Experiments were undertaken to determine molecular specificity of Vitamin E-effects on lifespan and fecundity in four bdelloid rotifers (Habrotrocha sp., Philodina sp., Pleuretra sp., and Rotaria sp.). Results indicate that lifespan and fecundity could be significantly increased by addition of any one of three tocopherol compounds (d--, -, and -tocopherol) to the rotifer medium. Life table functions were increased the most by the d--tocopherol form. Improvement of these life table functions was not achieved by substitution of tocopherol analogs or other antioxidants in the rotifer medium.  相似文献

Zusammenfassung Die Bildung von Pigmenten mit Redoxfunktion in Pseudomonas-Kulturen ist vom Eisengehalt des Nährbodens abhängig. In belüfteten Kulturen war die Menge der abgelagerten Farbstoffe stark herabgesetzt. Die bisher untersuchten Pigmente waren: Phenazin--carbonsäure aus Pseudomonas aureofaciens, Pseudoiodinin aus Ps. fluorescens var. pseudoiodinum und Pyocyanin aus Ps. aeruginosa.
Effect of iron and oxygen on the formation of pigments in some Pseudomonas spp.
Summary The formation of pigments having the function of redox catalysts in cultures of Pseudomonas spp. with gluconate as a sole carbon source was found to be related to the concentration of iron in the medium. In aerated cultures the accumulation of these pigments was extremely decreased.These pigments were: phenazine--carboxylic acid formed by Pseudomonas aureofaciens, pseudoiodinine formed by Ps. fluorescens var. pseudoiodinum, and pyocyanine formed by Ps. aeruginosa.

Summary A gene coding for amylase was cloned and sequenced from an alkalophilic Pseudomonas sp. KFCC 10818. The coding region for the amylase precursor contained 1,692 nucleotides. The presumed Shine-Dalgarno sequence, AAGG, was located at 8 nucleotides upstream from the ATG initiation codon. The precursor protein had a putative signal peptide of 25 amino acid residues at its amino terminus. The Pseudomonas amylase had four highly conserved regions as other -amylases. The amylase expressed from E. coli harboring the Pseudomonas gene produced maltose and maltotriose from soluble starch.The nucleotide sequence reported in this paper has been submitted in the GenBank/EMBL database with accession number(s) U40056.  相似文献

Summary Pseudomonas sp. strain T-12 transforms several substituted benzenes to catechols utilizing the two initial enzymes of the toluene degradative pathway, toluene-2,3-dioxygenase and toluene-2,3-dihydrodiol dehydrogenase. Several novel substrates for this catechol synthesizing system have been previously identified including cyclopropylbenzene, -methylstyrene, anisole, benzonitrile, ,,-trifluorotoluene, benzyl alcohol, 1-phenylethanol, 2-phenylethanol, p-difluorobenzene, and p-fluorobenzonitrile. The catechol products from these substrates are identified here as the 2,3-dihydroxy derivatives. Evidence is also presented which suggests that benzonitrile is metabolized like the halobenzenes and with 2,3-dihydroxybenzonitrile acting as a suicide substrate for catechol-2,3-dioxygenase. The scope and utility of Pseudomonas sp. strain T-12 catalyzed oxygenations is discussed.  相似文献

Nine different F2 families of peach [Prunus persica (L.) Batsch] were analyzed for linkage relationships between 14 morphological and two isozyme loci. Linkage was detected between weeping (We) and white flower (W), 33 cM; double flower (Dl) and pillar (Br), 10 cM; and flesh color (Y) and malate dehydrogenase (Mdh1), 26 cM. A leaf variant phenotypically distinct from the previously reported wavy-leaf (Wa) mutant in peach was found in progeny of Davie II. The new willow-leaf character (designated Wa2) was closely linked (0.4 cM) to a new dwarf phenotype (designated Dw3). Two families derived from the pollen-fertile cultivar White Glory segregated for pollen sterility, but segregation did not follow a 31 ratio. Evidence is presented suggesting that White Glory possesses a pollen-sterility gene (designated Ps2) that is non-allelic to the previously reported pollen-sterility gene (Ps) in peach. Ps2 was linked to both weeping (We-Ps2, 15.5 cM) and white flower (Ps2-W, 25.3 cM). A genomic map of peach containing 83 RAPD, one isozyme, and four morphological markers was generated using an F2 family obtained by selfing an NC174RL x Pillar F1. A total of 83 RAPD markers were assigned to 15 linkage groups. Various RAPD markers were linked to morphological traits. Bulked segregant analysis was used to identify RAPD markers flanking the red-leaf (Gr) and Mdh1 loci in the NC174RL x Pillar and Marsun x White Glory F2 families, respectively. Three markers flanking Mdh1 and ten markers flanking Gr were identified. The combination of RAPD markers and bulked segregant analysis provides an efficient method of identifying markers flanking traits of interest. Markers linked to traits that can only be scored late in development are potentially useful for marker-aided selection in trees. Seven specimens of rays of the genus Himantura which could not be identified to species were collected from waters near Dunwich, Stradbroke Island, Moreton Bay, Queensland, Australia. The five smallest specimens of Himanturasp. (disc width 218-302 mm; four female, one male) had a banded tail and the dorsal surface was uniformly grey/brown. The two largest individuals of Himantura sp. (disc widths 460, 533 mm; female and male, respectively) also had a banded tail but the grey/brown dorsal surface had white spots. Two new monogenean species (Monocotylidae: Monocotylinae) are described from both the plain and white-spotted specimens of Himantura. Dendromonocotyle lastin. sp. is distinguished from other species in the genus by the number of papillae on the haptor, by the morphology of the male copulatory organ and by the morphology of the proximal portion of the vagina. The muscular sheath which surrounds the male copulatory organ is also unique having sclerotised spines at the distal end. Dendromonocotyle species are skin parasites, but a total of five juvenile specimens of D. lasti were found on the gills of four rays. Monocotyle caseyae n. sp. from the gills is characterised by the morphology of the male copulatory organ and its accessory piece. One specimen of M. spiremae Measures, Beverley-Burton & Williams, 1990, originally described from the gills of Himantura fai Jordan & Seale off Heron Island, Great Barrier Reef, Queensland, Australia, was also found on the gills of one Himantura specimen. The site and host-specificity of the parasites and the identity of the hosts are discussed.  相似文献

Drought is an important abiotic stress that limits the plant growth and productivity. Present investigation was aimed that plant growth-promoting rhizobacteria (PGPR) isolated from moisture-stressed area impart drought tolerance in plants and tryptophan may improve their efficiency. Pseudomonas sp. (1), Bacillus cereus and Bacillus pumilus (B. pumilus) were isolated from maize rhizosphere grown in irrigated fields, semi-arid region and arid region, respectively. Proteus sp. and Pseudomonas sp. (2) were isolated from rice rhizosphere grown in irrigated fields and raised bed. B. pumilus produced 5× more abscisic acid (ABA) in culture media than Pseudomonas sp. (1) by the addition of l-tryptophan. These inoculants also modulated the phytohormone content of maize leaves in a pot experiment. Higher ABA was produced by the application of B. pumilus and Pseudomonas sp. (2), while indole 3-acetic acid and gibberellic acid were found higher in Pseudomonas sp. (1) and Proteus sp. treated plants. Addition of l-tryptophan increased the concentration of all phytohormones in soil and leaves of maize. Maximum increase in relative water content, osmotic potential, protein content and photosynthetic pigments was recorded in B. pumilus treated maize plants. Under irrigated condition, response of Pseudomonas sp. co-inoculated with B. pumilus from arid field superseded while under drought stress the effect of later predominated. Bacillus pumilus can be used in the formulation of biofertilizer to alleviate drought stress in arid and semi-arid regions.  相似文献

Summary Using disomic chromosome substitution lines based on the susceptible wheat cultivar Chinese Spring, loose smut resistance of wheat cultivars Hope and Thatcher was shown to be conferred in each case by a single dominant major gene carried on chromosome 7 A (Hope) or 7 B (Thatcher). Partial resistance was determined by genes on an additional eight Hope or seven Thatcher chromosomes, and similarities were evident between the partial resistance genotypes ofHope and Thatcher. Chinese Spring exhibited a mean infection value of approximately 50%, indicating a significant level of partial resistance, which was found to be due, in part, to genes on the homoeologous chromosome arms 1 As, 1 Es and 1 Ds, and to cytoplasmic genes. Substitution of the Chinese Spring nucleus into the cytoplasm of Aegilops squarrosa, Ae. variabilis or Ae. mutica resulted in increased susceptibility to Ustilago tritici. Several alloplasmic lines of the resistant wheat cultivars Selkirk and Chris exhibited race-specific susceptibility to U. tritici.  相似文献

The plant rhizosphere is a dynamic environment in which many parameters may influence the population structure, diversity and activity of the microbial community. Two important factors determining the structure of microbial community present in the vicinity of plant roots are plant species and soil type. In the present study we assessed the structure of microbial communities in response to four plant species (i.e. maize (Zea mays L.), oat (Avena sativa L.), barley (Hordeum vulgare L.) and commercial grass mix) planted in soil with different land use history (i.e. arable land under crop rotation, maize monoculture and permanent grassland). Both factors, plant species and land use history, showed clear effects on microbial community and diversity as determined by PCR-DGGE fingerprinting with universal and group-specific bacterial primers. Moreover, we explored the rhizosphere effect of these plant species on the abundance of bacterial antagonists of the potato pathogen Rhizoctonia solani AG3. The data showed that the abundance and taxonomic composition of antagonists differed clearly between the different plants. The highest percentages of antagonists were found in maize and grass rhizosphere. When antagonistic Pseudomonas populations were compared, the highest, abundance and diversity of antagonists were detected in barley and oat rhizospheres, as compared to maize and grass rhizosphere. The results obtained in our study demonstrate clearly that plant species and soil type are two important factors affecting the structure of total bacterial, Pseudomonas and Bacillus community.  相似文献

Summary From root exudates of three cultivars of chilli (Capsicum annuum L.) 12 amino acids and 7 sugars were detected. Methionine, d-1- phenylalanine, citrulline and d-xylose were detected only from the root exudates of resdistant cultivars. The root exudates of resistant variety inhibited spore germination of the pathogen (Fusarium oxysporum f. sp.capsici), but that of susceptible variety enhanced spore germiantion of the same. Spore germiantion of antagonistic fungi (Trichderma viride andAspergillus sydowi) was also influenced by the root exudates of resistant and susceptible varieties, but the influence was different.Spore germiantion of a number of rhizosphere fungi was studied and in general root exudate of susceptible cultivar enhanced spore germiantion of majority of fungi, but spore germination of antagonistic fungi against the pathogen was inhibited. However, root exudate of resistant cultivar stimulated spore germination of antagonistic fungi.  相似文献

Summary The probability of negative analysis of variance estimates of genetic variance components due to sampling error (Ps) was investigated. The objectives were to evaluate the magnitude of Ps, to compare Ps for estimates of A 2 and D 2 , and to compare Ps for genetic variance component estimates from the nested and factorial mating designs. Ps was defined in terms of ratios of mean squares and the F distribution was used to calculate probabilities of the negative estimates. The results indicated that Ps is often greater than 0.20 for D 2 . Internal motions of d-ribose selectively 2H-labeled at the 2 position were measured using solid state 2H NMR experiments. A sample of d-ribose-2 -d was prepared in a hydrated, non-crystalline state to eliminate effects of crystal-packing. Between temperatures of –74 and –60°C the C2–H2 bond was observed to undergo two kinds of motions which were similar to those of C2–H2/H2 found previously in crystalline deoxythymidine (Hiyama et al. (1989) J. Am. Chem. Soc., 111, 8609–8613): (1) Nanosecond motion of small angular displacement with an apparent activation energy of 3.6 ± 0.7 kcal mol–1, and (2) millisecond to microsecond motion of large amplitude with an apparent activation energy 4 kcal mol–1. At –74°C, the slow, large-amplitude motion was best characterized as a two-site jump with a correlation time on the millisecond time scale, whereas at –60°C it was diffusive on the microsecond time scale. The slow, large-amplitude motions of the C2–H2 bond are most likely from interconversions between C2-endo and C3-endo by way of the O4-endo conformation, whereas the fast, small-amplitude motions are probably librations of the C2–H2 bond within the C2-endo and C3-endo potential energy minima.  相似文献

Doubled haploid (DH) progeny from a cross between the scald susceptible barley (Hordeum vulgare L.) cultivar Ingrid and the resistant accession CI 11549 (Nigrinudum) was evaluated for resistance in the pathogen Rhynchosporium secalis (Oudem) J.J. Davis. Two linked and incompletely dominant loci confer resistance CI 11549 against isolate 4004. One is an allele at the complex Rrs1 locus on chromosome 3H close to the centromere; the other is located 22 cM distally on the long arm. The latter locus is designated Rrs4. In BC3-lines into Ingrid from CI 2222 (another Nigrinudum) resistance seems governed by one locus close to the telomeric region of chromosome 7H, probably allelic to Rrs2. In neither case did we find any trace of the recessive gene rh8 reported to be present in Nigrinudum. Various resistance donors of Ethiopian origin designated as Nigrinudum, Jet or Abyssinian were identical to a great extent with respect to markers, but differed in resistance to different isolates of scald or in barley yellow dwarf virus (BYDV) resistance. The implications for their use as differentials in scald tests and screening of germplasm collections are discussed.  相似文献

The relationship between bacterial cell volume and temperature was examined for field data collected over a 4-year period and through controlled chemostat incubations of aPseudomonas sp. Volumes of planktonic bacteria were found to decrease as water temperature increased. Changes in temperature accounted for 38% of the variation in average cell volume (P<0.001). Average planktobacterial cell volume fell 42% from 0.217m3 in mid-winter to 0.127m3 in mid-summer. Similar results were found for the size distribution of epibacterial cells. Controlled chemostat incubations of aPseudomonas sp. indicated that cell volume was significantly affected by temperature, growth rate, and the interaction of temperature and growth rate. The data suggest that a change in cell volume as a result of a change in temperature is an intrinsic property of planktonic bacteria.  相似文献

We found that -caprolactam is a new powerful inducer for the formation of Rhodococcus rhodochrous J1 nitrilase. When Rhodococcus rhodochrous J1 cells were cultivated at 28°C for 120 h in a nutrient medium supplemented with 0.5% (w/v) -caprolactam, an enormous amount of nitrilase was formed in the cells which corresponded to approximately 30% of all soluble protein. The level of -caprolactam in the culture broth barely decreased in the course of cultivation. -Butyrolactam and -valerolactam also caused effective induction. The induction of nitrilase formation by -caprolactam was also observed in some other Rhodococcus strains.  相似文献

Summary We studied changes in autolytic activity of cells in the course of mating, using heterothallic haploid strains of Saccharomyces cerevisiae. Autolytic activity was determined by measuring protein and sugar released in the medium. The autolytic activity increased very rapidly after mixing a and type haploid cells, while such a conspicuous change was not observed with separate cultures of a or type cells. Increase due to mating in release of sugar was more conspicuous than that of protein. Increase in autolytic activity preceded the appearance of conjugating cells.  相似文献

Summary The changes in some kinetic properties of pyruvate kinase from the mantle muscle and the hepatopancreas of Patella caerulea, over a time-course of 16 or 8 h of acute hypoxia were studied. Up to the second hour of anaerobiosis PK exhibited changes of the kinetic properties in the direction of activation, whereas in the late stages of anaerobiosis in both tissues examined these changes occurred in the direction of inactivation. Although PKs from these tissues seemed to be regulated in a similar pattern during the early stages of anaerobiosis, they exhibited different kinetic properties under both normoxic and hypoxic conditions. Two forms of PK were found to be present in both the mantle muscle and the hepatopancreas of P. caerulea with different kinetic properties. The underwater light field in the Bellingshausen andAdmundsen Seas was characterised using data collectedduring the R/V Polarstern cruise ANT XI/3, from12.1.94 to 27.3.94. The euphotic zone varied from 24to 100 m depth. Spectral diffuse vertical attenuationcoefficients (K d ())were determined for 12narrow wavebands as well as for photosyntheticallyavailable radiation (PAR, 400–700 nm): K d (490)ranged from 0.03 to 0.26 m1; K d (550) from0.04 to 0.17 m1; K d (683) from 0.04 to0.17 m1; and K d (PAR) varied from 0.02 to0.25 m1. K d () for wavelengths centred at412 nm, 443 nm, 465 nm, 490 nm, 510 nm, 520 nm and550 nm were significantly correlated with chlorophyllconcentration (ranging from 0.1 to 6 mg m3). Thevertical attenuation coefficients for 340 nm and380 nm ranged from 0.10 to 0.69 m1 and from 0.05to 0.34 m1, respectively, and were also highlycorrelated with chlorophyll concentrations. These K d values indicate that the 1% penetration depthmay reach maxima of 46 m and 92 m for 340 nm and380 nm, respectively. The spectral radiancereflectances (Rr()) for 443 nm, 510 nm and 550 nmwere less than 0.01 sr1. Rr() for 665 nm and683 nm increased with depth up to 0.2 sr1 because ofchlorophyll fluorescence. Using a model that predicts downwardirradiances by taking into account the attenuation bywater and absorption by chlorophyll, we show thatchlorophyll fluorescence has a significant influenceon the red downward irradiance (E d (633, 665, 683))in deeper layers. The ability of the phytoplanktonpopulation to influence the light environment byautofluorescence and absorption processes depends onthe light conditions and on the photoacclimation ofthe cells, represented by the in vivo crosssection absorption coefficient of chlorophyll (a*). Theobtained mean chlorophyll-specific light attenuationcoefficients of phytoplankton in situ (k d ) are higherthan the in vivo absorption coefficient of chlorophyll,more than to be excepted from the scattering. a*(), m2 mg chl1, decreased due topackaging effect with increasing chlorophyllconcentrations.  相似文献

Two novel extracellular cholesterol oxidases designated CO1 and CO2, from Bacillus sp. SFF34, were purified 5.6 and 5.9-fold giving Mr values of 36 and 37 kDa. The optimum temperature for the activity was 60 °C (CO1) and 40 °C (CO2), and the optimum pH was 6.25 (CO1) and 6 (CO2) over 30 min reaction time. The apparent K m values for cholesterol were 6.76 mM (CO1) and 4.50 mM (CO2). Both the enzymes could oxidize 5-cholestane, 5-cholestane-3-ol-7-one, coprostane, dihydrocholesterol, hecogenin, -sitosterol and stigmasterol.  相似文献

Strain B51 capable of degrading polychlorinated biphenyls (PCB) was isolated from soil contaminated with wastes from the chemical industry. Based on its morphological and chemotaxonomic characteristics, the strain was identified as a Microbacterium sp. Experiments with washed cells showed that strain B51 is able to degrade ortho- and para-substituted mono-, di-, and trichlorinated biphenyls (MCB, DCB, and TCB, respectively). Unlike the known PCB degraders, Microbacterium sp. B51 is able to oxidize the ortho-chlorinated ring of 2,2-DCB and 2,4-DCB and the para-chlorinated ring of 4.4-DCB. The degradation of 2,4-DCB and 4,4-DCB was associated with the accumulation of 4-chlorobenzoic acid (4-CBA) in the medium in amounts comprising 80–90% of the theoretical yield. The strain was able to utilize 2-MCB, 2,2-DCB, and their intermediate 2-CBA and to oxidize the mono(ortho)-chlorinated ring of 2,4,2-TCB and the di(ortho-para)-chlorinated ring of 2,4,4-TCB. A mixed culture of Microbacterium sp. B51 and the 4-CBA-degrading bacterium Arthrobacter sp. H5 was found to grow well on 1 g/l 2,4-DCB as the sole source of carbon and energy.  相似文献
