The sympathomimetic drugs noradrenaline, methoxamine, tyramine and norephedrine caused rhythmic contractions in isolated human vasa deferentia. Provided the drug was not washed out, these contractions lasted for the entire duration of the experiment (4-6 h). These contractions were mediated via alpha-adrenoreceptors. Intravenous administration of methoxamine or oxymetazolene to rats or guinea-pigs produced contractions of the vas deferens in vivo in some experiments but was accompanied by severe cardiovascular side effects. A local method of application was developed, using mixtures of tyramine with Silastic prepared as collars specially designed to fit round the vas deferens. Acute and chronic insertion of these slow-releasing devices around the vas deferens of rats produced rhythmic contractions of the vas deferens without any serious side effects.  相似文献

Norethisterone (NET) and its metabolite 5alpha-norethisterone (5alpha-NET) are competitors for the androgen receptor. The sensitivity of the rat vas deferens to the contractile action of methoxamine and serotonin is regulated by hormonal and anatomical factors. The aim of this study was to evaluate the ability of NET and 5alpha-NET to induce the androgen-regulated contractile response to methoxamine and serotonin in the epididymal and prostatic portions of rat vas deferens. Adult male rats either intact, castrated or steroid-treated castrated were used. The contractility was recorded isometrically, and non-cumulative concentration-response curves to either methoxamine or serotonin were obtained. NET and 5alpha-NET partially restored the sensitivity to methoxamine and serotonin in the epididymal portion of castrated rats. The maximal responses to both agonists were significantly higher than those observed in castrated rats, and significantly lower than the responses observed in either intact or androgen-treated castrated rats. The prostatic portion was less responsive to both agonists than the epididymal portion, in all groups but castrated rats, as castration induced sensitivity to both agonists. NET and 5alpha-NET displayed a partial though similar androgenic activity in the rat vas deferens. These results contrast with previous reports where a decrease of androgenic effect due to the 5alpha-reduction of NET has been found.  相似文献

本文介绍一种高分子聚合物HFMC注入输精管,遇体液即合成有孔隙(孔径2-16 μ)的固态聚合物,附着管壁并缓慢释放适度的H[+],以改变精子存活环境,从而达到狒猴非阻断性的输精管避孕。在狒猴配对试验中证明,输精管注入HFMC 30 mg×2(n=2),在注射后47-50天精液品质即逐渐恢复,已有致孕能力;输精管注入HFMC 60 mg×2(n=4),在300天后,有些个体精液品质才逐渐恢复,注后348-353天之间已有50%的雄猴配对致孕;另有50%的雄猴在注入854天时精液品质仍处于不育水平。实验结果还表明:HFMC对精子具有致死,致畸率高及使精子活力显著下降,这种作用随HFMC剂量的增大而加强,这种影响也随时间的后移而逐渐减弱;HFMC对输精管具有可通性及可复性。今后如能在注入剂量,自溶稳定性方面再改进,HFMC有可能成为男性节育较为理想的一种新型避孕剂。  相似文献

Xylamine (XYL) administered to intact rats caused a 70-80% reduction in norepinephrine (NE) uptake by the vas deferens but had little or no effect on NE content in that tissue. The vas deferens accumulates 3H-XYL in vitro by a desmethylimipramine (DMI)-sensitive mechanism. Vasa deferentia from 6-hydroxydopamine (6OHDA) pretreated animals exhibited a 80% reduction in both NE content and XYL uptake activity. These results indicate that XYL is taken up by sympathetic nerve terminals and can reduce NE uptake activity without depleting terminals of neurotransmitter.  相似文献

Action of ethanol on animals with different predisposition to alcohol consumption depended on the level of ethanol metabolism or sensitivity of nervous system to alcohol. There was studied the ethanol effects on contraction of rat vas deferens. Such an approach enables to distinguish the metabolic effects from action on nervous system. It was shown, that dose of ethanol, which inhibits the contraction of rat vas deferens of by 50% is the same in both groups of rats, but the ED50 of adenosin and beta-endorphin were 1.7 and 4.7 times lower for vas deferens of predisposed rats. It's possible to suggest the differences between two groups of rats to be connected with the effects of ethanol on purinergic and endorphinergic system, but not with ethanol itself.  相似文献

Polyacrylamide gel electrophoresis analysis revealed that the vas deferens of adult mouse contains a major protein. Mouse vas deferens protein is a basic glycoprotein with a molecular weight of 34,800 +/- 300. The protein represents 17 +/- 0.7% and 42 +/- 2.4% of soluble proteins from homogenate and luminal fluid respectively, an estimate based on densitometric scanning of polyacrylamide gels. The protein originated from the vas deferens since it was not detected in blood plasma or in sexual organs and it was still present after ligation of the epididymis. Changes in androgen status of the animal markedly affected the vas deferens protein. After castration a progressive decrease in the protein was observed and its relative percentage dropped to 2 +/- 0.4% after 45 days. The concentration of the protein returned to precastration levels after 2 weeks of testosterone treatment but oestradiol, progesterone and corticosterone were ineffective in this respect. The vas deferens protein was not synthesized in significant amounts until animals were 20 days old and its concentration increased rapidly from 20 to 30 days in concert with the pubertal increase of androgens in the vas deferens.  相似文献

The motility of sperm was examined in vivo in the vas deferens, the spermatophore, and the spermatheca of the red-spotted newt and in in vitro preparations with variations in osmolality, hydrogen ion concentration, and concentrations of specific osmolytes. Sperm were motile within the spermatophore, but little or no evidence of motility was seen in the spermatheca or the vas deferens. Approximately 25% of sperm from the vas deferens became motile when dispersed in spermatic fluid plasma, the sperm-bearing liquid of the vas deferens, indicating crowding to be a possible motion-restraining factor. Fewer than 50% were motile in several saline media isosmotic with spermatic fluid plasma, whereas more than 90% became motile in distilled water or media at osmolalities near that of pond water. Motility in isosmotic solutions persisted beyond 12 hours, but at low osmolality ceased by 6 hours. When dispersed at higher osmolalities initial motility was low but increased to isosmotic levels by 12 hours. Responses to immersion in solutions of mannitol were similar to ones observed in saline solutions of equivalent osmolality. Dispersion in hydrogen ion concentrations between pH 4 and 9 did not affect the initial motility of sperm, but after 12 hours at pH 9, pH 4 or 5 movement was inhibited. In general, these data indicate a major role for osmolality in the enforced quiescence of sperm during storage and demonstrate that the low osmolality of pond water is primarily responsible for the activation of sperm in the spermatophore of the newt.  相似文献

This study focused on the role of sodium-bicarbonate cotransporter (NBC1) in cAMP-stimulated ion transport in porcine vas deferens epithelium. Ion substitution experiments in modified Ussing chambers revealed that cAMP-mediated stimulation was dependent on the presence of Na(+), HCO, and Cl(-) for a full response. HCO-dependent current was unaffected by acetazolamide, bumetanide, or amiloride but was inhibited by basolateral 4,4'-diisothiocyanostilbene-2,2'-disulfonic acid. Na(+)-driven, HCO-dependent, stilbene-inhibitable anion flux was observed across the basolateral membrane of selectively permeabilized monolayers. Results of radiotracer flux studies suggest a 4,4'-dinitrostilbene-2,2'-disulfonate-sensitive stoichiometry of 2 base equivalents per Na(+). Antibodies raised against rat kidney NBC epitopes (rkNBC; amino acids 338-391 and 928-1035) identified a single band of ~145 kDa. RT-PCR detected NBC1 message in porcine vas deferens epithelia. These results demonstrate that vas deferens epithelial cells possess the proteins necessary for the vectoral transport of HCO and that these mechanisms are maintained in primary culture. Taken together, the results indicate that vas deferens epithelia play an active role in male fertility and have implications for our understanding of the relationship between cystic fibrosis and congenital bilateral absence of the vas deferens.  相似文献

Comparisons among spontaneously hypertensive (SHR), Kyoto Wistar (KW), and Wistar (W) rats were made of the functional states of central nervous system (CNS) alpha 2-adrenoceptors (clonidine-induced mydriasis) and nonvascular peripheral presynaptic alpha 2-adrenoceptors (clonidine-induced inhibition of the neurogenic twitch of the isolated vas deferens). While there were no differences among the strains of rats in the concentration of clonidine required to produce a 50% inhibition of the electrically evoked contractile response of the vas deferens, there was a significant reduction in the mean effective concentration (ED50) of clonidine to induce mydriasis in SHR as compared with KW and W rats. These observations indicate that CNS alpha 2-adrenoceptors may be functionally more sensitive in SHR. The data also suggest that the sensitivity of nonvascular presynaptic alpha 2-adrenoceptors, at least in the vas deferens, is not altered in hypertensive animals.  相似文献

In the isolated rat vas deferens, morphine caused an increase in the neuromuscular twitch evoked by electrical stimulation. In rats chronically treated with morphine, the concentration of the opiate required to cause a 50% increase in the twitch was about 3 times larger than needed to elicit the same response in vasa from rats treated with a placebo. The simultaneous administration of naloxone plus morphine partially antagonized tolerance development by about 22%. Morphine tolerance was extended to other opiate- like alkaloids such as etonitazene and a derivative of azidomorphine (CAM). In contrast to the effects of morphine, β-endorphin inhibited neuromuscular transmission. Vasa from rats chronically treated with morphine were about 10 times less sensitive to the inhibitory effect of β-endorphin as compared to paired placebo treated controls. Chronic naloxone treatment in conjuction with morphine significantly reduced the cross-opiate tolerance by 66%. Present results suggest that morphine may interact at two different sites in the nerve terminals of the rat vas deferens.  相似文献

The effect of cocaine (10 μM) on the kinetics of contractile response to noradrenaline (NA) was studied in the rat epididymal and prostatic vas deferens. Cocaine caused an acute increase in vas deferens adrenergic sensitivity primarily due to blockage of NA neuronal capture. The presynaptic action prevailed in the epididymal half: the EC 50 value decreased 32-fold with a slight increase of the maximum adrenergic response more evident in the prostatic half. In the presence of cocaine, the prostatic contraction to NA was mediated not by a single pool of α1-adrenoceptors like in epididymal vas deferens but by two. Its high affinity pool had the same affinity as α1-adrenoceptors of the epididymal half, the affinity value of the low one was 36-fold less, and the total maximal response of both pools increased 4.5-fold. The differences in cocaine effect on the rat epididymal and prostatic vas deferens contractions to NA appear to be caused by the different sources of Ca2+ involved in these responses.  相似文献

Clonidine induces contractile effects on the isolated rat vas deferens, but not on rat uterus or guinea-pig ileum. However, we have observed that if clonidine is incubated for about 10 min with a nutrient solution containing an isolated rat vas deferens, the resulting solution can contract an isolated rat uterus, or guinea-pig ileum indicating the involvement of a substance released from the vas. This contractile effect was partially reduced by naloxone and by serotonin antagonists, and by using a denervated vas, indicating that opioids, serotonin and eventually other substances released from nerve tissue of the vas can be involved.  相似文献

The zinc and copper content in the different epididymal segments and vas deferens of castrated rats were investigated with the help of atomic absorption spectrophotometer. The vas deferens showed maximum zinc content as compared to that of different parts of epididymis in all groups whether castrated unilaterally, bilaterally or in the intact control. Zinc content was reduced in the epididymis and vas deferens ipsilateral to the castrated side as compared to that of contralateral control and intake animals. Lowest zinc content was observed in the epididymis and vas deferens of bilaterally castrated animals from that of other groups. Absence of sperms was observed in all segments of epididymis and vas in bilaterally castrated animals and from the unilaterally castrated side. Copper content was unaltered in all epididymal segments and vas deferens. There appears to be a correlation between the absence of sperms in the male genital tract and the decrease in zinc content.  相似文献

Experiments were conducted to characterize the effects of oxytocin (OT) and vasopressin (VP) on epithelial cells isolated from human (1 degree HVD) and porcine (1 degree PVD) vas deferens and an immortalized epithelial cell line derived from porcine vas deferens (PVD9902 cells). Cultured monolayers were assessed in modified Ussing flux chambers and the OT- or VP-induced change in short circuit current (I(SC)) was recorded. All cell types responded to basolateral OT or VP with a transient increase in I(SC) that reached a peak of 3-5 microA cm(-2). Concentration-response curves constructed with 1 degree PVD and PVD9902 cells revealed that the apparent K(D) (k(app)) for OT was approximately 100-fold less than the k(app) for VP. Amplicons for the OT receptor (OXTR) and vasopressin type 2 and type 1a receptors (AVPR2 and AVPR1A) were generated with RT-PCR and the identification of each amplicon confirmed by sequence analysis. A selective antagonist for OXTR and AVPR1A fully blocked the effects of OT and partially blocked the effects of VP when assessed in both 1 degree PVD and PVD9902 monolayers. APVR2 antagonists blocked the effects of low (< or =30 nM) but not high concentrations of VP, indicating that VP was affecting both AVPR2 and a second receptor subtype, likely OXTR or AVPR1A. Experiments employing chelerythrine demonstrated that OT stimulation of vas deferens monolayers requires PKC activity. Alternatively, VP (but not OT) increased the accumulation of cytosolic cAMP in vas deferens epithelial cells. Results from this study demonstrate that OT and VP can modulate ion transport across vas deferens epithelia by independent mechanisms. OT and VP have the potential to acutely change the environment to which sperm are exposed and thus, have the potential to affect male fertility.  相似文献

Biological activity of synthetic beta-endorphin (beta-EP) analogs containing dermorphin or dynorphin-A-(1-13) structure has been investigated using the guinea pig ileum and the vas deferens of the mouse, rat and rabbit. Replacement of NH2-terminal 1-7 segment of camel beta-EP [beta c-EP-(1-7)] with dermorphin caused a great increase in opiate potency of the analog. [Dermorphin (1-7)]-beta c-EP was 120 times more potent than beta c-EP in the guinea pig ileum assay, 49 times more potent in the mouse vas deferens assay; and only 4 times more potent in the rat vas deferens assay. Replacement of NH2-terminal 1-13 segment of human beta-EP [beta h-EP-(1-13)] with dynorphin-A-(1-13) caused an increase in opiate potency in both the guinea pig ileum and rabbit vas deferens assays, a complete loss of potency in the rat vas deferens assay, and no change in the mouse vas deferens assay. In comparison with dynorphin-A-(1-13), the hybrid peptide was less potent in the guinea pig ileum assay as well as in mouse and rabbit vas deferens assay. It is suggested that beta c-EP-(8-31) facilitates the dermorphin moiety to act on opiate mu and delta receptors but not on the epsilon receptor, while beta h-(14-31) reduces the action of dynorphin on mu, delta and kappa receptors.  相似文献

The different segments of the guinea pig vas deferens circular muscle exhibit differential response patterns upon pharmacological stimulation. Namely, apart from barium chloride, the affinity and intrinsic activity of certain agonists and the strength of maximum contractions they induce appear to decrease along the path from the epididymis toward the prostate. If one subdivides the vas deferens into 3 parts of equal length such as epididymal, medial and prostatic portions, then adrenaline, acetylcholine, acetyl-beta-methylcholine, dopamine, histamine and bradykinin induce contractions on each of the 3 parts; whereas tyramine, ephedrine elicit responses in the epididymal and medial portions; amphetamine, DMPP, serotonin and PGF2 alpha in turn provoking contractions exclusively on the epididymal portion. The effects of adrenaline and noradrenaline are blocked by phentolamine and tolazoline; the responses to acetylcholine, acetyl-beta-methylcholine and carbamyl-beta-methylcholine are antagonized by atropine over a specific concentration range. The effects of tyramine, ephedrine and amphetamine are inhibited by phentolamine in an remarkably low dose range (pA2 = 13.51 +/- 0.09; 14.54 +/- 0.31; 14.35 +/- 0.12). The situation was the same when tyramine-dibenamine and tyramine-phenoxybenzamine combinations were tested (pD'2 = 14.03 +/- 0.37; 13.26 +/- 0.03). Based on these findings the presence of a peculiar alpha adrenergic receptor is suggested on the sympathetic postganglionic fibres. In addition to the already identified alpha adrenergic, muscarinic cholinergic and histamine H1 receptors, we could show the presence of dopaminergic receptors too in the vas deferens circular muscle.  相似文献

The effects of agents that modulate intracellular release of calcium and protein kinase C (PKC) activation on noradrenaline (NA)-induced contractions of epididymal vas deferens in calcium-free/EGTA (1 mM) medium were investigated. NA (100 microM) or methoxamine (100 microM) evoked repeatable contractions. Clonidine (100-300 microM) was ineffective. The contractions to NA were reduced by procaine (1-10 mM) but not by thapsigargin (0.1-30 microM), ryanodine (1-30 microM) or TMB-8 (1-30 microM). Contractions to cumulative additions of NA (1-100 microM) were enhanced in the presence of cyclopiazonic acid (10 & 30 microM) but not ryanodine (10 & 30 microM). Sequential contractions to NA were not blocked by PKC inhibitors, calphostin C (1 microM) or Ro 31-8220 (1-30 microM) but were reduced by H-7 (1-30 microM), a broad spectrum protein kinase inhibitor. Although RT-PCR experiments detected mRNA for some Ca2+-dependent/DAG-activated and Ca2+-independent/DAG-activated PKC isoforms in epididymal vas deferens, the PKC activators, phorbol 12, 13-dibutyrate (100 microM) or phorbol 12-myristate 13-acetate (100 microM) failed to activate the tissues in calcium-free medium but enhanced subsequent contractions to NA. These results indicate a limited role for intracellular calcium stores and phorbol ester/DAG-sensitive PKC isoforms in NA-induced contraction of epididymal rat vas deferens in calcium-free medium. The results suggest that pharmacomechanical coupling triggered by NA may involve the sensitization of contractile myofilaments to Ca2+ or a Ca2+-independent mechanism. The possible involvement of Ca2+-independent/DAG-insensitive PKC isoforms and agonist-dependent but PKC-independent sensitization pathway is discussed.  相似文献

Absence of the vas deferens is a rare cause of male infertility, associated with mutations in the cystic fibrosis transmembrane regulator (CFTR) gene in about 80% of cases. Only limited published data are available concerning the correlation between genotype and reproductive tract abnormalities observed in this disease: presence or absence of seminal vesicles and parts of the epididymis, symmetrical or asymmetrical lesions, testicular volumes. We screened 47 patients for the 13 most common CFTR mutations on the cystic fibrosis gene and for the 5-thymidine variant of the polythymidine tract of intron 8. Renal, scrotal and transrectal ultrasonography was performed in each patient to explore the testes and reproductive tract. All patients presented absence of the ampullae of the vas deferens. Forty patients presented bilateral absence of the vas deferens and 7 presented unilateral absence of the vas deferens. At least one mutation of the cystic fibrosis gene was present in 64% of cases: 47% had the ΔF 508 mutation and 63% had the 5T allele. No mutation was detected in seventeen patients, including 3 patients with unilateral renal agenesis and 3 patients with unilateral absence of the vas deferens. No differences were observed for seminal vesicles and symmetry of vesicular and epididymal abnormalities between patients with or without CFTR gene mutations, but epididymal abnormalities were significantly more frequent in the group without mutation (p=0.01). Testicular volumes were significantly lower in the patients without mutation or with the 5T allele only, than in the patients with at least one CFTR gene mutation: 10.7±4.1 ml versus 15.1±4.5 ml, respectively (p<0.001). In conclusion, in cases of isolated absence of the vas deferens, there is no difference in sperm duct abnormalities between patients with or without CFTR gene mutation. These results suggest that other genetic or environmental determinants are required to explain a common pathogenesis for these malformations. The decreased testicular volume of patients without CFTR gene mutation or with the 5T allele only suggests the existence of an unidentified secretory or mixed factor involved in these forms of absence of the vas deferens.  相似文献

The effect of an opiate alkaloid and an opioid-like peptide was studied on the electrically evoked twitching of the vas deferens of 3 common laboratory rodents. Normorphine and the synthetic opioid peptide D-Alanine2 methionine enkephalinamide (D-Ala2) produced dose dependent inhibitions of the twitching response in the mouse vas deferens. In the rat vas, while β-endorphin (β-EP) caused an inhibitory effect in three strains of rats to a similar degree, morphine produced a dose related enhancement of the twitching. In the guinea pig, both morphine and β-EP caused an increased in the muscular twitch. The results are interpreted in terms of an heterogenous mixture of opiate receptors present in the vas deferens from these rodents. The mouse appears to contain mainly δ receptors while the rat has mostly ε receptors characterized by their specificity and sensitivity to the action of β-EP. The guinea pig vas deferens has apparently lost the sensitivity to the inhibitory influence of the opioids, suggesting the absence of μ or δ opiate receptors in this tissue.  相似文献
