The BeWo trophoblastic cell line was employed to assess the requirement for microtubules and cellular energy in human chorionic gonadotropin (hCG) secretion. In contrast to the general inhibitory effect of colchicine and vincristine on hormone secretion in systems involving exocytosis, wide concentration ranges of these antimicrotubule agents caused enhancement of hCG secretion. Similarly, cytochalasin B, an agent that interferes with microfilament function, doubled both basal hCG secretion, and secretion of hCG stimulated by 1 mM dibutyryl cyclic AMP plus 1 mM theophylline (dbT). Inhibitors of cellular energy production (2,4-dinitrophenol, malonate, azide) decreased both secreted and cellular levels of hormone. High concentrations of K+ gave no enhancement of basal or dbT-stimulated hCG secretion, nor any reduction of cellular hCG levels. These findings contrasted with observations of others in secretory systems involving exocytosis, in which high K+ potentiated basal or stimulated hormone release and depleted cellular stores of hormone. It was concluded that the process of hCG secretion in the malignant trophoblast is fundamentally different from the mechanism of protein hormone secretion in other tissues.  相似文献

Summary The rate of active K+ transport by the isolated lepidopteran midgut shows a rectangular hyperbolic relation to [K+] over the range 20 to 70mm K+ in the absence of any divalent cation. Addition of Ba++ to the hemolymph (K+ uptake) side introduces a linear component to the concentration dependence, such that active K transport is decreased at [K+] of 55mm or less, but increased transiently at higher [K+]. As [Ba++] is increased over the range 2 to 8mm the linear component increases and the saturating component decreases; in 8mm Ba++ the concentration dependence is dominated by the linear component. The effect of Ba++ cannot easily be accounted for by simple competition with K+ for basal membrane uptake sites. Similar effects might be exercised by other alkali earth cations, since the concentration dependence of active K+ transport possesses a substantial linear component in solutions containing 5mm Ca++ and 5mm Mg++ (the alkali earth metal concentrations of standard lepidopteran saline).  相似文献

Summary To study the physiological role of the bidirectionally operating, furosemide-sensitive Na+/K+ transport system of human erythrocytes, the effect of furosemide on red cell cation and hemoglobin content was determined in cells incubated for 24 hr with ouabain in 145mm NaCl media containing 0 to 10mm K+ or Rb+. In pure Na+ media, furosemide accelerated cell Na+ gain and retarded cellular K+ loss. External K+ (5mm) had an effect similar to furosemide and markedly reduced the action of the drug on cellular cation content. External Rb+ accelerated the Na+ gain like K+, but did not affect the K+ retention induced by furosemide. The data are interpreted to indicate that the furosemide-sensitive Na+/K+ transport system of human erythrocytes mediates an equimolar extrusion of Na+ and K+ in Na+ media (Na+/K+ cotransport), a 1:1 K+/K+ (K+/Rb+) and Na+/Na+ exchange progressively appearing upon increasing external K+ (Rb+) concentrations to 5mm. The effect of furosemide (or external K+/Rb+) on cation contents was associated with a prevention of the cell shrinkage seen in pure Na+ media, or with a cell swelling, indicating that the furosemide-sensitive Na+/K+ transport system is involved in the control of cell volume of human erythrocytes. The action of furosemide on cellular volume and cation content tended to disappear at 5mm external K+ or Rb+. Thein vivo red cell K+ content was negatively correlated to the rate of furosemide-sensitive K+ (Rb+) uptake, and a positive correlation was seen between mean cellular hemoglobin content and furosemide-sensitive transport activity. The transport system possibly functions as a K+ and waterextruding mechanism under physiological conditiosin vivo. The red cell Na+ content showed no correlation to the activity of the furosemide-sensitive transport system.  相似文献

The ionic dependencies of stimulated and unstimulated Locusta tubules have been studied. K+, Na+, Cl? are essential to both basal and stimulated secretion. K+ is secreted against a concentration gradient in unstimulated tubules. In response to diuretic hormone or cAMP application, there is a dramatic influx of K+ into the lumen. A high level of Na+ and Cl? in the bathing medium is required to allow maximal fluid secretion. The tubules show an apparent impermeability to Na+; its concentration in the secreted fluid is always much less than in the bathing medium. If Na+ is omitted from the medium and excess K+ added (80 mM K), then although basal secretion occurs (2.5 nl/min), the tubules fail to respond to stimulation. Clearly Na+ has an important indirect role to play in stimulated fluid secretion.  相似文献

Neurosecretory granule exocytosis from axon terminals in the crayfish (Orconectes virilis) sinus gland can be effected by alterations in intracellular ionic concentrations through modification of the bathing medium or by electrical stimulation. This may result in increased numbers of exocytotic profiles involving one granule (single exocytosis) or several granules (compound exocytosis), which we interpret as evidence of stimulated neurosecretion. Our results suggest that increased free intracellular Ca++ together with a decrease in intracellular K+ are prerequisite for and sufficient to elicit increased exocytosis from sinus gland terminals. Similar release, stimulated by horseradish peroxidase, indicates that local surface permeability changes are involved. A minor response to high-K+-Ringer, and secretion via compound exocytosis, are uncharacteristic of cells with a neural ancestry.  相似文献

Summary Single K+-selective channels were studied in excised inside-out membrane patches from dissociated mouse toe muscle fibers. Channels of 74 pS conductance in symmetrical 160mm KCl solutions were blocked reversibly by 10 m internal ATP and thus identified as ATP-sensitive K+ channels. The channels were also blocked reversibly bymm concentrations of internal adenosine, adenine and thymine, but not by cytosine and uracil. The efficacy of the reversible channel blockers was higher when they were present in internal NaCl instead of KCl solutions. An irreversible inhibition of ATP-sensitive K+ channels was observed after application of several sulphydryl-modifying substances in the internal solution: 0.5mm chloramine-T, 50mm hydrogen peroxide or 2mm n-ethylmaleimide (NEM). Largeconductance Ca-activated K+ channels were not affected by these reagents. The presence of 1mm internal ATP prevents the irreversible inhibition of ATP-sensitive K+ channels by NEM. The results suggest that internal Na+ ions increase the affinity of the ATP-sensitive K+ channel to ATP and to other reversible channel blockers and that a functionally important SH-group is located at or near the ATP-binding site.  相似文献

Summary The concept that interaction between sodium-dependent transport systems represents competition for energy inherent in the transmembrane sodium gradient was examined with the use of isolated intestinal epithelial cells. The isolated cells exhibit transport interactions which are more significant in magnitude than those which have been described for intact tissue preparations. Accumulation of 1mm valine is inhibited 60% by 10mm 3-OMG. Conversely, uptake of 1mm 3-OMG is inhibited only 20% by 10mm valine. These data suggest that 3-OMG must discharge the cellular Na+ gradient more effectively than valine, if Na+ gradient dissipation can be taken as a basis for the inhibitory interaction. However, entry of 10mm 3-OMG is significantly slower than the entry of 10mm valine. Even if appropriate corrections are made for passive substrate entry and for differences in Na/substrate entry stoichiometry, it appears that valine should be somewhat more effective than 3-OMG in discharging the Na+ gradient. In light of these facts, it seems unlikely that the mechanistic basis for interaction between sugar and amino acid transport systems can be related to concomitant co-entry of Na+. It is suggested that the interaction results instead from competition for energized intermediates generated at limited rates by basic energy transduction events associated with the cell membrane which serve in support of a variety of active transport systems.  相似文献

Summary To investigate the voltage dependence of the Na/K pump, current-voltage relations were determined in prophasearrested oocytes ofXenopus laevis. All solutions contained 5mm Ba2– and 20mm tetraethylammonium (TEA) to block K channels. If. in addition, the Na+/K+ pump is blocked by ouabain, K+-sensitive currents no larger than 50 nA/cm2 remain. Reductions in steady-state current (on the order of 700 nA/cm2) produced by 50 m ouabain or dihydro-ouabain or by K+ removal, therefore, primarily represent current generated by the Na/K pump. In Na-free solution containing 5mm K+, Na+/K+ pump current is relatively voltage independent over the potential range from –160 to +40 mV. If external [K+] is reduced below 0.5mm, negative slopes are observed over this entire voltage range. Similar results are seen in Na+- and Ca2+-free solutions in the presence of 2mm Ni2+, an experimental condition designed to prevent Na+/Ca2+ exchange. The occurrence of a negative slope can be explained by the voltage dependence of the apparent affinity for activation of the Na+/K+ pump by external K+, consistent with the existence of an external ion well for K binding. In 90mm Na+, 5mm K+ solution, Na+/K+ pump current-voltage curves at negative membrane potentials have a positive slope and can be described by a monotonically increasing sigmoidal function. At an extracellular [K+] of 1.3mm, a negative slope was observed at positive potentials. These findings suggest that in addition to a voltage-dependent step associated with Na+ translocation, a second voltage-dependent step that is dependent on external [K+], possibly external K+ binding, participates in the overall reaction mechanism of the Na+/K+ pump.  相似文献

Summary The effects of external Rb+ on the efflux of42K+ from whole frog sartorius muscles loaded with 305mm K+ and 120mm Cl were studied. K+ efflux is activated by [Rb+] o less than about 40mm according to a sigmoid relation similar to that for activation by [K+] o . At [Rb+]o greater than 40mm, K+ efflux declines, although at [Rb+] o =300mm it is still greater than at [Rb+] o =0mm. For low concentrations, the increment in K+ efflux over that in K+- and Rb+-free solution, k, is described by the relation k=a[X+] o n , for both K+ and Rb+. The value ofa is larger for Rb+ than for K+, while the values ofn are similar; the activation produced by a given [Rb+] o is larger than that by an equal [K+] o for concentrations less than about 40mm. Adding a small amount of Rb+ to a K+-containing solution has effects on K+ efflux which depend on [K+] o . At low [K+] o , adding Rb+ increases K+ efflux, the effect being greatest near [K+] o =30mm and declining at higher [K+] o ; at [K+] o above 40mm, addition of Rb+ decreases K+ efflux. At [K+] o above 75mm, where K+ efflux is largely activated, Rb+ reduces K+ efflux by a factorb, described by the relationb=1/(1+c[Rb+] o ). Activation is discussed in terms of binding to at least two sites in the membrane, and the reduction in K+ efflux by Rb+ at high [K+] o in terms of association with an additional inhibitory site.  相似文献

Summary Insulin release and membrane potential fluctuations in response to increased extracellular potassium [K+] o have been measured in single perifused islets of Langerhans from normal mice. An increase in [K+] o from 5mm to 50mm induced a transient insulin release with a peak at about 1 min. The peak value was [K+] o -dependent but the half-timet 1/2 for the decline was constant at nearly 1 min. 2.5mm cobalt completely inhibited the potassium-induced stimulation of insulin release. The insulin release elicited by 28 and 50mm [K+] o was similar in terms of peak, total release and half-time from maximum release. Stepwise increase in [K+] o from 10 to 28 to 50mm resulted in a normal response to 28mm but no peak of release after the 28 to 50mm increase. The results indicate good correlation between excess voltage noise, thought to reflect calcium channel activity, and insulin release evoked by changing extracellular potassium.  相似文献

Summary Human red cells were prepared with various cellular Na+ and K+ concentrations at a constant sum of 156mm. At maximal activation of the K+ conductance,g K(Ca), the net efflux of K+ was determined as a function of the cellular Na+ and K+ concentrations and the membrane potential,V m , at a fixed [K+]ex of 3.5mm.V m was only varied from (V m E K)25 mV and upwards, that is, outside the range of potentials with a steep inward rectifying voltage dependence (Stampe & Vestergaard-Bogind, 1988).g K(Ca) as a function of cellular Na+ and K+ concentrations atV m =–40, 0 and 40 mV indicated a competitive, voltage-dependent block of the outward current conductance by cellular Na+. Since the present Ca2+-activated K+ channels have been shown to be of the multi-ion type, the experimental data from each set of Na+ and K+ concentrations were fitted separately to a Boltzmann-type equation, assuming that the outward current conductance in the absence of cellular Na+ is independent of voltage. The equivalent valence determined in this way was a function of the cellular Na+ concentration increasing from 0.5 to 1.5 as this concentration increased from 11 to 101mm. Data from a previous study of voltage dependence as a function of the degree of Ca2+ activation of the channel could be accounted for in this way as well. It is therefore suggested that the voltage dependence ofg K(Ca) for outward currents at (V m E K)>25 25 mV reflects a voltage-dependent Na+ block of the Ca2+-activated K+ channels.  相似文献

Summary The lepidopteran midgut is a model for the oxygendependent, electrogenic K+ transport found in both alimentary and sensory tissues of many economically important insects. Structural and biochemical evidence places the K+ pump on the portasome-studded apical plasma membrane which borders the extracellular goblet cavity. However, electrochemical evidence implies that the goblet cell K+ concentration is less than 50mm. We used electron probe X-ray microanalysis of frozenhydrated cryosections to measure the concentration of Na, Mg, P, S, Cl, K, Ca and H2O in several subcellular sites in the larval midgut ofManduca sexta under several experimental regimes. Na is undetectable at any site. K is at least 100mm in the cytoplasm of all cells. Typicalin vivo values (mm) for K were: blood, 25; goblet and columnar cytoplasm, 120; goblet cavity, 190; and gut lumen, 180. The high K concentration in the apically located goblet cavity declined by 100mm under anoxia. Both cavity and gut fluid are Cl deficient, but fixed negative charges may be present in the cavity. We conclude that the K+ pump is sited on the goblet cell apical membrane and that K+ follows a nonmixing pathway via only part of the goblet cell cytoplasm. The cavity appears to be electrically isolated in alimentary tissues, as it is in sensory sensilla, thereby allowing a PD exceeding 180 mV (lumen positive) to develop across the apical plasma membrane. This PD appears to couple K+ pump energy to nutrient absorption and pH regulation.  相似文献

Summary We have measured the intracellular potassium activity, [K+]i and the mechanisms of transcellular K+ transport in reabsorptive sweat duct (RSD) using intracellular ion-sensitive microelectrodes (ISMEs). The mean value of [K+]i in RSD is 79.8±4.1mm (n=39). Under conditions of microperfusion, the [K+]i is above equilibrium across both the basolateral membrane, BLM (5.5 times) and the apical membrane, APM (7.8 times). The Na+/K+ pump inhibitor ouabain reduced [K+]i towards passive distribution across the BLM. However, the [K+]i is insensitive to the Na+/K+/2 Cl cotransport inhibitor bumetanide in the bath. Cl substitution in the lumen had no effect on [K+]i. In contrast, Cl substitution in the bath (basolateral side) depolarized BLM from –26.0±2.6 mV to –4.7*±2.4 mV (n=3;* indicates significant difference) and decreased [K+]i from 76.0±15.2mm to 57.7* ±12.7mm (n=3). Removal of K+ in the bath decreased [K+]i from 76.3±15.0mm to 32.3*±7.6mm (n=4) while depolarizing the BLM from –32.5±4.1 mV to –28.3*±3.0 mV (n=4). Raising the [K+] in the bath by 10-fold increased [K+]i from 81.7±9.0mm to 95.0*±13.5mm and depolarized the BLM from –25.7±2.4 mV to –21.3*±2.9 mV (n=4). The K+ conductance inhibitor, Ba2+, in the bath also increased [K+]i from 85.8±6.7mm to 107.0*±11.5mm (n=4) and depolarized BLM from –25.8±2.2 mV to –17.0*±3.1 mV (n=4). Amiloride at 10–6 m increased [K+]i from 77.5±18.8mm to 98.8*±21.6mm (n=4) and hyperpolarized both the BLM (from –35.5±2.6 mV to –47.8*±4.3 mV) and the APM (from –27.5±1.4 mV to –46.0* ±3.5 mV,n=4). However, amiloride at 10–4 m decreased [K+]i from 64.5±0.9mm to 36.0*±9.9mm and hyperpolarized both the BLM (from –24.7±1.4 mV to –43.5*±4.2 mV) and APM (from –18.3±0.9 mV to –43.5*±4.2 mV,n=6). In contrast to the observations at the BLM, substitution of K+ or application of Ba2+ in the lumen had no effect on the [K+]i or the electrical properties of RSD, indicating the absence of a K+ conductance in the APM. Our results indicate that (i) [K+]i is above equilibrium due to the Na+/K+ pump; (ii) only the BLM has a K+ conductance; (iii) [K+]i is subject to modulation by transport status; (iv) K+ is probably not involved in carrier-mediated ion transport across the cell membranes; and (v) the RSD does not secrete K+ into the lumen.  相似文献

Summary The effects of cAMP, ATP and GTP on the Ca2+-dependent K+ channel of fresh (1–2 days) or cold-stored (28–36 days) human red cells were studied using atomic absorption flame photometry of Ca2+-EGTA loaded ghosts which had been resealed to monovalent cations in dextran solutions. When high-K+ ghosts were incubated in an isotonic Na+ medium, the rate constant of Ca2+-dependent K+ efflux was reduced by a half on increasing the theophylline concentration to 40mm. This effect was observed in ghosts from both fresh and stored cells, but only if they were previously loaded with ATP. The inhibition was more marked when Mg2+ was added together with ATP, and it was abolished by raising free Ca2+ to the micromolar level. Like theophylline, isobutyl methylxanthine (10mm) also affected K+ efflux. cAMP (0.2–0.5mm), added both internally and externally (as free salt, dibutyryl or bromide derivatives), had no significant effect on K+ loss when the ghost free-Ca2+ level was below 1 m, but it was slightly inhibitory at higher concentrations. The combined presence of cAMP (0.2mm) plus either theophylline (10mm), or isobutyl methylxanthine (0.5mm), was more effective than cAMP alone. This inhibition showed a strict requirement for ATP plus Mg2+ and it, was not overcome by raising internal Ca2+. Ghosts from stored cells seemed more sensitive than those from fresh cells, to the combined action of cAMP and methylxanthines. Loading ATP into ghosts from fresh or stored cells markedly decreased K+ loss. Although this effect was observed in the absence of added Mg2+ (0.5mm EDTA present), it was potentiated upon adding 2mm Mg2+. The K+ efflux from ATP-loaded ghosts was not altered by dithio-bis-nitrobenzoic acid (10mm) or acridine orange (100 m), while it was increased two-to fourfold by incubating with MgF2 (10mm), or MgF2 (10mm)+theophylline (40mm), respectively. By contrast, a marked efflux reduction was obtained by incorporating 0.5mm GTP into ATP-containing ghosts. The degree of phosphorylation obtained by incubating membranes with (-32P)ATP under various conditions affecting K+ channel activity, was in direct correspondence to their effect on K+ efflux. The results suggest that the K+ channel of red cells is under complex metabolic control, via cAMP-mediated and nonmediated mechanisms, some which require ATP and presumably, involve phosphorylation of the channel proteins.  相似文献

Summary Using the patch-clamp technique we have identified a Ca2+-sensitive, voltage-dependent, maxi-K+ channel on the basolateral surface of rat pancreatic duct cells. The channel had a conductance of 200 pS in excised patches bathed in symmetrical 150mm K+, and was blocked by 1mm Ba2+. Channel openstate probability (P o ) on unstimulated cells was very low, but was markedly increased by exposing the cells to secretin, dibutyryl cyclic AMP, forskolin or isobutylmethylxanthine. Stimulation also shifted theP o /voltage relationship towards hyperpolarizing potentials, but channel conductance was unchanged. If patches were excised from stimulated cells into the inside-out configuration,P o remained high, and was not markedly reduced by lowering bath (cytoplasmic) Ca2+ concentration from 2mm to 0.1 m. However, activated channels were still blocked by 1mm Ba2+. ChannelP o was also increased by exposing the cytoplasmic face of excised patches to the purified catalytic subunit of cyclic AMP-dependent protein kinase., We conclude that cyclic AMP-dependent phosphorylation can activate maxi-K+ channels on pancreatic duct cells via a stable modification of the channel protein itself, or a closely associated regulatory subunit, and that phosphorylation alters the responsiveness of the channels to Ca2+. Physiologically, these K+ channels may contribute to the basolateral K+ conductance of the duct cell and, by providing a pathway for current flow across the basolateral membrane, play an important role in pancreatic bicarbonate secretion.  相似文献

Summary A simple procedure was developed for the isolation of a sarcolemma-enriched membrane preparation from homogenates of bullfrog (Rana catesbeiana) heart. Crude microsomes obtained by differential centrifugation were fractionated in Hypaque density gradients. The fraction enriched in surface membrane markers consisted of 87% tightly sealed vesicles. The uptake of86Rb+ by the preparation was measured in the presence of an opposing K+ gradient using a rapid ion exchange technique. At low extravesicular Rb+ concentrations, at least 50% of the uptake was blocked by addition of 1mm ouabain to the assay medium. Orthovanadate (50 m), ADP (2.5mm), or Mg (1mm) were also partial inhibitors of Rb+ uptake under these conditions, and produced a complete block of Rb+ influx in the presence of 1mm ouabain. When86Rb+ was used as a tracer of extravesicular K+ (Rb 0 + 40 m K 0 + =0.1–5mm) a distinct uptake pathway emerged, as detected by its inhibition by 1mm Ba2+ (K 0.5=20 m). At a constant internal K+ concentration (K in + =50mm) the magnitude of the Ba2+-sensitive K+ uptake was found to depend on K 0 + in a manner that closely resembles the K+ concentration dependence of the background K+ conductance (I Kl) observed electrophysiologically in intact cardiac cells. We conclude that K+ permeates passively this preparation through two distinct pathways, the sodium pump and a system identifiable as the background potassium channel.  相似文献

Summary K+ channels in inside-out patches from hamster insulin tumor (HIT) cells were studied using the patch-clamp technique. HIT cells provide a convenient system for the study of ion channels and insulin secretion. They are easy to culture, form gigaohm seals readily and secrete insulin in response to glucose. The properties of the cells changed with the passage number. For cell passage numbers 48 to 56, five different K+-selective channels ranging from 15 to 211 pS in symmetrical 140mm KCl solutions were distinguished. The channels were characterized by the following features: a channel with a conductance (in symmetrical 140mm KCl solutions) of 210 pS that was activated by noncyclic purine nucleotides and closed by H+ ions (pH=6.8); a 211 pS channel that was Ca2+-activated and voltage dependent; a 185 pS channel that was blocked by TEA but was insensitive to quinine or nucleotides; a 130 pS channel that was activated by membrane hyperpolarization; and a small conductance (15 pS) channel that was not obviously affected by any manipulation. As determined by radioimmunoassay, cells from passage number 56 secreted 917±128 ng/mg cell protein/48 hr of insulin. In contrast, cells from passage number 77 revealed either no channel activity or an occasional nonselective channel, and secreted only 29.4±8.5 ng/mg cell protein/48 hr of insulin. The nonselective channel found in the passage 77 cells had a conductance of 25 pS in symmetrical 140mm KCl solutions. Thus, there appears to be a correlation between the presence of functional K+ channels and insulin secretion.  相似文献

Summary The tight-seal whole-cell recording method has been used to studyNecturus choroid plexus epithelium. A cell potential of –59±2 mV and a whole cell resistance of 56±6 M were measured using this technique. Application of depolarizing step potentials activated voltage-dependent outward currents that developed with time. For example, when the cell was bathed in 110mm NaCl Ringer solution and the interior of the cell contained a solution of 110mm KCl and 5nm Ca2+, stepping the membrane potential from a holding value of –50 to –10 mV evoked outward currents which, after a delay of greater than 50 msec, increased to a steady state in 500 msec. The voltage dependence of the delayed currents suggests that they may be currents through Ca2+-activated K_ channels. Based on the voltage dependence of the activation of Ca2+-activated K+ channels, we have devised a general method to isolate the delayed currents. The delayed currents were highly selective for K+ as their reversal potential at different K+ concentration gradients followed the Nernst potential for K+. These currents were reduced by the addition of TEA+ to the bath solution and were eliminated when Cs+ or Na+ replaced intracellular K+. Increasing the membrane potential to more positive values decreased both the delay and the half-times (t 1/2) to the steady value. Increasing the pipette Ca2+ also decreased the delay and decreasedt 1/2. For instance, when pipette Ca2+ was increased from 5 to 500nm, the delay andt 1/2 decreased from values greater than 50 and 150 msec to values less than 10 and 50 msec. We conclude that the delayed currents are K+ currents through Ca2+-activated K+ channels.At the resting membrane potential of –60 mV, Ca2+-activated K+ channels contribute between 13 to 25% of the total conductance of the cell. The contribution of these channels to cell conductance nearly doubles with membrane depolarization of 20–30 mV. Summary The apical membrane K+ permeability of the newt proximal tubular cells was examined in the doubly perfused isolated kidney by measuring the apical membrane potential change (V a change) during alteration of luminal K+ concentration and resultant voltage deflections caused by current pulse injection into the lumen.V a change/decade for K+ was 50 mV at K+ concentration higher than 25mm, and the resistance of the apical membrane decreased bt 58% of control when luminal K+ concentration was increased from 2.5 to 25mm. Ba2+ (1mm in the lumen) reducedV a change/decade to 24 mV and increased the apical membrane resistance by 70%. These data support the view that Ba2+-sensitive K+ conductance exists in the apical membrane of the newt proximal tubule. Furthermore, intracellular K+ activity measured by K+-selective electrode was 82.4 ± 3.6 meq/liter, which was higher than that predicted from the Nernst equation for K+ across both cell membranes. Thus, it is concluded that cell K+ passively diffuses, at least in part, through the K+ conductive pathway of the apical membrane.  相似文献

Human chorionic gonadotropin (hCG) is a key autocrine/paracrine regulator of placental syncytiotrophoblast, the transport epithelium of the human placenta. Syncytiotrophoblast hCG secretion is modulated by the partial pressure of oxygen (pO2), reactive oxygen species (ROS) and potassium (K+) channels. Here we test the hypothesis that K+ channels mediate the effects of pO2 and ROS on hCG secretion. Placental villous explants from normal term pregnancies were cultured for 6 days at 6% (normoxia), 21% (hyperoxia) or 1% (hypoxia) pO2. On days 3–5, explants were treated with 5mM 4-aminopyridine (4-AP) or tetraethylammonium (TEA), blockers of pO2-sensitive voltage-gated K+ (KV) channels, or ROS (10–1000μM H2O2). hCG secretion and lactate dehydrogenase (LDH) release, a marker of necrosis, were determined daily. At day 6, hCG and LDH were measured in tissue lysate and 86Rb (K+) efflux assessed to estimate syncytiotrophoblast K+ permeability. hCG secretion and 86Rb efflux were significantly greater in explants maintained in 21% pO2 than normoxia. 4-AP/TEA inhibited hCG secretion to a greater extent at 21% than 6% and 1% pO2, and reduced 86Rb efflux at 21% but not 6% pO2. LDH release and tissue LDH/hCG were similar in 6%, 21% and 1% pO2 and unaffected by 4-AP/TEA. H2O2 stimulated 86Rb efflux and hCG secretion at normoxia but decreased 86Rb efflux, without affecting hCG secretion, at 21% pO2. 4-AP/TEA-sensitive K+ channels participate in pO2-sensitive hCG secretion from syncytiotrophoblast. ROS effects on both hCG secretion and 86Rb efflux are pO2-dependent but causal links between the two remain to be established.  相似文献
