Twelve species of microalgae, isolated from north Australian marine, freshwater and hypersaline environments, were grown under controlled conditions of temperature, pH, photon flux density and salinity, and analysed for ash, total protein, water soluble carbohydrates, chlorophylla, total lipids, total fatty acids and fatty acid composition. Highest levels of the polyunsaturated fatty acid eicosapentaenoic acid [20:5(n-3)] were found in the marine diatoms.Nitzschia (frustulum) andN. closterium (23.1% and 15.2% of total fatty acids, respectively). None of the species studied had levels of docosahexaenoic acid [22:6(n-3)] greater than 1.1 % of total fatty acids. None of the chlorophyte species contained significant levels of either 20:5(n-3) or 22:6(n-3). The highest total fatty acid concentration of all species in the study was found in the freshwater chlorophyte speciesScenedesmus dimorphus (105 mg g–1 dry wt). The hypersaline speciesDunaliella salina had the highest total lipid content (28.1% dry wt), followed byN. closterium, N. (frustulum) andNavicula sp. (24.2–27.8% dry wt).Chlamydomonas sp. had the highest protein content (66.9% dry wt).N. We cultured Streptocephalus proboscideus using rice bran as a food source, and compared its growth, mortality, fertility, and cyst hatching with published data on animals fed other food sources. We also analysed the fatty acid profile of animals and cysts. The performance of rice-bran fed shrimp was indistinguishable from that of shrimp fed most other formulated diets, but there are indications that some previously applied diets were either defective in essential fatty acids, or toxic.  相似文献

There are increasing interests in rearing edible insects in Africa, but information on how the feeds modify their fatty acids is largely lacking. In this work, the influence of artificial diets on the fatty acid contents and composition in the edible Ruspolia differens (Serville, 1838), in Uganda was assessed. R. differens was reared on the mixtures of six gradually diversified diets of two, three, four, six, eight and nine feeds. The diets were formulated from rice seed head, finger millet seed head, wheat bran, superfeed chicken egg booster, sorghum seed head, germinated finger millet, simsim cake, crushed dog biscuit pellet and shea butter. Fatty acid methyl esters were prepared using direct transesterification method, and analysed using gas chromatography. The contents of saturated, monounsaturated and polyunsaturated fatty acid differed significantly among the diets. The more diverse diets resulted in increased content of the polyunsaturated fatty acids. The n6:n3 ratio differed significantly among the diets and between the sexes, with R. differens fed on the four-feed diet having a higher n6:n3 ratio than those fed on other diets. Also, the fatty acid composition differed significantly among the diets, and diet diversification corresponded with the proportions of polyunsaturated fatty acids, especially linoleic acid. Overall, our results demonstrate that higher levels of essential fatty acids can be achieved by rearing R. differens on highly diversified diets. These findings are important in informing the design of future mass-rearing program for this edible insect.  相似文献

Tetraselmis sp. and Nannochloropsis oculata, cultivated in industrial‐scale bioreactors, produced 2.33 and 2.44% w/w lipid (calculated as the sum of fatty acid methyl esters) in dry biomass, respectively. These lipids contained higher amounts of neutral lipids and glycolipids plus sphingolipids, than phospholipids. Lipids of Tetraselmis sp. were characterized by the presence of eicosapentaenoic acid (that was located mainly in phospholipids), and octadecatetraenoic acid (that was equally distributed among lipid fractions), while these fatty acids were completely absent in N. oculata lipids. Additionally, lipids produced by 16 newly isolated strains from Greek aquatic environments (cultivated in flask reactors) were studied. The highest percentage of lipids was found in Prorocentrum triestinum (3.69% w/w) while the lowest in Prymnesium parvum (0.47% w/w). Several strains produced lipids rich in eicosapentaenoic and docosahexaenoic acids. For instance, docosahexaenoic acid was found in high percentages in lipids of Amphidinium sp. S1, P. parvum, Prorocentrum minimum and P. triestinum, while lipids produced by Asterionella sp. (?) S2 contained eicosapentaenoic acid in high concentration. These lipids, containing ω‐3‐long‐chain polyunsaturated fatty acids, have important applications in the food and pharmaceutical industries and in aquaculture.  相似文献

The total protein, carbohydrate, lipid and ash compositions, and fatty acid contents of two species of marine microalgae, the eustigmatophyte Nannochloropsis oculata (formerly ‘Chlorella sp., Japan’) and the chrysophyte Isochrysis sp. (Tahitian) used in tropical Australian mariculture, were studied. The microalgae were grown under a range of culture conditions (41 and 601 laboratory culture, 3001 bag culture, and 80001 outdoor culture) and four light regimes (100 to 107 μ E m−2 s−1, 240 to 390 μ E m−2 s−1, 340 to 620 μ E m−2 s−1, and 1100 to 1200 μE m−2 s−1 respectively) to determine the effect of light intensity on the chemical composition of large scale outdoor cultures. Laboratory and bag cultures were axenic and cultured in Walne medium while outdoor cultures were grown in a commercial medium designed for optimum nutrition in tropical outdoor aquaculture operations. Change in growth medium and photon flux density produced only small changes in the proximate biochemical composition of both algae. N. oculata and Isochrysis sp. both showed a trend towards slightly lower carbohydrate and higher chlorophyll a in shaded outdoor culture. Isochrysis sp. showed significant concentrations of the essential polyunsaturated fatty acid 22:6(n−3) (docosahexaenoic acid) from 5.3 to 10.3% of total fatty acid, and 20:5(n−3) (eicosapentaenoic acid) ranged from 0.6 to 4.1%. In contrast, N. oculata had high concentrations of 20:5(n−3) (17.8 to 39.9%) and only traces of 22:6(n−3). The fatty acid composition of Isochrysis sp. grown at high photon flux density (1100–1200 μE m−2 s−1) under outdoor culture showed a decrease in the percentage of several highly unsaturated fatty acids, including 20:5(n−3), and an increase in 22:6(n−3). N. oculata showed a similar decrease in the percentage of 20:5(n−3). High light intensity caused a decrease in the ratio of total C16 unsaturated fatty acids to saturated 16:0 in N. oculata, and a decrease in the ratio of total C18 unsaturated fatty acids to saturated 18:0 together with a decrease in the ratio of total unsaturated fatty acids to total saturated fatty acids in both microalgae.  相似文献

The tufted apple budmoth, Platynota idaeusalis (Walker), was reared non-axenically for two successive generations on a casein-based semisynthetic diet. The qualitative essential fatty acid requirement for growth, development and normal pupal-adult ecdysis was studied using the non-axenic casein-based semisynthetic diets with and without various 99% pure fatty acids. Linoleic or linolenic acids caused accelerated larval development; linoleic, linolenic and arachidonic acids showed similar activity in body weight gain and survival to pupal-adult ecdysis. Linoleic or linolenic acids were active in alleviating wing deformities; arachidonic acid was partially active in alleviating wing deformities at the one dietary concentration evaluated. Activity of arachidonic acid as an essential fatty acid for P. idaeusalis is unique among insects, except for mosquitoes. The essential fatty acid deficiency syndrome of the adult, resulting from the larvae feeding on fat-deficient diets, was greatly reduced when larvae were fed on a diet adequate in essential fatty acid during either their early or late development.  相似文献

The role of intracontinental migration patterns of European eel (Anguilla anguilla) receives more and more recognition in both ecological studies of the European eel and possible management measures, but small-scale patterns proved to be challenging to study. We experimentally investigated the suitability of fatty acid trophic markers to elucidate the utilization of feeding habitats. Eight groups of juvenile European eels were fed on eight different diets in a freshwater recirculation system at 20°C for 56 days. Three groups were fed on freshwater diets (Rutilus rutilus, Chironomidae larvae, and Gammarus pulex) and four groups were reared on diets of a marine origin (Clupea harengus, Crangon crangon, Mysis spec., and Euphausia superba) and one on commercial pellets used in eel aquaculture. Fatty acid composition (FAC) of diets differed significantly with habitat. FAC of eel muscle tissue seemed to be rather insensitive to fatty acids supplied with diet, but the general pattern of lower n3:n6 and EPA:ARA ratios in freshwater prey organisms could be traced in the respective eels. Multivariate statistics of the fatty acid composition of the eels resulted in two distinct groups representing freshwater and marine treatments. Results further indicate the capability of selectively restraining certain fatty acids in eel, as e.g. the n3:n6 ratio in all treatments was <4, regardless of dietary n3:n6. In future studies on wild eel, these measures can be used to elucidate the utilization of feeding habitats of individual European eel.  相似文献

The lipid and fatty acid compositions of Chlamydomonas sp. isolated from a volcanic acidic lake and C. reinhardtii were compared, and the effects of pH of the medium on lipid and fatty acid components of Chlamydomonas sp. were studied. The fatty acids in polar lipids from Chlamydomonas sp. were more saturated than those of C. reinhardtii. The relative percentage of triacylglycerol to the total lipid content in Chlamydomonas sp. grown in medium at pH 1 was higher than that in other cells grown at higher pH. A probable explanation might be that Chlamydomonas sp. has two low pH adaptation mechanisms. One mechanism is the saturation of fatty acids in membrane lipids to decrease membrane lipid fluidity, and the other is the accumulation of triacylglycerol, as a storage lipid, to prevent the osmotic imbalance caused by high concentrations of H2SO4.  相似文献

Fatty acids are essential for numerous cellular functions. They serve as efficient energy storage molecules, make up the hydrophobic core of membranes, and participate in various signaling pathways. Caenorhabditis elegans synthesizes all of the enzymes necessary to produce a range of omega-6 and omega-3 fatty acids. This, combined with the simple anatomy and range of available genetic tools, make it an attractive model to study fatty acid function. In order to investigate the genetic pathways that mediate the physiological effects of dietary fatty acids, we have developed a method to supplement the C. elegans diet with unsaturated fatty acids. Supplementation is an effective means to alter the fatty acid composition of worms and can also be used to rescue defects in fatty acid-deficient mutants. Our method uses nematode growth medium agar (NGM) supplemented with fatty acidsodium salts. The fatty acids in the supplemented plates become incorporated into the membranes of the bacterial food source, which is then taken up by the C. elegans that feed on the supplemented bacteria. We also describe a gas chromatography protocol to monitor the changes in fatty acid composition that occur in supplemented worms. This is an efficient way to supplement the diets of both large and small populations of C. elegans, allowing for a range of applications for this method.  相似文献

The influence of diet on the fatty acid composition of the hepatopancreas of Mytilus trossulus was studied. Three groups of mollusks were fed monocultures of the microalgae Phaeodactylum tricornutum, Chaetoceros muelleri (Bacillariophyceae), and Nannochloropsis sp. (Eustigmatophyceae) for 10 days. After 10 days, the proportion of polyunsaturated fatty acids, mainly eicosapentaenoic and docosahexaenoic, increased in the total lipids of the hepatopancreas in all mollusk groups. The content of saturated fatty acids in the mussel tissues decreased and was not dependent on the amount in the algal diet. Toward the end of the experiment, the fatty acid composition of the hepatopancreas of mussels was similar irrespective of the fatty acid composition of their food. The fatty acid analysis of M. trossulus feces suggests a selective assimilation by mussels of predominantly the n-3 polyunsaturated fatty acids. The role of fatty acid metabolism in M. trossulus is discussed.  相似文献

Consumers are aware of foods containing microcomponents that may have positive effects on health maintenance and disease prevention. In ruminant milk, meat, and milk products; these functional food components include eicosapentaenoic acid (20:5n3), docosahexaenoic acid (22:6n3), 9c11t-conjugated linoleic acid, and vaccenic acid (11t-18:1). Modifying ruminal microbial metabolism of fatty acid in rumen through animal diet formulation is an effective way to enhance these functional fatty acids in ruminant-derived food products. However, it requires an understanding of the interrelationship between supply of lipid through the diet and rumen fermentation. Lipids in ruminant diets undergo extensive hydrolysis and biohydrogenation in the rumen. Apparent transfer efficiency of eicosapentaenoic acid and docosahexaenoic acid from feed to milk is very low (1.9 to 3.3%), which is, to a large extent, related to their extensive biohydrogenation in the rumen. Therefore, feeding a rumen-protected supplement containing eicosapentaenoic acid and docosahexaenoic acid, can be used to bypass the rumen. Ruminant-derived foods also contain different types of conjugated linoleic acid isomers, which are intermediates of rumen biohydrogenation of linoleic acid (9c12c-18:2). The predominant isomer of conjugated linoleic acid is 9c11t, which has numerous health benefits in animal models. The concentration of conjugated linoleic acid in ruminant-derived food products can be significantly enhanced through animal diet modification. We conclude that most current functional food products from ruminants have potential for their health-supporting properties, and for this market to succeed, an evidence-based approach should be developed in humans.  相似文献

Six isonitrogenous (approximately 35% crude protein) and isocaloric (approximately 4.0 kcal g−1) diets were formulated incorporating raw and fermented black gram, Phaseolus mungo, seed meal at 20%, 30% and 40% levels by weight into a fishmeal‐based control diet fed to rohu, Labeo rohita, fingerlings (mean weight, 1.81 ± 0.21 g) for 80 days for a study of fish performance. A particular bacterial strain (Bacillus sp.) isolated from the intestine of adult common carp (Cyprinus carpio) reared in the wild having significant amylolytic, cellulolytic, lipolytic and proteolytic activities was used for fermentation of seed meal for 15 days at 37 ± 2°C. Fermentation of P. mungo seed meal was effective in significantly reducing the crude fibre content and antinutritional factors such as tannins and phytic acid, and enhancing available free amino acids and fatty acids. In terms of growth, feed conversion ratio and protein efficiency ratio, the 30% fermented black gram seed meal incorporated diet resulted in a significantly (P < 0.05) better performance of rohu fingerlings. In general, growth and feed utilization efficiencies of diets containing fermented seed meal were superior to diets containing raw seed meal. The apparent protein digestibility (APD) values decreased with increasing levels of raw seed meal in the diets. The APD for raw seed meal was lower at all levels of inclusion in comparison to those for the fermented seed meals. The maximum deposition of protein in the carcass was recorded in fish fed the diet containing 40% fermented seed meal. The results indicate that fermented black gram seed meal can be incorporated in carp diets up to the 30% level compared to the 10% level of raw seed meal.  相似文献

The suitability of cysts of the brine shrimp Artemia sp. as a factitious food for the predator Macrolophus pygmaeus Rambur was investigated. The influence of decapsulation time and hydration of the cysts on the performance of the predator were studied in the absence of plant material. A longer time of decapsulation had a positive influence on the development of the predator. Hydration of cysts had a significant impact on nymphal survival when cysts where non‐decapsulated or poorly decapsulated. An experiment in which nymphs were switched from a diet of hydrated cysts to non‐hydrated cysts showed that in the absence of plant material the relative importance of hydrating the cysts decreased with nymphal age. Especially, the first instar and to a lesser extent the second instar appear to be susceptible to water shortage. Effects of prolonged rearing on development and reproduction on brine shrimp cysts from different origins were tested in the presence of plant material. Rearing M. pygmaeus on Artemia sp. (Jingyu Lake) cysts yielded similar survival, development, adult weight and fecundity in the fourth as in the second generation. In contrast, for Artemia franciscana cysts, an increase in nymphal development was notable. Biochemical analyses showed that total amino acid content and the concentration of the different amino acids did not differ among diets and generations. There were, however, differences in total fatty acid content between the different diets and generations and in the concentration of certain fatty acids, indicating that insects fed brine shrimp cysts may show nutritional deficiencies compared to those reared on a diet of Ephestia kuehniella eggs. Our results indicate that decapsulated brine shrimp cysts are an economically viable alternative food source in at least part of the rearing process for M. pygmaeus.  相似文献

Malaria parasites scavenge nutrients from their host but also harbour enzymatic pathways for de novo macromolecule synthesis. One such pathway is apicoplast‐targeted type II fatty acid synthesis, which is essential for late liver‐stage development in rodent malaria. It is likely that fatty acids synthesized in the apicoplast are ultimately incorporated into membrane phospholipids necessary for exoerythrocytic merozoite formation. We hypothesized that these synthesized fatty acids are being utilized for apicoplast‐targeted phosphatidic acid synthesis, the phospholipid precursor. Phosphatidic acid is typically synthesized in a three‐step reaction utilizing three enzymes: glycerol 3‐phosphate dehydrogenase, glycerol 3‐phosphate acyltransferase and lysophosphatidic acid acyltransferase. The Plasmodium genome is predicted to harbour genes for both apicoplast‐ and cytosol/endoplasmic reticulum‐targeted phosphatidic acid synthesis. Our research shows that apicoplast‐targeted Plasmodium yoelii glycerol 3‐phosphate dehydrogenase and glycerol 3‐phosphate acyltransferase are expressed only during liver‐stage development and deletion of the encoding genes resulted in late liver‐stage growth arrest and lack of merozoite differentiation. However, the predicted apicoplast‐targeted lysophosphatidic acid acyltransferase gene was refractory to deletion and was expressed solely in the endoplasmic reticulum throughout the parasite life cycle. Our results suggest that P. yoelii has an incomplete apicoplast‐targeted phosphatidic acid synthesis pathway that is essential for liver‐stage maturation.  相似文献

Shrimp farming accounts for more than 40% of the world shrimp production. Luminous vibriosis is a shrimp disease that causes major economic losses in the shrimp industry as a result of massive shrimp kills due to infection. Some farms in the South Asia use antibiotics to control Vibrio harveyi, a responsible pathogen for luminous vibriosis. However, the antibiotic-resistant strain was found recently in many shrimp farms, which makes it necessary to develop alternative pathogen control methods. Short-chain fatty acids are metabolic products of organisms, and they have been used as food preservatives for a long time. Organic acids are also commonly added in feeds in animal husbandry, but not in aquaculture. In this study, growth inhibitory effects of short-chain fatty acids, namely formic acid, acetic acid, propionic acid, and butyric acid, on V. harveyi were investigated. Among four acids, formic acid showed the strongest inhibitory effect followed by acetic acid, propionic acid, and butyric acid. The minimum inhibitory concentration (MIC) of 0.035% formic acid suppressed growth of V. harveyi. The major inhibitory mechanism seems to be the pH effect of organic acids. The effective concentration 50 (EC50) values at 96 h inoculation for all organic acids were determined to be 0.023, 0.041, 0.03, and 0.066% for formic, acetic, propionic, and butyric acid, respectively. The laboratory study results are encouraging to formulate shrimp feeds with organic acids to control vibrio infection in shrimp aquaculture farms.  相似文献

Fish are the only major dietary source for humans of -3 highly unsaturated fatty acids (HUFAs) and with declining fisheries farmed fish such as Atlantic salmon (Salmo salar) constitute an increasing proportion of the fish in the human diet. However, the current high use of fish oils, derived from wild capture marine fisheries, in aquaculture feeds is not sustainable in the longer term and will constrain continuing growth of aquaculture activities. Greater understanding of how fish metabolize and biosynthesize HUFA may lead to more sustainable aquaculture diets. The study described here contributes to an effort to determine the molecular genetics of the HUFA biosynthetic pathway in salmon, with the overall aim being to determine mechanisms for optimizing the use of vegetable oils in Atlantic salmon culture. In this paper we describe the cloning and functional characterization of 2 genes from salmon involved in the biosynthesis of HUFA. A salmon desaturase complementary DNA, SalDes, was isolated that include an open reading frame of 1362 bp specifying a protein of 454 amino acids. The protein sequence includes all the characteristics of microsomal fatty acid desaturases, including 3 histidine boxes, 2 transmembrane regions, and an N-terminal cytochrome b5 domain containing a heme-binding motif similar to that of other fatty acid desaturases. Functional expression in the yeast Saccharomyces cerevisiae showed SalDes is predominantly an -3 5 desaturase, a key enzyme in the synthesis of eicosapentaenoic acid (20:5n-3) from -linolenic acid (18:3n-3). The desaturase showed only low levels of 6 activity toward C18 polyunsaturated fatty acids. In addition, a fatty acid elongase cDNA, SalElo, was isolated that included an open reading frame of 888 bp, specifying a protein of 295 amino acids. The protein sequence of SalElo included characteristics of microsomal fatty acid elongases, including a histidine box and a transmembrane region. Upon expression in yeast SalElo showed broad substrate specificity for polyunsaturated fatty acids with a range of chain lengths, with the rank order being C18 > C20 > C22. Thus this one polypeptide product displays all fatty acid elongase activities required for the biosynthesis of docosahexaenoic acid (22:6n-3) from 18:3n-3.  相似文献

Forty psychrophile or psychrotrophic crude-oil-utilizing marine bacteria were investigated for their ability to accumulate lipid storage compounds in the cytoplasm during cultivation under nitrogen-limiting conditions. Most of them (73%) were able to accumulate specialized lipids like polyhydroxyalkanoic acids (PHA) while other lipids such as wax esters occurred in two isolates. Accumulation of PHA occurred predominantly at low temperatures (4–20 °C) as demonstrated for three isolates. Electron microscopy revealed polyphosphate inclusions occurring in two isolates in addition to PHA. Cells of the isolate Acinetobacter sp. 211 were able to synthesize and accumulate lipid inclusions during growth on acetate, ethanol, olive oil, hexadecanol and heptadecane. The composition of the lipid inclusions depended on the compounds provided as carbon source. Wax esters and acylglycerols occurred mainly during the cultivation on olive oil; in contrast, wax esters and free alcohols occurred during cultivation on hexadecanol. Total fatty acids in cells of the Acinetobacter sp. 211 amounted to 25% of the cellular dry weight in olive-oil-grown cells. Palmitic acid was the main fatty acid in the lipids when the cells were cultivated on acetate or ethanol (44% and 32% of total fatty acids respectively). In contrast, fatty acids occurring in the lipids during cultivation on hexadecanol, heptadecane or olive oil were related to the carbon source. The fatty acids present in the accumulated lipids consisted predominantly of saturated and unsaturated straight-chain fatty acids with a chain length ranging from 12 to 18 carbon atoms. Analysis of the lipid-granule-associated proteins in cells of Acinetobacter sp. 211 revealed a protein of 39 kDa as the predominant protein species. In this investigation, we used stable isotope and fatty acid biomarker analyses to estimate and compare the potential food sources that support macrobenthos (Nuttallia olivacea, Corbicula japonica, and Hediste sp.) in the Natori estuarine tidal flats of Japan. The δ13C and δ15N mean values for the sediment organic matter (SOM) were ?23.6‰ and 6.1‰, respectively, which were due to the contribution of terrestrial and/or aquatic vascular plant particulate organic matter (POM) from upper stream river or surrounding areas. Furthermore, from the results of the IsoSource mixing model, the contributions of estuarine POM to the diets of Hediste sp., C. japonica, and N. olivacea were 85.1%, 74.9%, and 48.9%, respectively. Moreover, essential fatty acids such as 20:5ω3, 18:2ω6 and 18:3ω3 highly contributed to the diets of macrobenthos from benthic diatoms, terrestrial and/or aquatic vascular plants. The contents of fatty acid markers of terrestrial OM (e.g., long chain fatty acids [LCFAs]) in the 3 species of macrobenthos were low in comparison to those of other food sources. Overall, the marine POM dietary contribution was minimal, while terrestrial OM, bacteria, and benthic diatoms constituted a significant portion of the macrobenthos diet, although the contribution varied among the 3 species of macrobenthos.  相似文献
