A clamped cantilever beam test was developed to determine the fatigue crack propagation rate of the CoCr alloy/PMMA cement interface at high crack tip phase angles. A combination of finite element and experimental methods was used to determine the fatigue crack growth rates of two different CoCr alloy/PMMA cement surfaces. A crack tip phase angle of 69 deg was found, indicating that loading at the crack tip was mixed-mode with a large degree of in-plane shear loading. The energy required to propagate a crack at the interface was much greater for the plasma-sprayed CoCr surface when compared to the PMMA-precoated satin finish (p < 0.001). Both interface surfaces could be modeled using a Paris fatigue crack growth law over crack propagation rates of 10(-4) to 10(-9) m/cycle.  相似文献

Background and Aims

Experimental evidence in the literature suggests that O2•− produced in the elongation zone of roots and leaves by plasma membrane NADPH oxidase activity is required for growth. This study explores whether growth changes along the root tip induced by hyperosmotic treatments in Zea mays are associated with the distribution of apoplastic O2•−.

Methods

Stress treatments were imposed using 150 mm NaCl or 300 mm sorbitol. Root elongation rates and the spatial distribution of growth rates in the root tip were measured. Apoplastic O2•− was determined using nitro blue tetrazolium, and H2O2 was determined using 2′, 7′-dichlorofluorescin.

Key Results

In non-stressed plants, the distribution of accelerating growth and highest O2•− levels coincided along the root tip. Salt and osmotic stress of the same intensity had similar inhibitory effects on root elongation, but O2•− levels increased in sorbitol-treated roots and decreased in NaCl-treated roots.

Conclusions

The lack of association between apoplastic O2•− levels and root growth inhibition under hyper-osmotic stress leads us to hypothesize that under those conditions the role of apoplastic O2•− may be to participate in signalling processes, that convey information on the nature of the substrate that the growing root is exploring.Key words: Root tip growth, Zea mays, salt stress, reactive oxygen species, ROS  相似文献

Aluminium toxicity has been recognized as a primary growth-limiting factor in acid soil, resulting in a decrease in plant growth and production. In this experiment we have studied the induction of oxidative stress and changes in antioxidant gene expression in pea (var. ALASKA) under aluminium (Al) stress. We have found that Al treatment affected the growth of pea plant and induced oxidative stress with a change in antioxidant gene expression profile. While the expression of glutathione-s-transferase (GST) and catalase (CAT) was more in root, cytosolic Ascorbate peroxidase (cAPX) expression increased in shoot under aluminium stress. Copper- Zinc Superoxide dismutase (Cu-Zn SOD) gene expression was higher after 24 h but decreased after 48 h along with elevated expression of manganese superoxide dismutase (MnSOD) and iron-superoxide dismutase (FeSOD) at higher aluminium contentrations after 24 and 48 h. Aluminium stress elevated hydrogen peroxide (H2O2) level and affected the growth. The proline content did not change significantly, whereas glutathione content increased with a decreased ascorbate content under Al stress. The present study indicates that aluminium treatment affected the antioxidant gene expression and induced oxidative stress in pea plant.  相似文献

Background and Aims

Plants growing on serpentine bedrock have to cope with the unique soil chemistry and often also low water-holding capacity. As plant-soil interactions are substantially modified by arbuscular mycorrhizal (AM) symbiosis, we hypothesise that drought tolerance of serpentine plants is enhanced by AM fungi (AMF).

Methods

We conducted a pot experiment combining four levels of drought stress and three AMF inoculation treatments, using serpentine Knautia arvensis (Dipsacaceae) plants as a model.

Results

AMF inoculation improved plant growth and increased phosphorus uptake. The diminishing water supply caused a gradual decrease in plant growth, accompanied by increasing concentrations of drought stress markers (proline, abscisic acid) in root tissues. Mycorrhizal growth dependence and phosphorus uptake benefit increased with drought intensity, and the alleviating effect of AMF on plant drought stress was also indicated by lower proline accumulation.

Conclusions

We documented the role of AM symbiosis in plant drought tolerance under serpentine conditions. However, the potential of AMF to alleviate drought stress was limited beyond a certain threshold, as indicated by a steep decline in mycorrhizal growth dependence and phosphorus uptake benefit and a concomitant rise in proline concentrations in the roots of mycorrhizal plants at the highest drought intensity.  相似文献

Pitting wear is a dominant form of polyethylene surface damage in total knee replacements, and may originate from surface cracks that propagate under repeated tribological contact. In the present study, stress intensity factors, KI and KII, were calculated for a surface crack in a polyethylene-CoCr-bone system in the presence of rolling or sliding contact pressures. Variations in crack length and load location were studied to determine probable crack propagation mechanisms and modes. The crack tip experienced a wide range of mixed-mode conditions that varied as a function of crack length, load location, and sliding friction. Positive KI values were observed for shorter cracks in rolling contact and for all crack lengths when the sliding load moved away from the crack. KII was greatest when the load was directly adjacent to the crack (g/a = +/- 1), where coincidental Mode I stresses were predominantly compressive. Sliding friction substantially increased both KImax and KIImax. The effective Mode I stress intensity factors, Keff, were greatest at g/a = +/- 1, illustrating the significance of high shear stresses generated by loads adjacent to surface cracks. Keff trends suggest mechanisms for surface pitting by which surface cracks propagate along their original plane under repeated reciprocating rolling or sliding, and turn in the direction of sliding under unidirectional sliding contact.  相似文献

Fracture mechanics studies have characterized bone's resistance to fracture in terms of critical stress intensity factor and critical strain energy release rate measured at the onset of a fracture crack. This approach, although useful, provide a limited insight into fracture behavior of bone because, unlike classical brittle materials, bone is a microcracking solid that derives its resistance to fracture during the process of crack propagation from microfracture mechanisms occurring behind the advancing crack front. To address this shortfall, a crack propagation-based approach to measure bone toughness is described here and compared with crack initiation approach. Post hoc analyses of data from previously tested bovine and antler cortical bone compact specimens demonstrates that, in contrast to crack initiation approach, the crack propagation approach successfully identifies the superior toughness properties of red deer's antler cortical bone. Propagation-based slope of crack growth resistance curve is, therefore, a more useful parameter to evaluate cortical bone fracture toughness.  相似文献

Aim

Characterization of polyhydroxyalkanoates (PHA) accumulated by halophilic bacteria isolated from solar salterns.

Methods and Results

Twenty‐six halophilic isolates were obtained from solar salterns of Goa, India. They were screened for accumulation of PHA by Sudan black B, Nile blue A and Nile red stains. Strains H15, H16 and H26 were selected based on their intensity of Nile blue A/Nile red fluorescence. On the basis of phenotypic and genotypic characterization, the three isolates were identified as Bacillus megaterium. Growth kinetics and polymer accumulating capacity of strain H16 were studied in E2 mineral media with 2% glucose with/without NaCl. In the absence of NaCl, strain H16 accumulated PHA to 40·0% (w/w) of cell dry weight (CDW) at 42 h of growth, whereas in presence of 5% w/v NaCl, the culture showed longer lag phase of up to 24 h and accumulated a maximum PHA of 39% (w/w) CDW at 54 h of growth. The infrared spectra of both the polymers exhibited peaks at 1733·9 cm?1 characteristic of C=O. Scans of 1H nuclear magnetic resonance (NMR) showed a doublet at 2·5 ppm corresponding to methylene group (‐CH2), the signal at 5·3 ppm corresponded to methine group (‐CH‐), and another signal at 1·3 ppm corresponded to the methyl group (‐CH3). Scans of 13C NMR showed prominent peaks at 20, 40, 67–68 and 170 ppm, indicating the polymer to be homopolymer of 3‐hydroxybutyrates. The polymer is stable up to a temperature of 160°C.

Conclusion

Three moderately halophilic isolates (strain H15, H16 and H26) capable of accumulating PHA were isolated from solar salterns of Ribandar Goa, India, and identified as B. megaterium based on phenotypic and genotypic characterization. Strain H16 accumulated polyhydroxybutyrate in the presence and absence of NaCl up to 40% of its CDW.

Significance and Impact of the Study

This strain would be better suited for production of PHA at industrial level due to its tolerance to high concentration of NaCl.  相似文献

Third-generation mechanical analogue bone models and synthetic analogue cortical bone materials manufactured by Pacific Research Laboratories, Inc. (PRL) are popular tools for use in mechanical testing of various orthopedic implants and biomaterials. A major issue with these models is that the current third-generation epoxy-short fiberglass based composite used as the cortical bone substitute is prone to crack formation and failure in fatigue or repeated quasistatic loading of the model. The purpose of the present study was to compare the tensile and fracture mechanics properties of the current baseline (established PRL "third-generation" E-glass-fiber-epoxy) composite analogue for cortical bone to a new composite material formulation proposed for use as an enhanced fourth-generation cortical bone analogue material. Standard tensile, plane strain fracture toughness, and fatigue crack propagation rate tests were performed on both the third- and fourth-generation composite material formulations using standard ASTM test techniques. Injection molding techniques were used to create random fiber orientation in all test specimens. Standard dog-bone style tensile specimens were tested to obtain ultimate tensile strength and stiffness. Compact tension fracture toughness specimens were utilized to determine plane strain fracture toughness values. Reduced thickness compact tension specimens were also used to determine fatigue crack propagation rate behavior for the two material groups. Literature values for the same parameters for human cortical bone were compared to results from the third- and fourth-generation cortical analogue bone materials. Tensile properties of the fourth-generation material were closer to that of average human cortical bone than the third-generation material. Fracture toughness was significantly increased by 48% in the fourth-generation composite as compared to the third-generation analogue bone. The threshold stress intensity to propagate the crack was much higher for the fourth-generation material than for the third-generation composite. Even at the higher stress intensity threshold, the fatigue crack propagation rate was significantly decreased in the fourth-generation composite compared to the third-generation composite. These results indicate that the bone analogue models made from the fourth-generation analogue cortical bone material may exhibit better performance in fracture and longer fatigue lives than similar models made of third-generation analogue cortical bone material. Further fatigue testing of the new composite material in clinically relevant use of bone models is still required for verification of these results. Biomechanical test models using the superior fourth-generation cortical analogue material are currently in development.  相似文献

Background  

Angiogenesis, the growth of new blood vessels from the pre-existing vasculature is associated with physiological (for example wound healing) and pathological conditions (tumour development). Vascular endothelial growth factor (VEGF), fibroblast growth factor-2 (FGF-2) and epidermal growth factor (EGF) are the major angiogenic regulators. We have identified a natural product (cheiradone) isolated from a Euphorbia species which inhibited in vivo and in vitro VEGF- stimulated angiogenesis but had no effect on FGF-2 or EGF activity. Two primary cultures, bovine aortic and human dermal endothelial cells were used in in vitro (proliferation, wound healing, invasion in Matrigel and tube formation) and in vivo (the chick chorioallantoic membrane) models of angiogenesis in the presence of growth factors and cheiradone. In all cases, the concentration of cheiradone which caused 50% inhibition (IC50) was determined. The effect of cheiradone on the binding of growth factors to their receptors was also investigated.  相似文献

An adaptive coarse-grained (kinetic) Monte Carlo (ACGMC) simulation framework is applied to reaction and diffusion dynamics in inhomogeneous domains. The presented model is relevant to the diffusion and dimerization dynamics of epidermal growth factor receptor (EGFR) in the presence of plasma membrane heterogeneity and specifically receptor clustering. We perform simulations representing EGFR cluster dissipation in heterogeneous plasma membranes consisting of higher density clusters of receptors surrounded by low population areas using the ACGMC method. Background and aims

The beneficial effects of Si have mainly been observed in herbaceous plants, while little is known about its role in deciduous trees. The aim of this work was to evaluate the effect of foliar application of Si on chestnut leaf growth, photosynthesis and water relations in the presence of short, but intense water deficit.

Methods

Sili-K® solution (containing 0.12 % Si and 0.15 % K) was repeatedly (× 3) sprayed onto leaves of potted chestnut plantlets and irrigation was suspended 7 weeks later, for 8 days. Leaf growth, anatomy, as well as physiological and biochemical traits of the plantlets were studied.

Results

Si application enhanced chestnut growth, due to increased photosynthetic traits, including higher chlorophyll content and chlorophyll a to b ratio, photochemical efficiency of PSII, gas exchange (stomatal conductance, transpiration rate, net CO2 assimilation) and oxygen evolution rate. Meanwhile, Si yielded larger and thinner leaves, higher xylem, specific leaf area and transpiration rate, thus being beneficial to the tree in absorbing sunlight energy for photosynthesis and in alleviating heat stress. However, Si also lowered leaf sap osmotic pressure, causing the plant to lose water more quickly, thus being more susceptible to water stress.

Conclusions

Si improved chestnut photosynthesis, growth, and heat stress tolerance, but it also increased the susceptibility to drought.  相似文献

The analysis of crack growth in titanium was performed using molecular dynamics simulation with Embedded Atom Method potentials. The effect of temperature and strain rate on the mechanism of crack growth and the change of microstructure were discussed. After setting an initial crack, the specimen was subjected to uniaxial tension strain up to the total strain level of 0.2 with a constant strain rate. During the period, the shape and the microstructure of crack tip as well as the stress–strain curves were monitored. In the simulation, the gather of voids and stress concentration leading to the crack growth occurred, which are in agreement with experimental results observed by transmission electron microscopy. The transformation from HCP to BCC also occurred at crack tip. The remarkable effect of temperature and strain rate on the growth direction and rate of stacking fault of crack tip was observed. Moreover, initial crack greatly lowered the tension yield point of pure titanium. In the stage of deformation, simulation results showed that loading strain rate and temperature strongly influenced peak stress point, which was increased by the low temperature and high strain, whereas the initial slope of the stress strain curve was independent of loading strain rate.  相似文献

The effect of ultraviolet-B (UV-B) radiation on Antarctic phytoplankton has become an attractive ecological issue as a result of annual springtime ozone depletion. The effects of UV-B radiation on the growth and antioxidant enzymes were investigated using Antarctic sea ice microalgae Chlamydomonas sp. ICE-L as the material in this study. The results demonstrated that UV-B radiation could notably inhibit the growth, especially at high UV-B radiation intensity (70 μW cm−2). Malondialdehyde and O2 ·− content in ICE-L increased rapidly in early days (1–3 days) exposed to UV-B radiation enhancement, then decreased rapidly. In the stress of UV-B radiation enhancement, the superoxide dismutase, peroxidase and Catalase activities of 1–4 days in ICE-L were obviously higher than those in the control, and their activities became higher at high UV-B radiation intensity (70 μW cm−2). These enzymes activity of 7 days would kept stable at low UV-B radiation intensity (35 μW cm−2), but kept high level at high UV-B radiation intensity (70 μW cm−2). However, the ascorbate peroxidase activity in ICE-L kept stable under the stress of UV-B radiation enhancement. The above experimental results indicated that the antioxidant enzyme system played an important role in the adaptation of Antarctic ice microalgae under the UV-B radiation change of Antarctic ecosystems.  相似文献

Abstract In the absence of inhibitory concentrations of copper, the photoautotrophic growth of Chlorella vulgaris INETI58C at 27°C exhibited a higher specific growth rate and reached a higher maximal concentration of biomass, under irradiance of 150 W m−2, compared with 100 W m−2. However, when the mineral growth medium was supplemented with CuSO4 (range 40–80 μM), algal growth was significantly affected at the higher light intensity. In the presence of Cu2+, the increase in dry biomass was uncoupled from the increase in cell concentration since more than 16 autospores gathered together, inside the enlarged mother cell, suggesting that copper arrested the normal bursting of the mother cell wall. At the higher irradiance, growth medium supplementation with 80 μM of CuSO4 led to bleaching of photosynthetic pigments. No growth was observed, while, under the lower irradiance, growth was only slightly inhibited. Results clearly showed that copper toxicity to growth of C. vulgaris was strongly influenced by light intensity. Higher light intensity elicits lethal or sublethal Cu2+ damage at concentrations lower than the threshold level for damage at lower light intensities. Cu2+ may elicit lethal or sublethal light damage at irradiances lower than the threshold level for unpolluted aquatic systems. Received: 17 January 1997; Accepted 15 April 1997  相似文献

Lipoxygenases (EC 1.13.11.12) catalyse the formation of hydroperoxy derivates by oxygenation of polyunsaturated fatty acids. They act as signal molecules, triggering several developmental processes and defence responses under stress conditions. Incubation of Cd-, IAA- or H2O2-short-term treated seedlings in the presence of LOX inhibitors efficiently inhibited both Cd-, IAA- or H2O2-induced LOX activity and root swelling in a concentration dependent manner, suggesting a key role of LOX or LOX signalling pathway in radial expansion of root cells. Application of antioxidants (ascorbate or N-acetyl cysteine) to the treated seedlings at low 2 mM concentration did not affect the Cd-, IAA- or H2O2-induced LOX activity and root swelling. At higher, 10 mM concentration antioxidants markedly inhibited root growth, significantly increased the activity of LOX and evoked the radial expansion of root cells leading to root swelling with well developed root hairs already in control roots. By contrast, the lipophilic antioxidant trolox, a scavenger of hydroperoxides, severely inhibited the development of Cd-, IAA- or H2O2-induced root swelling, indicating that not directly LOX, but probably oxylipins, products of LOX pathway, are involved in the induction of root swelling in barley root tip. The results of this study suggest a strong connection between abiotic stress-induced alteration in redox and hormone status caused root growth inhibition and LOX pathway mediated radial expansion of root tip cells.  相似文献

Endothelial cell proliferation and migration is initiated by growth factors including FGF and VEGF that bind to specific transmembrane receptor tyrosine kinases. Mechanisms that regulate in vivo expression of fibroblast growth factor receptors (FGFR) and vascular endothelial growth factor receptors (VEGFR) are not well understood. Since it is well known that different matrices influence the proliferation and migration of endothelial cells in culture, we hypothesized that changes in the extracellular matrix environment can regulate growth factor receptors on endothelial cells. We cultured human microvascular endothelial cells on different matrices (vitronectin, laminin, fibronectin, fibrin, and collagen IV) and examined for the presence of growth factor receptors (FGFR-1, FGFR-2, VEGFR-1, and VEGFR-2). We show that vitronectin increased the presence of all four growth factor receptors and most notably, VEGFR-1. In contrast, fibrin decreased all four receptors, especially FGFR-1 and FGFR-2. Inhibiting phosphotyrosine signaling abolished immunostaining for all four receptors, regardless of the matrix, but was not dependent on activating the Fyn-Shc pathway. Cells plated on vitronectin in the presence of blocking antibodies to integrins v3 and v5 similarly decreased presence of these growth factor receptors. Our data suggests a possible mechanism of how matrix-integrin interactions regulate endothelial cell responsiveness to growth factors and anchorage-dependent cell growth.  相似文献
