Sandy sediment beaches covering 70% of non-ice-covered coastlines are important ecosystems for nutrient cycling along the land-ocean continuum. Subterranean estuaries (STEs), where groundwater and seawater meet, are hotspots for biogeochemical cycling within sandy beaches. The STE microbial community facilitates biogeochemical reactions, determining the fate of nutrients, including nitrogen (N), supplied by groundwater. Nitrification influences the fate of N, oxidising reduced dissolved inorganic nitrogen (DIN), making it available for N removal. We used metabarcoding of 16S rRNA genes and quantitative PCR (qPCR) of ammonia monooxygenase (amoA) genes to characterise spatial and temporal variation in STE microbial community structure and nitrifying organisms. We examined nitrifier diversity, distribution and abundance to determine how geochemical measurements influenced their distribution in STEs. Sediment microbial communities varied with depth (p-value = 0.001) and followed geochemical gradients in dissolved oxygen (DO), salinity, pH, dissolved inorganic carbon and DIN. Genetic potential for nitrification in the STE was evidenced by qPCR quantification of amoA genes. Ammonia oxidiser abundance was best explained by DIN, DO and pH. Our results suggest that geochemical gradients are tightly linked to STE community composition and nitrifier abundance, which are important to determine the fate and transport of groundwater-derived nutrients to coastal waters.  相似文献

A series of laboratory-based and field experiments was conducted to address the effects of sunlight-exposed resuspended sediments on dissolved nutrient fluxes in two different water bodies. In suspensions of tidal creek sediments in 0.2 μm-filtered creek water, measurable increases in dissolved nutrients occurred after only 2 h of exposure to simulated sunlight. During a 6-h irradiation, nutrient release rates for total dissolved nitrogen (TDN) and phosphate were 2.2 ± 0.5 (standard error; S.E.) μmol g?1 h?1 and 0.09 ± 0.005 μmol g?1 h?1 (S.E.), compared to no significant changes in dark controls. The majority of nitrogen was released as dissolved organic nitrogen (87% on average) with lesser amounts of ammonium (13%). Continental shelf sediments resuspended in unfiltered seawater also released phosphate and TDN when exposed to sunlight, suggesting that this process can occur in a variety of marine and estuarine environments. The source material for inorganic nutrients appears to be associated with sediments rather than dissolved organic matter, as no significant changes in nutrient concentrations occurred in experiments with 0.2 μm-filtered creek water or seawater alone. Results suggest that photoproduction of dissolved nutrients from resuspended sediments could be an episodically significant and previously unrecognized source of dissolved organic and inorganic nutrients to coastal ecosystems. This process may be especially important for continental margins where episodic resuspension events occur, as well as in regions experiencing high riverine sediment fluxes resulting from erosion associated with deforestation and desertification.  相似文献

Headwater streams are foci for nutrient and energy loading from terrestrial landscapes, in situ nutrient transformations, and downstream transport. Despite the prominent role that headwater streams can have in regulating downstream water quality, the relative importance of processes that can influence nutrient uptake have not been fully compared in heterotrophic aquatic systems. To address this research need, we assessed the seasonality of dissolved organic carbon (DOC) and nitrate (NO3?) uptake, compared the relative influence of hydrologic and biogeochemical drivers on observed seasonal trends in nutrient uptake, and estimated the influence of these biological transformations on watershed scale nutrient retention and export. We determined that seasonal reductions in DOC and NO3? concentrations led to decreases in the potential for the biotic community to take up nutrients, and that seasonality of DOC and NO3? uptake was consistent with the seasonal dynamics of ecosystem metabolism. We calculated that that during the post-snowmelt period (June to August), biotic retention of both dissolved organic carbon and nitrate exceeded export fluxes from this headwater catchment, highlighting the potential for biological processes to regulate downstream water quality.  相似文献

Subterranean estuaries (STEs), the zones in which seawater and subsurface groundwater mix, are recognized as hotspots for biogeochemical reactions; however, little is known of the microbial communities that control many of those reactions. This study investigated the potential functions of microbes inhabiting a cenote and an offshore submarine spring (Pargos) in the near-coastal waters of the Yucatan Peninsula, Mexico. The inland cenote (Cenote Siete Bocas; C7B) is characterized by a chemocline that is host to an array of physicochemical gradients associated with microbial activities. The chemocline includes an increasing gradient in sulfide concentrations with depth and a decreasing gradient in nitrate concentrations. The microbial community within the chemocline was dominated by Sulfurimonas and Sulfurovum of the Campylobacteria, which are likely responsible for sulfide oxidation coupled with nitrate reduction. Although C7B has not been directly connected with Pargos Spring, water discharging from the spring has physicochemical characteristics and microbial community structures similar to C7B, strongly suggesting biogeochemical processing in the STE impacts groundwater composition prior to discharge. This work yields insight into the microbial communities and biogeochemical reactions in STEs in karstic aquifers and provides evidence for the importance of Campylobacteria in controlling nitrate concentrations exported to marine springs.  相似文献

Invasive plants, such as the hybrid cattail Typha × glauca, can reduce biodiversity and alter the ability of wetlands to provide critical ecosystem services, including nutrient cycling and carbon storage. Several approaches have been used to reduce Typha dominance and restore invaded wetlands, but long‐term studies assessing benefits of these restoration efforts are limited. A previous study demonstrated that aboveground harvesting of Typha × glauca stems and litter reduced Typha dominance 2 years post‐treatment in a Great Lakes coastal wetland. In the current study, we extended monitoring of experimental aboveground Typha harvest to 4 years post‐treatment and added assessments of treatment effects on soil nutrients, carbon emissions, and microbial community composition. Aboveground harvest treatment resulted in a dramatic reduction in Typha litter cover that persisted for 4 years, increased soil temperature, and increased abundance of the native plant genus Carex. However, aboveground harvest treatment did not significantly reduce Typha abundance, nor did it have significant effects on soil nutrient concentrations, carbon fluxes, or the taxonomic composition of soil microbial communities. We did observe differences in bacterial community composition between plots based on time since Typha invasion, which may indicate some legacy effects of Typha invasion. At the scale of this experiment (4 × 4 m plots), our results indicate that a single aboveground removal of Typha × glauca is not sufficient to restore a heavily invaded freshwater wetland ecosystem, and that periodic harvesting of Typha stems and litter may be required to maintain native plant abundance.  相似文献

Dissolved oxygen concentration plays a major role in shaping biotic interactions and nutrient flows within marine ecosystems. Throughout the global ocean, regions of low dissolved oxygen concentration (hypoxia) are a common and expanding feature of the water column, with major feedback on productivity and greenhouse gas cycling. To better understand microbial diversity underlying biogeochemical transformations within oxygen-deficient oceanic waters, we monitored and quantified bacterial and archaeal community dynamics in relation to dissolved gases and nutrients during a seasonal stratification and deep water renewal cycle in Saanich Inlet, British Columbia, a seasonally anoxic fjord. A number of microbial groups partitioned within oxygen-deficient waters including Nitrospina and SAR324 affiliated with the δ- proteobacteria , SAR406 and γ- proteobacteria related to thiotrophic gill symbionts of deep-sea clams and mussels. Microbial diversity was highest within the hypoxic transition zone decreasing dramatically within anoxic basin waters and temporal patterns of niche partitioning were observed along defined gradients of oxygen and phosphate. These results provide a robust comparative phylogenetic framework for inferring systems metabolism of nitrogen, carbon and sulfur cycling within oxygen-deficient oceanic waters and establish Saanich Inlet as a tractable model for studying the response of microbial communities to changing levels of water column hypoxia.  相似文献

Climate warming is expected to have particularly strong effects on tundra and boreal ecosystems, yet relatively few studies have examined soil responses to temperature change in these systems. We used closed‐top greenhouses to examine the response of soil respiration, nutrient availability, microbial abundance, and active fungal communities to soil warming in an Alaskan boreal forest dominated by mature black spruce. This treatment raised soil temperature by 0.5 °C and also resulted in a 22% decline in soil water content. We hypothesized that microbial abundance and activity would increase with the greenhouse treatment. Instead, we found that bacterial and fungal abundance declined by over 50%, and there was a trend toward lower activity of the chitin‐degrading enzyme N‐acetyl‐glucosaminidase. Soil respiration also declined by up to 50%, but only late in the growing season. These changes were accompanied by significant shifts in the community structure of active fungi, with decreased relative abundance of a dominant Thelephoroid fungus and increased relative abundance of Ascomycetes and Zygomycetes in response to warming. In line with our hypothesis, we found that warming marginally increased soil ammonium and nitrate availability as well as the overall diversity of active fungi. Our results indicate that rising temperatures in northern‐latitude ecosystems may not always cause a positive feedback to the soil carbon cycle, particularly in boreal forests with drier soils. Models of carbon cycle‐climate feedbacks could increase their predictive power by incorporating heterogeneity in soil properties and microbial communities across the boreal zone.  相似文献

Biogeochemical changes in marine sediments during coastal water hypoxia are well described, but less is known about underlying changes in microbial communities. Bacterial and archaeal communities in Louisiana continental shelf (LCS) hypoxic zone sediments were characterized by pyrosequencing 16S rRNA V4‐region gene fragments obtained by PCR amplification of community genomic DNA with bacterial‐ or archaeal‐specific primers. Duplicate LCS sediment cores collected during hypoxia had higher concentrations of Fe(II), and dissolved inorganic carbon, phosphate, and ammonium than cores collected when overlying water oxygen concentrations were normal. Pyrosequencing yielded 158 686 bacterial and 225 591 archaeal sequences from 20 sediment samples, representing five 2‐cm depth intervals in the duplicate cores. Bacterial communities grouped by sampling date and sediment depth in a neighbor‐joining analysis using Chao–Jaccard shared species values. Redundancy analysis indicated that variance in bacterial communities was mainly associated with differences in sediment chemistry between oxic and hypoxic water column conditions. Gammaproteobacteria (26.5%) were most prominent among bacterial sequences, followed by Firmicutes (9.6%), and Alphaproteobacteria (5.6%). Crenarchaeotal, thaumarchaeotal, and euryarchaeotal lineages accounted for 57%, 27%, and 16% of archaeal sequences, respectively. In Thaumarchaeota Marine Group I, sequences were 96–99% identical to the Nitrosopumilus maritimus SCM1 sequence, were highest in surficial sediments, and accounted for 31% of archaeal sequences when waters were normoxic vs. 13% of archaeal sequences when waters were hypoxic. Redundancy analysis showed Nitrosopumilus‐related sequence abundance was correlated with high solid‐phase Fe(III) concentrations, whereas most of the remaining archaeal clusters were not. In contrast, crenarchaeotal sequences were from phylogenetically diverse lineages, differed little in relative abundance between sampling times, and increased to high relative abundance with sediment depth. These results provide further evidence that marine sediment microbial community composition can be structured according to sediment chemistry and suggest the expansion of hypoxia in coastal waters may alter sediment microbial communities involved in carbon and nitrogen cycling.  相似文献

The biogeochemistry of mangrove sediments was investigated in several mangrove forest communities in Gazi Bay, a coastal lagoon in Kenya, Africa. Carbon dioxide fluxes, sediment median grain sizes, sedimentary organic carbon, nitrogen and phosphorus contents and pore-water characteristics (ammonium, nitrate, sulfate and chloride) could be related to forest type. Mangrove sediments have pH values that range from 3.5 to 8.3 due to the limited buffer capacity of these sediments and intense acidifying processes such as aerobic degradation of organic matter, oxidation of reduced components, ammonium uptake by roots and root respiration. The mangrove sediments are nitrogen-rich compared to mangrove litter, as a result of microbial nitrogen retention, uptake and fixation, and import of nitrogen-rich material. It appears that mangrove sediments in Gazi Bay act as a nutrient and carbon sink rather than as a source for adjacent seagrass and reef ecosystems.  相似文献

The quantification of greenhouse gas sources and sinks is important to understanding the impact of climate change. Methane (CH4) is a potent greenhouse gas, which, on a global scale, is released largely as a product of anaerobic microbial decomposition and predominantly from wetlands. A zone of intense CH4 production just below the water table is thought to contribute significantly to the overall flux from peat bogs. We describe the use of membrane inlet quadrupole mass spectrometry (QMS) to confirm the existence of bubbles, their gaseous concentrations and their localization at a fine spatial resolution within intact peat cores. We use the distribution of the noble gas argon (Ar) and the distinct QMS responses to dissolved and gaseous (bubble) phases to identify trapped bubbles with a resolution of 0.6 mm. Bubbles with CH4 concentrations of up to 20 kPa were widely distributed in the upper 300 mm of the cores with ~11% of all profiles comprising bubbles. The dissolved concentrations responsible for the bubbles were on average 83±80 μm , indicating lower concentrations relative to other QMS studies. We suggest that if the distinction between dissolved and gaseous phases is not made in studies of CH4 within peat profiles then the prominence of bubbles is likely to result in overestimates of dissolved CH4 concentrations. Fluxes of CH4 from peat as a result of drawdown or other perturbation are likely to be large, rapid and short lived because of bubble burst, and also larger than from peat without bubbles. We suggest that the dynamics of fluxes need to be modelled taking into account both gaseous and dissolved phases. Estimates of potential fluxes that assume CH4 is dissolved are likely to overestimate fluxes if the gaseous phase has not been taken into account.  相似文献

16S rRNA gene molecular analysis elucidated the spatiotemporal distribution of bacterial biofilm communities along a water quality gradient. Multivariate statistics indicated that terrestrial runoff, in particular dissolved organic carbon and chlorophyll a concentrations, induced shifts of specific bacterial communities between locations and seasons, suggesting microbial biofilms could be suitable bioindicators for water quality.  相似文献

土壤是地球表层最为重要的碳库也是温室气体的源或汇。自工业革命以来,对土壤温室气体的容量、收支平衡和通量等已有较多研究和估算,但对关键过程及其源/汇的研究却十分有限。微生物是土壤碳氮转化的主要驱动者, 在生态系统碳氮循环过程中扮演重要的角色,对全球气候变化有着响应的响应、适应及反馈,然而其个体数量,群落结构和多样性如何与气候扰动相互关联、进而怎样影响生态系统过程的问题仍有待进一步探索。从微生物介导的碳氮循环过程入手,重点讨论微生物对气候变化包括温室气体(CO2,CH4,N2O)增加、全球变暖、大气氮沉降等的响应和反馈,并由此提出削减温室气体排放的可能途径和今后发展的方向。  相似文献

Inland waters are increasingly recognized as critical sites of methane emissions to the atmosphere, but the biogeochemical reactions driving such fluxes are less well understood. The Prairie Pothole Region (PPR) of North America is one of the largest wetland complexes in the world, containing millions of small, shallow wetlands. The sediment pore waters of PPR wetlands contain some of the highest concentrations of dissolved organic carbon (DOC) and sulfur species ever recorded in terrestrial aquatic environments. Using a suite of geochemical and microbiological analyses, we measured the impact of sedimentary carbon and sulfur transformations in these wetlands on methane fluxes to the atmosphere. This research represents the first study of coupled geochemistry and microbiology within the PPR and demonstrates how the conversion of abundant labile DOC pools into methane results in some of the highest fluxes of this greenhouse gas to the atmosphere ever reported. Abundant DOC and sulfate additionally supported some of the highest sulfate reduction rates ever measured in terrestrial aquatic environments, which we infer to account for a large fraction of carbon mineralization in this system. Methane accumulations in zones of active sulfate reduction may be due to either the transport of free methane gas from deeper locations or the co‐occurrence of methanogenesis and sulfate reduction. If both respiratory processes are concurrent, any competitive inhibition of methanogenesis by sulfate‐reducing bacteria may be lessened by the presence of large labile DOC pools that yield noncompetitive substrates such as methanol. Our results reveal some of the underlying mechanisms that make PPR wetlands biogeochemical hotspots, which ultimately leads to their critical, but poorly recognized role in regional greenhouse gas emissions.  相似文献

ABSTRACT. The abundance, sizes, and when appropriate, diversity of gymnamoebae were documented at approximately monthly intervals for four years (1995–1998) at a grassy, terrestrial site slightly upslope from a freshwater pond. Soil samples were analyzed for viable gymnamoebae using a standard laboratory culturing protocol. The mean density of gymnamoebae based on the total data set was ca. 1,600/g (s.e. ± 190). Minimum densities of gymnamoebae (156/g) occurred in January 1995, and a maximum for the sampling period (5,838/g) occurred in July 1997, when a rainy period followed an extended period of drought. Among the environmental variables monitored (precipitation, soil moisture, organic content, and temperature) only precipitation correlated significantly with abundance of gymnamoebae (r= 0.34 , p = 0.02 ). During the mild, moist El Niño winter of 1997–1998, a larger than usual number of gymnamoebae was recorded at the site (~3,800/g) compared to a mean density of ~900/g for comparable periods in preceding years. The mean sizes were also larger. Since gymnamoebae are increasingly recognized as major members of soil microbial communities enhancing soil fertility through nutrient mineralization, it is important to document environmental variables that influence their abundance and activity in terrestrial ecosystems.  相似文献

Dissolved Organic Carbon in Terrestrial Ecosystems: Synthesis and a Model   总被引:37,自引:3,他引:34  
The movement of dissolved organic carbon (DOC) through soils is an important process for the transport of carbon within ecosystems and the formation of soil organic matter. In some cases, DOC fluxes may also contribute to the carbon balance of terrestrial ecosystems; in most ecosystems, they are an important source of energy, carbon, and nutrient transfers from terrestrial to aquatic ecosystems. Despite their importance for terrestrial and aquatic biogeochemistry, these fluxes are rarely represented in conceptual or numerical models of terrestrial biogeochemistry. In part, this is due to the lack of a comprehensive understanding of the suite of processes that control DOC dynamics in soils. In this article, we synthesize information on the geochemical and biological factors that control DOC fluxes through soils. We focus on conceptual issues and quantitative evaluations of key process rates to present a general numerical model of DOC dynamics. We then test the sensitivity of the model to variation in the controlling parameters to highlight both the significance of DOC fluxes to terrestrial carbon processes and the key uncertainties that require additional experiments and data. Simulation model results indicate the importance of representing both root carbon inputs and soluble carbon fluxes to predict the quantity and distribution of soil carbon in soil layers. For a test case in a temperate forest, DOC contributed 25% of the total soil profile carbon, whereas roots provided the remainder. The analysis also shows that physical factors—most notably, sorption dynamics and hydrology—play the dominant role in regulating DOC losses from terrestrial ecosystems but that interactions between hydrology and microbial–DOC relationships are important in regulating the fluxes of DOC in the litter and surface soil horizons. The model also indicates that DOC fluxes to deeper soil layers can support a large fraction (up to 30%) of microbial activity below 40 cm. The influence of long-term chronic ozone exposure on carbon fluxes from young beech trees (Fagus sylvatica L.) into the phospholipid fraction of microbial communities (PLFA) in the rhizosphere and into the dissolved organic carbon (DOC) fraction was studied in a lysimeter experiment using 13C depleted CO2 over one vegetation period to identify possible changes in below ground carbon translocation processes due to the plant stress. It could be shown that microbial biomass as well as individual microbial communities and their activity pattern in the rhizosphere of young beech trees are mainly driven by the vegetation period. An increase in total microbial biomass as well as individual microbial communities was detected during the vegetation period from June to September. However, also a clear ozone effect was visible mainly at the end of the vegetation period. Enzyme activities and PLFA data indicated earlier induced plant senescence as a response to the elevated ozone treatment. Furthermore higher microbial biomass and abundance of plant C utilizing microbes was observed in elevated ozone treatments over the whole vegetation period.  相似文献

1. Animals play a major role in nutrient cycling via excretory processes. Although the positive indirect effects of grazers on periphytic algae are well understood, little is known about top‐down effects on decomposers of shredders living on leaf litter. 2. Nutrient cycling by shredders in oligotrophic forest streams may be important for the microbial‐detritus compartment at very small spatial scales (i.e. within the leaf packs in which shredders feed). We hypothesised that insect excretion may cause local nutrient enrichment, so that microorganism growth on leaves is stimulated. 3. We first tested the effect of increasing concentration of ammonium (+10, +20 and +40 μg NH4+ L?1) on fungal and bacterial biomass on leaf litter in a laboratory experiment. Then we performed two experiments to test the effect of the presence and feeding activity of shredder larvae. We used two species belonging to the trichopteran family Sericostomatidae: the Palaearctic Sericostoma vittatum and the Neotropical Myothrichia murina, to test the effect of these shredders on fungal and bacterial biomass and decomposition on leaves of Quercus robur and Nothofagus pumilio, respectively. All experiments were run in water with low ammonium concentrations (2.4 ± 0.34 to 14.47 ± 0.95 μg NH4+ L?1). 4. After 5 days of incubation, NH4 concentrations were reduced to near‐ambient streamwater concentrations in all treatments with leaves. Fungal biomass was positively affected by increased ammonium concentration. On the other hand, bacteria abundance was similar in all treatments, both in terms of abundance (bacteria cells mg?1 leaf DW) and biomass. However, there was a tendency towards larger mean cell size in treatments with 20 μg NH4 L?1. 5. In the experiment with S. vittatum, fungal biomass in the treatment with insects was more than twice that in the control after 15 days. Bacteria were not detected in treatments with insects, where hyphae were abundant, but they were abundant in treatments without larvae. In the decomposition experiment run with M. murina, leaf‐mass loss was significantly higher in treatments with larvae than in controls. 6. Our hypothesis of a positive effect of shredders on fungal biomass and decomposition was demonstrated. Insect excretion caused ammonium concentration to increase in the microcosms, contributing to microbial N uptake in leaf substrata, which resulted in structural and functional changes in community attributes. The positive effect of detritivores on microbes has been mostly neglected in stream nutrient‐cycling models; our findings suggest that this phenomenon may be of greater importance than expected in stream nutrient budgets.  相似文献

A common source of disturbance for coastal aquatic habitats is nutrient enrichment through anthropogenic activities. Although the water column bacterioplankton communities in these environments have been characterized in some cases, changes in α-diversity and/or the abundances of specific taxonomic groups across enriched habitats remain unclear. Here, we investigated the bacterial community changes at three different nutrient-enriched and adjacent undisturbed habitats along the north coast of Crete, Greece: a fish farm, a closed bay within a town with low water renewal rates, and a city port where the level of nutrient enrichment and the trophic status of the habitat were different. Even though changes in α-diversity were different at each site, we observed across the sites a common change pattern accounting for most of the community variation for five of the most abundant bacterial groups: a decrease in the abundance of the Pelagibacteraceae and SAR86 and an increase in the abundance of the Alteromonadaceae, Rhodobacteraceae, and Cryomorphaceae in the impacted sites. The abundances of the groups that increased and decreased in the impacted sites were significantly correlated (positively and negatively, respectively) with the total heterotrophic bacterial counts and the concentrations of dissolved organic carbon and/or dissolved nitrogen and chlorophyll α, indicating that the common change pattern was associated with nutrient enrichment. Our results provide an in situ indication concerning the association of specific bacterioplankton groups with nutrient enrichment. These groups could potentially be used as indicators for nutrient enrichment if the pattern is confirmed over a broader spatial and temporal scale by future studies.  相似文献
