Caveolae are specialized lipid microdomains, forming small invaginations in the plasma membrane, known to be implicated in multiple functions including lipid storage, cell signaling and endocytosis. Formation of these wide flask-shaped invaginations is dependent on the expression of a caveolar coat protein, namely caveolin. Until now, the accepted paradigm was that caveolin was the sole and only structural protein of caveolae since its expression was necessary and sufficient to drive caveolae biogenesis. The recent characterizations of PTRF/cavin-1 and subsequently other cavin family members in caveolae formation have highlighted additional levels of complexity in the biogenesis of these plasma membrane invaginations. In this review, recent advances on the role of the different cavin family members in the regulation of caveolae structures as well as potential new functions will be discussed.  相似文献

Trichodorid nematodes (Nematoda: Trichodoridae) are vectors of tobacco rattle virus (TRV), one of the causal agents of spraing disease in potato. Root bacteria from nematicidal plants and their control potential against Trichodoridae were the focus of this study. Bacteria isolated from the roots of 12 nematicidal plants and potato were characterized for their production of hydrolytic enzymes, hydrogen cyanide, phenol oxidation ability and antifungal activity towards the potato pathogen Rhizoctonia solani. Based on these functional traits, bacteria isolates were selected and tested in greenhouse conditions on potato (cv. Saturna) for their effect on plant growth, and screened for nematicidal activity against Paratrichodorus pachydermus and Trichodorus primitivus in naturally infested soil. Sixteen bacteria isolates out of 44 reduced nematode densities by 50–100%. Nine selected isolated were further tested by bacterizing potato tubers (cv. King Edward) which were planted in a trichodorid and TRV-infested soil. Four bacterial isolates consistently reduced nematode densities (by 56.7–74.4%) with no visual negative effect on plant growth. These isolates were tentatively identified, partly by fatty acid methyl ester (FAME) analysis as: Stenotrophomonas maltophilia, Bacillus mycoides, Pseudomonas sp., and one unidentified bacterium. The isolates originated from potato, Plantago major, Thymus vulgaris and Asparagus officinalis, respectively. Two Pseudomonas isolates obtained from Zinnia elegans and selected for their strong nematicidal activity in soil screening tests, did not reduce the nematode population when tested on potato. It is concluded that plants releasing nematicidal compounds may harbour nematode-antagonistic bacteria as well.  相似文献

Summary Three species of free-living Antarctic fellfield nematodes, Eudorylaimus coniceps, E. spaulli and E. pseudocarteri exhibited differing degrees of both strategies of cold-hardiness; freeze-tolerance and freeze-avoidance. Bimodal distributions of supercooling points were obtained from monthly field samples of both E. coniceps and E. spaulli. Individuals found in the low group of this distribution (supercooling to <-15 °C) were capable of avoiding freezing by extensive supercoolint (to a mean temperature of ca-22 °C), sufficient to with-stand the environmental extremes of the maritime Antarctic. The high groups of both these species, and almost all E. pseudocarteri, were likely to have frozen at some stage during winter. Survival of freezing increased with the temperature at which nucleation occurred, and has been described by a Gompertz model. Estimates of the supercooling points at which survival fell to 50% were-10.4,-7.1 and-6.1 °C for E. coniceps, E. pseudocarteri and E. spaulli, respectively.  相似文献

Entomopathogenic steinemematid and heterorhabditid nematodes are increasingly used to control insect pests of economically important crops. The rhabditid nematode Phasmarhabditis hermaphrodita (a parasite capable of killing pest slugs) was grown in vitro, in association with a mixed bacterial flora on foam chips impregnated with a kidney‐based nutrient medium in aerated bags, to provide sufficient numbers for laboratory and field experiments. The feasibility of producing nematodes in liquid culture was investigated using 250 ml flasks. Baffled flasks containing 25 or 50 ml or liquid were found to be better than baffled flasks containing 100 ml or unbaffled flasks. Inoculum densities ranging from 50 to 330 ml‐1 did not affect final yield. Dauer larvae in aerated water died rapidly at temperatures of 26–35°C. Survival was progressively better at 22°C and 15°C, and best at 5°C or 10°C.  相似文献

Vitelline-coat lysins from two species of marine mollusc of the genus Tegula, Tegula lischkei and Tegula sp., were purified and their properties compared with those of Tegula pfeifferi. Cross-reaction tests among these three species proved that the lysin action on the vitelline coat was species specific. Each of the lysins from these three species is a single, basic polypeptide with a molecular weight of 16000-17000 and an isoelectric point of pH 10.5. All of them share a common antibody recognition site(s) for the anti-T. pfeifferi lysin antibody. Their amino acid sequences were analyzed using intact lysins and peptides separated by reverse phase high-performance liquid chromatography after V8 proteinase digestion. The amino acid sequences of the N-terminus (Nos 1-66) from the three species showed 98% homology, and those of the C-terminus (Nos 91-118) showed 89% homology. It was concluded that the species specificity of the vitelline coat lysin appears to be determined by a sequence of 24 residues (Nos 67-90) in the middle region of the molecule.  相似文献

Ludwigia adscendens (L.) Hara, Ludwigia parviflora Roxb., and Ludwigia octovalvis (Jacq.) Raven (Onagraceae) are abundant weeds in rice fields in India. These weeds compete with rice for resources in fields and this results in reduction of grain yield. Altica cyanea (Weber) (Coleoptera: Chrysomelidae) is a biocontrol agent of the three rice-field weeds. Hence, it is relevant to study host preference of A. cyanea using volatile cues of these three weeds. Therefore, we attempted to identify volatiles from leaves of the three Ludwigia species attracting A. cyanea, which could be used as an attractant during early emergence of the weeds in rice fields. In Y-tube olfactometer assays, A. cyanea females were more attracted to natural volatiles of plants after 48 h of feeding by adults than to volatiles of undamaged plants. The volatile organic compounds from undamaged plants, and plants after 6 and 48 h of feeding by A. cyanea were identified and quantified by gas chromatography and mass spectrometry (GC–MS) and GC-flame ionization detection (FID), respectively. In total, 25, 29, and 29 volatile compounds were detected in headspaces of undamaged L. adscendens, L. parviflora, and L. octovalvis, respectively, whereas 32, 35, and 34 compounds, respectively, were detected after 48 h of feeding by A. cyanea. Methyl jasmonate predominated among the volatile compounds in all treatments, but this compound was not attractive to A. cyanea. Females were attracted by synthetic blends of 3-hexanol, α-pinene, linalool oxide, and phytol in amounts mimicking those in each of the three Ludwigia species after 48 h of feeding by A. cyanea. The blends mimicking L. adscendens and L. parviflora included geraniol, whereas the blend mimicking L. parviflora also included 1-tridecanol. These synthetic blends may be helpful to monitor A. cyanea in biocontrol programmes.  相似文献

Mastomys rats are present as three distinct karyomorphs in Senegal. Specific assignations were accorded to each form on the basis of chromosomal, reproductive and distribution characteristics. The nematode Phasmarhabditis hermaphrodita is a commercially available biocontrol agent against slugs. This product is especially interesting for use in organic farming, where products containing metaldehyde or carbamates cannot be used for controlling pest slugs. We investigated the potential of P. hermaphrodita for the control of the pest slugs Deroceras reticulatum and Arion lusitanicus. These two species are the most harmful slug pests in Switzerland. At different times of the year, we collected slug specimens of different weight and assessed their susceptibility to P. hermaphrodita in the laboratory. Batches of five slugs were subjected to five different doses of nematodes plus an untreated control and replicated three times. During six weeks, feeding and survival of the slugs were recorded. D. reticulatum was strongly affected by increasing nematode doses, irrespective of the slugs' body weight. In small specimens of A. lusitanicus, feeding and survival were strongly affected by the nematodes, while larger specimens remained almost unaffected. Because A. lusitanicus has an asynchronous development in Switzerland, it seems difficult to control the entire population with a single nematode application. To what extent nematodes will be used in practice for slug control depends on their effectivity against the pest slugs of major importance, on the longevity of the molluscicidal effect and on the price of nematodes.  相似文献

Listeria monocytogenes, the causative agent of listeriosis, has been implicated in increasing foodborne outbreaks worldwide. The disease is manifested in various forms ranging from severe sepsis in immune-compromised individuals, febrile gastroenteritis, still birth, abortions and meningoencephalitis. In India, data from studies on the detection and molecular epidemiological analysis of L. monocytogenes are only recently emerging. The presence of Listeria in different ecological niches has been recorded from India, including foods, soil, vegetables, mangrove swamps, seafood, freshwater fishes, clinical cases, and also insects. The organism has also been isolated from women with spontaneous abortions, miscarriage or recurrent obstetric history, aborted foetuses, animal clinical cases and wildlife samples. A novel species of Listeria has also been characterized. Listeria monocytogenes strains isolated from clinical, environmental, and foods showed biofilm-forming abilities. Listeria monocytogenes serotype 4b isolates of ST328, a predominant and unique ST observed in India, was repeatedly isolated from different sources, times, and geographical locations. Here, we reviewed the occurrence of Listeria in different sources in India, its resistance to biocides, and provide epidemiological analysis on its genomic landscape.  相似文献

Karyotype analyses of three Chirostoma species, C. estor, C. patzcuaro, and C. jordani from Mexico, are presented. C. estor and C. jordani have both a diploid number of 2N = 48 and a fundamental number of NF = 68, but different karyotype formulae. The co-occurrence of these karyotypes with morphometric and allozymatic primitive features so far reported suggests that the ancestral karyotype of the genus Chirostoma was similar to the one shown by these species. The divergent karyotype of C. patzcuaro (2N = 44 and NF = 44) could be related to its endemism and to the relatively small size of its populations.
