Analysis of allozyme data of the European freshwater fish Cottus gobio showed marked genetic differentiation across drainage basins in northeastern Bavaria, which points to the existence of at least two cryptic taxa. Genetic variability within populations differed significantly between these two taxa, which could be due to historical (bottlenecks) or ecological reasons (population size). To distinguish between these two hypotheses we sampled 12 distinct populations from Rhine, Elbe and Danube drainages. Using allozyme data we examined the influence of population size and isolation on genetic variability within populations. We used spatial extent of populations (patch size) as a measure for population size. To estimate isolation we calculated a compound measure which took into account patch size and distance to all neighbouring populations. Both patch size and isolation were highly correlated with genetic variability, explaining ≈95% of the variance of genetic variability within populations. Furthermore, analysis of covariance suggests that the difference in genetic variability between taxa may be explained by differences in population size.  相似文献

In order to understand and protect ecosystems, local gene pools need to be evaluated with respect to their uniqueness. Cryptic species present a challenge in this context because their presence, if unrecognized, may lead to serious misjudgement of the distribution of evolutionarily distinct genetic entities. In this study, we describe the current geographical distribution of cryptic species of the ecologically important stream amphipod Gammarus fossarum (types A, B and C). We use a novel pyrosequencing assay for molecular species identification and survey 62 populations in Switzerland, plus several populations in Germany and eastern France. In addition, we compile data from previous publications (mainly Germany). A clear transition is observed from type A in the east (Danube and Po drainages) to types B and, more rarely, C in the west (Meuse, Rhone, and four smaller French river systems). Within the Rhine drainage, the cryptic species meet in a contact zone which spans the entire G. fossarum distribution range from north to south. This large-scale geographical sorting indicates that types A and B persisted in separate refugia during Pleistocene glaciations. Within the contact zone, the species rarely co-occur at the same site, suggesting that ecological processes may preclude long-term coexistence. The clear phylogeographical signal observed in this study implies that, in many parts of Europe, only one of the cryptic species is present.  相似文献

Spatial patterns of genetic diversity provide insight into the demography and history of species. Morphologically similar but genetically distinct “cryptic” species are increasingly being recognized in marine organisms through molecular analyses. Such species are, on closer inspection, often discovered to display contrasting life histories or occasionally minor morphological differences; molecular tools can thus be useful indicators of diversity. Bostrychia intricata, a marine red alga, is widely distributed throughout the Southern Hemisphere and comprises many cryptic species. We used mitochondrial cytochrome c oxidase I gene sequences to assess the genetic variation, population genetic structure, and demographic history of B. intricata in New Zealand. Our results supported the existence of three cryptic species of B. intricata (N2, N4, and N5) in New Zealand. Cryptic species N4, which was found throughout New Zealand, showed a higher genetic diversity and wider distribution than the other two species, which were only found in the North Island and northern South Island. Our analyses showed low to moderate genetic differentiation among eastern North Island populations for cryptic species N2, but high differentiation among North and South Island populations for N4, suggesting different population structure between these cryptic species. Data also indicated that N2 has recently undergone population expansion, probably since the Last Glacial Maximum (LGM), while the higher genetic diversity in N4 populations suggests persistence in situ through the LGM. The contrasting population structures and inferred demographic histories of these species highlight that life history can vary greatly even among morphologically indistinguishable taxa.  相似文献

Spatio-temporal patterns and processes of genetic differentiation in passively dispersing zooplankton are drawing much attention from both ecologists and evolutionary biologists. Two opposite phylogeographical scenarios have already been demonstrated in rotifers, which consist of high levels of genetic differentiation among populations even on small geographical scales on the one hand and the traditionally known cosmopolitanism that is associated with high levels of gene flow and long-distance dispersal via diapausing stages on the other hand. Here, we analysed the population genetic structure and the phylogeography of the Brachionus calyciflorus species complex in eastern China. By screening a total of 318 individuals from ten locations along a 2320-km gradient and analysing samples from two growing seasons, we aimed at focusing on both small- and large-scale patterns. We identified eight cryptic species and verified species status of two of these by sexual reproduction tests. Samples in summer and winter yielded different cryptic species. The distribution patterns of these genetically distinct cryptic species were diverse across eastern China, from full cosmopolitanism to local endemism. The two most abundant cryptic species BcWIII and BcSW showed a pattern of strong genetic differentiation among populations and no significant isolation by distance. Long-distance colonization, secondary contact and recent range expansion are probably responsible for the indistinct pattern of isolation by distance. Our results suggest that geographical distance is more important than temporal segregation across seasons in explaining population differentiation and the occurrence of cryptic species. We explain the current phylogeographical structure in the B. calyciflorus species complex by a combination of recent population expansion, restricted gene flow, priority effects and long-distance colonization.  相似文献

 利用9对SSR引物对中华猕猴桃(Actinidia chinensis)和美味猕猴桃(A. deliciosa)两近缘种的5个同域分布复合体和各自1个非同域分布居群进行了遗传多样性、居群遗传结构的分析以及种间杂交渐渗的探讨。结果表明:1)两物种共有等位基因比例高达81.13%,物种特有等位基因较少(中华猕猴桃:13.27%,美味猕猴桃:5.61%),但共享等位基因表型频率在两近缘种间存在差异,而且与各同域复合体中两物种样本的交错程度或间距存在关联;2)两种猕猴桃均具有极高遗传多样性,美味猕猴桃的遗传多样性(Ho=0 .749, PIC=0.818)都略高于中华猕猴桃(Ho=0.686,PIC=0.799);3)两猕 猴桃物种均具有较低的Nei’s居群遗传分化度,但AMOVA分析结果揭示种内异域居群间(FST=0.091 5)和同域复合体种间(FST=0.111 5)均存在一定程度的遗传分化;中华猕猴桃居群遗传分化(GST=0.086; FST=0.212 1)高于美味猕猴桃(GST= 0.080;FST=0.142 0);4)同域分布复合体两物种间的遗传分化(GST=0.020)低于物种内异域居群间的遗传分化(中华猕猴桃:GST=0.086; 美味猕猴桃:GST=0.080),同域复合体物种间的基因流(Nm=7.89 -29.75)远远高于 同种异域居群间(中华猕猴桃:Nm =2.663; 美味猕猴桃:Nm=2.880); 5)居群UPGMA聚类揭示在同一地域的居群优先聚类,个体聚类结果显示多数个体聚在各自居群组内,但各地理居群并不按地理距离的远近聚类,这与Mantel相关性检测所揭示的居群间遗传距离与地理距离没有显著性相关的结果一致。进一步分析表明两种猕猴桃的遗传多样性和居群遗传结构不仅受其广域分布、远交、晚期分化等生活史特性的影响,同时还与猕猴桃的染色体基数高 (x=29)、倍性复杂和种间杂交等因素密切相关,其中两种猕猴桃的共享祖先多态性和同域分布种间杂交基因渗透对两猕猴桃的居群遗传结构产生了重要影响。  相似文献

Analysis of mitochondrial ( ND-1/3/4 genes) and nuclear ( GH-1 gene, four microsatellite loci) DNA markers of European grayling Thymallus thymallus populations identified strong genetic differentiation between the Rhine/Main, Elbe and Danube drainages in Bavaria, as a result of both present processes and past history. The Danube and the Main-Elbe group diverged about five times earlier than the populations of Main and Elbe. These data suggest that exchange and transfer of grayling stocking material between these drainages should be avoided, to maintain the genetic diversity and integrity of populations.  相似文献

1. Determined by landscape structure as well as dispersal‐related traits of species, connectivity influences various key aspects of population biology, ranging from population persistence to genetic structure and diversity. Here, we investigated differences in small‐scale connectivity in terms of gene flow between populations of two ecologically important invertebrates with contrasting dispersal‐related traits: an amphipod (Gammarus fossarum) with a purely aquatic life cycle and a mayfly (Baetis rhodani) with a terrestrial adult stage. 2. We used highly polymorphic markers to estimate genetic differentiation between populations of both species within a Swiss pre‐alpine catchment and compared these results to the broader‐scale genetic structure within the Rhine drainage. Landscape genetic approaches were used to test for correlations of genetic and geographical structures and in‐stream barrier effects. 3. We found overall very weak genetic structure in populations of B. rhodani. In contrast, G. fossarum showed strong genetic differentiation, even at spatial scales of a few kilometres, and a clear pattern of isolation by distance. Genetic diversity decreased from downstream towards upstream populations of G. fossarum, suggesting asymmetric gene flow. Correlation of genetic structure with landscape topography was more pronounced in the amphipod. Our study also indicates that G. fossarum might be capable of dispersing overland in headwater regions and of crossing small in‐stream barriers. 4. We speculate that differences in dispersal capacity but also habitat specialisation and potentially the extent of local adaptation could be responsible for the differences in genetic differentiation found between the two species. These results highlight the importance of taking into account dispersal‐related traits when planning management and conservation strategies.  相似文献

We used sequences of the mitochondria control region to assess the distribution of genetic variation within and among populations of the poeciliid fish species Xiphophorus birchmanni . We collected 122 X. birchmanni samples from 11 sites in three drainage systems comprising the distribution of the species. We found low levels of polymorphism among aligned sequences and low levels of genetic variation within populations but high levels of genetic differentiation among populations. Haplotypes are exclusive to three river drainages (Los Hules, Calabozo and San Pedro). Mantel tests revealed correlations between geographical (both straight-line and river distances) and genetic distance, consistent with an isolation by distance scenario, while nested clade analysis suggested allopatric fragmentation between haplotypes from two of the major drainages, and isolation by distance with restricted gene flow within those drainages. Finally, monophyly of X. birchmanni is strongly supported while the previous hypothesis of the evolutionary origin of this species from X. malinche is not.  相似文献

Despite the fact that mollusc species play an important role in many aquatic ecosystems, little is known about their biodiversity and conservation genetics. Freshwater pearl mussel (Margaritifera margaritifera L.) populations are seriously declining all over Europe and a variety of conservation programs are being established to support the remaining endangered central European populations. In order to provide guidelines for conservation strategies and management programs, we investigated the genetic structure of 24 freshwater pearl mussel populations originating from five major central European drainages including Elbe, Danube, Rhine, Maas and Weser, representing the last and most important populations in this area. We present a nondestructive sampling method of haemolymph for DNA analyses, which is applicable for endangered bivalves. The analyses of nine microsatellite loci with different levels of polymorphism revealed a high degree of fragmented population structure and very different levels of genetic diversity within populations. These patterns can be explained by historical and demographic effects and have been enforced by anthropogenic activities. Even within drainages, distinct conservation units were detected, as revealed from high F(ST) values, private alleles and genetic distance measures. Populations sampled close to contact zones between main drainage systems showed lowest levels of correct assignment to present-day drainage systems. Populations with high priority for conservation should not only be selected by means of census population size and geographical distance to other populations. Instead, detailed genetic analyses are mandatory for revealing differentiation and diversity parameters, which should be combined with ecological criteria for sustainable conservation and recovery programs.  相似文献

Bluemask darters (Etheostoma akatulo) were sampled from the four drainages where extant populations of this narrowly endemic freshwater fish are known to exist. Population genetic diversity and structure were assessed at 10 microsatellite loci. All populations exhibited low levels of genetic variation, with expected heterozygosity ranging from 0.2 to 0.35. Significant population subdivision was found among most tributaries, and genetic divergence was strongly correlated with geographic distance. Bayesian population assignment and pairwise population differentiation measures both identified a lack of differentiation between E. akatulo populations inhabiting Cane Creek and the Caney Fork. This observation reduced the number of distinct breeding populations of this species to three. We also used approximate Bayesian computation to compare three models of demographic history in this species. A constant population size model was favored over models that included historic or recent population reductions. Our results suggest that impoundment of the Caney Fork and its tributaries, by completion of Great Falls Dam in 1916, was not responsible for the reduced genetic diversity in the sampled populations. Given the low levels of genetic diversity within populations and the limited geographic distribution, future conservation efforts should seek to maximize available habitat while simultaneously limiting the influences of anthropogenic stressors in the system.  相似文献

The climbing habit is a key innovation in plants: climbing taxa have higher species richness than nonclimbing sister groups. We evaluated the hypothesis that climbing plant species show greater among‐population genetic differentiation than nonclimber species. We compared the among‐population genetic distance in woody climbers (eight species, 30 populations) and trees (seven species, 29 populations) coexisting in nine communities in a temperate rainforest. We also compared within‐population genetic diversity in co‐occurring woody climbers and trees in two communities. Mean genetic distance between populations of climbers was twice that of trees. Isolation by distance (increase in genetic distance with geographic distance) was greater for climbers. Climbers and trees showed similar within‐population genetic diversity. Our longevity estimate suggested that climbers had shorter generation times, while other biological features often associated with diversification (dispersal and pollination syndromes, mating system, size, and metabolic rate) did not show significant differences between groups. We hypothesize that the greater population differentiation in climbers could result from greater evolutionary responses to local selection acting on initially higher within‐population genetic diversity, which could be driven by neutral processes associated with shorter generation times. Increased population genetic differentiation could be incorporated as another line of evidence when testing for key innovations.  相似文献

The Driftless Area of the upper Mississippi River drainage is a unique geographic region because of its complex geological history and the influence of recent, intensive human activities. The longnose dace, Rhinichthys cataractae, is a relatively common, small freshwater fish that is distributed in swift, cool streams within the region. The aim of this study was to determine the spatial genetic differentiation of the longnose dace and define the broad scale environmental variables that shape the distribution of the species in the southwestern portion of the Driftless Area. Genotypic data from seven microsatellite loci were analyzed for 276 individuals from 15 localities representing major drainages within the region in northeast Iowa. Broad scale environmental variables including hydrologic, soil, and climatic factors were evaluated to construct an ecological niche model (ENM) to predict the suitability of habitat for the species within the region. Results of the genetic analyses revealed two distinct, but somewhat admixed genetic clusters of longnose dace in Iowa. The genetic differentiation between localities and between drainages was low to moderate with some evidence of isolation by distance. Most of the variation was observed by differences between individuals within local populations. The ENM generated largely reflected the known distribution of the species in Iowa with a decreasing probability of suitable habitat from northern to southern drainages. Geologic factors played a key role in the model. The distribution and population structure of the longnose dace in the northeast Iowa revealed that isolation by distance, historical processes and the underlying geology are primarily responsible for the observed spatial distribution of genetic variation.  相似文献

为了揭示裸果木的遗传多样性和遗传结构,采用SCoT分子标记对新疆7个裸果木自然种群201个植株进行分析。结果表明12条引物共扩增出145个位点,在物种和种群水平上裸果木多态位点百分率分别为97.24%和76.45%,Nei's遗传多样性指数分别为0.263 8和0.213 7,Shannon's信息多样性指数分别为0.408 1和0.331 6,说明裸果木具有较高的遗传多样性,且物种水平遗传多样性略高于种群水平;遗传分化系数为0.183 9,说明其遗传变异主要来自种群内,种群间存在一定程度的分化;Mantel检验结果表明裸果木种群地理距离与遗传距离不存在显著相关性;运用主坐标分析(PCoA)和STRUCTURE分析对裸果木个体间亲缘关系聚类分析的结果存在一定差异,但总体上结果是一致的。裸果木的遗传多样性和遗传结构可能与进化历史、繁育系统、地理隔离和人为干扰有关。  相似文献

Stipa capillata L. (Poaceae) is a rare grassland species in Central Europe that is thought to have once been widespread in post‐glacial times. Such relict species are expected to show low genetic diversity within populations and high genetic differentiation between populations due to bottlenecks, long‐term isolation and ongoing habitat fragmentation. These patterns should be particularly pronounced in selfing species. We analysed patterns of random amplified polymorphic DNA (RAPD) variation in the facultatively cleistogamous S. capillata to examine whether genetic diversity is associated with population size, and to draw initial conclusions on the migration history of this species in Central Europe. We analysed 31 S. capillata populations distributed in northeastern, central and western Germany, Switzerland and Slovakia. Estimates of genetic diversity at the population level were low and not related to population size. Among all populations, extraordinarily high levels of genetic differentiation (amova : φST = 0.86; Bayesian analysis: θB = 0.758) and isolation‐by‐distance were detected. Hierarchical amova indicated that most of the variability was partitioned among geographic regions (59%), or among populations between regions when the genetically distinct Slovakian populations were excluded. These findings are supported by results of a multivariate ordination analysis. We also found two different groups in an UPGMA cluster analysis: one that contained the populations from Slovakia, and the other that combined the populations from Germany and Switzerland. Our findings imply that Scapillata is indeed a relict species that experienced strong bottlenecks in Central Europe, enhanced by isolation and selfing. Most likely, populations in Slovakia were not the main genetic source for the post‐glacial colonization of Central Europe.  相似文献

Habitat fragmentation may reduce gene flow and population viability of rare species. We tested whether riparian corridors enhanced gene flow and if human habitat modification between riparian corridors subsequently reduced dispersal and gene flow of a wetland butterfly, the US federally endangered St. Francis’ satyr butterfly (Neonympha mitchellii francisci). We surveyed nine populations throughout the taxon’s range using five polymorphic microsatellite loci. We found that genetic diversity of N. m. francisci was relatively high despite its restricted distribution, and that there is little evidence of population bottlenecks or extensive inbreeding within populations. We found substantial gene flow and detectable first generation migration, suggesting that N. m. francisci is unlikely to be currently endangered by genetic factors. Pairwise population differentiation and clustering indicate some structuring between populations on different drainages and suggest that dispersal probably occurs mainly via a stepping stone from the closest riparian corridors. However, genetic differentiation between geographically close populations suggests that isolation by distance is not solely responsible for population structure, and that management actions should be targeted at maintaining connectivity of riparian and upland habitats.  相似文献

Understanding levels of population differentiation and inbreeding are important issues in conservation biology, especially for social Hymenoptera with fragmented and small population sizes. Isolated populations are more vulnerable to genetic loss and extinction than those with extended continuous distributions. However, small populations are not always a consequence of a recent reduction of their habitat. Thus, determining the history of population isolation and current patterns of genetic variation of a species is crucial for its conservation. Rossomyrmex minuchae is a slave-making ant with patchy distribution in South Eastern Spain and is classified as vulnerable by the IUCN. In contrast, the other three known species of the genus are presumed to show more uniform distributions. Here we investigate the genetic diversity and population structure of R. minuchae and compare it with that found in two other species of the genus: R. anatolicus and R. quandratinodum. We conclude that although genetic diversity of R. minuchae is low, there is no evidence of a recent bottleneck, suggesting a gradual and natural fragmentation process. We also show extreme population differentiation at nuclear and mitochondrial markers, and isolation by distance at a local scale. Despite some evidence for inbreeding and low genetic variation within populations, we found almost no diploid males, a finding which contrasts with that expected in inbred Hymenoptera with single locus complementary sex determination. This could mean that sex is determined by another mechanism. We argue that continued low population size means that detrimental effects of inbreeding and low genetic variation are likely in the future. We suggest that a policy of artificial gene flow aimed at increasing within population variation is considered as a management option.  相似文献

海南鲌(Culter recurviceps)是我国华南地区重要经济鱼类, 由于受到近些年水利开发、过度捕捞、环境污染等诸多因素的影响, 其资源量快速下降, 亟需得到更多的关注和保护。为保护和合理开发海南鲌种质资源, 本研究采集了华南地区23个地理群体207尾海南鲌样本, 测定了2个线粒体基因(CytbND2)并从Barcode of Life Data System数据库获得相对应线粒体COI基因, 结合多种分析方法(系统发育分析、分化时间估算、单倍型网状图、群体遗传分析和Mantel检验)对海南鲌的遗传结构和遗传多样性展开研究。系统发育分析和单倍型网状图表明华南地区海南鲌群体被分成3个谱系(I、II和III), 其中谱系I和III由珠江的群体组成, 谱系II由海南岛的群体组成。分化时间估算发现3个谱系之间的分化时间介于0.028-0.251 Ma之间, 表明华南地区更新世气候变化可能是造成海南鲌谱系分化的重要原因。群体遗传分析发现海南鲌群体之间存在极显著的遗传分化(FST = 0.511, P < 0.001), 并且符合距离隔离模式(R = 0.348, P = 0.0010)。群体动态历史分析表明, 海南鲌群体可能在0.010-0.025 Ma经历了群体扩张, 表明更新世的气候波动也影响了海南鲌的群体大小和分布。综上所述, 海南鲌群体由3个谱系组成, 更新世气候变化是导致3个谱系分化和影响海南鲌群体动态历史的重要因素。此外, 海南鲌群体之间的遗传分化也可能受到了空间距离的影响。  相似文献

The Iberian cyprinid fauna, characterized by the presence of numerous endemic species, has suffered from significant habitat degradation. The critically endangered Squalius aradensis is restricted to small drainages of southern Portugal, habitats that typically exhibit a characteristic Mediterranean-type heterogeneous hydrological system throughout the year, including alternation of flooding events during winter and complete drought in large river sections during summer. To assess the effect of historical and recent processes on genetic diversity in S. aradensis we examined within- and among-population variability in cytochrome b and six polymorphic microsatellite loci. Estimates of genetic diversity in time and space through the combined use of traditional Phi-/F-statistics, phylogenetic trees, ordination methods and nested clade analysis indicated significant and congruent structuring among populations. Data suggest that the Arade drainage represent the evolutionary centre of the species, with subsequent allopatric fragmentation across drainages. Factors other than isolation by distance strongly affected the within-drainage genetic differentiation observed in these Mediterranean-type drainages, including recent population expansion from a bottleneck event and restricted gene flow imposed by a long-term barrier (brackish water area). Significant correlation was found between S. aradensis allelic diversity and upstream drainage area. The relevance of findings for conservation issues is discussed in relation to local intermittent hydrological conditions, the highly restricted distribution and the critically endangered status of the species.  相似文献

Allozyme data were used to assess the genetic structure between 37 sympatric populations of the species-pair Stlene vulgaris and S uniflora ssp petraea, and to infer levels of intra- and interspecific gene flow in the two species Silene vulgaris is a geographically widespread weed of disturbed habitats whereas S uniflora ssp petraea is endemic to the Baltic islands of Oland and Gotland On Oland, Silene vulgaris forms extensive linear populations along roads while S uniflora ssp petraea occurs in sparse and spatially-separated populations in open limestone habitats Despite the differences in population size and structure between the two species, both species show extremely low levels of between-population differentiation Between-site differences account for <2% of the total allozyme diversity within Oland in S vulgaris, and < 1% in S uniflora ssp petraea Indirect estimates of gene flow are high for both species (Nm = 11 and 27, respectively) There is no relationship between genetic distance and geographic distance within either species, and the lack of genetic structure is consistent with the pollination biology of the species - both of which are predominantly moth-pollinated The two species hybridize in intermediate habitats, and the geographic distribution of species-characteristic alleles indicates a potential for spatially extensive interspecific gene flow Nevertheless, there are significant differences in allele frequencies between the two species and multivariate analyses show no overlap between populations of the two species The species are ecologically separated by their different habitat preferences and by differences in their flowering phenology There is no evidence that the endemic S uniflora ssp petraea is threatened by genetic contamination or assimilation by the widespread weed, S vulgaris  相似文献
