荔枝雌蕊发育过程中钙分布变化与细胞程序性死亡   总被引:1,自引:0,他引:1  
应用焦锑酸钾沉淀法研究了荔枝雌花和雄花雌蕊发育过程中钙的分布变化。在大孢子母细胞阶段,雌花近珠孔内珠被细胞和花柱细胞的钙沉淀颗粒主要分布在细胞壁和细胞间隙,少部分在液泡;雌花花柱维管细胞中含有很多的钙沉淀颗粒;在雄花的近珠孔内珠被细胞钙沉淀颗粒大多在液泡中;雄花花柱细胞和维管细胞中钙沉淀颗粒很少。大孢子母细胞减数分裂后,雌花雌蕊继续发育,雄花雌蕊败育。雌花维管中的钙沉淀颗粒数量减少,可能被转运到将要发生花粉萌发和受精的部位。雌花近珠孔内珠被细胞壁的钙沉淀颗粒分布增加,花柱细胞从上(近柱头)到下(近子房)钙沉淀颗粒量递增。雄花近珠孔内珠被细胞发生程序性死亡:液泡中的钙进入细胞核启动细胞程序性死亡,核周隙与质膜腔形成连续的通道,钙在核与细胞质之间的流动不受限制;在特定的时间段,钙沉淀颗粒出现在线粒体、过氧化物体和线型内质网的外膜上。钙在细胞中重新分布可能触发和调节细胞程序性死亡的进程。缺乏钙沉淀颗粒的雄花花柱细胞迅速解体。  相似文献

The calcium distribution in the ampullary electroreceptor and the type B electrore-ceptor organ (gymnarchomast) of Gymnarchus niloticus (Gymnarchidae) and in the tuberous organ of Apteronotus leptorhynchus (gymnotidae) was studied. Endogenous calcium appeared as electron-dense precipitates when the cutaneous organs were pre-fixed with phosphate-buffered glutaraldehyde and postfixed with osmium tetroxide plus potassium bichromate. Calcium precipitates were localized in both intracellular compartments of sensory cells and afferent nerve fibers. In contrast to sensory cells, small amounts of calcium precipitates were found in the cytoplasm of accessory cells. In sensory cells, electron-dense deposits were apparent mainly in synaptic vesicles near synaptic ribbons, inside vacuoles of the endoplasmic reticulum, and between the layers of the nuclear membrane. Very few deposits were found in mitochondria. Precipitates were also observed within the axons of afferent nerves and between the layers of the myelin sheath. The synaptic cleft was devoid of calcium. Calcium deposits have a specific cellular distribution in electro-receptor organs of teleost fish.  相似文献

运用焦锑酸钾沉淀法研究了华北落叶松(Larix principis-rupprechtii Mayr)小孢子发育过程中不同阶段Ca2 的分布情况.减数分裂时期,小孢子囊壁表皮和中层细胞的细胞壁及细胞间隙Ca2 分布较多,绒毡层只有外切向面的细胞膜有Ca2 分布,小孢子母细胞的各部位则很少有Ca2 ;四分体时期,包围四分小孢子的胼胝质壁上有大量的Ca2 分布,在四分孢子壁上也有较多沉淀;游离小孢子时期,钙离子在小孢子壁的分布较四分体时期有所减少,而到花粉成熟时又逐渐增多;从四分体到花粉成熟,乌氏体周围的Ca2 有增多的趋势.对四分体外壁Ca2 的大量分布与花粉壁的形成及信号物质在花粉表面贮存的关系,以及小孢子囊的外壁、绒毡层和乌氏体在Ca2 向花粉运输中所起的作用进行了讨论.  相似文献

Calcium plays a crucial role in regulating the growth and differentiation of cultured keratinocytes. However, the mechanism(s) of this regulation is not clear. Prior studies have shown that intracellular free calcium (Cai) increases with keratinocyte differentiation. In this study, in order to evaluate the role of cytosolic free calcium and organelle-bound calcium in keratinocyte differentiation, we quantitated and localized calcium pools in keratinocytes, utilizing the fluorescence probe indo-1 and ion-capture cytochemistry, respectively. Cai of undifferentiated keratinocytes was 80–120 nM, whereas Cai of differentiated keratinocytes was 200–300 nM depending on the extent of differentiation. The Cai of individual cells in an undifferentiated colony was heterogeneous (60–160 nM) with larger cells displaying higher Cai. Heterogeneity also was observed in the intracellular calcium-containing precipitates in the different layers of stratifying keratinocyte cultures using the cytochemical technique. Calcium precipitates were abundant in the lower cell layers, progressively decreasing apically, with the uppermost layer devoid of precipitates. Calcium-containing precipitates appeared as fine-tocoarse electron-dense granules on the plasma membrane, within the cytosol, mitochondria, nucleus, and vacuolar organelles. Whereas ionomycin in the presence of extracellular calcium increased the amount of intracellular calcium precipitates, EGTA removed calcium precipitates from organelles. Unlike intact epidermis, keratinocytes displayed no extracellular calcium reservoirs. Putative calcium binding sites, visualized by trivalent lanthanum (La) binding, were abundant on cell membranes and desmosomes of basaloid cells, but decreased in the upper cell layers. These studies revealed differences in the distribution of free ionic calcium (as determined by the fluorescence technique) and organelle-bound calcium (as determined by the cytochemical technique). Striking differences were also observed in calcium localization between intact epidermis and cultured epidermal cells. The localization pattern of calcium in cultured keratinocytes may reflect the hyperproliferative state of these cells, as in psoriatic epidermis, and/or the absence of a normal permeability barrier in these submerged cultures. © 1993 Wiley-Liss, Inc.  相似文献

采用超薄切片技术,在透射电镜下观察麻疯树(Jatropha curcasL.)花药发育过程中Ca2 的分布特征。在孢原细胞时期的花药中几乎看不到Ca2 沉淀,但花药维管束周围的细胞中有较多的Ca2 沉淀;到小孢子母细胞时期,细胞质中Ca2 沉淀依然较少,绒毡层壁上Ca2 沉淀明显增多;四分体形成时,小孢子细胞质和绒毡层细胞质中出现了较多的Ca2 沉淀;在小孢子发育早期,细胞质中Ca2 沉淀增加不明显,花粉壁部位累积有很多的Ca2 沉淀,绒毡层中Ca2 沉淀数量达到最多;到小孢子发育晚期,小孢子大液泡的液泡膜上有大量的Ca2 沉淀,绒毡层中Ca2 沉淀明显减少;随着二胞花粉中的大液泡消失,细胞质中积累淀粉粒以后,花粉中看到的Ca2 沉淀极少,同时,在花药维管束周围的薄壁细胞中,又出现了较多的Ca2 沉淀,表明花粉对Ca2 的需求可能降低。麻疯树花药发育过程中钙的动态分布特征暗示着钙参与了调控花粉发育过程,Ca2 的运输途径是由药隔薄壁组织运输到绒毡层,再进一步转移到小孢子表面和细胞质中,整个花药发育过程中,Ca2 沉淀表现为少—增加—减少的变化趋势。  相似文献

Plasmodia of the acellular slime mold, Physarum polycephalum, were treated with an osmium tetroxide fixative containing potassium pyroantimonate to precipitate calcium and thereby localize calcium binding sites and sites of increased calcium concentration. Dense calcium pyroantimonate precipitates were detected within the nucleoli. The distribution of these precipitates during interphase and mitosis coincides with the distribution of the unique minichromosomes in Physarum, i.e., the numerous short pieces of extrachromosomal nucleolar chromatin containing segments of amplified DNA coding for ribosomal RNA. Calcium pyroantimonate precipitates were present as frequent dense granules in the mitochondrial matrix and as fine precipitates in the mitochondrial nucleoid. Large calcium-containing precipitates were seen within cytoplasmic vacuoles, confirming reports by others. In addition, we have identified calcium binding sites along the cytoplasmic surface of the plasma membrane. The distribution of calcium within the plasmodium is discussed in relation to the assembly of the mitotic spindle and the regulation of cell motility.  相似文献

采用焦锑酸钾沉淀钙离子技术,对洋葱(Alliumcepa)花药发育中Ca^2+分布进行了研究。在小孢子母细胞时期,小孢子母细胞中的钙沉淀颗粒很少,但绒毡层细胞的内切向壁已出现明显的钙沉淀颗粒。在四分体时期,四分体小孢子的胼胝质壁中出现较多的钙沉淀颗粒;绒毡层细胞内切向壁的钙沉淀颗粒消失,而在外切向壁和径向壁部位的钙沉淀颗粒增加。在小孢子早期,小孢子中也出现了钙沉淀颗粒,而绒毡层细胞内切向壁表面出现了很多絮状物,其上附有细小钙沉淀颗粒。到小孢子晚期,小孢子中出现一些小液泡,细胞质中的钙沉淀颗粒有所下降。此时绒毡层细胞已明显退化,但在绒毡层膜上仍有一些乌氏体和钙沉淀颗粒。在二胞花粉早期,营养细胞中的液泡收缩、消失,细胞质中又出现了较多的钙沉淀颗粒,在质体和其内部的淀粉粒表面上附有较多的钙沉淀颗粒。到二胞花粉晚期,花粉中的钙沉淀颗粒已明显下降,仅在花粉外壁中还有一地钙沉淀颗粒.  相似文献

Summary Calcium distribution in ovules ofTorenia fournieri was studied by electron energy loss spectroscopy and transmission electron microscopic visualization of calcium antimonate precipitates. High calcium levels were found in the ovules ofT. fournieri. Calcium is situated mainly in extracellular regions before fertilization, including the surface of embryo sac, in the mucilage, and among the cells of the egg apparatus. Intracellular calcium was found only in the nucellar cells around the embryo sac and in the epidermis of the central axis and funiculus. After pollination, a labyrinthine structure (coralloid-like cell wall formation) develops on the micropylar surfaces of the egg apparatus that contain high levels of calcium. Calcium levels increase in the degenerating synergid after the penetration of the pollen tube. Summary Mineral components of the scales in an amoeba, Cochliopodium sp., were examined by energy-dispersive X-ray microanalysis. The precipitates in potassium antimonate-treated material detected calcium in the scales. Calcium was also clearly detected in freeze-substituted thin sections. Similar deposits of calcium antimonate were detected in scales in formation within vacuoles, and also in Golgi cisternae, Golgi vesicles and special granules near the nucleus. There were only minute amounts of magnesium and potassium. This suggests that calcium is the main mineral component of the scales and that it is added in the Golgi complex during scale formation.  相似文献

用焦锑酸盐沉淀法对莴苣开花前后的卵细胞、合子与原胚细胞中的钙颗粒分布变化进行了观察。结果表明,开花前三天,刚形成的卵细胞内钙颗粒很少,开花前二天的卵细胞内钙颗粒开始增多,开花前一天的卵细胞形成了大液泡,建立了极性,细胞内的钙颗粒又减少。开花后、受精前的卵细胞的钙颗粒主要聚集在细胞核中。受精后合子中的钙颗粒又明显增多,在核质中分布一些较大的钙颗粒,在珠孔端大液泡中聚集了较多的絮状钙。二胞原胚中的钙颗粒又开始减少,多胞原胚细胞中的钙进一步减少,但原胚表面分布一层丰富的钙颗粒。探讨了钙在卵细胞分化成熟、受精以及原胚发育初期中的作用。  相似文献

Abstract

The stigma (tip of the pistil) of medlar is wet and covered with stigmatic exudate at anthesis. The exudate contains many vesicles with abundant calcium precipitates. After deposition on the stigma, the pollen grain undergoes hydration, displaying signs of calcium ion (Ca2+) transfer from the exudate vesicles into the pollen grains. Calcium precipitates in the pollen cytoplasm are concentrated into small vacuoles that fuse to form large vacuoles, which provide turgor pressure to push the cytoplasm to the apical region of the growing pollen tube. Many calcium precipitates are present in the stylar transmitting tract, which displays a calcium gradient: fewer precipitates are localised in the distal (upper) transmitting tissue below the stigma, and more precipitates are present in the transmitting tract at the style base. The emporal and spatial distribution of calcium in the stigma and style of medlar suggests that it satisfies the demand for calcium in vivo and played some functional significance.  相似文献

该研究以金线莲不同发育时期的花药为材料,采用电子显微镜观察花粉块中的钙离子分布,以揭示钙离子在金线莲花药发育中的相关生理功能。结果发现:(1)在造孢细胞时期,较多的钙沉淀颗粒出现在花药表皮和药室内壁细胞的液泡中,暗示钙离子与植物细胞的液泡发生和形成有关。(2)在减数分裂前期,小孢子母细胞核中聚集了较多的钙沉淀颗粒,当小孢子母细胞分裂时,在二组染色体之间有大量的钙沉淀颗粒,显示钙离子与细胞分裂有关。(3)在合成淀粉的质体表面覆盖了较多的钙沉淀颗粒,显示钙离子与质体中的糖代谢有关。研究表明,开花时在花粉块表面的花粉外壁上和成熟花粉中仍保持有大量的钙沉淀颗粒,为花粉萌发所需钙离子做好了储备。  相似文献

采用焦锑酸钾沉淀法研究了温敏雄性不育水稻(Oryza sativa L.)培矮64S在高温引起雄性不育与正常可育花药发育过种中Ca2+的分布变化.结果表明,当培矮64S生长在较高温度条件下引起雄性不育,与可育花药相比,不育花粉母细胞中有较多的液泡、较多的Ca2+沉积和较少的线粒体,并且有较多的Ca2+沉积在不育花药的中间层、表皮层和绒毡层中.到四分体与单细胞花粉时期,不育花药的木质部细胞的次生加厚壁上有较多的Ca2+沉淀,连接组织中的Ca2+沉淀也大大增加,所有不育花粉外壁较厚而发育都不正常.在单核细胞早期,不育花粉的四分体细胞中有较明显的大液泡出现.不育花药中的Ca2+在花药发育的各时期均比可育花药要多.这些结果说明在高温生长条件下,花粉母细胞发育的异常、花药中Ca2+沉积的增加、绒毡层与花粉外壁发育的异常可能与培矮64S花粉败育相关.  相似文献

Calcium was detected in CaCl (10 m M )-pretreated roots of Zea mays L. (cv. LG 11) by means of electron probe X-ray microanalysis with energy-dispersive spectrometry (EDS) using embedded samples, fixed by the antimonate staining procedure. A high level of calcium was found where large amounts of antimonate precipitates were observed by light or transmission electron microscopy. In the elongation zone, after 20 h in humid air following a 2 h CaCl2 pretreatment, the level of calcium was higher in trichoblasts than in atrichoblasts. In these cells it was detected mainly in the walls and nucleus, and antimonate staining was observed in the walls. Abundant precipitates containing calcium were associated with the nucleus, vacuoles, mitochondria and endoplasmic reticulum of non-differentiated cells, whereas they were confined to the walls of these cells just after the CaCl2 pretreatment. The involvement of calcium in the formation of root hairs is discussed.  相似文献

It has been reported that PG is a key enzyme related to the tomato fruit ripening and that the application of calcium can dramatically decrease the PG activity and delay the ripening of fruits. In this paper the effects of calcium treament at various ripening stages on the transformation of absorbed calcium, PG activity and PG synthesis in tomato fruits were studicd. According to the analysis of calcium by atomic absorption spectroscopy, it was shown that the soluble and total calcium contents in pericarp of fruits treated with calcium at mature-green stage were increased significantly, and that more soluble calcium was transformed into bound calcium. Both the absorption and transformation of calcium decreased in fruits treated with calcium at later stage of ripening. The inhibition of calcium on PG activity was most effective by treatment at mature-green stage, but less effective at later stage of ripening. One reason for the decrease of calcium inhibition was probably due to the decline of calcium absorption as fruit ripening. The polyacrylamide gel electrophoresis of PG showed that PG with a molecular weight of 46.7 kD was absent in mature-green fruits, and PG synthesis occurred only at the later stage of ripening. It seems that the earlier the treatment was done the more effective of the calcium inhibition of PG synthesis. Based on the above results, it was concluded that the PG plays a major role in ripening and senescence of tomato fruits, and both PG synthesis and its activity were inhibited by calcium. In order to delay the ripening and senescence of tomato fruits, the treatment with calcium should be done at mature-green stage.  相似文献

Calcium ions have important roles in cellular processes including intracellular signaling, protein folding, enzyme activation and initiation of programmed cell death. Cells maintain low levels of calcium in their cytosol in order to regulate these processes. When activation of calcium-dependent processes is needed, cells can release calcium stored in the endoplasmic reticulum (ER) into the cytosol to initiate the processes. This can also initiate activation of plasma membrane channels that allow entry of additional calcium from the extracellular milieu. The change in calcium levels is referred to as calcium flux. A key protein involved in initiation of calcium flux is Stromal Interaction Molecule 1 (STIM1), which has recently been identified as a sensor of ER calcium levels. STIM1 is an ER transmembrane protein that is activated by a drop in ER calcium levels. Upon activation, STIM1 interacts with a plasma membrane protein, ORAI1, to activate ORAI-containing calcium-selective plasma membrane channels. Dysregulation of calcium flux has been reported in cancers, autoimmune diseases and other diseases. STIM1 is a promising target in drug discovery due to its key role early in calcium flux. Here we review the involvement and importance of STIM1 in diseases and why STIM1 is a viable target for drug discovery. 应用焦锑酸钾原位定位法对大熊猫精子获能和顶体反应过程中进行钙定位研究,发现未获能精子的 Ca2+主要结合于顶体前区和赤道段质膜外侧和顶体内膜内侧(核膜侧);随着获能的进行,Ca2+进入精子内部并主要结合于顶体区质膜内侧和顶体外膜外侧;顶体反应的精子,Ca2+结合于顶体内膜外侧、顶体后区质膜外侧和分散存在于释放的顶体内容物中,有些顶体反应精子的顶体内膜外侧结合的Ca2+特别丰富。精子尾部的Ca2+主要分布于中段线粒体内,且其内所含Ca2+含量随着获能和顶体反应而增加。另外尾部致密纤维和轴丝处也有少量Ca2+分布。  相似文献
