Changes in Salicylic and Abscisic Acid Contents during Heat Treatment and Their Effect on Thermotolerance of Grape Plants 总被引:1,自引:0,他引:1
Heat treatment (38°C) of young grape plants (Vitis vinifera L., cv. Jingxiu) or leaf spraying with 100 µM salicylic acid (SA) increased leaf thermotolerance. Both treatments led to an increase in ABA content and a decrease in lipoxygenase (LOX) activity. ABA content showed a drastic rise by one hour after treatments and then sharply declined while LOX activity continuously decreased. In the course of heat treatment of grape plants, endogenous SA level and phenylalanine ammonia-lyase activity rapidly increased during the first hour, then declined. Leaves from annual young grape plants (Vitis vinifera L. cv. Jingxiu) were used as experimental materials. The ultrastructural characteristics of mesophyll cells in chilling-treated plants after heat acclimation (HA) and in heat-treated plants after cold acclimation (CA) were observed and compared using transmission electron microscopy. The results showed that slight injury appeared in the ultrastructure of mesophyll cells after either HA (38℃ for 10 h) or CA (8℃ for 2.5 d), but the tolerance to subsequent extreme temperature stress was remarkably improved by HA or CA pretreatment. The increases in membrane permeability and malondialdehyde concentration under chilling (0℃) or heat (45℃) stress were markedly inhibited by HA or CA pretreatment. The mesophyll cells of plants not pretreated with HA were markedly damaged following chilling stress. The chloroplasts appeared irregular in shape, the arrangement of the stroma lamellae was disordered, and no starch granules were present. The cristae of the mitochondria were disrupted and became empty. The nucleus became irregular in shape and the nuclear membrane was digested. In contrast, the mesophyll cells of HA-pretreated plants maintained an intact ultrastructure under chilling stress. The mesophyll cells of control plants were also severely damaged under heat stress. The chloroplast became round in shape, the stroma lamellae became swollen, and the contents of vacuoles formed clumps. In the case of mitochondria of control plants subjected to heat stress, the outer envelope was digested and the cristae were disrupted and became many small vesicles. Compared with cellular organelles in control plants, those in CA plant cells always maintained an integrated state during whole heat stress, except for the chloroplasts, which became round in shape after 10 h heat stress. From these data, we suggest that the stability of mesophyll cells under chilling stress can be increased by HA pretreatment. Similarly, CA pretreatment can protect chloroplasts, mitochondria, and the nucleus against subsequent heat stress; thus, the thermoresistance of grape seedlings was improved. The results obtained in the present study are the first, to our knowledge, to offered cytological evidence of cross-adaptation to temperature stresses in grape plants. 相似文献

以抗性葡萄品种‘F-242’组培苗为材料, 利用同源克隆法克隆了葡萄VvBAP1基因。测序结果显示, VvBAP1扩增片段大小为531 bp, 可编码176个氨基酸序列。利用生物信息学分析VvBAP1基因编码的蛋白序列显示, 该蛋白分子量为19.43 kDa, 含有保守的钙离子依赖性的C2结构域; 等电点pI为9.42; 不稳定系数为37.09, 推测为稳定的亲水性蛋白; 含有多个丝氨酸/苏氨酸磷酸化位点。实时荧光定量PCR表明, 该基因在根茎叶中均有表达, 其中在叶片中表达量较高; 盐胁迫、低温等逆境因子及逆境相关的信号物质, 如水杨酸和一氧化氮均可诱导VvBAP1的表达, 其中低温对其表达量影响更为显著, 推测该基因参与了葡萄抵御逆境胁迫的过程, 尤其是与低温相关的过程。 相似文献

以巨峰葡萄果皮为外植体,在添加2.0 mg/L 6-苄基嘌呤(6-BA)和0.1 mg/L 2,4-二氯苯氧基(2,4-D)的B5培养基上诱导葡萄愈伤组织; 以50 g/L的初始接种量在添加1.0 mg/L 6-BA和0.05 mg/L 2,4-D的B5液体培养基上建立葡萄悬浮培养体系。在25~27 ℃下,摇床振荡暗培养(120~130 r/min)18 d后,葡萄细胞生物量和白藜芦醇含量达到最大值(16.17 g/L、95.69 μg/g干质量)。在培养第12天时,向培养基中添加100 μmol/L茉莉酸甲酯(MeJA),经过6 d处理,细胞中白藜芦醇含量达235.73 μg/g干质量。 相似文献

A portable leaf greenness meter (SPAD-501) has been used to provide a rapid and non-destructive measurement of leaf chlorophyll in Vitis vinifera. Leaf extracted chlorophyll was related linearly to SPAD readings. It is suggested that separate linear equations should be developed for each cultivar so as to maximise the accuracy of estimating leaf chlorophyll content as a function of SPAD readings. 相似文献

We present here characterization data for seven new microsatellite markers designed from new microsatellite loci isolated from a microsatellite‐enriched DNA library from Vitis vinifera. The observed heterozygosity varied from 0.73 up to 0.93 and the number of alleles per locus ranged from 12 to 26. This high polymorphism makes these new markers interesting for use in genotyping studies and completing the set of microsatellite markers already available for V. vinifera. Additionally these seven new markers appear to be conserved in four other Vitis species and 15 Vitis hybrids used as rootstocks for V. vinifera cultivation. 相似文献

以葡萄品种‘左优红’组培苗叶片为材料,利用同源克隆法获得其病程相关蛋白1基因VvPR1的cDNA全长序列。扩增片段大小为486bp,编码161个氨基酸,分子量17.5kDa,等电点PI=8.69,含有6个保守半胱氨酸,4个allergenV5/Tpx-1related保守结构域。VvPR1与多种植物PR1高度同源。实时定量PCR检测结果表明VvPR1在葡萄叶片中相对表达量最高;霜霉病菌、低温、盐和干旱胁迫均可显著诱导其表达;水杨酸、脱落酸、茉莉酸、一氧化氮、过氧化氢和硫化氢等亦可诱导其大量表达,据此推测,VvPR1参与了多种生物胁迫和非生物胁迫过程。 相似文献

Somatic embryogenesis was observed in callus initiated from tendril explants of Vitis vinifera L. cvs. Thompson, Sonaka and Tas-e-Ganesh on Emershad and Ramming medium supplemented with 1 μm 6-benzylaminopurine. Low-frequency conversion to shoots was obtained in the third and fourth subculture on the same medium.
Emerging shoots subsequently formed complete plantlets on liquid rooting medium containing 1 μm indole-3-acetic acid. The possible use of tendrils as a novel explant for somatic embryogenesis in grape is discussed.

采用3种需冷量估算模型(≤7.2℃模型、0~7.2℃模型和犹他模型)和2种需热量估算模型(生长度小时模型和有效积温模型)分别测定了14个设施葡萄品种的需冷量和需热量,并分析了两者的相互关系。结果表明,不同葡萄品种的需冷量和需热量存在一定差异,需冷量值介于754~1489h(≤7_2℃模型)、497-757h(0~7.2℃模型)或192~755.5CU(犹他模型),需热量值为18491~24070GDH℃(生长度小时模型)或120~377D℃(有效积温模型),且欧亚种品种需热量普遍高于欧美杂种品种。另外,本研究还表明,用生长度小时模型估算出来的需热量值与需冷量值之间呈负相关关系,而有效积温模型估算出来的需热量值与需冷量值之间则呈正相关关系。 相似文献

Sucrose was found to modulate polyphenol accumulation in Vitis vinifera cell cultures. The production of anthocyanins increased 12-fold after addition of 0.15 m sucrose, while that of stilbenes was only slightly affected. Sucrose did not play a physical role because metabolic sugars
were required for the induction of polyphenol accumulation. Indeed, the polyols, mannitol and sorbitol, had no effect on this
accumulation. We established a model system to investigate the mechanism of sucrose regulation of polyphenol production without
inhibition of grape cell growth. After addition of sucrose to the culture medium, the major sugars accumulated in grape cells
were glucose and fructose, reaching 40% of the dry weight. The increase in the level of these hexoses closely coincided with
the increase in anthocyanin accumulation in grape cells.

Here the effect of jasmonic acid, methyljasmonate and Na-orthovanadate on the production of resveratrol was studied in Vitis vinifera cv. Barbera cell suspension cultures. Na-orthovanadate at 0.1 mm and 1 mm concentration was efficient in promoting the production and/or accumulation and release in the culture medium of cis-resveratrol while trans-resveratrol levels were not affected by this treatment. Methyljasmonate was highly effective in stimulating both trans- and cis-resveratrol endogenous accumulation, as well as their release into the culture medium. Cis-resveratrol was absent or detected in very low amounts in the controls. Jasmonic acid was less efficient than methyljasmonate in promoting endogenous resveratrol accumulation, but it stimulated the release in the culture medium especially of cis-resveratrol. Gel analysis was performed on control and 10 microm MeJA treated cell suspensions. Results showed an up-regulation of the stilbene synthase demonstrating that MeJA stimulated the synthesis ex-novo of this protein. 相似文献

A comparative analysis of differentially expressed proteins in a susceptible grapevine (Vitis vinifera ‘Cabernet Sauvignon’) during the infection of Erysiphe necator, the causal pathogen of grapevine powdery mildew (PM), was conducted using iTRAQ. The quantitative labeling analysis revealed 63 proteins that significantly changed in abundance at 24, 36, 48, and 72 h post inoculation with powdery mildew conidiospores. The functional classification of the PM‐responsive proteins showed that they are involved in photosynthesis, metabolism, disease/defense, protein destination, and protein synthesis. A number of the proteins induced in grapevine in response to E. necator are associated with the plant defense response, suggesting that PM‐susceptible Cabernet Sauvignon is able to initiate a basal defense but unable to restrict fungal growth or slow down disease progression. 相似文献

苯丙氨酸前体饲喂分别和环糊精、葡聚糖、茉莉酸甲酯、黑曲霉和直喙镰孢菌提取液五种诱导子联合作用,其中以与茉莉酸甲酯的联合作用对葡萄细胞培养生产花青素的影响最大,可使单位鲜细胞花青素含量提高2.7倍,花青素产量提高3.4倍,实验证明两者在培养后第4天加入效果最好。在30μmol/L苯丙氨酸、218μmol/L茉莉酸甲酯和3000~4000lx光照条件下,不同花青素产量的细胞株都能显著提高花青素产量,但低产株VV06比高产株VV05具有更大的产率提高潜力。该条件下VV05和VV06花青素产量分别达到2975和4090CV/L,是对照组的2.5倍和5.2倍。 相似文献

采用生物信息学方法从葡萄(Vitis vinifera Linn.)全基因组中鉴定Actin基因家族,并对各基因的染色体定位和结构特征,编码蛋白质的理化性质、亚细胞定位、二级结构、三级结构和系统进化,以及不同组织的基因表达进行研究.结果表明:葡萄Actin基因家族16个基因分布在12条染色体上.16个基因的结构特征及其编码蛋白质的理化性质差异较大.16个基因的长度及其内含子总长度的变化范围较大,编码序列(CDS)和外显子总长度的变化范围较小.除登录号GSVIVG01008254001和GSVIVG01014035001的基因外,其他14个基因的GC含量均低于其CDS的GC含量.除登录号GSVIVG01008254001的基因外,其他15个基因编码的蛋白质的理论相对分子质量为12534.54~82612.33,理论等电点为pI 4.92~pI 9.13.16个基因编码蛋白质的消光系数为14105~73645,脂肪族氨基酸指数为65.54~92.06,其中9个为稳定蛋白,7个为不稳定蛋白.除登录号GSVIVG01014035001的基因外,其他15个基因编码的蛋白质均为亲水性蛋白.登录号GSVIVG01016517001的基因编码的蛋白质定位于细胞质和细胞核,其他15个基因编码的蛋白质定位于细胞质.二级结构和三级结构显示:葡萄Actin基因家族16个基因编码的蛋白质均由α螺旋、无规则卷曲和延伸链构成,且总体以无规则卷曲为主.系统进化分析和不同组织的基因表达分析结果显示:与拟南芥〔Arabidopsis thaliana(Linn.)Heynh.〕相似,葡萄Actin基因家族16个基因编码的蛋白质分为3个亚家族,ClassⅡ亚家族(营养型)包括登录号GSVIVG01003099001和GSVIVG01026580001的基因编码的蛋白质,这2个基因在所有组织中的表达均较高;ClassⅢ亚家族(生殖型)包括登录号GSVIVG01033494001、GSVIVG01024980001和GSVIVG01016550001的基因编码的蛋白质,这3个基因在花粉、雄蕊和花中的表达均较高;ClassⅠ亚家族包括其他11个基因编码的蛋白质,这11个基因在各组织中的表达总体上较低.研究结果显示:葡萄Actin基因家族的表达具有组织特异性. 相似文献

目的:从葡萄中克隆白藜芦醇合酶基因vrs1并对其序列进行生物信息学分析.方法:利用葡萄总RNA为模板,采用RT - PCR技术克隆白藜芦醇合酶基因vrs1并亚克隆进T- Vector.利用生物信息学工具对其核酸和蛋白序列进行分析.结果:测序结果显示其cDNA序列全长为1 257bp,含有一个1 179bp的开放阅读框.生物信息学分析表明葡萄白藜芦醇合酶基因编码392个氨基酸,分子量为42.9kDa,理论等电点为5.97,具有芪合酶家族固有的氨基酸保守结构域,二级结构主要由α-螺旋、无规则卷曲、延伸链和β-转角组成.结论:该基因的克隆、生物信息学分析为进一步研究其功能奠定了基础. 相似文献

Abstract Using light and electron microscopy, we have studied the microsporogenesis and tapetal development of the feathers in two different low producing clones of Picolit giallo (sp. Vitis vinifera). In these clones while the productivity of the main branches (fertile branches originated from buds, formed in the previous year, that remained silent during the winter) is very low, that of the feathers (fertile branches derived from annual buds) is always normal. The microsporogenesis and tapetal development proceed normally in almost all the examined anthers; it is remarkable that at the tetrad stage the tapetal cells appear well structured without any degeneration symptom, unlike what observed for the main branches. Moreover in most of the mature anthers the pollen grains are numerous, pleinty of organelles and show sometimes thickenings in the callose layer under their wall. The tapetal cells of these anthers have disappeared. Only in few anthers we observed the presence of collapsed pollen grains and tapetal cells with anomalous development, that are still present when the pollen grains are mature. This rare situation for the feathers is on the contrary frequent for the main branches. 相似文献

In this study, the differences in the aroma compounds released after the free-run and pressed juices of cv. Emir grape (Vitis vinifera L.) were evaluated. Aroma compounds were obtained by liquid-liquid extraction with CH(2) Cl(2) , and then analyzed by gas chromatography (GC) and gas chromatography/mass spectrometry (GC/MS). According to the results, pressing uniformly increased the levels of the aromatic constituents, but this treatment lowered the grape juice quality for winemaking by increasing the total phenolic compounds, browning index, and C(6) -alcohol levels (green-herbaceaous odor) as compared to the free-run juice. From all the aroma compounds identified in both juices, hexan-1-ol, (E)-hex-2-en-1-ol, isobutanol, isoamyl alcohol, and 2-phenylethanol were the most abundant volatile compounds. 相似文献
