Several bacterial species express surface proteins with affinity for the constant region (Fc) of immunoglobulin (Ig) of different animal species. Previous studies from our group have reported the presence of an IgG-binding protein in various serotypes of Streptococcus suis . This molecule was also shown to bind in a non-immune fashion chicken IgY and to our knowledge this characteristic is unique. In the present study, by dot-blotting, we showed that the native protein, obtained by affinity chromatography, reacted more strongly with IgG from various animal species than the denatured material. Using a competitive enzyme-linked immunosorbent assay the affinity of the native 60-kDa protein (previously identified as a 52-kDa protein) towards IgG of various animal species was compared to pig IgG. Bovine, goat and human IgG were able to compete effectively with pig IgG whereas chicken IgY constituted a poor competitor. Peptide mapping analysis using denatured protein indicated that pig and bovine IgG recognized the same proteolytic fragment whereas chicken IgY did not. The smallest proteolytic fragment that retained the binding activity towards the IgG of the different animal species tested had a molecular mass of approximately 40 kDa. Fragments with M r<40 kDa showed specific binding activities. That is, the smallest fragment binding pig and bovine IgG had a M r of 30 kDa whereas for goat and human IgG a fragment of less than 16 kDa still showed binding activity. Finally, we observed that antisera raised against a heat-shock protein of Pseudomonas aeruginosa reacted with the 60-kDa S. suis protein indicating that the S. suis 60-kDa protein is a member of the 60-kDa hsp family that possesses the characteristic of binding in a non-immune way mammalian IgG and chicken IgY.  相似文献

Cholesterol-dependent cytolysins (CDC) are pore forming toxins. A prototype of the CDC family members is perfringolysin O (PFO), which directly binds to cholesterol rich cell membrane and lyses the cell. However, as an exception of this general observation, Streptococcus intermedius intermedilysin (ILY) requires human CD59 as its receptor in addition to cholesterol when exhibiting hemolytic activity. It was attempted to explain this functional difference based on a conformational variation in the C-terminal domain of the two toxin proteins, particularly a highly conserved undecapeptide termed tryptophan rich motif. Here, we present the crystal structure of suilysin, a CDC toxin from the swine infectious pathogen Streptococcus suis. Like PFO, suilysin does not require a host receptor for hemolytic activity; yet in the suilysin crystal it shares a similar conformation in the tryptophan rich motif with ILY. This observation suggests that current views of structure-function relationship of CDC proteins in membrane association are still far from complete.  相似文献

The Dps-like peroxide resistance protein (Dpr) is an aerotolerance and hydrogen peroxide resistance agent found in the meningitis-associated pathogen Streptococcus suis. Dpr is believed to act by binding free intracellular iron to prevent Fenton chemistry-catalysed formation of toxic hydroxyl radicals. The crystal structure of Dpr has been determined to 1.95 A resolution. The final model has an Rcyst value of 18.5% (Rfree = 22.4%) and consists of 12 identical monomers (each of them comprising a four alpha-helix bundle) that form a hollow sphere obeying 23 symmetry. Structural features show that Dpr belongs to the Dps family of bacterial proteins. Twelve putative ferroxidase centers, each formed at the interface of neighboring monomer pairs, were identified in the Dpr structure with structural similarities to those found in other Dps family members. Dpr was crystallized in the absence of iron, hence no bound iron was found in the structure in contrast to other Dps family members. A novel metal-binding site approximately 6A from the ferroxidase centre was identified and assigned to a bound calcium ion. Two residues from the ferroxidase centre (Asp63 and Asp74) were found to be involved in calcium binding. Structural comparison with other family members revealed that Asp63 and Asp74 adopt different conformation in the Dpr structure. The structure of Dpr presented here shows potential local conformational changes that may occur during iron incorporation. A role for the metal-binding site in iron uptake is proposed.  相似文献

Wall-associated protein antigens of Streptococcus mutans.   总被引:64,自引:0,他引:64  
When heat-killed whole organisms of Streptococcus mutans strain Ingbritt (serotype c) were injected into rabbits, antibodies to at least 12 antigens were detectable by crossed immunoelectrophoresis. In contrast, when rabbits were immunized with organisms which had been subjected to extraction with the detergent sodium dodecyl sulphate (SDS), antibodies to only two protein antigens were found. These two proteins (A and B), while existing in a form apparently closely associated with peptidoglycan, could also be recovered from homogenates of whole organisms after sonication and from culture filtrates. Antigenic material was excreted throughout growth. SDS-polyacrylamide gradient gel electrophoresis showed A to have a molecular weight of 29 000, while B had a molecular weight of 190 000. Antigen B was purified to apparent homogeneity as judged by SDS-polyacrylamide gel electrophoresis and isoelectric focusing. All of six strains of serotype c examined produced antigen B. Strains of serotypes e and f also produce antigenically identical proteins and strains of serotypes d and g produce proteins which cross-reacted with antigen B. Antigen B was specifically precipitated by rabbit antiserum to human heart tissue.  相似文献

Streptococcus suis serotype 2 (SS2) is a porcine and human pathogen with adhesive and invasive properties. The shortage of studies on the pathogenesis mechanisms of SS2 has made it very difficult to control infection with this pathogen. Three novel in vivo -induced antigens were identified using serological proteome analysis of the SS2 cell wall-associated proteins – SecA, autolysin and ZnuA – the upregulation of which in vivo relative to in vitro was shown by real-time PCR analysis. The in vivo -induced expression of znuA was also confirmed by the fact that the purified recombinant ZnuA could be recognized not by hyperimmune sera but by convalescent sera. This is, to the best of the authors' knowledge, the first report using an immunoproteomic method to identify in vivo -induced antigens of SS2.  相似文献

Hydrochloride extracts obtained from group B streptococcal strains of different serotypes have proved to be the source of type-nonspecific protein antigens, precipitated with ethanol and studied by gel chromatography and spectrophotometric scanning in ultraviolet rays. Thus, 2 or 3 antigens, one of them found to be common for streptococci of groups A, B and G, as well as the admixture of group-specific polysaccharide, have been detected. In extracts obtained from group B streptococcal strains of different serotypes a common protein antigen, specific only for group B, has been detected. The suitability of gel chromatography with the use Toyopearl gel HW-55F for the preparative isolation of the specific fraction of protein type-nonspecific antigen with a view to the subsequent study of immune response to group B streptococci has been shown.  相似文献

Recent epidemiological studies on Streptococcus suis infections in pigs indicated that, besides serotypes 1, 2 and 9, serotype 7 is also frequently associated with diseased animals. For the latter serotype, however, no rapid and sensitive diagnostic methods are available. This hampers prevention and control programs. Here, we describe the development of a type-specific PCR test for the rapid and sensitive detection of S. suis serotype 7. The test is based on DNA sequences of capsular (cps) genes specific for serotype 7. These sequences were identified by cross-hybridization of several individual cps genes with the chromosomal DNAs of 35 different S. suis serotypes.  相似文献

Novel vaccine strategies with protein antigens of Streptococcus pneumoniae   总被引:5,自引:0,他引:5  
Infections caused by Streptococcus pneumoniae (pneumococcus) are a major cause of mortality throughout the world. This organism is primarily a commensal in the upper respiratory tract of humans, but can cause pneumonia in high-risk persons and disseminate from the lungs by invasion of the bloodstream. Currently, prevention of pneumococcal infections is by immunization with vaccines which contain capsular polysaccharides from the most common serotypes causing invasive disease. However, there are more than 90 antigenically distinct serotypes and there is concern that serotypes not included in the vaccines may become more prevalent in the face of continued use of polysaccharide vaccines. Also, certain high-risk groups have poor immunological responses to some of the polysaccharides in the vaccine formulations. Protein antigens that are conserved across all capsular serotypes would induce more effective and durable humoral immune responses and could potentially protect against all clinically relevant pneumococcal capsular types. This review provides a summary of work on pneumococcal proteins that are being investigated as components for future generations of improved pneumococcal vaccines.  相似文献

Streptococcus suis serotype 2 (SS2) is a zoonotic pathogen that can cause infections in pigs and humans. Bacterial surface proteins are often investigated as potential vaccine candidates and biomarkers of virulence. In this study, a novel method for identifying bacterial surface proteins is presented, which combines immunoproteomic and immunoserologic techniques. Critical to the success of this new method is an improved procedure for generating two-dimensional electrophoresis gel profiles of S. suis proteins. The S. suis surface proteins identified in this study include muramidase-released protein precursor (MRP) and an ABC transporter protein, while MRP is thought to be one of the main virulence factors in SS2 located on the bacterial surface. Herein, we demonstrate that the ABC transporter protein can bind to HEp-2 cells, which strongly suggests that this protein is located on the bacterial cell surface and may be involved in pathogenesis. An immunofluorescence assay confirmed that the ABC transporter is localized to the bacterial outer surface. This new method may prove to be a useful tool for identifying surface proteins, and aid in the development of new vaccine subunits and disease diagnostics.  相似文献

Three thermosensitive (Ts) suicide vectors, pSET4s, pSET5s, and pSET6s, have been constructed for gene replacement in Streptococcus suis. Each vector contains an antibiotic-resistance gene (spc or cat), a Ts replication origin of pWV01 lineage, multiple cloning sites, lacZ', and the ColE1 replication origin of pUC19. These vectors could be propagated at 37 degrees C in Escherichia coli, but their replication was blocked above 37 degrees C in S. suis. Moreover, the thermosensitivity of the replication origin was confirmed in S. equi ssp. equi, S. equi ssp. zooepidemicus, and S. dysgalactiae by using pSET4s. For inactivation of the sly gene, which encodes a thiol-activated hemolysin of S. suis, pSLYK, in which the sly gene was interrupted by the cat gene, was constructed using pSET4s and introduced into S. suis DAT2. After growth at the nonpermissive temperature under the antibiotic pressure, the chromosomal sly gene was replaced with the sly::cat gene of pSLYK by a double-crossover event at a rate of 2.6% among chloramphenicol-resistant cells. Moreover, complementation of the sly gene by use of the previously reported S. suis-E. coli shuttle vector pSET2 was demonstrated. These results indicate that the Ts suicide vectors described here will facilitate the genetic analysis of S. suis and other streptococci of veterinary importance by means of allelic exchange of the genes of interest via homologous recombination.  相似文献

Streptococcus suis serotype 2 ( S. suis 2 or SS2) is the causative agent of several diseases in both pigs and humans. 6-Phosphogluconate dehydrogenase (6PGD) is a cell surface protein in SS2. In this study, the immunogenicity and protective efficacy of recombinant 6PGD (r6PGD) from SS2 was evaluated in piglets. Immunization with an r6PGD-containing adjuvant induced a vigorous immunoglobulin G (IgG) response, with the titer of IgG2 being higher than that of IgG1. Immunization of piglets with r6PGD yielded 50% survival upon an intravenous challenge with a lethal dose of SS2. Piglets immunized with the r6PDG vaccine were better protected than those immunized with the adjuvant control. The clinical signs and histopathological changes in the piglets were recorded. Collectively, these results suggest that r6PGD can confer partial protection against SS2 infection and could be useful for the development of subunit vaccines against SS2.  相似文献

Outbreaks in humans, caused by Streptococcus suis serotype 2 (SS2), were reported in 1998 and 2005 in China. However, the mechanism of SS2-associated infection remains unclear. For the first time, a 2-D gel approach combined with MS was used to establish a comprehensive 2-D reference map for aiding our understanding of the pathogenicity of SS2. The identification of 694 out of 834 processed spots revealed 373 proteins. Most of the identified proteins were located in the cytoplasm and were involved in energy metabolism, protein synthesis, and cellular processes. Proteins that were abundant in the 2-DE gels could be linked mainly to housekeeping functions in carbohydrate metabolism, protein quality control and translation. 2-DE of secretory proteins was performed using IPG strips of pH 4-7. Among the 102 protein spots processed, 87 spots representing 77 proteins were successfully identified. Some virulence-associated proteins of SS2 were found, including arginine deiminase, ornithine carbamoyl-transferase, carbamate kinase, muramidase-released protein precursor, extracellular factor, and suilysin. Enolase and endopeptidase have been proposed as putative virulence-associated factors in this study. The 2-D reference map might provide a powerful tool for analyzing the virulence factor and the regulatory network involved in the pathogenicity of this microorganism.  相似文献

Suilysin is a cholesterol-binding cytolysin encoded by sly in Streptococcus suis. DNA sequence determination of the sly locus in a strain lacking sly revealed the presence of another gene, designated orf102, in the place of sly. No transposable element or long-repeat sequence was found in the close vicinity. Except for six strains whose corresponding loci have been rearranged, all of the remaining 62 strains examined had either sly or orf102 at the same locus and their flanking regions were conserved. The genetic organizations having either sly or orf102 were found in the strains whose 16S rRNA sequences were identical. These results suggest that S. suis acquired sly or orf102 from a foreign source and that these genes subsequently spread among S. suis strains by homologous recombination.  相似文献

用DTT、H2O2、DEPC、EDC、NAI、NBS、PCMB、2,3Diacetyl和SA 等9种化学修饰剂,处理猪链球菌2型江苏分离株提纯的溶血素,研究其分子中氨基酸侧链基团与其溶血活性的关系。结果表明,巯基、氨基、羧基和酪氨酸残基等与其溶血活性无关,而色氨酸、组氨酸和精氨酸的化学修饰引起溶血活性的大幅度下降,二硫键的化学修饰引起溶血活性的大幅度增强。显示色氨酸、组氨酸和精氨酸残基是该溶血素的活性必需基团,二硫键的断裂可引起其活性增强。  相似文献

The characterization of 61 Streptococcus suis strains isolated from Chinese slaughter pigs was investigated. S. suis serotypes 1, 2, 3, 4, 7, 9, 10, 11, 12, 13, 14, 15, 16, 22, 23, 25, 26, 28, 29, and 1/2 were found in the isolates by serum agglutination. Of all the prevalent serotypes, S. suis serotype 7 is the most predominant circulating in Chinese slaughter pigs. The virulence-associated genes profile and multilocus sequence typing scheme of the isolates were analyzed. The mrp-/epf-/sly- virulence-associated genes type is the most prevalent in the isolates from slaughter pigs. It is the first time to find S. suis serotypes 7 and 9 isolates with epf. The serotypes 7 and 9 isolates with mrp and/or epf genes did not express MRP and/or EF in the present research. Thirteen new ST types were identified for the first time. ST1 complex and ST27 complex of S. suis are prevalent in China. This paper supplied information to understand the characteristics, such as capsular serotypes, virulence factors, and gene backgrounds of S. suis carried by slaughter pigs.  相似文献
