Intraspecific genetic variation of Echinococcus multilocularis, the etiologic agent of human alveolar echinococcosis, has been evaluated among 76 geographic isolates from Europe, Asia and North America by using sequence data of mitochondrial and nuclear DNA. Relatively low genetic variation was found only in the mitochondrial DNA sequence consisting of 3 protein-coding genes. Pairwise divergence among the resultant 18 haplotypes ranged from 0.03 to 1.91%. Phylogenetic trees and parsimony network of these haplotypes depicted a geographic division into European, Asian and North American clades, but 1 haplotype from Inner Mongolia was unrelated to other haplotypes. The coexistence of the Asian and North American haplotypes could be seen, particularly on the St. Lawrence Island in the Bering Sea. These data suggest an evolutionary scenario in which distinct parasite populations derived from glacial refugia have been maintained by indigenous host mammals. The nuclear DNA sequence for the immunodominant B cell epitope region of ezrin/radixin/moesin-like protein (elp) was extremely conservative, indicating that the elp antigen is available for immunodiagnosis in any endemic areas. 相似文献

To understand the relationships between the distribution of Chosenia arbutifolia and Salix sachalinensis and their mycorrhizal colonization, changes in the quality and types of ectomycorrhizas and arbuscular mycorrhizas of the seedlings of two species were studied at five different sites with different soil conditions in the floodplain of the Satsunai River, Hokkaido. High ectomycorrhizal and low arbuscular mycorrhizal colonization were found in roots of both plants. Ectomycorrhizal colonization of S. sachalinensis in wet sandy or muddy soil conditions was at the same level as that in dry gravelly sites. In contrast, ectomycorrhizal colonization of C. arbutifolia seedlings was lower from wet sandy sites than that from dry gravelly sites. In all study sites, the same three morphological types of ectomycorrhizas were dominant. 相似文献

Ten isolates of six species of ectomycorrhizal fungi were grown in vitro at nine concentrations of three sodium salts (NaCl, Na2SO4, Na3C6H5O7) for 4 weeks. Colony diamater, biomass and protein content of fungi were evaluated. Isolates of Pisolithus tinctorius and Suillus luteus were more tolerant of NaCl and Na2SO4 than of Na3C6H5O7. Fungi in the genera Cenococcum, Laccaria, and Thelephora were highly intolerant of Na3C6H5O7 and Na2SO4 in vitro. Biomass and protein content of fungi generally declined with increasing substrate salinity in solution culture. In situ ectomycorrhizal colonization by Laccara laccata and P. tinctorius and the dry weight of Pinus taeda seedlings were significantly reduced by 80 mM NaCl after 14 weeks. Only select ectomycorrhizal fungi appear capable of growth and symbiosis in saline soils. 相似文献

Th effect of fenvalerate 20% EC (a synthetic pyrethroid) on the growth of two ectomycorrhizal fungi, Thelephora terrestris and Laccaria laccata, was studied in vitro. In the case of T. terrestris, mycelial growth was promoted at lower concentrations of the insecticide but inhibited at higher concentrations; mycelial growth of L. laccata decreased with increase in concentration. The effect of the insecticide at three concentrations (0.25, 0.5, 1.5 ppm) was also studied on ectomycorrhizal formation in 4-month-old seedlings of Pinus patula in nursery trials. At lower concentrations the insecticide had no significant effect on mycorrhizal formation. 相似文献

Where its populations are isolated in higher mountain regions, Abies mariesii is one of the more important conifers of Japan's alpine forest zone. In this study we tried to clarify the genetic variation of chloroplast DNA (cpDNA) in A. mariesii. Cones and fresh needles were collected from seven mountain regions. Total DNAs were extracted from individual seedlings, and these were digested by 15 restriction endonucleases. Southern hybridization was then done using cpDNA clones of Cryptomeria japonica and tobacco as probes. CpDNA variation was detected with enzyme-probe combinations: HindIII+pCS10 probe, HindIII+pCS7, and BglII+pCS7 in preliminary screening. These variations were considered to be caused by the same insertion, deletion or inversion. All populations surveyed for the combination HindIII+pCS10 resulted in only two frequency variations in each population. This indicates a gradual cline along latitude and longitude. 相似文献

Forest ecosystems of the Pacific Northwest of the USA are changing as a result of climate change. Specifically, rise of global temperatures, decline of winter precipitation, earlier loss of snowpack, and increased summer drought are altering the range of Pinus contorta. Simultaneously, flux in environmental conditions within the historic P. contorta range may facilitate the encroachment of P. ponderosa into P. contorta territory. Furthermore, successful pine species migration may be constrained by the distribution or co-migration of ectomycorrhizal fungi (EMF). Knowledge of the linkages among soil fungal diversity, community structure, and environmental factors is critical to understanding the organization and stability of pine ecosystems. The objectives of this study were to establish a foundational knowledge of the EMF communities of P. ponderosa and P. contorta in the Deschutes National Forest, OR, USA, and to examine soil characteristics associated with community composition. We examined EMF root tips of P. ponderosa and P. contorta in soil cores and conducted soil chemistry analysis for P. ponderosa cores. Results indicate that Cenococcum geophilum, Rhizopogon salebrosus, and Inocybe flocculosa were dominant in both P. contorta and P. ponderosa soil cores. Rhizopogon spp. were ubiquitous in P. ponderosa cores. There was no significant difference in the species composition of EMF communities of P. ponderosa and P. contorta. Ordination analysis of P. ponderosa soils suggested that soil pH, plant-available phosphorus (Bray), total phosphorus (P), carbon (C), mineralizable nitrogen (N), ammonium (NH4), and nitrate (NO3) are driving EMF community composition in P. ponderosa stands. We found a significant linear relationship between EMF species richness and mineralizable N. In conclusion, P. ponderosa and P. contorta, within the Deschutes National Forest, share the same dominant EMF species, which implies that P. ponderosa may be able to successfully establish within the historic P. contorta range and dominant EMF assemblages may be conserved. 相似文献

Summary The seasonal and diurnal water relations were investigated within arctic and alpine populations of the dwarf willow Salix arctica. Marked differences that were habitat dependent (e.g. xeric vs. mesic) occured both within and between the populations. The environmental variables that most affected plant water balance and the bulk tissue water relations were soil water potential (soil) and the leafto-air water vapor pressure gradient (w), however, low soil temperature (<4.0° C) also had a marked effect in the wet to mesic habitats. The effects of declining soil and increased w were most pronounced in the plants growing in xiric habitats in both populations. Stomatal response to increased w was two-fold greater in alpine versus arctic plants and is hypothesized to have arisen in response to more frequent exposure to the higher evaporative conditions of alpine existance. Seasonal fluctuations in the osmotic potential closely followed changes in soil, suggesting that these were active rather that passive changes. Additionally, plants from xeric habiats had a lower bulk tissue elastic modulus (more elastic tissues) in both arctic and alpine populations. The osmotic and elastic properties enhanced turgor maintenance over a broad range of leaf water potentials and during periods when w was high. Turgor maintenance also correlated to continued transpiration despite fluctuations in soil and atmospherically induced water deficits. Arctic habitats have a shorter growing season, lower soil temperatures due to the presence of permafrost, but higher soil water potentials and lower leaf-to-air vapor pressure gradients than alpine habitats. The observed variation in patterns of stomatal conductance and in values of tissue water relations characteristics between arctic and alpine populations of S. arctica is hypothesized to have arisen in response to these different environmental regimes which represent different selective regimes that occur along the arctic-alpine environmental continuum inhabited by this wide ranging species. 相似文献

Silicone fouling-release coatings represent a non-toxic alternative to biocide-containing ship hull paints. These coatings allow fouling organisms to attach to the hull surface, but prevent firm adhesion. Adhesive tenacity to fouling-release materials varies both among and within species. We quantified broad-sense genetic and environmental sources of intraspecific variation in tenacity to two silicone substrata, for the barnacle Balanus amphitrite. For both materials tenacity varied over an order of magnitude; however, the partitioning of this variation differed between the substrata. For International Veridian, a commercially-available fouling-release coating, removal stress varied significantly among maternal families and replicate barnacle cultures. Variation among the maternal families was associated with previously observed differences among these families in the condition of the adhesive plaque. Additional experiments suggested that variation among the replicate cultures arose from heterogeneity between replicate coatings in properties that affect tenacity. We could not attribute variation in removal stress for Dow Corning Silastic T-2, a silicone rubber used for mold-making, to any of the genetic or environmental sources tested. Instead, variation may have been due to measurement error or heterogeneity within replicate coatings in properties affecting tenacity. Differences among maternal families in removal stress may stem from variation in the interaction between the adhesive and the substratum, or in the viscoelastic properties of the adhesive plaque. 相似文献

Many studies have analysed plant responses to flooding or drought separately, without addressing the relations between plant resistance to each of these factors. In this paper, we compare the responses to drought and flooding under glasshouse conditions of three populations of Paspalum dilatatum, a perennial C4 grass dominant at different positions along a topographic gradient in the flooding pampa of Argentina. Our results showed that flooding effects on yield were negative on an upland, null on an intermediate, and positive on a lowland population, whereas drought reduced yield equally across populations, showing that resistance to flooding was not related to resistance to drought at a population level. Drought decreased height and aerenchyma, and increased the proportion of roots, while flooding had opposite effects on these traits. The responses of the single clones that made up each population showed a positive relation between the resistances to both factors: along the ecocline formed by 58 clones, those more resistant to drought were also more resistant to flooding. In addition, the combined resistance of each clone to both factors was negatively related to yield at field capacity, (i.e. the most resistant clones were less productive) and unrelated to the proportion of roots and aerenchyma. This result agrees with predictions of Grime's plant strategy theory and differs from a few previous studies, which showed negative relations between the resistances to flooding and drought among genera and species. 相似文献

Picea sitchensis and Pseudotsuga menziesii seedlings were grown in containers, inoculated with ectomycorrhizal fungi, and planted in British forestry sites. Root samples taken during the year after planting were assessed for mycorrhiza formation. Survival and shoot height were assessed at the end of each year. Observations were made each autumn on the occurrence of sporophores of ectomycorrhizal fungi. Pot experiments were used to assess the colonization potential of soils from the experimental locations. Assessment of mycorrhiza formation by the inoculant fungi both before planting and the following year showed much variation among the fungi used. Similar variation was found among field sites. Inoculation with Laccaria isolates was most successful. Height measurements are reported for the first 2 years after planting, at which time there were few significant effects on growth of Picea sitchensis or Pseudotsuga menziesii seedlings. Experimental assessment of colonization potential was of little value in this work for predicting events in the forest. 相似文献

Summary Mycelial inocula of the two ectomycorrhizal fungi,Pisolithus tinctorius (Pt) andThelephora terrestris (Tt) were artificially introduced into fumigated nursery soil before seed sowing. At the end of the first growing season, Pt stimulated the height growth ofPinus densiflora, P. thunbergii, P. rigida, andP. rigida×taeda by 55, 36, 69, and 37%, respectively, compared with control seedlings (with no fumigation and no inoculation). When growth stimulation was expressed by dry weight, Pt increased dry weight ofP. densiflora andP. rigida×taeda by 143% and 128%, respectively, over control seedlings. Total absorption (per plant top) of nitrogen and phosphorus of inoculatedP. densiflora seedlings was also increased by 188 and 144%, respectively, even though the concentration of these two elements in the tissue was not significantly different from that of control seedlings.Tt failed to stimulate growth of the inoculated plants possibly due to its lower level of infection (40–60% mycorrhizal tips) compared with the relatively high infection (over 80%) of Pt.Pinus koraiensis did not respond to inoculation during the first growing season, probably due to its inherent characteristics of producing a small number of short roots and its slow shoot growth during the first few years after germination.This study was supported by the International Foundation for Science in Stockholm, Sweden. 相似文献

The main ecological factors that are hypothesized to explain the striking variation in the size of social groups among large herbivores are habitat structure, predation, and forage abundance and distribution; however, their relative roles in wild populations are not well understood. I combined analyses of ecological correlates of spatial variation in group size with analyses of individual behaviour in groups of different sizes to investigate factors maintaining variation in group size in an Indian antelope, the blackbuck Antilope cervicapra. I measured group size, habitat structure, forage, and the occurrence of predators in ten blackbuck populations, and, at a smaller spatial scale, within an intensively studied population. To examine the processes by which these ecological factors influence group size, I used behavioural observations and an experiment to estimate the shape of the relationship between group size and potential costs and benefits to individuals. Group size varied extensively both among and within populations. Analyses of spatial variation in group size suggested that both forage and habitat structure influence group size: large-scale, among-population variation in group size was primarily related to habitat structure, while small-scale, within-population variation was most closely related to forage abundance. Analyses of individual behaviour suggested that larger groups incur greater travel costs while foraging. However, individuals in larger groups appeared to experience greater benefits, namely the earlier detection of a “predator”, a reduction in vigilance, and an increase in the time spent feeding. Overall, these findings suggest that individuals in groups experience a trade-off between predation-related benefits and costs arising from feeding competition. Habitat structure and forage likely influence the nature of this trade-off; thus, variation in these ecological factors may maintain variation in group size. The role of predation pressure and other factors in explaining the remaining variation needs further exploration. Genetic variation in dominant species can affect plant and ecosystem functions in natural systems through multiple pathways. Our study focuses on how genetic variation in a dominant riparian tree (Populus fremontii, P. angustifolia and their natural F1 and backcross hybrids) affects whole-tree water use, and its potential ecosystem implications. Three major patterns were found. First, in a 12-year-old common garden with trees of known genetic makeup, hybrids had elevated daily integrated leaf-specific transpiration (Etl ; P=0.013) and average canopy conductance (Gc ; P=0.037), with both Etl and Gc ~30% higher in hybrid cross types than parental types. Second, 13C values of leaves from these same trees were significantly more negative in hybrids (P=0.004), and backcross hybrids had significantly more negative values than all other F1 hybrid and parental types (P <0.001). Third, in the wild, a similar pattern was found in leaf 13C values where both hybrid cross types had the lowest values (P <0.001) and backcross hybrids had lower 13C values than any other tree type (P <0.001). Our findings have two important implications: (1) the existence of a consistent genetic difference in whole-tree physiology suggests that whole-tree gas and water exchange could be another pathway through which genes could affect ecosystems; and (2) such studies are important because they seek to quantify the genetic variation that exists in basic physiological processes—such knowledge could ultimately place ecosystem studies within a genetic framework. 相似文献

We studied herbivory of two species of willows (Salix sericea and S. eriocephala) and their interspecific hybrids to test alternative hypotheses concerning the effects of hybridization on plant resistance. Individually marked plants were identified using morphological traits in the field and random amplified polymorphic DNA (RAPD) band analysis was used to verify the genetic status of many parental and hybrid plants. The desities of 12 herbivore species on plants in the field were compared between two parents and their F2-type hybrids. We found about equal support for the additive, dominance, and hybrid susceptibility hypotheses over 4 years. In one year, one species supported the hybrid resistance hypothesis. Guild membership was not a good predictor of similar responses of species to hybrid versus parental plants. There were marked differences in support for particular hypotheses among years for four herbivore species. This study demonstrates the diversity of responses of phytophages in response to interspecific hybridization, and indicates that year-to-year variation in relative resistance of hybrid plants can be important. 相似文献

Summary A study of the effects of defoliation by insects on the chemistry of lodgepole pine (Pinus contorta), and on the performance of Panolis flammea (Lepidoptera; Noctuidae) larvae, was carried out in a forest in northwest Scotland I year after a severe outbreak of P. flammea had caused extensive defoliation. Larval weight and survival were not significantly different on trees that had experienced different levels of defoliation in 1986. The nitrogen and tannin content of current and previous years' pine needles was not significantly affected by defoliation (although both were slightly greater in the foliage of defoliated trees). Phosphorus content of young pine foliage was lower (but not significantly lower except on one occasion) on heavily defoliated trees. On all sampling occasions, however, the nitrogen: phosphorus ratio was significantly higher on heavily defoliated trees. There were large differences in monoterpene composition of the previous year's shoots associated with defoliation intensity, but these differences had largely disappeared in the new growth. The results are discussed in relation to other studies on the effects of insect damage on plant chemistry and insect performance and in relation to the abundance of P. flammea in Scotland. 相似文献

Summary Chloroplast DNA variation has been used to examine some of the maternal lineages involved in the evolution of the intraspecific polyploid complex, Dactylis glomerata L. Diploid (2x) and tetraploid (4x) individuals were collected from natural populations of the subspecies glomerata (4x), marina (4x) and lusitanica (2x), as well as from sympatric 2x/4x populations of the Galician type. Digestion of their ctDNA with 11 restriction endonucleases revealed enough variation to characterise three ctDNA variants, designated MBMK, MBmK and mBMK. The distribution of these ctDNA variants reflects different stages in their spread among the populations. The MBMK ctDNA variant predominated at both ploidy levels in subspecies glomerata, lusitanica and marina, and in recent tetraploid Galician/glomerata hybrids. The MBmK variant was detected in a single tetraploid individual and probably results from a relatively recent mutation. Fixation of the mBMK minority variant in the diploid and tetraploid Galician populations adds to the evidence concerning the possible origin of the Galician tetraploids. It means that the Galician diploids were maternal ancestors of the tetraploids. This result complements evidence from earlier studies based on morphology or biochemical markers, and reduces the likelihood that the tetraploids arose by hybridisation between an ancient Galician diploid and an alien tetraploid. It is, however, consistent with a true autopolyploid origin of the tetraploids. 相似文献

Interspecific and intraspecific morphological and fertilization variations were studied in three sibling species of Echinometra (known as sp. B, C and D) found off the coast of Okinawa, Japan. Eggs from C and D were readily fertilized by sperm from all three color morphs of B when the sperm concentrations were high, but no fertilization was observed when sperm of the former were mixed with eggs from the latter. Under limiting sperm concentrations, however, both C and D were incapable to fertilize reciprocally with B. In contrast, crossing between C and D produced fertilization membrane at high and limiting sperm concentrations in both directions. Interspecific crosses between B vs. C and B vs. D clearly showed that these combinations were reproductively isolated in contrast to that observed for crossing between C and D. Interestingly, intraspecific fertilization of B showed considerable morphological variation in addition to variability in fertilization success. Intraspecific fertilization and morphological variations may occur due to a number of genetic and/or non-genetic factors. While the underlying cause(s) remain to be elucidated, the results of the present study suggest that B is now speciating very slowly. 相似文献

Genetic diversity has been investigated by the application of molecular markers in, for the first time, all the taxa recognised in recent treatises of the genusHippophae. RAPD (random amplified polymorphic DNA) analyses were conducted with 9 decamer primers, which together yielded 219 polymorphic markers. We found 16 fixed RAPD markers, i.e. markers that either occurred in all plants of a population or were absent from all plants. Several of these markers were useful for analysis of interspecific relationships, whereas others can be considered as taxon-specific markers. Clustering of taxa and populations in our neighbour-joining based dendrogram was in good agreement with some recently suggested taxonomic treatises ofHippophae. Amount and distribution of genetic variability varied considerably between species. Partitioning of molecular variance withinH. rhamnoides supported earlier findings that a considerable part of the total variance resides among subspecies (59.6%) Within-population variability also differed considerably. Percentage polymorphic RAPD loci and Lynch and Milligan within-population gene diversity estimates showed relatively high values for some species close to the geographic centre of origin in Central Asia, e.g.H. tibetana and the putatively hybridogenousH. goniocarpa. Spatial autocorrelation analyses performed on 12 populations ofH. rhamnoides revealed positive autocorrelation of allele frequencies when geographic distances ranged from 0 to 700 km, and no or negative autocorrelation at higher distances. At distances between 700 and 1900 km, we observed deviations from the expected values with strongly negative autocorrelation of allele frequencies. A corresponding relationship between geographic and genetic distances could not be found when the analysis instead was based on one population from each of 8 species. 相似文献

Between-tree variation in the terpenoid composition in the foliage of six species of Eucalyptus was investigated in relation to its effects on herbivory by Christmas beetles (Anoplognathus spp.). All six eucalypt species showed considerable intraspecific variation in terpenoid composition; the cineole content ranged from 13% to 78% of the total oil in different E. melliodora trees, from 0% to 67% in E. conica, 3% to 79% in E. sideroxylon, 1% to 76% in E. camaldulensis, 3% to 60% in E. rubida and 20% to 79% in E. blakelyi. Levels of defoliation by Christmas beetles of nine E. melliodora and eight E. conica trees were quantified from frass traps placed under each tree, and were used to confirm the reliability of visual ratings of defoliation. Defoliation was assessed visually for all six species and was found to be unrelated to the total amount of terpenoids in the foliage of each tree, but strongly associated with the percentage of cineole in the terpenoid mixture. Levels of most terpenoid components were significantly inter-correlated, so it was not possible to determine which components directly affected defoliation. The dominant Christmas beetle at all sites was A. montanus. 相似文献

Manipulation of water levels, andthe presence/ absence of competitorspecies, produced significant morphologicalresponses (e.g. resource allocation toabove- and below-ground structures; plantheight; leaf size; tiller production) inmature individuals of five freshwaterwetland plant species characteristic ofBritish northern poor-fen habitats. Theseresponses provide evidence for potentialadvantages in survival and ability tospread vegetatively.
