The population dynamics and production ecology of the two dominant copepod species, Acanthocyclops robustus and Mesocyclops leuckarti, in Tjeukemeer (The Netherlands) were studied for three successive years. Since copepods in Tjeukemeer show continuous recruitment, a population dynamics model INSTAR was developed and used to integrate field data on population density, population structure and fecundity and laboratory data on development rates and length-weight relationships. The pattern of size specific mortality indicated that both invertebrate and vertebrate predation were important in the regulation of population numbers.  相似文献

A suite of experiments are presented for the measurement of H–C, C–C, C–C and HN–N couplings from uniformly 15N, 13C labeled proteins. Couplings are obtained from a series of intensity modulated two-dimensional HN–N spectra equivalent to the common 1H–15N–HSQC spectra, alleviating many overlap and assignment issues associated with other techniques. To illustrate the efficiency of this method, H–C, C–C, and HN–N isotropic scalar couplings were determined for ubiquitin from data collected in less than 4.5 h, C–C data collection required 10 h. The resulting couplings were measured with an average error of ±0.06, ±0.05, ±0.04 and ±0.10 Hz, respectively. This study also shows H–C and C–C couplings, valuable because they provide orientation of bond vectors outside the peptide plane, can be measured in a uniform and precise way. Superior accuracy and precision to existing 3D measurements for C–C couplings and increased precision compared to IPAP measurements for HN–N couplings are demonstrated. Minor modifications allow for acquisition of modulated HN–C 2D spectra, which can yield additional well resolved peaks and significantly increase the number of measured RDCs for proteins with crowded 1H–15N resonances.  相似文献

The TROSY principle has been introduced into a HNCA experiment, which is designed for measurements of the intraresidual and sequential H-C/HN-N dipole/dipole and H-C/N dipole/CSA cross-correlated relaxation rates. In addition, the new experiment provides values of the 3,4 J H HN coupling constants measured in an E.COSY manner. The conformational restraints for the and angles are obtained through the use of the cross-correlated relaxation rates together with the Karplus-type dependencies of the coupling constants. Improved signal-to-noise is achieved through preservation of all coherence transfer pathways and application of the TROSY principle. The application of the [15N,13C]-DQ/ZQ-[15N,1H]-TROSY-E.COSY experiment to the 16 kDa apo-form of the E. coli Heme Chaperon protein CcmE is described. Overall good agreement is achieved between and angles measured with the new experiment and the average values determined from an ensemble of 20 NMR conformers.  相似文献

In this study we use a demographic approach to analyse the differing abundance of three congeneric columnar cacti: Neobuxbaumia macrocephala (the rarest), Neobuxbaumia tetetzo (intermediate), and Neobuxbaumia mezcalaensis (the most common). Populations of these species were studied in the Tehuacan Valley (Central Mexico) over a 3-year period. We employed traditional models and life table response experiments (LTRE) to explore the association between particular demographic traits and the degree of rarity of each species. Most matrices showed population growth rate () values close to unity; the only exception was N. mezcalaensis in 2001–2002 (=1.091±0.088). In the three species the highest elasticity values corresponded to entries referring to the stasis of pre-reproductive plant and small adults. However, the LTRE indicated that most important differences in the values between years and species were associated with variation in fruit production and seedling recruitment. N. mezcalaensis yielded the highest mean due to its high fecundity in 2001–2002. This result suggests that the variation in demographic behaviour observed between species is sufficient to explain the higher abundance of N. mezcalaensis compared to the rare N. macrocephala. The high fecundity values and recruitment success observed in the former, even if they occur only seldom, may result in higher average values accounting for its high population densities.  相似文献

Summary A genomic clone of a wheat -amylase gene (Amy3/33) was identified, on the basis of hybridisation properties, as different from -Amy1 and -Amy2 genes which had been characterised previously. The nucleotide sequence revealed that this gene has the normal sequence motifs of an active gene and an open reading frame interrupted by two introns. The protein sequence encoded by this open reading frame is recognisably similar to that of -amylase from the -Amy1 and -Amy2 genes and there is high sequence homology in all three proteins at the putative active sites and Ca++ binding region. In addition, the introns are at positions equivalent to the position of introns in the -Amy1 and -Amy2 genes. However, the sequence was less similar to -Amy1 and -Amy2 than these are to each other. Southern blot analysis showed that the Amy3/33 DNA is one of a small multigene family carried on a different chromosome (group 5) from either the -Amy1 or -Amy2 genes. A further difference from the -Amy1 and -Amy2 genes was the pattern of expression. Amy3/33 was expressed only in immature grains and, unlike the -Amy1 and -Amy2 genes, not at all in germinating aleurones. These data suggested therefore that this gene represents a third type of -amylase gene, not described before, which shares a common evolutionary ancestor with the -Amy1 and -Amy2 genes.  相似文献

The effect of interspecific competition and element additions (N and P) on four grassland species (Poa pratensis, Lolium perenne, Festuca valida, Taraxacum officinale) grown under field conditions was studied. Two grasses (L. perenne, F. valida) grown in monoculture (absence of competition) showed lower carbon isotope discrimination (13C) and enriched 15N values. Nitrogen addition (as urea) had inconsistent effects on species 13C while caused enrichment of 15N of P. pratensis and F. valida but strong depletion of 15N of T. officinale. Phosphorous had no significant effect on 13C but depleted 15N of all species.  相似文献

Summary Olfactometer tests with Asobara tabida (Nees 1834), a larval endo-parasitoid of frugivorous Drosophilidae showed that females are attracted to the odour of host food: a suspension of living yeast. This attraction decreased as the fermenting medium grew older and became less likely to contain suitable host stages. Olfactometer tests with — what was considered to be — A. tabida from two different microhabitats (fermenting fruits and decaying plants) showed a genetically determined difference in microhabitat odour preference between the two microhabitat strains. Each strain preferred the odour of its own microhabitat. This odour preference was not modified by larval conditioning. Hybridization tests indicated that we were dealing with two sibling species: A. tabida and A. rufescens (Foerster 1862), reproductively isolated by a pre-mating isolation mechanism only. Enforced matings resulted in fertile female offspring. Some small morphological differences were detected. The two species live sympatrically, although each inhabits and is most attracted to its own microhabitat.  相似文献

A recently described protocol for thein vitro production of ascomata was employed to determine the sexual incompatibility systems of five species ofCapronia. The formation of mature ascomata in isolates derived from single ascospores demonstrated thatC. epimyces, C. mansonii, andC. munkii n. sp. are homothallic. In contrast, fertile ascomata were observed only in mass-ascospore isolates and pairwise crosses between specific single-ascospore isolates inC. dactylotricha n. sp. andC. moravica. TheExophiala anamorphs ofC. dactylotricha andC. munkii are described and aPhialophora-like synanamorph is reported for the former species. Germinating ascospores ofC. munkii formed conidiogenous cells directly, while the ascospores of the remaining species germinated to produce germ tubes and hyphae. The application of the terms microcyclic conidiation to secondary conidium production and sclerotial body and stroma to the multicellular structures produced by species ofCapronia andExophiala are discussed.  相似文献

Summary The ability of adventitious and basal root systems of watercress (Rorippa nasturtium-aquaticum (L) Hayek) to absorb mineral nutrients from surrounding media has been demonstrated using radioisotopes 32P, 86Rb and 59Fe. Controlled experiments on single whole plants cultured in a dualmedium-apparatus, indicate that both root systems have a capacity for nutrient absorption. Analysis of axillary shoots formed during a seven day experimental period show that a greater proportion of phosphate and potassium, gained from the ambient media, was absorbed by the adventitious root system, although there was a greater mass of basal root tissue. The patterns of clomazone (2-[(2-chlorophenyl) methyl-4,4-dimethyl-3-isoxazolidinone) absorption, translocation, and metabolism and their contribution to the plant selectivity of this herbicide were studied in tolerant soybean [Glycine max (L.) Merr.] andAmaranthus hybridus and in susceptibleA. retroflexus andA. lividus. Differential root absorption appeared to play a significant role in the differential response of these four plant species to clomazone. Absorption of root-applied14C-clomazone was greater by the two sensitiveAmaranthus weeds than by the tolerant soybean andA. hybri-dus. Following application of14C-clomazone to roots, most of the absorbed radioactivity was translocated to the leaves of all four species. Approximately 50% of the absorbed14C-clomazone was metabolized by all four plant species as early as 12 h after treatment. Thin layer Chromatographic (TLC) analysis of plant tissue extracts from all four species revealed the formation of two major metabolites of clomazone. These unidentified metabolites had Rf values of 0.4 and 0.8, respectively, in a butanolacetic acidwater (1235, vol/vol/vol) developing system. The Rf value of unaltered clomazone in this system was 0.95. Doubled haploid (DH) progeny from a cross between the scald susceptible barley (Hordeum vulgare L.) cultivar Ingrid and the resistant accession CI 11549 (Nigrinudum) was evaluated for resistance in the pathogen Rhynchosporium secalis (Oudem) J.J. Davis. Two linked and incompletely dominant loci confer resistance CI 11549 against isolate 4004. One is an allele at the complex Rrs1 locus on chromosome 3H close to the centromere; the other is located 22 cM distally on the long arm. The latter locus is designated Rrs4. In BC3-lines into Ingrid from CI 2222 (another Nigrinudum) resistance seems governed by one locus close to the telomeric region of chromosome 7H, probably allelic to Rrs2. In neither case did we find any trace of the recessive gene rh8 reported to be present in Nigrinudum. Various resistance donors of Ethiopian origin designated as Nigrinudum, Jet or Abyssinian were identical to a great extent with respect to markers, but differed in resistance to different isolates of scald or in barley yellow dwarf virus (BYDV) resistance. The implications for their use as differentials in scald tests and screening of germplasm collections are discussed.  相似文献

The copepod Calanus chilensis is an endemic component of the zooplankton community in northern Chile. Size distributions of adult females, relative frequency of copepodid stages and relative numbers of adult males and nauplii, suggest the presence of at least 6 generations during the year, although the species seems to continuously reproduce through all seasons. Temperature profiles from 0 to 175 m indicated the presence of the El Niño current during March and April. Surface temperature ranged between 21.1 and 13.1 °C. Warm waters during the El Niño event seemed to affect the body size of adult females and the normal course of cohort development, although the population tended to recover rapidly through the subsequent months. We discuss the role of oceanographic conditions in controlling the life cycle of this species, as well as its continuous growth through the year compared to other Calanus species.  相似文献

Membrane preparations, capable of high rates of respiration-linked ATP synthesis, have been obtained from a gram-positive methylotrophic bacterium Bacillus sp. MGA3. NADH, succinate, reduced TMPD and methanol were shown to be suitable substrates for the oxidative phosphorylation. Esterification of orthophosphate was dependent on electron transfer, as evidenced by the requirement for both substrate and oxygen. Phosphorylation was also dependent on ADP and was destroyed by boiling the membrane preparation. The phosphorylation was markedly uncoupled by carbonyl cyanide p-(trichloromethoxy)-phenylhydrazone (CCCP) and was inhibited by N,N-dicyclohexylcarbodiimide (DCCD). KCN caused strong inhibition of substrate oxidation as well as phosphorylation for all substrates tested. Rotenone, amytal and antimycin A caused inhibition when NADH or methanol were used as substrates. Antimycin A inhibited respiration and ATP synthesis with succinate as substrate and had no effect on ascorbate —N,N,N,N-tetramethyl-p-phenylenediimide (TMPD) oxidation by membrane preparations of Bacillus sp. MGA3. P/O ratios determined were 2.4 with NADH, 1.7 with succinate and 0.8 with reduced TMPD. Summary Zooplankton and water samples were collected at weekly intervals between April 25 and May 30, 1986 in Barrow Strait, N.W.T. (Canadian Arctic Archipelago). In tows from 0–30 m, the zooplankton community (>202 m) was dominated by Pseudocalanus. The population was apparently growing and developing as shown by an increase in the proportion of adults (stage VI) and decreases in the proportion of stages III, IV, and V as the season progressed. Respiration and excretion rates of the Pseudocalanus populations were probably linked, there being an immediate increase in excretion rate, accompanying an increase in feeding rate when chlorophyll concentrations increased, which was followed by a smaller increase in respiration rate after a time lag. Hence, there was a large decrease in the ON ratio. Increased metabolism coincided with changes in the population structure, as did protease and bodily protein, but could not be clearly linked to dietary acclimation. Only laminarinase activity could be statistically related to an identifiable fraction of potential nutritional value in the water, particulate soluble carbohydrate, but neither showed overall seasonal change.  相似文献

This paper describes the application of a stochastic model developed to analyse the tolerance of individuals to moisture levels the intensity of which changes with time. The model measures the instantaneous rate of escape () that the individuals exhibit when facing desiccation. The abrupt changes of are detected by measuring maximum-likelihood ratio. This allows us to categorise a set of individuals according to increasing levels of tolerance to desiccation (low, high, and at least one intermediate level).The model was then applied to the case of a community of several species of oribatid mites subjected to slow and progressive desiccation of their environment. Seven categories of individuals were recognized. The first category consisted of individuals which left the soil before the water content fell to 29% while the last category comprised individuals who left the soil when the water content fell below 11%, i.e. after the disappearance of all traces of liquid water (pF>5). The model was also applied to three numerically dominant species within the biotope. This model should be applicable to future research on the adaptations of microarthropods to epigeaic life.  相似文献

We report here studies which characterize further the interaction ofN,N-dicyclohexylcarbodiimide with cytochromec oxidase leading to inhibition of H+ translocation by the enzyme. Further evidence is presented to show that the inhibition results from a real interaction of DCCD with the enzyme and cannot be accounted for by uncoupling and, contrary to recent criticisms, this interaction occurs specifically with subunit III of the enzyme even at relatively high inhibitor-to-enzyme stoichiometries. Summary Different LDH isoenzyme patterns are present in oocytes or unfertilized egg cells of different mammals. While in species of the orders Rodentia and Lagomorpha only LDH1 (-subunits) has been found, species of the order Carnivora and Artiodactyla show further LDH isoenzymes, indicating the participation of -subunits.
Zusammenfassung In Oocyten oder unbefruchteten Eizellen verschiedener Säuger finden sich verschiedene LDH-Isoenzymmuster. Während bei Species der Ordnungen Rodentia und Lagomorpha stets nur LDH1 (-Untereinheiten) gefunden wurden, zeigten Arten der Ordnungen Carnivora und Artiodactyla weitere LDH-Isoenzyme, die auf Beteiligung von -Untereinheiten schließen lassen.

The effects of uncouplers (FCCP, DNF), oligomycin, and rotenone on the fluorescence of potential-sensitive dyes, rhodamine 123 and diS-C3-(5), in lymphocyte suspensions were compared. The fluorescence of these optical probes gradually increased at higher FCCP concentrations. The dependences of fluorescence intensities and FCCP concentrations were similar for both dyes, and only diS-C3-(5) fluorescence started increasing at lower FCCP concentrations. Rotenone (1 µM) significantly increased rhodamine 123 fluorescence. TMPD-induced and uncoupler-induced diS-C3-(5) fluorescence changes increased 1.5- to 2-fold if the incubation mixture was supplemented with oligomycin (0.1–0.2 µg/ml). The fluorescence responses of the dyes in the lymphocyte suspension correlate with the effects of mitochondrial energetics inhibitors on m in isolated mitochondria. Colonization by midges, and temporal changes in their community structure, were examined in slow sand filter beds. The replicated beds allow the development of communities to be traced from a known starting point.The filter beds (rectangular concrete containers filled with water) have a substratum of sand on which a rich coating of organic particles develops during passage of the water through the bed. The containers (ponds) are drained from time to time and the organic layer is then scraped off the sand surface. This occurs on average, once a month. The length of time the ponds were filled with water (bed run) during the present study ranged from 16 to 77 days.In long bed runs small midges with a short aquatic phase (Cricotopus sylvestris, Psectrocladius limbatellus, Tanytarsus fimbriatus) produced adults after 16–20 days; other, larger midges,e.g. Psectrocladius barbimanus and the Tanypodinae required a longer aquatic phase. Of the Tanypodinae, the smallAblabesmyia phatta, had the shortest duration of the four species found, and was much the most numerous member of this subfamily. Some Chironomini only appeared when the organic coating had developed over the sand surface. Midges of this tribe frequently failed to complete their larval development within the duration of bed runs and were thus trapped on the substratum at the time of cleaning. When ponds were drained after short bed runs the succession in community structure observed in long runs was arrested.Three small midgesC. sylvestris, P. limbatellus andT. fimbriatus, were collected in high numbers throughout the life of all beds, except towards the end of the longest runs in the study. This suggests that small size, short life cycles, and the ability to colonize clean substrata, are important characteristics for the development of primary chironomid communities in short-lived temporary habitats.  相似文献
