made of 81 starch-assimilating yeasts, representing 59 species and varieties, with respect to their capacity for the direct conversion of starch into SCP. The extent of starch conversion by the native amylases of the strains during exponential growth, expressed as yield on starch (final amount of dry biomass formed per unit mass of starch originally supplied), varied over a wide range (0.043–0.590) The highest yields were obtained with strains ofLipomyces starkeyi andL. kononenkoae which converted on the average respectively 84% and nearly 100% of the starch supplied. The rate of starch hydrolysis byL. kononenkoae did not limit its specific rate of growth and SCP production.  相似文献

The human placenta is a tumor-like tissue in which highly proliferative, migratory, and invasive extra-villous trophoblast cells, migrate and invade the uterus and its vasculature, to provide a vital link between the mother and the developing fetus. In the present article, we review our studies on a series of experiments, designed to identify molecular events responsible for the phenotypic changes during placental growth. Our observations illustrate that the human placenta is endowed with the biochemical machinery to proliferate indefinitely throughout gestation, yet, there are intrinsic mechanisms that effectively circumscribe the extent and duration of trophoblast proliferation. The placenta combines in itself the unique ability to produce a wide variety of protein, peptide and steroid hormones, but intricately interwoven in this process, is also the remarkable capacity to simultaneously regulate their synthesis and secretion. The placenta therefore represents an autonomous or a self-sufficient unit capable of modulating its own growth and function, while assisting the developing fetus until it is capable of independent existence.  相似文献

Several growth factors implicated in the process of cellular transformation were tested for their ability to induce anchorage-independent (AI) growth of primary rat embryo (RE) cells. Our results show that in the presence of 10% calf serum, platelet-derived growth factor (PDGF), 1-30 ng/ml, has the strongest effect of all growth factors tested on AI growth. Type-beta transforming growth factor (TGF-beta), by itself, does not stimulate AI growth, and it inhibits the PDGF-induced colony formation in a dose-dependent manner (ED50 approximately 0.03 ng/ml). Qualitatively similar responses are obtained by using an established line of fibroblasts, NIH 3T3 cells; the principal difference between the response of the primary cells and the established cell line is in colony-forming efficiency in soft agar culture (15% and 90%, respectively, for growth of colonies greater than 1,500 micron2 diameter in the presence of 10 ng/ml PDGF). Since AI growth has been shown to correlate well with tumorigenicity in vivo, our results suggest that the transforming potential of PDGF in an appropriate responsive cell can be controlled not only through its interaction with its own receptor, but also by the presence of inhibitory factors such as TGF-beta.  相似文献

The empirical and quantitative (linear Arrhenius) model of Davey (1989a) was assessed against a range of published lag phase data, some of which included combined temperature—water activity as influencing factors while others had temperature as the sole influencing factor. For two large ( n ≧ 27) sets of combined temperature and water activity data, the model explained 74·2 and 86·4% of the variation in results, and for eight wide ranging data sets with temperature as the sole factor, it explained between 80·8 and 99·7% of the variation in results, with an overall mean of 95·7%. These published data included both Gram-positive and Gram-negative bacteria and a spore former. The model has four coefficients which can be obtained by relatively unsophisticated users with linear regression analyses on most desk top calculators or personal computers.  相似文献

While bulk crystallization from impure solutions is used industrially as a purification step for a wide variety of materials, it is a technique that has rarely been used for proteins. Proteins have a reputation for being difficult to crystallize and high purity of the initial crystallization solution is considered paramount for success in the crystallization. Although little is written on the purifying capability of protein crystallization or of the effect of impurities on the various aspects of the crystallization process, recent published reports show that crystallization shows promise and feasibility as a purification technique for proteins. To further examine the issue of purity in macromolecule crystallization, this study investigates the effect of the protein impurities, avidin, ovalbumin, and conalbumin at concentrations up to 50%, on the solubility, crystal face growth rates, and crystal purity of the protein lysozyme. Solubility was measured in batch experiments while a computer controlled video microscope system was used to measure the ?110? and ?101? lysozyme crystal face growth rates. While little effect was observed on solubility and high crystal purity was obtained (>99.99%), the effect of the impurities on the face growth rates varied from no effect to a significant face specific effect leading to growth cessation, a phenomenon that is frequently observed in protein crystal growth. The results shed interesting light on the effect of protein impurities on protein crystal growth and strengthen the feasibility of using crystallization as a unit operation for protein purification.  相似文献

The dynamics of Pseudomonas fluorescens VKM-1472 growth was studied under the conditions of batch and continuous cultivation, and the behaviour of the organism was investigated upon nutrition shifts and starvation. At D greater than 0.375 h-1, just as in the batch culture with a substrate excess, the strain realised excessive metabolism [15]: the yield in terms of the substrate fell down (38% for the batch culture and 40% for the continuous culture), the titre of viable cells decreased to a considerable extent, and maintenance expenditures increased (3 times for qgluc and 7.5 times for qO2). When the culture was incubated under the oligotrophous conditions, the biomass yield decreased fourfold within 8 days and the titre of viable cells dropped down twofold within the same period of time. However, the organism was still capable of oxidising a wide range of substrates (17 substrates were studied). As soon as a substrate was added, it was oxidised at a high rate and changes in the macromolecular composition were characteristic of an "up-jump" in the growth rate [11]. It is possible that the growth and behaviour of this organism are associated with its ecological niche occupied in natural systems.  相似文献

Existing approaches to characterizing quantitative trait loci (QTL) utilize a paradigm explicitly focused on the direct effects of genes, where phenotypic variation among individuals is mapped onto genetic variation of those individuals. For many characters, however, the genotype of the mother via its maternal effect accounts for a considerable portion of the genetically based variation in progeny phenotypes. Thus the focus on direct effect QTL may result in an insufficient or misleading characterization of genetic architecture due to the omission of the potentially important source of genetic variance contributed by maternal effects. We analyze the relative contribution of direct and maternal effect (ME) QTL to early growth in mice using a three-generation intercross of the Small (SM/J) and Large (LG/J) inbred mouse lineages. Using interval mapping and composite interval mapping, direct effect (DE) QTL for early growth (change in body mass during the interval from week 1 to 2) were detected in the F(2) generation of the intercross (n = 510), where no maternal genetic effect variance is present (all individuals are progeny of genetically identical F(1) mothers). ME QTL were detected by treating the phenotypes of cross-fostered F(3) pups as a characteristic of their nurse-dam (n = 168 dams with cross-fostered progeny). Five DE QTL, significant at a chromosome wide level (alpha = 0.05), were detected, with two significant at a genome wide level. FourME QTL significant at the chromosome wide level were detected, with three significant at the genome wide level. A model containing only DE QTL accounted for 11.8% of phenotypic variance, while a model containing only ME QTL accounted for 31.5% of the among litter variance in growth. There was no evidence for pleiotropy of DE and ME loci since there was no overlap between loci detected in these two analyses. Epistasis between all pairs of loci was analyzed for both DEs and MEs. Ten pairs of loci showed significant epistasis for MEs (alpha = 0.05 corrected for multiple comparisons) while four pairs showed significant epistasis for DEs on early growth.  相似文献

以盆栽当年生广西地不容为材料,研究不同光照强度(100%、50%、30%和15%自然光强)对其光合特性和生长的影响。结果表明:随着光照强度的降低,广西地不容最大净光合速率(P_(max))、光饱和点(LSP)、光补偿点(LCP)先减小,而后稍有增大;表观量子效率(AQY)在30%和50%光强下显著高于100%和15%光强处理;叶片叶绿素总量(Chl)、叶绿素a(Chla)、叶绿素b(Chlb)、类胡萝卜素(Car)含量随光强的减弱而增大,Car/Chl随光强的减弱而减小,Chla/Chlb比值在各处理间无显著差异;单叶面积随生长光强的减弱而增大,比叶重(LMA)则随着生长光强的减弱而减小;30%光强下广西地不容块根生物量最高,光照过强和过弱都不利于其生物量的积累。广西地不容对光强的适应范围较广,但光照过强或过荫均对生长造成不良影响,光合速率降低,生长减缓,块根生物量积累下降,30%光强是其当年生苗生长的最佳光强。  相似文献

Transforming growth factor-beta (TGF-beta) is synthesized and secreted by a wide variety of cells, including cells of the immune system. Lymphocytes and monocytes possess high affinity TGF-beta receptors and the addition of TGF-beta to in vitro cell cultures results in significant modulation of immune function. TGF-beta inhibits the proliferation of thymocytes, T cells, B cells, and natural killer cells. Additionally, it inhibits certain differentiative functions of lymphocytes including a marked inhibition of immunoglobulin production by human B lymphocytes. TGF-beta has dichotomous actions on monocytes. It is a potent chemoattractant for monocytes and induces interleukin 1 mRNA expression while inhibiting generation of reactive oxygen intermediates and monocyte killing. Evidence is accumulating that TGF-beta regulates immune function in vivo and that overproduction of TGF-beta may be associated with immunosuppression.  相似文献

利用盆栽试验研究水分胁迫下AM真菌对黄顶菊生长和抗旱性的影响,揭示黄顶菊入侵过程中的微生物学机制。以苯菌灵为杀真菌剂,在土壤相对含水量为120%、80%、40%和20%条件下,分别设灭菌和不灭菌两种处理。结果表明,水分胁迫显著降低了黄顶菊株高、干重和主根长,而对AM真菌侵染率无显著影响。施用苯菌灵显著降低了菌根侵染率、叶片保水力、保护酶活性、可溶性糖和可溶性蛋白含量,提高了MDA含量。不灭菌处理下黄顶菊植株对土壤有效N和有效P的利用率较高,且植株全N、P含量显著高于灭菌处理,菌根贡献率随土壤相对含水量降低而逐渐提高,重度胁迫分别是渍水条件下的1.84和1.88倍。土壤水分状况和AM真菌的交互作用对黄顶菊生物量和生理指标影响显著。AM真菌共生能够促进黄顶菊根系对土壤水分和矿质营养吸收,改善植物代谢活动,提高抗旱性。实验结果为黄顶菊合理防控措施的制定提供了依据,同时作为丛枝菌根的基础性研究也具有重要的意义。  相似文献

Transforming Growth Factor-Beta (TGF-β) is the general name for a family of naturally-occurring polypeptides which have multiple regulatory effects on cell proliferation and differentiation. Over the last decade it has become apparent that TGF-βs can be produced by most cell types and exert a wide range of effects in a context-dependent autocrine, paracrine or endocrine fashion via interactions with distinct receptors on the cell surface. This review summarizes current knowledge concerning the molecular and cellular biology of TGF-β3, the most recently described mammalian isoform, and focuses on those physiological actions which may lead to clinical applications, particularly in the indication areas of wound healing and chemoprotection.  相似文献

Improvement in growth and meat quality is one of the main objectives in sire line pig breeding programmes. Mapping quantitative trait loci for these traits using experimental crosses and a linkage‐based approach has been performed frequently in the past. The Piétrain breed often was involved as a founder breed to establish the experimental crosses. This breed was selected for muscularity and leanness but shows relatively poor meat quality. It is frequently used as a sire line breed. With the advent of genome‐wide and dense SNP chips in pig genomic research, it is possible to also conduct genome‐wide association studies within the Piétrain breed. In this study, around 500 progeny‐tested sires were genotyped with 60k SNPs. Data filtering showed that around 48k SNPs were useable in this sample. These SNPs were used to conduct a genome‐wide association study for growth, muscularity and meat quality traits. Because it is known that a mutation in the RYR1 gene located on chromosome 6 shows a major effect on meat quality, this mutation was included in the models. Single‐marker and multimarker association analyses were performed. The results revealed between zero and eight significant associations per trait with P < 5 × 10?5. Of special interest are SNPs located on SSC6, SSC10 and SSC15.  相似文献

The precise neuroendocrine mechanisms underlying activation of hypothalamic-pituitary-gonadal (HPG) axis maturation are elusive. The wide age range of pubertal onset among normal individuals throughout the world may suggest that both genetic and environmental factors modulate the timing of puberty. Early activation of the HPG axis, termed central precocious puberty (CPP), causes psychosocial difficulties and may lead to compromised final height, especially if medical intervention is delayed. Although CPP is considered to be idiopathic in the majority of patients, we have recently reported a 27.5% prevalence of familial cases among 147 patients with idiopathic CPP. Segregation analysis of this cohort suggested an autosomal dominant transmission with incomplete sex-dependent penetrance. Allelic variants of candidate genes that regulate the timing of puberty may cause familial CPP. Detection of these genes will provide a tool for identification of children at risk of developing CPP, enabling early intervention with the aim of preventing its distressing outcomes.  相似文献

Heparin-binding EGF-like growth factor (HB-EGF) is a member of the EGF family and is an important therapeutic target in some types of human cancers. KM3566 is a mouse anti-HB-EGF monoclonal antibody that neutralizes HB-EGF activity by inhibiting the binding of HB-EGF to its receptors. Based on the results of our pharmacokinetics study, a humanized derivative antibody, KHK2866, is rapidly cleared from serum and shows nonlinear pharmacokinetics in cynomolgus monkeys. In this study, we examined the antigen-dependent clearance of an anti-HB-EGF monoclonal antibody in vivo and in vitro in order to pharmacokinetically explain the rapid elimination of KHK2866. We revealed tumor size-dependent clearance of KM3566 in in vivo studies and obtained good fits between the observed and simulated concentrations of KM3566 based on the two-compartment with a saturable route of clearance model. Furthermore, in vivo imaging analyses demonstrated tumor-specific distribution of KM3566. We then confirmed rapid internalization and distribution to lysosome of KM3566 at a cellular level. Moreover, we revealed that the amounts of HB-EGF on cell surface membrane were maintained even while HB-EGF was internalized with KM3566. Recycled or newly synthesized HB-EGF, therefore, may contribute to a consecutive clearance of KM3566, which could explain a rapid clearance from serum. These data suggested that the rapid elimination in pharmacokinetics of KM3566 is due to antigen-dependent clearance. Given that its antigen is expressed in a wide range of normal tissue, it is estimated that the rapid elimination of KHK2866 from cynomolgus monkey serum is caused by antigen-dependent clearance.  相似文献
