The mineral requirements of a strain ofAspergillus niger for the production of citric acid in a synthetic medium were studied. It was observed that K2HPO4 and MgSO4.7 H2O were required at concentrations of 0.1% and 0.02% respectively. The optimum level of each of the trace elements Fe, Mn and Zn was 1.0 μg/ml. NaCl and CaCl2 at lower concentrations had no effect on citric acid production. Trace elements, Cu, Co and Mo, had an adverse effect on the production of citric acid while Ni and V were without effect.  相似文献

Summary The recycled solid-state surface fermentation (SSF) culture ofAspergillus niger KCU520 was used for repeated batch production of citric acid from sugarcane molasses. The rate of citric acid production was doubled, reducing the fermentation time to half, compared to the normal single cycle batch submerged or surface fermentation process. About 80% sugar was converted to citric acid in five-day batch fermentation and three batches were carried out with the same fungal mat without any significant loss of productivity.  相似文献

Protoplasts from a prototrophicAspergillus niger strain were first inactivated at 55°C for 12 min with a regeneration frequency of 3.5×10−6, and then fused with living protoplasts from an auxotrophic strain. The fusion frequency was 1.1×10−5. Some fusants segregated sectors of prototrophic recombinants Citric acid production in submerged cultivation of these recombinants was examined. Summary A large number of submerged citric acid fermentations in a beet molasses substrate was studied. The development of Aspergillus niger from conidia to pellets was followed. Rheological characteristics of the fermentation broth including the pellets were determined. The results obtained confirm the fact that the non-Newtonian pseudoplastic behaviour of the fermentation broth was due to the presence of mycelial pellets. Several tens of mutants were obtained by UV irradiation of a spore suspension ofAspergillus niger. Producers yielding large amounts of citric acid were selected on a moditied Czapek agar containing methyl red as pH indicator. After a submerged cultivation in flasks with baffles, a mutant characterized by yellow pigmentation on wort agar and by yields of citric acid up to 74.6% in the medium containing glucose was chosen from 130 isolates tested.  相似文献

Aspergillus niger has been grown in glucose- and maltose-limited continuous cultures to determine the bioenergetic consequences of the production of the extracellular enzyme glucoamylase. Growth yields (g biomass per mol substrate) were high, indicating that growth was very efficient and protein production for biomass was not exceedingly energy consuming. It has been found that the energy costs for the production of this extracellular enzyme is very high. Depending on the efficiency of energy conservation the glucoamylase protein yield on ATP is between 1.3 and 2.6 g protein per mol ATP, which is equal or less than 10% of the theoretical maximum of 25.5. These high energy costs most probably have to be invested in the process of excretion. A comparison between an industrial over-producing strain and the wild typeAspergillus niger showed that this over-producing strain most probably is a regulatory mutant. Two regions of specific growth rates could be determined (one at specific growth rates lower and one at specific growth rates higher than 0.1 h-1), which are characterized by differences in mycelium morphology and a significant deviation from linearity in the linear equation for substrate utilization. Analysis of the region of specific growth rates higher than 0.1 h-1 yielded maintenance requirements of virtual zero. It has been concluded that for a good analysis of the growth behaviour of filamentour fungi the linear equation for substrate utilization is not suitable, since it contains no term for the process of differentiation.  相似文献

Work has been carried out on the production of citric acid in batch culture using Aspergillus foetidus. It is shown that the dry weight increases throughout the fermentation. The supply of nitrogen is exhausted early and the subsequent increase in dry weight is due to accumulation of carbon by the cells. It appears that the exhaustion of nitrogen is a necessary prerequisite for the production of citric acid, which is produced by carbon-storing cells. The pH history in the early part of the fermentation did not affect the final yields.  相似文献

Summary We report the derivation of Aspergillus niger strains having a lower standard deviation in citric acid production by the use of 60Co gamma radiation and by the parasexual cycle. On the basis of the results obtained by isolating diploids and segregants, diploidization seems to be sufficient for reducing variability. Auxotrophic strains ofAspergillus niger were obtained from citric-acid-producing strains of the fungus after irradiation with UV light. Protoplasts were isolated from young hyphae of the auxotrophic strains after treatment with snail enzyme and than treated with polyethylene glycol (30%,W/V), in a Ca2+ (10 mmol/L) solution. The pH value of the suspension was adjusted to 9.0. The frequency of the heterokaryons (related to the number of protoplasts reverting after PEG treatment) was 0.67%. Prototrophic heterozygous spores were isolated from a heterokaryon with the frequency of 1.2×10−6. Citric acid production in the best heterozygous strains was about 15% higher than that of the high-production parent strain.  相似文献

In view of the often-cited theory that citric acid accumulation is caused by an inhibition of aconitase activity, the equilibrium of the reaction of aconitase was investigated by comparing in vivo steady-state concentrations of citrate and isocitrate in Aspergillus niger grown under various citric acid-producing conditions. With the equilibrium catalyzed by the A. niger enzyme in vitro, similar values were obtained. The validity of our in vivo measurements was verified by the addition of the aconitase inhibitor fluorocitrate, which appreciably elevated the citrate:isocitrate ratio. The results strongly argue against an inhibition of aconitase during citric acid fermentation.  相似文献

Summary The spores of Aspergillus niger were entrapped in calcium-alginate beads and precultivated in growth media with various amounts of nitrogen. During the following citric acid production in shaking cultures an optimum of acid formation and yield was observed after the precultivation with 100–200 mg/l NH4NO3. The productivity of the immobilized Aspergillus was found to be 1.5 times higher than in the case of free pellets. The outgrowth of free mycelia into the medium could be provided by increasing the ratio particle-volume: medium volume, using a 1-l air-lift fermenter, by which means the productivity was increased twice as much as obtained in shaking culture.  相似文献

Citric acid production in surface culture process with reuse of mycelia for more than one batch of media has been reported in this work. A strategy for replacing media and thus increasing productivity in subsequent batches is discussed. The activity of glutamine synthetase fromAspergillus niger was significantly lowered under conditions of citric acid fermentation. The intracellular pH of the organism as determined by bromophenol blue dye distribution and fluorescein diacetate uptake methods was relatively constant between 6·0–6·5, when the pH of the external medium was varied between 2·3–7·0.Aspergillus niger glutamine synthetase was rapidly inactivated under acidic pH conditions and Mn2+ ions partially protected the enzyme against this inactivation. Mn2+-dependent glutamine synthetase activity was higher at acidic pH (6·0) compared to Mg2+-supported activity. While the concentration of Mg2+ required to optimally activate glutamine synthetase at pH 6·0 was very high (≥ 50 mM), Mn2+ was effective at 4 mM. Higher concentrations of Mn2+ were inhibitory. The inhibition of both Mn2+ and Mg2+-dependent reactions by citrate, 2-oxoglutarate and ATP were probably due to their ability to chelate divalent ions rather than as regulatory molecules. This suggestion was supported by the observation that a metal ion chelator, EDTA also produced similar effects. Of the end-products of the pathway, only histidine, carbamyl phosphate, AMP and ADP inhibitedAspergillus niger glutamine synthetase. The inhibitions were more pronounced when Mn2+ was the metal ion activator and greater inhibition was observed at lower pH values. These results permit us to postulate that glutamine synthesis may be markedly inhibited when the fungus is grown under conditions suitable for citric acid production and this block may result in delinking carbon and nitrogen metabolism leading to acidogenesis  相似文献

The present investigation explored the possible use of a rarely used agro-industrial by-product, maize starch-hydrolysate, for economic production of citric acid. To achieve this, seventeen strains of Aspergillus niger were screened for their capacity to produce citric acid using starch-hydrolysate as a substrate. The most efficient strain, ITCC-605 was selected for further improvement in citric acid content by mutation. Mutants developed by treatment with EMS and UV, singly and in combination, produced citric acid in the range of 0.51-64.7 g kg(-1) of glucose consumed. The mutant UE-1 produced the maximum citric acid which was about 130 times more than that produced by the parent strain, ITCC-605. For further increase in citric acid production from this substrate, the cultural conditions were optimized: concentration of starch-hydrolysate, 15% (glucose equivalent); ammonium nitrate, 0.25%; KH2PO4, 0.15%; nicotinic acid, 0.0001% and initial pH of 2.0. Under these conditions, the mutant strain UE-1 yielded 490 g citric acid kg(-1) of glucose consumed in 8 days of incubation at 30 degrees C. The productivity of 341 mgl(-1)h(-1) corresponded to 49% substrate conversion to citric acid.  相似文献
