Cyclamen persicum Mill, seeds germinate in a narrow range oftemperature and germination is strongly inhibited by continuousirradiation with white light. The thermal optimum is approx.15 °C in both darkness and light. Seed germination is alsovery sensitive to oxygen deprivation and this sensitivity ismore pronounced at 20 °C than at the optimum 15 °C.Very immature seeds cannot germinate at any temperature, butgerminability increases during seed maturation Seedling development is unusual since seed reserves are usedimmediately for tuber formation. Effects of dehydration, storage temperature and humidificationon germination of Salix alba andS. matsudana seeds were studied.Newly released seeds showed 100% germination before and afterdehydration to 11–12% moisture content. Germination ofthe high vigour lot (100% initial normal germination) was notaffected by dehydration to 6.7% moisture content but germinationdecreased with further dehydration to 4.3%. The lower vigourlot (75% initial normal germination) was more susceptible todehydration and germination decreased following dehydrationto 6.7% moisture content. Dry seeds of both species survivedimmersion in liquid nitrogen without loss of viability. Thegermination of seeds stored with 9% moisture content decreasedto 35–40% in 5 months at -20°C or in 2 months at 5°C.However, at 25°C seeds entirely lost viability within 2weeks. Seeds showed improved performance when stored at -70°C> - 20°C > 5°C > 25°C and tolerated dehydrationto a moisture content in equilibrium with 15% relative humidity.Results suggest that they are orthodox in storage behaviouralthough they are short-lived. Humidification treatment of lowvigour seed lots resulted in a remarkable increase in germinationpercentage. Studies were made on bambarranut seeds (Voandzeia subterraneaThouars) after 0, 6, 12, 18 and 24 months of storage in gunnybags under laboratory conditions (25–35 °C). Seeddeterioration during storage was indicated by delayed germination,reduced germinability, reduced growth of seedlings and increasednumber of stunted seedlings culminating in a total failure ofgermination after two years. Slight depletion of food reserves occurred during seed storage.The loss in fat was higher than starch or protein. Total solublesugars decreased while the content of total fatty acids andamino acids and soluble protein increased. Total nitrogen (N)remained unaffected while soluble-N and amino-N increased. Allthese components showed a rapid change (increase or decrease)from 12 months to 18 months of storage which was associatedwith commencement of rapid decline in germinability of the seedsand growth of the seedlings. Initial rapid imbibition of water was observed in viable aswell as non-viable seeds, though at a higher rate in the latterand followed by a lag period in both. Seed conditioning and germination in witchweed (Striga asiatica(L.) Kuntze) were temperature-dependent. With higher conditioningtemperatures, shorter conditioning time was required for germinationwith terminal dl-strigol (strigol) treatment at 30 °C. Maximumgermination (80–100%) was obtained by conditioning inwater at 20, 25, 30 and 35 °C for 14, 7, 5 and 3 d, respectively,and terminally treating with 10–6 M strigol at 30 °C.Seeds conditioned in 10–8 M strigol instead of water germinatedmuch less with the same terminal strigol treatment. Generally,conditioning was slower when seeds were conditioned in strigolrather than water. The reduction in germination rate by pretreatmentin strigol or pretreatment at low temperatures could be overcomeby increasing the terminal strigol concentration in the germinationtest. Conditioned seeds did not germinate at 10 and 15 °Cwith a terminal 10–6 M strigol treatment but yielded closeto maximum germination at 25, 30 and 35 °C with the sameterminal strigol treatment. To obtain maximum germination, boththe minimum conditioning temperature and the minimum germinationtemperature for conditioned seeds were 20 °C. Photoinhibition of Seed Germination in Mediterranean Maritime Plants   总被引:4,自引:1,他引:3  
Photoinhibition of seed germination was shown for Allium staticiforme,Brassica tournefortii, Cakile maritima and Otanthus maritimus,all plant species inhabiting sandy coasts of the MediterraneanSea. Germination of A. staticiforme was found to be typicallyMediterranean in regard to its temperature range (>0–20°C), while B. tournefortii germinated optimally at intermediatetemperatures (15–25 °C). Light sensitivity was morepronounced in the latter species and 50 % inhibition of seedgermination was obtained with photon flux densities of approximately0.1 and 0.015 mol m–2 d–1 in A. staticiforme andB. tournefortii, respectively. However, the slopes of the regressionlines of germination plotted against the logarithm of whitelight flux density are similar in both species. From monthlyexperiments performed under fluctuating conditions of temperatureand light, simulating the elimate of Athens throughout the year,optimal germination response peaked in ‘winter’conditions for A. staticiforme, while a bimodal pattern wasobserved for B. tournefortii (maxima at the warm ends of therainy season). In contrast to the other plants, a fifth speciestested, Crithmum maritimum, showed an absolute light requirement,probably related to the rocky habitats of the species; the optimumgermination period coincided with the rainy season. Pot experimentswith seeds of the five species buried at various depths confirmedthat maximum emergence is favoured by shallow depths (0.5–1cm). It is concluded that seed germination in maritime plantsof the Mediterranean rim is mediated by a photoinhibition mechanismwhich can be considered an adaptation strategy against surfaceseedling establishment at the harsh, sandy or shingle, sea coasthabitats. Effects of temperature, light, NaCl and polyethylene glycol(PEG)-6000 on seed germination and radicle growth in a halophyticshrub, Kalidium caspicum(L.) Ung.-Sternb. were investigated.When seeds were incubated in deionized water at constant temperaturesbetween 10 and 30°C, the percentage germination in the darkexceeded 75%; light suppressed seed germination at alternatingtemperatures. Incubating seeds with a hypersaline solution ofNaCl for 30 d had no adverse effect on their germinability.The percentage germination of seeds incubated with a –0.8MPa NaCl solution was 73, 80 and 54% at 10, 20 and 30°C,respectively, but all radicles died before their length exceeded5 mm. In contrast, when seeds were incubated with a –0.8MPa PEG solution at 20°C, 68% of seeds germinated, and 95%of the emerging radicles survived beyond 5 mm. Biflavonoids from the leaves of Wollemia nobilis (Wollemi Pine) have been investigated and compared with data on other members of the family Araucariaceae. Based on 1H and 13C NMR as well as High Resolution Mass Spectrometry (HRMS) the structures of seven isolated biflavonoids were fully determined. Although all of them were previously characterised especially at Agathis and in lesser degree at Araucaria genera this is the first report on their occurrence at Wollemi Pine, one of the world's oldest and rarest plants.  相似文献

Immature Glycine max (L.) Merr. seeds initially at 50–70mg fresh weight were successfully grown and matured in vitroin detached pods. Surface sterilized pods were floated in aliquid medium containing 5 per cent sucrose, minerals, and glutaminein 125 ml Erlenmeyer flasks and incubated at 25 °C under350–400 µE m–1 s–1 white light. Seedswhich were matured in vitro increased tenfold in dry weight,were visually similar to commercial seeds of the same size,were tolerant to desiccation and germinated with normal seedlinggrowth. Excised pods transported dye from the pedicel to thegrowing seed within 120 min. Soya bean pod culture is a usefultechnique to study the influence of single or combinations ofchemical or environmental parameters on regulation of seed growth,seed maturation, and subsequent germination events without theconfounding interactions with the mother plant. Seeds of two lettuce cultivars (Lactuca sativa L., cv. Meikoninginand cv. Grand Rapids) were hermetically stored with constantmoisture contents ranging between 3.6 and 17.9 per cent (freshweight basis) at constant temperatures ranging between 5 and75 °C. The decline with time in percentage germination andpercentage normal seedlings was determined for each storagetreatment. The data were fitted to an equation which containsthe constants: K1, the probit of the initial percentage germinationor normal seedlings; KE, a species constant; CW, the constantof a logarithmic moisture term; CH, the constant of a lineartemperature term and CQ, the constant of a quadratic temperatureterm. Regression analysis of data from storage periods up to5.5 years at temperatures of 5–75 °C and seed moisturecontents of 3.6–13.6 per cent yielded the following values:KE= 8.218, CW=4.797±0.163, CH=0.0489±0.0050 andCQ=0.000365±0.000056. Although this equation consistentlyprovided a better fit, simplified equations, assuming eithera log-linear relationship between seed longevity and temperature,or a log-linear relationship between seed longevity and bothmoisture content and temperature, accounted for more than 94per cent of the variation at the restricted temperature rangeof 5–40 °C. Longevity of the same seed lots at sub-zero temperatures (–5,–10 and –20 °C) was studied in separate tests.Freezing damage, resulting in abnormal seedlings in the germinationtest, occurred at –20 °C when the moisture contentof the seeds exceeded 12 per cent. No decline in percentagenormal seedlings was observed after a storage period of 18 monthsor longer at –20 °C, provided the seed moisture contentdid not exceed 9.5 per cent. For seeds stored at –5 and–10 °C with 9.6–12.5 per cent moisture content,the observed rate of decline of percentage normal seedlingswas adequately predicted by the viability equation, using theabove values for the constants. This suggests that for low moisturecontents the viability equation can be applied to estimate longevityat sub-zero temperatures. This paper highlights recent advances and improved scientific understanding of conservation technologies through selected case studies on threatened plant species indigenous to Eastern Australia. This includes investigations into seed desiccation, storage responses and cryopreservation in rainforest species, particularly the socio-economically important Australian native Citrus spp., Davidsonia spp. (Davidson’s plum) and Syzygium spp. This work also (1) increases our understanding of ecological correlates of seed desiccation sensitivity for predictive use and (2) improves restoration practice through better understanding of seed storage and germination requirements. The use of in vitro conservation technologies in support of conservation actions for endangered species is outlined in case studies on Wollemia nobilis (Wollemi pine), epiphytic and terrestrial orchid species, and an endangered fern species.  相似文献

Various photon doses (net number of photons per unit area perday), provided by varying both photon flux density and photoperiod,were applied to imbibing seeds of seven lots of four speciesof Compositae in various germination test regimes. In all fourspecies germination was dependent upon photon dose, the productof photon flux density and daily duration of exposure. The responsewas quantified by linear relations between the probit of percentagegermination and the logarithm of photon dose. In general, photonflux density and photoperiod only influenced the stimulationof germination by the low energy reaction indirectly (as factorsof daily photon dose), whereas there was a tendency for photoperiodto have a direct influence on the inhibition of germinationby the high irradiance reaction. Reducing the germination testtemperature from 25?C to 20?C and 15?C not only increased thedark germination of L. sativa L., but also broadened the photondose range at which full germination occurred by reducing theminimum value necessary for the germination of the most dormantseeds, and increasing the maximum value which failed to inhibitthe germination of any seeds. Differences between L. sativaand L. serriola L. in the response of germination to white lightwere only quantitative, rather than qualitative. The singlemost promotory dose for all four species was 3 ? 10–3mol m–2 d–1, although the inhibitory action of dosesup to 10– mol m– d– was generally only slight. Moisture Content and the Longevity of Seeds of Phaseolus vulgaris   总被引:3,自引:0,他引:3  
The lower limit to the negative logarithmic relation betweenseed longevity and moisture was determined in bean (Phaseolusvulgaris L.). Sub-lots of seed were hermetically stored at 65°C and 11 moisture contents (3.26–13.6% f. wt) forup to 80 d, tested for germination and the seedlings evaluated.In accordance with the seed viability equation, there was anegative logarithmic relation between moisture content (%, f.wt) and longevity. Two different criteria for estimating theslope constant of this relation gave similar values of 4.76(s.e. 0.26) and 4.82 (s.e. 0.24). The calculated lower moisturecontent limits to the relation were 5.7 and 5.6%, respectively,values at equilibrium with 10.6–10.8% relative humidity(rh). Further drying to 3.26% had little additional effect onlongevity, but initial germination was slightly reduced. Theresults are discussed in relation to water potential and comparedwith other crops. The effects of storage conditions on the germination of developingmuskmelon (Cucumis melo L.) seeds were tested to determine whetherafter-ripening is required to obtain maximum seed vigour. Seedswere harvested at 5 d intervals from 35 (immature) to 60 (fullymature) days after anthesis (DAA), washed, dried, and storedat water contents of 3·3 to 19% (dry weight basis) at6, 20, or 30°C for up to one year. Germination was testedin water and in polyethylene glycol 8000 solutions ( –0·2to –1·2 MPa osmotic potential) at 15, 20, 25 or30°C. Germination percentages and rates (inverse of meantimes to radicle emergence) were compared to those of newlyharvested, washed and dried seeds. For 40 and 60 DAA seeds,one year of storage at 20°C and water contents <6·5%significantly increased germination percentages and rates at20°C, but had little effect on germination at 25 and 30°C.Storage reduced the estimated base temperature (Tb) and meanbase water potential (b) for germination of both 40 and 60 DAAseeds by approximately 5°C and 0·3 MPa, respectively.Immature 35 DAA seeds showed the greatest benefit from storageat 3 to 5% water content and 30°C, as germination percentagesand rates increased at all water potentials (). Storage underthese same conditions had little effect on the germination ofmature seeds in water, but increased germination percentagesand rates at reduced 's. Accelerated ageing for one month at30°C and water contents from 15 to 19° increased germinationrates and percentages of mature seeds at reduced 's, but longerdurations resulted in sharp declines in both parameters. Immatureseeds lost viability within one month under accelerated ageingconditions. An after-ripening period is required at all stagesof muskmelon seed development to expand the temperature andwater potential ranges allowing germination and to achieve maximumgerminability and vigour. Post-harvest dormancy is deepest atthe point of maximum seed dry weight accumulation and declinesthereafter, both in situ within the ripening fruit and duringdry storage. The influence of prehydration in water or priming in –1.5 MPa polyethylene glycol 8000 solution for various periods,followed by redrying, on germination rate and longevity of lettuce(Lactuca sativa L.) seeds (achenes) was determined during controlleddeterioration at 10% moisture content (fresh weight basis) and40°C. Short prehydration treatments (up to 1 h) had littleeffect on either germination rate or longevity, but significantlyimproved root growth rates. Increasing durations of prehydrationor priming reduced the mean time to germination by up to 61%relative to untreated seeds, but also reduced mean seed longevityby as much as 84% Prehydration and priming altered the relationshipsbetween germination rate and viability and between normal andabnormal seedlings during ageing. Prehydration in abscisic acidor at a temperature inhibitory to germination did not preventthe reduction in longevity under controlled deterioration conditions.While prehydration or priming treatments effectively acceleratesubsequent germination rates of lettuce seeds, the redried seedsare nonetheless highly susceptible to deterioration in storage. The effects of three temperature regimes, 25°C day/20°Cnight, 20°C day/15°C night and 15°C day/10°Cnight upon seed development in S.22 Italian ryegrass, S.24 perennialryegrass, S.215 meadow fescue and a Swiss ecotype of meadowfescue were investigated. The largest ovaries and largest seedswere obtained from the 15/10°C environment and floret fertilitywas greatest at 20/15°C. The largest seed weight per inflorescencewas at 20/15°C, except for the Swiss ecotype of meadow fescuewhere it was greatest at 15/10°C. The percentage germinationwas highest from those seeds developed at 25/20°C and thiswas particularly marked at germination temperatures of 13–20°C.There was a low rate of germination of seeds from the 15/10°Ctreatment, but they produced seedlings with the largest dryweights. It is concluded that year to year temperature differenceswill affect the yield and quality of seed crops of forage grasses.  相似文献

Background and Aims: The branch-base xylem structure of the endangered Wollemia nobiliswas anatomically investigated. Wollemia nobilis is probablythe only extant tree species that produces only first-orderbranches and where all branches are cleanly abscised. An investigationwas carried out to see if these unusual features might influencebranch-base xylem structure and water supply to the foliage. Methods: The xylem was sectioned at various distances along the branchbases of 6-year-old saplings. Huber values and relative theoreticalhydraulic conductivities were calculated for various regionsof the branch base. Key Results: The most proximal branch base featured a pronounced xylem constriction.The constriction had only 14–31 % (average 21 %)of the cross-sectional area and 20–42 % (average28 %) of the theoretical hydraulic conductivity of themore distal branch xylem. Wollemia nobilis had extremely lowHuber values for a conifer. Conclusions: The branch-base xylem constriction would appear to facilitatebranch abscission, while the associated Huber values show thatW. nobilis supplies a relatively large leaf area through a relativelysmall diameter ‘pipe’. It is tempting to suggestthat the pronounced decline of W. nobilis in the Tertiary isrelated to its unusual branch-base structure but physiologicalstudies of whole plant conductance are still needed.  相似文献

Immature barley embryos (Hordeum distichum var. Julia) of between0•20 and 0•80 mm in length, were isolated from thedeveloping grain and cultured in vitro on various culture media.The subsequent development of the embryos was followed overa period of weeks, and where germination ensued the growth rateof shoot and root meristems was compared with in vivo germinationrates. Various growth media were assessed for their abilityto support normal development of immature embryos. A numberof published media failed to support satisfactory developmentof young embryos. The addition of 1–15 per cent coconutmilk to Norstog's Medium I (mineral + vitamin solns) enhancedembryo development and lowered the threshold of viability fromembryos of 0•50 mm in length to 0•35 mm. Althoughin many cases germination ensued, embryo development was largelyabnormal. A slightly greater enhancement of growth was achievedwith 0•05–0•30 per cent casein hydrolysate asthe growth medium supplement, although abnormal developmentwas not eliminated. A further lowering of the viability thresholdto include embryos of 0•25 mm in length was obtained bycombining 2•7 mM glutamine with the casein hydrolysatesupplement. Normal development and germination of embryos assmall as O25 mm was however obtained on Norstog's Medium JJand the results were reproduced in four additional if . distichumvarieties. In each case the critical threshold of viabilitywas found to lie in embryos of 0•20–0•30 mmin length.  相似文献

Over 50 per cent germination has been obtained from Elaeis guineensisform pisifera seeds stored in unaerated sterile distilled waterfor 6 months. The moisture levels of the seeds and excised embryoswere of the same order (20–30 and 60–70 per cent,respectively) as those of fully imbibed fresh seeds. The rapid and uniform establishment of soya bean [Glycine max(L.) Merr.] stands is conducive to higher yields. This studywas undertaken to determine the effects of cultivar, temperature,and seed size on the rate of germination and emergence. No cultivar effect on the germination rate was observed. However,in an emergence study from a sand-soil-peat mixture, cultivardifferences in emergence rates were noted(‘Chippewa 64’> ‘Wayne’ > ‘Amsoy 71’). In anotheremergence study (sand media) the cvs ‘Calland’ and‘Williams’ emerged faster than the cv. 'Wayne or‘Wells’. Time required for 50 per cent germination decreased (18.8–4.0days) as the temperature increased from 10 to 30 °C (5 °Cincrements). Emergence (50 per cent) from a sand-soil-peat mixturewas more rapid (19.8–6.3 days) as the simulated plantingdate (growth chamber set to simulate field temperatures) wasdelayed from 16 April to 15 June with an intermediate date of16 May. In addition, time required for 50 per cent emergence of thecultivars from sand decreased (793–76 h) as the temperaturewas increased from 10 to 30 °C with no decrease from 30to 35 °C. Seed size effects were apparent, with the very small seed germinatingslower than the three larger seed sizes. In the emergence studieswith both the sand and sand-soil-peat mixture there was a generaltrend toward more rapid emergence with the smaller seeds. However,the absolute differences were small. Significant cultivar x temperature interactions were observedfor the germination and emergence rates. In most cases the cultivarsmerged in terms of germination and emergence rates at temperaturesbetween 10 and 20 °C and at the higher temperatures thecultivar rankings were different from those observed at temperaturesbelow the merging point. Immature Glycine max (L.) Merrill seeds, initially between 50and 450 mg f. wt, were grown and matured successfully in vitro.Excised seeds were floated in a liquid medium containing 5 percent sucrose, minerals and glutamine in flasks incubated at25 °C under 300 to 350 µE m–2 s–1 fluorescentlight. During 16 to 21 d in culture, seeds grew to a matured. wt of 100 to 600 mg per seed at an average rate of 5 to 25mg d. wt per seed d–1 depending on initial size. Growthrates were maximal during the first 8 to 10 d in vitro but declinedwith loss of green colour in the cotyledons. Seed coats rupturedwith rapid cotyledon expansion during the first 2 d in culture.Embryos were tolerant to desiccation and 80 to 90 per cent germinatedif removed from culture before complete loss of green colour.The growth of excised seeds in vitro exceeded the growth ofseeds in detached pods, but when windows were cut in pods topermit direct exposure of seeds to the medium, seed growth wascomparable.
