In two montane watersheds that receive minimal deposition of atmospheric nitrogen, 15–71% of dissolved organic nitrogen (DON) was bioavailable in stream water over a 2-year period. Discharge-weighted concentrations of bulk DON were between 102 and 135 μg/l, and the C:N ratio differed substantially between humic and non-humic fractions of DON. Approximately 70% of DON export occurred during snowmelt, and 40% of that DON was biologically available to microbes in stream sediments. Concentrations of bioavailable DON in stream water were 2–16 times greater than dissolved inorganic nitrogen (DIN) during the growing season, and bioavailable DON was depleted within 2–14 days during experimental incubations. Uptake of DON was influenced by the concentration of inorganic N in stream water, the concentration of non-humic DON in stream water, and the C:N ratio of the non-humic fraction of dissolved organic matter (DOM). Uptake of DON declined logarithmically as the concentration of inorganic N in stream water increased. Experimental additions of inorganic N also caused a decline in uptake of DON and net production of DON when the C:N ratio of non-humic DOM was high. This study indicates that the relative and absolute amount of bioavailable DON can vary greatly within and across years due to interactions between the availability of inorganic nutrients and composition of DOM. DOM has the potential to be used biotically at a high rate in nitrogen-poor streams, and it may be generated by heterotrophic microbes when DIN and labile DOM with low relative nitrogen content become abundant.  相似文献

The spatial distribution of bacterial abundance and production were measured every 4 h in a recently flooded oligo-mesotrophic reservoir (the Sep Reservoir, Puy-De-Dôme, France), in relation to concentrations of dissolved carbohydrates and combined amino acids. The concentration of dissolved organic matter (DOM) components in the recently flooded Sep Reservoir were higher than those measured in other lakes of similar trophic status. Short-term variations in the bacterial production in this new reservoir appeared cyclical and endogenous to bacterial communities. These results highlight the need for the evaluation of diel changes in bacterial production, if estimation of the daily production rate of bacteria is to be done accurately for a reliable model of carbon flow through bacterioplankton and ultimately through aquatic microbial food webs. Bacterial growth, measured over time and space, did not appear exclusively governed by DOM components from phytoplankton primary production.  相似文献

The trophic link between bacteria and bacterivorous protists is a complex interaction that involves feedback of inorganic nutrients and growth substrates that are immeadiately available for prey growth. These interactions were examined in the laboratory and in incubations of concentrated natural assemblages of bacterioplankton. Growth dynamics of estuarine and marine bacterivorous protists were determined in laboratory culture using Vibrio natriegens as prey and were compared to growth of protists on bacterioplankton assemblages concentrated by tangential flow filtration from four northwest Florida Estuaries. Biomass transfers from bacteria to protists were monitored by tracing elemental carbon and nitrogen in particulate fractions of protist added and grazer free controls. Gross growth efficiencies of the protists on naturally occurring bacteria were within the range determined in lab estimates of growth efficiency on cultured bacteria (50%). However, bacterial response to protist excretion products was different in the lab and field incubations, and bacterial growth contributed to the biomass available to protists in the field incubations. As determined by radioisotope-labeled substrate incorporation, a time lag in bacterial reponse to protist excretion products was observed for laboratory batch cultures, allowing accurate estimation of growth efficiency. In incubations with concentrated natural bacterial assemblages, bacterial growth response coincided with protist growth and excretion. The additional bacterial production on protist excretion products reached a maximum of 2–3-fold higher than protist-free controls. In addition, ammonium concentrations increased with protist grazing and growth in lab cultures, but ammonium excreted by protists in concentrates did not accumulate. The C:N values for the bacterial concentrates suggests that these bacteria were nitrogen limited. It is speculated that dissolved organic carbon, concentrated by tangential flow filtration (> 100,000 MW membrane) with the bacterioplankton, was utilized by bacteria when nitrogen was supplied as ammonium and amino acids from protist excretion. Thus, estimates of protist growth efficiency on naturally occurring bacterioplankton, corrected for protist-stimulated bacterial production, were in the range of 13–21%.  相似文献

Nitrogen frequently limits oceanic photosynthesis and the availability of inorganic nitrogen sources in the surface oceans is shifting with global change. We evaluated the potential for abrupt increases in inorganic N sources to induce cascading effects on dissolved organic matter (DOM) and microbial communities in the surface ocean. We collected water from 5 m depth in the central North Pacific and amended duplicate 20 liter polycarbonate carboys with nitrate or ammonium, tracking planktonic carbon fixation, DOM production, DOM composition and microbial community structure responses over 1 week relative to controls. Both nitrogen sources stimulated bulk phytoplankton, bacterial and DOM production and enriched Synechococcus and Flavobacteriaceae; ammonium enriched for oligotrophic Actinobacteria OM1 and Gammaproteobacteria KI89A clades while nitrate enriched Gammaproteobacteria SAR86, SAR92 and OM60 clades. DOM resulting from both N enrichments was more labile and stimulated growth of copiotrophic Gammaproteobacteria (Alteromonadaceae and Oceanospirillaceae) and Alphaproteobacteria (Rhodobacteraceae and Hyphomonadaceae) in weeklong dark incubations relative to controls. Our study illustrates how nitrogen pulses may have direct and cascading effects on DOM composition and microbial community dynamics in the open ocean.  相似文献

An understanding of the distribution of colored dissolved organic matter (CDOM) in the oceans and its role in the global carbon cycle requires a better knowledge of the colored materials produced and consumed by marine phytoplankton and bacteria. In this work, we examined the net uptake and release of CDOM by a natural bacterial community growing on DOM derived from four phytoplankton species cultured under axenic conditions. Fluorescent humic-like substances exuded by phytoplankton (excitation/emission [Ex/Em] wavelength, 310 nm/392 nm; Coble's peak M) were utilized by bacteria in different proportions depending on the phytoplankton species of origin. Furthermore, bacteria produced humic-like substances that fluoresce at an Ex/Em wavelength of 340 nm/440 nm (Coble's peak C). Differences were also observed in the Ex/Em wavelengths of the protein-like materials (Coble's peak T) produced by phytoplankton and bacteria. The induced fluorescent emission of CDOM produced by prokaryotes was an order of magnitude higher than that of CDOM produced by eukaryotes. We have also examined the final compositions of the bacterial communities growing on the exudates, which differed markedly depending on the phytoplankton species of origin. Alteromonas and Roseobacter were dominant during all the incubations on Chaetoceros sp. and Prorocentrum minimum exudates, respectively. Alteromonas was the dominant group growing on Skeletonema costatum exudates during the exponential growth phase, but it was replaced by Roseobacter afterwards. On Micromonas pusilla exudates, Roseobacter was replaced by Bacteroidetes after the exponential growth phase. Our work shows that fluorescence excitation-emission matrices of CDOM can be a helpful tool for the identification of microbial sources of DOM in the marine environment, but further studies are necessary to explore the association of particular bacterial groups with specific fluorophores.  相似文献

In aquatic ecosystems, carbon (C) availability strongly influences nitrogen (N) dynamics. One manifestation of this linkage is the importance in the dissolved organic matter (DOM) pool of dissolved organic nitrogen (DON), which can serve as both a C and an N source, yet our knowledge of how specific properties of DOM influence N dynamics are limited. To empirically examine the impact of labile DOM on the responses of bacteria to DON and dissolved inorganic nitrogen (DIN), bacterial abundance and community composition were examined in controlled laboratory microcosms subjected to various combinations of dissolved organic carbon (DOC), DON, and DIN treatments. Bacterial communities that had colonized glass beads incubated in a stream were treated with various glucose concentrations and combinations of inorganic and organic N (derived from algal exudate, bacterial protein, and humic matter). The results revealed a strong influence of C availability on bacterial utilization of DON and DIN, with preferential uptake of DON under low C concentrations. Bacterial DON uptake was affected by the concentration and by its chemical nature (labile versus recalcitrant). Labile organic N sources (algal exudate and bacterial protein) were utilized equally well as DIN as an N source, but this was not the case for the recalcitrant humic matter DON treatment. Clear differences in bacterial community composition among treatments were observed based on terminal restriction fragment length polymorphisms (T-RFLP) of 16S rRNA genes. C, DIN, and DON treatments likely drove changes in bacterial community composition that in turn affected the rates of DON and DIN utilization under various C concentrations.  相似文献

Understanding the quantity and quality of dissolved organic matter (DOM) in potential watershed sources is critical for explaining and quantifying the exports of DOM in stream runoff. Here, we examined the concentration and quality of DOM for ten watershed sources in a 12?ha forested catchment over a two-year period. DOM composition was evaluated for: throughfall, litter leachate, soil water (zero and tension), shallow and deep groundwater, stream water, hyporheic zone, and groundwater seeps. DOM quality was measured using a suite of optical indices including UV–visible absorbance and PARAFAC modeling of fluorescence excitation-emission matrices (EEMs). DOM concentrations and quality displayed a pronounced trend across watershed sources. Surficial watershed sources had higher DOM concentrations and more humic-like DOM with higher molecular weight whereas deeper groundwater sources were rich in % protein-like fluorescence. The greater % contribution of protein-like fluorescence in groundwater suggested that a larger fraction of groundwater DOM may be bioavailable. DOM for wetland groundwater was more aromatic and humic-like than that at the well-drained riparian location. Principal component analyses (PCA) revealed that the differences in surficial watershed compartments were dictated by humic-like components while groundwater sources separated out by % protein-like fluorescence. Observations from optical indices did not provide any conclusive evidence for preferential association of dissolved organic carbon (DOC) or dissolved organic nitrogen (DON) with any particular DOM quality pools.  相似文献

The photochemical transformation of dissolved organic matter (DOM) in lakes and oceans has been shown to either reduce or enhance bacterial utilization. We compared the effects of UV radiation on the bacterial use of DOM in a wide range of lakes. Although complex DOM was converted in all irradiated samples into carboxylic acids that are readily utilized by bacteria, irradiation in several lakes resulted in a decreased ability of DOM to support bacterial growth. The effect of irradiation on the ability of DOM to promote bacterial growth was a positive function of the terrestrial humic matter, and a negative function of indigenous algal production. We suggest that the net effect of irradiation is a result of counteracting but concurrent processes rendering DOM either labile or recalcitrant. Humic DOM is predominantly transformed into forms of increased lability, whereas photochemical transformation into compounds of decreased bacterial substrate quality dominates in algal-derived DOM. Hence, solar-induced photochemical reactions interact with microbial degraders in different ways, depending on the origin and nature of the organic matter, affecting the transfer of energy within aquatic food webs, as well as the degradation and preservation of detrital organic matter, in different directions.  相似文献

Urbanization has the potential to dramatically alter the biogeochemistry of receiving freshwater ecosystems. We examined the optical chemistry of dissolved organic matter (DOM) in forty-five urban ponds across southern Ontario, Canada to examine whether optical characteristics in these relatively new ecosystems are distinct from other freshwater systems. Dissolved organic carbon (DOC) concentrations ranged from 2 to 16 mg C L-1 across the ponds with an average value of 5.3 mg C L-1. Excitation-emission matrix (EEM) spectroscopy and parallel factor analysis (PARAFAC) modelling showed urban pond DOM to be characterized by microbial-like and, less importantly, by terrestrial derived humic-like components. The relatively transparent, non-humic DOM in urban ponds was more similar to that found in open water, lake ecosystems than to rivers or wetlands. After irradiation equivalent to 1.7 days of natural solar radiation, DOC concentrations, on average, decreased by 38% and UV absorbance decreased by 25%. Irradiation decreased the relative abundances of terrestrial humic-like components and increased protein-like aspects of the DOM pool. These findings suggest that high internal production and/or prolonged exposure to sunlight exerts a distinct and significant influence on the chemistry of urban pond DOM, which likely reduces its chemical similarity with upstream sources. These properties of urban pond DOM may alter its biogeochemical role in these relatively novel aquatic ecosystems.  相似文献

Soil organic matter (SOM) is the largest terrestrial C pool, and retention and release of dissolved organic matter (DOM) cause formation and loss of SOM. However, we lack information on how different sources of DOM affect its chemical composition, and how DOM chemical composition affects retention. We studied seasonal controls on DOM production and chemical controls on retention in soils of a temperate coniferous forest. The O horizon was not usually the dominant source for dissolved organic C (DOC) or N (DON) as has been reported for other sites. Rather, net production of both DOC and DON was often greater in the shallow mineral soil (0–10 cm) than in the O horizon. DOM production in the shallow mineral soil may be from root exudation as well as turnover of fine roots and microflora in the rhizosphere. In the field, the two acid fractions (hydrophobic and hydrophilic acids) dominated the soil solution at all depths. A major portion of net production and removal of total DOC within the soil column was explained by increases and decreases in these fractions, although a shift in chemical composition of DOM between the O and mineral soil horizons suggested different origins of DOM in these layers. A larger loss of the free amino fraction to deep soil water at this study site than at other sites suggested lower retention of labile DON. Field DOM removal measurements suggest that field-measured parameters may provide a good estimate for total DOM retained in mineral soil.  相似文献

Production, transport, and degradation of terrestrial dissolved organic matter (DOM) influence carbon (C) and nutrient cycling in both soils and downstream aquatic ecosystems. Here, we assessed the impacts of wildfire on DOM production, composition, and reactivity (biodegradation versus UV degradation) from soils of upland forest and peatland ecosystems. Soil C solubility was lowest for upland char samples, highest from surface soils in unburned spruce stands and decreased with a higher degree of peat humification regardless of fire history. Soil nitrogen (N) became relatively more soluble in both upland and peat soils post-fire, as leachate C/N decreased. Biodegradability was lower for DOM leachates from burned than unburned soils, both in upland and peatland sites. Several DOM composition indices were related to biodegradability; with the strongest relationship for specific UV absorbance at 254 nm (indicator of aromaticity). Parallel factor analysis revealed distinctive characteristics of leachates from burned soils and char that were related to low biodegradability and high UV-mediated losses. Relative to dark incubations, incubation under UV conditions led to greater C losses for highly aromatic leachates, but reduced losses for leachates with low aromaticity. This suggests that UV-mediated degradation could provide a pathway for highly stable terrestrial organic matter, including char, to become rapidly mineralized and released to the atmosphere once it reaches aquatic ecosystems in dissolved form. Together our results demonstrate that wildfire can potentially alter both turnover of DOM in terrestrial soils and linkages between terrestrial and aquatic C cycling through its influence on terrestrial DOM production and composition.  相似文献

Incubation experiments were performed to examine the processing of fresh autochthonous dissolved organic matter (DOM) produced by coastal plankton communities in spring and autumn. The major driver of observed DOM dynamics was the seasonally variable inorganic nutrient status and characteristics of the initial bulk DOM, whereas the characteristics of the phytoplankton community seemed to have a minor role. Net accumulation of dissolved organic carbon (DOC) during the 18-days experiments was 3.4 and 9.2 µmol l?1 d?1 in P-limited spring and N-limited autumn, respectively. Bacterial bioassays revealed that the phytoplankton-derived DOC had surprisingly low proportions of biologically labile DOC, 12.6% (spring) and 17.5% (autumn). The optical characteristics of the DOM changed throughout the experiments, demonstrating continuous heterotrophic processing of the DOM pool. However, these temporal changes in optical characteristics of the DOM pool were not the same between seasons, indicating seasonally variable environmental drivers. Nitrogen and phosphorus availability is likely the main driver of these seasonal differences, affecting both phytoplankton extracellular release of DOM and its heterotrophic degradation by bacteria. These findings underline the complexity of the DOM production and consumption by the natural planktonic community, and show the importance of the prevailing environmental conditions regulating the DOM pathways.  相似文献

Mineralization of dissolved organic matter (DOM) in thermokarst lakes plays a non-negligible role in the permafrost carbon (C) cycle, but remains poorly understood due to its complex interactions with external C and nutrient inputs (i.e., aquatic priming and nutrient effects). Based on large-scale lake sampling and laboratory incubations, in combination with 13C-stable-isotope labeling, optical spectroscopy, and high-throughput sequencing, we examined large-scale patterns and dominant drivers of priming and nutrient effects of DOM biodegradation across 30 thermokarst lakes along a 1100-km transect on the Tibetan Plateau. We observed that labile C and phosphorus (P) rather than nitrogen (N) inputs stimulated DOM biodegradation, with the priming and P effects being 172% and 451% over unamended control, respectively. We also detected significant interactive effects of labile C and nutrient supply on DOM biodegradation, with the combined labile C and nutrient additions inducing stronger microbial mineralization than C or nutrient treatment alone, illustrating that microbial activity in alpine thermokarst lakes is co-limited by both C and nutrients. We further found that the aquatic priming was mainly driven by DOM quality, with the priming intensity increasing with DOM recalcitrance, reflecting the limitation of external C as energy sources for microbial activity. Greater priming intensity was also associated with higher community-level ribosomal RNA gene operon (rrn) copy number and bacterial diversity as well as increased background soluble reactive P concentration. In contrast, the P effect decreased with DOM recalcitrance as well as with background soluble reactive P and ammonium concentrations, revealing the declining importance of P availability in mediating DOM biodegradation with enhanced C limitation but reduced nutrient limitation. Overall, the stimulation of external C and P inputs on DOM biodegradation in thermokarst lakes would amplify C-climate feedback in this alpine permafrost region.  相似文献

Attenuation of ultraviolet (UV)-radiation into the water column is highly correlated with the concentration of the dissolved organic matter (DOM). Thus UV penetrates deeper into marine waters than into freshwater systems. DOM is efficiently cleaved by solar surface radiation levels consuming more oxygen than bacterial metabolism. This photolytically cleaved DOM exhibits higher absorbance ratios (250/365 nm) than untreated DOM. Natural bacterioplankton reach higher abundance if inoculated in previously solar-exposed DOM than in untreated DOM; during bacterial growth the absorbance ratio declines steadily indicating the utilization of the photolytically cleaved DOM. On the other hand, bacterioplankton are greatly reduced in their activity if exposed to surface solar radiation levels. Photoenzymatic repair of DNA induced by UV-A radiation, however, leads to an efficient recovery of bacterial activity once the UV-B stress is released. Turbulent mixing of the upper layers of the water column leads to a continuous alteration of the UV exposure regime. Close to the surface, bacteria and DOM are exposed to high levels of UV-B leading to a reduction in bacterial activity and to photolysis of DOM. Once mixed into deeper layers where UV-B is attenuated, but sufficient UV-A is remaining to allow photoenzymatic repair, the photolytically cleaved DOM is efficiently taken up by bacterioplankton leading to even higher bacterial activity than prior to the exposure. Thus, the overall effect of UV on bacterioplankton is actually an enhancement of bacterial activity despite their lack of protective pigments.  相似文献

Photolysis of dissolved organic matter (DOM) leads to contrasting effects on bacterioplankton dynamics, i.e. stimulation and inhibition of bacterial activity. In particular, the role of short‐lived reactive oxygen species (ROS), e.g. singlet oxygen (1O2), in altering microbial activity and species composition has scarcely been investigated. Therefore, we have artificially increased the natural rate of 1O2 formation in short‐term (~4 h) in situ and long‐term (72 h) laboratory incubations of surface water samples from a humic acid‐rich lake. Denaturing gradient gel electrophoresis (DGGE) patterns revealed significant changes in occurrence of abundant bacterioplankton phylotypes upon 1O2 exposure. Cluster analysis of DGGE patterns showed that a moderate increase in 1O2 exposure leads to similar changes in different years indicating the establishment of bacterial communities adapted to 1O2 exposure. Bacterioplankton phylotypes favoured under these conditions belonged to Betaproteobacteria of the beta II cluster (e.g. Polynucleobacter necessarius) and the beta I cluster related to Limnohabitans (R‐BT subcluster) as well as Alphaproteobacteria affiliated to Novosphingobium acidiphilum. In contrast, Actinobacteria of the freshwater acI‐B cluster were sensitive even against moderate 1O2 exposure. We conclude that 1O2 exposure due to DOM photolysis represents an important natural selective factor affecting bacterial species dynamics in aquatic ecosystems in many ways.  相似文献

利用水生植物修复受污染水体中,水生植物在秋冬季节腐烂分解会释放大量溶解性有机物(DOM),DOM可影响水环境中污染物的迁移转化,对水体中的化学和生物过程产生一定影响。因此利用紫外可见光光谱(UV-vis)结合平行因子分析法(PARAFAC)和主成分分析法(PCA)来表征和分析水生植物腐解产物中DOM的组分及其特点。UV-vis的研究表明随着腐解时间的增加,样品中DOM的腐殖化程度逐渐升高,当腐殖化程度达到最高值时,腐解进入矿化阶段,此时腐殖化程度逐渐降低。通过PARAFAC分析可以分离出3种类蛋白组分(C1、C2和C5)和2种类腐殖酸组分(C3和C4)。由PCA可以得出在腐解初期,类蛋白组分占据主导地位;随着腐解时间的增加,类蛋白组分含量逐渐降低,而类腐殖酸组分含量逐渐升高。  相似文献

Dissolved substances released during decomposition of the white water lily (Nymphaea odorata) can alter the growth rate of Okefenokee Swamp bacterioplankton. In microcosm experiments dissolved compounds released from senescent Nymphaea leaves caused a transient reduction in the abundance and activity of water column bacterioplankton, followed by a period of intense bacterial growth. Rates of [H]thymidine incorporation and turnover of dissolved d-glucose were depressed by over 85%, 3 h after the addition of Nymphaea leachates to microcosms containing Okefenokee Swamp water. Bacterial activity subsequently recovered; after 20 h [H]thymidine incorporation in leachate-treated microcosms was 10-fold greater than that in control microcosms. The recovery of activity was due to a shift in the composition of the bacterial population toward resistance to the inhibitory compounds present in Nymphaea leachates. Inhibitory compounds released during the decomposition of aquatic macrophytes thus act as selective agents which alter the community structure of the bacterial population with respect to leachate resistance. Soluble compounds derived from macrophyte decomposition influence the rate of bacterial secondary production and the availability of microbial biomass to microconsumers.  相似文献

Abstract In a study of bacterioplankton in an oligotrophic lake in northern Wisconsin, a community fingerprinting technique, automated ribosomal intergenic spacer analysis (ARISA), was used to determine the effect of resources and trophic interactions on bacterioplankton diversity. Inorganic nitrogen and phosphorus (NP), carbon in the form of glucose (G) or dissolved organic matter extracted from peat (DOM), and carbon and NP in combination were added to two types of experimental systems. Ten-liter mesocosms contained all components of the original aquatic community except for large zooplankton. One-liter dilution cultures were prepared so that the effects of grazers and phytoplankton were removed. During a 3-day incubation, bacterial production showed the greatest response to the carbon plus NP treatment in both experimental systems, but bacterial diversity was strikingly different between them. In the mesocosms, the number of ARISA-PCR fragments averaged 41 per profile, whereas the dilution culture communities were highly reduced in complexity, dominated in most cases by a single PCR fragment. Further analysis of the mesocosm data suggested that whereas the NPDOM addition caused the greatest aggregate bacterial growth response, the addition of NP alone caused the largest shifts in community composition. These results suggest that the measurement of aggregate responses, such as bacterial production, alone in studies of freshwater bacterial communities may mask the effects of resources on bacterioplankton. Synergy of Fresh and Accumulated Organic Matter to Bacterial Growth   总被引:1,自引:0,他引:1  
The main goal of this research was to evaluate whether the mixture of fresh labile dissolved organic matter (DOM) and accumulated refractory DOM influences bacterial production, respiration, and growth efficiency (BGE) in aquatic ecosystems. Bacterial batch cultures were set up using DOM leached from aquatic macrophytes as the fresh DOM pool and DOM accumulated from a tropical humic lagoon. Two sets of experiments were performed and bacterial growth was followed in cultures composed of each carbon substrate (first experiment) and by carbon substrates combined (second experiment), with and without the addition of nitrogen and phosphorus. In both experiments, bacterial production, respiration, and BGE were always higher in cultures with N and P additions, indicating a consistent inorganic nutrient limitation. Bacterial production, respiration, and BGE were higher in cultures set up with leachate DOM than in cultures set up with humic DOM, indicating that the quality of the organic matter pool influenced the bacterial growth. Bacterial production and respiration were higher in the mixture of substrates (second experiment) than expected by bacterial production and respiration in single substrate cultures (first experiment). We suggest that the differences in the concentration of some compounds between DOM sources, the co-metabolism on carbon compound decomposition, and the higher diversity of molecules possibly support a greater bacterial diversity which might explain the higher bacterial growth observed. Finally, our results indicate that the mixture of fresh labile and accumulated refractory DOM that naturally occurs in aquatic ecosystems could accelerate the bacterial growth and bacterial DOM removal.  相似文献
