The virulence of 8 isolates of entomopathogenic hyphomycetes against adult and 5th instar nymph of Clavigralla tomentosicollis was evaluated in the laboratory at 4 different concentrations of inoculum. At all concentrations, Beauveria bassiana CPD 9 and Metarhizium anisopliae CPD 5 caused the highest mortality in adult bug ranging from 58 to 97% and 53 to 100%, respectively at 7 days post inoculation. The same isolates had the shortest LT50 (3.5 and 4.1 days, respectively) and the lowest LC50 (1.8 × 105 and 9.8 × 104 conidia ml-1 values in adult insects. In nymphs, M. anisopliae CPD 5 was the most virulent isolate causing mortality of between 43 to 92% with the shortest LT50 of 2.7 days and the lowest LC50of 4.6 × 105 conidia ml-1 which however did not differ significant from LC50 observed in B. bassiana CPD 9 isolate at 5 days post inoculation. A significant reduction in feeding in both developmental stages treated with fungi was observed at 2 days after treatment with the greatest reduction occurring in insects treated with B. bassiana CPD 9 and M. anisoplia CPD 5. In adult insects treated with these isolates, some bugs ceased feeding 24 h before death. When these two isolates were compared in caged experiment with an untreated control using a susceptible, tolerant and moderately resistant variety of cowpea, percentage pod and seed damage were significantly lower in fungal treated cages than in the control cages on all varieties tested. Grain yield per plant was also significantly higher in fungal treated cages than in the control cages on all varieties. The performance of M. anisopliae CPD 5 was however superior to B. bassiana CPD 9. Application of the fungi on moderately resistant variety of cowpea was found to enhance the performance of the pathogen. In a two-choice test, moreS. furcifera females settled more often on exposed plants than on parafilm-masked ones, regardless of the susceptibility of rice varieties. This indicates that rice volatiles play an important role in the insect's short-range orientation to its host. The fact that more insects settled on exposed resistant Rathu Heenati (RHT) than to masked susceptible Taichung Native 1 (TN1) suggests that there must be certain common volatiles released by both varieties. Few females landed on masked plants of either RHT or TN1. This implies that the insect could not recognize at a distance that a plant was resistant or susceptible without olfactory stimuli.S. furcifera excreted less honeydew on masked plants than on exposed ones for both varieties and more on masked TN1 than on exposed RHT. The electronic monitoring of feeding behavior demonstrates that the insect made more frequent probes and had shorter phloem ingestion durations on exposed RHT than on exposed TN1 and on masked RHT than on masked TN1. Moreover, the insect had longer phloem ingestion durations on masked TN1 than on exposed RHT. These results suggest that volatile chemicals given off by resistant RHT plants have a negative effect on feeding.  相似文献

为了明确防卫基因PAL与美洲南瓜抗西瓜花叶病毒(watermelon mosaic virus,WMV)之间的关系,通过室内接种和实时荧光定量PCR技术,测定了WMV侵染对不同抗性美洲南瓜体内防卫基因PAL表达的影响。结果表明:(1)室内测定显示,抗病品种GBRV-8发病率和病情指数(15.6%和14.2)显著低于感病品种‘光板’(91.1%和65.9)。(2)实时荧光定量PCR表明,接种WMV后不同抗感品种不同组织部位PAL基因相对表达量随着接种时间增加,整体呈现出先增加后降低的趋势,而且不同组织部位PAL基因相对表达量总体呈现出叶片较高,叶柄和茎秆次之。(3)接种后5个品种不同组织部位PAL基因相对表达量与对照相比均存在显著差异,且抗病和中抗品种不同组织部位PAL基因相对表达量显著高于感病品种,尤其抗病品种GBRV-8不同组织部位PAL基因相对表达量最高,感病品种光板最低。研究认为,防卫基因PAL表达量与美洲南瓜品种抗病毒病强弱密切相关。  相似文献

New Zealand has extensive alpine and subalpine habitats where, together with some lowland sites, insects are exposed to subzero temperatures. Studies of cold tolerance in New Zealand insects have centred on an alpine weta (Hemideina maori), which is the world's largest freezing tolerant insect, and an alpine cockroach (Celatoblatta quinquemaculata). Both of these insects are moderately freezing tolerant and have ice nucleating agents in their haemolymph and guts. There is some evidence for the survival of intracellular ice formation in the isolated gut tissue of C. quinquemaculata. Trehalose is a suggested cryoprotectant in both H. maori and C. quinquemaculata whilst proline also provides this role in H. maori. Cells and tissues of both insects maintain viability and physiological function during freezing to moderately low temperatures but viability declines at lower temperatures, the most vulnerable tissue presumably setting the limit to the survival of the animal. Antifreeze proteins are found in the gut tissue of C. quinquemaculata and may protect this tissue when freezing occurs in the gut. Several other New Zealand insects are also moderately freezing tolerant and the apparent dominance of this cold tolerance strategy in the New Zealand fauna may reflect the relatively mild climate but unpredictable exposure to subzero temperatures that is typical of many Southern Hemisphere environments.  相似文献

Sunflower head rot is a major disease caused by Sclerotinia sclerotiorum. Sunflower varieties which are tolerant to the fungus have been developed. The changes occurring in flower parts at different times after inoculation with pathogen ascospores were studied for two sunflower varieties (tolerant HA 302 and susceptible HA 89). In variety HA 302 there was cell collapse, changes in cell wall composition, and an increase in phenolic compounds in the tissues of corolla and style, which prevented the pathogen from advancing. This response was weaker in susceptible variety HA 89, and occurred only in the style, so did not stop the pathogen from developing and reaching the ovary. Phenolic compounds were found in HA 302 corolla and style tissues only when the pathogen was present, constituted an induced response that prevented further development of the fungus. Principal component analysis (PCA) showed that at the beginning of the infection there was no difference in behavior between the two varieties. The difference arose during the final observation times, when in variety HA 89, the pathogen colonized ovary, style and base of filaments and produced noticeable colonization of the corolla.  相似文献

Abstract

Six varieties of lentil (Lens culinaris Medik.) – Asha, Subrata, IPL 406, IPL 81, Lv and Sehore – were exposed to temperatures ranging from 30–50°C which resulted in retarded germination and seedling growth at higher temperatures. Tolerance index and membrane stability tests revealed Sehore and Lv to be susceptible to elevated temperatures while IPL 406, IPL 81, Asha and Subrata were tolerant. Catalase, ascorbate peroxidase and superoxide dismutase showed an initial increase before declining at 50°C, while peroxidase and glutathione reductase activities declined at all temperatures. Lipid peroxidation significantly increased in all varieties. In the tolerant varieties, there was an initial decrease in accumulation of H2O2 followed by an increase from 40°C onwards; however, in the susceptible varieties, accumulation was enhanced at all high temperatures. Ascorbate and glutathione also showed initial increase followed by a decline. Total antioxidant activity was at a maximum at 35–40°C in the tolerant varieties and at 30°C in the susceptible ones. Oxidative stress induced by high temperature was ameliorated by treatment with salicylic acid, abscisic acid or CaCl2, of which salicylic acid was the most effective.  相似文献

The reaction of 15 varieties of maize to the growth and cyclopiazonic acid (CPA) production ofPenicillium griseofulvum was examined as a means of identifying varieties which would be resistant to this infestation. Only one variety, MMEH-25, was resistant to the fungus, seeds containing little CPA 30 days after infestation. Callus cultures from salt tolerant (CSR-10) and susceptible (Swarnadhan) varieties of Oryza sativa L. were established in Murashige and Skoog’s (MS) medium containing lethal concentrations (50 mM) of rubidium chloride (RbCl) as a selective agent. While 95–100% cells were viable in callus cultures grown without RbCl, viability was 75% in 50 mM RbCl selected cultures. Growth of RbCl selected calli in presence of salt was comparable to that of callus grown without it. Cells tolerant to RbCl showed more vacuoles and accumulated more K+ in comparison with their corresponding controls. Suspension cultures were established and uptake of 86Rb+ was measured at 10 and 20 min intervals, which revealed a linear relationship between the absorption of K+ and time. Callus cultures (560-day-old) tolerant to 50 mM RbCl regenerated shoots with 35–40% frequencies in both the varieties, but the same age-old callus grown in the medium devoid of RbCl did not show any organogenesis. Callus cultures that are tolerant to 50 mM RbCl when exposed to 25 mM LiCl, 50 mM NaCl, 50 mM KCl and 25 mM CsCl also exhibited cross tolerance in both the varieties. This is the first time that a callus line of rice resistant to RbCl was raised and shown to accumulate a major cation K+ and also an increased influx of it.  相似文献

The cotton aphid, Aphis gossypii Glover (Homoptera: Aphididae), is an important cotton pest in northern China, especially in the seedling stage of cotton. After large scale commercial use of transgenic Bt cotton, cotton aphids became one of the most important cotton pests. A 2‐year study was conducted to evaluate the role of four winter wheat varieties that were resistant or susceptible to wheat aphid, Sitobion avenae Fabricius (Homoptera: Aphididae), in conserving arthropod natural enemies and suppressing cotton aphids in a wheat–cotton relay intercropping system in northern China. The results indicated that wheat–cotton intercropping preserved and augmented natural enemies more than a monoculture of cotton. The density of natural enemies in cotton was significantly different among relay‐intercropping fields with different wheat varieties. The highest density of natural enemies and low cotton aphid populations were found in the treatment of cotton in relay intercropped with the wheat variety Lovrin10, which is susceptible to wheat aphid. The lowest density of predators and parasitoids associated with high cotton aphid populations were found with the wheat variety KOK1679, which is resistant to wheat aphid. The results showed that wheat varieties that are susceptible or moderately resistant to wheat aphid might reduce cotton aphids more effectively than an aphid‐resistant variety in the intercropping system by enhancing predators to suppress cotton aphids during the cotton seedling stage.  相似文献

Abstract. Investigations of the responses to acclimation of upper and lower lethal limits and limits to activity in insects have focused primarily on Drosophila. In the present study, Halmaeusa atriceps (Staphylinidae) is examined for thermal tolerance responses to acclimation, and seasonal acclimatization. In summer and winter, lower lethal temperatures of adults and larvae are approximately −7.6 ± 0.03 and −11.1 ± 0.06 °C, respectively. Supercooling points (SCPs) are more variable, with winter SCPs of −5.4 ± 0.4 °C in larvae and −6.3 ± 0.8 °C in adults. The species appears to be chill susceptible in summer and moderately freeze tolerant in winter, thus showing seasonal acclimatization. Similar changes cannot be induced solely by acclimation to low temperatures in the laboratory. Upper lethal temperatures show a weaker response to acclimation. There are also significant responses to acclimation of critical thermal limits. Critical thermal minima vary between −3.6 ± 0.2 and −0.6 ± 0.2 °C in larvae, and from −4.1 ± 0.1 to −0.8 ± 0.2 °C in adults. By contrast, critical thermal maxima vary much less within adults and larvae. These findings are in keeping with the general pattern found in insects, although this species differs in several respects from others found on Marion Island.  相似文献

The selection of TSWV resistant individuals can be facilitated by molecular markers. RAPD analysis was carried out on three forms (Stevens × Rodade — resistant; Rey de los Tempranos — moderately tolerant; Potentat — susceptible) with the use of 271 primers. Out of 271 primers 28 generated stable polymorphism and so they were tested for linkage to resistance gene. Bulk segregant analysis (BSA) was applied to F2 segregating progeny developed from resistant × susceptible parents. As a result, 5 primers enabling us to distinguish between resistant and susceptible forms were detected. Only one of them had previously been reported by Chague et al. (1996). The analysis should be repeated on a larger population to confirm the results obtained.  相似文献

为探明辽宁地区水稻品种对本地灰飞虱Laodelphax striatellus(Fallén)的抗性水平及其抗虫机制,本研究利用改进的苗期集团鉴定法,以IR36为抗虫对照品种、武育粳3号为感虫对照品种,对42份辽宁地区主栽水稻品种和研究待推广品种进行了水稻苗期对灰飞虱抗性鉴定,并从中选取20份不同抗性水平的品种进行了排趋性和抗生性的测定。结果表明:从42份水稻材料中仅筛选出1份抗虫材料辽优5218,中抗品种11份,其余均为感虫或高感品种。在不同水稻类型中,杂交稻的抗虫性普遍较常规稻强,而从水稻株型上看,抗性品种大多为披散型。抗虫机制研究发现,抗虫品种辽优5218和中抗品种港育129兼具排趋性和抗生性,是非常理想的抗性种质资源,中抗品种港源8号和粳优558具有很强的排趋性,也是较为理想的抗性资源,为抗性机制的深入研究提供了材料。但大部分省内主栽主推品种不具备对灰飞虱的抗性,应引起重视。  相似文献

【背景】香蕉枯萎病是世界性的香蕉毁灭性病害,尚无有效药剂防控,筛选抗病品种是目前理想的防治方法。【方法】采用组培苗伤根接种法,研究了18份香蕉种质对香蕉枯萎病菌4号生理小种的抗性水平,并根据病情指数进行抗性分级。【结果】在供试的18份香蕉种质中,2份(东莞大蕉、抗枯5号)高抗,2份(碧盛、大丰)抗病,3份(抗枯1号、粉杂、农科1号)中抗,7份(粤优抗1号、广东-741、泰国B9、大蕉、台湾8号、海贡蕉、威廉斯8818)感病,4份(巴西、广东2号、广粉1号、粉蕉)高感。【结论与意义】不同香蕉种质对香蕉枯萎病菌4号生理小种的抗病性存在较大差异,本研究初步筛选出7份抗枯萎病的香蕉种质,为香蕉枯萎病抗病育种提供了依据,为病区种植香蕉品种提供了有效参考。  相似文献

The short-term response of redlegged earth mite, Halotydeus destructor (Tucker) (Acarina: Penthaleidae) to cotyledons of different varieties of subterranean clover (Trifolium subterraneum subsp. subterraneum L.) was assessed by means of paired choice tests, and no-choice tests. H. destructor had lower numbers and fed less on detached cotyledons of resistant than susceptible varieties, yielding a correlation between the numbers of mites and feeding damage to the cotyledons during a three hour test period. For a number of resistant and susceptible varieties, there was a negative correlation between cotyledon deterrence in the three hour choice test and feeding damage to seedling after a two week period. Since the response of the mites to different subclover varieties occurred within three hours, it is concluded that the resistance is based on antixenosis.No evidence was obtained for antifeedant activity in organic solvent extracts from the variety DGI007 (resistant) in comparison with those from the variety Dalkeith (susceptible). Water soluble compounds from DGI007 cotyledons were preferred by mites, in feeding tests in terms of numbers, over those from Dalkeith (susceptible). Squeezed sap from the cotyledons of both varieties showed the same effects on mites as 5% glucose and were more phagostimulatory than water extracts. Mechanically damaged cotyledons of Dalkeith and DGI007 attracted more mites than the undamaged counterparts. The toughness of cotyledons in 17 varieties of T. subterraneum subsp. subterraneum was measured with a manual penetrometer. Results showed a negative correlation between toughness values and mite feeding damage scores (r2=0.752) for all varieties except S3615D (resistant). This implies a likely involvement of epidermal toughness as a contributor in the antixenotic resistance of these varieties. Other mechanisms may be involved in the resistance of S3615D.  相似文献

Preference studies of the white-backed planthopper, Sogatella furcifera (Horvath) (Delphacidae: Homoptera) (WBPH) for different growth stages of rice, Oryza sativa L., indicated that WBPH selected its preferred variety for feeding by alighting on the plant and feeding briefly, tasting the plant. Differential preference for shelter, food or oviposition by WBPH were observed for different rice varieties and for different stages of a variety. With the advancement of plant age, rice varieties became more unsuitable for feeding and a process progressing more rapidly in resistant varieties. Most of the resistant varieties viz. Pundia, Landi Sarakanti and Ptb-33, unsuitable for shelter and feeding were usually not preferred for oviposition. However, the tolerant variety Mahia bankoi, though suitable for shelter and feeding was also not preferred for oviposition. Therefore, when screening varieties by only measuring feeding preference sources having tolerance mechanisms of resistance may be overlooked.  相似文献

Sugarcane is an important international commodity as a valuable agricultural crop especially in tropical and subtropical countries. Two bulked DNA used to screen polymorphic primers from commercial hybrids (varieties) with moderately resistant and highly susceptible to red rot disease. Among 145 simple sequence repeat and unigene primers screened, 37 (25%) were found to be highly robust and polymorphic with Polymorphism Information Content values ranging from 0.50 to 1.00 with the mean value of 0.82. Among these microsatellites, twenty one were used in the study of genetic relationships and marker identification in sugarcane varieties for red rot resistance. A total of 105 polymorphic DNA bands were identified, with their fragment size ranging from 54 to 1,280 bp. Jaccard’s similarity coefficient value recorded between closely related hybrids was 0.986 while lowest coefficient value of 0.341 was detected with distantly related hybrids. The average similarity coefficient among these hybrids was 0.663. Cluster analysis resulted in a dendrogram with two major clusters separating the moderately resistant varieties from highly susceptible varieties. Three group specific fragments amplified by unigene Saccharum microsatellite primers viz; two markers UGSM316850 and UGSM31660 were closely associated with moderately resistant varieties by appearing bands in this region but the bands were absent in highly susceptible varieties. Similarly UGSM316400 marker was tightly linked with highly susceptible varieties by amplifying uniformly in sugarcane varieties showing highly susceptible reaction to red rot but it was absent in moderately resistant varietal groups. Validation of red rot resistance/susceptibility associated markers on a group of different mapping populations for red rot resistant/susceptible traits is in progress.  相似文献

Three spring wheat genotypes, susceptible, moderately resistant or resistant to Pyrenophora tritici-repentis (tan spot fungus) were exposed to charcoal-filtered air and to approx. 80, 160, 240 (g m?3 ozone for five consecutive days (7 h per day). Visible leaf injury on seedling plants (three-leaf stage) was only observed after fumigation with 160 or 240 (g m?3 O3. Amount of injury was four-fold and 10-fold on the susceptible genotype when compared to resistant or moderately resistant genotype at the two highest concentration of ozone, respectively. Genotypic differences to O3 tolerance were detected at the seedling growth stage (three-leaf stage) and flowering stage but not at the stem elongation stage. A significant increase in tan spot lesion area was observed only on O3 predisposed second top most leaves of the susceptible genotype at all the three levels of ozone. Predisposition did not enhance tan spot development in resistant and moderately resistant genotypes. In a test with 12 wheat genotypes, a highly significant positive correlation (r = 0· 986, p < 0· 0001) was observed between ozone sensitivity (percent leaf area damaged due to 240 (g m?3 ozone exposure) and tan spot development (mm2 lesion area) following inoculation with P. tritici-repentis. It indicates that wheat genotypes resistant to the tan spot fungus might be tolerant to ozone damage.  相似文献

本文运用苗期抗性筛选法,测定了9个水稻品种对褐稻虱的抗性及其与秧龄的关系,表明秧龄对高抗品种的抗性表现影响不显著,而部分中抗品种的抗性则随秧龄增长而加大。抗性机制研究证明:高抗品种7105具有很强的不嗜性和抗生性;而包选2号和辐包矮21等中抗品种则以耐虫性为主,兼有不同程度的抗生性。  相似文献

对3个类型稻田早、晚稻捕食性节肢动物群落调查,结果表明:共有捕食性节肢动物67种,丰富度以蜘蛛高于捕食性昆虫类;早稻高于晚稻;中抗品种高于抗虫和感虫品种。早稻以皿蛛科Linyphiidae的食虫沟瘤蛛UlmmeliatainsecticepesBoes.etStr.占优势,晚稻则以狼蛛科Lycosidae的相对丰盛度最高。早稻捕食性天敌数量动态以中抗品种峰期最长;感性样田中天敌平均密度与中抗样田差异不显著,与抗虫品种差异显著;捕食性节肢动物多样性随着品种抗性的增强呈降低趋势,优势度随着品种抗性的增强而增强。晚稻3个品种样田中天敌的平均密度差异不显著;各品种样田的多样性明显高于早稻,且波动幅度小于早稻,品种之间多样性差异较小,晚稻群落可能具有较高的稳定性。  相似文献

The giant willow aphid Tuberolachnus salignus Gmelin is a large phloem-feeding insect which colonizes the stems of willow trees. This aphid is a new invasive species in New Zealand and there is limited knowledge of its inter-annual population patterns and the damage it can cause to willow hosts. Our study investigated the T. salignus population dynamics and its effects on the flowering parameters of fifteen willow species and hybrids in a field trial. The aphid population levels were found to vary among the willow species and hybrids. Based on the aphid population levels, the willow species and hybrids were classified as resistant, moderately resistant, susceptible or highly susceptible. T. salignus infestation had no effect on the flowering of resistant and moderately resistant willows, but significantly delayed the flowering time, extended the flowering duration, and decreased the catkin length in susceptible species and hybrids. Interestingly, aphid infestation was found to increase the catkin number and total floral output of some willow species and hybrids. It can be concluded that aphid populations and their effects on flowering are host-specific, with large differences between resistant and susceptible host plants.  相似文献
