Cytoplasmic vacuoles seen in methanol-fixed, Giemsa's-stained ookinetes of Leucocytozoon simondi, Parahaemoproteus fringillae and Plasmodium gallinaceum, when studied with the electron microscope, were found to correspond with crystalloid inclusions of similar structure, particle size, and arrangement. Cytochemical examination of these “crystalloids” revealed their lipid-protein nature. Morphologically similar inclusions were found also in ookinetes of Leucocytozoon ziemanni and Parahaemoproteus velans. In L. simondi, crystalloid is formed rapidly after fertilization, from amorphous electron dense material seen in mature macrogametocytes. The arrangement and distribution of crystalloids in the zygote, ookinete, oocyst, and sporozoite are described. On the basis of differences in structure and particle size, it is proposed that the crystalloid inclusions in Haemosporina be divided into 2 types. Type I—lipid-protein in nature, characterized by electron dense irregularly spherical particles, 25–40 nm in diameter, with individual particles not invested by membrane. Type II—probably virus, characterized by electron dense, irregularly spherical, membrane-bounded particles, with a diameter usually greater than 40 nm.  相似文献

SYNOPSIS. Microgametes of Haemoproteus columbae were studied by light microscopy and electron microscopy, using the single-stage replication technic. The filiform microgamete has a swelling or vesicle near the round end; the opposite end is asymmetrically tapered. The gamete is polarized, for it travels in sinuous movements with the round end (the putative anterior end) directed forward. There is no flagellum or undulating membrane and no mitochondrion. The vesicle may contain energy reserves for locomotion or lytic enzymes for penetration. Tubular material often is seen coming from the vesicle and anterior end and may be a kind of acrosomal filament.  相似文献

Ookinete differentiation begins in vitro~1 hr after blood infected with mature gametocytes of Haemoproteus columbae is withdrawn from a pigeon. In the undifferentiated zygote, dense material accumulates at the point under the plasma membrane. The conoid and conoidal rings condense from this material. The nucleus is drawn out to a point with the intranuclear spindle (INS) at the peak. Atypical centrioles lie under the forming conoid in the cytoplasm next to the INS. Fibrous material under the inner membrane forms the polar ring from which subpellicular microtubules originate. One hr later the centrioles have disappeared and the nucleus has returned to the center of the organism. The conoidal complex forms the tip of a growing cytoplasmic projection, the anterior end of the ookinete. During this time an elaborate pellicle is differentiating antero-posteriorly; crystalloid formation begins with an extensive proliferation of rough endoplasmic reticulum (ER) continuous with the outer membrane of the nuclear envelope. Crystalloid particles are formed between the lamellae of the ER and collected in a sphere that is later partially surrounded by a small amount of ER. Ookinetes, differentiated 2 hr longer than the ookinetes in vitro, were obtained from the gut of the pigeon fly, Pseudolynchia maura. The differentiated pellicle of these ookinetes consists of a plasma membrane, an inner membrane layer composed of 2 appressed membranes, and in the anterior end, an electron-opaque lamina immediately under the inner membrane. Anterior to the polar ring, this lamina forms a canopy which, posteriorly, is drawn out into projecting ribs which diminish and disappear in the first third of the organism. Fifty to 60 subpellicular microtubules insert on the polar ring. Ookinetes differentiated in vitro were no more than 4 hr old. They lacked micronemes and retained a pellicular cytostome and “internal cytostomes.” The differentiation of micronemes probably occurs at a later time because they are visible after 6 hr in ookinetes in the fly gut. So many degenerating organisms appeared in vitro after 5 hr that this material was discarded.  相似文献

SYNOPSIS. Pekin ducks were infected naturally and experimentally. The life cycle of Leucocytozoon simondi was followed in the ducks and the vector, a new species of Simulium, close to Simulium dogieli. The prepatent period in the ducks was 4–5 days and developing megaloschizonts appeared in 6–7 days. Schizonts were found in liver, spleen, brain, and kidney. In the kidneys they were located in the glomeruli. Sporogony in some individuals was completed in 7 days at 13–14 C, other individuals developed more slowly, as ookinetes were found in flies 8 days after feeding. The rapid asexual cycle combined with a sporogonic cycle, in which some ookinetes develop rapidly and others more slowly, favors the maintenance of the parasite in an environment with relatively low daily temperatures. This and the size of the megaloschizonts indicate differences between this strain of the parasite and the one occurring in North America.  相似文献

SYNOPSIS. The fine structure of Haemoproteus columbae sporozoites has been studied and compared to sporozoite structure as revealed by the light microscope. The sporozoites are ultrastructurally similar to those of other Haemosporidia in that they possess a 3-layered pellicle, subpellicular microtubules, polar ring, micropore, free ribosome-like particles, micronemes, a structure resembling a Golgi complex, an irregular mitochondrion, and a large nucleus. In the anterior region of the sporozoite there are 21–22 regularly arranged longitudinal subpellicular microtubules located peripherally around the cell. In the apical region the microtubules appear thickened on 1 side. The sporozoite of H. columbae has a microneme system in which 1–3 micronemes are associated with the outer pellicular membrane at the anterior end. Micronemes are found throughout the cytoplasm, but occur in greater concentration in the anterior region of the sporozoite. A clear pellicular cavity, located between the polar ring and the termination of the inner pellicular layer, is present at the anterior end of the sporozoite. Vesicular invaginations of the inner pellicular layer have been observed in the anterior region; their function is unknown. Spherical osmophilic bodies are found throughout the cytoplasm.  相似文献

From 1969 through 1972, 605 wild turkeys (Meleagris gallopavo Linnaeus) from 10 localities in Florida were examined for blood protozoans. The prevalence in turkeys more than a month old was 84% for Haemoproteus meleagridis Levine, 1961 and 72% for Leucocytozoon smithi Laveran and Lucet, 1905. Sixty-three percent of the birds were infected by both parasites. Infections were most prevalent in juveniles 8–12 months of age. No infection of H. meleagridis or L. smithi was found in 111 poults younger than 35 days. Prevalences were similar in both sexes. Haemoproteus meleagridis was more prevalent (87%) in the southern part of the state; L. smithi was more prevalent (75%) in the north. The prevalence of H. meleagridis did not change during the 4 year period, but L. smithi decreased markedly in 1971, a year of low rainfall. None of the 605 blood films was positive for Plasmodium, but 24 (75%) of 32 blood samples were found by subinoculation technics to be positive for a species of Plasmodium morphologically similar to P. durae Herman, 1941. No trypanosomes were seen on the 605 blood films or in bone marrow cultures (saline-neopeptone-blood) made from 11 turkeys.  相似文献

SYNOPSIS. Ultrastructure of cysts of Naegleria gruberi, Naegleria fowleri, and Naegleria jadini was compared by transmission electron microscopy. Pores in the cyst wall were observed in all 3 species. In N. gruberi they were surrounded by a collar and sealed with a relatively large mucoid plug; no such collar was seen around the pores in the other 2 species, in which the plug was smaller than that in N. gruberi. An electron-dense plaque serving as an additional pore closure was present in all 3 species. In N. gruberi, the cyst wall consisted of an inner thick and an outer thin layer; however, only the inner component was present in cysts of N. fowleri and N. jadini, which had a smooth appearance. At the ultrastructural level, excystment of N. fowleri involved digestion of the mucoid plug and emergence of the trophozoite through the pore. Some digestion of the cyst wall also appeared to take place during excystment.  相似文献

When blood is withdrawn from a pigeon (Columba livia) infected with gametocytes of Haemoproteus columbae, differentiation of the gametes begins immediately. This study examines the formation of the macrogamete and its fertilization. The first visible signs of differentiation are the elongation of the nucleus along with the appearance of an intranuclear spindle and atypical centrioles. Then maturation bodies, the products of nuclear reduction, form in both erythrocytic macrogametocytes and macrogametocytes free of their host cells. Penetration of the macrogamete by the microgamete occurs rapidly. Their plasma membranes fuse, and the microgamete's nucleus, axonemes and cytoplasm enter the macrogamete. The nucleus of the microgamete expands and migrates to lie at an angle to the macrogamete nucleus. The 2 fuse across a small area. The nuclear envelope and the plasma membrane of the zygote are a mosaic of the membranes of the 2 gametes.  相似文献

SYNOPSIS. Blood films were examined from 1477 birds of Taiwan (193 species, 49 families). Haemoproteus Kruse was by far the commonest parasite, with Leucocytozoon Danilewski a not very close second. It is probable that some of the Haemoproteus infections represented new species, and 1 occurring in the Bamboo Partridge (Bambusicola thoracica sonorivox Gould) seemed characteristic enough to justify recognition as such; the name Haemoproteus bambusicolae sp. n. is proposed for this organism. Malaria was found in 77 birds, the greatest number of infections occurring in the Bamboo Partridge. Most of them were caused by Plasmodium juxtanucleare Versiani & Gomes, a pathogen of chickens, but a number were due to an undetermined species of Plasmodium. The Bamboo Partridge may be a reservoir host of the former. A few other identified species (P. rouxi Sergent & Sergent, P. hexamerium Huff, P. tenue Laveran & Mesnil) were seen, as well as some unidentified ones. Plasmodium tenue was seen in Garrulax canorus taewanus Swinhoe, a babbler: until now it was known only from the Pekin Robin (Leiothrix luteus Scopoli), also a babbler, in which we have found it extremely common. Sixty-four microfilarial infections were identified; they were especially frequent in the Button Quail (Turnix suscitator rostrata Swinhoe).  相似文献

Several factors shape lifetime reproductive success, including genetic background, body condition, environmental conditions and ecological interactions such as parasitism. Adults often show higher reproductive success than their young conspecifics, especially in long-lived bird species, and this may be explained by the cumulative effects of an increase in reproductive experience and the selection of high-quality individuals from one year to the next. To test whether this pattern also exists in short-lived bird species, we used 13 years of monitoring data from two Great Tit Parus major populations. The effects of male and female age on several reproductive parameters were analysed in 419 pairs of Great Tits, while accounting for body condition and infection by haemosporidian parasites. Reproductive success was mainly affected by the age-class of males. Pairs containing a sub-adult male fledged one-third fewer chicks than pairs containing an adult male. The difference was not caused by variation in male fertility but could have been caused by better parental care provided by adult birds. In addition to lower reproductive success, first-year males also had reduced access to mating compared with adult males, suggesting an avoidance of sub-adult males by females. Nestling body condition was positively correlated with parental body condition, and the body condition of male and female members of breeding pairs was positively correlated. Finally, the number of fledged chicks was mainly affected by the infection status of males. These results temper our previously published results showing an effect of infection on Great Tit reproduction regardless of their sex. In our previous study, and as in most cases, the status of the partner was not taken into account and we show here that this is essential because it can lead to a biased interpretation of the results.  相似文献

We investigated the degree of geographical shifts of transmission areas of vector-borne avian blood parasites (Plasmodium, Haemoproteus and Leucocytozoon) over ecological and evolutionary timescales. Of 259 different parasite lineages obtained from 5886 screened birds sampled in Europe and Africa, only two lineages were confirmed to have current transmission in resident bird species in both geographical areas. We used a phylogenetic approach to show that parasites belonging to the genera Haemoproteus and Leucocytozoon rarely change transmission area and that these parasites are restricted to one resident bird fauna over a long evolutionary time span and are not freely spread between the continents with the help of migratory birds. Lineages of the genus Plasmodium seem more freely spread between the continents. We suggest that such a reduced transmission barrier of Plasmodium parasites is caused by their higher tendency to infect migratory bird species, which might facilitate shifting of transmission area. Although vector-borne parasites of these genera apparently can shift between a tropical and a temperate transmission area and these areas are linked with an immense amount of annual bird migration, our data suggest that novel introductions of these parasites into resident bird faunas are rather rare evolutionary events.  相似文献

Research in avian blood parasites has seen a remarkable increase since the introduction of polymerase chain reaction-based methods for parasite identification. New data are revealing complex multihost-multiparasite systems which are difficult to understand without good knowledge of the host range and geographical distribution of the parasite lineages. However, such information is currently difficult to obtain from the literature, or from general repositories such as GenBank, mainly because (i) different research groups use different parasite lineage names, (ii) GenBank entries frequently refer only to the first host and locality at which each parasite was sampled, and (iii) different researchers use different gene fragments to identify parasite lineages. We propose a unified database of avian blood parasites of the genera Plasmodium, Haemoproteus and Leucocytozoon identified by a partial region of their cytochrome b sequences. The database uses a standardized nomenclature to remove synonymy, and concentrates all available information about each parasite in a public reference site, thereby facilitating access to all researchers. Initial data include a list of host species and localities, as well as genetic markers that can be used for phylogenetical analyses. The database is free to download and will be regularly updated by the authors. Prior to publication of new lineages, we encourage researchers to assign names to match the existing database. We anticipate that the value of the database as a source for determining host range and geographical distribution of the parasites will grow with its size and substantially enhance the understanding of this remarkably diverse group of parasites.  相似文献

A total of 86 adult bluethroats (Luscinia svecica) from nine different localities, covering the full length of the Fennoscandian mountain range, were screened for blood parasites of the three genera Haemoproteus, Plasmodium and Leucocytozoon using a recently developed polymerase chain reaction method. The overall occurrence of infection was 59.3%. Prevalence of Leucocytozoon spp. (47.7%), Plasmodium spp. (23.3%) and Haemoproteus spp. (1.2%) was detected. Of the infected birds, 15.1% carried mixed infections. Five different mitochondrial DNA-lineages of Leucocytozoon spp., eight lineages of Plasmodium spp. and one lineage of Haemoproteus spp. were found. Due to large sequence divergence these corresponded to at least five different species, but with the possibility of all 14 being independent evolutionary units with the potential of evolving different effects on the host. Of the lineages of Leucocytozoon spp., the most common was found throughout the range. The occurrence of the second most common lineage of Leucocytozoon spp. showed significant variation in prevalence between sites. The data also showed molecular evidence of one lineage of Leucocytozoon sp. existing in more than one species of avian host, thus challenging the use of host taxon as a taxonomic character when distinguishing between different species leucocytozoids.Communicated by F. SYNOPSIS As determined by their ability to exflagellate and round up, it took the macrogametocyte and the microgametocyte of Haemoproteus columbae in pigeons 68 and 116 h, respectively, after patency to reach maturity. Pigment granules appeared in the undifferentiated gametocytes 8 h after invasion of blood. Vacuoles were observed in young gametocytes and persisted in the older forms. The growth curve of H. columbae is close to the sigmoidal curve for growth in protozoa. Multiple infection was noticed in pigeons with high levels of parasitemia, but no more than 2 gametocytes reached maturity; such multiple infections were rare in relapses. The sex ratio of the gametocytes was 1:1. Strong lateral displacement of the nuclei of infected erythrocytes was the rule; hypertrophy was negligible.  相似文献

Corticosterone (CORT) is the main glucocorticoid hormone of amphibians, reptiles, birds and some mammals. This hormone may have evolved as an adaptive metabolic mechanism, in part because increased concentrations of CORT are essential for individuals to manage energy resources and thus cope with negative perturbations such as predation and storms. The benefits of CORT are offset by costs, because elevated levels can suppress inflammatory responses of individuals, making them more susceptible to parasites and pathogens. In this study, we investigated the relationships between feather CORT levels, infection status and diversity of haemosporidian parasites in the Blue-crowned Manakin Lepidothrix coronata, considering possible effects related to the sex and age of individuals. We predicted higher levels of feather CORT in infected individuals. We observed that feather CORT levels were similar among individuals of different sexes and ages. Although haemosporidian infection status did not vary among sexes, occurrence probability was higher among younger individuals, which may indicate that the less developed immune system of these individuals makes them more susceptible to avian malaria. Contrary to expectations, we found that feather CORT levels were not associated with the infection status and diversity of haemosporidian parasites. That haemosporidian occurrence probability does not increase with elevated feather CORT levels suggests that individuals are not immunosuppressed by elevated levels of this hormone, at least to the extent that feather CORT truly reflects individual differences in the level of this hormone.  相似文献

SYNOPSIS. The structural changes in macro and microgametocytes of Parahaemoproteus velans following removal of infected blood from the avian host were studied in the light and electron microscope. Gametocytes of both sexes round up and soon escape from their host cells. Shortly thereafter they assume a dumbbell shape. The microgametocyte undergoes exflagellation forming 8 slender microgametes. During fertilization the entire microgamete appears to enter the female. The most striking ultrastructural change in the formation of the macrogamete is the condensation and enclosure by a membrane of abundant amophorus dense material seen in the cytoplasm of the immature gametocyte. Maturation of the microgametocyte begins prior to its escape from the host cell. Axonemes are present in the cytoplasm and nuclear reorganization occurs while the parasite is intracellular. Bundles of microtubules associated with condensed chromatin are found in the peripheral cytoplasm of maturing forms and apparently participate in the formation of small compact microgamete nuclei. Each of these filiform structures consists of a dense, centrally located nucleus and a single axoneme lying in flocculent cytoplasm. The nucleus and axoneme of the microgamete are seen free in the cytoplasm of a fertilized macrogamete.  相似文献

Land use changes including deforestation, road construction and agricultural encroachments have been linked to the increased prevalence of several infectious diseases. In order to better understand how deforestation affects the prevalence of vector-borne infectious diseases in wildlife, nine paired sites were sampled (disturbed vs. undisturbed habitats) in Southern Cameroon. We studied the diversity, prevalence and distribution of avian malaria parasites ( Plasmodium spp.) and other related haemosporidians (species of Haemoproteus and Leucocytozoon ) from these sites in two widespread species of African rainforest birds, the yellow-whiskered greenbul ( Andropadus latirostris , Pycnonotidae) and the olive sunbird ( Cyanomitra olivacea , Nectariniidae). Twenty-six mitochondrial cytochrome b lineages were identified: 20 Plasmodium lineages and 6 Haemoproteus lineages. These lineages showed no geographic specificity, nor significant differences in lineage diversity between habitat types. However, we found that the prevalence of Leucocytozoon and Haemoproteus infections were significantly higher in undisturbed than in deforested habitats ( Leucocytozoon spp. 50.3% vs. 35.8%, Haemoproteus spp. 16.3% vs. 10.8%). We also found higher prevalence for all haemosporidian parasites in C. olivacea than in A. latirostris species (70.2% vs. 58.2%). Interestingly, we found one morphospecies of Plasmodium in C. olivacea , as represented by a clade of related lineages, showed increased prevalence at disturbed sites, while another showed a decrease, testifying to different patterns of transmission, even among closely related lineages of avian malaria, in relation to deforestation. Our work demonstrates that anthropogenic habitat change can affect host–parasite systems and result in opposing trends in prevalence of haemosporidian parasites in wild bird populations.  相似文献

SYNOPSIS. The erythrocytic stages of Plasmodium gallinaceum in chicken embryos injected with parasitized blood either from a syringe-passaged infection in chickens or from a chicken infected with sporozoites, were characterized by abnormal structure. Particularly evident were large, unstained vacuoles within the cytoplasm; these occurred with greatest frequency in schizonts. The presence of myelin bodies within these vacuoles was revealed by transmission electron microscopy; abnormal cytokinesis and aberrant merozoites provided additional evidence of the parasite's inability to develop naturally within the milieu of the embryonic erythrocytes. Fifty-five passages were necessary to restore normal structure of the parasites in embryos, while only 5 passages were required for such restoration in neonate chicks. The probable adaptation of the parasite to the proportions of hemoglobin of the adult chicken may be responsible for the abnormal growth in the immature host.  相似文献

Understanding the drivers of infection risk helps us to detect the most at-risk species in a community and identify species whose intrinsic characteristics could act as potential reservoirs of pathogens. This knowledge is crucial if we are to predict the emergence and evolution of infectious diseases. To date, most studies have only focused on infections caused by a single parasite, leaving out co-infections. Yet, co-infections are of paramount importance in understanding the ecology and evolution of host-parasite interactions due to the wide range of effects they can have on host fitness and on the evolutionary trajectories of parasites. Here, we used a multinomial Bayesian phylogenetic modelling framework to explore the extent to which bird ecology and phylogeny impact the probability of being infected by one genus (hereafter single infection) or by multiple genera (hereafter co-infection) of haemosporidian parasites. We show that while nesting and migration behaviours influenced both the probability of being single- and co-infected, species position along the slow-fast life-history continuum and geographic range size were only pertinent in explaining variation in co-infection risk. We also found evidence for a phylogenetic conservatism regarding both single- and co-infections, indicating that phylogenetically related bird species tend to have similar infection patterns. This phylogenetic signal was four times stronger for co-infections than for single infections, suggesting that co-infections may act as a stronger selective pressure than single infections. Overall, our study underscores the combined influence of hosts’ evolutionary history and attributes in determining infection risk in avian host communities. These results also suggest that co-infection risk might be under stronger deterministic control than single infection risk, potentially paving the way toward a better understanding of the emergence and evolution of infectious diseases.  相似文献

SYNOPSIS. Gametocytes differentiated from ring-stage parasites in microcultures of human blood infected with Plasmodium falciparum. Immature gametocytes could be distinguished morphologically from late asexual trophozoites after ~ 40 h of culture. Differentiation into crescentic forms took several days and the incorporation of [3H]-isoleucine by developing gametocytes was demonstrated. About 1% of red cells contained gametocytes at the maximum densities attained. Differentiation of gametocytes occurred either directly from rings placed in culture or from the progeny of subsequent cycles of schizogony and invasion in vitro. The latter occurrence was confirmed by the development of gametocytes in marker fetal red cells added to cultures, although fetal red cells provide a less favorable environment than those with HbA for growth of the parasites.  相似文献
