Laboratory experiments utilizing ecologically relevant irradiances of ultraviolet radiation (UVR) known to occur in shallow Gulf of Maine waters were conducted on the planktonic embryos and larvae of two common benthic echinoids; the green sea urchin Strongylocentrotus droebachiensis and the sand dollar Echinarachnius parma. Significant decreases in survivorship were observed in freshly fertilized embryos of both species with greater mortality in E. parma that was associated with the absence of UVR-absorbing compounds, the mycosporine-like amino acids. Experiments on blastula, gastrula, and prism larval stages of S. droebachiensis also showed significant decreases in survivorship, delays in development, and abnormal embryos and larvae associated with exposure to UVR. Additionally, all developmental stages of S. droebachiensis experimentally exposed to UVR resulted in significant increases in DNA damage, measured as cyclobutane pyrimidine dimer photoproducts. The observed delays in early cleavage and subsequent developmental stages for S. droebachiensis are correlated with DNA damage. It is postulated that cell cycle arrest at critical checkpoints after DNA damage, mediated by a suite of cell cycle genes, is a component of the observed UVR induced developmental delays.  相似文献

Activation of the dopamine type-D2 receptor in late gastrula of sea urchins is known to decrease the growth rate of post-oral arms of larvae, and, as a result, the phenotype of these larvae mimics that of larvae developing in the abundance of food. Our data indicate that the effect of dopamine on sea urchin larvae is stage-dependent. In our experiment, the early four-armed plutei (96 hours post fertilization, hpf) of Strongylocentrotus intermedius had substantially shorter post-oral arms if they developed from the larvae treated with dopamine at the early pluteus stage (48 hpf), when they had already formed the first dopaminergic neurons, as compared to the plutei from the larvae treated with dopamine at the mid to late gastrula stage (24 hpf), when they did not have any neurons yet. The pre-treatment of larvae in 6-hydroxydopamine, a neurotoxic analog of dopamine that specifically disrupts activity of dopaminergic neurons, prevented the development of the short post-oral arms phenotype in larvae. These results confirm the assumption that dopaminergic neurons play an important role in the development of the short post-oral arms phenotype in sea urchin larvae. Another finding of our study is that the dopamine treatment also affects the growth of the body rods and the overall larval body growth. Based on these observations, we suggest researchers to carefully select the developmental stage, pharmacological agents, and incubation time for experimental manipulation of sea urchin larvae phenotypes through dopaminergic nervous system.  相似文献

Lipid peroxidation (LP) and glutathione content were studied at different developmental stages of the sea urchinStrongylocentrotus intermedius: egg cell, fertilization, 4 blastomers, blastula, hatching, gastrula, prism, pluteus. A high rate of LP in the total membrane fraction of sea urchin embryos and larvae at the stages from the egg cell to hatching was observed at enzymatic and nonenzymatic activation of LP. The LP rate was significantly reduced at the gastrula stage and at subsequent stages, there was practically no further development of the process. The glutathione concentration remained unchanged at different stages. The alterations in LP seem to reflect participation of free radicals in regulation of individual development.  相似文献

Sea-urchin embryo RNAs of 9 kb and 7 kb hybridise with a collagen-coding probe. The delta Tm of the hybrids indicates a 70% sequence identity between these RNA regions. Both RNAs are localised in the pluteus endomesoderm, but accumulate over different developmental periods: the 9 kb RNA first appears in the blastula and reaches a maximum concentration during the gastrula stages, while the 7 kb RNA is first detected in the gastrula and is at maximal concentration in the pluteus larva. Animalization by transient exposure of the early stage embryo to Zn2+ alters the developmental profile of the 9 kb collagen mRNA in a way that is clearly different from responses of other mRNAs whose accumulations are initiated during the blastula stage (Nemer, M. Five developmentally regulated sea urchin mRNA sequences which increase in abundance between the blastula and pluteus stages of development were isolated by molecular cloning of cDNA. The regulated sequences all appeared in moderately abundant mRNA molecules of pluteus cells and represented 4% of the clones tested. There were no regulated sequences detected in the 40% of the clones which hybridized to the most abundant mRNA, and the screening procedures were inadequate to detect possible regulation in the 20 to 30% of the clones presumably derived from rare-class mRNA. The reaction of 32P[cDNA] from blastula and pluteus mRNA to dots of the cloned DNAs on nitrocellulose filters indicated that the mRNAs complementary to the different cloned pluteus-specific sequences were between 3- and 47-fold more prevalent at the pluteus stage than at the blastula stage. Polyadenylated RNA from different developmental stages was transferred from electrophoretic gels to nitrocellulose filters and reacted to the different cloned sequences. The regulated mRNAs were undetectable in the RNA of 3-h embryos, became evident at the hatching blastula stage, and reached a maximum in abundance by the gastrula or pluteus stage. Certain of the clones reacted to two sizes of mRNA which did not vary coordinately with development. Transfers of RNA isolated from each of the three cell layers of pluteus embryos that were reacted to the cloned sequences revealed that two of the sequences were found in the mRNA of all three layers, two were ectoderm specific, and one was endoderm specific. Four of the regulated sequences were complementary to one or two major bands and one to at least 50 bands on Southern transfers of restriction endonuclease-digested total sea urchin DNA.  相似文献

Total cell number and number of the primary mesenchyme cells of 1/2 and 1/4 larvae were counted at several developmental stages after hatching in comparison with those of a whole larva, using Clypeaster japonicus as material. To obtain partial larvae, blastomeres were isolated at the 2- or 4-cell stage in Ca-free sea water and cultured in natural sea water at around 23°C. Isolated blastomeres cleaved as in situ, namely, as a part of an embryo. Although each partial embryo tended to spread into a plate, it acquired spherical shape prior to hatching of control whole embryo and developed normally in terms of both developmental rate and morphogenesis. Total cell number of a whole larva was about 620 just after hatching and increased almost linearly until i t reached 1850 at the pluteus stage. A half and quarter larvae contained roughly 1/2 and 1/4, respectively, of the number of cells of whole larva through all stages counted. Numbers of the primary mesenchyme cells in the partial larvae, however, tended to be slightly larger than a half or a fourth of that in whole larva. In whole larva, 35, 50, 56 and 58 was counted at the mesenchyme blastula, early gastrula, late gastrula and pluteus stage, respectively.  相似文献

Larvae of the sea urchin, Evechinus chloroticus, at varying stages of development, were assessed for their potential to survive cryopreservation. Ethylene glycol (EG) and dimethyl sulphoxide (Me2SO), at concentrations of 1-2 M, were evaluated as cryoprotectants (CPAs) in freezing regimes initially based on methods established for freezing larvae of other sea urchin species. Subsequent work varied cooling rate, holding temperature, holding time, and plunge temperature. Ethylene glycol was less toxic to larvae than Me2SO. However, no larvae survived freezing and thawing in EG. Larvae frozen in Me2SO at the gastrula stage and 4-armed pluteus stage regained motility post-thawing. The most successful freezing regime cooled straws containing larvae in 1.5 M Me2SO from 0 to -35 degrees C at 2.5 degrees C min(-1), held at -35 degrees C for 5 min, then plunged straws into liquid nitrogen. Motility was high 2-4 h post-thawing using this regime but decreased markedly within 24 h. Some 4-armed pluteus larvae that survived beyond this time developed through to metamorphosis and settled. Different Me2SO concentrations and supplementary trehalose did not improve long-term survival. Large variation in post-thaw survival was observed among batches of larvae produced from different females.  相似文献

The presence of serotonin binding sites in blastula, gastrula, prism, and pluteus embryos of the sea urchin, Arbacia punctulata, was investigated by the binding of radiolabelled serotonin to dissociated embryo cells. 2. [3H]serotonin binding sites were identified in prism, early pluteus, and advanced pluteus larvae, but not in blastula or gastrula embryos. 3. The ontogeny of [3H]serotonin binding activity closely parallels that of serotonin content as previously reported in Paracentrotus lividus embryos (Toneby, 1977a). 4. The combined effects of temperature (8, 12, 14, 17, 20, 22 and 25°C) and a salinity decrease from 36 to 12‰ on the development of the sea urchin Echinocardium cordatum (Pennant) were studied. Embryonic development proved to be the process most vulnerable to a salinity decrease. It was completed successfully at 8–20°C within a narrow salinity range of 36–28‰ Larvae at the most resistant stage, the blastula, survived at 12–22°C and a salinity of 36–18‰. Larvae at the most sensitive stage, pluteus I with the first pair of arms, died even in a favorable environment, a temperature of 17–20°C and a salinity of 34–28‰. That may be related to qualitative alterations during skeleton formation and to transition to phytoplankton feeding. The resistance of larvae to variations in environmental factors gradually increased in the pluteus II and III stages; however, it significantly decreased before the settling of the larvae. Larvae that were 37 days old survived at a temperature of 14–20°C within a salinity range of 36–22‰ and at 22 and 25°C, they survived at a salinity of 36–24‰; however, all the larvae became abnormal at 25°C. The larvae settled earlier on sand inhabited by adult individuals of E. cordatum than on sand from other locations, and they settled faster at 20–25°C, than at 14 and 17°C. The juveniles, if lacking an opportunity to burrow in the sand, died within 14 days after settling.  相似文献

The sea urchinStrongylocentrotus nudus is highly sensitive to oil pollution. Experiments were performed in winter, spring and summer over periods of 15 to 45 days. Experimental urchins were kept in water with hydrocarbon concentrations of 10 to 30 mg l−1, and control urchins in pure sea water. Thermal stimulation by Evdokimov's method was applied to obtain mature sexual products during winter and spring tests. Summer investigations were conducted at temperatures of 17 to 18 °C. The gonads were studied histologically and morphometrically, and the sexual cells obtained were analyzed at the embryological level. No histological and morphometrical differences were recorded between sexual cells of controls and experimentals. However, marked hydrocarbon effects were observed in the embryonic development of artificially fertilized cells from experimental urchins. Control embryos developed normally. Embryogenesis of artificially fertilized gametes from control females and experimental males, and vice versa, was found to be distinctly abnormal. Many abnormalities were identified at the first cleavage stage, as well as in blastula, gastrula and pluteus. Fertilization of experimental eggs with experimental sperm resulted in serious disturbances of embryos, followed by the development of non-viable larvae. On the whole, embryogenesis of sexual cells from experimental urchins was characterized by prominent delay, asynchronism and presence of abnormal non-viable larvae. Consequently, long-term effects of sublethal hydrocarbon concentrations resulted in the formation of defective sex cells and high larval mortality.  相似文献

Changes in the activity of DNA polymerase-α and in subcellular distribution were studied during gastrulation of the sea urchin, Hemicentrotus pulcherrimus. Although the activity of DNA polymerase-α for each embryo was constant up to the blastula stage as reported previously, the enzyme activity increased during gastrulation by about twofold prior to an increase in its DNA content. Thereafter the enzyme activity remained constant at a high level until the early pluteus stage. During gastrulation, an increase in the fraction of DNA polymerase-α was associated with the rough endoplasmic reticulum. During the period between the gastrula and pluteus stages, the cytoplasmic DNA polymerase-α activity decreased gradually with a concomitant increase of activity in the nucleus fraction. The timing of this increase in the nucleus coincided with the increase of DNA content per embryo. These results suggest that DNA polymerase-α accumulates on the rough endoplasmic reticulum during gastrulation and then translocates to the nucleus for DNA synthesis as seen before the blastula stage. DNA polymerase-α obtained from gastrula nuclei did not associate with the endoplasmic reticulum from gastrulae. DNA polymerase-α obtained from the gastrula endoplasmic reticulum membranes became bound to the salt-washed membranes from gastrulae but not to those from unfertilized eggs. Likewise, DNA polymerase-α from the rough endoplasmic reticulum of unfertilized eggs became attached to salt-washed membranes from unfertilized eggs, but not to those from gastrulae. This suggests that DNA polymerase-α is synthesized anew, and a transition of both DNA polymerase-α and endoplasmic reticulum occurs at the gastrula stage.  相似文献

A rapid, gentle technique is described for the isolation of nuclei from sea urchin embryos. Using this technique, we have analyzed the synthesis and accumulation of nonhistone nuclear proteins during sea urchin development by two-dimensional gel electrophoresis. Most nuclear proteins fall into one of three patterns of synthesis, which are distinguished by maximal rates of accumulation at early (prior to hatching blastula), middle (hatching blastula/gastrula), or late (prism/pluteus) stages of development. Over 60% of observed nuclear proteins undergo apparent qualitative changes in synthesis and accumulation between the 64-cell and pluteus stages. Most of these changes represent appearances of new proteins. A large number of qualitative changes occur very early in development; the period of greatest change is between the 64-cell and 200-cell stages. Over half of the proteins which first appear in the nucleus subsequent to the 64-cell stage are synthesized at stages prior to the time of their initial appearance in nuclei, but are excluded from nuclei for some time.  相似文献

The temperature tolerances of embryonic and early larval development stages of Tripneustes gratilla were investigated from 13-34°C under laboratory conditions. Zygotes showed unequal cleavage at 13°C, whereas cleavage did not occurred at 34°C. Hatching was observed between 16–31°C with maximum hatching rates observed at 22–29°C. The lower and higher temperature limits for embryonic development were approximately 22°C and 29°C, respectively. Outside of this temperature range, embryos showed abnormality at different incubation times. Early larvae of this species have the ability to survive the higher temperature limit for short periods of time. Prism and 2 arm pluteus larvae survived at temperatures between 30 and 33°C, whereas 4 arm pluteus larvae survived at temperatures between 30 and 36°C for 2 h. These results suggest that the larval temperature tolerance capability of T. gratilla is stage dependent. These findings are important for understanding the life history strategy of this sea urchin in the shallow open water environment.  相似文献

The effect of constant and fluctuating salinity on larval development and metamorphosis of the sand dollar Dendraster excentricus was investigated in the laboratory. Sand dollar larvae at different stages of development were kept either at 32‰ (controls), exposed to constant low salinity (22‰) throughout development, or exposed to fluctuating salinity (i.e. transferring larvae from 32‰ to 22‰ for 7 days then back to 32‰ for the rest of their development). Larvae exposed to constant low salinity were significantly smaller but developed all larval arms at a slower rate than larvae in all other treatments. Larvae exposed to fluctuating salinity recovered and developed significantly longer larval arms and bigger rudiments than larvae kept at constant low salinity. Larvae exposed to fluctuating salinity produced more juveniles than larvae at constant high salinity (32‰), while those at constant low salinity produced few or no juveniles. Four-arm larvae exposed to fluctuating salinity produced significantly more juveniles than six-arm larvae exposed to the same treatment. Transferring competent 8-arm larvae from 31‰ to 15‰ for 2 days then back to 31‰, induced metamorphosis with juvenile production being significantly higher than for those kept at a constant salinity of 20, 25 and 31‰. This study indicates that a short-term decrease in salinity might induce metamorphosis for this species.  相似文献

Pesticides are now chronically found in numerous ecosystems incurring widespread toxic effects on multiple organisms. For insects, the larvae are very exposed to pesticide pollution and the acute effect of insecticides on larvae has been characterized in a range of species. However, the carry‐on effects in adults of sublethal exposure occurring in larvae are not well characterized. Here, we use a collection of strains of Drosophila melanogaster differing in their larval resistance to a commonly used insecticide, imidacloprid, and we test the effect of larval exposure on behavioural traits at the adult stage. Focusing on locomotor activity and on courtship and mating behaviour, we observed a significant carry‐on effect of imidacloprid exposure. The heritability of activity traits measured in flies exposed to imidacloprid was higher than measured in controls and in these, courtship traits were genetically less correlated from mating success. Altogether, we did not observe a significant effect of the larval insecticide resistance status on adult behavioural traits, suggesting that selection for resistance in larvae does not involve repeatable behavioural changes in adults. This lack of correlation between larval resistance and adult behaviour also suggests that resistance at the larval stage does not necessarily result in increased behavioural resilience at a later life stage. These findings imply that selection for resistance in larvae as well as for behavioural resilience to sublethal exposure in adult will combine and impose a greater evolutionary constraint. Our conclusions further substantiate the need to encompass multiple trait measures and life stages in toxicological assays to properly assess the environmental impact of pesticides.  相似文献

Coral reefs across the Caribbean are undergoing unprecedented rates of decline in coral cover during the last three decades, and coral recruitment is one potential process that could aid the recovery of coral populations. To better understand the effects of climate change on coral larval ecology, the larvae of Porites astreoides were studied to determine the immediate and post-settlement effects of elevated temperature and associated oxidative stress. Larvae of Porites astreoides were exposed to 27 °C (ambient) and +3.0 °C (elevated temperature) seawater for a short duration of 24 h; then, a suite of physiological parameters were measured to determine the extent of sublethal stress. Following the +3.0 °C treatment, larvae did not show a significant difference in maximum quantum yield of PSII (F v/F m) or respiratory demand when compared to controls maintained at 27 °C. The addition of micromolar concentrations of hydrogen peroxide did not impact respiration or photochemical efficiency. Catalase activity in the larvae increased (>60 %) following exposure to elevated temperature when compared to the controls. Short-term larval survival and settlement and metamorphosis were not affected by increased temperature or the H2O2 treatment. However, the settled spat that were exposed to elevated temperature underwent a 99 % reduction in survival compared to 90 % reduction for the control spat when examined 24 days following the deployment of 4-day-old settled spat on settlement tiles in the field. These results show that short-term exposure to some stressors might have small impacts on coral physiology, and no effects on larval survival, settlement and metamorphosis. However, due to post-settlement mortality, these stressors can cause a significant reduction in coral recruitment.  相似文献
