Sylvatic plague vaccine (SPV) is a virally vectored bait-delivered vaccine expressing Yersinia pestis antigens that can protect prairie dogs (Cynomys spp.) from plague and has potential utility as a management tool. In a large-scale 3-year field trial, SPV-laden baits containing the biomarker rhodamine B (used to determine bait consumption) were distributed annually at a rate of approximately 100–125 baits/hectare along transects at 58 plots encompassing the geographic ranges of four species of prairie dogs. We assessed site- and individual-level factors related to bait uptake in prairie dogs to determine which were associated with bait uptake rates. Overall bait uptake for 7820 prairie dogs sampled was 70% (95% C.I. 69.9–72.0). Factors influencing bait uptake rates by prairie dogs varied by species, however, in general, heavier animals had greater bait uptake rates. Vegetation quality and day of baiting influenced this relationship for black-tailed, Gunnison’s, and Utah prairie dogs. For these species, baiting later in the season, when normalized difference vegetation indices (a measure of green vegetation density) are lower, improves bait uptake by smaller animals. Consideration of these factors can aid in the development of species-specific SPV baiting strategies that maximize bait uptake and subsequent immunization of prairie dogs against plague.  相似文献

Oral vaccination is an emerging management strategy to reduce the prevalence of high impact infectious diseases within wild animal populations. Plague is a flea-borne zoonosis of rodents that often decimates prairie dog (Cynomys spp.) colonies in the western USA. Recently, an oral sylvatic plague vaccine (SPV) was developed to protect prairie dogs from plague and aid recovery of the endangered black-footed ferret (Mustela nigripes). Although oral vaccination programs are targeted toward specific species, field distribution of vaccine-laden baits can result in vaccine uptake by non-target animals and unintended indirect effects. We assessed the impact of SPV on non-target rodents at paired vaccine and placebo-treated prairie dog colonies in four US states from 2013 to 2015. Bait consumption by non-target rodents was high (70.8%, n?=?3113), but anti-plague antibody development on vaccine plots was low (23.7%, n?=?266). In addition, no significant differences were noted in combined deer mice (Peromyscus maniculatus) and western harvest mouse (Reithrodontomys megalotis) abundance or community evenness and richness of non-target rodents between vaccine-treated and placebo plots. In our 3-year field study, we could not detect a significant positive or negative effect of SPV application on non-target rodents.  相似文献

Plague, caused by the bacterium Yersinia pestis, triggers die-offs in colonies of black-tailed prairie dogs (Cynomys ludovicianus), but the time-frame of plague activity is not well understood. We document plague activity in fleas from prairie dogs and their burrows on three prairie dog colonies that suffered die-offs. We demonstrate that Y. pestis transmission occurs over periods from several months to over a year in prairie dog populations before observed die-offs.  相似文献

Plague, caused by Yersinia pestis, is an exotic disease in North America circulating predominantly in wild populations of rodents and their fleas. Black-tailed prairie dogs (Cynomys ludovicianus) are highly susceptible to infection, often experiencing mortality of nearly all individuals in a town as a result of plague. The fleas of black-tailed prairie dogs are Oropsylla tuberculata cynomuris and Oropsylla hirsuta. We tested the efficiency of O. tuberculata cynomuris to transmit Y. pestis daily from 24 to 96 h postinfection and compared it to previously collected data for O. hirsuta. We found that O. tuberculata cynomuris has over threefold greater transmission efficiency (0.18 infected fleas transmit Y. pestis at 24 h postinfection) than O. hirsuta (0.05 fleas transmit). Using a simple model of flea-borne transmission, we combine these laboratory measurements with field data on monthly flea loads to compare the seasonal vectorial capacity of these two flea species. Coinciding with seasonal patterns of flea abundance, we find a peak in potential for flea-borne transmission in March, during high O. tuberculata cynomuris abundance, and in September–October when O. hirsuta is common. Our findings may be useful in determining the timing of insecticidal dusting to slow plague transmission in black-tailed prairie dogs.  相似文献

A field study was conducted to determine how atmospheric and edaphic conditions influenced the water relations of avocado trees (Persea americana Mill. cv. Bacon). With high and low levels of incident photosynthetically active radiation (PAR, 400–700 nm wave length), and either wet or dry soil, leaf conductance decreased as the absolute humidity difference from leaf to air increased. For any water stress treatment, conductance was higher at high PAR than at low PAR. Both conductance and transpiration were higher in well-watered trees than in stressed trees, and in prestressed trees levels were intermediate to unstressed and stressed trees. A model for water flux through the soil-plant-atmosphere continuum was used to examine the relationship of leaf xylem pressure potential to transpiration in well-watered trees and in trees stressed by dry soil. There was a close linkage between leaf xylem pressure potential and transpiration in unstressed and previously stressed trees with high or low PAR, i.e. similar potentials occurred with equivalent transpiration regardless of previous treatment or time of day. In stressed trees, xylem pressure potential was lower than in unstressed trees both during the day and night, and at a given transpiration rate the potential was lower after 1400 h than before that time. The model indicated that in stressed trees xylem pressure potential was uncoupled from transpiration, presumably because of altered resistance in the soil-root portion of the transport system.  相似文献

The sources of water and corresponding delivery mechanisms to groundwater-fed fens are not well understood due to the multi-scale geo-morphologic variability of the glacial landscape in which they occur. This lack of understanding limits the ability to effectively conserve these systems and the ecosystem services they provide, including biodiversity and water provisioning. While fens tend to occur in clusters around regional groundwater mounds, Ives Road Fen in southern Michigan is an example of a geographically-isolated fen. In this paper, we apply a multi-scale groundwater modeling approach to understand the groundwater sources for Ives Road fen. We apply Transition Probability geo-statistics on more than 3000 well logs from a state-wide water well database to characterize the complex geology using conditional simulations. We subsequently implement a 3-dimensional reverse particle tracking to delineate groundwater contribution areas to the fen. The fen receives water from multiple sources: local recharge, regional recharge from an extensive till plain, a regional groundwater mound, and a nearby pond. The regional sources deliver water through a tortuous, 3-dimensional “pipeline” consisting of a confined aquifer lying beneath an extensive clay layer. Water in this pipeline reaches the fen by upwelling through openings in the clay layer. The pipeline connects the geographically-isolated fen to the same regional mound that provides water to other fen clusters in southern Michigan. The major implication of these findings is that fen conservation efforts must be expanded from focusing on individual fens and their immediate surroundings, to studying the much larger and inter-connected hydrologic network that sustains multiple fens.  相似文献

Based on regional stakeholder preferences and planning guidelines as allocation criteria for SRC, this study aims at providing a transparent approach to evaluate multiple environmental effects and the regional significance of SRC systems. Using the example of two poplar SRC-systems (4-year rotation, 9-year rotation) the potential effects on ground water supply, wind erosion, and biodiversity aspects are evaluated in comparison to arable land for two selected municipalities in the district of Uelzen, Germany. Building on fuzzy membership functions and simple fuzzy-logic rules, the qualitative multi-criteria assessment is transparent and easily to adapt. This approach is transferable to other regions and spatial levels, since it derives from commonly available data and scientific evidence. Results show that implementation of SRC could provide multiple beneficial environmental effects, especially in areas with low landscape heterogeneity. The tools provided allow for a multi-criteria evaluation of environmental effects, and reveal the sensitivity to distinct allocation patterns. Physiographical conditions of the study area implicate a preference for mini-SRC systems. This is supported by smaller decline of annual deep percolation water compared to maxi-SRC. On average, decline in groundwater recharge of mini-SRC (92mm a−1) is comparable to irrigated arable land (80mm a−1), which is common practice in the study area. Currently, the utilization of beneficial environmental SRC effects is quite limited, since only 3 % of arable land is suitable for SRC implementation regarding farmers’ preferences for SRC allocation. Allocation preferences could however change substantially with increasing incentives for SRC, e.g., due to regional bioenergy schemes or “Greening” initiatives within the European Common Agricultural Policy, which is to be reformed by 2013.  相似文献

Canine vaccination has been successful in controlling rabies in diverse settings worldwide. However, concerns remain that coverage levels which have previously been sufficient might be insufficient in systems where transmission occurs both between and within populations of domestic dogs and other carnivores. To evaluate the effectiveness of vaccination targeted at domestic dogs when wildlife also contributes to transmission, we applied a next-generation matrix model based on contract tracing data from the Ngorongoro and Serengeti Districts in northwest Tanzania. We calculated corresponding values of R(0), and determined, for policy purposes, the probabilities that various annual vaccination targets would control the disease, taking into account the empirical uncertainty in our field data. We found that transition rate estimates and corresponding probabilities of vaccination-based control indicate that rabies transmission in this region is driven by transmission within domestic dogs. Different patterns of rabies transmission between the two districts exist, with wildlife playing a more important part in Ngorongoro and leading to higher recommended coverage levels in that district. Nonetheless, our findings indicate that an annual dog vaccination campaign achieving the WHO-recommended target of 70% will control rabies in both districts with a high level of certainty. Our results support the feasibility of controlling rabies in Tanzania through dog vaccination.  相似文献

Effects of Grazing on Restoration of Southern Mixed Prairie Soils   总被引:6,自引:0,他引:6  
A comparative analysis of soils and vegetation from cultivated areas reseeded to native grasses and native prairies that have not been cultivated was conducted to evaluate restoration of southern mixed prairie of the Great Plains over the past 30 to 50 years. Restored sites were within large tracts of native prairie and part of long‐term grazing intensity treatments (heavy, moderate, and ungrazed), allowing evaluation of the effects of grazing intensity on prairie restoration. Our objective was to evaluate restored and native sites subjected to heavy and moderate grazing regimes to determine if soil nutrients from reseeded cultivated land recovered after 30 years of management similar to the surrounding prairie and to identify the interactive influence of different levels of grazing and history of cultivation on plant functional group composition and soils in mixed prairies. For this mixed prairie, soil nitrogen and soil carbon on previously cultivated sites was 30 to 40% lower than in uncultivated native prairies, indicating that soils from restored sites have not recovered over the past 30 to 50 years. In addition, it appears that grazing alters the extent of recovery of these grassland soils as indicated by the significant interaction between grazing intensity and cultivation history for soil nitrogen and soil carbon. Management of livestock grazing is likely a critical factor in determining the potential restoration of mixed prairies. Heavy grazing on restored prairies reduces the rate of soil nutrient and organic matter accumulation. These effects are largely due to changes in composition (reduced tallgrasses), reduced litter accumulation, and high cover of bare ground in heavily grazed restored prairies. However, it is evident from this study that regardless of grazing intensity, restoration of native prairie soils requires many decades and possibly external inputs to adequately restore organic matter, soil carbon, and soil nitrogen.  相似文献

Fish communities in prairie wetlands are extremely dynamic. Due to complete winterkills and periodic colonization, individual basins alternate between supporting a fish population and being fishless. Here we assess the ecological consequences of colonization and subsequent extinction of a fathead minnow population in a prairie wetland. We used a BACI-type approach (before-after control impact) in which data from the colonized wetland were paired with data from a similar fishless site first when both wetlands were fishless (1996), then when the minnow population reached moderate densities in the colonized site (1998), and then again when the colonized site became fishless after treatment with rotenone (1999). Fish colonization resulted in significant increases in turbidity, total phosphorus, and chlorophyll a in the water column; it also caused significant decreases in the abundance of aquatic insects and large cladocerans. Elimination of the minnow population largely reversed the effects of minnow colonization. Our results indicate that characteristics of prairie wetlands can vary as they alternate between supporting fathead minnow populations and being fishless and that ecological characteristics may change rapidly in response to minnow colonization or elimination. Background

Climate change directly affects species by altering their physical environment and indirectly affects species by altering interspecific interactions such as predation and competition. Recent studies have shown that the indirect effects of climate change may amplify or counteract the direct effects. However, little is known about the the relative strength of direct and indirect effects or their potential to impact population persistence.

Methodology/Principal Findings

We studied the effects of altered precipitation and interspecific interactions on the low-density tiller growth rates and biomass production of three perennial grass species in a Kansas, USA mixed prairie. We transplanted plugs of each species into local neighborhoods of heterospecific competitors and then exposed the plugs to a factorial manipulation of growing season precipitation and neighbor removal. Precipitation treatments had significant direct effects on two of the three species. Interspecific competition also had strong effects, reducing low-density tiller growth rates and aboveground biomass production for all three species. In fact, in the presence of competitors, (log) tiller growth rates were close to or below zero for all three species. However, we found no convincing evidence that per capita competitive effects changed with precipitation, as shown by a lack of significant precipitation × competition interactions.

Conclusions/Significance

We found little evidence that altered precipitation will influence per capita competitive effects. However, based on species'' very low growth rates in the presence of competitors in some precipitation treatments, interspecific interactions appear strong enough to affect the balance between population persistence and local extinction. Therefore, ecological forecasting models should include the effect of interspecific interactions on population growth, even if such interaction coefficients are treated as constants.  相似文献

ALK (anaplastic lymphoma kinase) is a transmembrane receptor tyrosine kinase, initially discovered as part of the NPM-ALK fusion protein, resulting from a chromosomal rearrangement frequently associated with anaplastic large cell lymphomas. The native ALK protein is normally expressed in the developing and, at a weaker level, adult nervous system. We recently demonstrated that ALK is a novel dependence receptor. As such, in the absence of ligand, the ALK receptor is kinase inactive and its expression results in enhanced apoptosis, whereas kinase activation, due to a ligand or constitutive as in NPM-ALK, decreases apoptosis. Unligated/kinase unactivated ALK receptor facilitates apoptosis via its own cleavage by caspases, a phenomenon allowing the exposure of a proapoptotic juxta-membrane intra-cellular domain. This review summarizes the biological significance of the ALK receptor in cancer and development, in perspective with its dependence receptor function. The dual function of ALK in the physiology of development is illustrated in the visual system of Drosophila. In this part of the nervous system, ALK in the presence of ligand appears essential for axonal guidance, whereas in the absence of ligand, ALK expression can lead to developmental neuronal apoptosis. ALK is also found expressed in neural crest-derived tumors such as human neuroblastomas or glioblastomas but its role is not fully elucidated. However, an excessive or constitutive ALK tyrosine kinase activation can lead to deregulation of cell proliferation and survival, therefore to human cancers such as lymphomas and inflammatory myofibroblastic tumors. Our observations could have important implications in the therapy of ALK-positive tumors harboring the chimeric or wild type ALK protein.  相似文献

Cornelia de Lange syndrome (CdLS), a disorder caused by mutations in cohesion proteins, is characterized by multisystem developmental abnormalities. PDS5, a cohesion protein, is important for proper chromosome segregation in lower organisms and has two homologues in vertebrates (PDS5A and PDS5B). Pds5B mutant mice have developmental abnormalities resembling CdLS; however the role of Pds5A in mammals and the association of PDS5 proteins with CdLS are unknown. To delineate genetic interactions between Pds5A and Pds5B and explore mechanisms underlying phenotypic variability, we generated Pds5A-deficient mice. Curiously, these mice exhibit multiple abnormalities that were previously observed in Pds5B-deficient mice, including cleft palate, skeletal patterning defects, growth retardation, congenital heart defects and delayed migration of enteric neuron precursors. They also frequently display renal agenesis, an abnormality not observed in Pds5B−/− mice. While Pds5A−/− and Pds5B−/− mice die at birth, embryos harboring 3 mutant Pds5 alleles die between E11.5 and E12.5 most likely of heart failure, indicating that total Pds5 gene dosage is critical for normal development. In addition, characterization of these compound homozygous-heterozygous mice revealed a severe abnormality in lens formation that does not occur in either Pds5A−/− or Pds5B−/− mice. We further identified a functional missense mutation (R1292Q) in the PDS5B DNA-binding domain in a familial case of CdLS, in which affected individuals also develop megacolon. This study shows that PDS5A and PDS5B functions other than those involving chromosomal dynamics are important for normal development, highlights the sensitivity of key developmental processes on PDS5 signaling, and provides mechanistic insights into how PDS5 mutations may lead to CdLS.  相似文献

Semen cryopreservation is an essential biotechnology in canine reproduction and during the cryopreservation process commonly egg yolk are used. The discrepancy in the egg yolk composition and the potential risk of disease dissemination are obstacles for semen exportation and use. Therefore, studies aiming to substitute egg yolk are extremely important. In this context, soy lecithin contains a low-density lipoprotein fraction, is an interesting alternative. Thus, the objective of this study was to compare extenders based on soy lecithin (several concentrations and forms) with egg yolk during the cryopreservation process of dog sperm. For this purpose, we used twelve dogs. Semen was evaluated at different time points (after refrigeration, glycerolization, and thawing), by motility analysis (CASA) and functional tests (e.g., membrane integrity—eosin/nigrosin, acrosome integrity—fast green/Bengal rose, mitochondrial activity—3’3 diaminobenzidine, Chromatin susceptibility to acid-induced denaturation—SCSA, and susceptibility to oxidative stress—thiobarbituric acid reactive substances). The results indicated that egg yolk and lower concentrations of lecithin had similar effects on mitochondrial activity and motility. Thus, soy lecithin is a potentially viable alternative to egg yolk for the cryopreservation of dog semen.  相似文献

Disturbed coral reefs are often dominated by dense mat- or canopy-forming assemblages of macroalgae. This study investigated how such dense macroalgal assemblages change the chemical and physical microenvironment for understorey corals, and how the altered environmental conditions affect the physiological performance of corals. Field measurements were conducted on macroalgal-dominated inshore reefs in the Great Barrier Reef in quadrats with macroalgal biomass ranging from 235 to 1029 g DW m−2 dry weight. Underneath mat-forming assemblages, the mean concentration of dissolved oxygen was reduced by 26% and irradiance by 96% compared with conditions above the mat, while concentrations of dissolved organic carbon and soluble reactive phosphorous increased by 26% and 267%, respectively. The difference was significant but less pronounced under canopy-forming assemblages. Dissolved oxygen declined and dissolved inorganic carbon and alkalinity increased with increasing algal biomass underneath mat-forming but not under canopy-forming assemblages. The responses of corals to conditions similar to those found underneath algal assemblages were investigated in an aquarium experiment. Coral nubbins of the species Acropora millepora showed reduced photosynthetic yields and increased RNA/DNA ratios when exposed to conditions simulating those underneath assemblages (pre-incubating seawater with macroalgae, and shading). The magnitude of these stress responses increased with increasing proportion of pre-incubated algal water. Our study shows that mat-forming and, to a lesser extent, canopy-forming macroalgal assemblages alter the physical and chemical microenvironment sufficiently to directly and detrimentally affect the metabolism of corals, potentially impeding reef recovery from algal to coral-dominated states after disturbance. Macroalgal dominance on coral reefs therefore simultaneously represents a consequence and cause of coral reef degradation.  相似文献

This article reports the first known study on dog breeding in an Afro-Caribbean community. The study obtained the information on 517 matings through interviews with dog caregivers. Few litters (6.8%) from mongrels (potcakes) resulted from planned matings, whereas 66.5% of matings between purebred dogs were planned. Confinement of the female is often inadequate, and roaming dogs may have been responsible for 24.8% of the litters. The lack of confinement of potcakes has resulted in the perception that potcakes are “responsible” for the companion animal (pet) overpopulation problem; however, potcakes made up the minority (29.4%) of the breeding females. Until regulations concerning dog breeding are introduced, caregivers can be expected to continue exploiting their nonhuman animals to supplement their incomes from the sale of puppies. A consequence of unregulated breeding may also be inbred offspring of “purebred” dogs as few self-styled “professional” breeders appeared to use dogs who were not their own.  相似文献

Fire frequency has significant effects on the biota of tallgrass prairie, including mammals, vascular plants and birds. Recent concern has been expressed that widespread annual burning, sometimes in combination with heavy livestock grazing, negatively impacts the biota of remaining prairie remnants. A common management recommendation, intended to address this problem, is to create a landscape with a mosaic of different burn regimes. Pitfall trapping was used to investigate the impacts of fire pattern on the diversity and species composition of ground beetles (Coleoptera: Carabidae) at Konza Prairie Biological Station in eastern Kansas, USA. Trapping was conducted over three seasons in landscape units burned on average every 1, 4, or 20 years, and in a fourth season across the available range of vegetative structure to assess the variability of the community within the study system. In the fifth season communities were also followed immediately after two fire events to detect within-season effects of fire and to study short-term patterns of post-disturbance community assembly. Fire frequency had comparatively minimal effects on ground beetle diversity measures, and most numerically common species were observed widely across habitat and management types. Fire frequency effects were manifested primarily in changes in abundance of common species. Colonization of burned areas apparently did not occur from juxtaposed non-burned areas, but from underground or from long distances. While these results suggest that widespread annual burning of tallgrass prairie remnants may not have dramatic effects on prairie ground beetles, we urge caution regarding the application of these results to other taxa within tallgrass prairie.  相似文献
