Here we report the biosynthetic pathway for the neoantimycin and present three novel neoantimycin analogues, neoantimycin D (1), E (2) and F (3), from this assembly system from Streptoverticillium orinoci. Identification of these novel neoantimycin variants was achieved by selective MS/MS interrogation of natural product extracts using diagnostic fragments of the known neoantimycins. Their structures, including the absolute configurations, were elucidated using a combination of NMR experiments, detailed MS/MS experiments and the advanced Marfey’s method. The biosynthetic pathway of neoantimycin was dissected by genome sequencing data analysis for the first time, which includes a hybrid nonribosomal peptide synthetase (NRPS) and polyketide synthetase (PKS) assembly lines.  相似文献

Hepatocytes were isolated at specified times from livers of diabetic and insulin-treated diabetic rats during the course of a 48-h refeeding of a fat-free diet to previously fasted rats. The rates of synthesis of fatty acid synthetase and acetyl-CoA carboxylase in the isolated cells were determined as a function of time of refeeding by a 2-h incubation with l-[U-14C]leucine. Immunochemical methods were employed to determine the amount of radioactivity in the fatty acid synthetase and acetyl-CoA carboxylase proteins. The amount of radioactivity in the fatty acid synthetase synthesized by the isolated cells was also determined following enzyme purification of the enzyme to homogeneity. Enzyme activities of the fatty acid synthetase and acetyl-CoA carboxylase in the cells were measured by standard procedures. The results show that isolated liver cells obtained from insulintreated diabetic rats retain the capacity to synthesize fatty acid synthetase and acetyl-CoA carboxylase. The rate of synthesis of the fatty acid synthetase in the isolated cells was similar to the rate found in normal refed animals in in vivo experiments [Craig et al. (1972) Arch. Biochem. Biophys. 152, 619–630; Lakshmanan et al. (1972) Proc. Nat. Acad. Sci. USA69, 3516–3519]. In addition the relative rate of synthesis of fatty acid synthetase was stimulated greater than 20-fold in the diabetic animals treated with insulin. Immunochemical assays, when compared with enzyme activities, indicated the presence of an immunologically reactive, but enzymatically inactive, form or “apoenzyme” for both the fatty acid synthetase and acetyl-CoA carboxylase. The synthesis of these immunoreactive and enzymatically inactive species of protein, as well as the synthesis of the “holoenzyme” forms of both enzymes, requires insulin.  相似文献

Phytochemical investigation on the fruiting bodies of Ganoderma resinaceum led to the isolation of five new meroterpenoids, namely ganoresinains A–E (15), and four known analogues (69). The new compounds were identified by extensive analyses of spectroscopic data (NMR, MS, UV, and IR) and comparison with the literature data. Compound 1 and 6 were isolated as enantiomeric mixture, which were separated over analytical chiral HPLC chromatography. Compounds 6–9 were isolated from G. resinaceum for the first time.  相似文献

Three new phenanthraquinones (13), together with seven known phenanthraquinone derivatives (410) were isolated from Heleocharis dulcis for the first time. Their structures were established on extensive NMR, MS, UV, IR experiments in conjunction with their references. Compounds (110) were evaluated for their hypolipidemic activities for the first time. Compounds 1, 3, 7, and 9 (10 μM) exhibited significant hypolipidemic activities by measuring the triglyceride content in HepG2 cells with simvastatin as positive control. The chemotaxonomic significance of compounds (110) was also discussed.  相似文献

Sixteen compounds were isolated from the whole herbs of Peperomia tetraphylla (Forst. F.) Hooker et Arnott by phytochemical methods, including eight flavonoids (1–3, 6, 7, 1416), three lignans (8–10), three beta sitosterols (4, 5, 11), and two phenolic acids (12, 13). Their structures were identified by the analysis of NMR and MS, as well as the comparisons to the reported data. Among them, 2″-O-xylosylisoswertisin (14) was firstly isolated from the Piperaceae family, as well as ten compounds (1–4, 7, 10–11, 13, 15–16) were isolated from P. tytraphylla for the first time. Moreover, the chemotaxonomic significance of constituents isolated from P. tytraphylla was also discussed.  相似文献

A Xylaria sp. was isolated as an endophyte from a surface-sterilized Concord grape leaf (Vitis labrusca). Dereplication and LC–MS analysis of the filtrate extracts identified two new structures, compound 1, 13-O-methyl-(5R) diplosporin, and 2 a new agistatine. Additionally, five known compounds 37 including an agistatine, coriloxin, (5R)-diplosporin, and two mellein derivatives were also isolated. This is the first report of these structurally-related metabolites isolated from the same fungus, suggesting they share a related biosynthesis. Metabolites were isolated using liquid chromatography-solid phase extraction (LC-SPE) and semi-preparative HPLC. Structures were elucidated on the basis of 1D and 2D NMR analysis, HRMS measurements and X-ray structural analysis for compound 1.  相似文献

Three known (13) and a novel (4) monoterpene indole alkaloids have been isolated from the methanol extract of leaves of Tabernaemontana elegans and their structures were elucidated by a series of spectroscopic experiments, involving NMR, MS, UV, and IR techniques. The isolated monoterpene indole alkaloids along with previously described β-carbolines (57) from the same specimen were studied for their apoptosis induction activity in human hepatoma HuH-7 cells. Methodology for apoptosis induction studies included cell viability assays, nuclear morphology assessments, and general caspase-3-like activity assays. The monoterpene indole alkaloids, tabernaemontanine (1) and vobasine (3) showed the most promising apoptosis induction profile in HuH-7 cells.  相似文献

A sensitive and selective assay for the determination of N-{1(R)-[(1,2-dihydro-1-methylsulfonylspiro[3H-indole-3,4′-piperidin]-1′-yl)carbonyl]-2-(phenylmethoxy)-ethyl}-2-hydroxyamino-2-methylpropanamide (I), a hydroxyl amine metabolite of a novel growth hormone secretagouge (II) has been developed utilizing high-performance liquid chromatography with ion spray tandem mass spectrometric detection (HPLC–MS–MS). The analyte and an internal standard (III) were isolated from the basified biological matrix using a liquid–liquid extraction with methyl tert.-butyl ether (MTBE). The organic extract was evaporated to dryness at room temperature. The residue was reconstituted in the mobile phase and injected into the HPLC–MS–MS system. Multiple reaction monitoring using the precursor→product ion combinations of m/z 545→267 and 543 →267 was used to quantify I and III, respectively, after chromatographic separation under isocratic conditions. The assay was validated in the concentration range of 0.5 to 500 ng/0.1 ml in both human and dog plasma. The precision of the assay, expressed as relative standard deviation, was less than 10% over the entire concentration range with the exception of the low concentration of 0.5 ng/0.1 ml which was 14.0% for human plasma. The HPLC–MS–MS method provided sufficient sensitivity to completely map the pharmacokinetic time course of I following a single 5 mg dose of II to human subjects and a 0.5 mg/kg dose to beagle dogs.  相似文献

Two new labdane diterpenoids cinereanoid C (1), cinereanoid D (2), a new β-lactam, cinerealactam E (3) as well as six known flavonoid glycosides (4–9) were isolated from the aerial parts of Roylea cinerea (Lamiaceae). The structures of (1–9) were all determined by MS, IR and NMR spectroscopy. The structure of cinereanoid D (2) was further proven by single crystal X-ray diffraction. Six known flavonoid glycosides (4–9) were also isolated for the first time from this plant. 2, 5, 6 and 7 were found to significantly inhibit the ATP binding of a tumour growth-promoting heat shock protein, Hsp90.  相似文献

An effective screening method for inhibitors of NO production in natural products using LC-QTOF MS/MS coupled with a cell-based assay was proposed. The ethyl acetate fraction of Catalpa ovata exhibited a strong inhibitory effect on NO production in lipopolysaccharide-induced BV2 microglia cells. We attempted to identify the active constituents of C. ovata by using LC-QTOF MS/MS coupled with a cell-based assay. Peaks at approximately 14–15 min on the MS chromatogram were estimated to be the bioactive constituents. A new iridoid compound, 6-O-trans-feruloyl-3β-hydroxy-7-deoxyrehamaglutin A (4), and nine known compounds (13, 510) were isolated from the ethyl acetate fraction of C. ovata by repeated column chromatography. Compounds 3, 4, 5, 7, and 8 significantly attenuated lipopolysaccharide-stimulated NO production in BV2 cells. Our results indicate that LC-QTOF MS/MS coupled with a cell-based NO production inhibitory assay successfully predicted active compounds without a time-consuming isolation process.  相似文献

Chemical investigation on the roots of Chloranthus serratus (Chloranthaceae) afforded 11 terpenoids, including four lindenane-type sesquiterpenoid dimers (14), and seven labdane diterpenoids (511). Two new eremophilane sesquiterpenes, compounds 1 and 2, were isolated from the endophytic fungus Microdiplodia sp. KS 75-1, together with the known compounds phomadecalins C (3) and D (4). Their structures were determined by extensive 1D– and 2D–NMR and MS spectral analyses. The previously reported stereochemistry at C-8 of 3 and 4 were revised on the basis of NOEs experiments. The phytochemical investigation of stems of Fissistigma glaucescens (Hance) Merr. led to the isolation of a new aristolactam derivative aristolactam AIb (1), together with eleven known aristolactam derivatives (212). Structure elucidation of these compounds were performed on the basis of NMR spectral data and MS. Compounds 27 were isolated from the F. glaucescens for the first time. Three new compounds, 17β-cevanin-6-oxo-5α,20β-diol yibeinine (1), 2-(tetrahydro-5-(2-hydroxyphenyl)-2H-pyran-3-yl) phenol (2), 1,3-O-diferuloyl-2-methoxypropane diol (3), as well as four known compounds (47), have been isolated from the ethanol extract of dried bulbs of Fritillaria pallidiflora Schrenk. All structures were determined based on their spectroscopic data (1D and 2D NMR (including 1H NMR, 13C NMR, 1H-1H COSY, HMBC, HSQC, HSQC-TOCSY, and NOESY experiments), and MS). Biological evaluation showed that compounds 14 inhibited the production of nitric oxide (NO) in LPS-stimulated RAW 264.7 cells with IC50 values of 18.0, 38.7, 29.5, and 47.1 μM, respectively. Five new cis-ent-clerodanes, linarenones A–E (15) and two known compounds (6 and 7) were isolated from whole plant of Linaria japonica. The structures of these compounds were determined by various spectroscopic analyses (UV, IR, HR–ESI–MS, 1D and 2D NMR). The absolute configuration of five new diterpenoids was confirmed by circular dichroism (CD) analysis and chemical conversion. Cytotoxicity of the isolated compounds against A549 cell lines and Leishmania major were evaluated. The new cis-ent-clerodane 3 was found to be moderately active against A549 cell lines, and new cis-ent-clerodanes 1, 6 and desacetyl-linarienone (7) were active against L. major.  相似文献

The chemical investigation of whole plants Piper boehmeriifolium (Miq.) Wall. ex C. DC. led to the isolation of 22 compounds, including two lignans (12), sixteen amide alkaloids (318), one diterpene (19), two monoterpenes (2021), and one phenylpropanoid (22). Their structures were elucidated by extensive spectroscopic analyses including NMR, MS, and by comparison with the literature. Compounds 12, 67, 1112, 14, and 1722 were firstly isolated from P. boehmeriifolium, while compounds 2, and 1920 were isolated from Piper genus for the first time.
