目的:构建融合表达GST和结核分枝杆菌16kDa蛋白的表达系统,摸索表达条件及制备方法.方法:根据16kDa蛋白的基因序列设计引物,从结核菌基因组DNA中扩增该基因,酶切后插入表达载体,测序证实后转入大肠杆菌JM109菌株,经IPTG诱导表达,产物经谷胱甘肽-琼脂糖凝胶法纯化.结果:16kDa基因产物大小、酶切片断大小和载体的大小与设计一致、16kDa基因测序的结果与目的基因完全符合.表达纯化的蛋白的大小与蛋白质分子量标准相比较一致.结论:构建的表达载体成功,纯化的蛋白即为目的蛋白.  相似文献

克隆嗜热枯草芽孢杆菌WY-34普鲁兰酶基因并在大肠杆菌中进行表达,对重组酶进行纯化和酶学性质研究,根据枯草芽孢杆菌的普鲁兰酶蛋白序列,设计PCR引物对WY-34的普鲁兰酶基因进行克隆及异源表达.对表达蛋白的最适pH、pH稳定性及最适温度、温度稳定性等特性进行研究,并测定重组普鲁兰酶的底物特异性.将普鲁兰酶基因pluA克隆及分析序列后,发现基因长度为2.2 kb,编码718个氨基酸,在大肠杆菌中异源表达.通过Ni-IDA亲和层析一步纯化得到比活力为93.2 U/mg的纯酶,SDS-PAGE和凝胶层析测定的分子量分别为76.2 kD和74.3 kD.酶学性质研究表明,该酶的最适温度为40℃,在温度不高于45℃条件下稳定;最适pH为6.0,同一温度下pH 6.0-9.0范围内处理30 min可以保持80%以上的酶活力,此酶对普鲁兰糖有很强的底物特异性.此重组普鲁兰酶的酶学性质表明此酶具有一定的工业化应用价值.  相似文献

以产酸性淀粉酶菌株Bacillus sp.CN7的基因组DNA为模板,PCR扩增到α淀粉酶成熟肽基因,将该基因插入表达质粒pSE380中,构建重组质粒pSE380-cn7a。将重组质粒导入到Escherichia coli JM109中,IPTG诱导表达。重组酶经Sephacryl S300、Ni-NTA纯化后测定其酶学性质。重组酶CN7A的最适温度为65°C,最适pH为5.5?6.0,对可溶性淀粉的Km值为3.784g/L,最大反应速度为101.2mg/(L·min),该酶的热稳定性不依赖钙离子。  相似文献

[目的]克隆、表达小麦蓝矮病(WBD)植原体胸苷酸激酶基因(tmk),并分析酶活性,进一步研究胸苷酸激酶在植原体感染宿主及繁殖过程中的功能和作用机理,更好地防治植原体病害.[方法]PCR方法扩增tmk基因并进行序列分析,连接pET30a( )表达载体后原核表达,经Ni-NTA柱层析纯化后进行酶催化活性分析.[结果]首次从小麦蓝矮病(WBD)植原体基因组中分离出胸苷酸激酶基因(tmk),该基因包含tmk-1和tmk-2两种,大小分别为630 bp和624 bp,其编码的氨基酸序列均包含3个与结合NTP/NMP相关的保守功能区.表达的融合蛋白TMK-1活性极低,酶活仅16.4 U/mg,而 TMK-2酶活高达112.41 U/mg,且其最适催化条件为32℃、pH 7.3、1.5 mmol/L Mg2 和 1 mmol/L ATP.[结论]分析了胸苷酸激酶活性中心的一级结构序列及其催化活性随条件变化而改变的性质,为深入研究小麦蓝矮病植原体胸苷酸激酶在侵染寄主及其在宿主体内增殖的转录性质奠定基础.  相似文献

从氧化葡萄糖酸杆菌(Gluconobacter oxydans)的基因组DNA上扩增出木糖醇脱氢酶基因xdh,构建了诱导型表达载体pSE-xdh,导入E.coli JM109后获得了高效表达木糖醇脱氢酶基因的重组菌JM109/pSE-xdh。通过HisTrap HP亲和层析和SephacrylS 300分子筛两步纯化从细胞中得到纯酶,并对酶学性质进行研究。XDH最适还原反应的pH值为5.0,最适还原反应的温度为35℃;最适氧化反应的pH值为11.0,最适氧化反应的温度为30℃。重组菌中的XDH依赖NADH,对NADH的米氏常数Km=57.8 mmol/L,最大反应速率Vmax=1209.1 mmol/(ml·min)。重组菌的XDH酶活力为13.9 U/mg。利用重组菌和原始菌混合静止细胞转化D 木酮糖,16 h 28.0 g/L D木酮糖生成16.7 g/L木糖醇,而原始菌单独转化只生成8.3 g/L木糖醇。  相似文献

为了研究P.pastoris Mpr1的生理特性,在E.coli JM109胞内中成功表达来源于P.pastoris GS115的Mpr1酶,并利用响应面分析法对诱导温度、IPTG诱导浓度、起始诱导OD进行发酵优化,酶活达到(610.3±9.5)m U/m L。酶学性质显示,Mpr1酶的最适反应p H范围为7.0-7.5,最适反应温度是30℃。通过分析重组表达Mpr1菌株和对照菌株的培养过程,显示重组菌株的生长能力显著增强,原因是Mpr1降低了细胞内的ROS水平。  相似文献

根据Gen Bank数据库中已报道的灰色链霉菌(Streptomyces griseus)全基因组序列,分析得到假定的乙酰木聚糖酯酶基因序列并设计引物;利用分子克隆的方法得到该菌株基因组中乙酰木聚糖酯酶基因,并构建原核表达载体p ET28a-Sgraxe,经IPTG诱导表达重组Sgr Axe,Ni-NTA亲和层析法纯化该蛋白。结果显示,克隆得到乙酰木聚糖酯酶基因axe,其序列全长1 008 bp,编码336个氨基酸。SDS-PAGE检测带有p ET28a-Sgraxe转化菌株诱导表达产物相对分子量约为37 k D,与理论值相符。纯化的重组Sgr Axe酶学性质表明,该酶最适反应温度为50℃,最适p H8.0,热稳定性较强,p H作用范围广;金属离子对酶均表现为抑制作用,尤其是Zn2+严重抑制酶活力;重组酶特征的分析揭示了其在工业中潜在的应用价值。  相似文献

以北京红篓菌基因组DNA为供体,pKC505 cosmid质粒为载体系统,λ噬菌体体外包装重组质粒并转染感受态细胞E.coli JM109,构建了该菌株基因组DNA文库。以聚羟基烷酸(PHA)合成酶基因通用简并引物扩增的PHA合成酶部分基因为探针,经DIG标记后利用菌落原位杂交技术对文库进行筛选,获得3个阳性克隆菌株,PCR检测证明这3个克隆具有PHA合成酶基因片段,酶活测定表明3个克隆都具有PHA合成酶及与PHA合成相关的乙酰乙酰辅酶A还原酶的活性。  相似文献

[目的] 从芽胞杆菌(Bacillus sp.)YX-1基因组中克隆出一种有机溶剂耐受型的葡萄糖脱氢酶基因,实现了该基因在大肠杆菌中的高效表达,研究了重组蛋白的酶学性质.[方法] 依据芽胞杆菌属中葡萄糖脱氢酶氨基酸序列的保守性,设计合理引物,钓取来源于Bacillus sp.YX-1的葡萄糖脱氢酶基因,构建诱导型表达载体pET28a-gdh,于大肠杆菌中进行表达.镍柱亲和层析法纯化重组蛋白,考察了重组蛋白的酶学性质.[结果] 葡萄糖脱氢酶基因全长为786 bp,编码261个氨基酸.酶学研究结果表明:该酶最适反应温度为45℃,最适pH值为8.0;具有良好的有机溶剂耐受性,于50%的辛烷、环己烷、癸烷中室温放置1h后,酶活仍能保持90%以上;具有较宽的底物谱,对多种糖均具有一定的催化活性,其中催化D-葡萄糖的活力最高,产生还原型辅酶因子;对辅酶NADH和NADPH具有相似的依赖性,对NAD+和NADP+的催化比活分别为8.37 U/mg和8.62 U/mg.[结论]利用生物信息学成功地挖掘出Bacillus sp.YX-1一种耐有机溶剂的葡萄糖脱氢酶,为氧化还原酶在有机相反应中的的辅酶再生循环提供了新型的生物催化剂.  相似文献

以弗氏柠檬酸菌(Citrobacter freundii)基因组DNA为模板,通过PCR得到甘油脱水酶(glycerol dehydratase)基因dhaB、dhaC、dhaE,克隆到表达载体pSE380上,得到重组质粒pSn-dhaBCE。将此重组质粒转化到E.coli JM109中,重组菌株SDS-PAGE结果显示有明显的61kD、22kD、16kD三条特异性蛋白条带出现。重组菌株经诱导表达,酶活力为11.59U/mL。  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献

采用原位分子杂交方法检测HCV RNA及HBV X基因;采用免疫组织化学方法研究HCV核心抗原,非结构区C33c抗原及HBxAg在肝细胞肝癌中的定位及分布.结果表明(1)HCV RNA、HBV X基因在肝细胞肝癌组织检出率分别为40%(55/136)和82%(112/136).HCV RNA定位于癌细胞的胞浆内,阳性细胞呈散在、灶状及弥漫分布三种形式;HBV X基因在肝癌细胞中的分布呈胞浆型、核型及核浆型,阳性细胞也呈上述三种分布形式;(2)HCV C33c抗原、核心抗原在肝细胞肝癌中的阳性率为81%(133/164)及86%(141/164).C33c抗原定位于癌细胞及肝细胞的胞浆内;核心抗原既定位于癌细胞核中,又可定位于胞浆中.C33c抗原阳性细胞以灶状分布为主;而核心抗原阳性细  相似文献

For a plant selection model with frequency-independent viabilities, fertilities and selfing rates, it is shown that apart from global fixation, for certain parameter combinations a protected polymorphism and facultative fixation (either allele may become fixed according to initial frequencies) may both occur. Facultative fixation requires different selling rates for the dominant and recessive type. Protection of the polymorphism requires resource allocation for male and female function. In this connection the problem of purely genetically caused population extinction is discussed.
For general frequency dependence and regular segregation, the chances for establishment of a completely recessive gene are compared to those of a completely dominant gene. It is proven that the process of establishment of the recessive gene, despite a fitness advantage, may be considerably endangered by drift effects if random mating prevails. The recessive gene may reach the same effectivity in establishment as a dominant gene, only if the recessive homozygote mates exclusively with its own type during the period of establishment.  相似文献
