Development rates were determined for three pteromalid parasitoids of houseflies under constant and varying temperatures from 15 to 35°C.Muscidifurax raptorGirault and Sanders was the fastest developing species, with females completing development in 13.8 days at 32.5°C and 66.5 days at 15°C.Spalangia geminaBoucek females completed development in 20.8 days at 30.0°C and 161 days at 15.0°C, whereasS. cameroniPerkins females completed development in 20.6 days at 30.0°C and 155.5 days at 15.0°C. Male development times were 90.3% of those for femaleS. geminaand 92.7 and 88.6% of those for femaleS. cameroniandM. raptor,respectively. Parasitoid survival was very low at 35°C for all species and noSpalangiasurvived constant exposure to 15.0°C. Exposure to these lethal temperatures for shorter periods indicated that the parasitoids can tolerate them well under conditions more typical of the field. Development rates were modeled using biophysical and degree-day models and the models were tested for their ability to predict development under fluctuating conditions (24–36°C). Neither model was superior for all three species because of interspecific differences in the parasitoids' responses to high temperatures. Agreement between predicted and observed development times for all three species was achieved by small empirical adjustments of a key parameter in the biophysical model.  相似文献

Food concentration (0.5 × 103 – 5 × 105 Scenedesmus cells m1–1) significant influenced the somatic growth, maturation and survivorship ofS. proboscideus larvae. A density of 5 × 104 cells m1–1 was optimal for early larval stages. Of four temperature tested (20–35 °C), 30 °C resulted in the best growth and survival. Maturation time was inversely related to temperature, and was size- rather that age-dependent. Larvae were tolerant of a wide conductivity range, but optimal growth and survival were observed at 260 µS cm–1. Nitrate-Nitrogen (NO2-N) caused a larval mortality of 50% after 24 h at 0.58 mg1–1.  相似文献

Three micro algae, Ankistrodesmus convolutus, Scenedesmus incrassatulus and Chlorella vulgaris, at three concentrations, were tested as diets for Moina macrocopa. Their effect on reproduction, fecundity, growth and survival was evaluated. All three algae satisfied the nutritional requirements of M. macrocopa, despite their difference in size. The best concentration, expressed in dry weight, was 5 mg l–1 for all three. Time to first reproduction was 4 days with all diets. Average time between clutches was 33 hours, and maximum number of clutches was 12; the highest average number of offspring per brood was 27. The largest broods were the fourth to sixth, depending on the diet. Ephippium hatching depends on temperature, taking only 24 hours at 30 °C and 48 hours at 27 °C. This species could be an important test organism in aquatic bioassays, and a live food in aquaculture.  相似文献

The red alga Stictosiphonia hookeri is epilithic in shaded habitats of the upper intertidal zone from 30 to 55° S. Thalli of this species from Argentina, Chile, South Africa and Australia, usually without reproductive structures when collected, all developed tetrasporangia in culture. Although good vegetative growth occurred in all nine isolates at 20–25 °C, 12:12 light: dark cycle, 10–30 µmol photons m–2 s–1, none reproduced in these conditions except one isolate from Australia. At 15 °C the four South African (34 °S) isolates developed tetrasporangial stichidia, and three completed a Polysiphonia-type life history. Gametophytes were unisexual or bisexual. At 15 °C one isolate from Chile (36 °S) formed tetrasporangia, but sporelings were not viable. At 10 °C isolates from Argentina and Chile (53 °S and 54 °S) formed tetrasporangia; however, only the Chile isolate completed a Polysiphonia-type life history with unisexual gametophytes. The temperature required to induce sporogenesis correlates with the range of water and air temperatures in the natural habitats of each isolate. In irradiances >50 µmol m–2 s–1 the thalli became yellow- brown within two weeks because of phycobiliprotein loss, but this did not impair growth or reproduction. The Argentina and Chile isolates were resistant to freezing in seawater for at least two days, showing no cell damage. The protein cuticle of the outer cell wall is repeatedly shed in culture. This may serve to minimize the attachment of epiphytes in the field.  相似文献

The KT 2 strain of thermophilic spore-forming bacteria was isolated from a biofilm on the surface of a corroded pipeline in an extremely deep well (4680 m, 40–72°C) in the Urals. The cells are rod-shaped, motile, gram-variable. They grow on a complex medium with tryptone and yeast extract and on a synthetic medium with glucose and mineral salts without additional growth factors. The cells use a wide range of organic substances as carbon and energy sources. They exhibit a respiratory metabolism but are also capable of anaerobic growth on a nitrate-containing medium. Growth occurs within the 40–75°C temperature range (with an optimum of 65°C) and at pH 5–9. The minimum generation time (15 min) was observed at pH 7.5. Ammonium salts, nitrates, and arginine are used as nitrogen sources. The G+C content of the DNA is 54.5 mol %. From the morphological, physiological, and biochemical properties and the nucleotide sequence of the 16S rRNA gene, it was concluded that the isolate KT 2 represents a new species of the genus Geobacillus, Geobacillus uralicus.  相似文献

The intrinsic rate of natural increase(rm) is useful to estimate the populationgrowth potential of insects and mites, whichmay help predict the outcome of pest-naturalenemy interactions. This study was conductedto determine how 12 constant temperatureregimes between 10–38 °C (± 0.5 °C) may differentially affect therm of the McDaniel spider mite, Tetranychus mcdanieli McGregor (Acarina: Tetranychidea), a common pest of raspberry, andits coccinellid predator, Stethoruspunctillum Weise (Coleoptera: Coccinellidae). Tetranychus mcdanieli survived tomaturity in the 14–36 °C range, comparedto the 14–32 °C range for S.punctillum. Survival above 24 °Cremained high for the spider mite, butdecreased markedly for the coccinellid. Tetranychus mcdanieli's range forreproduction was similar to its survival range,but S. punctillum failed to reproduce at14 °C and reproduced only poorlyat 16 °C. Offspring production peakedat 24 °C for both T. mcdanieli(average 152 eggs per female), and S.punctillum (280 eggs per female). At alltemperatures suitable for reproduction, femalelongevity of the coccinellid was greater thanthe spider mite, which was characterized byearlier/faster reproduction than thecoccinellid. As temperature increased, therm followed a typical asymmetricaldome-shape pattern, with maximum values of0.196 d–1 and 0.385 d–1at 30 °C and 34 °C, for S.punctillum and T. mcdanieli,respectively. For each species, therm-temperature relation was successfullymodelled using a curvilinear regressionequation previously shown to predictdevelopment rate. In both species, thedevelopment rate response to temperature has amajor influence on the temperature-rmrelationship. In the 16–32 °C rangesuitable for population growth of both species,the rm of T. mcdanieliwas 1.9 (30 °C) to 8 (16 °C) times greaterthan S. punctillum. These growthpotential ratios are consistently in favor ofthe prey, suggesting a limitation of thecoccinellid with respect to its capacity totrack T. mcdanieli populations. However,under short season conditions, the inferiorreproductive dynamics of S. punctillum'svs. spider mite prey should not have aprevailing influence in determining impact, andmay be compensated by high voracity incombination with a strong aggregativeresponse.  相似文献

Synchaeta kitina Rousselet reached population densities of up to 5000 ind.l–1 in Loch Leven, between January 1977 and December 1982. The species was found over the entire range of temperatures recorded (0.4–21.4 °C), but was most abundant at temperatures above 7 °C. Embryo development times, determined under laboratory conditions, ranged from 122 h at 2 °C to 12.5 h at 20 °C. There was a marked inverse relationship between populations of S. kitina and Daphnia hyalina var. lacustris in the loch. It seems unlikely that this was due to interference competition from Daphnia. S. kitina can be cultured on Rhodomonas minuta var. lacustris and there is some evidence that this rotifer also feeds on small flagellates in its natural environment.  相似文献

A prolonged incubation of Escherichia, Salmonella or Pseudomonas at 48°C with nalidixic acid selected mutants (T48) able to grow at 48°C. A prolonged incubation at 54°C of the T48 mutants selected mutants (T54) able to grow at 54°C. These mutants were susceptible to the same bacteriophages as the original mesophilic strains. Auxotrophic phenotypes of Escherichia coli and Salmonella typhimurium mesophilic parents were demonstrated by these mutants if they were cultivated on minimal agar with cellobiose at 48°C or 54°C or on a minimal agar with glucose at 37°C. The T48 alleles mapped in the gyrA region of E. coli or S. typhimurium chromosome. In S. typhimurium the T54 alleles, which permit growth at 54°C, were shown by cotransductional analysis to be linked to gyrA.  相似文献

Effects of temperature on the activity of flucycloxuron on larval stages of Panonychus ulmi (Koch), based on LC50 values, were highly significant (P < 0.001) with temperature coefficients of-1.7 in both the ranges of 15° to 25°C and 20° to 30°C. The slopes of probit regression lines at 15° and 20°C were significantly steeper than those at 25° and 30°C. As a consequence the temperature coefficients based on LC90 values were-4.4 and-2.2, for the 2 temperature ranges. The ovicidal activity of flucycloxuron on P. ulmi was low and was only statistically detectable at 20°C (LC90 of 84 mg a.i./l). In studies with larvae of Aedes aegypti (Linnaeus), Leptinotarsa decemlineata (Say), Plutella xylostella (Linnaeus), Spodeptera exigua (Hübner) and Spodoptera littoralis (Boisduval) probit regression lines were parallel over temperature. The activity of flucycloxuron on these five insect species was not affected by temperature. Based on LC50 values, diflubenzuron showed positive temperature coefficients on P. xylostella of + 2.1 at 15° to 25°C and + 2.5 at 20° to 30°C. For S. littoralis the temperature coefficient was positive (+ 2.4) at 15° to 25°C but negative (-1.9) at the 20° to 30°C range. Temperature coefficients of diflubenzuron were neutral for A. aegypti, L. decemlineata and S. exigua. In the design and analysis of these studies special allowance was made for date effects and variation in natural mortality over temperature.  相似文献

The survival kinetics of two clinical isolates of Salmonella senftenberg were studied after heat and acid stress. The strains survived better at 53 and 55 °C after heat shock of 30 min at 50 °C or overnight heat adaptation at 45 °C. An increase in the decimal reduction time, D, of heat-shocked [10.2 min (53 °C) and 9.37 min (55 °C)] and heat-adapted [8.12 min (53 °C) and 7.8 min (55 °C)] cells was observed compared with the non-stressed cells [6.87 min (53 °C), 6.56 min 55 °C)]. A significant difference was also observed in the survival of acid-adapted to acid non-adapted S. senftenberg bacteria.  相似文献

The production of unicellular algae is laborious and is a major constraint for the culturing of aquatic filter-feeders. Because of their small particle size and their high protein content yeasts are considered as a promising substitute for micro-algae. Furthermore, recent work has shown that baker's yeast can be converted into a digestible diet for Artemia by chemical treatment. The present study documents the use at laboratory scale of this manipulated yeast as an algal substitute for the culture of two anostracan species. The experiments were conducted with the brine shrimp artemia franciscana and the fairy shrimp Streptocephalus proboscideus. A similar experimental set-up was used for both species. The algal diet, consisting of Dunaliella tertiolecta for A. franciscana and Selenastrum capricornutum for S. proboscideus, was substituted at various levels by two types of treated baker's yeast: a fresh form and a dried product which was rich in highly unsaturated fatty acids (HUFA). The chemically-treated yeast offers promising possibilities as an algal substitute for Artemia; i.e. replacing 75% of the algae by the dried yeast resulted in similar survival and even higher growth rates in comparison with the reference algal diet; for the treated fresh yeast similar results could be achieved by up to 95% substitution. For S. proboscideus, a substitution of 75% by either of the yeast products resulted in good survival, though growth did not exceed 80% of the observed growth in the algal control. A diet consisting solely of yeast resulted in poor survival for larvae of both species. Experiments were run to investigate whether this was due to a sub-optimal feeding regime, nutritional deficiencies, or deterioration of the water quality.  相似文献

The combined effects of host age and rearing temperature upon the encapsulation of eggs of Metaphycus swirskii Annecke & Mynhardt (Hymenoptera: Encyrtidae) by the hemispherical scale, Saissetia coffeae (Walker) (Homoptera: Coccidae), were determined. Encapsulation incidence at 16, 20, 24 and 28°C increased with the age of the host, being 5.5–19.6% in 3rd instar nymphs, 45.4–74.2% in young female scales and 88.8–100% in preovipositing female scales. Within each host physiological age range, encapsulation frequency was not materially influenced by most of the temperatures. At 32°C, regardless of host age, S. coffeae encapsulated 99.5 to 100% of parasitoid eggs. Complete immunity to successful parasitism by M. swirskii occurred at 20, 24, 28 and 32°C in preovipositing female scales and at 32°C in 3rd-instar nymphs and young female scales of S. coffeae.
Résumé Cette étude a déterminé les effets combinés de l'âge de l'hôte et de la température sur l'encapsulation des oeufs de M. swirskii Annecke & Mynhardt (Hym. Encyrtidae) par S. coffeae Walker (Homopt. Coccidae). La fréquence d'encapsulation à 16, 20, 24 et 28°C a augmenté avec l'âge de l'hôte, 5,5–19,6% dans les larves de 3ème stade, 45,4–74,2% chez les jeunes femelles et 88,8–100% chez les femelles avant la ponte. Dans chaque gamme d'âge physiologique de l'hôte, la fréquence d'encapsulation n'est pas influencée par la plupart des températures. A 32°C, quel que soit l'âge de l'hôte, S. coffeae a encapsulé 99,5–100% des oeufs du parasitoïde. Une immunité complète au parasitisme par M. swirskii a été obtenue à 20, 24, 28 et 32°C pour les femelles de cochenilles dans la période précédant la ponte, et à 32°C pour les larves de 3ème stade et les jeunes femelles de S. coffeae.

We fed prairie voles (Microtus ochrogaster) rat chow diluted with variable amounts of -cellulose to determine 1) how much fiber the voles could tolerate in their diet; 2) changes in food intake and digestibility of dry matter and of fiber; 3) the extent to which voles utilized fiber as an energy source; and 4) whether any of these variables differed between groups of animals maintained at 5 or 22°C. Fiber content of the diets ranged from 20 to 84%. Animals held at 5°C maintained body mass through a diet containing 69% fiber, while animals held at 22°C maintained body mass through the 84% fiber diet. Dry matter intake increased with fiber level from 9.3 to 15.0 g·day-1 for animals at 5°C and from 5.6 to 14.0 g·day-1 for animals at 22°C; intake on the highest fiber diet eaten by either group was not different. Dry matter digestibility decreased significantly as the fiber in the diets increased, but was not affected by temperature treatments. Digestible dry matter intake for each group remained constant regardless of diet quality, but on each diet digestible dry matter intake for animals at 5°C was significantly higher than that of the animals held at 22°C. Digestibility of the fiber portion of the experimental diets remained constant as food quality decreased, so the percent of daily energy need met by fiber utilization increased with higher food intake. We cultured Streptocephalus proboscideus using rice bran as a food source, and compared its growth, mortality, fertility, and cyst hatching with published data on animals fed other food sources. We also analysed the fatty acid profile of animals and cysts. The performance of rice-bran fed shrimp was indistinguishable from that of shrimp fed most other formulated diets, but there are indications that some previously applied diets were either defective in essential fatty acids, or toxic.  相似文献

Preimaginal development, mortality, aphid consumption rate, and size and weight upon reaching the adult stage of the aphidophagous coccinellids Hippodamia convergens Guérin-Méneville and Coccinella septempunctata L. collected from Karditsa, central Greece, were examined at four constant temperatures (14, 17, 20 and 23 °C) and L16:D8. The coccinellids fed on the tobacco aphid, Myzus persicae nicotianae Blackman. Egg, larval and pupal mortality was highest at 14 °C reaching 85.0, 73.8 and 29.4% in H. convergens and 49.3, 75.4 and 58.8% in C. septempunctata, respectively. Total preimaginal development ranged from 57.2 to 70.4 days at 14 °C, and to 16.9 and 22.1 days at 23 °C in H. convergens and C. septempunctata, respectively. Heavier and larger adults of H. convergens were obtained at 17 and 20 ° C. In C. septempunctata temperature did not affect adult weight while the lowest size was observed at 14 and 17 °C. Day-degrees requirements for preimaginal development in H. corvengens were 212.9 above a developmental threshold of 11.0 °C. The corresponding values for C. septempunctata were 281.5 and 10.7 °C. In H. convergens total and daily aphid consumption ranged from 46.8 aphids at 14 °C to 85.0 aphids at 23 °C and from 1.5 aphids at 14 °C to 9.2 aphids at 23 °C, respectively. The corresponding values for C. septempunctata were 112.0 and 2.7 at 14 °C and 157.7 and 12.4 at 23 °C. The results show the high potential of both predators as biological control agents against the tobacco aphid. The knowledge obtained could be essential for their appropriate use and for the improvement of mass rearing systems.  相似文献

Two micronized waste products were evaluated in a closed recirculation system for their suitability to replace a costly die of live algae in the culture of the Sudanese fairy shrimp,Streptocephalus proboscideus. The test population was kept at a density of 50 individuals 1–1 (sex ratio:1/1). An agricultural waste product (YM20; mixture of pea and corn), and an industrial waste product (POME; Palm Oil Mill Effluent) were fed at two regimes: 0.1 and 0.2mg DW animal–1 h–1. The microalgaSelenastrum capricornutum, used as a reference diet at a density of 2.0±0.82 × 105 cells ml–1, proved adequate in preliminary screening experiments. The effect of the diets and feeding regimes on selected biological variables and water quality were followed by weekly observations and measurements. Results in terms of growth (=increase in length), cyst production, and mortality were more successful when animals were supplied high densities of YM20 than in all other treatments: mean brood size was 155±6 cysts with a maximum of 266. Length after 6 weeks about 2 cm while this ranged between1.4–1.7 cm for the other treatments. Weekly mortality rate was comparable under high food conditions. Mortality rate gradually increased from 5% in the first week to 15% in the last week. Water quality, especially nitriate concentration (measured as NO2-N), was slightly better in the dry food fed than in the algae fed cultures. Present results are promising for large-scale culturing ofS. proboscideus in a cost-effective way by making use of agro-industrial waste products.  相似文献

Salmonella typhimurium is a major foodborne microbial pathogen which primarily contaminates poultry products causing salmonellosis in humans. S. typhimurium LT2 cultures, when transferred from 37 °C to 5 °C or 10 °C, showed an initial lag period in growth with an approximate generation time of 10–25 h. Western blot assay using E. coli CS7.4 antibody and analysis of radiolabeled total cellular proteins from S. typhimurium cultures after exposure to 10 °C or 5 °C showed elevated expression of a major cold shock protein, CS7.4. Identification of a decreased level of CS7.4 at 37 °C suggests that the expression of this protein may require a large temperature downshift. Putative regulatory protein binding segment on the 5-untranslated region referred as Fragment 7 in S. typhimurium exhibited a 90.6% and a 56.25% nucleotide sequence identity when compared with the Fragment 7 of E. coli and S. enteritidis, respectively. The differences in the nucleotide sequence within the Fragment 7 between S. typhimurium and S. enteritidis may explain the differential expression of CspA at 37 °C. The nucleotide sequence of the open reading frame of S. typhimurium cspA gene showed a single base difference at 816 bp position from a G to a C which altered the amino acid residue from a glycine to an alanine. In addition to CspA, an elevated expression of a 105 kDa, and decreased expression of 6 proteins were evidenced when cultures of S. typhimurium were exposed to 10 °C or 5 °C. Differential expression of the CspA and other proteins in S. typhimurium following exposure to cold temperatures suggest that adaptation and continued growth and survival at cold temperatures in this pathogen may be aided by these cold-responsive proteins.  相似文献

Several hot springs in the Rotorua-Taupo regions, North Island, New Zealand, were tested for the presence of extremely thermophilic acidophilic bacteria. In the majority of the springs, ranging in temperature from 43–96°C and in pH from 2.1–6.9, direct microscopic observations revealed the presence of both rod-shaped and spherical bacteria. Isolations were attempted at 70°C and pH 2.0 and 7.0, with either yeast extract for heterotrophic growth, or elemental sulfur as the sole source of energy for autotrophic growth. Eight of the samples produced grwoth at pH 2.0 with either yeast extract or sulfur, but none of the samples grew at pH 7.0. All the isolates obtained, resembled Sulfolobus acidocaldarius, a thermophilic acidophilic bacterium which has previously been reported from various regions in the Northern Hemisphere. Immunofluorescence examination of six of these isolates revealed varying degrees of cross reactions with two already characterized Sulfolobus isolates from the Yellowstone National Park, U.S.A. This paper is the first published record of Sulfolobus from the Southern Hemisphere.  相似文献

Plants of Solanum tuberosum L. potato do not cold acclimate when exposed to low temperature such as 5°C, day/night. When ABA (45 M) was added to the culture medium, stem-cultured plantlets of S. tuberosum, cv. Red Pontiac, either grown at 20°C/15°C, day/night, or at 5°C, increased in cold hardiness from –2°C (killing temperature) to –4.5°C. The increase in cold hardiness could be inhibited in both temperature regimes if cycloheximide (70 M) was added to the culture medium at the inception of ABA treatment. Cycloheximide did not inhibit cold hardiness development, however, when it was added to the culture medium 3 days after ABA treatment.When pot-grown plants were foliar sprayed with mefluidide (50 M), ABA content increased from 10 nmol to 30 nmol g–1 dry weight and plants increased in cold hardiness from –2°C to about –3.5°C. The increases in free ABA and cold hardiness occurred only in plants grown at 20°C/15°C; neither ABA nor cold hardiness increased in plants grown at 5°C.The results suggest that an increase in ABA and a subsequent de novo synthesis of proteins are required for the development of cold hardiness in S. tuberosum regardless of temperature regime, and that the inability to synthesize ABA at low temperature, rather than protein synthesis, appears to be the reason why S. tuberosum does not cold acclimate.  相似文献
