采用RAPD技术,对5个柞蚕品种的遗传差异进行比较研究.结果表明,所采用的40个随机引物中,有27个引物扩增谱带清晰且重复性较好,扩增总片段数253条,单个引物的扩增片段数在4~16之间,片段大小在0.33~3.0kb之间.不同柞蚕品种间的遗传差异较小,遗传距离(D)在0.066~0.1659之间,根据D值,由UPGMA聚类分析软件绘制了它们的分子进化树。 Abstract:Random amplified Polymorphic DNA (RAPD) was used to analyze the genetic diversity among Antheraea pernyi.The genetic variance of five Antheraea pernyi was studied.The result showed that:27 of 40 arbitrary primers could amplify clear and repeating bands.A total of 262 fragments were obtained.Each primer gave 4~16 bands and the average was 9.7.The length of the band was 0.33~3.0kb.The D value between different breeds of Antheraea pernyi was 0.066~0.1659.The D value was used to construct a dendrogram by UPGMA.  相似文献

广东省水稻纹枯病菌遗传多样性与致病力分化的研究   总被引:6,自引:0,他引:6  
利用随机引物扩增多态性DNA(PAPD)分子标记技术分析了来自广东省7个县市48个水稻纹枯病菌菌株的遗传多样性,以筛选出的10个随机引物对菌株进行PAPD-PCR扩增,共产生了98个PAPD分子标记,其中89.9%的片段具有多态性。48菌株间的遗传相似性系数(以Nei基因一致度表示)在0.56-0.949之间,用UPGMA和类分析可将它们分为5个RAPD遗传聚类群(A、B、C、D、E),相同地区来源的菌株基本上聚类在同一组群内。在温室中对供试菌株进行致病性测定,结果表明所有菌株对水稻品种Tetep都有致病性,菌株间致病力差异显著(α=0.05),病情指数范围为0.73-18.7,平均感指病指数为5.24。试验分析结果表明水稻纹枯病菌具有丰富的遗传多样性,不同县市的菌株存在很大的遗传分化现象(FST=0.579),RAPD遗传聚类群的划分与菌株的地理来源有明显的相关性,但菌株的致病力差异与菌株的来源、遗传聚类组群的划分没有明显的相关性。  相似文献

取30个枣树品种进行ISSR分子标记分析,其扩增条带分别进行琼脂糖和聚丙烯酰胺凝胶电泳检测,以期获得不同产地品种之间的遗传多态性。从100条选择扩增的ISSR引物中筛选出17条扩增清晰、重复性和稳定性好的引物,选取其中8条扩增条带多态性强的引物进行遗传聚类分析。结果表明:(1)1%琼脂糖凝胶电泳和5%聚丙烯酰胺凝胶电泳检测扩增总条带数分别为72和127条,其中多态性条带分别为51条和113条,多态性条带比率(PPB)分别为70.8%和88.9%。(2)基于UPGMA软件对30个品种的遗传差异性分析表明,8个ISSR引物可以将枣树品种之间遗传差异明显区分开来。两种电泳检测方法相比较,聚丙烯酰胺凝胶电泳检测可获得较为精细的枣树品种间遗传图谱。其遗传相似系数范围在0.56~1.00之间,以0.62为最低遗传相似系数,可将30个枣树品种分成3个大类,6个亚类,为进一步研究枣树品种间分类、起源进化关系和分子辅助育种奠定基础。  相似文献

利用SSR和AFLP两种分子标记技术,分析了52份转基因抗虫棉品种(系)的遗传多样性。结果表明:在61对SSR引物中,有4对引物在供试材料中表现出多态性,共扩增出102个标记,其中多态性标记25个,多态性百分率为24.51%,每对引物的扩增带数变化在17~30之间;在100对AFLP引物中,有9对引物在供试材料中产生多态性,共扩增出618个标记,多态性标记33个,占总数的5.34%,每对引物组合扩增的标记数分布于47~81之间。成对品种的欧式距离变化在2.00~5.57之间,平均值为4.21,单一品种欧氏距离的平均值分布在3.73~4.75之间,表明不同品种之间遗传差异不大。基于SSRs和AFLPs多态性数据的聚类分析,可以将供试材料划分为3个类群(SAGs),但类群划分与品种地理来源不十分吻合。  相似文献

利用22对SSR引物的扩增结果计算品种间的Jaccard相似系数,在此基础上用UP(GMA方法进行了聚类分析,检测了43个春小麦品种间在DNA水平上的遗传变异。22对引物共扩增出102条多态性带,平均每对引物可扩增出4.64条多态性带,具有较好的多态性。SSR水平上43个品种间遗传距离变异范围为0.2222~0.8393,平均遗传距离GD%=0.6055。43个品种聚为两大类,除佛手麦自成一类外,其余42个品种聚为第二大类。聚类结果真实地反映了品种间基因型差异。历史上地方品种间SSR水平上的遗传变异最大,育成品种遗传多样性水平总体上呈下降趋势,且低于地方品种和引进品种。1BL/1RS易位片段特异性引物Rye检测结果显示,共有7个品种含有1RS片段,结果需进一步证实和深入研究。  相似文献

采用RAPD技术,利用200个引物筛选的22个10碱基随机引物对桃182个变种,类型,品种进行DNA扩增。对扩增位点上的带型分析,观察到有的引的在某一位点上明显有带的分离,且S65,S459,S167,S60引物在这一位点可以包含一个类群或几个类群全部的供试品种,变种或类型,但各类群都没有各自独特的特征带。聚类图分析表明,来源无性系的品种遗传一致度最大达0.990;有的品种也表现出较大遗传一致度的为0.985;也有的品种的遗传一致度较低,范围在0.686-0.831之间。对桃的分类,以水蜜桃,寿星桃,垂枝桃作为类群划分较明晰,其它品种的类群划分则较困难,从而也说明了原产中国的桃种质资源的遗传多样性丰富。  相似文献

肌肉生长抑制素是控制骨骼肌生长发育的重要细胞因子,采用PCR-SSCP和测序的方法发现了5个位于Myostatin基因5′-和3′-调控区的单核苷酸多态性位点,对北京油鸡、白耳鸡、石歧杂、矮小黄鸡、小型黄鸡、惠阳胡须鸡、隐性白羽鸡、海兰、AA鸡等不同鸡种的该单核苷酸多态性分析结果表明:Myostatin基因的5′调控区引物P60/P61扩增片段多态性是由3个核苷酸的改变而产生的[分别是G→A(304位)、A→G(322位)、G→(344位)],引物P93/P94扩增片段的多态性是由G→A(167位)突变造成的,引物P117。P118PC扩增片段多态性是由T→C(177位)造成的。3′调控我引物P80/P81扩增片段多态性是由第7263位A突变为T造成的,引物P76/P77扩增片段多态性是由A→G(6935位)造成的。不同鸡种群体遗传学分析表明,5′-调控区引物60/P61扩增片段多态性片段多态性是由A→G(6935位)造成的。不同鸡种群体遗传学分析表明,5′-调控区引物P60/P61扩增片段多态性位点在北京油鸡的基因型频率分布与其他的品种有很大的差异,其BB型频率为0.700,AA基因型频率仅为0.033,而其他鸡种中以A基因优势;对于引物P93/P94,品种间的基因型频率差异极显著(P<0.01),北京油鸡和AA鸡的EE型频率鸡种中以A基因占优势;对于引物P93/P94,品种间的基因型频率差异极显著(P<0.01),北京油鸡和AA鸡的EE型频率低于其他品种,白耳鸡和海兰蛋鸡以EE型为主,其频率高于其他品种;3′-调控区引物P80/P81多态怀位点在9个鸡种中都是等位基因C占优势。引物P76/P77,总体上MM型的频率较低,杂合子MN型的频率较高。  相似文献

利用RAPD技术对香蕉(Musa nana Lour.)33个品种的遗传变异进行了研究,从249个随机引物中筛选出18个有效引物,用它们共扩增出192条DNA带,其中183条为多态性带,占95.31%,平均每个引物扩增的DNA带数为10.67条,利用18个有效引物扩增的192条DNA带对香蕉33个品种间的亲缘关系进行UPGMA聚类分析,计算出33个品种间的平均遗传距离为0.3412。在此基础上建立了香蕉33个品种的DNA分子系统树状图。该系统将香蕉33个品种划归A,B,C和D4个群,其中A群20个品种,B群5个品种,C群2个品种,D群6个品种;A群又可以分为3个亚群。对香蕉遗传多样性分子基础进行了探讨。  相似文献

本文对家蚕、野桑蚕、蓖麻蚕、柞蚕和天蚕等5种绢丝昆虫进行了随机扩增多态DNA(RAPD)分析。40个引物中有27个引物能扩增出536个清晰且重复性强的条带,其中可变条带数为520个,单个引物扩增的条带数在11~28之间,平均为19.9,各片段分子量大小在0.29~2.67kb之间。每个样本都能找出其独特的分子标记。家蚕与野桑蚕的遗传距离(D)最小,为0.3760;家蚕与蓖麻蚕的遗传距离(D)最大,为0.7488。根据遗传距离,用UPGMA聚类分析方法构建了它们的分子树。 Abstract:Five species of silk insects including Bombyx mori, B. manolarina, Philosamia cynthia, Autheraea pernyi and A. yamamai were analyzed by RAPD method using 40 arbitrary primers. In these primers, 27 of them could amplify clear and repeating bands. 536 fragments were obtained and the variable bands were 520. Each primer gave 11~28 bands and the average was 19.9. The length of the fragments is 0.29~2.67 kb. Some distinctive bands were found in every species. The genetic distance(D) between bombyx mori and B. manolarina is 0.3760, which is the lowest. The highest D value is 0.7488, which between Bombyx mori and Philosamia cynhia. The D value was then used to construct a dendrogram by unweighted pair-group method with arithmetical averages(UPGMA).  相似文献

The immune system in the Chinese oak silk moth, Antheraea pernyi, has been compared with that of the Cecropia moth which has been characterized earlier. Antibacterial activity against Escherichia coli was induced in diapausing pupae by injection of viable E. coli or Enterobacter cloacae. The activity reached a maximum on day 7-8 after which the response gradually declined. The pupae produced a set of immune proteins with P4 and P5 as major labelled components similar to that earlier found in Cecropia. The major antibacterial factor in A. pernyi was cecropin D. A procedure is described for the isolation of cecropin B and D, which is in principle similar to the one used for the isolation of the corresponding cecropins from Cecropia pupae. Amino acid sequence analyses of the A. pernyi cecropins show the D form to contain 36 amino acid residues and that both cecropins have blocked C-termini. The general structure of cecropins having a charged N-terminal region (residues 1-21) followed by a long hydrophobic stretch (residues 22-32) is well conserved. Cecropin B and D from A. pernyi differ from the corresponding proteins in Cecropia by four and three conservative amino acid replacements, respectively. The homology between the cecropins from the two insects suggests that they orginate from a single ancestral gene. The antibacterial activity was tested against nine different bacterial species. Evolutionary aspects of the cecropins are discussed.  相似文献

利用~(60)Co-γ射线对柞蚕卵进行辐射,探讨了~(60)Co-γ射线辐射对柞蚕生物学特征的影响.结果表明:120 Gy以下辐射剂量对柞蚕卵孵化率及柞蚕死亡率影响不大,但随着~(60)Co-γ射线辐射剂量的增加,孵化率明显降低而死亡率显著升高;血液蛋白质图谱和相关基因序列在辐射前后亦存在差异.这些生物性状的改变说明~(60)Co-γ射线对柞蚕生长发育具有重要的影响.  相似文献

The genetic diversity and genetic structure of three Chinese silkworm species Bombyx mori L., Antheraea pernyi Guérin-Meneville and Samia cynthia ricini Donovan were comparatively assessed based on RAPD markers. At the species level, A. pernyi and B. mori showed high levels of genetic diversity, whereas S. cynthia ricini showed low level of genetic diversity. However, at the strain level, A. pernyi had relatively highest genetic diversity and B. mori had lowest genetic diversity. Analysis of molecular variance (AMOVA) suggested that 60% and 72% of genetic variation resided within strains in A. pernyi and S. cynthia ricini, respectively, whereas only 16% of genetic variation occurred within strains in B. mori. In UPGMA dendrogram, individuals of A. pernyi and B. mori formed the strain-specific genetic clades, whereas those of S. cynthia ricini were distributed in a mixed way. The implications of these results for the conservation and utilization in breeding programs of three silkworm species are discussed.  相似文献

本实验测定了柳蚕卵表面的胶着物质的含量以及粘液腺的形状、大小,并对粘液腺的内容物进行了SDS一聚丙烯酰胺凝胶电泳分析。结果表明:柳蚕卵表面的胶着物质的含量为1.11mg/卵,与柞蚕(1.01mg/卵)相当。柞蚕、柳蚕雌蛾的粘液腺整体呈较细长的管状结构且几乎无枝状部分枝,柳蚕粘液腺的干状部长约为2.0cm,柞蚕为1.8cm,二者相近。柳蚕粘液腺的内容物经SDS-聚丙烯酰胺凝胶电泳分析出现了7条蛋白质谱带,分子量分别为114kD,92kD,78kD,53kD,50kD,23kD,12kD,与柞蚕存在差别。  相似文献

了解柞蚕Antheraea pernyi感染微孢子虫初期血淋巴内免疫系统及刺激应答相关蛋白质种类, 本研究以柞蚕5龄雌幼虫的起蚕(结束4眠, 刚完成蜕皮的幼虫)添食柞蚕微孢子虫Nosema pernyi为材料, 对感染后血淋巴利用SDS-PAGE进行分离后, 利用LC-MS/MS质谱技术和蛋白质组学分析对差异蛋白质条带进行鉴定。结果显示: 感染微孢子虫144 h后, 血淋巴中分子量约为44 kD (AP44)和28 kD (AP28)的蛋白质条带表达量增高。质谱分析AP28和AP44蛋白质条带样品, 共鉴定117个不重复蛋白质, 其中2个样品共有蛋白质12个, AP28独有蛋白质52个, AP44独有蛋白质53个。对质谱数据利用COG数据库进行搜寻鉴定, 显示AP28和AP44的鉴定蛋白质中涉及柞蚕免疫系统及刺激应答生物过程的蛋白质共有29个, 其中AP28中包括热激蛋白、 泛素样蛋白、 泛素结合酶E2、 保幼激素环氧水解酶、 微管结合蛋白、 溶菌酶、 ADP-核糖基化因子、 防御蛋白、 肽聚糖识别蛋白等15个, AP44中包括DRK、 酚氧化酶原、 类免疫球蛋白等10个; 二者共有热激蛋白hsp21.4、 酚氧化酶原、 抗菌肽等4个。本研究结果可以为今后研究柞蚕对微孢子虫的免疫应答及防御机制提供参考。  相似文献

Molecular tools opened a new vista to understand nature's bio-diversity and its relevance and the same approach was availed of to build-up the foundation work on the bio-diversity of silkworm spp in India. It is well established that the heritage of usage of silk for dress materials in India, Russia and China dates back to premedieval period and in spite of industrial development resulting to de-forestation, India still can claim as the owner of wide bio-diversity, espicially in northern India for silkworm spp. The molecular diversity was assessed among Antheraea mylitta, A. assama, A. pernyi, A. provlei, A. roylei and Philcosomia cynthia wiht 11 ISSR and 8 non-random primers on agarose gel. Nei's statistics as also Euclidean distance matrix was applied to find the genetic diversity between the six species, wherein the closest relationship between A. pernyi and A. proylei is established. With the help of POPGEN statistics, the average genetic heterozygosity appeared as 0.271 while Shanon's index is 0.4312 and alleles with segregation ratios of 3:1, 2:1, 1:1, 9:1 (generated with ISSR primers) were identified which can be utilized for future molecular breeding porgram. Further, and attempt was made to isolate a number of bands generated with 3 ISSR and six non-random primers, specific for different species and 22 such markers have been characterized through sequencing which will be made available through international public domain database.  相似文献
