Osmolarity not only plays a key role in cellular homeostasis but also challenges cell survival. The molecular understanding of osmosis has not yet been completely achieved, and the discovery of aquaporins as molecular entities involved in water transport has caused osmosis to again become a focus of research. The main questions that need to be answered are the mechanism underlying the osmotic permeability coefficients and the extent to which aquaporins change our understanding of osmosis. Here, attempts to answer these questions are discussed. Critical aspects of the state of the state of knowledge on osmosis, a topic that has been studied since 19th century, are reviewed and integrated with the available information provided by in vivo, in vitro and in silico approaches.  相似文献

How extracellular signals communicate with the cell cycle is poorly understood. In this issue, two papers address this problem by reporting phosphorylation of the cyclin-dependent kinase inhibitor p27Kip1 on a tyrosine residue by nonreceptor tyrosine kinases, which decreases p27 stability. This new mechanism could explain how cells enter the cell cycle from a quiescent state.  相似文献

Involvement of the mitochondrial permeability transition pore (PTP) in apoptosis and PTP structure are highly controversial. In this issue of Molecular Microbiology, experiments based on yeast genetics analyse the roles of the three proteins commonly considered to form the PTP, i.e. porin, ADP/ATP carrier (ACC) and mitochondrial cyclophilin, on apoptosis-like cell death. Whereas knocking out cyclophilin had no effect, the porin-1 knockout yeast showed enhanced apoptosis, suggesting that porin-1 has an antiapoptotic role. Loss of the ACC proteins afforded protection against some causes of death, but enhanced death induced by H(2)O(2), suggesting a more complex role for the ACC proteins in regulating apoptosis-like death in yeast.  相似文献

Although much newsprint is devoted to the subject of reducing the United States and other major developed countries dependence on their respective foreign energy sources; the most challenging issues for society is to provide long-term, sustainable energy sources to accommodate the global population as a whole. The projected population of planet Earth for the year 2050 is estimated to be in excess of 9 billion. With hydrocarbon-based energy becoming limiting it is unlikely that one type of energy will alone replace our dependence on this source. So-called "green" technologies that include solar, wind and wave powers are now being explored to reduce on traditional hydrocarbon-based fuel sources. The diverse and functional properties of microbes, and in particular anaerobes, are now being utilized in the production of biofuels and may provide one piece of the jigsaw for future energy requirements. Here we present some results of a screening program to identify and characterize a number of carbon monoxide oxidizing, ethanol-producing acetogenic anaerobes phylogenetically located within the Clostridiales.  相似文献

The habitat use, seasonality and demography of the broadnose sevengill shark Notorynchus cepedianus were investigated in central Patagonia, where a data gap exists for the species. Catch and effort and video‐derived indices indicated high relative abundance of sharks during warm months. Video stations revealed differences in the spatial use by sharks, being more frequently observed in the inner section of the bay. Complementary tagging efforts evidenced both a seasonal residence pattern and site fidelity between consecutive warm seasons. Juveniles outnumbered adults, which suggests that they may use the study area as a secondary nursing ground. Evidence from spontaneous regurgitation further suggests that prey abundance may be driving the seasonal occurrence of sharks in the region. This study allows for a more comprehensive understanding of the population structure and dynamics of sevengill sharks in the Southwest Atlantic. Many models of human evolution propose that key behavioral innovations were involved in the divergence of the human line from the ape line. An ecological reason, the shift from dense forests to a more open habitat, is suggested as the basis for these innovations. Primate models can be useful to our understanding of how environmental factors can affect such key behaviors. New results on forest chimpanzees demonstrate that for most patterns of behavior considered to favor a savanna model, the environmental influences are in opposition to the expectations. A new evaluation of the environmental influences on human evolution is required. We propose that either patterns of key human behaviors were inaccurately distinguished or that the hominization process started while the common ancestor of the chimpanzee and man was alive.  相似文献

Alzheimer's disease (AD) is a complex neurodegenerative disorder with multiple etiologies. The presence of the E4 isoform of apolipoprotein E (apoE) has been shown to increase the risk and to decrease the age of onset for AD and is the major susceptibility factor known for the disease. ApoE4 has been shown to intensify all the biochemical distrubances characteristic of AD, including beta amyloid (Aβ) deposition, tangle formation, neuronal cell death, oxidative stress, synaptic plasticity and dysfunctions of lipid homeostasis and cholinergic signalling. In contrast, other apoE isoforms are protective. Here we review and discuss these major hypotheses of the apoE4-AD association.  相似文献

TDP-43 mislocalization and aggregation are implicated in the pathogenesis of ALS and FTLD-U. Valosin containing protein (VCP) mutations also lead to TDP-43 deposition, resulting in Inclusion Body Myopathy, Paget disease, and Frontotemporal Dementia (IBMPFD). In this issue of Neuron, Johnson et?al. used whole-exome capture to identify VCP mutations in familial ALS. This extends the VCP phenotype to include motor neuron degeneration and provides another molecular tool to explore neurodegeneration disease mechanisms underlying the TDP-43 proteinopathies.  相似文献
