A1R-A2AR heterodimers regulate striatal glutamatergic neurotransmission. However, few researches about kinetics have been reported. Here, we combined Iem-spFRET and E-FRET to investigate the kinetics of A1R and A2AR interaction. Iem-spFRET obtains the energy transfer efficiency of the whole cell. E-FRET gets energy transfer efficiency with high spatial resolution, whereas, it was prone to biases because background was easily selected due to manual operation. To study the interaction with high spatio-temporal resolution, Iem-spFRET was used to correct the deviation of E-FRET. In this paper, A1R and A2AR interaction was monitored, and the changes of FRET efficiency of the whole or/and partial cell membrane were described. The results showed that activation of A1R or A2AR leads to rapid aggregation, inhibition of A1R or A2AR leads to slow segregation, and the interaction is reversible. These results demonstrated that combination of Iem-spFRET and E-FRET could measure A1R and A2AR interaction with high spatio-temporal resolution. 相似文献

Model glycopeptides of the general formula Boc-Ala-Thr(G-D)-A(1)-A(2)-Leu-Leu-Lys(N)-Ala-OMe, where D = dansyl (dimethyl aminonaphthalenesulphonyl), G = glucosyl and N = naphthyl, while A(1)-A(2) = Ala-Leu or Aib-Aib, and denoted as D-G-Ala-N and D-G-Aib-N, respectively, were used to investigate glycoprotein-membrane interactions. They carry two fluorophores (D and N), covalently linked to the glucose ring and the lysine side chain, respectively, while the threonine side chain is O-glycosylated. CD spectra in different solvent media suggest that both glycopeptides attain an ordered structure, possibly a helix-like conformation. By combining FRET (fluorescence resonance energy transfer) experiments with molecular mechanics data, the most probable structures of both glycopeptides were built up, starting from both a right-handed (rh) alpha- and 3(10)-helix. They were found to populate an alpha-helical conformation, a result further confirmed by the very good agreement between theoretical and experimental quenching efficiency only observed when the backbone chain was in alpha-helix. The association of D-G-Ala-N with model membranes (liposomes) was studied by CD, fluorescence decay, fluorescence anisotropy, and collisional quenching experiments. The binding does not alter the structural features of the peptide because the CD spectral patterns are unaffected by the association. The peptide orientation inside the phospholipidic bilayer is guided by the polar glucose molecule lying in the water phase. The insertion of the hydrophobic backbone chain into the membrane, seeing the probes only partially accessible from the external solution, is characterized by a significant degree of heterogeneity, an increase in vesicles size, and a relevant stabilizing effect on the membrane itself against rupture by methanol. 相似文献

Background information. The idea that GPCRs (G‐protein‐coupled receptors) may exist as homo‐ or hetero‐oligomers, although still controversial, is now widely accepted. Nevertheless, the functional roles of oligomerization are still unclear and gaining greater insight into the mechanisms underlying the dynamics of GPCR assembly and, in particular, assessing the effect of ligands on this process seems important. We chose to focus our present study on the effect of MT7 (muscarinic toxin 7), a highly selective allosteric peptide ligand, on the oligomerization state of the hM1 (human M1 muscarinic acetylcholine receptor subtype). Results. We analysed the hM1 oligomerization state in membrane preparations or in live cells and observed the effect of MT7 via four complementary techniques: native‐PAGE electrophoresis analysed by both Western blotting and autoradiography on solubilized membrane preparations of CHO‐M1 cells (Chinese‐hamster ovary cells expressing muscarinic M1 receptors); FRET (fluorescence resonance energy transfer) experiments on cells expressing differently tagged M1 receptors using either an acceptor photobleaching approach or a novel fluorescence emission anisotropy technique; and, finally, by BRET (bioluminescence resonance energy transfer) assays. Our results reveal that MT7 seems to protect the M1 receptor from the dissociating effect of the detergent and induces an increase in the FRET and BRET signals, highlighting its ability to affect the dimeric form of the receptor. Conclusions. Our results suggest that MT7 binds to a dimeric form of hM1 receptor, favouring the stability of this receptor state at the cellular level, probably by inducing some conformational rearrangements of the pre‐existing muscarinic receptor homodimers. 相似文献

Caspases play important roles in cell apoptosis. Measurement of the dynamics of caspase activation in tumor cells not only facilitates understanding of the molecular mechanisms of apoptosis but also contributes to the development, screening, and evaluation of anticancer drugs that target apoptotic pathways. The fluorescence resonance energy transfer (FRET) technique provides a valuable approach for defining the dynamics of apoptosis with high spatio-temporal resolution. However, FRET generally functions in the single-cell level and becomes ineffective when applied in the high throughput detection of caspase activation. In the current study, a FRET sensor was combined with capillary electrophoresis (CE) to achieve a high throughput method for cellular caspase detection. The FRET-based CE system is composed of a homemade CE system and a laser source for detecting the dynamics of caspase-3 in various cells expressing sensors of caspase-3 that have been treated with anticancer drugs, such as cell cycle-independent drug cisplatin and specific cell cycle drugs camptothecin and etoposide, as well as their combination with tumor necrosis factor (TNF). A positive correlation between the caspase-3 activation velocity and drug concentration was observed when the cells were treated with cisplatin, but cells induced by camptothecin and etoposide did not show any apparent correlation with their concentrations. Moreover, different types of cells presented distinct sensitivities under the same drug treatment, and the combination treatment of TNF and anticancer drugs significantly accelerated the caspase-3 activation process. Its high throughput capability and detection sensitivity make the FRET-based CE system a useful tool for investigating the mechanisms of anticancer drugs and anticancer drug screening. 相似文献

The interactions of caffeine (CF) with chlorogenic acid (CGA) and caffeic acid (CFA) were investigated by fluorescence quenching, UV/vis and Fourier transform infrared (FTIR) spectroscopic techniques. The results of the study indicated that the fluorescence quenching between caffeine and hydroxycinnamic acids could be rationalized in terms of static quenching or the formation of non‐fluorescent CF–CFA and CF–CGA complexes. From fluorescence quenching spectral analysis, the quenching constant (KSV), quenching rate constant (kq), number of binding sites (n), thermodynamic properties and conformational changes of the interaction were determined. The quenching constants (KSV) between CF and CGA, CFA are 1.84 × 104 and 1.04 × 104 L/mol at 298 K and their binding site n is ~ 1. Thermodynamic parameters determined using the Van't Hoff equation indicated that hydrogen bonds and van der Waal's forces have a major role in the reaction of caffeine with caffeic acid and chlorogenic acid. The 3D fluorescence, UV/vis and FTIR spectra also showed that the binding of CF with CFA and CGA induces conformational changes in CFA and CGA. Copyright © 2015 John Wiley & Sons, Ltd. 相似文献

The detection of protein-protein binding on microarrays using the fluorescence lifetime as a dynamic analytical parameter was investigated in a model system. The assay is based on F?rster resonance energy transfer (FRET) and carried out with biotinylated Bovine Serum Albumin and streptavidin, labeled with the commonly used microarray dyes Alexa 555 and Alexa 647, respectively. This efficient FRET donor/acceptor pair was employed in a competitive assay format on three different microarray surfaces. The fluorescence was excited by 200ps laser pulses from a mode-locked and cavity-dumped argon-ion laser, adapted to an intensified CCD camera as detection unit allowing time resolution with subnanosecond precision. Lifetime maps were recorded according to the Rapid Lifetime Determination (RLD) scheme. Interaction between the proteins could clearly be detected on all formats and resulted in almost complete quenching on CEL Epoxy surfaces upon addition of excess streptavidin labeled the FRET acceptor dye. In this case, the fluorescence lifetimes dropped by 90%, whereas on ARChip Epoxy and ARChip Gel the reduction was 54% and 47%, respectively. Good linearity of the quenching curve was obtained in all cases. The method is applicable to all types of protein interaction analysis on microarrays, particularly in cases where evaluation of fluorescence intensity is prone to erroneous results and a more robust parameter is required. 相似文献

In this study, CdS quantum dots (QDs) capped with mercaptosuccinic acid (MSA) were prepared in one step. The size, shape, component and spectral properties of MSA‐capped CdS QDs were characterized by transmission electron microscopy (TEM), photoluminescence (PL) and infrared (IR) spectrometry. The results showed that the prepared QDs with an average diameter of 6 nm have favorable fluorescence, which is greatly influenced by the pH of the environment. The interaction of some heavy metal ions including Ag+, Hg2+, Cu2+, Ni2+ and Co2+ with MSA‐capped CdS QDs was investigated in different buffering pH media. Based on the fluorescence quenching of the QDs in the presence of each of the metal ions, the feasibility of their determinations was examined according to the Stern–Volmer equation. The investigations showed that Hg(II) ions can be determined in the presence of many co‐existing metal ions at a buffering pH of 5. The three types of structure of the pyrenebutyric acid of fiber optic chemical film sensor were stud-ied by fluorescence multiple quenching. They are, for different test samples and purposes, respectively general, three-way and combined. A tri-cup method was designed to demonstrate the multiple quenching of response mechanism, and a relationship formula of mathematical approach was established. The response mechanism was shown to include the dynamic quenching , inner-filter effects and/or resonance energy transfer. To show the response characterization in a series of organic and inorganic quenchers, a new concept of apparent quenching coefficient Kq was advanced. This kind of sensor has been used in continuous and in situ monitoring of the dissolution rate of drug tablets, on line and in situ monitoring of some organic therapeutic drugs in biological fluid and Cr( VI ) in industrial waste water. To develop an analytical system for single-nucleotide polymorphisms (SNPs), the fluorescence resonance energy transfer (FRET) technique was employed on a bacterial magnetic particle (BMP) surface. A combination of fluorescein isothiocyanate (FITC; excitation 490 nm/emission 520 nm) labeled at the 5' end of DNA and an intercalating compound (POPO-3, excitation 534 nm/emission 570 nm) was used to avoid the interference from light scattering caused by nanoparticles. After hybridization between target DNA immobilized onto BMPs and FITC-labeled probes, fluorescence from POPO-3, which was excited by the energy from the FITC, was detected. The major homozygous (ALDH2*1), heterozygous (ALDH2*1/*2), and minor homozygous (ALDH2*2) genotypes in the blood samples were discriminated by this method. Curcumin (Cur) has medicinal properties, undergoes hydrolysis, and has low water solubility that limits its bioavailability and industrial usage. Different host molecules such as carbon nanotubes (CNT) can be useful in improving solubility and stabilizing Cur, therefore understanding the interaction of Cur with host molecules such as CNT is crucial. In this study, UV–visible light absorption and fluorescence spectroscopic techniques have been applied to reveal the interaction of Cur with CNT. Visible light absorption of Cur increases with CNT concentration, whereas fluorescence intensity of Cur is quenched in the presence of CNT. The obtained results confirm that fluorescence reduction is due to both static and dynamic quenching and is a result of the ground state and excited‐state complex formation. The pH environment influences the quenching rate due to deprotonation of Cur at higher pH; excess OH‐ ion concentration in the solution further discourages electrostatic interaction between the deprotonated form of Cur with CNT. It is found that at lower temperatures (<35°C) dynamic quenching is much more dominant and at higher temperatures (45°C) static quenching is more dominant. The interaction is further supported using X‐ray diffraction patterns and Fourier transform infrared spectra in the solid state, and suggests encapsulation of curcumin within the CNT. It is further evident that fluorescence quenching of Cur using CNT is further enhanced in the presence of several salts, as increase in ionic strength of the solution pushes the hydrophobic Cur molecule towards the CNT core by increasing the proximity between them. 相似文献

Selective turn‐on fluorescence assay of 6–thioguanine by using harmine‐modified silver nanoparticles 下载免费PDF全文
This article reports on a novel fluorescence resonance energy transfer (FRET) system between harmine and silver nanoparticles (AgNPs), in which harmine acts as the donor and AgNPs act as the acceptor. As a result of FRET, harmine fluorescence is quenched efficiently with a corresponding Stern–Volmer constant of 3.61 × 1011 L/mol. It was found that upon addition of the anticancer drug, 6–thioguanine (6–TG), the fluorescence was recovered due to the competitive adsorption of this compound onto AgNPs. Based on this effect, a selective turn‐on fluorescence sensor was developed for the determination of 6–TG. Under optimum conditions, the enhanced fluorescence intensity displays a linear relationship with the concentration of 6–TG in the range 1.5 × 10‐8–7.5 × 10‐7 M with a detection limit of 9.7 nM. The interacting-heads motif (IHM) is a structure of myosin that has been proposed to modulate cardiac output by occluding myosin molecules from undergoing the force-generating cycle. It is hypothesized to be the structural basis for the super-relaxed state (SRX), a low-ATPase kinetic state thought to be cardioprotective. The goal of the present study was to test this hypothesis by determining directly and quantitatively the fractions of myosin in the IHM and SRX under the same conditions in solution. To detect the structural IHM, we used time-resolved fluorescence resonance energy transfer to quantitate two distinct populations. One population was observed at a center distance of 2.0 nm, whereas the other was not detectable by fluorescence resonance energy transfer, implying a distance greater than 4 nm. We confirmed the IHM assignment to the 2.0-nm population by applying the same cross-linking protocol used previously to image the IHM by electron microscopy. Under the same conditions, we also measured the fraction of myosin in the SRX using stopped-flow kinetics. Our results show that the populations of SRX and IHM myosin were similar, unless treated with mavacamten, a drug that recently completed phase III clinical trials to treat hypertrophic cardiomyopathy and is proposed to act by stabilizing both the SRX and IHM. However, we found that mavacamten had a much greater effect on the SRX (55% increase) than on the IHM (4% increase). We conclude that the IHM structure is sufficient but not necessary to produce the SRX kinetic state. 相似文献

基于荧光共振能量转移(fluorescence resonance energy transfer, FRET)原理,以新冠病毒主蛋白酶(main protease, Mpro)为靶标,建立并应用Mpro小分子抑制剂FRET高通量筛选模型,以期快速筛选新型Mpro小分子抑制剂。利用大肠杆菌原核表达与分离纯化高活性的Mpro,再以FRET法进行比活力测定。基于FRET原理,以7-甲氧基香豆素-4-乙酸(7-methoxycoumarin-4-acetic acid, MCA)与2,4-二硝基苯酚(2,4-dinitropheno, Dnp)标记的多肽作为Mpro水解底物,通过优化反应缓冲液、Mpro反应浓度、反应温度与时间及DMSO耐受浓度,建立并应用Mpro小分子抑制剂FRET高通量筛选模型进行苗头化合物的筛选。利用大肠杆菌实现了高活性Mpro的原核表达与分离纯化,且比活力不低于40 000 U/mg。通过一系列优化实验,使用0.4μmol/L Mpro与5μmol/L底物建立了Z′因子值为0.79的Mpro小分子抑制剂FRET高通量筛选模型,且反应体系中含有的二硫苏糖醇(1,4-dithiothreitol,DTT)是影响FRET筛选模型可靠性的重要因素。通过对天然产物化合物库进行高通量筛选,发现白花丹素与银杏酸在体外对Mpro酶活性具有良好的抑制作用。本研究建立了基于FRET原理的Mpro小分子抑制剂高通量筛选模型,初步证实了白花丹素与银杏酸是一类新型苗头化合物,为抗新型冠状病毒药物先导化合物的筛选与发现奠定了基础。 相似文献

ATP-binding cassette exporters use the energy of ATP hydrolysis to transport substrates across membranes by switching between inward- and outward-facing conformations. Essentially all structural studies of these proteins have been performed with the proteins in detergent micelles, locked in specific conformations and/or at low temperature. Here, we used luminescence resonance energy transfer spectroscopy to study the prototypical ATP-binding cassette exporter MsbA reconstituted in nanodiscs at 37 °C while it performs ATP hydrolysis. We found major differences when comparing MsbA in these native-like conditions with double electron-electron resonance data and the crystal structure of MsbA in the open inward-facing conformation. The most striking differences include a significantly smaller separation between the nucleotide-binding domains and a larger fraction of molecules with associated nucleotide-binding domains in the nucleotide-free apo state. These studies stress the importance of studying membrane proteins in an environment that approaches physiological conditions. 相似文献
