Nucleotide sequences of mitochondrial DNA (mtDNA) of 50 domestic cats (Felis catus) obtained from the Tsushima Islands were determined and the genetic diversity was analyzed. In the cats, six haplotypes of the complete cytochrome b sequences (1,140 base-pairs, bp) and ten haplotypes of the partial control region sequences (350 bp) were identified. Haplotypes obtained from both genes showed existence of at least 11 maternal lineages of domestic cats in Tsushima. Mean values of polymorphic site numbers and sequences differences in the control region were 2.4 times and 1.8 times higher than those in the cytochrome b gene, respectively. Our results support the idea that the evolutionary rate of the control region was faster than that of the cytochrome b as reported in other mammals. Molecular phylogenetic trees showed the similar clustering of haplotypes for both genes. Meanwhile, no individual variations within the Tsushima leopard cat (Felis bengalensis euptilura), which is native to Tsushima, were observed, possibly as a result of genetic drift in the small ancestral population by geographical isolation. In contrast, the diversity of the domestic cat population was higher than that of the leopard cats, because the genetic variability of the former's founders, which were repeatedly brought to Tsushima in the past, still remains. In addition, no sequences of the leopard cat mtDNA were detected in any domestic cats. However, because the possibility that the domestic cat would crossbreed with the leopard cat cannot be denied, genetic monitoring of two species is necessary to biological conservation in Tsushima.

Evolutionary shape changes in skull and mandibular anatomy was analysed in 223 specimens of pantherine felids (Neofelis nebulosa, Panthera leo, Panthera onca, Panthera pardus, Panthera tigris, Panthera uncia) compared to a small‐felid outgroup, consisting of 86 specimens of nine different species, using digital surface morphometry on 25 (skull) and 17 (mandible) landmarks. Shape evolution in the pantherine species is complex and nonlinear, and involves both large‐scale and small‐scale shape changes. Shape changes frequently differ among the ingroup species, but the four large Panthera species (leo, onca, pardus, tigris) bear some resemblance to each other. The leopard and jaguar bear the closest resemblance to each other, and several shape changes are common to the lion and tiger, but have probably evolved convergently as a result of large size. The lion has undergone the largest and most numerous shape changes from a small‐felid outgroup. Certain shape changes in the skull and, in some respects, the mandible of the clouded leopard bear resemblance to those in the four large Panthera species. The snow leopard is often regarded as the most primitive of the extant Panthera, and skull and mandibular shape changes often diverge markedly from those observed in the other five ingroup taxa; its overall skull shape is rather similar to the small‐felid outgroup. This indicates that the shape changes in the clouded leopard are convergent with those of the four large Panthera species. Landmark integration showed no significant correlation with molecular phylogeny, chiefly owing to the snow leopard being placed among the four large Panthera species. A traditional phylogenetic topology with the snow leopard as the basal‐most species of Panthera yielded a weak but nonsignificant phylogenetic signal. © 2008 The Linnean Society of London, Biological Journal of the Linnean Society, 2008, 95 , 766–778.

Coat colours and patterns are highly variable in cats and are determined mainly by several genes with Mendelian inheritance. A 2‐bp deletion in agouti signalling protein (ASIP) is associated with melanism in domestic cats. Bengal cats are hybrids between domestic cats and Asian leopard cats (Prionailurus bengalensis), and the charcoal coat colouration/pattern in Bengals presents as a possible incomplete melanism. The complete coding region of ASIP was directly sequenced in Asian leopard, domestic and Bengal cats. Twenty‐seven variants were identified between domestic and leopard cats and were investigated in Bengals and Savannahs, a hybrid with servals (Leptailurus serval). The leopard cat ASIP haplotype was distinguished from domestic cat by four synonymous and four non‐synonymous exonic SNPs, as well as 19 intronic variants, including a 42‐bp deletion in intron 4. Fifty‐six of 64 reported charcoal cats were compound heterozygotes at ASIP, with leopard cat agouti (APbe) and domestic cat non‐agouti (a) haplotypes. Twenty‐four Bengals had an additional unique haplotype (A2) for exon 2 that was not identified in leopard cats, servals or jungle cats (Felis chaus). The compound heterozygote state suggests the leopard cat allele, in combination with the recessive non‐agouti allele, influences Bengal markings, producing a darker, yet not completely melanistic coat. This is the first validation of a leopard cat allele segregating in the Bengal breed and likely affecting their overall pelage phenotype. Genetic testing services need to be aware of the possible segregation of wild felid alleles in all assays performed on hybrid cats.

Food habits of three sympatric carnivore mammals in the Tsushima islands of Japan were studied during 1986–91. Scats of the Tsushima marten (n=1236), the Siberian weasel (n=218) and the Tsushima leopard cat (n=350) were collected monthly and the food items were determined by scat contents analysis. Marten was omnivorous showing a high level of diversity of food throughout the year. The important foods for marten were fruits and berries from spring to autumn, insects in summer and autumn and small mammals all year round. Leopard cat preferred to hunt wood/mice and birds, and remained a flesh meat specialist throughout the year. Weasel was intermediate between marten and leopard cat, but was slightly biased towards the flesh meat eater. The three carnivores do not compete against one another for food, except for small rodents. A conflict for food between leopard cat and weasel was suggested to be more intense than that of other combinations based on diet overlapping. Marten may be characterized as an opportunistic generalist. When interspecific competitors existed, or human disturbance to the habitat occurred, the preferential flexibility of the marten to alternative food resources might become more advantageous than the other two species.

The estimated population of the Tsushima leopard cat Prionailurus bengalensis euptilura is only 80–110 individuals. However, the cause of the population decline is not clear. We investigated temporal changes in the food habits of the cat and two other species of Carnivora (marten and weasel) inhabiting the Tsushima Islands by measuring δ13C and δ15N values in hair samples. Hair samples of the cat were collected not only from specimens and furs, but also from feces. The gathering of hair from cat feces was most efficient when the feces were collected in the spring. The food habit of male cats seemed to be more diverse and tended to comprise prey of higher trophic levels than the food habits of the females. The δ13C and δ15N measurements suggested that the trophic level of the food sources has been decreasing over the last several decades for the cat and weasel, but not for the marten. Increased consumption of prey from lower trophic levels in the food habit of the cat seems to be related to the decline of the cat population because these phenomena occurred simultaneously.

Impact of predation by domestic cats Felis catus in an urban area
  • 1 As companion animals, domestic cats Felis catus can attain very high densities, and have the potential to exert detrimental effects on prey species. Yet, there is a paucity of information on the impact of cat predation in urban areas, where most cats are likely to be present.
  • 2 We quantified the minimum number of animals killed annually by cats in a 4.2-km2 area of Bristol, UK, by asking owners to record prey animals returned home by their pets. The potential impact of cat predation on prey species was estimated by comparing the number of animals killed with published estimates of prey density and annual productivity.
  • 3 Predator density was 229 cats/km2.
  • 4 Five mammal, 10 bird and one amphibian prey species were recorded. Mean predation rate was 21 prey/cat/annum. The most commonly recorded prey species was the wood mouse Apodemus sylvaticus.
  • 5 Predation on birds was greatest in spring and summer, and probably reflected the killing of juvenile individuals. For three prey species (house sparrow Passer domesticus, dunnock Prunella modularis, robin Erithacus rubecula), estimated predation rates were high relative to annual productivity, such that predation by cats may have created a dispersal sink for juveniles from more productive neighbouring areas. The impact of cats on these species therefore warrants further investigation.

Anthropogenic landscape change can lead to increased opportunities for pathogen transmission between domestic and non-domestic animals. Pumas, bobcats, and domestic cats are sympatric in many areas of North America and share many of the same pathogens, some of which are zoonotic. We analyzed bobcat, puma, and feral domestic cat samples collected from targeted geographic areas. We examined exposure to three pathogens that are taxonomically diverse (bacterial, protozoal, viral), that incorporate multiple transmission strategies (vector-borne, environmental exposure/ingestion, and direct contact), and that vary in species-specificity. Bartonella spp., Feline Immunodeficiency Virus (FIV), and Toxoplasma gondii IgG were detected in all three species with mean respective prevalence as follows: puma 16%, 41% and 75%; bobcat 31%, 22% and 43%; domestic cat 45%, 10% and 1%. Bartonella spp. were highly prevalent among domestic cats in Southern California compared to other cohort groups. Feline Immunodeficiency Virus exposure was primarily associated with species and age, and was not influenced by geographic location. Pumas were more likely to be infected with FIV than bobcats, with domestic cats having the lowest infection rate. Toxoplasma gondii seroprevalence was high in both pumas and bobcats across all sites; in contrast, few domestic cats were seropositive, despite the fact that feral, free ranging domestic cats were targeted in this study. Interestingly, a directly transmitted species-specific disease (FIV) was not associated with geographic location, while exposure to indirectly transmitted diseases--vector-borne for Bartonella spp. and ingestion of oocysts via infected prey or environmental exposure for T. gondii--varied significantly by site. Pathogens transmitted by direct contact may be more dependent upon individual behaviors and intra-specific encounters. Future studies will integrate host density, as well as landscape features, to better understand the mechanisms driving disease exposure and to predict zones of cross-species pathogen transmission among wild and domestic felids.

Feline immunodeficiency virus (FIV) was isolated from a wild-caught Tsushima cat (Felis bengalensis euptilura), an endangered Japanese nondomestic subspecies of leopard cat (F. bengalensis). Phylogenetic analysis of the env gene sequences indicated that the FIV from the Tsushima cat belonged to a cluster of subtype D FIVs from domestic cats. FIVs from both the Tsushima cat and the domestic cat showed similar levels of replication and cytopathicity in lymphoid cell lines derived from these two species. The results indicated the occurrence of interspecies transmission of FIV from the domestic cat to the Tsushima cat in the wild.

The sabertoothed felids were among the most unusual predators in the late Tertiary ecosystems, and the sabertooth morphology is regarded as being absent from the modern ecosystems. In recent years, the primitive Paramachairodus has become well known and has yielded much valuable information on the primitive skull morphology among sabercats, providing the first evidence-based scenarios for the evolution of skull morphology in later sabercats. However, comparison of craniomandibular morphology of the extant clouded leopard Neofelis nebulosa and Paramachairodus reveals numerous similarities and subsequent divergence from other extant great cats. In several key aspects, the clouded leopard has approached a primitive sabercat craniomandibular morphology and has diverged markedly from its sister group, the Panthera lineage. A primitive sabertooth condition arose six times in the Tertiary period, not five as is traditionally advocated. The clouded leopard appears to be a useful model for understanding primitive sabercat morphology and could shed important light on sabercat evolution. The unusual nature of the clouded leopard implies that increased efforts should be spent on insuring the continuing survival of this rare and endangered species.

Understanding attitude of local people towards big cats is vital for conservation interventions to succeed. Taking tigers and leopards as focal species, we investigated local peoples' attitude towards four subjects—tiger, tiger conservation, leopard, and leopard conservation—considering demographic and socio-economic factors as well as past experience with such predators in Nepal's first national park and a world heritage site, Chitwan National Park. The data were collected from 414 local people using structured questionnaires and their attitude towards the four subjects determined. We performed ordinal logistic regression analysis to identify the best fitted model and significant variables affecting attitudes. While majority of the people (51%) strongly liked tigers, fewer people (38%) had similar view while it came to leopard. However, a greater proportion of people strongly agreed that the conservation of tigers (61%) and leopards (53%) is important. About 12% people had negative attitude towards both big cats. We found women and low income respondents to likely have negative attitudes and higher caste Hindus to have positive attitudes towards both big cats and their conservation. Better educated persons and the owners to larger herds of livestock only agreed on conservation of tiger but not leopard. Past experience with the predator negatively affected attitude towards tiger but not leopard. We suggest the identified cohort of people with negative attitudes be more targeted in conservation initiatives. The reasons behind the similarities and differences in peoples’ attitudes are discussed and designation of species-specific programmes for both cats is recommended.

Introgression can be an important evolutionary force but it can also lead to species extinction and as such is a crucial issue for species conservation. However, introgression is difficult to detect, morphologically as well as genetically. Hybridization with domestic cats (Felis silvestris catus) is a major concern for the conservation of European wildcats (Felis s. silvestris). The available morphologic and genetic markers for the two Felis subspecies are not sufficient to reliably detect hybrids beyond first generation. Here we present a single nucleotide polymorphism (SNP) based approach that allows the identification of introgressed individuals. Using high‐throughput sequencing of reduced representation libraries we developed a diagnostic marker set containing 48 SNPs (Fst  > 0.8) which allows the identification of wildcats, domestic cats, their hybrids and backcrosses. This allows assessing introgression rate in natural wildcat populations and is key for a better understanding of hybridization processes.

The endangered European wildcat (Felis silvestris silvestris) is represented, today, by fragmented and declining populations whose genetic integrity is considered to be seriously threatened by crossbreeding with widespread free-ranging domestic cats. Extensive and recent hybridisation has been described in Hungary and Scotland, in contrast with rare introgression of domestic alleles in Italy and Germany. In Portugal, the wildcat is now listed as VULNERABLE in the Red Book of Portuguese Vertebrates. Nevertheless, genetic diversity of populations and the eventual interbreeding with domestic cats remain poorly studied. We surveyed genetic variation at 12 autosomal microsatellites for 34 wild and 64 domestic cats collected across Portugal. Wild and domestic cats were significantly differentiated both at allele frequencies and sizes (F ST=0.11, R ST = 0.18, P < 0.001). Population structure and admixture analyses performed using Bayesian approaches also showed evidence of two discrete groups clustering wild and domestic populations. Results did not show significant genetic divergence among Northern, Central and Southern wildcats. Six morphologically identified wildcats were significantly assigned to the domestic cluster, revealing some discrepancy between phenotypic and genetic identifications. We detected four hybrids (approximately 14%) using a consensus analysis of different Bayesian model-based software. These hybrids were identified throughout all sampled areas, suggesting that hybridisation is of major concern for the appropriate implementation of wildcat conservation strategies in Portugal.

Cat predation upon bat species has been reported to have significant effects on bat populations in both rural and urban areas. The majority of research in this area has focussed on observational data from bat rehabilitators documenting injuries, and cat owners, when domestic cats present prey. However, this has the potential to underestimate the number of bats killed or injured by cats. Here, we use forensic DNA analysis techniques to analyze swabs taken from injured bats in the United Kingdom, mainly including Pipistrellus pipistrellus (40 out of 72 specimens). Using quantitative PCR, cat DNA was found in two‐thirds of samples submitted by bat rehabilitators. Of these samples, short tandem repeat analysis produced partial DNA profiles for approximately one‐third of samples, which could be used to link predation events to individual cats. The use of genetic analysis can complement observational data and potentially provide additional information to give a more accurate estimation of cat predation.

Blastomyces dermatitidis, the etiologic agent of blastomycosis, a potentially life-threatening systemic mycosis of humans and animals, is acquired from a yet incompletely defined environmental niche. There is controversy regarding the potential for contact with the fungus in or near one’s home, particularly in urban areas. We investigated an outbreak of blastomycosis among five urban, indoor cats diagnosed at three veterinary clinics March 3–July 13, 2005, in suburban Chicago, Illinois, by owner interviews, site visits, environmental cultures for B. dermatitidis, GIS analysis, and analysis of local weather data. There were no environmental exposures common to the five cats that lived a median of 300 m from nearest body of water, in homes on a loam soil. Closest and farthest case home sites were 3.4 and 26.1 km, respectively. All cats were confined indoors except one cat that averaged 15 min/week in his backyard and was exposed to excavation. B. dermatitidis was not isolated from any of 60 environmental samples. The annualized incidence rate March through July 2005 among 6,761 cats in these practices was 178/100,000, compared to none in the previous 4 years, and 0.14/100,000 cat visits from a nationwide animal hospital registry. Precipitation January through June 2005 was 9.30 versus period mean of 14.05 ± 1.69 inches the previous 4 years (P = 0.01). Circumstantial evidence suggests acquisition of B. dermatitidis from the home site environment in five cats. Relative drought may have contributed to an apparent outbreak of blastomycosis in this urban locale.

Cats and their fleas collected in Guatemala were investigated for the presence of Bartonella infections. Bartonella bacteria were cultured from 8.2% (13/159) of cats, and all cultures were identified as B. henselae. Molecular analysis allowed detection of Bartonella DNA in 33.8% (48/142) of cats and in 22.4% (34/152) of cat fleas using gltA, nuoG, and 16S–23S internal transcribed spacer targets. Two Bartonella species, B. henselae and B. clarridgeiae, were identified in cats and cat fleas by molecular analysis, with B. henselae being more common than B. clarridgeiae in the cats (68.1%; 32/47 vs 31.9%; 15/47). The nuoG was found to be less sensitive for detecting B. clarridgeiae compared with other molecular targets and could detect only two of the 15 B. clarridgeiae‐infected cats. No significant differences were observed for prevalence between male and female cats and between different age groups. No evident association was observed between the presence of Bartonella species in cats and in their fleas.

Domestic cats exhibit physiological proteinuria due to the excretion of cauxin, a carboxylesterase, into the urine. In the present report, we demonstrate that cauxin is excreted in a species-, sex-, and age-dependent manner. Although the cauxin gene is conserved in mammals, including human, mouse, and dog, urinary cauxin was found only in member of the genus Felis and lynx (bobcat, and lynx) and not in other Felidae (genus: Panthera and puma) tested. In mature cats, cauxin excretion was higher in intact males than in castrated males or in intact or spayed females. Daily cauxin excretion decreased immediately after castration. Immunohistochemistry confirmed that cauxin expression in the kidney proximal straight tubules was higher in intact males than in castrated males. Urinary cauxin was detectable by Western blotting in cats older than about 3 months, and its excretion increased with age. In a zymographic esterase assay, urine contained a major cauxin band; by contrast, kidney homogenates contained three major bands, comprising two carboxylesterases and an unidentified esterase, and one minor cauxin band. These results suggest that 1. cauxin excretion is regulated by sex hormones, such as testosterone, 2. cauxin functions as an esterase in the urine rather than in kidney cells, and 3. the decomposition products by cauxin are excreted in a species-, sex-, and age-dependent manner, as is cauxin itself.
