Two polysaccharide fractions (PSPO-1a and PSPO-4a) were isolated from the fruiting bodies of Pleurotus ostreatus using ethanol precipitation, anion-exchange chromatography and gel permeation chromatography. Both fractions were heteropolysaccharide containing protein and uronic acid. PSPO-1a was composed of mannose, glucose, galactose, xylose and rhamnose with a molar ratio of 2.47:0.91:1.00:1.66:3.87. PSPO-4a was composed of only three monosaccharides: rhamnose, mannose and galactose with a molar ratio of 0.92:2.69:1.00. The average molecular weight of PSPO-1a and PSPO-4a determined by HPLC were estimated to be 1.8 × 10(4)Da and 1.1 × 10(6)Da respectively. The in vitro tests revealed that two polysaccharides were natural potential antioxidant. Both polysaccharides presented stronger DPPH radical and superoxide anion radical scavenging activity with increasing concentrations, but less effective on scavenging hydroxyl radical. Compared with PSPO-4a, PSPO-1a was the more effective free-radical scavenger. In conclusion, the two polysaccharides may be useful as a naturally potential antioxidant agent for application in food and medicinal fields.  相似文献

To gain insights into dimorphism, cell wall polysaccharides from Tremella fuciformis strains were obtained from alkali-extracted water-soluble fractions PTF-M38 (from the mycelial form), PTF-Y3 and PTF-Y8 (from the yeast form) of T. fuciformis strains were used to gain some insights into dimorphism study. Their chemical properties and structural features were investigated using gel permeation chromatography, gas chromatography, UV and IR spectrophotometry and Congo red binding reactions. The results indicated that the backbones of PTF-M38, PTF-Y3 and PTF-Y8 were configured with α-linkages with average molecular weights of 1.24, 1.08, and 1.19 kDa, respectively. PTF-M38 was mainly composed of xylose, mannose, glucose, and galactose in a ratio of 1:1.47:0.48:0.34, while PTF-Y3 and PTF-Y8 were mainly composed of xylose, mannose and glucose in a ratio of 1:1.65:4.06 and 1:1.21:0.44, respectively. The sugar profiles of PTF-M38, PTF-Y3 and PTF-Y8 were also established for further comparison. These profiles showed that all three polysaccharides contained the same sugars but in different ratios, and the carbon sources (xylose, mannose, glucose, and galactose) affected the sugar ratios within the polysaccharides.  相似文献

Mature fruit of Chorisia speciosa yield an exudate (E-I) following mechanical injury. The polysaccharide contains rhamnose, arabinose, xylose, mannose, glucose, galactose and glucuronic acid in molar ratios of 20:11:1:3:2:40:23. The main chain of the structure is composed by beta-galactopyranosyl units linked (1 --> 3) and (1 --> 6) as indicated by NMR spectra and methylation data. Arabinosef and rhamnose are terminal residues. In order to compare E-I with the polysaccharides from the fruit mesocarp, the latter was submitted to different extractions. The water fraction contains rhamnose, arabinose, xylose, mannose, glucose, galactose and uronic acid in molar ratios of 18:4:1:2:3:44:28. It was treated with CTAB yielding a precipitate which was decomplexed with NaCl, giving four fractions. The fraction obtained using 0.15 M NaCl had a quantitative composition similar that of E-I.  相似文献

In this paper, improvement in the method for purifying glycoprotein from green tea (Camellia sinensis) was described; some properties and chemical compositions of tea glycoprotein (TGP) were determined by HPGPC, FT-IR, GC–MS technologies. Compared to existing methods, a more compatible method for purifying TGP was proposed. This method was faster, simpler, more effective and easier to be extended to the industrial production than the method that used in our previous work. The molecular weight of TGP was 126,513 Da using HPGPC. GC–MS analysis of TGP showed that TGP was composed of seven kinds of monosaccharides, namely ribose, rhamnose, arabinose, xylose, mannose, glucose, galactose in molar ratios of 1.71:5.88:13.70:1.99:1.00:1.84:33.75. Eighteen amino acids were identified in TGP by amino acid analysis. The FT-IR spectrum of the TGP revealed also typical characteristics of polysaccharides, protein and uronic acid.  相似文献

用热水从香菇子实体中浸提出香菇多糖,采用两种超滤陶瓷膜将粗多糖分级成三部分Le1,Le2和Le3。所有的这三种多糖都由两组分所组成,采用凝胶过滤色谱测定了多糖分子量,13CNMR和IR光谱测定显示多糖Le1为含α糖甙键的多糖,多糖Le3为含β糖甙键的多糖。采用气相色谱法测定了三种多糖的单糖组成,结果显示三种多糖都由葡糖糖,阿拉伯糖,木糖,甘露糖和半乳糖组成,Le1,Le2和Le3中阿拉伯糖、木糖、甘露糖、半乳糖、葡萄糖的摩尔比分别为0.15∶0.52∶1.00∶1.20∶7.20、0.21∶0.68∶1.00∶1.02∶11.56、0.29∶0.42∶1.00∶0.85∶16.20。三种多糖Le1,Le2和Le3的平均分子量分别为4.02×104、2.16×105和8.93×105。  相似文献

Two extracellular polysaccharides, ENP1 and ENP2, were isolated from the fermentation liquid of the marine fungus Epicoccum nigrum JJY-40 by anion-exchange chromatography and gel-filtration chromatography, and their structures were investigated using chemical and spectroscopic methods including methylation analysis and NMR spectroscopy. The results demonstrated that ENP1 was composed of mannose, glucose, and galactose in the molar ratio of 5.0:2.1:1.0, and the main chain of the polysaccharide consisted of (1?→?2)-linked mannose, (1?→?3)-linked mannose, terminal mannose, (1?→?6)-linked glucose, (1?→?4)-linked glucose, and (1?→?4)-linked galactose. ENP2 was composed of mannose, galactose, glucose, and glucuronic acid in a molar ratio of 12.4:11.2:8.3:1.0, and its glycosidic linkage patterns included terminal mannose, (1?→?6)-linked glucose, (1?→?4)-linked galactose, and (1?→?3)-linked mannose. The two polysaccharides had a partially branched structure with branch point located at C-3 position of (1?→?6)-linked glucose residue. The molecular weights of ENP1 and ENP2 were 19.2 kDa and 32.7 kDa, respectively. Antioxidant properties of the two polysaccharides were evaluated with hydroxyl, superoxide, and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activities and lipid peroxidation inhibition in vitro, and results showed that ENP2 and ENP1 had good antioxidant activities, especially ENP2. Hot water-soluble polysaccharides were extracted from field colonies and suspension cultures of Nostoc commune Vaucher, Nostoc flagelliforme Berkeley et Curtis, and Nostoc sphaeroides Kützing. Excreted extracellular polymeric substances (EPS) were isolated from the media in which the suspension cultures were grown. The main monosaccharides of the field colony polysaccharides from the three species were glucose, xylose, and galactose, with an approximate ratio of 2:1:1. Mannose was also present, but the levels varied among the species, and arabinose appeared only in N. flagelliforme. The compositions of the cellular polysaccharides and EPS from suspension cultures were more complicated than those of the field samples and varied among the different species. The polysaccharides from the cultures of N. flagelliforme had a relatively simple composition consisting of mannose, galactose, glucose, and glucuronic acid, but no xylose, as was found in the field colony polysaccharides. The polysaccharides from cultures of N. sphaeroides contained glucose (the major component), rhamnose, fucose, xylose, mannose, and galactose. These same sugars were present in the polysaccharides from cultures of N. commune, with xylose as the major component. Combined nitrogen in the media had no qualitative influence on the compositions of the cellular polysaccharides but affected those of the EPS of N. commune and N. flagelliforme. The EPS of N. sphaeroides had a very low total carbohydrate content and thus was not considered to be polysaccharide in nature. The field colony polysaccharides could be separated by anion exchange chromatography into neutral and acidic fractions having similar sugar compositions. Preliminary linkage analysis showed that 1) xylose, glucose, and galactose were 1→4 linked, 2) mannose, galactose, and xylose occurred as terminal residues, and 3) branch points occurred in glucose as 1→3,4 and 1→3,6 linkages and in xylose as a 1→3,4 linkage. The polymer preparations from field colonies had higher kinematic viscosities than those from correspondingsuspension cultures. The high viscosities of the polymers suggested that they might be suitable for industrial uses.  相似文献

Quinoa crude polysaccharides (QPS) were extracted from Chenopodium quinoa Willd. The soluble non-starch polysaccharide fraction (QPS1) was subsequently purified by DEAE-52 cellulose and Sephadex G-50 gel chromatography, using QPS as raw materials. Its chemical structure was identified using FT-IR, NMR, AFM, SEM and Congo red staining. High performance gel permeation chromatography (HPGPC) was used to determine molecular weight, and composition by HPLC. QPS1, with a molecular weight of 34.0 kDa, was mainly composed of mannose, rhamnose, galacturonic acid, glucose, galactose, xylose and arabinose at a molar ratio of 2.63:2.40:1.64:6.28:1.95:2.48:5.01. In addition, we evaluated the ameliorative effects of QPS1 on the improvement of anti-cyclophosphamide (CTX)-induced immunosuppression in ICR mice. The result exhibited significantly immune-enhancing activity: QPS1 successfully improved the content of IFN-γ, IL-6, IFN-ɑ, IgM and lysozyme (LYSO) in serum for three weeks, enhanced the phagocytic function of mononuclear macrophages and ameliorated delayed allergy in mice.  相似文献

Changes in mechanical properties and chemical nature of the cell walls of the different zones along elongating maize ( Zea mays L. cv. LG 11) roots were analyzed and the following results were obtained. (1) The apical region 2 to 5 mm from the tip of 15 mm long roots showed rapid elongation whereas the region 8–10 mm from the tip showed very little growth. (2) The minimum stress-relaxation time (To) and the mean stress-relaxation rate (R) of the cell wall were small whereas the maximum stress-relaxation time (Tm) was large in the region where cell elongation was optimum. The To and R increased and the Tm decreased gradually towards the base of the root. (3) The amounts of non-cellulosic polysaccharides of the cell wall were highest in the region 1.5–2.5 mm from the tip, decreasing until 5 mm from the tip, and then increasing towards the base. However, the proportion of this fraction in the total cell wall polysaccharides was highest in the extreme tip (cap and meristem, 0–1 mm) and decreased towards the base. (4) Major neutral sugars constituting the non-cellulosic polysaccharides of the cell wall were xylose, arabinose, galactose and glucose, with minor amounts of rhamnosc. mannose and fucose. The 1–15 mm region was on the whole rich in glucose and xylose and contained arabinose to a lesser extent. However, the chemical nature in the apical region, (0–2 mm, was rather special, being rich in galactose and fucose. (5) The cell wall of maize roots contained, as a whole, only little pectic substances but was high in hemicellulose 1 (rich in xylose, arabinose and glucose) and hemicellulose 2 (rich in glucose and xylose). (6) It appeared that in the elongating region (apical 2 to 5 mm) the cell elongation rate (CET) showed a rather good correlation with the parameters of mechanical properties (To, Tm and R) and with neutral sugar compositions in the non-cellulosic polysaccharides.  相似文献

The extracellular polysaccharides (EPS) of six strains of cowpea rhizobia were examined. The strains (MI50A, M6-7B, IRC253) produced polysaccharides containing glucose, galactose and mannose in a molar ratio of 2:1.1:1, 1:1.3:3.1 and 1:1.3:3.5 respectively. Two strains (513-B and Ez-Aesch) produced polysaccharides containing galactose and mannose in a molar ratio of 2:3. Mannose was the only sugar detected in the EPS of strain IRC291. Four neutral polysaccharides (ESBP1-1, ESBP1-2, ESBP2-1 and ESBP3-1) were successfully purified from the water extracted crude polysaccharides of Erythronium sibiricum bulbs through the combination of DEAE Sepharose CL-6B and Sephadex G-100 chromatography; their average molecular weights were 1.3?×?104, 1.7?×?104, 9.4?×?105 and 4.1?×?105 Da, respectively. Monosaccharide component analysis indicated that ESBP1-1 and ESBP1-2 were mainly composed of glucose (Glc). ESBP2-1 was composed of Glc, galactose (Gal) and arabinose, with a molar ratio of 24.3:1.1:1, whereas ESBP3-1 comprised Glc and Gal at a molar ratio of 14.8:1. In-vitro study showed that all of the four polysaccharides were able to considerably promote the proliferation and neutral red phagocytosis of RAW 264.7 macrophage cell. They could also stimulate the production of the cell lines’ secretory molecules [nitric oxide, tumour necrosis factor-α (TNF-α) and interleukin-1β (IL-1β)] in a dose-dependent manner. However, ESBP1-2 was not included in IL-1β. Overall, these results suggested that polysaccharides from E. sibiricum bulbs can be developed as immunomodulatory ingredients for complementary medicines or functional foods. However, further animal or clinical studies are required.

In this paper, the composition and biological activities of polysaccharides from Inula britannica flower IBP obtained by water extraction were investigated. The properties and chemical compositions of IBP were analyzed with HPLC and IR methods. The results showed that IBP consisted of two kinds of polysaccharides with the molecular weight of 3500Da, 700Da. IBP consisted of mannose, glucuronic acid, rhamnose, galacturonic acid, glucose, galactose, arabinose with a molar ratio of 4.1:1:1.4:2.7:14.6:6.3:7.9. The IR spectrum of IBP revealed the typical characteristics of polysaccharides and protein. IBP was administered orally at three doses [100, 200 and 400mg/kg body weight] for 14 days to the diabetic mice induced by alloxan. The body weight, plasma glucose, serum triglyceride (TG), total cholesterol (TC), low density lipoprotein cholesterol (LDL-C), high density lipoprotein cholesterol (HDL-C) and liver glycogen were evaluated in normal and alloxan-induced diabetic mice. IBP could dose-dependently significantly increase the body weight of diabetic mice, and reverse the decrease of plasma glucose, glycogen and the decrease of blood lipid of diabetic mice as compared to those in control group. These results indicated that IBP could be developed to a potential anti-diabetic drug in the future.  相似文献

Aeromonas salmonicida grown in a medium with excess glucose as carbon source produces both capsular and exocellular polysaccharides. The capsular polysaccharide is composed of glucose, mannose, rhamnose, N-acetylmannosamine and mannuronic acid in the molar ratios of approximately 5:3:0.75:2:1. The extracellular polysaccharide is similarly constituted, but in the molar ratios of approximately 4.75:10.5:1.5:2:1. The capsular and exocellular polysaccharides did not cross-react with monoclonal antibodies against the A-layer or the O-antigen lipopolysaccharide.  相似文献

Three polysaccharide fractions (DDP1-1, DDP2-1 and DDP3-1) were successfully purified from the crude polysaccharide of Dendrobium denneanum by DEAE-Cellulose and Sephadex G-200 column chromatography. The average molecular weights (Mws) of these fractions were 51.5, 26.1 and 6.95 kDa, respectively. Monosaccharide components analysis indicated that DDP1-1 and DDP2-1 were composed of arabinose, xylose, mannose, glucose and galactose in a molar ratio of 1.00:2.82:57.11:140.82:7.76 and 1.00:1.62:1.18:77.5:7.79. DDP3-1 was composed of arabinose, mannose, glucose and galactose in a molar ratio of 1.00:1.03:8.84:2.00. On the basis of antioxidant test in vitro, DDP2-1 exhibited the highest antioxidant ability among these samples.  相似文献

The free monosaccharide content of C. lipolytica (strain 4 124) cells grown on n-hexadecane was identified and found to be only glucose. The chromatographic analysis of the hydrolysate of intracellular cell wall polysaccharides indicated the presence of glucose: mannose: galactose: xylose in a ratio of 1 : 1.32 : 1.07 : 0.35. Paper and dise electrophoresis of extracellular polysaccharid from the culture broth was found to be heterogeneous. Ethanol fractionation separated it to a major component F (I) 81.99% and a minor one F (II) 13.04%. Analysis of the major fraction showed that it consisted of galactose and mannose only while the minor polysaccharide consisted of galactose, glucose and mannose. Thus it was concluded that the predominant sugar in both extracellular and intracellular polysaccharides was mannose. Xylose was detected in the intracellular polysaccharide only.  相似文献

The shiitake polysaccharides were obtained from shiitake mushroom. Four fractions were isolated from the polysaccharides using a Sephadex G-100 gel column. Chemical components of the two main fractions were determined by thin layer chromatography (TLC), and high performance liquid chromatography (HPLC). F1 was composed of rhamnose, glucose, and mannose. F3 was composed of xylose, mannose, arabinose and galactose. The obtained results still showed that administration of shiitake polysaccharides could improve muscle's comfortability of animals under a long period of vibration. The above findings might be applicable to studies of vibration ergonomics.  相似文献

提取纯化绣球菌多糖(Sparassis latifolia polysaccharides,SCPs),研究其表征和功能活性,探索绣球菌多糖表征与其抗氧化及免疫活性之间的关系。以绣球菌子实体为原料,采用聚能超声波辅助水提醇沉法提取绣球菌多糖,经DEAE-52、SephadexG-100纯化,用高效凝胶渗透色谱法、离子色谱法、傅里叶红外色谱法、扫描电镜、原子力显微镜对绣球菌多糖进行初步表征,检测绣球菌多糖清除DPPH、·OH、O2^-·自由基能力以及总还原力,用MTT法检测绣球菌多糖对巨噬细胞RAW264.7增殖的影响。结果表明,SCPs分子量范围为215Da–393kDa,由葡萄糖、甘露糖、半乳糖、木糖、果糖构成,摩尔比13:4:1:2:3,其表观形貌为簇状堆积,交织,结构规律性不强,表面光滑,呈一定的网络状结构,分子呈现链状构象,具有高度的分支结构,链间形成小环且伴随一定的球形颗粒。SCPs具有一定的还原能力和清除DPPH、·OH、O2^-·自由基的能力,且能够促进巨噬细胞RAW264.7的增殖。绣球菌多糖的抗氧化及免疫活性可能与其分子量、单糖组成、糖链分支及分子构象有关。  相似文献

The present study was designed to investigate the effect of Angelica sinensis polysaccharides (ASP) and Tai Chi exercise on free radical generation and lipid peroxidation and its modulatory effect on immunity activity in middle-aged women subjects at a dosage levels of 125 mg/kg. ASP was characterized by high performance liquid chromatography (HPLC). HPLC analysis showed that ASP was composed of eight kinds of monosaccharides, namely mannose, rhamnose, glucuronic acid, galacturonic acid, glucose, galactose, arabinose and fucose in molar ratios of 1.2:4.5:1:10.5:17.8:37.5:8.7:4.9. Antioxidant activities such as superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx) and reduced glutathione (GSH) and lipid peroxidation levels in both ASP and Tai Chi exercise groups were significantly enhanced or reduced compared to the control group. In addition, immunity activities such as serum vascular cell adhesion molecule-1 (s-VCAM-1), interleukin-1 beta (IL-1β), interleukin-6 (IL-6) and tumor necrosis factor (TNF-α) levels in both ASP and Ta Chi exercise groups were significantly reduced compared to the control group. It can be concluded that ASP intake and Tai Chi exercise decrease oxidant stress induced injury and improve immunity activities in middle-aged women subjects, indicating a obvious healthy promotion effect.  相似文献

采用高效液相色谱法,测定金钱菇多糖的单糖组成.用超声辅助提取金钱菇多糖,通过1-苯基-3-甲基-5-吡唑酮(PMP)衍生水解后的单糖,高效液相色谱法检测衍生物.结果表明:金钱菇多糖由甘露糖(Man)、核糖(Rib)、鼠李糖(Rha)、葡萄糖(Glc)、半乳糖(Gal)、木糖(Xyl)组成,其摩尔为1.00∶0.90∶0.91∶28.03∶1.58∶0.11.该方法快速、简便、重现性好,可用于测定金钱菇多糖的单糖组成.  相似文献

Culture filtrates of four basidiomycete fungi, Stereum strigoso-zonatum, Fomes australis, Trametes lilacinogilva and Polyporus tumulosus were fractionated and examined for polysaccharide content. Acid hydrolysis showed the presence of galactose, mannose, xylose, fucose and glucose. Their relative amounts were estimated by gas chromatography of the corresponding alditol acetates. Galactose and mannose were the major constituent sugars, amounting to more than 50% of the total. One of the polysaccharides, a fucogalactomannan elaborated by P. tumulosus, was isolated in a purified form. It was shown to have [alpha]D +42 degrees and contained galactose, mannose, fucose and xylose in the relative proportions 2 : 1 : 1 : 0-2.  相似文献
