The comparison between measurements effected by different apparati shows that a given class of instruments provides an integral set of measurements. This characteristic has a certain disadvantage: the modification of the spectral bandwidth is limited to an attenuation of 3 dB. However, it also has certain advantages. The integral measurement (or the counting measurement) makes it possible to satisfy Shannon's sampling criteria by avoiding the necessary anti-aliasing filtering which is commonly impossible to realize on measurements of a biological nature. A second advantage is linked to the reduction of the background noise on the band.  相似文献

Mass balance of a glacier is an accepted measure of how much mass a glacier gains or loses. In theory, it is typically computed by integral functional and empirically, it is approximated by arithmetic mean. However, the variability of such an approach was not studied satisfactory yet. In this paper we provide a dynamical system of mass balance measurements under the constrains of 2nd order model with exponentially decreasing covariance. We also provide locations of optimal measurements, so called designs. We study Ornstein–Uhlenbeck (OU) processes and sheets with linear drifts and introduce K optimal designs in the correlated processes setup. We provide a thorough comparison of equidistant, Latin Hypercube Samples (LHS), and factorial designs for D- and K-optimality as well as the variance. We show differences between these criteria and discuss the role of equidistant designs for the correlated process. In particular, applications to estimation of mass balance of Olivares Alfa and Beta glaciers in Chile is investigated showing that simple application of full raster design and kriging based on inter- and extrapolation of points can lead to increased variance. We also show how the removal of certain measurement points may increase the quality of the melting assessment while decreasing costs. Blow-ups of solutions of dynamical systems underline the empirically observed fact that in a homogenous glaciers around 11 well-positioned stakes suffices for mass balance measurement.  相似文献

The method of photosensitized chemiluminescence (PCL) allows the quantification of water- and lipid-soluble antioxidants and activity of superoxide dismutase (SOD) in the same measuring system. However, it needs a special device, which we have described in a previous paper in this series. Another method suitable for the assay of water- and lipid-soluble antioxidants is the thermo-initiated decay of azo-compounds combined with the measurement of O2 consumption (Niki, 1985; Wayner et al., 1985). Its long duration and the complicated measuring procedure is not acceptable for routine medical applications. We show that a modification using CL detection of free radicals with luminol, has results comparable with PCL for the determination of non-enzymic water- and lipid-soluble antioxidants, SOD activity and oxidative modification of proteins. In contrast to PCL, it is possible to use any luminometer with a heatable measuring cell and to investigate coloured samples. While the new method has an overall higher sensitivity and is scalable to microtitre plates, PCL measurements can be made at different pH. The advantages and analytical information content of certain components of the integral antioxidative capacity of blood plasma are discussed in comparison with other methods.  相似文献

The ultrafine particle measurements in the Augsburger Umweltstudie, a panel study conducted in Augsburg, Germany, exhibit measurement error from various sources. Measurements of mobile devices show classical possibly individual–specific measurement error; Berkson–type error, which may also vary individually, occurs, if measurements of fixed monitoring stations are used. The combination of fixed site and individual exposure measurements results in a mixture of the two error types. We extended existing bias analysis approaches to linear mixed models with a complex error structure including individual–specific error components, autocorrelated errors, and a mixture of classical and Berkson error. Theoretical considerations and simulation results show, that autocorrelation may severely change the attenuation of the effect estimations. Furthermore, unbalanced designs and the inclusion of confounding variables influence the degree of attenuation. Bias correction with the method of moments using data with mixture measurement error partially yielded better results compared to the usage of incomplete data with classical error. Confidence intervals (CIs) based on the delta method achieved better coverage probabilities than those based on Bootstrap samples. Moreover, we present the application of these new methods to heart rate measurements within the Augsburger Umweltstudie: the corrected effect estimates were slightly higher than their naive equivalents. The substantial measurement error of ultrafine particle measurements has little impact on the results. The developed methodology is generally applicable to longitudinal data with measurement error.  相似文献

Biomass measurement is one of the most critical measurements in biotechnological processes. The technologies developed for the measurement of biomass in situ have developed over the years. Because it has been over 10 years since the last review concentrating on practical issues concerning biomass measurements, it is time to evaluate recent developments in the field. This review concentrates on the applications of dielectric spectroscopy, optical density, infrared spectroscopy, and fluorescence for in situ measurement of biomass. Since the first demonstration by Van Herle et al. that thyroglobuling (Tg), the main iodo-protein of the thyroid gland, was detectable in the circulation of normal subjects by using specific radioimmunoassay, an impressive amount of papers has been produced, describing several clinical applications of Tg measurements. Now, measurement of Tg is the mainstay in the post-surgical follow-up of differentiated thyroid cancer and an integral part in the diagnostic evaluation of patients with benign thyroid diseases. In this article we will review the most relevant clinical application of Tg measurements in the blood and in other biological material, including: a) serum Tg in perinatal age and congenital hypothyroidism; b) serum Tg measurements in thyrotoxicosis factitia and in other thyrotoxic conditions associated with low iodine uptake; c) differential diagnosis of thyroid and parathyroid cysts; and d) clinical relevance of Tg measurement in thyroid cancer.  相似文献

A simple technique for the measurement of swimming speed of Chlamydomonas reinhardi is described. The technique is based on a rapid statistical counting method which treats the swimming cells as a dilute, kinetic gas and its accuracy has been confirmed by direct photomicrographic measurements. ABSTRACT Out of precaution, opportunism, and a general tendency towards thoroughness, researchers studying wildlife often collect multiple, sometimes highly correlated measurements or samples. Although such redundancy has its benefits in terms of quality control, increased resolution, and unforeseen future utility, it also comes at a cost if animal welfare (e.g., duration of handling) or time and resource limitation are a concern. Using principle components analysis and bootstrapping, we analyzed sets of morphometric measurements collected on 171 brown bears in Sweden during a long-term monitoring study (1984–2006). We show that of 11 measurements, 7 were so similar in terms of their predictive power for an overall size index that each individual measurement provided little additional information. We argue that when multiple research objectives or data collection goals compete for a limited amount of time or resources, it is advisable to critically evaluate the amount of additional information contributed by extra measurements. We recommend that wildlife researchers look critically at the data they collect not just in terms of quality but also in terms of need.  相似文献

An optical method for measuring the birefringence of muscle fibers was developed, which is realized on an automated Linnick interferometer microscope equipped with a laser. It was shown that the method has some advantages over the methods based on measurements of the intensity of light passing through a crossed polarizer, an analyzer, and a fiber (light polarized microscopy). The method involves direct phase measurements of optical path length at the parallel and perpendicular orientations of the polarization plane of probing radiation. The phase image is reconstructed automatically from interferograms with the use of the four-frame phase-shifting algorithm. The phase images of one and the same central part of the fiber at different orientations of the polarization plane represent two-dimensional numerical maps of the optical path length. The subtraction of these images gives a two-dimensional map of the phase shift, which includes information about the birefringence of the fiber. A formula for birefringence measurements was deduced, which has a certain advantage in comparison to that used earlier in that it does not take into account the thickness of a fiber that depends on the measurement point. The birefringence is normalized to a value of the half sum of phases, which are measured separately in the course of the experiment.  相似文献

Quality by design (QbD) is a current structured approach to design processes yielding a quality product. Knowledge and process understanding cannot be achieved without proper experimental data; hence requirements for measurement error and frequency of measurement of bioprocess variables have to be defined. In this contribution, a model-based approach is used to investigate impact factors on calculated rates to predict the obtainable information from real-time measurements (= signal quality). Measurement error, biological activity, and averaging window (= period of observation) were identified as biggest impact factors on signal quality. Moreover, signal quality has been set in context with a quantifiable measure using statistical error testing, which can be used as a benchmark for process analytics and exploitation of data. Results have been validated with data from an E. coli batch process. This approach is useful to get an idea which process dynamics can be observed with a given bioprocess setup and sampling strategy beforehand.  相似文献

 Non-linear time sequence analysis has been performed on infant sleep measurement data in order to obtain more information about the respiratory processes. As a first step, respiration data during REM sleep were analysed with methods from non-linear dynamics, especially, the correlation integral and the slope of its log-log plot, representing the correlation dimension. Before calculation of the correlation integral, a special kind of filtering has to be applied to the data. This filtering algorithm is a state space and singular value decomposition-based noise reduction method, and it is used to separate the noise and signal subspaces. The dynamics of a signal (in our case data from the respiratory process) and its degrees of freedom can be characterised by the correlation integral and by the correlation dimension, respectively. The main result of this study is that the highly irregular-looking breathing patterns during REM sleep could be described by a deterministic system, and finally the physiological significance of this finding is discussed. The detection of microbial concentration, essential for safe and high quality food products, is traditionally made with the plate count technique, that is reliable, but also slow and not easily realized in the automatic form, as required for direct use in industrial machines. To this purpose, the method based on impedance measurements represents an attractive alternative since it can produce results in about 10 h, instead of the 24–48 h needed by standard plate counts and can be easily realized in automatic form. In this paper such a method has been experimentally studied in the case of ice-cream products. In particular, all main ice-cream compositions of real interest have been considered and no nutrient media has been used to dilute the samples. A measurement set-up has been realized using benchtop instruments for impedance measurements on samples whose bacteria concentration was independently measured by means of standard plate counts. The obtained results clearly indicate that impedance measurement represents a feasible and reliable technique to detect total microbial concentration in ice-cream, suitable to be implemented as an embedded system for industrial machines.  相似文献

Protein translocation normally starts from an N-terminal signal peptide and proceeds in an N-to-C-terminal direction. However, in certain integral membrane proteins an N-terminal tail is translocated even though it is not preceded by a signal peptide. In eukaryotic cells this process involves the normal Sec-machinery. In contrast, recent studies in Escherichia coli show that translocation of such N-terminal tails occurs by a mechanism that does not appear to involve the Sec proteins and is most efficient for short tails lacking positively charged residues. These novel observations suggest that the Sec-machinery has an inherent N-to-C-terminal directionality and cannot work 'in reverse'.  相似文献

Laser light scattering in conjunction with measurement of the spectral width and integral intensity of light scattering was applied to studying the process of complex formation of antigens and antibodies. The system of polysaccharide group A streptococcus and antibody against it was examined under varying polysaccharide concentrations. The measurements were performed every 10 s for 70 min after combining polysaccharide and antibody solutions. Within the entire time interval, the size of the complexes were less than the wave length of exciting laser light (0.633 m). This made it possible to determine their average magnitude in terms of the Rayleigh model.  相似文献

In addition to reference measurement procedures and reference materials, reference or calibration laboratories play an integral role in the implementation of measurement traceability in routine laboratories. They provide results of measurements using higher-order methods, e.g. isotope dilution mass spectrometry and may assign values to materials to be used for external quality assessment programs and to secondary reference materials. The requirements for listing of laboratories that provide reference measurement services include a statement of the metrological level or principle of measurement, accreditation as a calibration laboratory according to ISO 15195 and the participation in a proficiency testing system (regular inter-laboratory comparisons) for reference laboratories. Ring trials are currently conducted for thirty well-defined measurands and the results are made available to all laboratories. Through the use of reference laboratory services that are listed by the Joint Committee for Traceability in Laboratory Medicine there is the opportunity to further promote traceability and standardisation of laboratory measurements.  相似文献

Measurements provide the basis for process monitoring and control as well as for model development and validation. Systematic approaches to increase the accuracy and credibility of the empirical data set are therefore of great value. In (bio)chemical conversions, linear conservation relations such as the balance equations for charge, enthalpy, and/or chemical elements, can be employed to relate conversion rates. In a pactical situation, some of these rates will be measured (in effect, be calculated directly from primary measurements of, e.g., concentrations and flow rates), as others can or cannot be calculated from the measured ones. When certain measured rates can also be calculated from other measured rates, the set of equations, the accuracy and credibility of the measured rates can indeed be improved by, respectively, balancing and gross error diagnosis. The balanced conversion rates are more accurate, and form a consistent set of data, which is more suitable for further application (e.g., to calculate nonmeasured rates) than the raw measurements. Such an approach has drawn attention in previous studies. The current study deals mainly with the problem of mathematically classifying the conversion rates into balanceable and calculable rates, given the subset of measured rates. The significance of this problem is illustrated with some examples. It is shown that a simple matrix equation can be derived that contains the vector of measured conversion rates and the redundancy matrix R. Matrix R plays a predominant role in the classification problem. In supplementary articles, significance of the redundancy matrix R for an improved gross error diagnosis approach will be shown. In addition, efficient equations have been derived to calculate the balanceable and/or calculable rates. The method is completely based on matrix algebra (principally different from the graph-theoretical approach), and it is easily implemented into a computer program. Consider the problem of estimating the correlation between two nutrient measurements, such as the percent energy from fat obtained from a food frequency questionnaire (FFQ) and that from repeated food records or 24-hour recalls. Under a classical additive model for repeated food records, it is known that there is an attenuation effect on the correlation estimation if the sample average of repeated food records for each subject is used to estimate the underlying long-term average. This paper considers the case in which the selection probability of a subject for participation in the calibration study, in which repeated food records are measured, depends on the corresponding FFQ value, and the repeated longitudinal measurement errors have an autoregressive structure. This paper investigates a normality-based estimator and compares it with a simple method of moments. Both methods are consistent if the first two moments of nutrient measurements exist. Furthermore, joint estimating equations are applied to estimate the correlation coefficient and related nuisance parameters simultaneously. This approach provides a simple sandwich formula for the covariance estimation of the estimator. Finite sample performance is examined via a simulation study, and the proposed weighted normality-based estimator performs well under various distributional assumptions. The methods are applied to real data from a dietary assessment study.  相似文献

The neutral theory of molecular evolution has been instrumental in organizing our thinking about the nature of evolutionary forces shaping variation at the DNA level. More importantly, it has provided empiricists with a strong set of testable predictions and hence, a useful null hypothesis against which to test for the presence of selection. Evidence indicates that the neutral theory cannot explain key features of protein evolution nor patterns of biased codon usage in certain species. Whereas we now have a reasonable model of selection acting on synonymous changes in Drosophila, protein evolution remains poorly understood. Despite limitations in the applicability of the neutral theory, it is likely to remain an integral part of the quest to understand molecular evolution.  相似文献

It was reported by others that hands-free accessories increase the absorption of RF energy in a human head compared to a handset alone. The results of this study show that the opposite is observed when proper dosimetric methods are employed. It is pointed out that for correct estimation of the exposure level it is necessary to use appropriate physical and experimental models and measurement instrumentation, following internationally recommended standards. The human phantoms used for measurements involving the hands-free accessories should include the torso; i.e., measurements should not be performed on the head phantom alone. This has a significant impact on the results because the RF energy coupled into the leads of hands-free accessories is strongly attenuated by the body. Numerical simulations using the Finite-Difference Time-Domain (FDTD) method and experimental measurements with a miniature electric-field probe are in good agreement and show a decrease, not an increase, in RF energy exposure in the human head from hands-free accessories.  相似文献
