Argopecten purpuratus is a bivalve filter feeder inhabiting protected areas of the Peruvian coastline and northern and central Chile. The species is commercially valuable and the natural stocks are over-exploited in Chile, mainly in the central region. Expansion of aquaculture farms to the geographically protected coastline of southern Chile is a potential solution to this problem. So, the objective of the present study was to determine the acute feeding and digestive response of A. purpuratus in relation to short acclimation periods (1, 3 and 7 days) and exposure to variable salinity (24 per thousand, 27 per thousand and 30 per thousand) with two different diets. The feeding behaviour of A. purpuratus follows that described for other species of bivalves whereby the decrease in salinity produces a reduction in the clearance and ingestion rates. This behaviour is not affected by the diet type or the 7 day acclimation period. Lower values of absorption efficiency are associated with increasing food concentration. The response of the AE at 24 per thousand salinity and low organic content diet indicates that the interaction of low salinity and low organic content diet reduces the AE to values approaching zero. The enzymatic investment of A. purpuratus at low salinity (24 per thousand) is similar for both diets and more reduced than for 30 per thousand and 27 per thousand salinity. The results show that osmotic stress leads to a reduction of the enzymatic response capacity. The multiple regression analysis indicates that different responses are observed as a function of the enzymatic activity and the organ. A reduction of the carbohydrase activities in the digestive gland was observed to low salinity (24 per thousand), specifically amylase and celullase. The results of this study are ecophysiologically relevant and highlight a relationship between digestive enzymatic activity and salinity, and may explain the possible differences in the energetic balance of A. purpuratus in response to changes in salinity.  相似文献

This study evaluates the effect of temperature, coupled with ingestion rate, on the dynamics of biochemical components and lipid classes in R. philippinarum. The data are discussed with regard to sexual development and energy balance. Experimental protocol developed in the present study used two groups of the clam R. philippinarum: L (temperatures of 14 degrees C and 18 degrees C) and H (temperatures of 18 degrees C and 22 degrees C). The intra-group ingestion level was similar, although the ingestion level of the clams in the group H was 2.4 times higher than group L. We observed that R. philippinarum conditioned at 18 degrees C (18L) shows higher protein content, furthermore an important loss of organic weight was observed after 48 days. In such a situation, the clams use their own reserves (carbohydrates and glycogen) for sexual development while in situations without food stress (positive energy balance) and low temperature (14 degrees C) an accumulation of reserves is produced. Strikingly dissimilar behaviour in biochemical composition was observed for the 18H and 22H treatments, both with a positive energy balance. Despite similar protein content, the highest levels of carbohydrates were observed at the lower temperature (18 degrees C). Glycogen was also higher for the 18 degrees C treatment, although the differences were significant only in the males. Although the total lipids in R. philippinarum showed no significant differences in any treatment, they became apparent and related to sex when considering the individual lipid classes. There was no variation in lipid classes in the males between the 14L and 22H treatments despite the large disparity in the degree of sexual development. However, in the females significant differences in lipid classes (phospholipids, triglycerides) were observed. The results of this study show that a positive energy balance permits R. philippinarum gonadal development and accumulation of reserves both in low and high temperature conditions. In low temperature situations, gonadal development is slower and the energy reserves are accumulated in the form of carbohydrates. When the clams are conditioned at high temperatures, gonadal development is fast and complete, carbohydrates are consumed and lipids are accumulated.  相似文献

Rainbow trout were fed for 10 weeks with either a carbohydrate-free diet (C-free) or with four experimental diets containing various levels (20 or 40%) and sources of starch (extruded wheat or peas) in order to examine metabolic utilisation of dietary vegetable carbohydrates and its endocrine control. The study was focused on the parameters described as limiting in glucose metabolism in fish. Feeding trials were conducted at 8 and 18 degrees C to establish whether carbohydrate-rich diets can be used in trout farming irrespective of water temperature. At both temperatures, pea diets (especially the highest level) resulted in a feed efficiency as high as the C-free diet. Fish had similar growth rates except when fed the low wheat content diet. Glycaemia values 6 h after feeding were significantly higher in trout fed carbohydrate diets than those given the C-free diet, whereas plasma insulin levels were similar independently of the levels of dietary starch. This study provides the first evidence that glucokinase (GK) activity and mRNA level in trout liver increase in proportion to the content of dietary starch. Nevertheless, these changes were not correlated with plasma insulin levels. Insulin-like growth factor-I (IGF-I) binding and number of receptors in skeletal muscle were consistently higher than those for insulin but no diet-induced differences were found for any of these parameters. Temperature clearly affected the postprandial profile of glucose and insulin, which both showed lower levels 6 h after feeding at 8 degrees C than at 18 degrees C, which was consistent with a lower feed intake. Glucose and insulin levels decreased markedly 24 h after feeding at 18 degrees C, while they were still high at 8 degrees C, an observation concordant with delayed transit rate. These findings indicate satisfactory adaptation of rainbow trout to diets with a relatively high vegetable starch content, especially when provided as extruded peas, and indicate that diets with increased levels of carbohydrates can be used in this species even when it is reared at low temperature.  相似文献

The influence of both the lipid composition of microalgal diets and commercial flours on the lipid classes and fatty acids of Ruditapes decussatus spat was studied. These aspects of the nutritional value of the diets were discussed in relation to the growth of the spat. Four diets were tested; Diet A, composed of 100% of the daily food ration of microalgae; Diet B, composed of 100% of wheatgerm; Diet C, composed of 50% of microalgae and 50% of wheatgerm; and Diet D, composed of 25% of microalgae and 75% of wheatgerm. The microalgal cells present a higher lipid content than that for wheatgerm. Tahitian Isochrysis cells have phospholipids and triacylglycerols as majority lipids, whereas in the wheatgerm particles, the lipids more abundant are triacylglycerols. Fatty acid content was higher in the microalgal cells than in the wheatgerm particles. The n-3 fatty acids were the most abundant acids in the microalgae, whereas the n-6 fatty acids were in the wheatgerm. The n-3 PUFA were not detected in wheatgerm. Phospholipids were the main lipids present in the clam spat, followed by triacylglycerols. Other lipid classes, detected in significantly lower amounts, included free fatty acids, sterols, and sterol ester + waxes. The composition of fatty acids in the spat was influenced by the fatty acid composition of the diet. Highest spat growth rates were observed with those diets that present a higher phospholipid/triacylglycerol relation. A negative correlation in the relation n-6/n-3 vs. growth has also been observed, with better growth rates in diets with a lower ratio. If the fatty acid 20:5n-3 and 22:6n-3 considered "essential" for marine animals were not present in the diet, they were not present in the spat either. Desaturation and elongation capabilities of R. decussatus spat were also discussed.  相似文献

Diets were computed to contain equal concentrations of digestible crude protein either of wheat gluten (diet 1) or of grieves (diets 2–8). Per kg dry diet, 41 g crystalline amino acids were supplemented. All diets contained at least 1.2 g Lys per MJ digestible energy (DE). In diet 2, ratios of Met + Cys, Trp, Leu, Ile and Phe to Lys were about equal to those in diet 1. In each of diets 3–7, one of the respective amino acids, in diet 8 all five were replaced by Glu in the supplemented mixture of amino acids.

Each diet was fed to triplciate groups of 20 trout during a trial lasting 66 days. Trout fed the diet containing wheat gluten consumed more dry matter and showed higher growth rates as well as higher protein contents in their gained body mass than trout fed diets based on grieves. Supplementing Met plus Trp significantly improved dry matter intake, growth rate and protein content of gain, though not to the level of trout fed the wheat gluten diet, whereas Leu, Ile and Phe showed no such effect. When grieves were not supplemented with both Met and Trp, gain in body mass contained significantly more lipids. DE required per kg gain by trout fed wheat gluten, grieves + Met + Trp or grieves without supplementation of Met and Trp was 20.1, 21.2 and 29.9 MJ, respectively.  相似文献

The aim of this study was to compare the effects of dietary oxidized cholesterol and pure cholesterol on plasma and very low density lipoprotein (VLDL) lipids and on some parameters of VLDL assembly and secretion in rats fed two different dietary fats. Four groups of male growing Sprague-Dawley rats were fed diets containing pure or oxidized cholesterol (5 g/kg diet) with either coconut oil or salmon oil as dietary fat (100 g/kg diet) for 35 days. Rats fed oxidized cholesterol supplemented diets had significantly lower concentrations of triglycerides and cholesterol in plasma and VLDL than rats fed pure cholesterol supplemented diets irrespective of the type of fat. In addition, rats fed oxidized cholesterol supplemented diets had significantly lower relative concentrations of microsomal triglyceride transfer protein messenger ribonucleic acid (mRNA) than rats fed pure cholesterol supplemented diets. In contrast, hepatic lipid concentrations and the relative concentration of apolipoprotein B mRNA were not influenced by the dietary factors investigated. Parameters of hepatic lipogenesis (relative mRNA concentration of sterol regulatory element binding protein-1c and activity of glucose-6-phosphat dehydrogenase) were significantly reduced by feeding fish oil compared to coconut oil, but were not affected by the type of cholesterol. In conclusion, the data of this study suggest, that dietary oxidized cholesterol affects VLDL assembly and/or secretion by reducing the synthesis of MTP but not by impairing hepatic lipogenesis or synthesis of apolipoprotein B.  相似文献

One-year-old bay scallops, Argopecten irradians irradians (58 ± 2 mm, 22 ± 1 g live weight) were exposed to four replicated photoperiod treatments (24D, 8L:16D, 16L:8D, and 24L where D = dark hours, L = light hours) in order to measure the effect on gonad weight and maturation during the conditioning process. Results indicated that day-lengths of more than 8 h are necessary to promote gonad maturation in bay scallops. After 6 wk, the mean gonad weight for scallops in the 16-h and 24-h light regimes was similar at 0.6 ± 0.1 g dry weight compared to a mean of 0.2 ± 0.1 g dry weight for those in the 8-h and 0-h light regimes. Histological assessment indicated significantly more follicular tissue development in both the male and female portion of the gonad in the two longer photoperiod treatments. Overall, gamete maturity was highest for the scallops in the 16-h light regime; the incidence of mature eggs was 50% compared to 35% in the 24-h light regime, 20% in the 8-h light regime and 10% in the 0-h light regime. Assessment of feeding rates indicated no significant difference in algal cell consumption among treatments. Total dry tissue weight doubled over the 6-wk conditioning trial with no significant differences among treatments. One-year-old bay scallops appear to be non-responsive to conditions suitable for gonad maturation (i.e. appropriate temperature and food levels) unless they receive more than 8 h of light exposure. This finding has important implications for northern hatcheries which typically condition broodstock indoors during the early spring.  相似文献

以紫扇贝DNA为模板,用已开发的147个海湾扇贝微卫星标记引物扩增,结果表明100个微卫星标记能成功扩增,其中有47个表现出多态性,等位基因数目从2到9不等.观测杂合度范围为0.128 0~1.000 0(平均0.660 4),期望杂合度范围为0.503 1~0.862 1(平均0.671 9),有6个位点偏离Hardy-Weinberg平衡(P<0.05).用7对引物分别对紫扇贝、海湾扇贝及其种间正反杂交F1各30个个体进行PCR扩增,发现它们可明确区分紫扇贝与海湾扇贝,且所检测60个杂交子代均同时含海湾扇贝与紫扇贝的相应种特异性条带,证明全部为种间杂交子代.将该7对引物的扩增产物克隆测序,发现这些位点在两种扇贝中的序列同源率为40.22%~91.95%,其中3个位点在紫扇贝中的扩增产物仍然含有微卫星.  相似文献

The effects of different protein, lipid and carbohydrate diets on growth and energy storage in tench, Tinca tinca L., were studied. Over a 2-month period fish were fed four different diets: control, protein-enriched, carbohydrate-enriched and lipid-enriched. The best growth rates were obtained with the control and protein-enriched diets; the carbohydrate diet produced the worst results (lowest specific growth rate, weight gain, nutritional index and hepatosomatic index). These results suggest that it is not advisable to reduce dietary fish protein below 35%, and that it is not possible to obtain a protein-sparing effect of either lipids or carbohydrates, at least in our experimental conditions. The high-protein diet resulted in the storage of energy excess as muscle proteins and hepatic glycogen. Tench fed the high-carbohydrate diet stored carbohydrates as muscle glycogen and reduced plasma triglycerides. Finally, both liver and muscle lipid content were in positive correlation to dietary lipid.  相似文献

Experiments on rats showed that inclusion of sucrose into the composition of diets with increased or normal carbohydrate content accelerated the synthesis in the liver of apoproteins of pre-beta-lipoproteins and their loading with endogenously formed lipids, promoted a more intensive secretion of lipoproteins of very low density into the blood and an increase in it of the triglyceride level. However, the rate of the lipid compensation with the protein of pre-beta-lipoproteins in the liver depends also on the qualitative ratio of the carbohydrate and fat components in the diet. In using the diet with sucrose with the physiological ratio of carbohydrates and fats (2:1) there was noted a lower load of apoproteins of pre-beta-lipoproteins with lipids, this leading to an increase of the latter in the liver.  相似文献

Our experiments were designed to test the hypotheses that dietary lipids can affect whole-animal physiological processes in a manner concordant with changes in the fluidity of cell membranes. We measured (1) the lipid composition of five tissues, (2) body temperatures selected in a thermal gradient (T(sel)), (3) the body temperature at which the righting reflex was lost (critical thermal minimal [CTMin]), and (4) resting metabolic rate (RMR) at three body temperatures in desert iguanas (Dipsosaurus dorsalis) fed diets enriched with either saturated or unsaturated fatty acids. The composition of lipids in tissues of the lizards generally reflected the lipids in their diets, but the particular classes and ratios of fatty acids varied among sampled organs, indicating the conservative nature of some tissues (e.g., brain) relative to others (e.g., depot fat). Lizards fed the diet enriched with saturated fatty acids selected warmer nighttime body temperatures than did lizards fed a diet enriched with unsaturated fatty acids. This difference is concordant with the hypothesis that the composition of dietary fats influences membrane fluidity and that ectotherms may compensate for such changes in fluidity by selecting different body temperatures. The CTMin of the two treatment groups was indistinguishable. This may reflect the conservatism of some tissues (e.g., brain) irrespective of diet treatment. The RMR of the saturated treatment group nearly doubled between 30 degrees and 40 degrees C. Here, some discrete membrane domains in the lizards fed the saturated diet may have been in a more-ordered phase at 30 degrees C and then transformed to a less-ordered phase at 40 degrees C. In contrast, the RMR of the unsaturated treatment group exhibited temperature independence in metabolic rate from 30 degrees to 40 degrees C. Perhaps the unsaturated diet resulted in membranes that developed a higher degree of disorder (i.e., a certain phase) at a lower temperature than were membranes of lizards fed the saturated diet. Our study demonstrates links between dietary fats and whole-animal physiology; however, the mechanistic basis of these links, and the general knowledge of lipid metabolism in squamate reptiles, remain poorly understood and warrant further study.  相似文献

Effects of dietary supplementation of orotic acid to a diet containing the casein protein were compared with diets containing egg protein, soy protein, or wheat gluten on lipid levels in the liver and serum and activities of ornithine carbamoyltransferase (OCT) and alanine aminotransferase in the serum of rats. We found that supplementation of orotic acid to each diet increased the contents of the liver total lipids, triacylglycerol, and phospholipids compared with those not supplemented. The contents of liver total lipids, triacylglycerol, cholesterol, and phospholipids in rats fed the casein diet were significantly higher than those of rats fed the other three diets when orotic acid was supplemented. The levels of triacylglycerol, cholesterol, and phospholipids in the serum of rats fed the casein diet were markedly decreased by addition of orotic acid. The supplementation of orotic acid significantly increased the activities of both serum OCT and alanine aminotransferase in rats fed the casein diet, but not in rats fed the other diets. In conclusion, liver lipid accumulation induced by dietary orotic acid depends on the type of dietary protein. The enhancement of serum OCT activity may result from liver lipid accumulation in rats fed the casein diet supplemented with orotic acid, demonstrating hepatic damage.  相似文献

Phytic acid was extracted from sweet potato (Ipomoea batatas) and fed to Wistar rats with or without zinc for 3 weeks. Animals were then sacrificed and bone and faecal minerals were assessed. The ultra-structure of the bones was examined via scanning electron microscopy. Phytic acid extract or commercial phytic acid supplemented diets (D + Zn + PE or D + PE) displayed reduced bone calcium levels (101.27 ± 59.11 and 119.27 ± 45.36 g/kg) compared to the other test groups. Similarly, reduced calcium were observed in the control groups (D + Zn and D) fed formulated diets with or without zinc supplementation (213.14 ± 15.31 and 210 ± 6.88 g/kg) compared to the other test groups. The group fed supplemented commercial phytic acid diet (D + CP) demonstrated the lowest femur magnesium (3.72 ± 0.13 g/kg) while the group fed phytic acid extract supplementation (D + PE) recorded the highest level (4.84 ± 0.26 g/kg) amongst the groups. Femur iron was highest in the group fed commercial phytic acid supplemented diet (D + CP −115.74 ± 2.41 g/kg) compared to the other groups. Faecal magnesium levels were significantly higher in the two test groups fed phytic acid extract with or without zinc (D + Zn + PE or D + PE) compared to all other groups. All the groups which had phytic acid supplemented diets had significantly thinner bone in the trabecular region, compared to the groups fed formulated diet or zinc supplemented formulated diet (D or D + Zn). These observations suggest that the consumption of foods high in phytic acid may contribute to a reduction in the minerals available for essential metabolic processes in rats.  相似文献

Although some external coloration and pigmentation patterns in molluscan shells may be attributable to environmental factors, most variation in these phenotypic characters depends on uncomplicated genetic mechanisms. Genetic research on inheritance of color variations in the north-Chilean scallop (Argopecten purpuratus) has now been expanded to analyze color segregation in juvenile scallops produced under controlled conditions employing self- and cross-fertilization. Calculations from the results were used for comparison with different numerical models based on Mendelian inheritance, and results were also obtained on the inheritance of a dorsoventral white line often observed on the left (upper) valve in this species. The results confirmed the hereditary basis for color variation in the shell of this scallop, suggesting a simple, dominant model of epistasis to explain the distribution of the different color variants observed (purple, brown, orange, yellow, and white). The presence of the white line may be controlled by a recessive allele with simple Mendelian traits on a locus distinct from those that control color variation.  相似文献

Nile tilapia were fed diets supplemented with one of the following lipid sources at 5% level: cod liver oil, corn oil, soybean oil, a coconut oil-based cooking oil or a combination of cod liver oil and corn oil (1 : 1). The control diet had no lipid supplement and tad fish meal as a sole protein source. A diet with soybean meal as a protein source was also tested. The number of females that spawned, spawning frequency, number of fry per spawning, and total fry production were increased at varying degrees by the supplemental lipid sources except for the cod liver oil. Fish fed the soybean oil diet tad the best overall reproductive performance over a 24-week period. Fish fed the cod liver oil diet had the highest weight gain but the poorest reproductive performance. The suplemental lipids significantly increased crude fat levels in the liver and ovaries. Both males and females Ld the cod liver oil diet had the highest levels of fat in the liver and muscle. The ratio of total n-6/n-3 fatty acid in the liver, ovaries and testes was influenced by the supplemental lipid sources. It was highest in fish fed either the soybean oil diet, the corn oil diet, or the soybean meal diet and lowest in fish fed the control diet or the cod liver oil diet.  相似文献

The use of chemically defined artificial diets has allowed researchers to examine questions within nutritional ecology about how macronutrients affect life‐history traits and resource‐based trade‐offs. Using a chemically defined diet, it is possible to manipulate both the total nutritional content and the ratio of macronutrients (i.e., proteins, carbohydrates, or lipids) within the diet. Studies using the geometric framework have made use of these diets to examine lifespan, fecundity, and immune responses. Here, we develop an artificial diet suitable for rearing lepidopteran larvae. We created diets with three proportions of non‐nutritive material (30, 50, and 70% indigestible cellulose) relative to protein and carbohydrate macronutrients, and compared these to standard wheat bran laboratory diet. We then examined the effects of variable nutrient content on lifespan and development time in Plodia interpunctella Hübner (Lepidoptera: Pyralidae). The artificial diets supported development (almost) as well as bran‐based laboratory diets. Total nutrient content affected development time: females that fed on the diet with the highest nutrient content took the longest time to reach eclosion. We also found evidence to support dietary restriction, with larvae receiving the fewest nutrients having the longest lifespan as adults. These findings are indicative of the usefulness of this diet as a tool to further investigate the effects of nutrient content and macronutrient imbalance on resource‐based trade‐offs and life‐history traits.  相似文献

Semimytilus algosus is a small mussel species that fouls artificial culture systems of the scallop Argopecten purpuratus (Lamarck, 1819) in the north of Chile. Since biofouling organisms are a serious problem in culture, competing with the scallops for food and oxygen, environmentally-friendly methods are required to mitigate the effects of this fouling in the culture systems. The present study reports the evaluation of the inhibitory effect of biofilms and extracellular products (EP) of the bacterium Alteromonas strain Ni1-LEM on the byssal formation of S. algosus juveniles. Laboratory bioassays were carried out to determine the reattachment, exploratory behaviour and/or byssal thread production of the mussel in plastic Petri dishes containing bacterial biofilms, different dilutions of EP, and EP incorporated in a test substratum. It was concluded from the results that culture supernatants of the Alteromonas tested had an inhibitory effect on reattachment by S. algosus.  相似文献

The effects of carbohydrate sources/complexity and rearing temperature on hepatic glucokinase (GK) and glucose-6-phosphatase (G6Pase) activities and gene expression were studied in gilthead sea bream juveniles. Two isonitrogenous (50% crude protein) and isolipidic (19% crude lipids) diets were formulated to contain 20% waxy maize starch or 20% glucose. Triplicate groups of fish (63.5 g initial body weight) were fed each diet to near satiation during four weeks at 18 degrees C or 25 degrees C. Growth, feed intake, feed efficiency and protein efficiency ratio, were higher at the higher water temperature. At each water temperatures fish growth and feed efficiency were higher with the glucose diet. Plasma glucose levels were not influenced by water temperature but were higher in fish fed the glucose diet. Hepatosomatic index and liver glycogen were higher at the lower water temperature and within each water temperature in fish fed the glucose diet. No effect of water temperature on enzymes activities was observed, except for hexokinase and GK which were higher at 25 degrees C. Hepatic hexokinase and pyruvate kinase activities were not influenced by diet composition, whereas glucose-6-phosphate dehydrogenase activity was higher in fish fed the glucose diet. Higher GK activity was observed in fish fed the glucose diet. GK gene expression was higher at 25 degrees C in fish fed the waxy maize starch diet while in fish fed the glucose diet, no temperature effect on GK gene expression was observed. Hepatic G6Pase activities and gene expression were neither influenced by dietary carbohydrates nor water temperature. Overall, our data suggest that in gilthead sea bream juveniles hepatocytes dietary carbohydrate source and temperature affect more intensively GK, the enzyme responsible for the first step of glucose uptake, than G6Pase the enzyme involved in the last step of glucose hepatic release.  相似文献

The Mediterranean fruit fly [Ceratitis capitata Wiedemann (Diptera: Tephritidae)], or medfly, is mass produced in many facilities throughout the world to supply sterile flies for sterile insect technique programs. Production of sterile males requires large amounts of larval and adult diets. Larval diets comprise the largest economic burdens in the mass production of sterile flies, and are one of the main areas where production costs could be reduced without affecting quality and efficacy. The present study investigated the effect of manipulating diet constituents on larval development and performance. Medfly larvae were reared on diets differing in the proportions of brewer's yeast and sucrose. We studied the effect of such diets on the ability of pupating larvae to accumulate protein and lipids, and on other developmental indicators. Except for diets with a very low proportion of brewer's yeast (e.g., 4%), pupation and adult emergence rates were in general high and satisfactory. The ability of pupating larvae to accumulate lipid reserves and proteins was significantly affected by the sucrose and yeast in the diet, and by the proportion of protein to carbohydrates (P/C). In contrast to previous nutritional studies conducted with other insects, low P/C in medfly larval diets (with excess dietary carbohydrates) resulted in pupating medfly larvae having a relatively reduced load of lipids; medfly larvae protein contents in these diets were, as expected, relatively low. Similarly, high P/C ratios in the diet produced larvae with high protein and lipid contents. Differences with other insects may be due to differential post‐ingestion regulation where a high dietary carbohydrate diet reduces the lipogenic activity of the larvae, and induces a shift from lipid to glucose oxidation. Larvae reared on low P/C diets spent more time foraging in the diet than larvae maintained on a high P/C diet, suggesting a compensatory mechanism to complement nutrient intake. The results suggest that the content of brewer's yeast, the most expensive diet component, could be fine‐tuned without apparently affecting fly quality.  相似文献
