Following centuries of exploitation, basking sharks (Cetorhinus maximus) are considered by IUCN as Endangered in the Northeast Atlantic, where they have now been substantially protected for over two decades. However, the present size of this population remains unknown. We investigated the use of photo-identification of individuals’ dorsal fins, combined with mark-recapture methodology, to investigate the size of populations of basking shark within the west coast of Scotland. From a total of 921 encounters photographed between 2004 and 2011, 710 sharks were found to be individually identifiable based on dorsal fin damage and natural features. Of these, only 41 individuals were re-sighted, most commonly both within days of, and close to the site of, the initial encounter. A smaller number were re-sighted after longer periods of up to two years. A comparison of the distinguishing features of individuals on first recording and subsequent re-sighting showed that in almost all cases these features remained little changed, suggesting the low re-sighting rate was not due to a loss of distinguishing features. Because of the low number of re-sighting we were not able to produce reliable estimates for the long-term regional population. However, for one 50 km diameter study area between the islands of Mull, Coll and Tiree, we were able to generate closed-population estimates for 6–9 day periods in 2010 of 985 (95% CI = 494–1683), and in 2011 of 201 (95% CI = 143–340). For the same 2011 period an open-population model generated a similar estimate of 213 (95% CI = 111–317). Otherwise the low rate and temporal patterning of re-sightings support the view that such local basking shark populations are temporary, dynamic groupings of individuals drawn from a much larger regional population than previously supposed. The study demonstrated the feasibility and limitations of photo-identification as a non-invasive technique for identifying individual basking sharks.  相似文献

Dynamin 2 and dynamin 3 are highly expressed in testis. However, their functions in the tissue remain unclear. Considering that dynamin 1, neuron-specific isoform of dynamin, plays a pivotal role in endocytosis, functions of dynamin 2 and dynamin 3 in testis must be essential. Cellular expression and subcellular localization of dynamin 2 and dynamin 3 in testis were investigated. Dynamin 2 and dynamin 3 were highly expressed in germ cells and Sertoli cells, constituents of seminiferous tubules. By immunofluorescence it was revealed that dynamin 2 colocalizes with clathrin both at the plasmamembrane and at Golgi in a cell line of Sertoli cells. Immunoreactivity for dynamin 3, on the other hand, appeared as finer puncta, which did not colocalize with clathrin, suggesting that these two dynamins have distinct functions in Sertoli cells. In the klotho deficient mouse testis, which demonstrates disorder in spermatogenesis, expression of dynamin 2 and dynamin 3 was drastically reduced indicating possible association of these proteins with spermatogenesis.  相似文献

The reference ranges of the trace elements Al, As, Be, B, Cd, Co, Cu, Fe, Mn, Mo, Ni, Pb, Li, Rb, Se, Sr, and Zn were determined by inductively coupled plasma-mass spectrometry (ICP-MS) in sera of a group of free-ranging plains viscachas of the pampa grasslands of Argentina. The values were compared with those of a small group of captive plains viscachas of the Zurich Zoo with diabetes and bilateral cataracts. In addition, a method for digestion of whole-blood samples is described for the trace element determination. Significant differences in the concentration of trace elements in the two groups of animals are discussed. No correlation was found between the levels of selenium and of other trace elements compared to the formation of cataracts.  相似文献

The intracellular localizations ofchitinase and β-N-acetylglucosaminidase were detected in turbot blood smears, using a novel method employing fluorogenic substrates. The two enzymes showed different distributions, with chitinase being more generally distributed and N-acetylglucosaminidase being strongly associated with distinct intracellular bodies, probably lysosomes. The fluorogenic substrates were used to analyse soluble and membrane fractions of homogenates of red and white blood cells prepared on Percoll gradients. In the leucocytes, the chitinase and N-acetylglucosaminidase activities were mostly in the soluble fraction. In the erythrocytes the activities were lower, at about one-hundredth and one-tenth specific activities, respectively, and were distributed between soluble and membrane-bound fractions at about 2 : 1 and 3 : 1, respectively. In contrast, lysozyme had a soluble distribution in leucocytes and was not detected in erythrocytes. Plasma was rich in chitinase and lysozyme activities but had no detectable N-acetylglucosaminidase. Two possible roles for the chitinolytic enzymes are discussed: defence against pathogens and processing of glycoproteins or glucosaminoglycans. Evidence for a defence role for the chitinase and lysozyme is provided by demonstrating that they had inhibitory activity against the chitinous fungus Mucor mucedo .  相似文献

Synopsis The final preferred temperatures (FPTs) of adult premigratory and migratory life-history phases of American eels, Anguilla rostrata, were determined by chronic tests in a horizontal thermal gradient. Mean FPTs were between 17 and 20°C and were not significantly different between life-history phases, acclimation temperatures, illumination regimes, photoperiods or sexual maturation states. Thermal behavior of eels was highly variable, both among individuals of the various test groups and among repeated tests of single individuals. Light inhibited behavioral thermoregulation by promoting shelter-seeking. The following inferences are drawn from the laboratory findings and observations of migrating A. rostrata and A. anguilla (European eels) in the North Atlantic: (1) decreasing temperatures may initiate downstream migration of silver eels, (2) eels may select temperatures close to their FPT in thermally stratified environments, but will tolerate higher and lower temperatures depending on illumination or other physical constraints, (3) the oceanic phase of the migration to the Sargasso Sea may take place at relatively shallow depths in the open ocean, probably within the upper 1000 meters. The strong eurythermality observed in eels may facilitate their occupation of and migration through thermally diverse and unpredictable habitats.  相似文献

Summary Pool sizes of ATP and ADP were analysed in freeze-stopped, lyophilised homogenates of needles from Norway spruce [Picea abies (L.) Karst.]. Control experiments in which possible changes in adenylate pools during sample acquisition were investigated did not reveal significant differences between needles taken from branches in situ or within a 30-min period after cutting off a branch. In addition, pool sizes of ADP and ATP were not affected by changes in light intensity (between 60 and 1500 E*-m-2*s-1), which inevitably occur when samples have to be taken from the upper region of older trees. Levels of ATP and ADP showed considerable seasonal changes (May through October) with the highest ratios of ATP/ADP in developing needles. In general, there was a tendency towards increased ratios of ATP/ADP with increasing needle age. This observation was corroborated by analyses of needles from spruce trees of different age and growing under different conditions. Needles from declining trees or from trees specifically fumigated with low concentrations of ozone and sulphur dioxide had significantly increased ratios of ATP/ADP compared to controls. The results are discussed with respect to physiological responses connected with natural senescence and induced ageing.  相似文献

The intrinsic viscosities, apparent molar volumes, apparentspecific volumes and apparent solution parachors of selectedsugars were determined with the aim of investigating the possiblerelationship of these solution properties to the published sweetnessvalues of sugars. For a given sugar, there is no clear relationshipbetween its intrinsic viscosity and other solution propertiesstudied. The values of apparent specific volume and intrinsicviscosity differ greatly even though both parameters reflectthe amount of water surrounding 1 g of the solute. There isalso no observable correlation between the intrinsic viscositiesand the literature values on the relative sweetness of sugars.The intrinsic viscosity of a solute molecule reflects its extentof hydration. It is greatly influenced by the conformation andconfiguration of the sugar molecules. Equatorial OH groups aremore favourable for hydration than the axial OH groups. Thus,the more OH groups in the equatorial position, the higher theintrinsic viscosity.  相似文献

The susceptibility of turbot, coho salmon, and rainbow trout to strains of Vibrio anguillarum of serotypes 01 and 02 and their extracellular products (ECP) was investigated in order to clarify the role of exotoxins in the mechanism of virulence of both serotypes. All V. anguillarum isolates were virulent for trout, salmon, and turbot. Despite the origin of the strains tested, rainbow trout was the most susceptible fish species to experimentally induced vibriosis. Coho salmon and turbot did not differ significantly in their susceptibility to V. anguillarum live cells. In contrast, the ECP from Vibrio strains of serotypes 01 and 02 exhibited similar lethal dose for turbot, salmon, and trout (ranging from 4.52 to 7.32 μg protein/g fish). Therefore, differences in susceptibility to vibriosis are not completely due to a differential sensitivity of fish to the extracellular products of Vibrio strains. The ECP from 7 of 10 V. anguillarum strains possessed vascular permeability factors, and all the extracts displayed proteolytic, hemolytic and cytotoxic activities. All the biological activities of ECP were lost after heat treatment at 80° C/10 min.  相似文献

Changes in the stem radius of young Norway spruce [Picea abies (L.) Karst.] were related to changes in stem water content in order to investigate the relationship between diurnal stem size fluctuations and internally stored water. Experiments were performed on living trees and on cut stem segments. The defoliated stem segments were dried under room conditions and weight (W), volume (V), and xylem water potential (Os) were continuously monitored for 95 h. Additionally, photos of cross-sections of fresh and air-dried stem segments were taken. For stem segments we found that the change in V was linearly correlated to the change in W as long as Os was >-2.3ǂ.3 MPa (phase transition point). Stem contraction occurred almost solely in the elastic tissues of the bark (cambium, phloem, and parenchyma), and the stem radius changes were closely coupled to bark water content. For living trees, it is therefore possible to estimate the daily contribution of "bark water" to transpiration from knowledge of the stem size and continuous measurements of the stem radius fluctuations. When Os reaches the phase-transition point, water is also withdrawn from the inelastic tissue of the stem (xylem), which - in the experiment with stem segments - was indicated by an increasing ratio between (V and (W. We assume that for Os below the transition point, air is sucked into the tracheids (cavitation) and water is also withdrawn from the xylem. Due to the fact that in living P. abies Os rarely falls below -2.3ǂ.3 MPa and the xylem size is almost unaffected by radius fluctuations, dendrometers are useful instruments with which to derive the diurnal changes in the bark water contents of Norway spruce trees.  相似文献

Human parathyroid hormone (hPTH) and several deletion analogues were examined for the presence of secondary structure using circular dichroism spectroscopy. The spectra of hPTH and the deletion analogues 8-84, 34-53, 53-84, 1-34, 13-34, 1-19, and 20-34, in neutral, aqueous buffer, gave no evidence for extensive secondary structure. An alpha-helical-like spectral contribution was found to arise from a region within peptide 13-34. This spectral contribution was speculated to arise from partial stability of a helix consisting of residues 17-29. Molecular dynamics simulations of peptide 1-34 suggested that this peptide tends to fold with a bend defined by residues 10-14, with the amino-terminal and carboxyl-terminal residues tending to be in more extended forms and the other residues in helical-like conformations. The addition of trifluoroethanol promoted the formation of alpha-helix, mainly in the 1-34 region. The putative helix comprised of residues 17-29 was stabilized by the addition of 10-20% TFE, while a second putative helix proximal to the amino terminus, and comprised of residues 3-11, was stabilized by slightly higher concentrations of TFE. An amphiphilic sequence was identified within the 20-34 fragment. The development of alpha-helix on binding this fragment, and other analogues containing this sequence, to palmitoyloleoylphosphatidylserine vesicles provided experimental evidence for the potential role of this amphiphilic sequence in binding to membranes or to a membrane receptor. The relationships between these alpha-helical regions in 1-34, either potentiated by trifluoroethanol or lipid vesicles, are discussed in terms of different receptor-binding regions within hPTH.  相似文献

Distribution, sexual maturity, fecundity, genital asymmetry, cyclic vitellogenesis and gestation are described in the angel sharks Squatina squatina (L., 1758) and S. oculata Bonaparte, I840 from Tunisian waters. With embryonic nutrition being purely lecithotrophic, lack of egg capsules, block of oocyte growth at the beginning of gestation, and inability to ovulate soon after parturition, Squatinidae can be compared to Torpedinidae and to Centroscymnus (Squalidae), except that their vitellogenesis starts again at mid-gestation. This new, special feature has been termed 'semi-delayed vitellogenesis' with regard to the delayed vitellogenesis of Torpedinidae and Centroscymnus .  相似文献
