Powdery mildew, caused by Blumeria graminis f.sp. tritici, is a major wheat disease in maritime and temperate climates. Breeding for race-non-specific or partial resistance is a cost-effective and environmentally friendly disease control strategy. The German spring wheat cultivar Naxos has proven to be a good source for partial resistance to powdery mildew. The objectives of the present study were to map the resistance loci in Naxos with use of high-density SNP markers in the Shanghai3/Catbird x Naxos inbred line population and validate the results in a different genetic background; Soru#1 x Naxos. Both populations were genotyped with the Illumina iSelect 90K wheat chip, and integrated linkage maps developed by inclusion of previously genotyped SSR and DArT markers. With the new linkage maps, we detected a total of 12 QTL for powdery mildew resistance in Shanghai3/Catbird x Naxos, of which eight were derived from Naxos. Previously reported QTL on chromosome arms 1AS and 2BL were more precisely mapped and the SNP markers enabled discovery of new QTL on 1AL, 2AL, 5AS and 5AL. In the Soru#1 x Naxos population, four QTL for powdery mildew resistance were detected, of which three had resistance from Naxos. This mapping verified the 1AS and 2AL QTL detected in Shanghai3/Catbird x Naxos, and identified a new QTL from Naxos on 2BL. In conclusion, the improved linkage maps with SNP markers enabled discovery of new resistance QTL and more precise mapping of previously known QTL. Moreover, the results were validated in an independent genetic background.  相似文献

Two dominant powdery mildew resistance genes introduced from Triticum carthlicum accession PS5 to common wheat were identified and tagged using microsatellite markers. The gene designated PmPS5A was placed on wheat chromosome 2AL and linked to the microsatellite marker Xgwm356 at a genetic distance of 10.2 cM. Based on the information of its origin, chromosome location, and reactions to 5 powdery mildew isolates, this gene could be a member of the complex Pm4 locus. The 2nd gene designated PmPS5B was located on wheat chromosome 2BL with 3 microsatellite markers mapping proximally to the gene: Xwmc317 at 1.1 cM; Xgwm111 at 2.2 cM; and Xgwm382 at 4.0 cM; and 1 marker, Xgwm526, mapping distally to the gene at a distance of 18.1 cM. Since this gene showed no linkage to the other 2 known powdery mildew resistance genes on wheat chromosome 2B, Pm6 and Pm26, we believe it is a novel powdery mildew resistance gene and propose to designate this gene as Pm33.  相似文献

Powdery mildew significantly affects grain yield and end-use quality of winter wheat in the southern Great Plains. Employing resistance resources in locally adapted cultivars is the most effective means to control powdery mildew. Two types of powdery mildew resistance exist in wheat cultivars, i.e., qualitative and quantitative. Qualitative resistance is controlled by major genes, is race-specific, is not durable, and is effective in seedlings and in adult plants. Quantitative resistance is controlled by minor genes, is non-race-specific, is durable, and is predominantly effective in adult plants. In this study, we found that the segregation of powdery mildew resistance in a population of recombinant inbred lines developed from a cross between the susceptible cultivar Jagger and the resistant cultivar 2174 was controlled by a major QTL on the short arm of chromosome 1A and modified by four minor QTLs on chromosomes 1B, 3B, 4A, and 6D. The major QTL was mapped to the genomic region where the Pm3 gene resides. Using specific PCR markers for seven Pm3 alleles, 2174 was found to carry the Pm3a allele. Pm3a explained 61% of the total phenotypic variation in disease reaction observed among seedlings inoculated in the greenhouse and adult plants grown in the field and subjected to natural disease pressure. The resistant Pm3a allele was present among 4 of 31 cultivars currently being produced in the southern Great Plains. The genetic effects of several minor loci varied with different developmental stages and environments. Molecular markers associated with these genetic loci would facilitate incorporating genetic resistance to powdery mildew into improved winter wheat cultivars.  相似文献

Powdery mildew, caused by Blumeria graminis f. sp. tritici, is a major fungal disease in common wheat (Triticum aestivum L.) worldwide. The Chinese winter wheat cultivar Lumai 21 has shown good and stable adult plant resistance for 19 years. The aim of this study was to map quantitative trait loci (QTLs) for resistance to powdery mildew in a population of 200 F3 lines from the cross Lumai 21/Jingshuang 16. The population was tested for powdery mildew reaction in Beijing and Anyang in the 2005–2006 and 2006–2007 cropping seasons, providing data for 4 environments. A total of 1,375 simple sequence repeat (SSR) markers were screened for associations with powdery mildew reactions, initially in bulked segregant analysis. Based on the mean disease values averaged across environments, broad-sense heritabilities of maximum disease severity and area under the disease progress curve were 0.96 and 0.77, respectively. Three QTLs for adult plant resistance were detected by inclusive composite interval mapping. These were designated QPm.caas-2BS, QPm.caas-2BL and QPm.caas-2DL, respectively, and explained from 5.4 to 20.6% of the phenotypic variance across environments. QPm.caas-2BS and QPm.caas-2DL were likely new adult plant resistance QTLs flanked by SSR markers Xbarc98Xbarc1147 and Xwmc18Xcfd233, respectively. These markers could be useful for improving wheat powdery mildew resistance in breeding programs.  相似文献

Fourteen Bacillus thuringiensis isolates having both insecticidal activity and in vitro antifungal activity were selected and tested for in vivo antifungal activity against tomato late blight, wheat leaf rust, tomato gray mold, and barley powdery mildew in growth chambers. All the isolates represented more than 70% disease control efficacy against at least one of four plant diseases. Specifically, 12 isolates exhibited strong control activity against barley powdery mildew. Under glasshouse conditions, four (50-02, 52-08, 52-16, and 52- 18) of the isolates also displayed potent control efficacy against cucumber powdery mildew. To our knowledge, this is the first report of B. thuringiensis isolates that have disease control efficacy against powdery mildew of barley and cucumber as well as insecticidal activity.  相似文献

Segregation analysis of resistance to powdery mildew in a F2 progeny from the cross Chinese Spring (CS) × TA2682c revealed the inheritance of a dominant and a recessive powdery mildew resistance gene. Selfing of susceptible F2 individuals allowed the establishment of a mapping population segregating exclusively for the recessive resistance gene. The extracted resistant derivative showing full resistance to each of 11 wheat powdery mildew isolates was designated RD30. Amplified fragment length polymorphism (AFLP) analysis of bulked segregants from F3s showing the homozygous susceptible and resistant phenotypes revealed an AFLP marker that was associated with the recessive resistance gene in repulsion phase. Following the assignment of this AFLP marker to wheat chromosome 7A by means of CS nullitetrasomics, an inspection of simple sequence repeat (SSR) loci evenly spaced along chromosome 7A showed that the recessive resistance gene maps to the distal region of chromosome 7AL. On the basis of its close linkage to the Pm1 locus, as inferred from connecting partial genetic maps of 7AL of populations CS × TA2682c and CS × Virest (Pm1e), and its unique disease response pattern, the recessive resistance gene in RD30 was considered to be novel and tentatively designated mlRD30.Communicated by C. Since it was commercialized in 2008, Liangxing 66 is one of the most widely grown cultivars of wheat (Triticum aestivum L.) in winter and facultative wheat-producing regions in northern China. This cultivar displays broad-spectrum resistance to isolates of powdery mildew. To identify the powdery mildew resistance gene in Liangxing 66, genetic analysis and molecular mapping were conducted using the F2 populations and F2:3 families derived from the reciprocal crosses of Liangxing 66 and the susceptible cultivar Jingshuang 16. A single dominant gene, tentatively designated PmLX66, conferred resistance in Liangxing 66 to the powdery mildew isolate E09. The results of molecular mapping indicated that this gene was located on the short arm of chromosome 5D and flanked by SCAR203 and Xcfd81 at genetic distances of 0.4 and 2.8?cM, respectively, which is similar to the position of locus Pm2. However, PmLX66 and Pm2 showed different reactions to five of the 42 isolates of powdery mildew tested. Together, these results indicated that PmLX66 was most likely an allele of Pm2. Based on its superior yield and agronomic performance, in combination with powdery mildew resistance, Liangxing 66 is useful as a promising parent for control of powdery mildew and for the development of new disease-resistant cultivars.  相似文献

To determine the influence of plant density and powdery mildew infection of winter wheat and to predict grain yield, hyperspectral canopy reflectance of winter wheat was measured for two plant densities at Feekes growth stage (GS) 10.5.3, 10.5.4, and 11.1 in the 2009–2010 and 2010–2011 seasons. Reflectance in near infrared (NIR) regions was significantly correlated with disease index at GS 10.5.3, 10.5.4, and 11.1 at two plant densities in both seasons. For the two plant densities, the area of the red edge peak (Σdr 680–760 nm), difference vegetation index (DVI), and triangular vegetation index (TVI) were significantly correlated negatively with disease index at three GSs in two seasons. Compared with other parameters Σdr 680–760 nm was the most sensitive parameter for detecting powdery mildew. Linear regression models relating mildew severity to Σdr 680–760 nm were constructed at three GSs in two seasons for the two plant densities, demonstrating no significant difference in the slope estimates between the two plant densities at three GSs. Σdr 680–760 nm was correlated with grain yield at three GSs in two seasons. The accuracies of partial least square regression (PLSR) models were consistently higher than those of models based on Σdr 680760 nm for disease index and grain yield. PLSR can, therefore, provide more accurate estimation of disease index of wheat powdery mildew and grain yield using canopy reflectance.  相似文献

Using genomic in situ hybidization, among the common wheat cultivars produced in West Siberia (Siberian Research Institute of Agriculture, Omsk) with the involvement of the winter wheat cultivar Kavkaz carrying the wheat-rye 1RS.1BL translocation we identified three cultivars with this translocation: Omskaya 29, Omskaya 37, and Omskaya 38. The protein and crude gluten contents in the grain of these cultivars are equal to or exceed the levels observed in cultivars without the wheat-rye translocation. The common wheat cultivars carrying the wheat-rye translocation were evaluated in terms of resistance of plants reaching wax ripeness to leaf rust and powdery mildew in the natural field conditions. The cultivars Omskaya 37 and Omskaya 38 displayed a high field resistance to leaf rust and were resistant to a variable extent to powdery mildew. The cultivar Omskaya 29 was susceptible to leaf rust and powdery mildew pathogens. Importance of the selection direction and the role of the genetic background in developing common wheat cultivars carrying the wheat-rye translocation is discussed.  相似文献

Wheat powdery mildew, caused by Blumeria graminis f. sp. tritici (Bgt), is one of the most important wheat diseases worldwide. The basis for wheat powdery mildew resistance breeding consists of screening diversified host genetic resources with a range of races of the powdery mildew pathogen. Spelt wheat (Triticum aestivum ssp. spelta 2n = 6x = 42, AABBDD) is a close relative of common wheat (T. aestivum ssp. aestivum) and contains several known disease resistance genes, including Pm1d, Yr5, and Lr65. Here, we report the identification and mapping of a powdery mildew resistance gene in spelt wheat cultivar Hubel, which was introduced to China from Europe and is resistant to Chinese Bgt isolate E09 at the seedling stage. Genetic analysis of a recombinant inbred line population derived from a cross of Hubel and a susceptible early maturing mutant line indicated that Hubel possessed a recessive powdery mildew resistance gene (temporarily designated MlHubel). Markers linked to MlHubel were identified using bulked segregant analysis, simple sequence repeat, and expressed sequence tag-derived sequence tagged site methods. The linked markers were physically located on wheat chromosome 2D. Comparative genomic analysis indicated that the genetic interval covering MlHubel in wheat is highly colinear with the corresponding regions on Brachypodium distachyon chromosome 5 and Oryza sativa chromosome 4. Accordingly, the genetic map of MlHubel was established in comparison with B. distachyon 5L and O. sativa 4L, with the closest marker Xgwm265 being 0.4 cM from MlHubel. The identification of the recessive powdery mildew gene in spelt wheat suggests the potential of this accession along with its closely linked markers in breeding for resistance to powdery mildew.  相似文献

Wheat (Triticum aestivum L.) incurs significant yield losses from powdery mildew, a major fungal disease caused by Blumeria graminis f. sp. tritici (Bgt). enhanced disease resistance1 (EDR1) plays a negative role in the defense response against powdery mildew in Arabidopsis thaliana; however, the edr1 mutant does not show constitutively activated defense responses. This makes EDR1 an ideal target for approaches using new genome‐editing tools to improve resistance to powdery mildew. We cloned TaEDR1 from hexaploid wheat and found high similarity among the three homoeologs of EDR1. Knock‐down of TaEDR1 by virus‐induced gene silencing or RNA interference enhanced resistance to powdery mildew, indicating that TaEDR1 negatively regulates powdery mildew resistance in wheat. We used CRISPR/Cas9 technology to generate Taedr1 wheat plants by simultaneous modification of the three homoeologs of wheat EDR1. No off‐target mutations were detected in the Taedr1 mutant plants. The Taedr1 plants were resistant to powdery mildew and did not show mildew‐induced cell death. Our study represents the successful generation of a potentially valuable trait using genome‐editing technology in wheat and provides germplasm for disease resistance breeding.  相似文献

Disease resistance (R) gene, RPP13, plays an important role in the resistance of plants to pathogen infections; its function in resistance of wheat to powdery mildew remains unknown. In this study, a RNA-Seq technique was used to monitor expression of genes in susceptible wheat ‘Jing411’ and resistant near-isogenic line ‘BJ-1’ in response to powdery mildew infection. Overall, 413 differential expression genes were observed and identified as involved in disease resistance. RPP13 homologous gene on wheat chromosome 7D was preliminarily identified using the wheat 660K SNP chip. RPP13 was highly expressed in ‘BJ-1’ and encodes 1,027 amino acids, including CC, NB and LRR domain, termed TaRPP13-3. After inoculation with powdery mildew, expression of TaRPP13-3 in resistant wheat changed with time, but average expression was higher when compared to susceptible variety, thus indicating that TaRPP13-3 is involved in resistance to powdery mildew. Virus-induced gene silencing (VIGS) was used to inhibit expression of TaRPP13-3 in resistant parent ‘Brock’. Results indicated that silencing of TaRPP13-3 led to decreased disease resistance in ‘Brock’. Overall results of this study indicate that TaRPP13-3 gene is involved in the defence response of wheat to powdery mildew and plays a positive role in wheat powdery mildew interactions.  相似文献

Wheat powdery mildew is a severe foliar disease and causes significant yield losses in epidemic years. Breeding and using resistant cultivars is the most widely employed strategy to curb this disease. To identify and transfer powdery mildew resistance genes in wild emmer wheat accession TA1410 into common wheat, a resistant F3 line derived from the cross of TA1410 × durum wheat line Zhongyin1320 was crossed with common wheat cultivar Yangmai158. The homozygous resistant BC5F2 lines derived from the backcross with Yangmai158 exhibited susceptibility at seedling stage and conferred increasing resistance when the plants were closer to heading stage. In two segregating BC5F3 families investigated at heading stage, the segregation of the resistance fit a 3:1 ratio, suggesting that a single dominant gene controls the resistance. This resistance gene, designated HSM1, was mapped to the 0.6-cM Xmag5825.1–Xgwm344 interval on chromosome 7AL and co-segregated with Xrga-C3 and Xrga-C6. A mapping position comparison with other powdery mildew resistance genes on this chromosome suggested that HSM1 belongs to the Pm1 resistance gene cluster. HSM1 is a useful candidate gene for resistance breeding, particularly in winter-wheat growing areas.  相似文献

Powdery mildew, caused by Blumeria graminis f. sp. tritici is a major disease of wheat (Triticum aestivum L.) that can be controlled by resistance breeding. The CIMMYT bread wheat line Saar is known for its good level of partial and race non-specific resistance, and the aim of this study was to map QTLs for resistance to powdery mildew in a population of 113 recombinant inbred lines from a cross between Saar and the susceptible line Avocet. The population was tested over 2 years in field trials at two locations in southeastern Norway and once in Beijing, China. SSR markers were screened for association with powdery mildew resistance in a bulked segregant analysis, and linkage maps were created based on selected SSR markers and supplemented with DArT genotyping. The most important QTLs for powdery mildew resistance derived from Saar were located on chromosomes 7DS and 1BL and corresponded to the adult plant rust resistance loci Lr34/Yr18 and Lr46/Yr29. A major QTL was also located on 4BL with resistance contributed by Avocet. Additional QTLs were detected at 3AS and 5AL in the Norwegian testing environments and at 5BS in Beijing. The population was also tested for leaf rust (caused by Puccinia triticina) and stripe rust (caused by P. striiformis f. sp. tritici) resistance and leaf tip necrosis in Mexico. QTLs for these traits were detected on 7DS and 1BL at the same positions as the QTLs for powdery mildew resistance, and confirmed the presence of Lr34/Yr18 and Lr46/Yr29 in Saar. The powdery mildew resistance gene at the Lr34/Yr18 locus has recently been named Pm38. The powdery mildew resistance gene at the Lr46/Yr29 locus is designated as Pm39.  相似文献

CH1302是以来源于中间偃麦草的八倍体小偃麦TAI7047为桥梁亲本选育的高抗白粉病的小麦新品系,对白粉菌多个流行小种均表现出良好抗性。为了解其抗白粉病基因来源及其在染色体上的位置,对绵阳11×CH1302的F_1、F_2及F_(2∶3)家系进行了遗传分析,推断其抗白粉病基因可能来源于中间偃麦草,暂将其命名为PmCH1302。利用i Select 90K SNP芯片对抗、感病池进行扫描,发现位于2AL染色体上的多态性位点最多,为313个,占全部多态性位点的9.79%,且集中于2AL染色体100~105 c M和150~155 cM两个区域附近。在上述位点选取SSR标记,筛选出3对与Pm CH1302连锁的分子标记,Xwmc522、Xgwm356和Xgwm526,其中Xgwm356和Xgwm526位于Pm CH1302两侧,连锁距离分别为3.1 c M和7.8 cM。利用遗传图谱以及中国春缺体、双端体将PmCH1302定位于小麦2AL染色体上。进一步与位于2AL上的Pm4、Pm50比较发现,PmCH1302可能是位于2AL上的一个新基因或等位基因。  相似文献

鉴定了170份小麦近缘物种材料苗期对北京地区流行的小麦白粉菌小种的抗性表现,包括引自美国和欧洲的斯卑尔脱小麦81份,密穗小麦27份,中国的西藏半野生小麦4份,和引自 CIMMYT 的人工合成六倍体小麦58份。结果表明,3份斯卑尔脱小麦表现抗病,它们是瑞士品种 Hubel 和 Lueg 以及德国的原始品种69Z6.245(编号 PI348085)。人工合成六倍体小麦中有19份材料表现高抗至免疫。密穗小麦材料中有2份(即美国材料 DN-2263和 Coda)表现抗病。4份西藏半野生小麦苗期都不抗小麦白粉病。  相似文献

Question: Can recent satellite imagery of coarse spatial resolution support forest cover assessment and mapping at the regional level? Location: Continental southeast Asia. Methods: Forest cover mapping was based on digital classification of SPOT4‐VEGETATION satellite images of 1 km spatial resolution from the dry seasons 1998/1999 and 1999/2000. Following a geographical stratification, the spectral clusters were visually assigned to land cover classes. The forest classes were validated by an independent set of maps, derived from interpretation of satellite imagery of high spatial resolution (Landsat TM, 30 m). Forest area estimates from the regional forest cover map were compared to the forest figures of the FAO database. Results: The regional forest cover map displays 12 forest and land cover classes. The mapping of the region's deciduous and fragmented forest cover remained challenging. A high correlation was found between forest area estimates obtained from this map and from the Landsat TM derived maps. The regional and sub‐regional forest area estimates were close to those reported by FAO. Conclusion: SPOT4‐VEGETATION satellite imagery can be used for mapping consistently and uniformly the extent and distribution of the broad forest cover types at the regional scale. The new map can be considered as an update and improvement on existing regional forest cover maps.  相似文献

 Consensus linkage maps were constructed for all seven rye chromosomes using 12 basic RFLP maps. The maps presented contain a total of 413 markers. The number of markers per chromosome varies from 41 (chromosome 3R) to 83 (chromosome 1R). In addition to 374 RFLP and 24 isozyme markers 15 gene loci were incorporated, determining the traits reduced plant height, self fertility, male sterility restoration, vernalization response, resistance against powdery mildew, chlorophyll deficiency, hairy leaf sheath, hairy peduncle, waxy endosperm, waxless plant and absence of ligules. The maps presented allow the selection of markers for the fine mapping of certain regions of the rye genome. In terms of the known chromosomal rearrangements within the Triticeae its utilization can also be extended for mapping in wheat and barley.
