Preirradiation of mouse recipients with a dose of 1-2 Gy 24 and 48 h before lethal irradiation (8 Gy) made CFUs content of femur increase upon transplantation of bone marrow from exposed and intact donors. The same was with the long-term bone marrow culture: preirradiation of a stromal sublayer increased the number of CFUs in the transplanted bone marrow preirradiated with 6 Gy radiation. Retransplantation of bone marrow to irradiated donors after 5 day cultivation, a sublayer being activated, increased the number of CFUs in the femur in comparison with donors which were injected with the bone marrow from the culture without activation of the sublayer by low-level radiation.  相似文献

In studying the influence of thymocytes fractionated by their size in the ficoll density gradient on the CFUs content of the irradiated mouse bone marrow, two subpopulations of T-cells were isolated: the administration of the first thymocyte subpopulation decreased the CFUs content during the postirradiation recovery period while thymocytes of the second subpopulation increased the content of CFUs in the bone marrow. When thymocytes of mice exposed to low-level radiation were separated a considerable stimulatory effect was produced by certain thymus cell fractions on the number of CFUs in the bone marrow of exposed recipients; no inhibitory effect was registered.  相似文献

Intraperitoneal administration of a spleen extract from Testudo horsfieldi and its U-2 fraction increases the number of endogenous splenic haemopoietic colonies. The U-2 fraction administered to irradiated (4 Gy) mice increases the number of bone marrow CFUs. Bone marrow cells of exposed (4 Gy) mice preincubated in vitro with the U-2 fraction also increase the number of exogenous colonies in the recipient's spleen.  相似文献

Bone marrow cells were separated according to buoyant density, velocity sedimentation and cell surface charge. Fractionated (C3H x AKR)F1 bone marrow cells were transplanted into lethally-irradiated C3H recipients. In all fractions, the CFUs content and the capacity to restore the thymus cell population were determined. For all the physical parameters tested, the thymocyte progenitor cells show the same distribution as CFUs. The relationship between number of thymocyte progenitor cells and number of CFUs is dependent on density. Bone marrow progenitors of PHA responsive cells are of low buoyant density and show a distribution which resembles the distribution of the progenitors of Thy 1 positive cells. After transplantation of large numbers of bone marrow cells into irradiated mice, no significant change in the CFUs content of the thymus was observed.  相似文献

The influence of a tumor-necrotic factor (TNF) on the CFUs population has been studied normally and after irradiation. An inhibitory effect on the pool of the seven-day and doubling of the yield of the eleven-day colonies have been observed in mice received TNF 20 h before bone marrow removal as compared with the controls. The kinetics of restoration of bone marrow cellularity and CFUs number in mouse donors treated with TNF 20 h before irradiation (5.0 Gy) has demonstrated the stimulatory effect of the agent on both indices.  相似文献

We examined the effects of 5 soluble derivatives of yeast glucan on the formation of exogenous (CFU-S) and endogenous (E-CFU) colony-forming units in the spleens of sublethally irradiated (60Co, 6.5–7.0 Gy) mice of two inbred strains. For the estimation of CFU-S, glucans were administered intravenously either to donors or recipients of spleen cells 24 h prior to irradiation or removal of the spleen. The number of CFU-S was increased when both the donors and recipients were treated with glucan; the highest increase was obtained with glucans S, P and K. All glucan preparations increased significantly also the number of E-CFU even when administered 90 min after irradiation. There exist differences in the response to the stimulatory effect of glucans among individual mouse strains. Thus, for example, the stimulatory effect of glucan KM on the E-CFU number was significantly more pronounced in strain A/Ph than in strain C57B1/6.  相似文献

The data are presented on the stimulatory effect of interleukin-2 on the formation of spleen exocolonies from bone marrow irradiated in vitro with doses of 0.5 to 2.5 Gy. The results of 3 sets of experiments on the effects of 22 μT sinusoidal 50 Hz magnetic fields (MF), applied for 1 h on 5 successive days (1 h/5 days), on the level of host defense and on spleen colony formation are reported. The first set of experiments shows the effects on the number of colony‐forming units (CFUs) on the spleen and on the cellularity of the thymus in mice. The MF exposures resulted in an increase in CFUs which was statistically significant with respect to the controls, but not with respect to the shams. Statistically significant changes in the thymic weight and thymic index with respect to both the controls and the shams were measured 1 h after the last MF exposure. In the second set of experiments, the mice were given a sublethal dose of X‐rays (6 Gy), which was followed by exposure 2 h later to the MF. The MF exposure was repeated at the same time of day for 5 days. The number of colonies per spleen showed a consistent, statistically significant increase with MF exposure and the number of CFUs per femur was decreased. In the third set of experiments, bone marrow was taken from mice which had been exposed to 22 μT fields and injected into mice which had been exposed to a lethal dose of X‐rays (9 Gy). The number of CFUs per femur in the recipient mice was shown to be reduced by a statistically significant amount at 1 and 4 days after injection. The rate and the degree of recovery of committed precursors of granulocytes and monocytes (CFUc) following long-term fractionated irradiation were a function of a cumulative radiation dose. In rats exposed to doses of 9.7 and 19.4 Gy the number of CFUc of myelokaryocytes and granulocytes of blood reached the control values after 1-3 months. The increase in CFUc of animals exposed to a dose of 29.1 Gy was transient and did not provide the recovery of granulocytopoiesis.  相似文献

The influence of cystamine delivered in a radioprotective dose before and after irradiation of mouse-recipients (8 Gy) on the effectiveness of exogenous bone marrow cloning has been investigated. Cystamine administered prior to irradiation exerts a protective effect on CFUs and also causes an increase in the number of splenic colonies grown from CFUs of the transplanted bone marrow. With cystamine administered after irradiation the protective effect is absent, but the CFUs number in the femur increases in recipients transplanted with intact bone marrow in comparison with those transplanted without cystamine. It is believed, that in addition to the specific protective mechanism of action of radioprotectors, there is a nonspecific mechanism of increasing the proliferation of protected stem cells that is connected with the stimulatory effect of radioprotective agents on the haemopoietic stroma elements.  相似文献

Regulation of the proliferation of transplanted colony forming units (CFUs) was investigated in lethally irradiated mice, pretreated by methods known to accelerate hemopoietic recovery after sublethal irradiation. Prospective recipients were exposed to either hypoxia, vinblastine or priming irradiation and at different intervals thereafter lethally irradiated and transplanted with bone marrow. Repopulation of CFUs was determined by counting the number of splenic colonies in primary recipients or by retransplantation. Regeneration of grafted CFUs was greatly accelerated and their self-renewal capacity increased in mice grafted within two days after hypoxia. Also the number of splenic colonies formed by grafted syngeneic CFUs as well as by C57BL parent CFUs growing in BC3F1 hosts was significantly increased. The effect was not dependent on the seeding efficiency of CFUs and apparently resulted from hypoxia induced changes in the hosts physiological environment. Proliferative capacity of grafted CFUs increased remarkably in hosts receiving vinblastine two or four days prior to irradiation. Priming irradiation given six days before main irradiation accelerated, given two days before impaired regeneration of CFUs. The increased rate of regeneration was not related to the cellularity of hemopoietic organs at the time of transplantation. The growth of CFUs in diffusion chambers implanted into posthypoxic mice was only slightly improved which does indicate that the accelerated regeneration of CFUs in posthypoxic mice is mainly due to the changes in the hemopoietic microenvironment. A short conditioning of transplanted CFUs by host factor(s) was sufficient to improve regeneration. The results might suggest that the speed of hemopoietic regeneration depends on the number of CFUs being induced to proliferate shordy after irradiation, rather than on the absolute numbers of CFUs available to the organism.  相似文献

The authors have made an attempt to find out the reasons of IL-2 stimulation of spleen colony growth by in vitro (1 Gy) irradiated bone marrow cells. It was shown that the effect of IL on haemopoiesis manifests itself with merely small radiation doses implying that the influence of the preparation makes the process of haemopoietic organ repopulation start at a higher level of cell survival, which is presumably related to a more active repair of radiation-induced CFUs damages: this leads, with other things being equal (e.g. proliferation rate and f factor), to a higher yield of colonies than it is observed with the recipients protected with the exposed bone marrow only.  相似文献

A human recombinant granulocytic-and-macrophagic colony-stimulating factor (rGM-CSF) administered repeatedly to irradiated (10 Gy) CBA mice increased CFUs and CFU-GM content, the number of bone marrow granulocytes and erythronormoblasts, and spleen and peripheral blood cellularity. Prodigiozan injected to long-term cultures of mouse bone marrow 24 h before irradiation increased CFUs and CFU-GM number and colony-stimulating factor (CSF) level by the time of delivery of ionizing radiation. As early as 60 min following irradiation of bone marrow structures with a dose of 2 Gy the number of CFUs and CFU-GM decreased considerably, and from day 3 on after irradiation the indices under study were gradually restored. By day 14 the cultures preinjected with prodigiozan exhibited higher recovery levels. The decrease in the number of precursor cells 60 min after irradiation was accompanied by a drastic increase in the CSF content of cultures; the CSF release in cultures protected with prodigiozan was more moderate than in the irradiated controls.  相似文献

A study was made of adherence to plastic Petri dishes and viability (as non-pyknotic cell counts) of rat bone marrow cells cultured for 5 to 22 h in 199 medium containing 15 per cent fresh isologous serum. An overall decrease in the number of viable myelokaryocytes, including mature neutrophils, was observed in the cultures irradiated with doses of 3 to 12 Gy. In addition, gamma-irradiation increased substantially the adherence of neutrophils and, to a lesser extent, of other myelokaryocytes. A possibility of early radiation-induced disturbances in granulocytic maturation is discussed.  相似文献

重组人血小板生成素(rhTPO)是一种能促进巨核系祖细胞增殖、分化生成血小板的造血因子,研究表明它能促进射线照射小鼠造血功能恢复,前期工作证明rhTPO早期干预可显著提高致死剂量照射小鼠的活存率.本文以7.0Gy照射恒河猴为重度骨髓型急性放射病(ARS)模型,研究了rhTPO早期干预对重症ARS的治疗作用,并与WR2721和"500"的辐射防护作用进行了比较,结果发现rhTPO早期干预可明显促进ARS猴造血功能恢复,改善ARS猴症状,简化对症治疗措施,提高重度骨髓型ARS猴活存率,其对重度骨髓型ARS的防治作用优于现有的辐射防护药WR2721和"500",有望开发成安全有效的新型辐射防治药物.  相似文献

The effect of mouse serum interferon (IF) in vitro and an inducer in vivo on the proliferation of a pluripotent stem cell population with high turnover rate was studied. Proliferation rate was characterized by the number of CFUs in the S phase of the cell cycle. Increased proliferation of bone marrow stem cell populations was produced either by irradiating the donor mice with 3.36 Gy (336 rad) 60Co-gamma rays 7 days before the experiment or by incubating normal bone marrow cells with 10(-11) M concentration of isoproterenol. IF considerably reduced the number of CFUs in S phase in both cases without reducing the CFUs content of the samples. Injection of IF inducer (4 mg/kg poly I:C) into regenerating mice also inhibited the proliferation of CFUs without decreasing the femoral CFUs level. Regeneration kinetics of CFUs from irradiated poly I:C-treated mice ran parallel with that of irradiated untreated animals but showed a characteristic delay corresponding to approximately one CFUs doubling. A transient, non-cytotoxic proliferation inhibitory effect of IF or IF inducer is, therefore, proposed.  相似文献

It was shown that reactivity of the nuclear matrix of thymocytes for antibodies against chromatin of the control and irradiated thymocytes and PDN did not change immediately and increased markedly 2 h following irradiation of rats with a dose of 10 Gy. The method of immunoblotting failed to reveal any qualitative differences in the protein content of the thymocyte nuclear matrix of the control and exposed rats.  相似文献
