Bait formulations containing boric acid, chlorpyrifos, or hydramethylnon were evaluated in field and laboratory experiments against the German cockroach, Blattella germanica (L.). In continuous exposure tests, chlorpyrifos formulations produced 100% mortality in less than 24 h, boric acid formulations had LT50's of approximately 5.5 d, and hydramethylnon had an LT50 of 0.86 d. LT50's estimated in Ebeling choice boxes were 1.6 to 10.8 times greater than those from continuous exposure tests for chlorpyrifos formulations and 2.5 times greater for the hydramethylnon formulation. The same LT50's were produced by choice box and continuous exposure tests for boric acid formulations. Relative repellency, measured as the mean percentage of live cockroaches in the light side of the choice box, was greatest for chlorpyrifos formulations, followed by boric acid formulations, then hydramethylnon. A performance index (PI) that combined choice box repellency and mortality data indicated that hydramethylnon (Combat) had the greatest potential for field effectiveness. Only hydramethylnon significantly reduced German cockroach populations in the field. Apartments treated with the IT WORKS formulation of boric acid bait had significantly greater cockroach trap catches. The PI is an accurate predictor of the field performance of toxic baits for German cockroach control.  相似文献

The effect of topically applied chlorpyrifos on acetylcholinesterase and other esterases in heads and decapitated bodies of CSMA and Crawford German cockroaches was examined with spectrophotometric enzyme assay and native polyacrylamide gel electrophoresis. The toxicity of chlorpyrifos was greatly reduced in decapitated CSMA male cockroaches with LD50 value 17.1-fold higher than that of normal CSMA cockroaches. Acetylcholinesterase activity from heads was significantly higher in the Crawford compared with the CSMA strain and did not change until 24 h after chlorpyrifos in vivo treatment in both strains. The p-nitrophenyl butyrate (NPB) esterase activities from both heads and decapitated bodies of the resistant Crawford strain were significantly greater than the susceptible CSMA strain. The p-NPB esterase activity was significantly inhibited by chlorpyrifos in vivo treatment, and total p-NPB esterase activity was significantly reduced in decapitated bodies compared with heads of both strains. Native polyacrylamide gel electrophoresis (PAGE) analysis of extracts solubilized with Triton X-100 from heads and decapitated bodies revealed five major esterase bands and an acetylcholinesterase (AChE) band with a high capability of hydrolyzing alpha-naphthyl butyrate and acetylthiocholine, respectively. In the heads of susceptible CSMA male cockroaches, the activity of mobile isozymes d1 and d2 was completely inhibited at 24 h after chlorpyrifos application, and isozyme e was partially inhibited. In contrast, isozymes c1 and c2 from the decapitated bodies of CSMA cockroaches were mostly affected at 24 h after the topical application of chlorpyrifos. The activities of acetylcholinesterase and esterase isozymes a and b from the decapitated body remained uninhibited in both strains. Inhibition of isozymes d1 and d2 seems to be more important in chlorpyrifos intoxication than acetylcholinesterase.  相似文献

We have identified resistance mechanisms in the German cockroach, Blattella germanica (L.), for propoxur and chlorpyrifos in strains of cockroaches that display multiresistance to several organophosphate and carbamate insecticides. The resistance mechanisms involve the combined effects of increased oxidative and hydrolytic metabolism and both strains are resistant to chlorpyrifos and propoxur. Experiments designed to test for similarity in metabolic enzymes suggest that, although the mechanisms involve similar processes, the enzymes responsible for insecticide detoxification are different in the two strains. Both resistant strains exhibited enhanced activity toward alpha-naphtholic esters relative to a standard susceptible strain; however, analysis of the progeny from resistant X susceptible crosses suggests that this general esterase activity is inherited differently than propoxur or chlorpyrifos resistance. Hybrids of the propoxur-resistant strain displayed the highest activity of all cockroaches tested, in contrast to hybrids of the chlorpyrifos-resistant strain, which were similar to the susceptible strain. Native gel electrophoresis of cytosolic preparations provided further evidence for differences in the pattern of hydrolytic enzymes and inheritance of resistance in the two strains. Analysis of components of the cytochrome P450-dependent monooxygenase system and activities toward model substrates indicate that the two resistance mechanisms also involve different oxidative processes. The propoxur-resistant strain displayed significantly higher levels of total cytochrome P450, but no other components were correlated with resistance. In contrast with the chlopyrifos-resistant strain, which was similar to the susceptible strain in all parameters measured, activity toward model substrates was higher in the propoxur-resistant strain than in any of the other strains and hybrids tested.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Forty-five field-collected strains of German cockroaches, Blattella germanica were tested for resistance to 12 different insecticides by the time-mortality response method in comparison with a known susceptible strain. Only low to moderate resistance to diazinon, chlorpyrifos, and acephate was detected. Resistance to malathion was widespread; about half of the strains tested showed high resistance. High resistance to the carbamates propoxur and bendiocarb also occurred. High resistance was uncommon with propoxur, but about 35 strains were highly resistant to bendiocarb. High resistance to pyrethrins was observed in about of the strains tested. Resistance to the pyrethroids allethrin, permethrin, phenothrin, fenvalerate, and cyfluthrin was detected in some of the strains examined. All of the strains tested were susceptible to one or more of the insecticides used. These results indicate that, although resistance is a serious problem in this species, satisfactory control should be possible by selection of an appropriate insecticide.  相似文献

Three strains of German cockroach, Blattella germanica (L.) showed varying levels of resistance to chlorpyrifos, methyl parathion, propoxur, bendiocarb, and cypermethrin. The general esterase activity was at least twofold higher than susceptible strain. The subcellular distribution studies revealed that the majority of the esterase activity is present in the 100,000g cytosolic fraction. Only a small portion of the activity was membrane bound. Using non-denaturing gel electrophoresis, ten isozymes were identified in German cockroaches. These isozymes were isolated individually from the gels and analyzed for differences in activity. The isozymes E5, E6, and E7 of resistant strains had significantly higher specific activities when compared with the susceptible strain. The purification process using various column chromatography and preparative gel electrophoresis resulted in 9–11% of total esterase recovery. About double the amount of E6 was recovered from the resistant strains when compared with the susceptible strain. Kinetic analyses of E6 did not indicate differences in Km and Vmax values between the resistant and susceptible strains. Also, inhibition of esterase activity by paraoxon, chlorpyrifos, and propoxur did not suggest any structural differences in esterase E6 between strains. The results suggest that the increased production of E6 esterase contributes to insecticide resistance in German cockroaches. The physiological susceptibility to insecticides and the behavioral responses of four wild‐caught populations of female Anopheles epiroticus to synthetic pyrethroids (deltamethrin, permethrin, and alpha‐cypermethrin) were assessed. Test populations were collected from different localities along the eastern coast, Trat (TR), Songkhla (SK), and Surat Thani (ST) and one population from the western coast, Phang Nga (PN). Results showed that all four populations of An. epiroticus were susceptible to all three synthetic pyrethroids tested. Behavioral responses to test compounds were characterized for all four populations using an excito‐repellency test system. TR displayed the strongest contact excitation (‘irritancy’) escape response (76.8% exposed to deltamethrin, 74.1% permethrin, and 78.4% alpha‐cypermethrin), followed by the PN population (24.4% deltamethrin, 35% permethrin, and 34.4% for alpha‐cypermethrin) by rapidly escaping test chambers after direct contact with surfaces treated with each active ingredient compared with match‐paired untreated controls. Moderate non‐contact repellency responses to all three compounds were observed in the TR population but were comparatively weaker than paired contact tests. Few mosquitoes from the SK and ST populations escaped from test chambers, regardless of insecticide tested or type of trial. We conclude that contact excitation was a major behavioral response in two populations of An. epiroticus, whereas two other populations showed virtually no escape response following exposure to the three pyrethroids. The explanation for these large unexpected differences in avoidance responses between pyrethroid‐susceptible populations of the same species is unclear and warrants further investigation.  相似文献

Toxicity of bendiocarb, chlorpyrifos, cyfluthrin, cypermethrin, fenvalerate, hydramethylnon, malathion, propetamphos, propoxur, and pyrethrins against the adult German cockroaches, Blattella germanica (L.), was investigated. At LD50, cyfluthrin was the most toxic insecticide to adult males (0.53 microgram/g), adult females (1.2 micrograms/g), and gravid females (0.85 microgram/g). Malathion was the least toxic insecticide to adult males (464.83 micrograms/g), adult females (335.83 micrograms/g), and gravid females (275.90 micrograms/g). Males and gravid females were generally more sensitive than nongravid females to the insecticides that we tested. In tests with malathion, however, males were more tolerant. The order of toxicity of the insecticide classes varied among the stages of adult German cockroaches. The order of toxicity for males and nongravid females was pyrethroids greater than pyrethrins = organophosphates (except malathion) greater than carbamates = amidinohydrazone. The order of toxicity for gravid females was pyrethroids greater than pyrethrins = organophosphates (except malathion) greater than carbamates greater than amidinohydrazone. These differences in toxicity suggest that sex differences should be considered when determining insecticide toxicity for German cockroaches.  相似文献

Laboratory selection with azinphosmethyl had little effect on the chemical resistance in Cacopsylla pyri strains, in comparison to the original wild populations. The resistance ratio relative to a susceptible strain varied from 10- to 40-fold depending on the generation studied. Crosses between two resistant strains and the susceptible strain show resistance to be autosomally inherited and semi-dominant in expression. Backcrosses between F1 and the susceptible strain were unable to distinguish unabigously between monogenic and polygenic inheritance. In the majority of experiments, however, the overall dose-response relationship for backcross progeny was consistent with a single gene hypothesis. Additional bioassays showed azinphosmethyl-resistant strain to cross-resistant monocrotophos and phosmet, but not carbamates, pyrethroids, amitraz or the organophosphates chlorpyrifos and mevinphos.  相似文献

Late-instar German cockroaches from an insecticide-susceptible field strain (Fairbanks) and four pyrethroid-resistant field strains were exposed to vapors of a cyfluthrin flowable concentrate (FC) and the formulation blank (formulation without the active ingredient). Experiments with other pyrethroid formulations were done with the Fairbanks strain and an insecticide-resistant field strain (K-851) because these strains exemplified the maximum interstrain variation in experiments with the FC. In these experiments, I used a cyfluthrin wettable powder (WP) and its blank, a cyfluthrin emulsifiable concentrate (EC), and a permethrin aerosol and its blank. Vapors of the FC and its blank caused the most rapid dispersal. Dispersal induced by vapors of the WP, EC, and aerosol formulations was low and variable. In general, less of a dispersal response occurred in the resistant strains than in the susceptible strain. The strain mostly highly resistant to pyrethroids (K-851) dispersed slowly compared with the other resistant strains in experiments with the FC and its blank. When vapor-induced dispersal was relatively low (less than 50%), a trend was still evident towards less dispersal in the K-851 strain than in the Fairbanks strain. Although dispersal in experiments with the FC did not differ significantly from that in experiments with the FC formulation blank, mean percentage dispersal at the end of the experiments was higher with the formulation blanks than with the complete formulations in experiments with the WP and permethrin aerosol.  相似文献

Foraging cockroaches ingest insecticide baits, translocate them, and can cause mortality in untreated cockroaches that contact the foragers or ingest their excretions. Translocation of eight ingested baits by adult male Blattella germanica (L.) was examined in relation to the type of the active ingredient, formulation, and foraging area. Ingested boric acid, chlorpyrifos, fipronil, and hydramethylnon that were excreted by adults in small dishes killed 100% of first instars within 10 d and >50% of second instars within 14 d. Residues from these ingested baits were also highly effective on nymphs in larger arenas and killed 16-100% of the adults. However, when the baits and dead cockroaches were removed from the large arenas and replaced with new cockroaches, only residues of the slow-acting hydramethylnon killed most of the nymphs and adults, whereas residues of fast acting insecticides (chlorpyrifos and fipronil) killed fewer nymphs and adults. Excretions from cockroaches that ingested abamectin baits failed to cause significant mortality in cockroaches that contacted the residues. These results suggest that hydramethylnon is highly effective in these assays because cockroaches that feed on the bait have ample time to return to their shelter and defecate insecticide-laden feces. The relatively high concentration of hydramethylnon in the bait (2.15%) and its apparent stability in the digestive tract and feces probably contribute to the efficacy of hydramethylnon. To control for differences among baits in inert ingredients and the amount of active ingredient, we compared 1% chlorpyrifos with 1% hydramethylnon in identical baits. Again, hydramethylnon residues provided greater secondary kill, but the results highlighted the importance of the inert ingredients. We conclude that, in the absence of cannibalism and necrophagy, translocation of baits and secondary kill are most effective with slow acting insecticides in palatable baits that can traverse the digestive tract and be deposited within and around the cockroach aggregation.  相似文献

Crosses were made between the Asian cockroach,Blattella asahinai Mizukubo, and resistant strains of the German cockroach,B. germanica (L.), to assess the transfer of pyrethroid resistance to the progeny and to study the inheritance mechanism(s) involved. It was shown that the strain of Asian cockroaches studied was susceptible to four pyrethroids. F1 progeny were essentially susceptible to the same compounds. Tests with F2 progeny and those from backcrosses to the resistant parent indicated that the data for each pyrethroid fit an hypothesis of simple, autosomal, nearly completely recessive inheritance. The results are discussed from the standpoint of the impact of the Asian genome on the inheritance mechanism(s).  相似文献

A soil-less bioassay arena to test repellency of wireworms (A. obscurus) to insecticides and carrier solvents is described. The bioassay and variables measured distinguish between shorter-range (contact and/or volatile) and longer-range (volatile) repellency. Wireworm positions are recorded every 3 s for 20 min, and average speed, rate of slowing, and longer- and shorter-range repellent behaviors calculated. Shorter-range repellency is determined with a Wireworm Repellency Score (WRS, range 0-100), calculated before contact and after contact with test chemicals. Of two carrier solvents tested, wireworms were strongly repelled by acetone (WRS = 57) but not by water (WRS = 1) when introduced to the bioassay arenas immediately after chemical inoculation. When bioassay arenas were assembled/sealed 2 min after inoculation, acetone elicited no repellency (WRS = 2). When dissolved in acetone in bioassays assembled with a 2-min delay, imidacloprid, chlorpyrifos, lindane, and tefluthrin elicited slight to moderate repellency at the highest concentrations tested (WRS = 30, 48, 42, and 49, respectively). Both longer- and shorter-range repellency increased over the duration of the observation period for lindane and tefluthrin, and shorter-range repellency also increased over the duration of the observation period for chlorpyrifos. Removal of volatiles in the bioassay arena by vacuum considerably affected wireworm movement in the arena, with the repellency elicited by acetone and lindane being significantly reduced. Clothianidin elicited no longer- or shorter-range repellency.  相似文献

Resistance to insecticides of larval Culex pipiens populations in Israel has been monitored for ten years and the results were used for control planning. The insecticides tested were the organophosphates chlorpyrifos, fenthion, and temephos and the pyrethroids permethrin and cypermethrin. Over the years the relative resistance (R/R) values to chlorpyrifos in most populations tested were between 200-400, with records of up to 700 R/R. Practically no susceptible populations were found and the compound has been withdrawn from use, resulting in a decrease in the relative resistance values. In 1996 when fenthion was reintroduced, no resistant populations were found, but in the following years significant resistance appeared in an increasing number of populations with values lower than recorded for chlorpyrifos but still high (up to 100 R/R). All populations tested were found to be susceptible to temephos and accordingly the compound was reintroduced in 2002. Over the ten years, no significant resistance to cypermethrin was found in the majority of the populations tested, while a trend of increased resistance to permethrin was noted with a few records of >1000 R/R. The results demonstrate the importance of continuous monitoring of insecticide resistance for efficient mosquito control. Alternative insecticides and methods of control are discussed.  相似文献

Pyrethroid resistant and susceptible adults ofHelicoverpa armigera (Lepidoptera: Noctuidae) were screened for tolerance to pyrethroids after 6 wk or 12 wk pupal diapuse. Resistant larvae and F2 larvae from a cross between resistant and susceptible parents (two replicates), were reared under conditions to induce pupal diapause. After eclosion, adults were tested in glass vials coated with the pyrethroid fenvalerate, at a dose (DD) that is known to discriminate between susceptible and heterozygous resistant individuals. In all diapause experiments, the frequency of resistance was lower in the test groups that had experienced diapause compared with the non-diapausing control group. The underlying cause of the decline is not certain but selective mortality of resistant versus susceptible individuals could not account for all the difference in two of the three experiments — tolerance to the pyrethroid, fenvalerate, is most likely to have declined either as a consequence of diapause or from the extended time of development associated with diapause. These results indicate that monitoring programs could underestimate pyrethroid resistance frequencies when usingH. armigera adults emerging from diapause.  相似文献

Laboratory toxicity bioassays using chlorpyrifos (Dursban) confirmed the notion that development of resistance is responsible for widespread failures to control the California red scale, Aonidiella aurantii (Mask.) by applying organophosphorus (OP) compounds in citrus groves in Israel. Higher Vmax values of acetylcholinesterase (AChE) activity (9–13 fold) were measured in resistant strains collected from the field as compared to a susceptible line. No differences were found with respect to Km values using acetylthiocholine iodide as a substrate, or degree of inhibition (expressed by IC50 values) by the OP compounds chlorpyrifos-oxon and paraoxon and the carbamate pirimicarb. We suggest that resistance of the California red scale is caused by excess of AChE molecules able to bind and thus scavenge inhibitory OP compounds. This scavenging mechanism related to AChE may be similar in other insect species where elevated levels of detoxifying esterases were implicated in conferring OP resistance.  相似文献

In response to reports of failures of sprays to control Plutella xylostella infestations in vegetable-growing areas around Adelaide in 1991, the extent of resistance to six insecticides in several South Australian populations of P. xylostella was determined. Populations sampled from vegetable crops in the Adelaide region were five- to 200-fold resistant to organophosphates (chlorpyrifos, methamidophos and mevinphos), eight- to 400-fold resistant to pyrethroids (esfenvalerate and permethrin) and six-fold resistant to the carbamate methomyl compared with a susceptible strain. Mevinphos and esfenvalerate were the most effective of the tested insecticides. These, together with Bacillus thuringiensis products, have continued until 1998 to be used almost exclusively for the control of P. xylostella in South Australian vegetable crops in the absence of any resistance management strategy.  相似文献

Responses of German cockroaches, Blattella germanica (L.) (Dictyoptera: Blattellidae), to microencapsulated (ME) formulations of six insecticides (bifenthrin, chlorpyrifos, cyfluthrin, deltamethrin, lambda-cyhalothrin, and permethrin) were compared with emulsifiable concentrates (EC) (chlorpyrifos, cyfluthrin, deltamethrin, and permethrin) or ready-to-use (RTU) formulations (bifenthin and lambda-cyhalothrin). Two rates were tested per comparison. Baseline toxicity (LT50 value) was determined by continuous exposure to residual deposits. Repellency, toxicity (LT50), and performance index (PI) values were determined using Ebeling choice boxes. Baseline toxicity of the permethrin formulations was similar, but all other active ingredients had significant toxicity differences at one or both formulation x dose comparisons. Baseline toxicity and repellency were negatively correlated. Choice box LT50 and the time to reach 50% of the maximum PI were positively correlated. The maximum PI was positively correlated (P < 0.06) with baseline LT50 and negatively correlated (P < 0.07) with repellency. Chlorpyrifos had the lowest repellency except for the EC at 0.25%. Bifenthrin ME and lambda-cyhalothrin ME had greater PI values than comparative RTU formulations. Cyfluthrin EC at 0.03% and deltamethrin ME at 0.01% had significantly lower PI values than comparison treatments. Permethrin PI value for the EC at 0.03% exceeded that for the ME, but at 0.05% the ME had a significantly greater PI. These data demonstrate the difficulty in making generalizations about the relative performance of ME compared with EC or RTU formulations. Variable results observed within, and between, formulations may be influenced by application rate, formulation type, other formulation components, and the toxicity-repellency of the active ingredient.  相似文献

ABSTRACT The laboratory studies were carried out for evaluating control effects of four commercial insecticidal baits such as two different hydramethylnon [2.0%(AI)] products (DBK® and Combat‐Gold®), fipronil [0.05 %(AI)] (Combat‐Power®) and (0.6% chlorpyrifos [0.6%(AI)] (Raid‐Roachbait®) against German cockroaches (Blattella germanica). The control rates of four kinds of toxic baits were all 100.0% mortality of German cockroaches in 5 days after treatment. The results of chlorpyrifos and fipronil brought 100.0% mortalities in 2 to 3 days after treatment, respectively. After 3 day treatment, there was no significant difference of control effect among the four toxic baits. As a result of this study, fipnonil and chlorpyrifos showed faster killing action against German cockroaches than the two hydramethylnon formulation products. In the choice test, DBK® (hydramethylnon) (average 17.0 ind.) significantly attracted more German cockroaches than Combat‐Gold® (hydramethylnon) (avg. 7.0 ind.), Combat‐Power® (fipronil) (avg. 5.3 ind.) and Raid‐Roachbait® (chlorpyrifos) (avg. 3.3 ind.). The difference in attraction effects came 10 minutes after treatment. In order to evaluate the secondary killing effect of toxic baits by coprophagy against adult Blattella germanica, the comparative test was carried out. The adult mortality rates were significantly different among the four toxic baits showing 86.7% mortality for fipronil, 60.0% for hydramethylnon (DBK®), 30.0% for chlorpyrifos, and below 13.3% for hydramethylnon (Combat‐Gold®) in 6 days. During the first 12 days, fipronil showed the highest mortality rate (90.0%), followed by hydramethylnon (DBK®) (60.0%). The secondary killing effect of toxic baits by coprophagy appeared in all of the baits against adult Blattella germanica.  相似文献

Current control methods for the black carpenter ant, Camponotus pennsylvanicus (De Geer), include the use of remedial and preventative residual sprays as well as toxic baits. We evaluated the acceptance of three baits (Maxforce, Niban, and Baygon) to field colonies of the black carpenter ant in the spring and fall. Maxforce bait granules were more readily accepted than either Niban or Baygon bait granules in the spring. A change in food preference from protein to sugar by the black carpenter ant appeared to reduce the number of Maxforce bait granules removed in the fall, resulting in no differences in bait acceptability. The longevity of Dursban 50W and Tempo 20WP were evaluated in the summer and fall on painted wood panels. Panels aged outside for 15 d under prevailing weather conditions exhibited increased LT50 values. For each sampling period, panels aged on the south face (in the sun) exhibited less insecticidal activity (i.e., large LT50 values) than panels on the north face (shaded; small LT50 values). At each sampling period, Tempo 20WP provided smaller LT50 values than Dursban 50W. Because of changing dietary preferences, our data highlight the importance of using various bait types for carpenter ant control. Moreover, the application of residual spays should be made to locations protected from direct sunlight.  相似文献

Thirty‐two Aedes aegypti populations collected throughout Thailand and five populations of Aedes albopictus from southern Thailand were subjected to standard WHO contact bioassays to assess susceptibility to three commonly used synthetic pyrethroids: permethrin, deltamethrin, and lambda‐cyhalothrin. A wide degree of physiological response to permethrin was detected in Ae. aegypti, ranging from 56.5% survival (Lampang, northern Thailand) to only 4% (Kalasin in northeastern and Phuket in southern Thailand). All 32 populations of Ae. aegypti were found to have evidence of incipient resistance (62.5%) or levels of survival deemed resistant (37.5%) to permethrin. Four populations of Ae. albopictus were found with incipient resistance (97 – 80% mortality) and one with resistance (< 80%) to permethrin. The majority of Ae. aegypti populations (68.7%) was susceptible (> 98% mortality) to deltamethrin, with incipient resistance (observed 97–82% mortality) in other localities. In contrast, all populations of Ae. aegypti were completely susceptible (100% mortality) to the recommended operational dosage of lambda‐cyhalothrin. All five populations of Ae. albopictus were found completely susceptible to both deltamethrin and lambda‐cyhalothrin. Evidence of defined incipient or resistance to synthetic pyrethroids mandates appropriate response and countermeasures to mitigate further development and spread of resistance. In light of these findings, we conclude that routine and comprehensive susceptibility monitoring of dengue mosquito vectors to synthetic pyrethroids should be a required component of resistance management policies and disease control activities.  相似文献
