CALCIUM FLUXES IN CULTURED AND BULK ISOLATED NEURONAL AND GLIAL CELLS   总被引:3,自引:3,他引:0  
Abstract— The influx and efflux of 45Ca has been studied in cultured human glioma and mouse neuroblastoma cells and in isolated fractions enriched in synaptosomes, neuronal and astrocytic perikarya from rabbit brain.
The uptake of 45Ca was somewhat more efficient in glioma compared to neuroblastoma cells, whereas there was little difference in the rate of 45Ca uptake by isolated glial cells and neuronal perikarya. Isolated synaptosomes showed the highest rate of 45Ca accumulation. An increase of K concentration to 50 m m in the medium, with a corresponding lowering of Na, stimulated both glioma and glial as well as synaptosomal 45Ca uptake more markedly than the uptake by neuroblastoma cells and neuronal perikarya. Lowering the Na concentration and replacing it by choline had no effect on the cultured cells and astrocytes. Na-free media caused massive stimulation of 45Ca influx in all fractions and cells tested.
The efflux of 45Ca was studied after preloading of cells. Three phases could be resolved from the desaturation curves. All cells had nearly similar half-lives for 45Ca efflux under standard conditions. Pulses of media containing 50 m m -K stimulated 45Ca efflux from glioma cells and astrocytes more efficiently than from neuroblastoma cells, neuronal perikarya and synaptosomes. The stimulated release was exclusively seen in Ca-containing media in experiments with the cultured cells and in Ca-free media in experiments with cell perikarya. The effect of transmitter pulses on the release of 45Ca was examined in a limited series. Acetylcholine and isoproterenol were found to stimulate 45Ca release more actively from glia than from neurons.  相似文献

Abstract— Movement of 45Ca into and out of cerebral slices in vitro was altered by incubation in Na-free medium or in the presence of ouabain. 45Ca uptake into the 'non-inulin' space was more than doubled in slices that were losing Na into Na-deficient media. Replacement of Na in the incubation medium by choline chloride reduced the efflux of 45Ca into Ca-free medium. In the presence of ouabain (10-4M), the rate of efflux was also reduced. NaCN or NaCN plus ethacrynic acid increased the rate of 45Ca efflux. The data are compatible with a sodium-calcium exchange mechanism in cerebral slices.  相似文献

Ca-Cd interaction in the prymnesiophyte Cricosphaera elongata   总被引:1,自引:1,他引:0  
Abstract. 45Ca and 109Cd uptake were followed in Criscosphaera elongata Prymnesiophyceae. In both cases, after a rapid increase for the first 5 min, the incorporation rate slowed during the hour of observation. Verapamil, a blocker of voltage-dependent slow calcium channels, inhibited 45Ca uptake except during the first rapid phase when adsorption should predominate. Cadmium also decreased 45Ca labelling, suggesting that the two metals are antagonistic. However, verapamil was shown to augment 109Cd incorporation, contrary to what occurs in animals cells; this effect is detectable in continuous labelling as well as in pulse experiments. The data support the presence of calcium channels in the alga and suggest several processes in Cd accumulation: adsorption on peripheral envelopes and diffusion of uncharged Cd.  相似文献

The effects of AICI3 on uptake of Ca2+ and phosphate in roots of intact beech ( Fagus sylvatica L. provenance Maramures) plants were studied in nutrient solution and soil solution. Aluminium reduced the concentrations of Ca, Mg and P in plants and increased that of K. In short term experiments, uptake of Ca2+(45Ca) was reduced by exposure of the roots to Al. The effect of aluminium on Ca2+(45Ca) uptake was immediate and primarily of a competitive nature, preventing Ca2+ from being adsorbed. Uptake of 32P-phosphate increased with increasing Al concentration up to 0.1 m M and then decreased at higher Al concentrations. The effect of Al on 32P-phosphate uptake was most pronounced during the first hours of exposure. Growth of plants for 15 days in soil solution, collected from the upper A horizon of a beech forest soil, had no effect on uptake of Ca2+(45Ca) and 32P-phosphate, probably because of a low concentration of labile bound monomeric Al and binding of Al to organic compounds. Soil solution from the deeper B horizon reduced Ca2+(45Ca) uptake and increased 32P-phosphate uptake in a manner similar to that with Altreatment in nutrient solution. It is concluded that in soil solution from the deeper regions of the soil, mineral uptake by roots was affected by Al.  相似文献

Abstract: The activities mediated by the N -methyl-D-aspartate (NMDA) receptor were studied in cultured rat cerebellar granule cells. Micromolar concentrations of the metal binding compounds, EDTA, cysteine, and histidine, as well as serum albumin strongly potentiated receptor activity in the presence of millimolar concentrations of Ca2+ and Mg2+. The findings indicated that these agents remove an endogenous metal, probably Zn2+, which attenuates NMDA receptor-mediated 45Ca uptake and toxicity. Several added metal ions were therefore tested at low micromolar concentrations. Zn2+ was found to be the most potent inhibitor of NMDA-induced 45Ca uptake, followed by Cu2+ and Fe2+. Co2+, Cd2+, Fe3+, and AI3+ had no significant effect, whereas Ni2+ potentiated the 45Ca uptake but inhibited at much higher concentrations. The potentiating agents that remove the endogenous metal had a particularly dramatic effect in the presence of Mg2+, the voltage-dependent suppressor of the NMDA receptor. Mg2+ also played an important role in the inhibitory effect of added Zn2+. Much lower concentrations of Zn2+ were needed to achieve inhibition of NMDA-induced 45Ca uptake in the presence of Mg2+. Under a variety of conditions, a very good correlation was found between NMDA receptor-mediated 45Ca uptake and the magnitude of acute neurotoxicity.  相似文献

To examine otolith resorption induced by anaerobic stress, 45Ca-prelabelled goldfish, Carassius auratus , were kept in oxygen-deficient ambient water (O2 < 0.5 .1 1−1, 26° C) for 2 days and otoliths (asterisci) were analysed for 45Ca retention. In a second experiment, fish were anaerobically stressed for 2 days, and then received a single intraperitoneal injection of 45Ca. They were maintained under stress for one more day and killed to examine 45Ca deposition in otoliths. Plasma was analysed for total and radioactive calcium. Otoliths (lapilli) were examined for stress-induced check formation by scanning electron microscopy (SEM).
Stress significantly reduced plasma calcium levels and the rate of calcium retention in otoliths, which was calculated from the 45Ca specific activity of the plasma. On the other hand, the rate of calcium deposition in otoliths was the same in the stressed as in the control fish. SEM observation revealed that the applied stress resulted in a check formation in otoliths. These results indicate that a 48-h stress of oxygen deficiency induces calcium resorption in otoliths.  相似文献

The aims of this study were to evaluate the growth of Streptococcus sanguis on hydroxyapatite, bovine enamel and polytetrafluoroethylene substrata in a constant depth film fermentor, and to determine the effects of three antimicrobial-containing mouthwashes on biofilm formation and bacterial viability on hydroxyapatite and enamel. There was little difference in the final cell density (5 × 104 cfu mm−2) of the Strep. sanguis biofilm on the three substrata. When hydroxyapatite-grown biofilms were exposed to the mouthwashes for 1 min, the one containing triclosan (T) proved the most effective. The chlorhexidine-containing mouthwash (CX) also achieved significant kills. The T-containing mouthwash was the most effective at killing biofilms grown on enamel. Pre-treatment of hydroxyapatite with CX, cetylpyridium chloride (CPC) or T for 1 min resulted in undetectable biofilm formation after 8 h. After 8 h of growth, only biofilms grown on enamel discs pre-treated with CX showed a reduction in the number of viable organisms. In conclusion, the results of this study have shown that while growth of Strep. sanguis on hydroxyapatite and enamel were similar, the ability of antimicrobial agents to prevent the accumulation of viable bacteria depended on the nature of the substratum.  相似文献

Abstract: Cultured cerebellar granule cells were subjected to toxic activation of the NMDA receptor that was terminated by MK-801. Subsequent resuscitation experiments were mostly conducted in the presence of a physiological concentration of Ca2+. Addition of pyruvate and inorganic phosphate, in addition to glucose, which was always present, rescued ∼40% of the dying neurons. La3+ and ruthenium red were also effective resuscitating agents. The combination of pyruvate, inorganic phosphate, and ruthenium red rescued 65% of the dying neurons. Parallel studies with 45Ca indicated that La3+ and ruthenium red facilitated the decrease of 45Ca in the neurons, whereas inorganic phosphate, supported by energy-yielding pyruvate, formed perhaps, a less harmful Ca complex inside the neurons.  相似文献

Aims:  To conduct a pilot human clinical trial to assess the safety and to test the ability of a probiotic mouthwash, ProBiora3, to affect the levels of Streptococcus mutans and certain known periodontal pathogens in the mouth when administered twice daily over a period of 4 weeks.
Methods and Results:  The mouthwash contained three specific strains of naturally occurring oral bacteria and was tested at two dose levels: 106 and 108 colony forming units each of Strep. oralis strain KJ3sm, Strep. uberis strain KJ2sm, and the spontaneous lactic acid-deficient variant of Strep. rattus , strain JH145. Substantial decreases in the levels of the marker bacteria were observed. No safety issues were noted with the twice daily application of this mouthwash.
Conclusions:  Despite the small number of subjects and the use of young, orally healthy adults, along with the inherent variability in the microbiological measurements, the probiotic mouthwash was able to substantially affect the levels of dental pathogens in saliva and periodontal pathogens in subgingival plaque.
Significance and Impact of the Study:  The results of this pilot human study suggest that the probiotic mouthwash product may be safe for daily use as an aid in maintaining both dental and periodontal health.  相似文献

Electroporation methods for introduction of plasmid DNA into the ruminal bacteria Butyrivibrio fibrisolvens and Streptococcus bovis were developed. Electroporation of the strictly anaerobic B. fibrisolvens was carried out in an anaerobic glovebox with a buffer of 10% (v/v) glycerol and 1 mM MgCl2 in distilled water. Streptococcus bovis electroporation could be carried out aerobically with a buffer of 10% (v/v) glycerol in distilled water. The Escherichia coli/Bacillus subtilis shuttle vector pBS42 could be transformed into B. fibrisolvens strain H17c, selecting for chloramphenicol resistance. The Streptococcus sanguis/E. coli shuttle vector pVA838 could replicate and express erythromycin resistance in Strep. bovis. Both vectors were stable in each organism in the absence of antibiotic selection. While the efficiency was low (<102/μg DNA), the results demonstrate a means to introduce cloned genes into these organisms.  相似文献

All strains of oral streptococci tested and specially those of Streptococcus mutans, Strep. sanguis and Strep. minor produced more than one distinct bacteriocin-like substance with variable inhibitory activity on 20 indicator staphylococci. Inhibitory activity was comparatively higher on nasal strains of Staph. aureus and Staph. epidermidis than on strains of both species isolated from the mouth. Nineteen of 20 staphylococcal indicators were inhibited by 1–12 of the 12 effector streptococci. Sensitivity of nasal staphylococci to bacteriocins (frequency of positive inhibitory tests and total inhibition zone diameters) was significantly higher ( P < 0·001, χ2 test and P < 0·05, t test respectively) than that of oral ones. The sensitivity of nasal over oral Staph. aureus ( P < 0·001 and P < 0·01) and of oral Staph. epidermidis over oral Staph. aureus ( P < 0·01 and P < 0·05) was also significantly higher. The evaluation of variability of inhibitory patterns of bacteriocins produced by streptococci (p-typing), of sensitivity patterns of staphylococci to bacteriocins (s-typing) and of the significantly higher sensitivity of nasal over oral staphylococci to bacteriocins from the epidemiological and ecological veiwpoints are discussed.  相似文献

In cultured cells derived from micromeres isolated at the 16-cell stage of sea urchin embryos, the activity of H+, K+-ATPase became detectable after 15 hr of culture, when the cells started to form spicules, and then increased reaching a plateau from 25 hr of culture. The Na+, K+-ATPase activity of isolated micromeres increased to a maximum at 20 hr of culture and thereafter decreased gradually. Allylisothiocyanate, an inhibitor of H+, K+-ATPase, caused a decrease in intracellular pH (pHi) accompanied by blockage of 45Ca deposition in spicule rods in spicule-forming cells at 30 hr of culture. Ouabain and amiloride had scarcely any effect on the pHi or 45, deposition. In cultured cells exposed to nifedipine, which blocked 45Ca deposition in spicule rods, allylisothiocyanate did not cause any decrease in pHi. These results show that H+, which is generated in the overall reaction to produce CaCO3 from Ca2+ and HCO3, is probably released from the cells mainly in the reaction catalyzed by H+, K+-ATPase to maintain successive production of CaCO3.  相似文献

Streptococcus mutans and Streptococcus mitior were both capable of colonizing the surfaces of human teeth in an artificial mouth. Although fermentable carbohydrate was not essential for colonization, highest numbers were recovered after 5 d if 5–0°***0 (w/v) sucrose had been available intermittently. When grown together in mixed culture the interaction of the two species was affected profoundly by the available nutrients. In the presence of synthetic saliva alone, Strep. mitior was strongly antagonistic to Strep. mutans. When a nutrient broth containing sucrose was also provided intermittently there was slight inhibition of Strep. mutans accompanied by an increase in the proportion of Strep. mitior in the plaque, although the former was the dominant organism under these conditions. When 0–5% (w/v) glucose replaced the sucrose, mutual antagonism occurred and fewer organisms were recovered than if only synthetic saliva had been available. One reason why a high-sucrose diet encourages colonization by Strep. mutans may be that insoluble extracellular polysaccharide confers a competitive advantage upon it in the face of antagonistic agents such as the hydrogen peroxide produced by Strep. mitior.  相似文献

The effect of the copolymer M239, 144 with and without chlorhexidine on the adherence of oral streptococci to saliva-coated hydroxyapatite was investigated. At 1% w/v M239, 144 reduced the adherence of Streptococcus sanguis NCTC 7863 by 94%. It had a moderate effect on the adherence of other Strep. sanguis strains and a Streptococcus gordonii strain but had no effect on the adherence of Streptococcus oralis or Streptococcus mutans. Chlorhexidine did not influence the anti-adhesive properties of 1% w/v M239, 144.  相似文献

THE DISTRIBUTION OF GROUP D STREPTOCOCCI IN CATTLE AND SHEEP   总被引:3,自引:3,他引:0  
Numbers and types of Lancefield group D streptococci have been determined in samples from the colons of 17 cattle and 9 sheep. Mean total streptococcal counts of 8 × 104/g in cattle and 2 × 106/g in sheep were obtained. Streptococcus bovis was found in every sample and was the predominant species in 15 of the cattle and 6 of the sheep. Other group D streptococci ( Strep. faecalis, Strep. faecium and Strep. durans ) were rare in cattle, but in sheep they formed a significant proportion of the population. Of 60 Strep. faecium, Strep. durans and related strains, 51 fermented raffinose. Many of the strains of Strep. faecium were also atypical in that they fermented sorbitol and appreciably reduced tetrazolium in broth at pH 6.0.
Strep. bovis remained the predominant streptococcus in faeces samples from 4 dairy cows when they were tested again after an interval of 17 and 18 months.  相似文献

Dantrolene – an inhibitor of ryanodine receptors and calcium stabilizer – prevents ischemia- and excitotoxicity-evoked neurodegeneration. To elucidate the mechanisms of this phenomenon, we investigated effects of dantrolene on the NMDA- and glutamate-induced lesion and stimulation of 45Ca uptake in primary cultures of rat cerebellar granule neurons. Neurodegeneration was evaluated after 24 h with the propidium iodide staining. Bcl-2 immunoreactivity in cell homogenates was measured by immunoblotting. The results demonstrated that dantrolene applied at micromolar concentrations inhibits in a dose-dependent manner NMDA- and glutamate-evoked 45Ca uptake in neurones and induces neuroprotection. This effect was additive to known effects of DMSO, a vehicle to dantrolene. Dantrolene failed to induce changes in Bcl-2 immunoreactivity. Thus, dantrolene-induced neuroprotection against excitotoxicity may be at least partially mediated by its inhibitory effect on the NMDA receptors.  相似文献

Surface free energies of oral streptococci and their adhesion to solids   总被引:1,自引:0,他引:1  
Abstract The adhesion of 3 strains of oral streptococci from a buffered suspension onto 3 different solid substrata was studied. Representative strains of streptococci were selected on the basis of their surface free energy ( γ b), namely Streptococcus mitis L1 ( γ b= 37 mJ·m−2), Streptococcus sanguis CH3 (95 mJ·m−2) and Streptococcus mutans NS (117 mJ·m−2). Solid substrata were also selected on basis of their surface free energy ( γ s), and included polytetrafluorethylene ( γ s= 20 mJ·m−2), polymethylmethacrylate (53 mJ·m−2) and glass (109 mJ·m−2). Bacterial adhesion was measured as the number of bacteria adhering per cm2 at equilibrium. Equilibrium was usually obtained within 20 min. S. sanguis CH3, having an intermediate surface free energy did not show a clear preference for any of the 3 solids. S. mitis L1, however, the lowest surface free energy strain, adhered in highest numbers to the low energy solid PTFE, whereas the highest γ b strain, S. mutans NS, adhered in highest numbers to the highest γ s solid, glass. Calculation of the interfacial free energy of adhesion ( ΔF adh) for each bacterial strain showed that this parameter was predictive of bacterial adhesion to solid substrata.  相似文献

Abstract: Methyl 1,4-dihydro-2,6-dimethyl-3-nitro-4-(2-trifluoromethylphenyl)-pyridine-5-carboxylate (BAY K 8644), an analog of dihydropyridine calcium channel antagonists, stimulated 45Ca uptake into PC12 pheochromocytoma cells. Half-maximal stimulation occurred at 80 n M BAY K 8644. Enhancement of uptake was inhibited by cationic and organic calcium channel blockers, but not by tetrodotoxin, which is consistent with an effect on voltage-dependent calcium channels. Stimulation of 45Ca uptake by BAY K 8644 occurred only at elevated concentrations of extracellular K+, suggesting that BAY K 8644 may interact with calcium channels in the open (activated) state.  相似文献

Abstract Oral Streptococcus species experience carbohydrate limitation interrupted by periods of substrate excess following food intake by the host. To investigate the competitiveness of various streptococcal species under fluctuating carbohydrate supply, 2-membered chemostat cultures were run.
Under continuous limitation of glucose or sucrose, all 6 Streptococcus mutans test strains were outcompeted by Streptococcus sanguis P4A7 or Streptococcus milleri B448. This indicated that S. mutans had a lower affinity for glucose and sucrose than S. sanguis and S. milleri .
Mixed cultures were then subjected to hourly pulses with glucose. Under these conditions S. mutans Ny344 competed successfully with S. milleri B448, but still lost the competition against S. sanguis P4A7. The streptococci responded to pulses by taking up glucose at the maximum rate almost instantaneously. S. sanguis P4A7 had the highest rate of glucose uptake while the q max value of S. mutans Ny344 was higher than that of S. milleri B448. This suggested a causal relationship between q max and competitiveness.  相似文献

Abstract The interaction of salivary lysozyme with the surface protein antigen (PAc) of Streptococcus mutans and the interaction of lysozyme with the pathogen were examined by ELISA using S. mutans MT8148 (PAc+) and the PAc-defective mutant EM-2 (PAc). The lysozyme clearly bound to the S. mutans wild type but not to the S. mutans mutant. Furthermore, lysozyme bound directly in the fluid phase to the rPAc, of which the binding kinetics were determined ( K on= 3.63 ± 0.04 × 103M−1 s −1, K off= 1.72 ± 0.04 × 10−5s−1 and Kon / Koff= 2.11 × 108M−1) using surface plasmon resonance. The kinetics of both association and dissociation were relatively slow. In addition, anti-lysozyme antibody significantly inhibited the binding of salivary components to the rPAc. The present findings indicate that lysozyme is one of the major salivary components interacting with S. mutans PAc.  相似文献
