The grass Alloteropsis semialata (R.Br.) Hitchcock is uniquein that both Kranz and non-Kranz leaf anatomy has been reportedin this species. The present study investigates Kranz formsof A. semialata collected from a single ecological niche. Theseplants exhibit morphological and anatomical differences withrespect to leaf area, stomatal size and stomatal distribution.Carbon dioxide and water exchange measurements in the two formsshow the expected pattern of higher photosynthetic rate andhigher water utilization efficiency associated with Kranz anatomy.No intermediate physiological response or anatomical form wasobserved in this sample. Background and Aims: The grass Alloteropsis semialata is the only plant species withboth C3 and C4 subspecies. It therefore offers excellent potentialas a model system for investigating the genetics, physiologyand ecological significance of the C4 photosynthetic pathway.Here, a molecular phylogeny of the genus Alloteropsis is constructedto: (a) confirm the close relationship between the C3 and C4subspecies of A. semialata; and (b) infer evolutionary relationshipsbetween species within the Alloteropsis genus. Methods: The chloroplast gene ndhF was sequenced from 12 individuals,representing both subspecies of A. semialata and all four ofthe other species in the genus. ndhF sequences were added tothose previously sequenced from the Panicoideae, and used toconstruct a phylogenetic tree. Key Results: The phylogeny confirms that the two subspecies of A. semialataare among the most recently diverging lineages of C3 and C4taxa currently recognized within the Panicoideae. Furthermore,the position of the C3 subspecies of A. semialata within theAlloteropsis genus is consistent with the hypothesis that itsphysiology represents a reversion from C4 photosynthesis. Thedata point to a similar evolutionary event in the Panicum stenodesP.caricoidesP. mertensii clade. The Alloteropsis genusis monophyletic and occurs in a clade with remarkable diversityof photosynthetic biochemistry and leaf anatomy. Conclusions: These results confirm the utility of A. semialata as a modelsystem for investigating C3 and C4 physiology, and provide moleculardata that are consistent with reversions from C4 to C3 photosynthesisin two separate clades. It is suggested that further phylogeneticand functional investigations of the Alloteropsis genus andclosely related taxa are likely to shed new light on the mechanismsand intermediate stages underlying photosynthetic pathway evolution.  相似文献

In species of Clusia, switching from C3-photosynthesis (C3-PS)to crassulacean acid metabolism (CAM) may be a means of optimizingwater use, plant carbon balance and photon utilization duringperiods of stress. We ask whether, in perennial species of Clusia,the switch from CAM back to C3-PS is also of ecophysiologicalsignificance. Our objective was to investigate the performanceof C. minor L. during a short-term shift from CAM to C3-PS.During the transition from CAM to C3-PS, nocturnal malate andcitrate accumulation decreased whereas CO2uptake increased duringthe daytime. However, after 7 d, marked nocturnal accumulationof citrate and 24 h CO2uptake occurred. Total peroxidase activities and peroxidase isozymes were comparedamong 10 bush- and vine-squash cultivars of Cucurbita maximaDuch. and C. pepo (L.) Several qualitative variations of isozymeswere detected between C. maxima and C. pepo, and some differenceswere evident among varieties of C. pepo. There were no qualitativedifferences between genetically similar bush and vine strains.Quantitative differences were difficult to resolve, althoughit did appear that in varieties of C. maxima exhibiting lowlevels of peroxidase activity, isozymes C3, C4, and C6 wereless intense. Depending upon the cultivars compared, total peroxidase activityin bush forms was either higher or lower than, or the same astheir vine counterparts. Since the major bush genes in the varietiesstudied are believed to be allelic, the results indicate thatthe over-all genotype of squash varieties can influence therelative expression of peroxidase activity between bush andvine forms.  相似文献

Gas exchange measurements were undertaken on 2-year-old plantsof Clusia rosea. The plants were shown to have the ability toswitch from C3-photosynthesis to CAM and vice versa regardlessof leaf age and, under some conditions, CO2 was taken up continuously,throughout the day and night. The light response was saturatedby 120 µmol m–2 s–1 typical of a shade plant. Gas exchange patterns in response to light, water and VPD wereexamined. All combinations of daytime and night-time CO2 uptakewere observed, with rates of CO2 uptake ranging from 2 to 11µmol m–2 s–1 depending upon water status andlight. Categorization of this plant asC3, CAM or an intermediateis impossible. Differing VPD affected the magnitude of changesfrom CAM to C3-photosynthesis (0 to 0.5 and 0 to 6.0 µmolm–2 s–1 CO2, respectively) when plants were watered.Under well-watered conditions, but not under water stress, highPPFD elicited changes from CAM to C3 gas exchange. This is unusualnot only for a shade plant but also for a plant with CAM. Itis of ecological importance for C. rosea, which may spend theearly years of its life as an epiphyte or in the forest understorey,to be able to maximize photosynthesis with minimal water loss. RuBP carboxylase-oxygenase protein in three C3 species (Nicotianatabacum L., Solanum tuberosum L., Triticum aestivum L.) andthree C4 species (Panicum miliaceum L., Panicum texanum Buckl.,Zea mays L.) was quantitatively determined by sucrose densitygradient centrifugation and by immunochemical assay using antibodyraised to crystallized tobacco leaf RuBP carboxylase-oxygenase.The C3 species had 3- to 6-fold higher concentrations of RuBPcarboxylase-oxygenase than the C4 species when expressed oneither a chlorophyll or a leaf area basis. The C3 species alsoallocated a higher fraction of their total soluble protein tothis enzyme (from 25 to 60% for the C3 species compared to 8to 23% for the C4 species). There was no RuBP carboxylase-oxygenaseprotein or activity in the C4 mesophyll cells, while the enzymeconstituted from 20 to 40% of the total soluble protein in theC4 bundle sheath cells. A close correlation (r = +0·91)was found between catalytic activity and level of the enzymeprotein in the species examined.  相似文献

An hypothesis was formulated that phosphorus (P) partitioningin tissues of C4 leaves would permit C4 plants to resist P deficiencybetter than C3 plants. To test this hypothesis, 12 C3, C4, andC3–C4 intermediate species were grown at adequate, deficient,and severely deficient P supply in a solid-phase-buffered sandculture system to characterize photosynthetic and growth responses.Species differed considerably in response to P stress. The growthof C3 species was more sensitive to P supply than C4 species,but C3 and C4 species had similar photosynthetic P use efficiency,and C4 species did not have low leaf P content, contrary toour hypothesis. In fact, leaf photosynthetic rates were notcorrelated with growth responses. Moncots had lower leaf P contentand better maintenance of leaf production under P stress thandicots, because of greater inhibition of branching (dicots)than of tillering (monocots). The most P efficient species inthis survey was Brachiaria, a C4 monocot that increased rootbiomass allocation under stress while maintaining P allocationto the shoot. It is concluded that C4 species are not inherentlymore P efficient than C3 species, but that monocots are moreP efficient than dicots, because of contrasting P and biomassallocation under stress. Anatomical and morphological structures of leaf blades werecompared between C3 and C4 species in Panicum. Inter-specificvariation of stomatal density, longitudinal vein density andmesophyll thickness was highly correlative either plus or minuswithin respective groups. The two groups could not be distinguishedby a single character, since the variation ranges overlappedeach other. However, the quantitative relations between veindensity and the other two characters differentiated the twogroups well. In C3, stomatal density seemed to be a primaryfactor for regulating water balance, while in C4 vein systemwas considered to be important for the regulation. The specieswith intermediate photosynthesis behaved similar to the C3 species.In the C3 group, correlative variation was observed betweenleaf width, leaf angle and the three characters mentioned above.Variation of light-receiving area due to the changes of widthand angle of leaf blades was considered to be one of the adaptivestrategies of this group. Increase of light-receiving area wasin connection with the thinning of leaves. On the other hand,in the C4 correlations between length, width and angle of leaveswere low. Such loose character correlation may be achieved byits efficiency of CO2 utilization and its well developed veinsystems. Besides, NAD-me type species tended to have relativelylower stomatal and vein densities as compared with the otherdecarboxylation types in this group. The influence of varying light intensity and quality on thecarbon labelling patterns in Rumex vesicarius (a C3 plant),Setaria italica (a malate-formingC4 plant), and Amaranthus paniculatus(an aspartate-forming C4 plant) was studied. In A. paniculatusand B. vesicarius blue light decreased the transfer of radioactivityto sugars and starch but in S. italica only slightly decreasedradioactivity in sugar phosphates, sucrose, and insolubles.Negligible transfer was observed from the C4 acids to sugarphosphates, sucrose, and starch under dim blue-green and blue-yellowlights in S. italica and A. paniculatus. Blue light favouredthe formation of malate, aspartate, and alanine in all threeplants. The differential effect of blue and red light suggesteda variation in the mechanisms of C4-photosynthesis in Setariaand Amaranthus. Leaves of S. italica and A. paniculatus were allowed to photosynthesizein 14CO2 for 5 s and then the distribution of the labelled productsbetween the mesophyll and the bundle sheath cells was determinedduring subsequent photosynthesis in 12CO2. Malate and aspartatewhich appeared initially in the mesophyll layer moved rapidlyinto the bundle sheath cells. Phosphoglyceric acid originatingin the bundle sheath moved swiftly to the mesophyll layer. Sugarphosphates were recovered from both the mesophyll and the bundlesheath cells. Most of the starch was found in the bundle sheathcells while sucrose and alanine were localized in the mesophyllcells.  相似文献

Kleczkowski, L. A. and Randall, D. D. 1987. Immunologicallydistinct forms of adenylate kinase in leaves: comparison ofsubunit size of adenylate kinase from C3 and C4 plants.—J.exp. Bot. 38: 1440–1445. Antibodies prepared against maize leaf adenylate kinase (E.C.2.7.4.3 [EC] ) cross-reacted with the enzyme isolated from leavesof both C3 and C4 plants. The immunoreaction was very specificas judged by the presence of a single band on Western immunoblotscontaining proteins from leaf extracts of several species. Themolecular weight (M1) of adenylate kinase determined by meansof the immunoblotting was 29 kD and 27 kD for C4 and C3 species,respectively. For both C3 and C4 plants, the antibodies failedto precipitate all adenylate kinase activity in leaf extracts,while they were 100% effective in pelleting the enzyme frommaize mesophyll chloroplasts. This indicated the presence ofat least two immunologically distinct forms of adenylate kinasein leaves. It is suggested that the observed differences in molecular structure(M1s) of adenylate kinase from C3 and C4 species might be responsiblefor distinct catalytic and functional properties of the enzymein these two groups of plants. C4 photosynthesis is a complex trait resulting from a series of anatomical and biochemical modifications to the ancestral C3 pathway. It is thought to evolve in a stepwise manner, creating intermediates with different combinations of C4‐like components. Determining the adaptive value of these components is key to understanding how C4 photosynthesis can gradually assemble through natural selection. Here, we decompose the photosynthetic phenotypes of numerous individuals of the grass Alloteropsis semialata, the only species known to include both C3 and C4 genotypes. Analyses of δ13C, physiology and leaf anatomy demonstrate for the first time the existence of physiological C3–C4 intermediate individuals in the species. Based on previous phylogenetic analyses, the C3–C4 individuals are not hybrids between the C3 and C4 genotypes analysed, but instead belong to a distinct genetic lineage, and might have given rise to C4 descendants. C3 A. semialata, present in colder climates, likely represents a reversal from a C3–C4 intermediate state, indicating that, unlike C4 photosynthesis, evolution of the C3–C4 phenotype is not irreversible.  相似文献

Pascopyrum smithii (C3) andBouteloua gracilis (C4) are importantforage grasses native to the Colorado shortgrass steppe. Thisstudy investigated photosynthetic responses of these grassesto long-term CO2enrichment and temperature in relation to leafnonstructural carbohydrate (TNC) and [N]. Glasshouse-grown seedlingswere transferred to growth chambers and grown for 49 d at twoCO2concentrations (380 and 750 µmol mol-1) at 20 and 35°C, and two additional temperatures (25 and 30 °C) at750 µmol mol-1CO2. Leaf CO2exchange rate (CER) was measuredat a plant's respective growth temperature and at two CO2concentrationsof approx. 380 and 700 µmol mol-1. Long-term CO2enrichmentstimulated CER in both species, although the response was greaterin the C3,P. smithii . Doubling the [CO2] from 380 to 750 µmolmol-1stimulated CER ofP. smithii slightly more in plants grownand measured at 30 °C compared to plants grown at 20, 25or 35 °C. CO2-enriched plants sometimes exhibited lowerCER when compared to ambient-grown controls measured at thesame [CO2], indicating photosynthetic acclimation to CO2growthregime. InP. smithii , such reductions in CER were associatedwith increases in TNC and specific leaf mass, reductions inleaf [N] and, in one instance, a reduction in leaf conductancecompared to controls. InB. gracilis , photosynthetic acclimationwas observed more often, but significant changes in leaf metabolitelevels from growth at different [CO2] were generally less evident.Temperatures considered optimal for growth (C3: 20 °C; C4:35 °C) sometimes led to CO2-induced accumulations of TNCin both species, with starch accumulating in the leaves of bothspecies, and fructans accumulating only inP. smithii. Photosynthesisof both species is likely to be enhanced in future CO2-enrichedand warmer environments, although responses will sometimes beattenuated by acclimation. We report new information on silica deposition in 15 plant species,including nine grasses, two sedges and four composites. Thesilica depositional patterns found in seven of the grass speciesindicate that they are C4 plants. However the festucoid grassCortaderia selloana is a C3 plant with long leaf trichomes andoval silica structures in the leaves. In contrast the panicoidC4 grasses Chasmathium latifolium, Chasmathium sessiflorum,Imperata cylindrica, Panicum repens, Panicum commutatum andSetaria magna, all produce dumb-bell-shaped silica structuresin the leaves. The chloridoid grasses Spartina patens and Spartinacynosuroides have saddle-shaped structures and no dumb-bellor oval shaped ones. The sedges Rhynchospora plumosa and Scirpuscyperinus were found to have oval phytoliths and may be C3 plants.Our examination of these and other grasses strongly suggeststhat C4 grasses tend to produce the same type of silica cells.Grasses and sedges with C3 type photosynthesis tend to produceoval silica structures. The composite Grindelia squarrosa andsunflowers Helianthus angustifolia, Helianthus atrorubens andHelianthus tuberosus absorb relatively small amounts of siliconand larger amounts of calcium, where both elements deposit inleaf trichomes. We found no clear indicator for the C3 sunflowersor C4 types in the Asteraceae. Helianthus tuberosus leaves havemany trichomes on the adaxial surface. These trichomes havea higher concentration of silica than the surrounding leaf surface.Helianthus tuberosus leaves had much higher ash and silica contentsthan those of Helianthus angustifolia and Helianthus atrorubens.The composite Grindelia squarrosa has a usual deposition ofsilica in the basal cells around the guard cells. Silica depositionoften reflects the surface features of a leaf. An exceptionis Scripus cyperinus where the silica structures are deep inthe tissue and do not reflect the surface configurations. Theinforescence of Setaria magna had a 14.64 silica content. Thetufts of white, silky hairs characteristic of Imperata cylindricainflorescence have no silica. The maximum catalytic activities of several photorespiratoryand photosynthetic enzymes were determined in leaf extractsof three C3–C4 intermediates (Alternanthera ficoides,A. tenella and Parthenium hysterophorus) and were compared tothose of C3 (A. sessiles, Pisum sativum) and C4 (A. pungens,Zea mays and Amaranthus hypochondriacus) species. The activitylevels of key photorespiratory enzymes, glycolate oxidase, catalase,NADH-hydroxypyruvate reductase and glycerate kinase were less(28 to 35% reduced) in intermediates than those of typical C3species. Similarly, the activities of photorespiratory aminotransferasesin the C3–C4 intermediates were also partially reduced(23 to 37% reduction). The activities of phosphoenolpyruvatecarboxylase (PEPC), pyruvate, orthophosphate dikinase and NAD-malicenzyme were higher (2 to 7 times) in leaf extracts of the intermediatesthan those of C3 species. Enzymes of the C4, C3 pathway and photorespiration have beenanalyzed for P. hians and P. milioides, which have chlorenchymatousbundle sheath cells in the leaves. On whole leaf extracts thelevels of PEP carboxylase are relatively low compared to C4species, RuDP carboxylase is typical of C3 species, and enzymesof photorespiratory metabolism appear somewhat intermediatebetween C3 and C4. Substantial levels of PEP carboxylase, RuDPcarboxylase, and photorespiratory enzymes were found in bothmesophyll and bundle sheath cells. Low levels of C4-acid decarboxylatingenzymes may limit the capacity for C4 photosynthesis in P. hiansand P. milioides. C4 photosynthesis is a complex trait that boosts productivity in warm environments. Paradoxically, it evolved independently in numerous plant lineages, despite requiring specialised leaf anatomy. The anatomical modifications underlying C4 evolution have previously been evaluated through interspecific comparisons, which capture numerous changes besides those needed for C4 functionality. Here, we quantify the anatomical changes accompanying the transition between non‐C4 and C4 phenotypes by sampling widely across the continuum of leaf anatomical traits in the grass Alloteropsis semialata. Within this species, the only trait that is shared among and specific to C4 individuals is an increase in vein density, driven specifically by minor vein development that yields multiple secondary effects facilitating C4 function. For species with the necessary anatomical preconditions, developmental proliferation of veins can therefore be sufficient to produce a functional C4 leaf anatomy, creating an evolutionary entry point to complex C4 syndromes that can become more specialised.  相似文献

Fatty acids of chain length from C4 to C12 inhibited ethyleneproduction in wounded albedo tissue of Hassaku (Citrus hassakuHort. ex Tanaka) fruit. Of the fatty acids tested, caprylicacid (C8) and capric acid (C10) were the most effective. Lauricacid (C12) was less effective, and caproic acid (C6) and butyricacid (C4) were the least effective. Caprylic acid at 5 mM markedlyinhibited ethylene production in not only wounded albedo tissueof citrus fruit but also apple (Malus sylvestris Mill.) cortex,tomato (Lycopersicon esculentum Mill.) pericarp, cucumber (Cucumissativus L.) cortex, banana (Musa AAA group Cavendish subgroup)pulp, broccoli (Brassica oleracea L.) floret, spinach (Spinaciaoleracea L.) leaf, lettuce (Lactuca sativa L.) leaf and mungbean (Vigna radiata [L.] Wilczek) hypocotyl. Caprylic acid inhibitedethylene production at the step of conversion of l-aminocyclopropane-l-carboxylicacid to ethylene. Panicum hians and Panicum milioides were found to have characteristicsintermediate to those of C3 and C4 species with respect to CO2compensation point, percentage inhibition of photosynthesisby O2 at various O2/CO2 solubility ratios, and water use efficiency.C4 species have a higher carboxylation efficiency than eitherthe intermediate or C3 species. During photosynthesis, evenunder 2.5% O2, C4 species have a higher affinity for intercellularCO2 (Km 1.6 µM) apparently due to the initial carboxylationthrough PEP carboxylase. Under low O2 the intermediate and C3species had a similar affinity for intercellular CO2 duringphotosynthesis (Km 5–7 µM) consistent with carboxylationof atmospheric CO2 through RuDP carboxylase. There were considerablevariation in photosynthesis/unit leaf area at saturating CO2levels in the species examined which in part is due to differencesin RuDP carboxylase /unit leaf area. The highest rates of photosynthesis/unitleaf area under CO2-saturating conditions were with the C3 specieswhich had a correspondingly high level of RuDP carboxylase/unitleaf area. The kinetic properties of phosphoenolpyruvate (PEP) carboxylasehave been studied among several Flaveria species: the C3 speciesF. cronquistii, the C3–C4 species F. pubescens and F.linearis, and the C4 species F. trinervia. At either pH 7 or8, the maximum activities (in µmol.mg Chl–1.h–1)for F. pubescens and linearis (187–513) were intermediateto those of the C3 species (12–19) and the C4 species(2,182–2,627). The response curves of velocity versusPEP concentration were hyperbolic for the C3 and C3–C4species at either pH 7 or 8 while they were sigmoidal for theC4 species at pH 7 and hyperbolic at pH 8. The Km values forPEP determined from reciprocal plots were lowest in the C3 species,and of intermediate value in the C3–C4 species comparedto the K' values of the C4 species determined from Hill plotsat either pH 7 or 8. Glucose-6-phosphate (G6P) decreased theKm values for PEP at both pH 7 and 8 in the C3 and C3–C4species. In the C4 species, G6P decreased the K' values at pH8 but increased the K' values at pH 7. In all cases, G6P hadits effect by influencing the activity at limiting PEP concentrationswith little or no effect on the maximum activity. At pH 8 andlimiting concentrations of PEP the degree of stimulation ofthe activity by G6P was greatest in the C4 species, intermediatein F. linearis, a C3–C4 species, and lowest in the C3species. Bot. 38: 433–441. S. anglica is one of the few C4 species which occurs naturallyin cool temperate zones. It is known to attain photosyntheticrates which equal or exceed those of C3 grasses over the temperaturerange typical of the spring and summer in cool temperate climates.This study examines whether S. anglica can also attain comparablegrowth rates at these temperatures. Seedlings of S. anglicaand L. perenne cv. S23 were grown in controlled environmentsat 10,15,20 and 25 °C. Quantitative growth analysis wasconducted by taking frequent harvests to determine the progressionsof leaf area and plant weight of individual plants with time.Quadratic regressions were found to describe these progressionswell. Instantaneous derived growth parameters were calculatedfrom the fitted regressions. Both absolute and relative growthrates of S. anglica were significantly lower than for L. perenne,this being largely attributable to a lower ratio of leaf areaproduction per unit of plant dry weight. Although the amountof dry matter invested into leaves was similar, the leaf areaper unit of leaf dry weight was lower in S. anglica. In comparisonto L. perenne, the rate of dry matter accumulation per unitof leaf area (ULR) was higher in S. anglica at 25 °C andinitially equal at 10 °C. Prolonged exposure to 10 °Csteadily reduced ULR in S. anglica which approached zero at80 d. Although growth in S. anglica is reduced more by low temperaturethan it is in L. perenne, by comparison to other C4 speciesthe assimilatory capacity of S. anglica is more tolerant oflow temperature exposure.
