Apparent transinhibition of peptide uptake in the scutellum of barley grain   总被引:1,自引:0,他引:1  
The uptake of glycylsarcosine (Gly-Sar) into scutella separated from germinating grains of barley ( Hordeum vulgare L. cv. Himalaya) is inhibited by other peptides; in most cases the inhibition is not purely competitive but of a mixed type (simultaneous increase in the apparent Km and decrease in Vmax) (Sopanen, T. 1979. FEBS Lett. 108: 447–450). The aim of the present experiments was to elucidate the mechanism of the mixed inhibition by studying how peptides already taken up into the cells affect the uptake of Gly-Sar.
When scutella were preincubated in the presence of various peptides, 11 of the 13 peptides tested inhibited the subsequent uptake of Gly-Sar by 10 to 45%. The inhibition, studied in detail with leucylleucine and prolylproline, was due to a decrease in Vmax. The two peptides having no effect were glycylglycine and D-alanyl-L-alanine which are the only peptides known to date acting as purely competitive inhibitors when present together with the substrate Gly-Sar.
Preincubation with leucine, proline and alanine was not inhibitory, although preincubation with the corresponding dipeptides was. This result, together with the demonstration of intact leucylleucine in the scutella after preincubation with leucylleucine, indicates that the inhibition was caused by the intact peptides.
The results support the notion that in the mixed type inhibition the increase in the apparent Km is due to competition for the carrier at the outside of the membrane, while the decrease in Vmax is due to peptides taken up and binding to the carrier at the inside of the membrane.  相似文献

Some factors that may be concerned in determining final grain weight in barley ( Hordeum vulgare L. var. distichum ) have been investigated. Variation in endosperm fresh and dry weight, volume and starch content have been recorded at different stages of grain development between anthesis and harvest-ripeness for two barleys, cvs Kym and Golden Promise, differing in final grain weight. Results were recorded under both field and glasshouse conditions. The results suggest that the higher final dry weight of Kym, in comparison with Golden Promise, is a function of both rate and duration of grain filling. Only at later stages of endosperm development did the differences in volume become significant and the Kym endosperms continued to increased rapidly in volume for two to three days after endosperm volume had reached a maximum in Golden Promise. The rates of starch accumulation in both cultivars were very similar but starch deposition continued in Kym endosperms for four to five days after deposition in Golden Promise endosperms had slowed down.  相似文献

Abstract Treatment of barley leaf sections with 0.1 mol m?3 thioproline (L-thiazolidine-4-carboxylic acid) was found to induce a marked increase in proline together with some decrease in glutamate, whereas the levels of other andno acids were not influenced. This result is discussed in relation to the significance of the increase in proline in tissues treated with abscisic acid or subjected to water stress.  相似文献

The functional implications of root cortical senescence (RCS) are poorly understood. We tested the hypotheses that RCS in barley (1) reduces the respiration and nutrient content of root tissue; (2) decreases radial water and nutrient transport; and (3) is accompanied by increased suberization to protect the stele. Genetic variation for RCS exists between modern germplasm and landraces. Nitrogen and phosphorus deficiency increased the rate of RCS. Maximal RCS, defined as the disappearance of the entire root cortex, reduced root nitrogen content by 66%, phosphorus content by 63% and respiration by 87% compared with root segments with no RCS. Roots with maximal RCS had 90, 92 and 84% less radial water, nitrate and phosphorus transport, respectively, compared with segments with no RCS. The onset of RCS coincided with 30% greater aliphatic suberin in the endodermis. These results support the hypothesis that RCS reduces root carbon and nutrient costs and may therefore have adaptive significance for soil resource acquisition. By reducing root respiration and nutrient content, RCS could permit greater root growth, soil resource acquisition and resource allocation to other plant processes. RCS merits investigation as a trait for improving the performance of barley, wheat, triticale and rye under edaphic stress.  相似文献

Barley (Hordeum vulgare L.) is a major cereal grain widely used for livestock feed, brewing malts and human food. Grain yield is the most important breeding target for genetic improvement and largely depends on optimal timing of flowering. Little is known about the allelic diversity of genes that underlie flowering time in domesticated barley, the genetic changes that have occurred during breeding, and their impact on yield and adaptation. Here, we report a comprehensive genomic assessment of a worldwide collection of 895 barley accessions based on the targeted resequencing of phenology genes. A versatile target‐capture method was used to detect genome‐wide polymorphisms in a panel of 174 flowering time‐related genes, chosen based on prior knowledge from barley, rice and Arabidopsis thaliana. Association studies identified novel polymorphisms that accounted for observed phenotypic variation in phenology and grain yield, and explained improvements in adaptation as a result of historical breeding of Australian barley cultivars. We found that 50% of genetic variants associated with grain yield, and 67% of the plant height variation was also associated with phenology. The precise identification of favourable alleles provides a genomic basis to improve barley yield traits and to enhance adaptation for specific production areas.  相似文献

The kinetics of proline accumulation in barley ( Hordeum vulgare L. cv. Georgie) leaf segments showed a lag phase of ca 3 h when the increase was induced by abscisic acid (ABA), but not when the accumulation of the imino acid was promoted by isobutyric acid (IBA). Cycloheximide (CHI) supplied together with ABA, either from the beginning of the treatment or some time before the end of the lag phase, completely abolished ABA-induced proline accumulation, whereas no block was observed when the inhibitor was supplied after the lag phase. Cordycepin (COR) exibited a similar effect. The IBA-induced increase in proline was not influenced by CHI for at least 5 h.
When segments were pretreated with ABA for a period longer than the lag phase in the absence of salts in the external medium, there was no significant increase in proline. If KCI was added to the incubation medium after such a pretreatment, however, proline increased even after removal of the hormone from the external medium. This increase in proline occurred without any lag phase, and was only partially inbibited by CHI and rapidly and totally blocked by fusicoccin (FC). These results suggest that some protein, characterized by a fast turnover and possibly conferring the sensitivity to KCI, is synthesized during the early hours (lag phase) of the ABA treatment.
The synthesis of this protein(s) does not seem to be involved in the increase in proline induced by IBA and is thus a peculiar aspect of that mediated by ABA.  相似文献

Barley is used for food and feed, and brewing. Nondormant seeds are required for malting, but the lack of dormancy can lead to preharvest sprouting (PHS), which is also undesired. Here, we report several new loci that modulate barley seed dormancy and PHS. Using genome‐wide association mapping of 184 spring barley genotypes, we identified four new, highly significant associations on chromosomes 1H, 3H, and 5H previously not associated with barley seed dormancy or PHS. A total of 71 responsible genes were found mostly related to flowering time and hormone signalling. A homolog of the well‐known Arabidopsis Delay of Germination 1 (DOG1) gene was annotated on the barley chromosome 3H. Unexpectedly, DOG1 appears to play only a minor role in barley seed dormancy. However, the gibberellin oxidase gene HvGA20ox1 contributed to dormancy alleviation, and another seven important loci changed significantly during after‐ripening. Furthermore, nitric oxide release correlated negatively with dormancy and shared 27 associations. Origin and growth environment affected seed dormancy and PHS more than did agronomic traits. Days to anthesis and maturity were shorter when seeds were produced under drier conditions, seeds were less dormant, and PHS increased, with a heritability of 0.57–0.80. The results are expected to be useful for crop improvement.  相似文献

Dehydrins are a group of plant proteins that usually accumulate in response to environmental stresses. They are proposed to play specific protective roles in plant cells. Present study showed that the accumulation of dehydrins in water-stressed barley (Hordeum vulgare L.) seedlings was influenced by their treatment with salicylic acid (SA). The level of dehydrin proteins was increased by 0.20 mM SA, but decreased by 0.50 mM SA treatment. Both mRNA expression and protein accumulation of a typical barley dehydrin, DHN5, were enhanced by SA treatment when SA concentrations were lower than 0.25 mM. However, the higher SA concentrations significantly decreased the protein level of DHN5 despite of a stable mRNA level. Our results also showed that low SA concentrations (less than 0.25 mM) decreased the electrolyte leakage and malondialdehyde (MDA) and H2O2 contents in water-stressed barley seedlings. But high SA concentrations (more than 0.25 mM) enhanced H2O2 accumulation, tended to cause more electrolyte leakage, and increase MDA content. These data indicated that SA could up-regulate the dehydrin gene expression and protein accumulation. Since the protective role of dehydrins in plant cells, such effect could be an important reason for the SA-mediated alleviation on water stress injury. The distribution and fluctuation of sugars in germinating barley seeds were examined by 13C nuclear magnetic resonance (NMR) spectroscopy, 1H-NMR imaging and 1H-NMR localized spectroscopy in relation to morphology. Maltose, sucrose, fructose and oils were detected in intact imbibed seeds by 13C-NMR spectra. During the first 6 d of germination, the maltose content increased and the oil content gradually decreased, whilst the levels of sucrose and fructose remained constant. Sugars were located by 1H-NMR images and 1H-NMR localized spectra in the vascular bundle of the seeds as well as in the solubilized endosperm. They were also detected in the shoots. The sugars detected in an 80% ethanol shoot extract were sucrose and glucose, which were located in the vascular bundles but not in the mesophyll cells of the coleoptile. They were also located in the basal part of the shoot, but not above 7 mm from the scutellum. The data suggest that the sugars are primarily transported through the vascular bundles and, at the same time, rapidly incorporated into mesophyll cells in the leaves.  相似文献

Barleygrass ( Hordeum leporinum ) from Australian low-P (phosphorus) soils and commercial barley ( H. vulgare ) with high fertilizer requirements were grown in solution culture at 3 levels of P supply. The high-P-adapted barley produced more biomass at all levels of P supply and was more responsive to added P in terms of rate of tillering, rate of leaf production, final leaf size, and therefore total shoot weight compared to barleygrass. In both species root: shoot ratio decreased in response to improved tissue P status, even at P levels where total biomass did not respond to P supply. Removal of endosperm reserves of barley reduced total biomass to a greater extent than it altered phosphate absorption rate, thus increasing tissue P status and making plants less responsive to added P. Similarly, barleygrass had a slower growth rate but a comparable P absorption rate to that of barley. Thus barleygrass also accumulated tissue P and was unresponsive to added P. All phosphorus chemical fractions increased in response to improved tissue P status, but to differing extents (inorganic-P > nucleic acid-P > lipid-P > ester-P), suggesting that all P fractions (particularly inorganic P) serve, in part, a storage function. Both barleygrass and barley without endosperm had higher concentrations of all P fractions (particularly inorganic P) than did unaltered barley, but this was due entirely to their higher P status (due to slow growth) rather than to any major difference in P metabolism between species. We conclude that slow growth is more important than interspecific differences in P metabolism, P absorption, or efficiency of P utilization in explaining the success of barleygrass and other low-P-adapted species on infertile soils.  相似文献

Roots perform vital roles for adaptation and productivity under water-deficit stress, even though their specific functions are poorly understood. In this study, the genetic control of the nodal-root architectural and anatomical response to water deficit were investigated among diverse spring barley accessions. Water deficit induced substantial variations in the nodal root traits. The cortical, stele, and total root cross-sectional areas of the main-shoot nodal roots decreased under water deficit, but increased in the tiller nodal roots. Root xylem density and arrested nodal roots increased under water deficit, with the formation of root suberization/lignification and large cortical aerenchyma. Genome-wide association study implicated 11 QTL intervals in the architectural and anatomical nodal root response to water deficit. Among them, three and four QTL intervals had strong effects across seasons and on both root architectural and anatomical traits, respectively. Genome-wide epistasis analysis revealed 44 epistatically interacting SNP loci. Further analyses showed that these QTL intervals contain important candidate genes, including ZIFL2, MATE, and PPIB, whose functions are shown to be related to the root adaptive response to water deprivation in plants. These results give novel insight into the genetic architectures of barley nodal root response to soil water deficit stress in the fields, and thus offer useful resources for root-targeted marker-assisted selection.  相似文献

The pattern and sequence of cellular degradation during the course of leaf senescence remains obscure and the nature of the trigger that induces cell senescence is unknown. In order to probe the pre-mortem phase of senescence temporal changes in cell ultrastucture were studied in aging leaves of light-grown achlorophyllous Hordeum vulgare L. cv. Dyan mutant seedlings. Electron microscope examination of the ultrastructure of mesophyll cell plastids revealed the absence of ribosomes and a highly disorganized prolamellar body. Both the number and size of plastoglobuli increased with aging and this change coincided with depletion of starch grains and dilation of lamellar membranes. Aging of mesophyll cells occurred coincident with a decline in ribosome content of the cytoplasm and loss of matrix granularity. Loss of ribosomes associated with the outer nuclear envelope membrane and a reduction in chromatin were also apparent. Only after 10 days was there evidence of loss of internal membrane integrity and swelling of mitochondrial cristae. Compartmentation was thus maintained during the aging process with membrane dissolution occurring late in senescence. These results suggest that an inability to produce chlorophyll and carotenoids and form thylakoid stacks due to the absence of plastid ribosomes, contributes to the rapid onset of senescence in light-grown achlorophyllous seedlings. Furthermore, disruption of chloroplast ribosome synthesis/assembly may constitute part of the plastid signal involved in triggering cell senescence.  相似文献

The role of ABA in freezing tolerance and cold acclimation in barley   总被引:4,自引:0,他引:4  
The role of ABA in freezing resistance in nonacclimated and cold‐acclimated barley ( Hordeum vulgare L.) was studied. Eleven nonacclimated cultivars differed in their LT50, ranging from −10.8 to −4.8°C. Sugars, free proline, soluble proteins and ABA were analyzed in nonacclimated cultivars and during cold acclimation of one cultivar. There was an inverse correlation between LT50 and both ABA and sucrose contents. Exogenous ABA caused a decrease in the freezing point of leaf tissue in the cultivar with the lowest level of endogenous ABA, but not in the cultivar with the highest level, suggesting that ABA in the latter may be near the optimum endogenous level to induce freezing tolerance. Plants of cv. Aramir treated with ABA or allowed to acclimate to cold temperature increased their soluble sugar content to a similar level. The LT50 of leaves of cold‐acclimated cv. Aramir decreased from −5.8 to −11.4°C, with biphasic kinetics, accumulating proline and soluble sugars with similar kinetics. The biphasic profile observed during cold acclimation could be a direct consequence of cryoprotectant accumulation kinetics. ABA and soluble protein accumulation showed a single step profile, associated mainly with the second phase of the LT50 decrease. Thus, a significant increase in endogenous ABA is part of the response of barley to low temperature and may be required as a signal for the second phase of cold acclimation. Endogenous ABA contents in the nonacclimated state may determine constitutive freezing tolerance.  相似文献

Abstract Salt-stressed plants often show Ca deficiency symptoms. The effects of NaCl salinity (1 to 150 mol m-3) and supplemental Ca (10 mol m-3) on Na and Ca transport in barley (Hordeum vulgare L.) and their relationship to growth were investigated. The adjustment of Na and Ca transport was investigated by examining young seedlings exposed to short-term (immediate) and long-term (7 d) exposure to salinity. When the plants were exposed to long-term treatments of salinity, the rate of sodium accumulation in roots was approximately 10 to 15% of short-term treatments. No significant adjustment in the transport to the shoot was observed. Rates of tracer (22Na) transport were compared to calculated rates based on relative growth rates and tissue element concentrations. Comparisons between measured tracer and calculated rates of transport indicate that 22Na transport may underestimate transport to the shoot because of dilution of the tracer in the root cytoplasm. Calcium uptake showed only minor adjustment with time. Measured rates of tracer transport to the shoot correlated well with calculated values. The transport and tissue concentrations of Na were significantly affected by supplemental Ca. Calcium transport and tissue concentrations were markedly inhibited by salinity. Supplemental Ca increased Ca transport and accumulation at all NaCl treatments above that of control plants without supplemental Ca. Salinity inhibited plant growth at 150 mol m -3NaCl, but not at 75 mol m-3. Supplemental Ca significantly improved root length but not fresh weight after 7d of salinity, although differences in fresh weight were detected after 9d. There were significant Na-Ca interactions with ion transport, ion accumulation, and growth. The effects of salinity on Na and Ca transport to the shoot do not appear to play a major role in shoot growth of barley.  相似文献

The Russian barley cultivar Nevsky lacks 3 hordein and accumulates most of its hordein in the lumen of the endoplasmic reticulum and only a minor portion in the vacuole. In wild type barley and all other temperate cereals, storage proteins are deposited in the vacuole. F1 crosses revealed that the Nevsky phenotype is recessive; but the extent of hordein accumulation in the endoplasmic reticulum in F2 endosperm lacking 3 hordein was very much less than in the Nevsky parent. In order to study the Nevsky endosperm phenotype we have measured the levels of seven proteins and two mRNAs involved in protein folding in the ER lumen or ER to Golgi transport during endosperm development. The protein levels were unaltered in Nevsky as compared to the wild-type variety Bomi. When the levels of these seven proteins were correlated with the rate of hordein accumulation, four of these (HSP70, PDI, Sar1p and Sec18p) were consistently up-regulated with hordein synthesis. Accumulation of hordein in the endoplasmic reticulum appears to be determined by the absence of 3 hordein, or the product of a gene closely linked to it, plus one or more other recessive genes.  相似文献

Heat shock in barley ( Hordeum vulgare L. cv. Himalaya) aleurone layers induces the synthesis of heat shock proteins (hsps) and suppresses the synthesis and secretion of α-amylase, the principal secretory protein. This is accompanied by the destabilization of α-amylase mRNA and a concomitant dissociation of ER lamellae. In the absence of heat shock α-amylase mRNA is extremely stable (Belanger et al. 1986. Proc. Natl. Acad. Sci. USA 83: 1354–1358). In most organisms there is a direct correlation between the synthesis of hsps and thermotolerance. The ability of hsps to provide thermoprotection to secretory protein synthesis, α-amylase mRNA and ER lamellae was analyzed. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) of pulse-chased, [35S]-methionine-labeled proteins revealed that the half-life of hsps in barley aleurone cells recovering from heat shock was approximately 12 h. Within approximately 6 h, there was a recovery of α-amylase mRNA and a reformation of ER lamellae. Heat shock protein synthesis was induced by either heat shock (40°C) or arsenite, the cells were allowed to recover for 8 h, then were re-exposed to heat shock. Results from SDS-PAGE showed that, despite the presence of hsps, α-amylase synthesis was suppressed. Northern blot hybridizations showed that α-amylase mRNA levels were reduced in heat-shocked tissues. Transmission electron microscopy demonstrated that ER lamellar structures were dissociated. The synthesis of hsps did not enable barley aleurone cells to sustain the synthesis of any proteins at lethal temperature. In contrast, similar conditions established thermotolerance and provided thermoprotection to protein synthesis in germinating barley embryos. Our findings suggest that the aleurone layer does not become thermotolerant following the induction of hsp synthesis.  相似文献

Abstract. Carbonylcyanide, m -chlorophenyl hydra-zone (CCCP) decreased the ATP content of barley and corn roots by 80% within 5 min. The protonophore inhibited K and Cl absorption by largely unvacuolated root tips, and vacuolated root segments of barley and corn. The protonophore also inhibited Na absorption by root segments and Na extrusion by root tips of barley; it did not affect these Na fluxes in corn root tips and segments, and Na Influx in barley root tips. It was concluded that corn roots lack a metabolic mechanism for Na extrusion from the cytoplasm to the external solution or vacuole, which is functional in barley roots.  相似文献

Although much investigated, the factors constraining cereal grain protein accumulation are not well understood. As a result of the development of a new technique, new approaches to this question are now possible. A peduncle perfusion system was used to deliver a range of plant growth regulators (PGRs) and/or N solutions into barley (Hordeum vulgare) plants during the grain-filling period. The perfusion technique floods the peduncle interior with a treatment solution for periods of weeks to months, allowing the plant to take up administered substances from the perfused solution. The objectives of the present work were to determine: (1) whether some PGRs could alter the overall pattern of N allocation within barley plants, perhaps leading to higher protein accumulation in the seeds, (2) whether the addition of N through the peduncle could increase the seed N concentration even when the concentration of N in the rooting medium was high, and (3) whether or not PGR-stimulated elevations in grain protein levels and peduncle-added N increases in grain protein levels were additive. Three experiments were conducted to determine the physiological effects of (1) peduncle-administered PGRs (2) combinations of soil- and peduncle-applied N and (3) selected combinations of soil- and peduncle-administered N, and peduncle-applied PGRs on photosynthetic rate, dry matter partitioning and N accumulation of barley plants during grain filling. The first experiment tested four PGRs: abscisic acid (ABA), kinetin, gibberellic acid (GA3), and 2,4-dichlorophenoxy acetic acid (2,4-D) each at three concentrations. The second experiment tested three levels of soil N (NH4NO3) fertility, and two concentrations of peduncle-added N (urea). The third experiment tested four PGRs: ABA, kinetin, GA3, and 2,4-D with two soil N concentrations and two concentrations of peduncle-added N. ABA and 2,4-D decreased total seed weight of the perfused spike. The addition of peduncle-perfused N increased seed protein concentration and content under conditions of high soil N fertility, suggesting that seed protein accumulation is more limited by the ability of roots to take up N from the soil than by the seed to take up N from the rest of the plant. The effects of the PGRs on N allocation among plant parts varied with the amount of N available to the plant. Because it resulted in less protein stored in the flag leaf and more in the seeds, GA3 perfusion caused an overall change in the allocation of N among plant parts. Peduncle perfusion of kinetin and ABA affected some aspects of photosynthetic physiology.  相似文献
