Secondary metabolites of 22 fungal strains (genus Aspergillus, section Usti) isolated at diverse geographic regions, including the Arctic permafrost deposits, were studied. The studied strains were found to synthesize a variety of biologically active compounds, structurally identified as drimane sesqueterpenoids, isoquinoline alkaloids (TMC-120 A?C, derivative 1), meroterpenoids (austalides О and J), and anthraquinone pigments (averufin, versicolorin C). Desferritriacetylfusigen production by A. calidoustus isolates is reported for the first time. The individual spectra of secondary metabolites were used for reidentification of 17 strains, of which 15 were identified as A. calidoustus and two, as A. pseudodeflectus.  相似文献

Species identification of slow-growing fungi of the genus Penicillium isolated from ancient permafrost deposits was performed using micro- and macromorphological characteristics as well as the composition of secondary metabolites. The strains producing clavine ergot alkaloids fumigaclavines A and B and festuclavine were assigned to the species P. palitans Westling 1911, whereas the strains forming ochratoxins A and B were identified as P. verrucosum Dierckx 1901.  相似文献

The shore fly, Scatella stagnalis (Fallén) (Diptera: Ephydridae) is an important insect pest of greenhouse crops. We evaluated two different Spanish isolates of entomopathogenic nematodes, Steinernema feltiae (Filipjev) (Rhabditida: Steinernematidae) and Steinernema arenarium (Artyukhovsky) (Rhabditida: Steinernematidae), and two commercially available strains, Steinernema feltiae (Nemaplus®) and Heterorhabditis bacteriophora (Poinar) (Rhabditida: Heterorhabditidae) (Nematop®) against shore flies. In tests conducted in 24-well plate filter paper applied at 5, 11, 22, 44 and 88 nematodes per larva, all nematodes produced significant shore fly larval mortality. The lowest concentration tested was enough to obtain high larval mortality (65.2–87.0%). The nematodes Steinernema feltiae and Steinernema arenarium, which parasitized the shore fly larvae faster, also penetrated in higher number in the shore fly larva (4.6–8.8% penetration rate). In bioassays conducted in algae, Steinernema feltiae, applied at 50 nematodes/cm2, caused highest (100%) and Steinernema arenarium lowest shore fly mortality (94%). Our results suggest that entomopathogenic nematodes appear feasible for controlling shore flies but further tests are needed to determine their efficacy in the field.  相似文献

Earliest middle Cambrian rocks in the Franconian Forest, formerly known as the ‘Galgenberg Formation’, include a moderately diverse fauna with a characteristic West Gondwanan, Atlas-type trilobite assemblage with often surprisingly well-preserved specimens. The hitherto inadequately characterised and poorly described assemblage includes Kingaspidoides frankenwaldensis, K. sp. aff. usitata, K. alberti sp. nov., K. meieri sp. nov., K.? sp. A, Ornamentaspis cf. crassilimbata, Latikingaspis sp. aff. alatus, Enixus sp. aff. juvenis, Acadoparadoxides sp. A, Parasolenopleura wurmi sp. nov., Parasolenopleura parabolica sp. nov. and Acanthomicmacca franconica Geyer, 2016. In addition to precise documentation of the species’ morphology and ontogenetic development, this study exemplifies allometric developments during the ontogeny of ellipsocephaloid and early solenopleurid trilobites, particularly Kingaspidoides and Parasolenopleura, and effects of deformation and distortion caused by diagenesis and tectonics. It further discusses the aspects of the trilobites’ ecology and taphonomy, and it characterises generic differences within the Kingaspis clade, particularly of Kingaspidoides, Latikingaspis and Ornamentaspis.  相似文献

The bacterial species of the genus Xenorhabdus in the family Enterobacteriaceae have a mutualistic association with steinernematid entomopathogenic nematodes (EPNs), which have been used as biological control agents against soil insect pests. In this study we present the genetic and phenotypic characterizations of the Xenorhabdus species isolated from steinernematid nematodes in Japan. The 18 Japanese Xenorhabdus isolates were classified into five bacterial species based on 16S ribosomal RNA (16S rRNA) gene sequences: Xenorhabdus bovienii, Xenorhabdus hominickii, Xenorhabdus indica, Xenorhabdus ishibashii, and Xenorhabdus japonica. There was no genetic variation between the 16S RNA sequences among the three X. ishibashii isolates, 0–0.1% variation among the five X. hominickii isolates, and 0–0.5% among the eight X. bovienii isolates. Phenotypic characterization demonstrated that representative isolates of the five bacterial species shared common characteristics of the genus Xenorhabdus, and only X. hominickii isolates produced indole. Symbiotic association and co-speciation of Xenorhabdus bacteria with Steinernema nematodes from Japan are discussed.  相似文献

The present work describes the immune response of wireworm larvae, Agriotes lineatus (L.) (Coleoptera: Elateridae) when challenged with two species of entomopathogenic nematodes, Steinernema feltiae (Filipjev) (Strongyloidea: Steinernematidae) and Heterorhabditis bacteriophora (Poinar) (Rhabiditoidea: Heterorhabditidae). Two main immunological processes including cellular and humoral reactions have been addressed. Total haemocyte counts after infection with H. bacteriophora increased quickly in initial times, but decreased over time post-injection (at 12 and 16 h). Instead, haemocyte numbers after infection with S. feltiae was unchanged in the early stage, but significantly decreased until 16 h post-injection. Plasmatocytes and granulocytes showed more significant changes compared to other haemocytes. The encapsulation response to parasites was significantly different against two nematode species. Particularly, S. feltiae was almost unrecognized by host haemocytes (5.85 % of encapsulated parasites). Assays with H. bacteriophora showed 23.5 % of encapsulated nematodes. From 8 to 12 h after H. bacteriophora infection, an increase in phenoloxidase activity was detected, while in the larvae injected with S. feltiae the enzymatic activity decreased gradually reaching the lowest level 16 h post-injection. This is the first report on the modulation of immune response of wireworm larvae after infection with entomopathogenic nematodes.  相似文献

Deladenus (=?Beddingia) siricidicola (Tylenchida: Neotylenchidae) is the most effective biocontrol agent used against the invasive wood wasp, Sirex noctilio (Fabricius) (Hymenoptera: Siricidae). The nematodes feed and reproduce on the wood-inhabiting fungus, Amylostereum areolatum (Chaillet ex Fr.) Boidin (Russulales: Amylostereaceae) and parasitise larvae of S. noctilio. In the nematode biocontrol program, the nematodes are inoculated into herbicide-weakened ‘trap trees’. Recent declines in nematode parasitism of S. noctilio in Australia have coincided with an increased incidence of an exotic bark beetle, Ips grandicollis (Eichhoff) (Coleoptera: Curculionidae), attacking trap trees and vectoring a wood-inhabiting fungus, Ophiostoma ips (Rumbold) Nannfelt (Ophiostomatales: Ophiostomataceae), which may inhibit migration of the nematode within the tree to the detriment of S. noctilio biocontrol. Several in vitro and in vivo experiments were conducted to investigate the effect of fungal interactions on the ability of D. siricidicola to locate and reproduce on A. areolatum. Deladenus siricidicola showed preference to A. areolatum in the presence and absence of O. ips, but the presence of O. ips negatively affected the choice response and the number of eggs laid by the nematodes. Deladenus siricidicola was unable to survive and reproduce on O. ips. Results give a clearer understanding of the choice response of D. siricidicola in I. grandicollis infested trees, explaining the disruptive impact of bark beetles on biocontrol of S. noctilio, an effect that could extend from Australia to other important pine growing countries.  相似文献

Ectomycorrhizal (ECM) and ectendomycorrhizal fungal species associated with Pinus montezumae were recorded in 8 year-old trees established in microcosms and compared with those associated with 2 year-old trees, in order to determine their persistence over the long-term. Mycorrhizal root tips were morphologically and anatomically characterized and sequenced. The extension of extramatrical mycelium of ECM fungi with long exploration strategies was evaluated. In total, 11 mycorrhizal species were registered. Seven mycorrhizal species were detected on both 2 and 8 year-old pines: Atheliaceae sp., Rhizopogon aff. fallax, R. aff. occidentalis, Suillus pseudobrevipes, Tuber separans, Wilcoxina mikolae and Wilcoxina rehmii. One species, Thelephora terrestris, was exclusively associated with two year–old seedlings, while Cenococcum geophilum, Pezizaceae sp. and Pyrenomataceae sp. were exclusively found on 8 year-old trees. Atheliaceae sp. was the ECM fungal species that presented the most abundant mycelium. Finally, we report one new fungal species of Pezizaceae occurring as a symbiont of P. montezumae.  相似文献

Single infective juveniles of Heterorhabditis bacteriophora, H. megidis (Nematoda: Heterorhabditidae), Steinernema arenarium, S. carpocapsae and S. feltiae (Nematoda: Steinernematidae) were used to infect single Galleria mellonella (Lepidoptera: Pyralidae) larvae. Four parameters of entomopathogenic nematodes pathogenicity were assessed: the mortality of insects, infectivity of nematodes, number of nematodes established per single G. mellonella, and degree of infective juveniles colonization (percent of infective juveniles which intestine was colonized by symbiotic bacteria). The accuracy, repeatability, and versatility for different species of EPNs in bioassay arenas were compared. Our modifications of the original methods yielded ~ 50% higher efficiency of infective juveniles in cell culture plates and > 20% higher efficiency in centrifuge test tubes. The efficiency of nematodes in cell culture plates (39–77%) was relatively low, especially in the case of Heterorhabditis spp. In the bioassay arena, infective juveniles migrated between cells. The results of our studies indicate that the pathogenicity of EPNs should be assessed in centrifuge test tubes. In these arenas, the infectivity of single IJs was ~ 90% for Heterorhabditis spp. and ~ 95% for Steinernema spp. The degree of colonization of the EPN isolates by symbiotic bacteria was in the range of 96–98%.  相似文献

Turgor is very important for the invasive growth of fungal pathogens. Glycerol, a highly osmotic solvent, is considered to play an important role in turgor generation. The nematophagous fungus Arthrobotrys oligospora mainly lives as a saprophyte. In the presence of nematodes, A. oligospora enters the parasitic stage by forming three-dimensional networks (traps) to capture nematodes. In A. oligospora, we found that glycerol accumulated during nematode-induced trap formation. We demonstrated that deleting gph1, which encodes glycogen phosphorylase, decreased the glycerol content, compared with that of a wild-type strain. Although the number of traps induced by nematodes was not affected in the Δgph1 mutant, the capture rate was lower. Meanwhile, deleting gph1 also affected the growth rate and conidiation capacity of the fungus. These results indicate that glycerol derived from GPH1 is essential for the full virulence of A. oligospora against nematodes.  相似文献

Photorhabdus is an insect-pathogenic Gram negative enterobacterium found in the gut of Heterorhabditis nematodes. Photorhabdus is highly virulent to insects, and can kill insects rapidly upon injection at very low concentrations of one to few cells. We characterized the virulence of Photorhabdus symbionts isolated from the Heterorhabditis nematodes collected from various parts of India by injecting different concentrations of bacterial cells into fourth instar larval stage of insect Galleria mellonella. Photorhabdus luminescens ssp. akhurstii strain IARI-SGMG3 from Meghalaya was identified as the most virulent of all the tested strains on the basis of LT50 and LC50 values. This study forms a basis for further investigations on the genetic basis of virulence in Photorhabdus bacteria.  相似文献

The European Miocene records a wide diversity of hyaenid ecomorphotypes represented by multiple genera. Among these, Hyaenictis Gaudry, 1861, is one of the least known. This genus includes four species from the late Miocene and Pliocene of the Old World, but in Europe Hyaenictis is only represented by two species, recorded by scarce and fragmentary remains: Hyaenictis graeca Gaudry, 1861, from Pikermi (MN12; Greece) and Hyaenictis almerai Villalta Comella and Crusafont Pairó, 1948, from Sant Miquel de Toudell (MN10; Vallès-Penedès Basin, NE Iberia). Here, we describe a new skull of Hyaenictis aff. almerai from the Vallès-Penedès site of Ronda Oest Sabadell Sector D (MN10), representing the most complete European specimen of the genus. In the presence of m2 and virtual lack of m1 metaconid, the described cranium more closely resembles Hyaenictis rather than any other medium- to large-sized European hyaenid. However, the new skull does not fit well with previously known Hyaenictis species, more closely resembling the bone-cracking Adcrocuta Kretzoi, 1938, in the development of premolar accessory cuspids and the possession of relatively broad cheek teeth. These and other features (strong mandibular muscular insertions and enamel microstructure) denote more durophagous adaptations than previously documented in Hyaenictis (considered a cursorial/dog-like hyaena), and favor the inclusion of H. aff. almerai in the transitional bone-cracking hyaenid ecomorphotype.  相似文献

Ostracodes from the Gur'yano-ka Formation (Upper Ordo-ician, Sandbian and Katian stages) of Gorny Altai, Russia, are studied for the first time. Some localities with fossil ostracodes of this age are described briefly. Ele-en new species of the genera Ballardina, Eurychilina, Laccochilina, Bolbina, Retiprimites, Pseudozygobolbina, Bullaeferum, Soanella, Egoro-ella, and Allertemmysa are described and figured. The pre-iously described species Leperditella aff. par-ipunctata V. Ivanova, 1959, Grammolomatella valdari V. Ivanova et Melnikova 1977, Longidorsa? baltica Meidla, 1996, and Steusloffina cuneata (Steusloff, 1895) and ostracodes in open nomenclature, Bollia sp., Klimphores sp., Rectella sp., Longiscula sp., are only figured.  相似文献

Aggressive behaviour can ensure animal access to local resources. To reduce constant costs in the defence of territories, species could save energy with conflicts avoiding aggression with neighbour or in situations with abundance of resources. In the present study, we analysed the effect of distance among colonies and resource availability on the aggression level and responses to chemical cues of Nasutitermes aff. coxipoensis (Holmgren) (Termitidae: Nasutitermitinae). Manipulation of resource offer was conducted in the field, where nests with different distances were kept without addition of baits (control), with addition of three or 16 sugarcane baits/nest. After 3 months, aggressiveness, linear and Y-shaped trail-following bioassays were carried out with all pairwise combinations of colonies in each treatment. Our results showed that aggressive index of N. aff. coxipoensis was affected by the resource availability. However, individuals from colonies with 0 and 3 baits/nest showed a higher number of fighting with neighbours than those from non-neighbours colonies. Termite workers from colonies without baits (control) followed shorter distance in the linear trails compared to those from colonies with addition of baits. In all treatments, there was no preference of workers in relation to the choice of chemical cues from own or other colonies. The response of intercolonial aggressiveness in N. aff. coxipoensis seems to be resource-dependent. These results may contribute to the comprehension of the use of space by N. aff. coxipoensis and could be useful to explain patterns of termite co-occurrence at different spatial scales, from local (inside the nest—e.g. cohabitation of nests by inquilines) to regional (e.g. around the nest).  相似文献

Reactive oxygen species (ROS) produced by NADPH oxidases can serve as signaling molecules to regulate a variety of physiological processes in multi-cellular organisms. In the nematophagous fungus Arthrobotrys oligospora, we found that ROS were produced during conidial germination, hyphal extension, and trap formation in the presence of nematodes. Generation of an AoNoxA knockout strain demonstrated the crucial role of NADPH oxidase in the production of ROS in A. oligospora, with trap formation impaired in the AoNoxA mutant, even in the presence of the nematode host. In addition, the expression of virulence factor serine protease P186 was up-regulated in the wild-type strain, but not in the mutant strain, in the presence of Caenorhabditis elegans. These results indicate that ROS derived from AoNoxA are essential for full virulence of A. oligospora in nematodes.  相似文献

Exometabolites of 22 strains of the genus Penicillium, section Chrysogena isolated from low-temperature ecotopes of various geographical regions were analyzed. The ecotopes included permafrost deposits, frozen volcanic ash, a fossil horse, cryopeg, and water from an Antarctic lake. The studied strains were found to contain exometabolites belonging to the groups of penicillins (penicillin G), chrysogines (chrysogine, 3-acetyl-quinazolin-4(3H)-one, 2-pyruvoylaminobenzamide, 2-(2-hydroxypropionylamino)-benzamide, and questiomycin A), roquefortines (3,12-dihydroroquefortine, roquefortine, glandicolines A and B, and meleagrine), xanthocillins (xanthocillin X), and simple tryptophan derivatives (N-acetyltriptamine and indoleacetic acid). In five P. chrysogenum strains and three P. nalgiovense strains, a correlation was found between exometabolite spectra and morphological characteristics of the cultures isolated from modern ecotopes. For other strains species, identification was based on morphological features due to the absence of biosynthesis of penicillin G on the major chemotaxonomic markers for these species.  相似文献

Background

The karyotypes of Leptodactylus species usually consist of 22 bi-armed chromosomes, but morphological variations in some chromosomes and even differences in the 2n have been reported. To better understand the mechanisms responsible for these differences, eight species were analysed using classical and molecular cytogenetic techniques, including replication banding with BrdU incorporation.

Results

Distinct chromosome numbers were found: 2n = 22 in Leptodactylus chaquensis, L. labyrinthicus, L. pentadactylus, L. petersii, L. podicipinus, and L. rhodomystax; 2n = 20 in Leptodactylus sp. (aff. podicipinus); and 2n = 24 in L. marmoratus. Among the species with 2n = 22, only three had the same basic karyotype. Leptodactylus pentadactylus presented multiple translocations, L. petersii displayed chromosome morphological discrepancy, and L. podicipinus had four pairs of telocentric chromosomes. Replication banding was crucial for characterising this variability and for explaining the reduced 2n in Leptodactylus sp. (aff. podicipinus). Leptodactylus marmoratus had few chromosomes with a similar banding patterns to the 2n = 22 karyotypes. The majority of the species presented a single NOR-bearing pair, which was confirmed using Ag-impregnation and FISH with an rDNA probe. In general, the NOR-bearing chromosomes corresponded to chromosome 8, but NORs were found on chromosome 3 or 4 in some species. Leptodactylus marmoratus had NORs on chromosome pairs 6 and 8. The data from C-banding, fluorochrome staining, and FISH using the telomeric probe helped in characterising the repetitive sequences. Even though hybridisation did occur on the chromosome ends, telomere-like repetitive sequences outside of the telomere region were identified. Metaphase I cells from L. pentadactylus confirmed its complex karyotype constitution because 12 chromosomes appeared as ring-shaped chain in addition to five bivalents.

Conclusions

Species of Leptodactylus exhibited both major and minor karyotypic differences which were identified by classical and molecular cytogenetic techniques. Replication banding, which is a unique procedure that has been used to obtain longitudinal multiple band patterns in amphibian chromosomes, allowed us to outline the general mechanisms responsible for these karyotype differences. The findings also suggested that L. marmoratus, which was formerly included in the genus Adenomera, may have undergone great chromosomal repatterning.

Fungus gnats (FG) have been reported in Venezuelan greenhouses recently. Bradysia difformis Frey (Diptera, Mycetophilidae) was found attacking Daisy gerbera plants [Gerbera sp. L. (Asterales: Asteraceae)] and many other crops in the country causing serious damages. Heterorhabditis amazonensis Andaló et al. (Rhabditida: Heterorhabditidae), was used to assess the mortality of larvae and adults of FG, number of invader nematodes per larvae and to compare the mortality of FG using nematodes and/or ciromazine under laboratory conditions. In a commercial flower farm, different doses of these nematodes were applied for eight weeks to control FG. The results showed 95% of mortality of the B. difformis larvae under laboratory conditions and performed better in the field when 50,000 nematodes were applied in every pot as a base dose to initiate a control programme. According to our results, H. amazonensis has the potential to become a regular organism for controlling fungus gnat larvae in tropical greenhouses.  相似文献
