Genetic-parameter estimates and parental breeding-value predictions were compared from open-pollinated and control-pollinated progeny populations of Eucalyptus globulus and two populations of E. nitens. For E. globulus there were two types of open-pollinated populations (native stand open-pollinated and seed orchard open-pollinated) and two types of control-pollinated populations (intra-provenance and interprovenance full-sib families). For E. nitens there were two populations, a seed orchard open-pollinated population and intra-provenance full-sib families. Progeny tests were established across multiple sites and 2-year height and diameter were measured and volume calculated. Genetic parameters from native stand open-pollinated E. globulus were unlike the parameters from the other three E. globulus populations; heritability estimates were severely inflated, presumably due to high levels, and possibly differential levels, of inbreeding depression relative to the other populations. Estimates of dominance variance in the E. globulus full-sib populations were high, but were zero in the E. nitens population. Correlations among parental breeding values, predicted using data from the different populations, were generally low and non-significant, with two exceptions: predictions from the two E. globulus full-sib populations were significantly correlated (r=0.54, P = 0.001), as were predictions from the E. nitens seed orchard OP and full-sib population (r = 0.61, P = 0.08). There was some indication that superior parents of E. globulus native stand open-pollinated families also tended to have above-average breeding values based on the performance of intra-provenance full-sib offspring. The consequences of these results for exploitation of base-population collections from native stands are discussed.  相似文献

Using DNA fingerprint markers within species and populations of wild plants requires information on the relationship between fingerprint similarity and relatedness. We identified a hypervariable marker based on oliog(GATA)4-hybridization of DpnII-cut genomic DNA from Sea Lavender (Limonium carolinianum). Banding patterns were somatically stable and highly variable among unrelated individuals. Band molecular-weight sizing errors (as a percent of band molecular weight) were estimated at 0.44%±0.003 within gels and 0.76%±0.964 between gels. Band sizing errors defined a 99% confidence bin of ±0.95% (1.90% total) of molecular weight. Band-sharing estimates were based on this bin size and on variance estimates that compensate for non-independent comparisons. Band-sharing among nine unrelated individuals () was 0.198±0.O11. Experimental pollinations designed to produce selfed, fulland half-sib progeny groups led to five selfed progeny groups and no outcrossed progeny (mean band-sharing, ovS=0.468±0.074). A linear regression between band-sharing (S) and relatedness (r) assuming 17% inbreeding was r=0.006+0.914*S (R2=0.973) and established the maximum amount of inbreeding. ovS(0.392±0.022) estimated from wild pollinated seeds from four maternal families was intermediate to unrelated individuals and experimental selfed progeny, giving evidence for mixed mating in wild plants. More extensive plant pedigrees with known levels of inbreeding will be needed to measure variation in the relationship between S and r among populations and families.  相似文献

We present an approach to infer the number of pollen donors directly from genotype data of open- pollinated progeny of Quercus robur (pedunculate oak), a highly outcrossing tree species. The approach is based on closely linked, highly polymorphic codominant microsatellite markers. Initially the close linkages between three previously mapped microsatellite loci were confirmed by studies of linkage disequilibrium (LD). Then an approach to track the pollen donors contributing to maternal half-sib families (open-pollinated families) was developed by analysing haplotype arrays of closely linked microsatellite markers transmitted from the fathers to the progeny. Simulated data of five linked microsatellite loci segregating in eight open-pollinated families were used to study the relationship between the number of paternal chromosomes detected by this ”haplotype approach” and the number of diploid fathers contributing to the families. The results showed that the number of diploid pollen donors can be expressed as an exponential function of the number of paternal chromosomes inferred from the progeny. The 95% confidence interval of this regression function is used to determine the minimum number of fathers contributing to a genotyped open-pollinated family of Quercus robur. Finally this open-pollinated family is used to demonstrate the resolution obtained with the ”haplotype approach”. Six independent microsatellite loci were used to study relatedness among all pairs of pollen gametes that share a haplotype of three linked markers. The results suggest that the majority of such gametes are identical by descent from the same father. The ”haplotype approach” presented here can be used to monitor the number of contributing pollen donors in commercial seedlot samples from oak or any other outcrossing tree species for which closely linked, highly polymorphic, codominant genetic markers are available. The management of a genetic improvement program is based on the knowledge of the genetic parameters and their relationships to determine the genetic gains. Knowledge of the coefficient of coancestry (θ) is a requirement for efficient progeny testing scheme and for estimating additive variance components for any quantitative trait. When using open-pollinated families, most authors assume that the seedlings are related as half-sibs, but this is not always true. Our aim was to estimate a mean value of the coancestry coefficient of the families present in a maritime pine Pinus pinaster Ait. (maritime or cluster pine) progeny trial originating from seed collected in a clonal seed orchard and to study how deviations from the standard assumption of θ = 0.125 affect heritability estimations. Five highly polymorphic microsatellite markers were scored in 125 offspring from a subsample of five families from the progeny trial. The mean value of the coancestry coefficient of the families present in this progeny trial was 0.130. Differences between the unadjusted and adjusted heritability estimates were more pronounced in wood density (0.609 and 0.586, respectively) than in diameter (0.166 and 0.154, respectively). We conclude that in the trial, the associated error in heritability estimates due to the inclusion of full-sibs, when assuming a standard coefficient of relationship among open-pollinated sibs of 0.250, was low and that this result is robust with respect to the number of families sampled, given unbiased estimates of average relationship among offspring within sib families.  相似文献

Summary Precision measurement is an essential part of heritability estimate interpretation. Approximate standard errors are commonly used as measures of precision for heritability on a progeny mean basis (H). Their derivation, however, is not inferred from the distribution theory for H. F-distribution based exact confidence intervals have been derived for some onefactor mating design H estimators. Extension of the confidence interval results from one-factor to twofactor mating designs is reported in this paper. Functions of heritability on a full-sib or half-sib progeny mean basis from nested or factorial mating design parameters were distributed according to the F-distribution. Exact confidence intervals were derived for heritability on a full-sib progeny mean basis. Exact confidence intervals for heritability on a half-sib progeny basis were adapted from previous results. Maize (Zea mays L.) data were used to estimate confidence intervals. Statistical analyses of trees produced from wind-pollinated seeds to determine levels of additive variance need to be adjusted to compensate for the relationships among the progeny. The assumption that the coefficient of relationship among the progeny is 0.25 will lead to inaccurate estimation of the additive variance if the relationships among the progeny are not entirely half-sib. Foreign pollen intrusion into the orchard pollen cloud, selfing, and reproductive phenology variation (i.e., mating proportion differences) among the parent trees may lead to varying proportions of self, self-half, half-sib, and full-sib relationships among the progeny. The variance of the coefficient of relationship from 0.25 for two coniferous seed orchards subjected to simulated selfing and foreign pollen problems illustrate the need for caaution when estimation techniques are employed.Communicated by A. Linkage analysis is commonly used to find marker-trait associations within the full-sib families of forest tree and other species. Study of marker-trait associations at the population level is termed linkage-disequilibrium (LD) mapping. A female-tester design comprising 200 full-sib families generated by crossing 40 pollen parents with five female parents was used to assess the relationship between the marker-allele frequency classes obtained from parental genotypes at SSR marker loci and the full-sib family performance (average predicted breeding value of two parents) in radiata pine (Pinus radiata D. Don). For alleles (at a marker locus) that showed significant association, the copy number of that allele in the parents was significantly correlated, either positively or negatively, with the full-sib family performance for various economic traits. Regression of parental breeding value on its genotype at marker loci revealed that most of the markers that showed significant association with full-sib family performance were not significantly associated with the parental breeding values. This suggests that over-representation of the female parents in our sample of 200 full-sib families could have biased the process of detecting marker-trait associations. The evidence for the existence of marker-trait LD in the population studied is rather weak and would require further testing. The exact test for genotypic disequilibrium between pairs of linked or unlinked marker loci revealed non-significant LD. Observed genotypic frequencies at several marker loci were significantly different from the expected Hardy-Weinberg equilibrium. The possibilities of utilising marker-trait associations for early selection, among-family selection and selecting parents for the next generation of breeding are also discussed.  相似文献

To determine the effect of growing conditions on population parameters in wild radish, (Raphanus sativus L.: Brassicaceae), we replicated maternal and paternal half-sib families of seed across three planting densities in an experimental garden. A nested breeding design performed in the greenhouse produced 1,800 F1 seeds sown in the garden. We recorded survivorship, measured phenotypic correlations among and estimated narrow-sense and broad-sense heritabilities (h2) of: days to germination, days to flowering, petal area, ovule number/flower, pollen production/flower, and modal pollen grain volume. Survivorship declined with increasing density, but the relative abundances of surviving families did not differ significantly among densities. Seeds in high-density plots germinated significantly faster than seeds sown in medium- or low-density plots, but they flowered significantly later. Plants in high-density plots had fewer ovules per flower than those in the other treatments. Petal area and pollen characters did not differ significantly among densities. Densities differed with respect to the number and sign of significant phenotypic correlations. Analyses of variance were conducted to detect additive genetic variance (Va) of each trait in each density. At low density, there were significant paternal effects on flowering time and modal pollen grain volume; in medium-density plots, germination time, flowering time and ovule number exhibited significant paternal effects; in high-density plots, only pollen grain volume differed among paternal sibships. The ability to detect maternal effects on progeny phenotype also depended on density. Narrow-sense h2 estimates differed markedly among density treatments for germination time, flowering time, ovule number and pollen grain volume. Maternal, paternal and error variance components were estimated for each trait and density to examine the sources of variation in narrow-sense h2 across densities. Variance components did not change consistently across densities; each trait behaved differently. To provide qualitative estimates of genetic correlations between characters, correlation coefficients were estimated using paternal family means; these correlations also differed among densities. These results demonstrate that: a) planting density influences the magnitude of maternal and paternal effects on progeny phenotype, and of h2 estimates, b) traits differ with respect to the density in which heritability is greatest, c) density affects the variance components that comprise heritability, but each trait behaves differently, and d) the response to selection on any target trait should result in different correlated responses of other traits, depending on density.  相似文献

Willow is being developed as a crop for biomass plantations in the Northeast and North-central United States, but has only recently been the subject of controlled breeding to generate improved genotypes. Maximizing variability among progeny within full-sib families produced by controlled pollination may increase the probability of producing willow clones exhibiting desirable extreme phenotypes. Yet, predicting combinations of parents yielding highly variable progeny is not currently possible. Controlled pollinations were completed among 15 Salix eriocephala clones and the resulting progeny were vegetatively propagated and planted in a greenhouse progeny test. Heights of rooted cuttings were measured after 4 months of growth. Genetic similarity among parents was estimated based on 77 polymorphic AFLP bands. Strong negative correlation (r = -0.88) was detected between mean female-parent similarity indices and the standard deviation of height among half-sib progeny from those females. Parent combinations that had relatively low similarity indices tended to produce progeny that had greater variability in height. This negative relationship suggests that AFLP fingerprints of S. eriocephala parents may be useful for predicting parent combinations that will yield families with large variability.  相似文献

Summary First generation (S1) progeny, second generation (S2) progeny, and backcross (BC) progeny of a celery (Apium graveolens L. var. dulce) somaclonal variant, UC-T3, were evaluated for resistance to the fungus Fusarium oxysporum f. sp. apii, race 2 (FOA2). Chisquare analysis of S1 progeny showed that the expression of resistance did not fit a single-locus model. S2 progeny means were similar among families, except in a heavily infested field. The lowest ranking S2 family in both the lightly infested and heavily infested fields was significantly more resistant to FOA2 than individuals of the susceptible progenitor line Tall-Utah 527OR; therefore; it was concluded that the trait was heritable. The phenotypic distribution of the backcross progeny was broad, suggesting that the new resistance was conferred by at least two genes whose expression was dominant to susceptibility. The mean scores for disease resistance of the progeny of crosses between UC-T3 and the moderately resistant line, Tall-Utah 527OHK, generally equaled the resistance found among the progeny of the most resistant parent.  相似文献

Summary This study was conducted to assess the genetic basis of the variability observed for the glutamate oxaloacetate transaminase (GOT), Superoxide dismutase (SOD), esterase (EST), and malate dehydrogenase (MDH) isozyme systems in different open-pollinated Vicia faba varieties. Individual plants showing contrasting zymogram patterns were simultaneously selfed and cross-combined. Crossing was unsuccessful in producing progeny, and only selfed progenies were suitable for genetical analysis of isozyme variability. Three zones of GOT activity were made visible. The isozyme of GOT-2 and GOT-3 zones were dimeric and under the control of three alleles at the Got-2 locus and two alleles at the Got-3 locus, respectively. The isozymes of the GOT-1 zone did not show any variability. Three zones of SOD isozyme activity were made visible. The isozymes occurring in the SOD-1 (chloroplastic isozyme form) and SOD-2 (cytosol isozyme form) zones were dimeric and under the control of two alleles at the Sod-1 and Sod-2 loci. The isozyme visualized in the SOD-3 zone (mitochondrial isozyme form) were tetrameric and under the control of two alleles at the Sod-3 locus. Apparently the isozymes made visible in the most anodal esterase zones EST-1, EST-2, and EST-3 were monomeric, and the occurrence of two alleles at each of two different loci explained the variability observed in the EST-2 and EST-3 zones. For MDH, only two five-banded zymogram pattern types were found, and every selfed progeny showed only one of the two zymogram type, indicating that each individual possessed fixed alleles at the loci controlling MDH isozyme. Got-2, Got-3, Sod-1, Sod-2, and Sod-3 appear to be five new isozyme gene markers that can be useful in Vicia faba breeding for linkage study, varietal fingerprinting, outcrossing rate estimate, and indirect selection for quantitative characters.  相似文献

Summary A colchicine-doubled F1 hybrid (2n=118) of a cross between PI 360841 (Glycine max) (2n=40) x PI 378708 (G. tomentella) (2n=78), propagated by shoot cuttings since January 1984, produced approximately 100 F2 seed during October 1988. One-fourth of the F2 plants or their F3 progeny have been analyzed for chromosome number, pollen viability, pubescence tip morphology, seed coat color, and isoenzyme variation. Without exception, all plants evaluated possessed the chromosome number of the G. max parent (2n=40). Most F2 plants demonstrated a high level of fertility, although 2 of 24 plants had low pollen viability and had large numbers of fleshy pods. One F2 plant possessed sharp pubescence tip morphology, whereas all others were blunt-tipped. All evaluated F2 and F3 plants expressed the malate dehydrogenase and diaphorase isoenzyme patterns of the G. max parent and the endopeptidase isoenzyme pattern of the G. tomentella parent. Mobility variants were observed among progeny for the isoenzymes phosphoglucomutase, aconitase, and phosphoglucoisomerase. This study suggests that the G. The mating-system was investigated in a natural population of the tetraploid taxon Acacia nilotica ssp. leiocarpa using open-pollinated seeds from 15 families. Six-day-old germinated seeds were used for starch-gel electrophoresis. Three enzyme systems (ADH, EST-1, and 6PGD) were scored. Isozyme banding patterns and segregation of isozyme variants within families suggest that the species is an autotetraploid displaying tetrasomic inheritance. Estimates of single-locus (ts=0.358) and multilocus (tm=0.384) population outcrossing rates were homogeneous, and indicate substantial selfing in the population. Heterogeneity of outcrossing estimates among loci and families were marked, suggesting departure from the assumptions of the mixed-mating model. Implications of the result for the utilization of germplasm in tree-improvement programmes are noted.  相似文献

Mating system and inbreeding depression in quantitative traits of whitebark pine (Pinus albicaulis Engelm.) was determined using isozymes and a seedling common garden experiment. Simultaneous isozyme analysis of embryo and haploid megagametophyes from progeny arrays of families in three distinct geographic regions (Oregon, Montana, and southern British Columbia) was used to estimate parental and progeny inbreeding coefficients, as well as regional and family mean multilocus outcrossing rates (t m). Quantitative trait family means of seedlings from the same families growing in two temperature treatments in a common garden experiment were regressed on the estimated inbreeding coefficient to determine the presence and magnitude of inbreeding depression. Regional estimates of t m ranged from 0.73 to 0.93, with a mean over all regions of 0.86. Family mean t m values indicated predominant outcrossing; however, some individuals experienced substantial inbreeding. The Oregon region had a significant excess of heterozygotes in the parental generation relative to Hardy–Weinberg equilibrium, while both the Oregon and southern BC regions had a heterozygote deficiency in progeny, suggesting selection against inbred individuals. Biomass in the ambient temperature treatment for the southern BC region was the only trait significantly related to inbreeding coefficient. The mean inbreeding coefficient for this region was 0.25, and based on this relationship, mean predicted biomass would be reduced by 19.6% in this region if inbred individuals are not removed by selection. The estimated outcrossing rate of whitebark pine is slightly lower than most wind-pollinated conifers, and while most individuals are highly outcrossing, some experience substantial inbreeding.  相似文献

Summary Pollen-stigma compatibility relationship were studied in 50 cultivars and more than 800 seedlings of the European hazelnut (Corylus avellana L.). A total of 22 unique S-alleles have been identified. Dominance relationships in 75 of the possible 231 pairs of alleles have been determined in both pistil and pollen. In the pistil, all alleles exhibited independent action, whereas in the pollen, alleles exhibited either dominance or codominance. 以华北落叶松控制授粉群体的全部子代为材料,母本相同的个体视为同一家系,利用18对SSR分子标记对7个家系257个个体进行扩增,分析其遗传多样性及其遗传分化水平。结果表明:(1)18个SSR位点共检测到72个等位基因,平均等位基因数4个,有效等位基因数(Ne)为1.247~3.411。(2)7个家系的平均有效等位基因数为2.135个,观测杂合度(Ho)为0.518,期望杂合度(He)为0.502,Shannon信息指数0.846,其中55号家系遗传多样性水平最高,56号的遗传多样性水平最低。(3)遗传分化系数(GST)为0.113,各家系群体处于中等遗传分化水平,AMOVA分析结果显示82%的遗传变异存在于家系内,18%的遗传变异存在于家系间。(4)聚类分析结果表明,55号与59号家系的遗传距离最近,具有较近的亲缘关系,49号家系与其他家系的遗传距离最远。(5)结合遗传多样性及遗传分化水平结果,估算获得选择核心家系数及核心个体数,选择5个家系均可获得96%以上的遗传多样性,对于个体数较少的家系,选择15~20个个体;对于个体数较多的家系,选择35个个体,即可获得96%以上的遗传多样性。该研究结果对华北落叶松种子园育种群体的选择及其遗传多样性的保护具有重要意义。  相似文献

Summary The expressed activity in pollen and stigma was determined for both S alleles of sixteen S-alíele heterozygous genotypes and for one of the two S alleles of two additional heterozygotes. Activities were measured using pollen tube penetration and seed set data from reciprocal crosses between each S-allele heterozygote and its two corresponding S-allele homozygotes.In pollen the S-allele activities ranged from zero to 100% inhibition of pollen tube penetration and seed set, and in the stigma they ranged from 8 to 100% inhibition. Of the sixty-eight S-allele activities measured, thirty-three (48%) were 90 to 100% inhibition, nine (13%) were 80 to 89% inhibition and one to five were within each ten-unit range below 80% inhibition.In an S-allele heterozygote, each subset of two S alleles had an activity for each allele in both pollen and stigma which was highly repeatable among duplicate pollinations within and among successive years. Each subset of two S alleles had a specific S-allele interaction in the pollen, and the same or another specific interaction in the stigma. In pairings with six other S alleles, allele S 2 had four calculated levels of activity in pollen that ranged from 88 to 94%, and five levels in the stigmas between 15 and 94%. When paired in a heterozygote, alleles S 3 and S 5 had activities ranging between 42 and 59%, representing mutual weakening of S-allele activity. Also, heterozygote S 15 S 3 had pollen activities, respectively, of 25 and 6%, i.e. mutual weakening in the pollen.These results indicate that in heterozygous combination with a series of other S alleles, each S-allele may have activity in pollen and also in stigma that potentially is between zero and 100% inhibition. They further indicate that the defined sexual-organ X S-allele-interaction Types I, II, III and IV are extremes; all intermediate variations including complete weakening of both alleles are possible. Recessiveness is weakening of the activity of but one of the two S alleles. Habitat fragmentation has been shown to disrupt ecosystem processes such as plant-pollinator mutualisms. Consequently, mating patterns in remnant tree populations are expected to shift towards increased inbreeding and reduced pollen diversity, with fitness consequences for future generations. However, mating patterns and phenotypic assessments of open-pollinated progeny have rarely been combined in a single study. Here, we collected seeds from 37 Eucalyptus incrassata trees from contrasting stand densities following recent clearance in a single South Australian population (intact woodland=12.6 trees ha−1; isolated pasture=1.7 trees ha−1; population area=10 km2). 649 progeny from these trees were genotyped at eight microsatellite loci. We estimated genetic diversity, spatial genetic structure, indirect contemporary pollen flow and mating patterns for adults older than the clearance events and open-pollinated progeny sired post-clearance. A proxy of early stage progeny viability was assessed in a common garden experiment. Density had no impact on mating patterns, adult and progeny genetic diversity or progeny growth, but was associated with increased mean pollen dispersal. Weak spatial genetic structure among adults suggests high historical gene flow. We observed preliminary evidence for inbreeding depression related to stress caused by fungal infection, but which was not associated with density. Higher observed heterozygosities in adults compared with progeny may relate to weak selection on progeny and lifetime-accumulated mortality of inbred adults. E. incrassata appears to be resistant to the negative mating pattern and fitness changes expected within fragmented landscapes. This pattern is likely explained by strong outcrossing and regular long-distance pollen flow.  相似文献

The mating system, patterns of pollen mediated gene flow and levels of genetic contamination were investigated in a planted stand of Acacia saligna subsp. saligna via paternity analysis using microsatellite markers. High levels of outcrossing were detected within the stand (t m = 0.98), and the average pollen dispersal distance was 37 m with the majority of progeny sired by paternal trees within a 50-m neighbourhood of the maternal tree. Genetic contamination from the natural background population of A. saligna subsp. lindleyi was detected in 14% of the progeny of A. saligna subsp. saligna and varied among maternal trees. Long distance inter-subspecific pollen dispersal was detected for distances of over 1,500 m. The results provide information for use in the breeding and domestication programme aimed at developing A. saligna as an agroforestry crop for the low rainfall areas of southern Australia.  相似文献

Summary A series of experiments was conducted to determine the inheritance of seed weight in cucumber. Matings between a Cucumis sativus var. sativus (Cs) L. inbred line (USDA WI 1606; P1) and a C. sativus var. hardwickii (Royle) Kitamura (Ch) collection (PI 215589; P2) were made to produce seed of reciprocal F1, F2, and BC1 families. Families were grown under field and greenhouse conditions, and seeds were extracted from fruit 55 to 60 days post-pollination. Seed of F1 and F2 families was obtained using the Cs inbred WI2808 (P12) and the Ch collection LJ 90430 (P10), and seed of F1 families were produced using a North Carolina Design II mating scheme in which three Cs (P3= GY-14; P4=WI 1379; P5=WI 1909) inbreds were used as maternal parents and seven Ch collections (P2; P6= PI462369; P7=486336; P8=LJ91176; P9=273469; P10= 2590430; P11=PI187367) were used as paternal parents. Mean seed weights of F1 progeny reflected the dominance of genes of the C. sativus var. sativus parent. Transformation to number of seeds per unit weight resulted in increased variance homogeneity within generations and a broad-sense heritability ranging between 26% to 56%. Additive and dominance effects were important in the expression of seed weight in P1×P2 progeny produced in the greenhouse and additive effects were important in field grown progeny resulting from P1×P2 and P10×P12 matings. The estimated number of factors or loci involved ranged between 10 to 13, depending on the method of calculation.  相似文献
